Pineal glands secrets melatonin and various proteins and peptides which has many physiological functions. In keeping with this view, present experiment was conducted to know the effect of buffalo (Bubalus bubalis) pineal proteins (PP) at different dose level on fluoride-induced changes in plasma biochemicals and blood antioxidants enzymes in female rats. For this, we took 30 adult female Wistar rats (133–145 g body weights, BW) and divided into five groups (control, group I; 150 ppm fluoride (F), group II; F+ 50 µg pineal proteins, group III; F+ 100 µg PP, group IV; F+ 200 µg PP, group V). We administered fluoride (150 ppm, drinking water) and F+ pineal proteins at 50, 100, and 200 µg/kg BW, i.p. daily for 21 days. Blood samples were collected at the end of the experiments to estimate plasma glucose, proteins, F, lipid peroxidation (LPO), alkaline phosphatase (ALP), and acetyl cholinesterase (AChE) activity. Red blood cells (RBCs) were separated for analysis of LPO, AChE, catalase (CAT), superoxide dismutase (SOD), reduced glutathione (GSH), glutathione peroxidase (GPx), and glutathione reductase (GR) in different groups of animals. Total plasma glucose and protein level did not significantly change in F-treated rats. Plasma ALP and F level were significantly (p?<?0.05) high in group II as compared with control and groups III, IV, and V. Administration of PP at different dose level significantly (p?<?0.05) reduced the F concentration and ALP activity. Plasma and RBCs AChE activity was significantly (p?<?0.05) reduced in F-treated animals as compared with control rats and significantly (p?<?0.05) elevated on exogenous administration of PP (groups III and IV). Plasma and RBCs LPO level was significantly (p?<?0.05) high in F-alone-treated rats, and PP caused significant (p?<?0.05) reduction of LPO in groups IV and V. However, PP treatment in group IV brought better amelioration of F-induced high LPO than in groups III and V. At no dose level, PP-ameliorated F-induced depression of RBCs GSH, CAT, GR, and GPx level. Interestingly, SOD activity was elevated in dose-dependent manner at different dose level of PP in groups III, IV, and V than control and F-administered rats. These findings clearly indicate the beneficial effects of buffalo pineal proteins on fluoride-induced adverse changes in certain plasma biochemical and blood antioxidant systems of rats. It further indicates that PP has dose-dependent ameliorative function against F-induced adverse effects in plasma and blood.  相似文献

Brook trout (Salvelinus fontinalis) raised from eggs under two photoperiod and two feeding regimes were tested for physiological changes preparatory for transition from freshwater to seawater. Size, age, growth rate, photoperiod, and diel rhythms were examined for possible influences on plasma osmolarity, [Na+], [Cl], [K+], [Mg2+], thyroxine concentration, hematocrit, and gill Na+, K+-ATPase activity of brook trout in freshwater.
  • 2.2. Significant diel cycles were found in plasma osmolarity, [Na+] and thyroxine concentration.
  • 3.3. Significant size and/or age related changes occurred for plasma osmolarity, Na+], [K+] and hematocrit, but could explain little of their total variation (0.02 < r2 < 0.18).
  • 4.4. A sexually dimorphic response to photoperiod was observed in hematocrit for both mature and immature fish, with hematocrit of mature females declining in autumn and hematocrit of immature males increasing in autumn.
  • 5.5. Gill Na+, K+-ATPase activity did not respond to photoperiod or feeding treatment and showed no change with size or age.
  • 6.6. Plasma thyroxine levels responded to feeding and photoperiod treatment. There was a significant correlation between the percent mean difference in plasma thyroxine and the mean difference in growth rate between high and low feed fish (r2 =0.51), suggesting a relationship between thyroxine and growth.

A humoral ouabain-like plasma factor has been observed in patients with essential hypertension (EHT). In the present study, we hypothesized that this humoral factor might be responsible for the elevated cytosolic free calcium concentrations [Ca2+]i seen in these patients. Patients with mild to moderate EHT and their normotensive first degree blood relatives (NTBR) participated in the study. Platelet Na+, K+-ATPase activity was assayed in EHT patients and their NT first-degree relatives. To confirm the ouabain-like activity in plasma from EHT patients, control platelets were incubated with EHT and NTBR plasma and their Na+, K+-ATPase activity was measured. In addition, the effect of EHT plasma on platelet45Ca-uptake was studied. Thein vitro effects of ouabain (10 ΜM) on (i)45Ca-uptake and (ii) [Ca2+]i response in control platelets were also observed. A decreased Na+K+-ATPase activity (P< 0.05) was observed in platelet membranes from EHT patients. Incubation of control platelets with EHT plasma decreased their Na+, K+-ATPase activity (P< 0.01) and increased their45Ca-uptake (P< 0.05). C-18 Sep-Pak filtered hypertensive plasma extracts (containing the ouabain-like fraction) also decreased Na+, K+-ATPase activity (P< 001) in control platelet membranes.In vitro incubation of control platelets with ouabain increased45Ca-uptake (P< 005) and [Ca2+]i response (P< 0.05) in these platelets. Thus it appears that an ouabain-like factor in the EHT plasma may contribute to the elevated platelet [Ca2+]i observed in EHT patients.  相似文献

The aim of the present study is to evaluate the oxidative damage in rats of different ages. Weaned rats of 25 g and adults of 300 g were used in groups of 6, a single i.p. dose of morphine sulfate of 3, 6 or 12 mg/kg was administered. All animals were sacrificed to measure GSH and 5-HT levels in brain by liquid chromatography, as well as Na+, K+-ATPase and total ATPase enzymatic activity. 5-HT levels decreased significantly (p<0.05) in adult animals that received 3 and 6 mg morphine. Na+, K+-ATPase activity increased significantly (p<0.05) in all groups of weaned animals. In adult animals, Na+, K+-ATPase and total ATPase partially diminished. GSH levels diminished significantly (p<0.05) both in weaned and in adult groups. The results indicate age-induced changes in cellular regulation and biochemical responses to oxidative stress induced by morphine.  相似文献

We examined the metabolic and ionoregulatory responses of the Amazonian cichlid, Astronotus ocellatus, to 20 h exposure to severe hypoxia (0.37 ± 0.19 mg O2/l; 4.6% air saturation) or 8 h severe hypoxia followed by 12 h recovery in normoxic water. During 20 h exposure to hypoxia, white muscle [ATP] was maintained at normoxic levels primarily through a 20% decrease in [creatine phosphate] (CrP) and an activation of glycolysis yielding lactate accumulation. Muscle lactate accumulation maintained cytoplasmic redox state ([NAD+]/[NADH]) and was associated with an inactivation of the mitochondrial enzyme pyruvate dehydrogenase (PDH). The inactivation of PDH was not associated with significant changes in cytoplasmic allosteric modulators ([ADPfree], redox state, or [pyruvate]). Hypoxia exposure caused a ∼65% decrease in gill Na+/K+ ATPase activity, which was not matched by changes in Na+/K+ ATPase α-subunit protein abundance indicating post-translational modification of Na+/K+ ATPase was responsible for the decrease in activity. Despite decreases in gill Na+/K+ ATPase activity, plasma [Na+] increased, but this increase was possibly due to a significant hemoconcentration and fluid shift out of the extracellular space. Hypoxia caused an increase in Na+/K+ ATPase α-subunit mRNA abundance pointing to either reduced mRNA degradation during exposure to hypoxia or enhanced expression of Na+/K+ ATPase α-subunit relative to other genes.  相似文献

The aim of the study was to verify the hypothesis if copper could influence the activity of sodium-transporting systems in erythrocyte membrane that could be related to essential hypertension. The examined group of patients consisted of 15 men with hypertension. The control group was 11 healthy male volunteers. The Na+/H+ exchanger (NHE) activity in erythrocytes was determined according to Orlov et al. The activity of transporting systems (ATP-Na+/K+; co-Na+/K+/Cl; ex-Na+/Li+; free Na+ and K+ outflow [Na+, K+-outflow]) was determined according to Garay's method. The concentration of copper in plasma was assessed using atomic absorption spectrometry. The activity of ATP-Na+/K+ (μmol/L red blood cells [RBCs]/h) in hypertensive patients was 2231.5±657.6 vs 1750.5±291 in the control (p<0.05), the activity of co-Na+/K+/Cl (μmol/L RBCs/h) in hypertensives was 171.3±77.9 vs 150.7±53.9 in the control (NS). Na+-outflow (μmol/L RBCs/h) in hypertensives was 118.3±51.6 vs 113.3±24.4 in the control (NS). The K+-outflow (μmol/L RBCs/h) in hypertensives was 1361.7±545.4 vs 1035.6±188.3 in the control (NS). The activity of ex-Na+/Li+ (μmol/L RBCs/h) in hypertensive patients was 266.1±76.1 vs 204.1±71.6 in the control (p<0.05). NHE activity (mmol/L RBCs/h) in hypertensives was 9.7±2.96 vs 7.7±1.33 in the control (p<0.05). In hypertensive patients, negative correlation was found between the activity of Na+/K+/Cl co-transport and plasma copper concentration (R s=−0.579, p <0.05) and between the activity of ex-Na+/Li+ and plasma copper concentration (R s=−0.508, p<0.05). Plasma copper concentration significantly influences the activity of sodium transporting systems in erythrocyte membrane. Copper supplementation could be expected to provide therapeutic benefits for hypertensive patients.  相似文献

Summary The intestinal caeca reabsorb urinary sodium chloride (NaCl) and water (Rice and Skadhauge 1982). Free water may be generated if the reabsorbed NaCl is secreted via salt gland secretion (Schmidt-Nielsen et al. 1958). Therefore ceacal ligation should (a) reduce hingut NaCl and water reabsorption, (b) enhance the increase in plasma osmolality during saline acclimation, and (c) affect drakes more than ducks. Twelve Pekin drakes and 13 Pekin ducks, Anas platyrhynchos, were caecally ligated or sham operated before acclimation to 450 mmol · 1 NaCl. Body mass, hematocrit, plasma osmolality, and inonic concentrations of plasma, cloacal fluid, and salt gland secretion were measured after each increase in drinking water salinity. Osmoregulatory organ masses were determined. Caecal ligation did not effect plasma osmolality or ion concentrations of plasma, cloacal fluid, or salt gland secretion, but reduced salt gland size in ducks. Drakes and ducks drinking fresh water had the same hematocrit, plasma osmolality, and plasma concentrations of Na+ and Cl. In both sexes exposure to 75 mmol · 1-1 NaCl significantly decreased plasma [Na+] and doubled cloacal fluid [Na+]. Exposure to 450 mmol · 1-1 NaCl decreased body mass and increased hematocrit, plasma [Na+], [Cl], and plasma osmolality (more in drakes than in ducks); cloacal fluid osmolality nearly doubled compared to freshwater-adapted ducks, due mainly to osmolytes other than Na+ and Cl. Biochemical effects of Citrullus colocynthis in normal and diabetic rats   总被引:2,自引:0,他引:2  
Diabetes mellitus is one of the most common endocrine diseases. In UAE many traditional plants such as the Citrullus colocynthis (Handal) are used as antidiabetic remedies. The aim of this study was to examine the effect of the aqueous extract of the seed of C. colocynthis on the biochemical parameters of normal and streptozotocin (STZ)-induced diabetic rats. Diabetes mellitus was induced by a single intraperitoneal (60 mg/kg body wt1) injection of STZ. Normal and diabetic rats were fed with the plant extract daily by oral intubation for 2 weeks. Blood sample were collected at the beginning and end of the experiment for the measurement of biochemical parameters. The plasma level of alanine aminotranferase (ALT), alkaline phosphatase (ALP), aspartate aminotransferase (AST), gamma-glutamyl transferase (GGT), lactic dehydrogenase (LDH) increased significantly after the onset of diabetes. Oral administration of the plant extract reduced the plasma level of AST and LDH significantly. However, the plant extract failed to reduce the increased blood level of GGT and ALP in diabetic rats. Blood urea nitrogen (BUN) increased significantly after the onset of diabetes. No significant difference was observed in the blood creatinine, K+, Na+, Ca2+ and P levels of normal and diabetic rats. The plant extract did not have any effect on BUN level, however, it caused an increase in the level of K+, Na+ in diabetic rats. In conclusion, oral administration of the aqueous extract of the C. colocynthis can ameliorate some of the toxic effects of streptozotocin. (Mol Cell Biochem 261: 143–149, 2004)  相似文献

The process of regenerating liver is the result of a balance between stimulating factors and inhibitors of hepatocyte proliferation. Melatonin and its metabolites have been found to protect tissues against oxidative damage generated by a variety of toxic agents and metabolic processes. Furthermore, studies in liver of rats showed a decrease in the liver mitochondrial hydroxylation of drugs returning to the normal state after the administration of antioxidants. This study was designed to determine, in experimental animals, whether the administration of an antioxidant agent such as melatonin could prevent cells events leading to tissue injury and hepatic dysfunction after partial hepatectomy (PH). Biliary flow (BF), oxidative stress in hepatic tissue and Na+/K+ATPase activities in whole plasma membrane were determined. PH decreased the Na+/K+ATPase activity. PH significantly reduced the BF (36%) and promoted oxidative stress with an increase of lipoperoxidation and decrease of glutathione peroxidase and catalase activities. Treatment with melatonin prevented the decrease of BF in rats with hepatectomy and normalized the Na+/K+ATPase activity. Moreover, melatonin markedly attenuated oxidative stress produced by PH. This may be the results of the higher efficacy of melatonin in scavenging various free radicals and also because of its ability in stimulating the antioxidant enzymes. We suggest that oxidative stress before and during liver regeneration has a crucial role in cholestasis, apoptotic/necrotic hepatocellular damage and the impairment in liver transport function induced by PH and that melatonin could modulate the degree of oxidative stress and through it prevent the alterations in liver function carrier. In frog red blood cells, K-Cl cotransport (i.e., the difference between ouabain-resistant K fluxes in Cl and NO3) has been shown to mediate a large fraction of the total K+ transport. In the present study, Cl-dependent and Cl-independent K+ fluxes via frog erythrocyte membranes were investigated as a function of external and internal K+ ([K+] e and [K+] i ) concentration. The dependence of ouabain-resistant Cl-dependent K+ (86Rb) influx on [K+] e over the range 0–20 mm fitted the Michaelis-Menten equation, with an apparent affinity (K m ) of 8.2 ± 1.3 mm and maximal velocity (V max ) of 10.4 ± 1.6 mmol/l cells/hr under isotonic conditions. Hypotonic stimulation of the Cl-dependent K+ influx increased both K m (12.8 ± 1.7 mm, P < 0.05) and V max (20.2 ± 2.9 mmol/l/hr, P < 0.001). Raising [K+] e above 20 mm in isotonic media significantly reduced the Cl-dependent K+ influx due to a reciprocal decrease of the external Na+ ([Na+] e ) concentration below 50 mm. Replacing [Na+] e by NMDG+ markedly decreased V max (3.2 ± 0.7 mmol/l/hr, P < 0.001) and increased K m (15.7 ± 2.1 mm, P < 0.03) of Cl-dependent K+ influx. Moreover, NMDG+ Cl substitution for NaCl in isotonic and hypotonic media containing 10 mm RbCl significantly reduced both Rb+ uptake and K+ loss from red cells. Cell swelling did not affect the Na+-dependent changes in Rb+ uptake and K+ loss. In a nominally K+(Rb+)-free medium, net K+ loss was reduced after lowering [Na+] e below 50 mm. These results indicate that over 50 mm [Na+] e is required for complete activation of the K-Cl cotransporter. In nystatin-pretreated cells with various intracellular K+, Cl-dependent K+ loss in K+-free media was a linear function of [K+] i , with a rate constant of 0.11 ± 0.01 and 0.18 ± 0.008 hr−1 (P < 0.001) in isotonic and hypotonic media, respectively. Thus K-Cl cotransport in frog erythrocytes exhibits a strong asymmetry with respect to transported K+ ions. The residual, ouabain-resistant K+ fluxes in NO3 were only 5–10% of the total and were well fitted to linear regressions. The rate constants for the residual influxes were not different from those for K+ effluxes in isotonic (∼0.014 hr−1) and hypotonic (∼0.022 hr−1) media, but cell swelling resulted in a significant increase in the rate constants. Brook trout (Salvelinus fontinalis) of a single genetic stock, and hatched at the same time, were raised under two photoperiod and two feeding regimes to obtain fish of the same age but with different sizes and photoperiod experiences. In 11 experiments over 1.5 firs, fish were gradually exposed to 32 ppt seawater for 20 days to investigate the ontogeny of salinity tolerance.
  • 2.2. Daily changes in plasma osmolarity, [Na+], [Cl], [K+], [Mg2+], thyroxine, hematocrit and gill Na+,K+-ATPase during adaptation to 10, 20 and 32 ppt were examined in one experiment.
  • 3.3. Size was the primary determinant of seawater survival (r2 = 0.77) the effect of size on seawater survival slowed after fish reached a fork length of 14 cm. The effect of age on seawater survival (r2 = 0.65) was through its covariance with size.
  • 4.4. Photoperiod affected seawater survival only through its influence on the timing of male maturation, which decreased salinity tolerance.
  • 5.5. Regulation of plasma osmolarity, [Na+], [Cl], [K2+], [Mg2+] and hematocrit in sea water increased linearly with size over the entire range of sizes (6–32 em).

This study tested the hypothesis that melatonin (Mel) therapy preserved the brain architectural and functional integrity against ischaemic stroke (IS) dependently through suppressing the inflammatory/oxidative stress downstream signalling pathways. Adult male B6 (n = 6 per each B6 group) and TLR4 knockout (ie TLR4?/?) (n = 6 per each TLR4?/? group) mice were categorized into sham control (SCB6), SCTLR4?/?, ISB6, ISTLR4?/?, ISB6 + Mel (i.p. daily administration) and ISTLR4?/? + Mel (i.p. daily administration). By day 28 after IS, the protein expressions of inflammatory (HMBG1/TLR2/TLR4/MAL/MyD88/RAM TRIF/TRAF6/IKK‐α/p‐NF‐κB/nuclear‐NF‐κB/nuclear‐IRF‐3&7/IL‐1β/IL‐6/TNF‐α/IFN‐γ) and oxidative stress (NOX‐1/NOX‐2/ASK1/p‐MKK4&7/p‐JNK/p‐c‐JUN) downstream pathways as well as mitochondrial‐damaged markers (cytosolic cytochrome C/cyclophilin D/SRP1/autophagy) were highest in group ISB6, lowest in groups SCB6 and SCTLR4?/?, lower in group ISTLR4?/? + Mel than in groups ISTLR4?/? and ISB6 + Mel and lower in group ISB6 + Mel than in group ISTLR4?/? (all P < .0001). The brain infarct volume, brain infarct area and the number of inflammatory cells in brain (CD14/F4‐88) and in circulation (MPO+//Ly6C+/CD11b+//Ly6G+/CD11b+) exhibited an identical pattern, whereas the neurological function displayed an opposite pattern of inflammatory protein expression among the six groups (all P < .0001). In conclusion, TLR inflammatory and oxidative stress signallings played crucial roles for brain damage and impaired neurological function after IS that were significantly reversed by Mel therapy.  相似文献

Summary A membrane preparation enriched in the basolateral segment of the plasma membrane was isolated from the rat renal cortex by a procedure that included separation of particulates on a self-generating Percoll gradient. The uptake ofl-glutamate by the basolateral membrane vesicles was studied. A Na+ gradient ([Na+] o >[Na+] i ) stimulated the uptake ofl-glutamate and provided the driving force for the uphill transport of the acidic amino acid, suggesting a Na+-l-glutamate cotransport system in the basolateral membrane. A K+ gradient ([K+] i >[K+] o ) increased the uptake additionally. This effect was specific for K+ (Rb+). The action of the K+ gradient in enhancing the uptake ofl-glutamate had an absolute requirement for Na+. In the presence of Na+, but in the absence of a Na+ gradient. i.e., [Na+] o =[Na+] i , the K+ gradient also energized the concentrative uptake ofl-glutamate. This effect of the K+ gradient was not attributable to an alteration in membrane potential. The finding of a concentrative uptake system forl-glutamate energized by both Na+ ([Na+] o >[Na+] i and K+ ([K+] i >[K+] o ) gradients in the basolateral membrane, combined with previous reports of an ion gradient-dependent uphill transport system for this amino acid in the brush border membrane, suggests a mechanism by whichl-glutamate is accumulated intracellularly in the renal proximal tubule to extraordinarily high concentrations.  相似文献

Summary The intracellular distribution of Na+, K+, Cl and water has been studied in the Ehrlich ascites tumor cell. Comparison of the ion and water contents of whole cells with those of cells exposed to La3+ and mechanical stress indicated that La3+ treatment results in selective damage to the cell membrane and permits evaluation of cytoplasmic and nuclear ion concentrations. The results show that Na+ is sequestered within the nucleus, while K+ and Cl are more highly concentrated in the cell cytoplasm. Reduction of the [Na+] of the incubation medium by replacement with K+ results in reduced cytoplasmic [Na+], increased [Cl] and no change in [K+]. Nuclear concentrations of these ions are virtually insensitive to the cation composition of the medium. Concomitant measurements of the membrane potential were made. The potential in control cells was –13.7 mV. Reduction of [Na+] in the medium caused significant depolarization. The measured potential is describable by the Cl equilibrium potential and can be accounted for in terms of cation distributions and permeabilities. The energetic implications of the intracellular compartmentation of ions are discussed.  相似文献

Summary Ehrlich cells shrink when the osmolality of the suspending medium is increased and behave, at least initially, as osmometers. Subsequent behavior depends on the nature of the hyperosmotic solute but in no case did the cells exhibit regulatory volume increase. With hyperosmotic NaCl an osmometric response was found and the resultant volume maintained relatively constant. Continuous shrinkage was observed, however, with sucrose-induced hyperosmolality. In both cases increasing osmolality from 300 to 500 mOsm initiated significant changes in cellular electrolyte content, as well as intracellular pH. This was brought about by activation of the Na+/H+ exchanger, the Na/K pump, the Na++K++2Cl cotransporter and by loss of K+ via a Ba-sensitive pathway. The cotransporter in response to elevated [Cl] i (100mm) and/or the increase in the outwardly directed gradient of chemical potential for Na+, K+ and Cl, mediated net loss of ions which accounted for cell shrinkage in the sucrose-containing medium. In hyperosmotic NaCl, however, the net Cl flux was almost zero suggesting minimal net cotransport activity.We conclude that volume stability following cell shrinkage depends on the transmembrane gradient of chemical potential for [Na++K++Cl], as well as the ratio of intra- to extracellular [Cl]. Both factors appear to influence the activity of the cotransport pathway.  相似文献

Summary Na+–H+ exchange activity in renal brush border membrane vesicles isolated from hyperthyroid rats was increased. When examined as a function of [Na+], treatment altered the initial rate of Na+ uptake by increasingV m (hyperthyroid, 18.9±1.1 nmol Na+ · mg–1 · 2 sec–1; normal, 8.9±0.3 nmol Na+ · mg–1 · 2 sec–1), and not the apparent affinityK Na + (hyperthyroid, 7.3±1.7mm; normal, 6.5±0.9mm). When examined as a function of [H+] and at a subsaturating [Na+] (1mm), hyperthyroidism resulted in the proportional increase in Na+ uptake at every intravesicular pH measured. A positive cooperative effect on Na+ uptake was found with increased intravesicular acidity in vesicles from both normal and hyperthyroid rats. When the data were analyzed by the Hill equation, it was found that hyperthyroidism did not change then (hyperthyroid, 1.2±0.06; normal, 1.2±0.07) or the [H+]0.5 (hyperthyroid, 0.39±0.08 m; normal, 0.44±0.07 m) but increased the apparentV m (hyperthyroid, 1.68±0.14 nmol Na+ · mg–1 · 2 sec–1; normal 0.96±0.10 nmol Na+ · mg–1 · 2 sec–1). The uptake of Na+ in exchange for H+ in membrane vesicles from normal and hyperthyroid animals was not influenced by membrane potential. H+ translocation or debinding was rate limiting for Na+–H+ exchange since Na+–Na+ exchange activity was greater than Na+–H+ exchange activity. Hyperthyroidism caused a proportional increase and hypothyroidism caused a proportional decrease in Na+–Na+ and Na+–H+ exchange. We conclude that hyperthyroidism leads to either an increase in the number of functional exchangers in the membrane or exactly proportional increases in the rate-limiting steps for Na+–Na+ and Na+–H+ exchange activity.  相似文献

We used a central composite rotatable experimental design and response surface methodology to evaluate the effects of temperature (18–37 °C), salinity (0–20‰), and their interaction on specific growth rate (SGR), feed efficiency (FE), plasma osmolality, and gill Na+, K+-ATPase activity in GIFT tilapia juveniles. The linear and quadratic effects of temperature and salinity on SGR, plasma osmolality, and gill Na+, K+-ATPase activity were statistically significant (P<0.05). The interactive effects of temperature and salinity on plasma osmolality were significant (P<0.05). In contrast, the interaction term was not significant for SGR, FE, and gill Na+, K+-ATPase activity (P>0.05). The regression equations for SGR, FE, plasma osmolality, and gill Na+, K+-ATPase activity against the two factors of interest had coefficients of determination of 0.944, 0.984, 0.966, and 0.960, respectively (P<0.01). The optimal temperature/salinity combination was 28.9 °C/7.8‰ at which SGR (2.26% d1) and FE (0.82) were highest. These values correspond to the optimal temperature/salinity combination (29.1 °C/7.5‰) and the lowest plasma osmolality (348.38 mOsmol kg−1) and gill Na+, K+-ATPase activity (1.31 µmol Pi. h−1 g−1 protein), and resulted in an energy-saving effect on osmoregulation, which promoted growth.  相似文献

Glutamine, the preferred precursor for neurotransmitter glutamate, is likely to be the principal substrate for the neuronal System A transporter SAT1 in vivo. By measuring currents associated with SAT1 expression in Xenopus oocytes, we found that SAT1 mediates transport of small, neutral, aliphatic amino acids including glutamine, alanine and the System A‐specific analogue 2‐(methylamino) isobutyrate, each with K0.5 of 0.3–0.5 mm . Amino acid transport is driven by the Na+ electrochemical gradient. Kinetic data indicates that Na+/cotransport comprises the ordered binding first of Na+ (a voltage‐dependent step), then alanine, then simultaneous translocation. Li+ (but not H+) can substitute for Na+ but results in reduced Vmax. In the absence of amino acid, SAT1 mediates a cation leak with selectivity Na+, Li+, H+, K+. The temperature‐dependence of the leak current (Ea = 17 ± 3 kcal/mol) is consistent with carrier‐mediated Na+ uniport activity (cf 13 ± 2 kcal/mol for Na+/alanine cotransport) but the leak does not saturate at physiological [Na+], suggesting channel activity. Despite a Na+ Hill coefficient of 1, we obtained Na+/amino acid coupling coefficients greater than 1 from simultaneous measurement of charge and [3H]alanine or [3H]glutamine uptake. Interpretation of these data is model‐dependent and consistent with either (1) an all‐carrier model in which Na+/amino acid cotransport is thermodynamically coupled 2 : 1, cotransport is preferred over Na+ uniport, and in which there is little cooperativity between Na+ binding events, or (2) 1 : 1 coupling in parallel with an always‐on Na+ channel activity. In either scenario, the presence of SAT1 at the plasma membrane and resultant Na+ fluxes will place a significant energy burden on the cell.  相似文献

Abstract: The present study examines the interaction of Na+ and K+ with the binding of the cocaine analogue 3β-(4-[125I]iodophenyl)tropane-2β-carboxylic acid isopropyl ester to dopamine transporters (DATs) in rat striatal synaptosomal membranes at 37°C. The binding increases with [Na+] from 10 to 100 mM and decreases with higher [Na+]. The presence of K+ reduces the maximal stimulatory effect of Na+ and causes a nonlinear EC50 shift for Na+. K+ strongly inhibits the binding at low [Na+]. Increasing [Na+] produces a linear IC50 shift for K+. Saturation analysis indicates a single binding site changing its affinity for the radioligand depending on [K+]/[Na+] ratio in the assay buffer. A reduced Bmax was observed in the presence of 10 mM Na+ and 30 mM K+. Both high [Na+] and high [K+] accelerate the dissociation of the binding, and K+-induced acceleration was abolished by increasing [Na+]. Least squares model fitting of equilibrium data and kinetic analysis of dissociation rates reveal competitive interactions between Na+ and K+ at two sites allosterically linked on the DAT: One site mediates the stimulatory effect of Na+, and the other site involves the radioligand binding and the inhibitory effect of cations on the binding. Various uptake blockers and substrates, dopamine in particular, display reduced potency in inhibiting the binding at a higher [K+]/[Na+] ratio.  相似文献

Chalcalburnus tarichi is an anadromous cyprinid fish that has adapted to extreme conditions (salinity 22 ‰, pH 9.8 and alkalinity 153 mEq × l?1) in Lake Van in eastern Turkey. Changes in immunoreactivity of Na+/K+-ATPase in gill tissue and osmolarity and ion levels in plasma were investigated in C. tarichi during reproductive migration. Physicochemical characteristics and ion levels in Lake Van were high compared freshwater. Plasma osmolality and plasma ion concentrations ([Na+], [K+] and [Cl?]) increased after transfer from freshwater to Lake Van. The mitochondria-rich (MR) cells of the gill were stained in both filament and lamellar epithelia of C. tarichi by immunocytochemistry with a specific antiserum for Na+/K+-ATPase in river fish samples. Density and area of MR cells were decreased in lake-adapted fishes. These results indicated that freshwater acclimation capacity is correlated with the size and distribution of MR cells in C. tarichi, in contrast to many teleost fishes.  相似文献
