Polymorphism for the phenotypical expression of gene for LDH subunit A was demonstrated in sheep erythrocytes by means of starch gel electrophoresis. Three types of LDH were established differing in the relative activities of the second and third LDH isozymes. Familial analysis has shown that high level of LDH-2 and LDH-3 activities are determined by the autosomal gene Ldr-1a and low level of their activities is controlled by gene Ldr-1b. The frequencies of gene Ldr-1a in Altai thin-fleece, Romney Marsh and Linkoln breeds were 0,533; 0,509 and 0,425 respectively. Gene frequencies were also determined in crossbreeds. Possible mechanisms of the action of genes Ldr-1a and Ldr-1b in sheep erythrocytes are discussed.  相似文献

The spectrum of lactate dehydrogenase (LDH) isozymes in erythrocytes of silver foxes was investigated by means of electrophoretic and immunochemical methods. By means of electrophoresis it is shown that differences in LDH isozyme spectrum between the animals homozygous for the gene Ldr-1a and those homozygous for the gene Ldr-1b are most conspisions at the age of 90-100 days of postnatal development. By means of the immunochemical method three groups of animals are distinguished differing in the LDH content in erythrocytes: the animals with a high LDH content (81.0 mcg/ml) and with a low LDH content (54.14 mcg/ml), which are homozygous for the gene Ldr-1b and Ldr-1a respectively, and the animals with intermediate LDH content (64.58 mcg/ml), which are heterozygous. The data obtained suggest that the effect of the gene Ldr-1a is associated with the decrease of the quantity of A subunits of LDH. It is assumed that the mechanism of the gene Ldr-1a action is realized either by means of the decrease of the synthesis of the A LDH subunits, or by means of the increase of the rate of their degradation.  相似文献

Patterns of lactate dehydrogenase (LDH) isozymes of tissues from various mouse strains were examined. An interstrain polymorphism for LDH isozymes of liver was established. One phenotype (CBA/Lac and AKR/J mice) yielded a five-banded LDH pattern, another phenotype (DBA/1J, DBA/2J, C57BL/6J and C3H/He) showed a three-banded one. Immunochemical evidence was obtained indicating that differences in the LDH pattern are mainly due to different contents of the B subunit of LDH. Linkage tests indicated that the locus Ldr-2 determining the amounts of the LDH B subunit in mouse liver tissue is located in chromosome 6, 19 ± 4.1 cm away from the earlier described Ldr-1 locus. The effect of locus Ldr-2 is strictly tissue-specific; it is manifest only on days 6–8 after birth.  相似文献

Lactate dehydrogenase (LDH; E.C. 1.1.1.27) isozymes were compared in three inbred strains of mice, and two strains of wild mice, as well as the F1 hybrids and other genetic crosses involving two of the inbred strains. The strains examined were NZB/B1NJ, 129/J and C57BL/6J, Mus musculus molossinus and M. musculus castaneus. Genetic crosses were made between the xenotropic virus-producing NZB and the non-virus producing 129/J mice. Tissue specificity of LDH in these strains was studied using homogenates of kidney, liver, spleen and thymus. Polymorphism of the enzyme was studied by agarose gel electrophoresis. Enzyme polymorphism in the tissues of NZB and 129/J has not been previously reported. The liver and spleen tissues of 129/J showed the absence of LDH-1 and LDH-2 isozymes. Thymic homogenates of NZB showed a lack of expression of LDH-1, LDH-2 and LDH-3 isozymes. The F1, F2 and the backcross progeny from genetic crosses involving NZB, and 129/J mice showed an isozyme pattern more similar to the non-virus-producing 129/J strain than the virus-producing NZB. Evidence of genetic regulation at the LDH-B subunit appears to be the reason for the differential expression of the isozymes in NZB and 129/J strains. The other inbred strain of mice, C57BL/6J, also showed a greater similarity to the 129/J strain than NZB. The two strains of wild mice were similar in their expression of LDH-isozymes between each other and to the 129/J strain, with respect to the liver and spleen tissues.  相似文献

Normal tissues of DBA, CBA, CC57W, C3H, Balb/c, SHR mice and F1 hybrids CC57W/DBA appeared to differ in the ratios of mitochondrial and supernatant NADP-dependent isocitrate dehydrogenase (IDH). Tested inbred mice strains CC57W, C3H, SHR, Balb/c contain allelic form Idh-1a of supernatant IDH gene Idh-1, whereas allelic form Idh-1b is characteristic of mice strains DBA and CBA. In tumors IDH isozymes have the same mobility as do isozymes of homologous normal tissues; but their activity is lower. A high variability of each isozyme activity in the isozyme spectrum is revealed in various tissues of F1 hybrids CC57W/DBA. Allelic forms of gene Idh-1 were used as markers of normal and tumor cells for the experimental model: transplantation of sarcoma 37 (Idh-1a/Idh-1a) to subcutaneous tissue of the mouse strain DBA (Idh-1b/Idh-1b). It enables us to reveal isozymes of stromal cell in tumor IDH isozyme spectrum. The results indicate that the relation of normal and tumor isozymes vary in different tumors.  相似文献

The dynamics of changes in the lactate dehydrogenase (LDH) spectra during organogenesis in CBA mice has been studied by means of ultramicroelectrophoresis. The embryonic period of development is characterized by the predominance of cathodic isozymes (LDH-5 and LDH-4) in all the organs under study. The increase of anodic isozymes (LDH-1 and LDH-2) takes place in the heart, kidneys and brain as the development proceeds. The first reliable differences in the LDH spectra of different organs appear on the 11th day of embryogenesis. On the basis of comparison with the help of criterion gamma, the LDH spectra of the organs under study can be divided into two groups: I--heart, lungs, kidneys and brain (tendency towards the increase of H-subunits) and II--intestine, liver and muscles (tendency towards the increase of of M-subunits). The LDH spectra of adult animals are divided into 4 distinct groups: I--heart and kidneys, II--brain, III--lungs and muscles, IV--liver.  相似文献

1. Effect of pH on liver lactate dehydrogenase (LDH) and its isozymes was examined in the goldfish acclimated to different temperatures and some purification of the LDH was attempted. 2. The optimal pH and the Km value at 30 degrees C of the enzyme were independent of acclimation temperature. 3. the optimal pH of isozyme was more basic in the order of LDH-1, LDH-2, LDH-3, LDH-4 and LDH-5. Km values of isozymes at 30 degrees C were higher in the order of LDH-1, LDH-3 and LDH-5. 4. There was no change in the enzyme activity during thermal acclimation.  相似文献

Changes in the activity of serum lactate dehydrogenase (LDH) and the serum LDH isozyme pattern in healthy young men given a high carbohydrate diet (480-636 g/day, 80% of the total energy) for 21 days were examined. Serum total LDH activity showed no significant change in four healthy young volunteers who received high carbohydrate diet for 21 days. However, the percentage of LDH-4 increased significantly (P less than 0.01) from 8.5 +/- 2.4 to 10.9 +/- 1.9% after 21 days, as did also the percentage of LDH-5 (P less than 0.01) from 5.1 +/- 1.9 to 10.7 +/- 2.9%. The percentage of M-type LDH activity increased 30% during the experiment (P less than 0.05). It is concluded from the results that the high carbohydrate intake affects the percentage of LDH-4 and LDH-5, but not the total serum LDH activity.  相似文献

The spectrum of LDH isozymes was studied using different concentration of X-100 triton for enzyme extraction from the eggs and embryos, different systems of electrophoresis and different methods of staining electrophoregrams. Similar results were obtained in all cases. The mouse eggs and embryos were shown to contain only LDH-1; the treatment of oviducal fluid with X-100 triton strengthened the activity of LDH-4 and LDH-5. The detection of the activity of LDH-5 and hybrid isozymes appears to be related to the presence of oviducal fluid in the sample under study.  相似文献

Lengthened circadian period of locomotor activity is a characteristic of a congenic strain of mice carrying a nonsense mutation in exon 5 of the carbonic anhydrase II gene, car2. The null mutation in car2 is located on a DBA/2J inbred strain insert on proximal chromosome 3, on an otherwise C57BL/6J genomic background. Since reducing the size of the congenic region would narrow the possible candidate genes for period, two recombinant congenic strains (R1 and R2) were developed from the original congenic strain. These new congenic strains were assessed for period, genetic composition, and the presence of immunoreactive carbonic anhydrase II. R1 mice were homozygous DBA/2J for the distal portion of the original DBA/2J insert, while R2 mice were homozygous DBA/2J for the proximal portion. R1 mice had a significantly lengthened period compared to R2 mice and wild-type C57BL/6J mice, indicating that the gene(s) affecting period is likely found within the reduced DBA/2J insert (~1 cM) in the R1 mice. The R1 mice also possessed the null mutation in car2. This study confirmed the presence of a gene(s) affecting period on proximal chromosome 3 and significantly reduced the size of the congenic region and the number of candidate genes. Future studies will focus on identifying the gene influencing period.  相似文献

Summary Lactate dehydrogenase (LDH) isozyme patterns were analysed by polyacrylamide (PAA) slab gel electrophoresis in extracts prepared from various rabbit skeletal muscles of defined fibre composition and by PAA microelectrophoresis of microdissected, histochemically typed single muscle fibres. The results obtained by electrophoresis of whole muscle extracts generally agreed with the data obtained from single fibre electrophoresis, i.e. the LDH isozyme pattern corresponded to that of the predominant fibre type. Type I Fibres from soleus and semitendinosus muscles were characterized by a unique pattern of all 5 LDH isozymes with a predominance of LDH-1, 2 and 3. The major fraction (80%) of the type II fibres from extensor digitorum longus and tibialis anterior muscles contained only LDH-5 (M4). About 20% of the type II fibres contained in addition to LDH-5 small amounts of LDH-4 and LDH-3. The fraction of fibres containing LDH-5, LDH-4, and LDH-3 was similar (ca. 20%) in the histochemically defined IIA and IIB subpopulations In view of the fact that the major fractions of rabbit IIB fibres display low and of IIA fibres high aerobic oxidative capacities (Reichmann and Pette 1982), these data indicate that the expression of the H-subunit of LDH is not correlated with the aerobic-oxidative capacity of the fibre. It also appears not to be correlated with the presence of different myosin isoforms in IIA and IIB fibres.  相似文献

利用SOD和LDH同工酶电泳分析,研究大豆黄酮对衰老小鼠的抗氧化作用。结果显示大豆黄酮没有改变SOD和LDH同工酶谱的特征,但对因衰老引起的小鼠脑组织LDH和SOD同工酶活性、各组分的相对活性和比活力的变化有不同程度的改善作用,即LDH同工酶中LDH-2、LDH-3的活性明显下降,LDH-1的活性下降最为明显,而LDH-4的活性有所下降,但不显著,LDH-5的活性几乎没有变化,SOD同工酶的SOD-1和SOD-2的活性有不同程度的升高。这表明大豆黄酮是通过抑制LDH同工酶H亚基的合成来降低LDH的活性,而对M亚基的合成没有影响,并且能够促进SOD同工酶SOD-1和SOD-2的合成,不影响其遗传稳定性。  相似文献

An improved method is described for the isolation of isozymes 1 and 5 of lactate dehydrogenase (LDH) from heart and skeletal muscles of foxes. The method includes salt fractionation with ammonium sulphate, chromatography on DEAE- and CM-celluloses and affinity chromatography on AMP-Sepharose. The preparations of LDH isozymes 1 and 5 turned out to be homogeneous both in 7,5% polyacrylamide gel electrophoresis and under immunodiffusion analysis. It is shown that the pH optimum for LDH-1 is 10.2-10.4 for LDH-5 it is 9.5-9.6 in the case of the direct reaction, and the pH optimum is 7.6-7.8 for LDH-1 and 7.3-7.4 for LDH-5 in the case of reverse reaction. The values of Mikhaelis constants were determined for substrates and coenzymes in direct and reverse reactions. It is found that the excess of lactate and pyruvate causes substrate inhibition of both LDH-1 and LDH-5. The activities of LDH-1 and LDH-5 showed an unexpected similar sensitivity to the inhibitory effect of high pyruvate concentrations.  相似文献

This study finds lengthened circadian period in a congenic strain of mice homozygous for a null mutation in carbonic anhydrase isoenzyme-II gene on proximal Chromosome 3. Carbonic anhydrase II has the highest turnover rate of any constitutive enzyme. It catalyzes the reversible hydration of carbon dioxide to control intercellular acid/base balance. A strain of congenic mice has a carbonic anhydrase II null mutation within a DBA/2J inbred strain insert on a C57BL/6J inbred strain background. The locomotor activity levels and period of circadian rhythms were examined in the homozygous null mutants and their progenitors, mice heterozygous for the region around the carbonic anhydrase gene. The heterozygous mice siblings and the wild-type siblings served as the controls. During behavioral studies, male and female offspring and parents were housed singly in constant darkness. Locomotor activity was monitored using an infrared photobeam array. Mice homozygous for the carbonic anhydrase null mutation had a longer circadian period than either heterozygote or wild type littermates. Carbonic anhydrase null mutants also had low locomotor activity compared to either heterozygous or wild-type litter mates. This implies that either the physiological changes resulting from absence of carbonic anhydrase II isozyme or the presence of DBA/2J alleles around the carbonic anhydrase locus influence the circadian period and level of locomotor activity in laboratory mice.  相似文献

The lactate dehydrogenase isoenzyme pattern has been determined in different murine lymphocytic cell populations. In each cell population, the LDH activity was predominantly found in the LDH-4 and LDH-5 fractions. The percentage LDH-5 activity was significantly higher in B cells than in T cells. The same is true for lymphocytes from the spleen versus lymph node lymphocytes. The percentage LDH-5 activity is significantly higher in peripheral T lymphocytes than in thymocytes. Enrichment of the more mature thymocytes of the thymocyte cell pool by either cortisone treatment in vivo or gradient centrifugation on bovine serum albumin (BSA) results in a decrease of LDH-1 and LDH-2 fractions. In the cortisone-treated group, the shift in the LDH pattern is accompanied by a significant increase of LDH-5 and LDH-4 fractions, whereas in the BSA group only the LDH-4 fraction increases.  相似文献

Glaucoma is a leading cause of blindness, ultimatively resulting in the apoptotic death of retinal ganglion cells. However, molecular mechanisms involved in ganglion cell death are poorly understood. While the involvement of ionotropic glutamate receptors has been extensively studied, virtually nothing is known about its metabotropic counterparts. Here, we compared the retinal gene expression of metabotropic glutamate receptors (mGluR) in eyes with normal and elevated intraocular pressure (IOP) of DBA/2J mice, a model for secondary angle-closure glaucoma using RT-PCR and immunohistochemistry. Elevated IOP in DBA/2J mice significantly increased retinal gene expression of mGluR1a, mGluR2, mGluR4a, mGluR4b, mGluR6 and mGluR7a when compared to C57BL/6 control animals, while mGluR5a/b and mGluR8a were decreased and no difference was observed for mGluR3 and mGluR8b. Specific antibodies detected an increase of mGluR1a and mGluR5a/b in both synaptic layers and in the ganglion cell layer of the retina under elevated IOP. Because ganglion cell death in DBA/2J mice occurs most likely by apoptotic mechanisms, we demonstrated up-regulation of mGluRs in neurons undergoing apoptosis. In summary, we support the idea that the specific gene regulation of mGluRs is a part of the glaucoma-like pathological process that develops in the eyes of DBA/2J mice.  相似文献

We have previously shown that the adult DBA/2J and A/J strains of mice differ in carotid body volume and morphology. The question has arisen whether these differences develop during the prenatal or postnatal period. Investigating morphological development of the carotid body and contributing genes in these mice can provide further understanding of the appropriate formation of the carotid body. We examined the carotid body of these mice from 1 day to 4 wk old for differences in volume, morphology, and gene expression of Gdnf family, Dlx2, Msx2, and Phox2b. The two strains showed divergent morphology starting at 1 wk old. The volume of the carotid body increased from 1 wk up to 2 wk old to the level of 4 wk old in the DBA/2J mice but not in the A/J mice. This corresponds with immunoreactivity of LC3, an autophagy marker, in A/J tissues at 10 days and 2 wk. The differences in gene expression were examined at 1 wk, 10 days, and 2 wk old, because divergent growth occurred during this period. The DBA/2J's carotid body at 1 wk old showed a greater expression of Msx2 than the A/J's carotid body. No other candidate genes showed consistent differences between the ages and strains. The difference was not seen in sympathetic cervical ganglia of 1 wk old, suggesting that the difference is carotid body specific. The current study indicates the critical postnatal period for developing distinctive morphology of the carotid body in these mice. Further studies are required to further elucidate a role of Msx2 and other uninvestigated genes.  相似文献
