一株抗重金属铜镉细菌的分离、鉴定及其16S rDNA的序列分析   总被引:12,自引:0,他引:12  
从湖北省大冶市矿区土壤中分离到一株高抗铜和镉的菌株,命名为NTG-01。该菌株可以单抗4.5mmol/L的铜和2mmol/L的镉,因此它是研究抗铜或镉机制的重要菌株。对分离到的NTG-01进行了形态学观察和生理生化鉴定以及16S rDNA序列分析。结果显示菌株NTG-01为细菌,革兰氏染色阴性,短杆状,鞭毛周生,菌体大小约为0.8μm×2.0μm,V-P实验阳性,甲基红实验阴性,利用葡萄糖产酸产气;通过对菌株NTG-01的16S rDNA序列进行测定和同源性比对,发现它与产气肠杆菌的16S rDNA有高达99%的同源性,结合形态和生理生化指标,将其鉴定为产气肠杆菌(Enterobacter aerogenes)。通过测定NTG-01对9种重金属的M IC值,可知它对多种重金属具有普遍较高的抗性。  相似文献

从土壤微生物中筛选得到一株产神经节苷脂内切糖苷酶的菌株YW2112, 该酶能特异性水解神经节苷脂中连接神经酰胺和寡糖链之间的糖苷键,是研究神经节苷脂结构与功能的重要工具酶。对分离菌株YW2112进行了形态、生理生化鉴定及16S rDNA序列分析。菌株YW2112为细菌,革兰氏染色阴性,直杆状, 鞭毛周生,菌体大小为(0.6μm~1μm)×(1μm~3μm),V-P实验阳性,甲基红阴性,利用葡萄糖产酸产气。以16S rDNA同源性为基础构建了包括11株相关种属细菌在内的系统发育树,在系统发育树中,分离菌株YW2112 与Enterobacter cloacae 在同一分支,二者的序列相似性为98.6% ,结合形态和生理生化鉴定,将其鉴定为阴沟肠杆菌(Enterobacter cloacae)。  相似文献

采用富集筛选法从杜仲根中分离到5株具有ACC脱氨酶活性的内生细菌, 利用纸片法测定它们的抑菌活性, 通过形态特征、生理生化试验和16S rRNA序列分析对分离菌株进行鉴定。结果显示, 5株杜仲内生细菌均具有较高的ACC脱氨酶活性, 其中4株菌对大肠杆菌CGMCC1.1103和枯草芽孢杆菌CGMCC1.769均有较好的抑菌活性, 通过生理生化试验和16S rRNA序列分析, 将菌株JDM-2、JDM-8、JDM-11、JDM-14和JDM-19分别鉴定为Pseudomonas koreensis、肺炎克雷伯氏菌(Klebsiella pneumoniae)、路德维希肠杆菌(Enterobacter ludwigii)、变栖克雷伯氏菌(Klebsiella variicola)和阿氏肠杆菌(Enterobacter asburiae)。  相似文献

本文对大肠杆菌(Escherichia coli)不同菌株和肠杆菌科(Enterobacteriaceae)不同属其他三种菌株,即普通变形菌(Proteus vulgaris)、粘质沙雷氏菌(Serratia marcescens)和产气肠杆菌(Enterobacter aerogenes),分别进行4.5S RNA基因聚合酶链式反应(PCR),然后对扩增产物作依赖于序列的单链构象多态性(SSCP)分析.实验结果表明,上述细菌4.5S RNA基因的大小和正链构象均无可觉察的差异,仅产气肠杆菌的负链构象有明显不同.由此可见4.5S RNA基因在进化上相当保守,产气肠杆菌4.5S RNA基因的序列虽有改变,仍能维持其有义链的基本构象.  相似文献

【目的】筛选具有1-氨基环丙烷-1-羧酸(简称ACC)脱氨酶活性的菌株,并探索该类菌的促生作用,有助于研发微生物肥料,实现农业增产。【方法】以ACC为唯一氮源,从土壤中富集和分离ACC脱氨酶产生菌;测定ACC脱氨酶的比活力,对酶活性最强的菌株根据形态和培养特征、生理生化特征及16S rDNA序列进行分类鉴定;分别采用菌液培养接种法与菌液浸种接种法初步研究该菌株对紫花苜蓿幼苗生长的促生作用。【结果】筛选得到6株ACC脱氨酶阳性细菌,其中菌株ACC 30酶活性最高,为0.217 U/mg,根据培养特征观察和生理生化指标测定结果,结合16S rDNA序列比对分析,确定ACC 30为产气肠杆菌(Enterobacter aerogenes)。促生试验表明,ACC 30可促进紫花苜蓿幼苗根伸长,菌液培养接种法与菌液浸种接种法两种处理方法下ACC 30分别使幼苗根相对伸长135%、136%,促生作用均明显且基本一致。但是,两种方法处理下ACC 30均抑制幼苗下胚轴伸长。【结论】筛选获得ACC脱氨酶活性菌株ACC 30,其酶活性较高且促生作用明显,有望进一步研发成为微生物肥料。  相似文献

从土壤中分离到一株产红色素的细菌H31,根据该菌株的16S rDNA序列与GenBank中已有序列比对的结果,并结合菌落形态和常规生理生化鉴定方法对该菌株进行鉴定,结果表明:该菌在细菌分类学上属于肠杆菌科,其与黏质沙雷氏菌(Serratia marcescens)同源性最高,但该菌株生理生化实验中的赖氨酸脱羧酶、鸟氨酸脱羧酶以及精氨酸双水解酶实验与报道的S. marcescens的结果不一致,为一株新的黏质沙雷氏菌S. marcescens H31。通过紫外光谱、质谱和核磁共振等方法分析,确定其产生的红色素为灵菌红素。  相似文献

从池塘患烂鳃病的斑鳠(Mystus guttatus)亲鱼病灶中分离出一株毒力较强的致病菌-Mg2,经形态学观察、生理生化特性和16S rRNA基因序列分析鉴定,其主要特征为:菌体细长,无鞭毛和荚膜,革兰氏阴性,大小0.5×(6.5-11)μm,菌落黄色,边缘不整齐呈假树根状。氧化酶、过氧化氢酶阳性,分解酪素和明胶,硝酸盐还原阳性;不分解纤维素、几丁质、酪氨酸、七叶灵和淀粉,吲哚和葡萄糖产气阴性。对恩诺沙星、诺氟沙星、萘啶酸、红霉素、洁霉素敏感,菌株的16S rRNA基因序列分析结果表明:Mg2菌株与柱状黄杆菌聚类,基因序列的同源性在97.5%以上,综合生理生化、分子生物学两方面的分类鉴定结果,Mg2菌株应归类鉴定为柱状黄杆菌(Flavobacterium Columnare)。  相似文献

从受石油污染的红树林湿地土样中分离筛选对烷烃具较高降解能力的细菌菌株, 以期应用于被石油污染滨海湿地的生物修复。采用富集培养方法, 富集、分离和筛选烷烃降解菌;观察各菌落及菌体形态特征;测试菌株Z2的生理生化特征, 并用16S rDNA序列分析方法进行该菌种鉴定。分离筛选得到Z1、Z2和Z3这3个能以正十六烷为唯一碳源生长的菌株, 其降解率依次为63.4%、82.5%和78.3%, 其中菌株Z2的降解率最高。经过形态学观察、生理生化特性实验和16S rDNA序列分析鉴定, 菌株Z2为不动杆菌(Acinetobacter sp.)。  相似文献

从活性污泥中分离筛选得到一株能代谢甘油生产1,3-丙二醇(1,3-PD)的菌株2-1,通过形态学鉴定、生理生化试验、16S rRNA序列分析对菌株分类学地位进行鉴定,用MEGA 4.1软件构建的系统发育树显示菌株2-1与Klebsiella pneumoniae(CP001891)的亲缘关系最近。16S rDNA序列同源性比较发现,菌株2-1与模式菌株同源率为95.4%,疑似为新种。对菌株2-1在5 L发酵罐中进行发酵特性研究,分批补料发酵时得到较高的1,3-PD终浓度,达到63.5 g/L,此时生产强度为2.19 g/(L.h),底物转化率0.64 mol/mol。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
