Tomicus species (Coleoptera: Scolytidae) are serious pests of pines with a wide distribution in Europe, Asia and America. In Yunnan, south-western China,T. piniperda has destroyed more than 0.5 million ha ofPinus yunnanensis in the past 15 years. A blue stain fungus belonging to the genusLeptographium is associated with both the shoot-feeding and trunk-attacking stages of the beetle's life cycle. The fungus is morphologically similar to the anamorph ofOphiostoma crassivaginatum and toL. pyrinum, which are both characterised by short robust conidiophores and hyphae covered by a granular layer. Both these species have been isolated from conifers and are associated with insects. After comparing the fungus fromT. piniperda with similarLeptographium species, using light and scanning electron microscopy, we concluded that it represents a new taxon, which is described here asL. yunnanense sp. nov.  相似文献

Sydowia polyspora occurs often as an epiphyte or endophyte of conifers and is widely distributed around the world. It is also reported as a fungus associated with bark beetles. In this study, based on ITS sequence data and morphological characteristics, 14 strains collected from the beetle bodies and galleries of Tomicus minor and Tomicus yunnanensis infesting Pinus yunnanensis in Southwestern China were identified as S. polyspora. To assess the virulence of S. polyspora to P. yunnanensis, pathogenicity tests were conducted by inoculating the fungus in the stems and on needles of P. yunnanensis. The results after 30 days showed that the lesion lengths in the phloem after inoculation with S. polyspora were significantly longer than for the control. In addition, the fungus caused partly distinct chlorisis or discolouration of needles 14 days after inoculation. This suggests that S. polyspora is able to colonize stem phloem and pine needles of P. yunnanensis and thus may cause secondary damages to this host after being spread with bark beetles.  相似文献

Phlebiopsis gigantea (Fr.) Jül. is one of the most common fungal species in coniferous forests and commonly used as a biological control agent to prevent aerial infections of conifers by Heterobasidion fungi. We used feeding experiments to examine whether Hylobius abietis L. (Coleoptera: Curculionidae) could serve as a vector for P. gigantea and associated viruses by disseminating the fungus in its faecal pellets. Two different P. gigantea strains were used in the experiments: the virus-free Rotsop biocontrol strain and P. gigantea 93073 infected with the virus strain PgLV-1. The Rotstop strain showed 100% viability during insect feeding, while the viability of the virus-infected strain 93073 was only 35%. Virus persistence was 100% during the passage of the host fungus through the alimentary tract. Based on growth experiments using virus-infected and virus-free strains obtained by hyphal tip or oidial isolation, the presence of PgLV-1 did not significantly affect the growth rate of the host fungus.  相似文献

Mycelial growth of Phellinus weirii (Murr.) Gilb., a root pathogen of conifers in the Pacific Northwest, was studied in defined liquid media with different carbon and nitrogen sources and vitamins. The fungus grew significantly more on glucose, xylose, maltose, or fructose than on other carbon sources. Starch did not support growth. Maximum growth occurred in 4 weeks on all carbon sources except dextrin and sucrose, with which maximum growth occurred in 3 weeks. Of the nitrogen sources, peptone supported the best growth of the fungus; glutamic acid, serine, aspartic acid, alanine, leucine, ammonium sulphate, and urea supported significantly better growth than tyrosine, arginine, methionine, threonine, and glycine. Potassium nitrate, phenylalanine, sodium nitrate, lysine, proline, and cysteine inhibited growth. Thiamine hydrochloride was not absolutely required for growth of P. weirii, but better growth was obtained with its addition. The fungus showed no significant responses to a range of other vitamins.  相似文献

【目的】筛选高效降解木质素的菌株,并研究其以玉米秸秆为底物时木素降解酶活性。【方法】本研究以愈创木酚培养基和苯胺蓝培养基从吉林省不同经纬度的自然朽木及腐朽玉米秸秆土壤样品中分离、筛选得到高效降解木质素的菌株,并对其形态学鉴定,通过ITS序列分析构建系统发育树,分析菌株的分类地位。通过秸秆固体发酵过程产生的胞外木质素酶的活性分析,选出高效秸秆降解菌。【结果】筛选出1株高效降解秸秆的真菌,对其进行形态学特征和ITS序列分析,命名为白囊耙齿菌W2(Irpex lacteus W2)。该菌株在4–8 d内产生的锰过氧化物酶(Manganese peroxidase)呈上升趋势,并且在8 d达到峰值86.31 U/mL,与黄孢原毛平革菌(Phanerochaete chrysosporium)的最高酶活力45.86 U/mL相比,高出了88.20%(P0.01);该菌株的漆酶(Laccase)活力8 d时达到20.60 U/mL,比对照高40.76%(P0.05)。【结论】本研究分离到一株具有较强降解秸秆能力的真菌,初步鉴定为Irpex lacteus W2,具有较强的降解秸秆能力,其降解秸秆过程中产生较高的锰过氧化物酶与漆酶活力。  相似文献

A targeted genomics approach was used to construct a cDNA array of potential pathogen-regulated genes for investigating host–pathogen interactions in pine trees (Pinus species). This array, containing a nonredundant set of 311 cDNAs, was assembled by combining smaller sets of cDNAs generated by differential display or suppression subtraction hybridization using a variety of pathogen treatments and elicitors. The array was probed to identify host genes regulated by Fusarium circinatum, a necrotrophic fungus that incites pitch canker disease on pine stems. A set of 29 cDNAs were induced during the disease state. Notably, cDNAs on the array that were derived from experiments with fusiform rust, incited by Cronartium quercuum f. sp. fusiforme (a biotrophic fungus) were unregulated by Fusarium. The results imply distinct genetic responses in pine to diseases incited by necrotrophs and biotrophs. This cDNA collection expands the genomics toolkit for understanding interactions between conifers and their microbial associates in forest ecosystems.  相似文献

Larches (Larix spp.), deciduous conifers, occur in the northern hemisphere in cold-temperate and boreal climates – an environment normally thought to favor evergreen tree species. We compare foliar carbon isotope discrimination (Δ), instantaneous water use efficiency, total foliar nitrogen concentration, and specific leaf area (for a subset of sites) between Larix spp. and co-occurring evergreen conifers at 20 sites throughout the natural range of larches. Except for Larix occidentalis in the xeric Intermountain West, USA, Δ is significantly (P < 0.05) greater for larches than co-occurring evergreen conifers at 77% of the sites, suggesting that larches use water less efficiently. At elevations greater than 3000 m, the Δ of Larix spp. and co-occurring conifers converge, suggesting that water is not the limiting resource. Foliar nitrogen concentration and specific leaf area are two ecophysiological characteristics that are positively correlated with high photosynthetic capacity. Foliar nitrogen concentration is significantly greater for larches than evergreen conifers at 88% of the sites and specific leaf area is approximately three times greater for larches than co-occurring conifers. Future studies should examine the potential effect that global warming may have on the distribution of larch forests because the water use efficiency of larches is commonly less than co-occurring evergreen conifers and the boreal and high-latitude environments are likely to experience the greatest climate warming. Future increases in air temperature resulting from human activities may increase the water vapour pressure deficit (VPD) of the atmosphere. Understanding the responses of trees to spatial variation in VPD can strengthen our ability to predict how trees will respond to temporal changes in this important variable. Using published values, we tested the theoretical prediction that conifers decrease their investment in photosynthetic tissue (leaves) relative to water‐conducting tissue in the stem (sapwood) as VPD increases. The ratio of leaf/sapwood area (AL/AS) decreased significantly with increasing VPD in Pinus species but not in Abies, Pseudotsuga, Tsuga and Picea, and the average AL/AS was significantly lower for pines than other conifers (pines: 0.17 m2 cm?2; nonpines: 0.44 m2 cm?2). Thus, pines adjusted to increasing aridity by altering above‐ground morphology while nonpine conifers did not. The average water potential causing a 50% loss of hydraulic conductivity was ?3.28 MPa for pines and ?4.52 MPa for nonpine conifers, suggesting that pines are more vulnerable to xylem embolism than other conifers. For Pinus ponderosa the decrease in AL/AS with high VPD increases the capacity to provide water to foliage without escalating the risk of xylem embolism. Low AL/AS and plasticity in this variable may enhance drought tolerance in pines. However, lower AL/AS with increasing VPD and an associated shift in biomass allocation from foliage to stems suggests that pines may expend more photosynthate constructing and supporting structural mass and carry less leaf area as the climate warms.  相似文献

Abstract. Population structures were used to infer regeneration patterns of the conifers Podocarpus nubigena and Saxegothaea conspicua and associated angiosperm tree species in six stands in the temperate rainforests of the coastal ranges of southern Chile. On poorly-drained, nutrient-poor and high altitude sites, where most of the associated species were fairly shade-intolerant and light-crowned, dense all-aged populations and the presence of numerous saplings beneath the canopy suggested continuous regeneration of both P. nubigena and S. conspicua. On more favourable sites, where several of the associated angiosperms were highly shade-tolerant and densecrowned, both conifers, particularly P. nubigena, were less abundant, and their regeneration from seed appeared to be sporadic. S. conspicua's great longevity (≥ 750 yr) and frequent vegetative reproduction are probably crucial to its persistence in competition with shade-tolerant broadleaved species in undisturbed stands on favourable sites. Results of this study, and a review of related literature on southern temperate forests are not consistent with the hypothesis that heavily-shaded, infrequently-disturbed habitats are an evolutionary refuge for conifers. Sites likely to have high leaf area indices and infrequent disturbance are better exploited by shade-tolerant angiosperms.  相似文献

More than 50 kinds of endophytic fungi associated with Taxus chinensis were isolated and examined as a potential source of the imposing anticancer drug taxol. Of these, four isolates show ability to produce taxane when measured with the competitive inhibition enzyme immunoassay method. The most promising clone, DA10, identified as Mucor rouxianus sp., is the first rouxianus reported as taxol production fungus. The presence of taxol and its important precursors, such as 10-diacetyl baccatinIII (10-DAB) and baccatinIII, in theculture of this fungus was confirmed by reactivity with a taxane-specific monoclonal antibody, comparative chromatographic and mass spectrometric behavior, cytotoxity to liver carcinoma 7402, and molecular cloning of kernel fragment of taxadiene synthase gene. 【背景】开发生物甲烷资源是减轻化石燃料供求紧张的有效措施,而秸秆类原料的预处理及甲烷生产方法需要不断创新,从而进一步满足可持续发展。厌氧真菌与甲烷菌共培养能够通过假根侵入及纤维降解酶双重预处理秸秆并生产甲烷,但目前全世界被报道的骆驼胃肠道来源的厌氧真菌分离培养物仅有1株。【目的】从新疆准噶尔双峰驼瘤胃内容物中分离出新型厌氧真菌和甲烷菌共培养物,研究其在降解秸秆并联合生产生物甲烷方面的应用潜力。【方法】采用Hungate滚管纯化技术将从骆驼胃肠道中分离的厌氧真菌和甲烷菌共培养,对其进行形态学及分子学鉴定,随后厌氧发酵5种底物(稻秸、芦苇、构树叶、苜蓿秆和草木樨),研究产甲烷量、降解效果及主要代谢产物等方面的特性。【结果】筛选到的共培养物中的厌氧真菌为Oontomyces sp. CR1,甲烷菌为Methanobrevibacter sp. CR1。其在降解稻秸时表现出最高的木聚糖酶酶活力(21.64 IU/mL)及甲烷产量(143.39 mL/g-DM),甲烷生产特性较分离自其他动物宿主的厌氧真菌共培养物更优。【结论】共培养厌氧真菌与甲烷菌菌株CR1是一种新型高效降解菌株资源,其在利用木质纤维素生物质生产生物甲烷方面具有良好的应用前景。  相似文献

The microbial transformation of artemisinic acid (1) using cell culture of endophytic fungus Trichothecium roseum was investigated. Previously, we have reported two major metabolites, 3β-hydroxyartemisinic acid (2) and 3β,15-dihydroxyartemisinic acid (3) from the biotransformation of artemisinic acid by the fungus T. roseum CIMAPN1. Here in the present paper, we obtained a new minor compound 4 (5.2% in yield) along with compounds 2 and 3 through scale-up of biotransformation process of artemisinic acid using the same fungus. The structure of compound 4 was established as 3-oxoartemisinic acid on the basis of its IR, ESI-MS, HRMS, 1?D (1H and 13C, DEPT), and 2?D (COSY, HSQC, HMBC) NMR spectral data analysis. The possible reaction mechanism of the formation of 3-oxoartemisinic acid from artemisinic acid was proposed. Furthermore, all the three metabolites along with the artemisinic acid were evaluated for their antifungal activity against the three fungal strains Candida albicans (ATCC 14053), Candida albicans clinical isolates and Candida kefyr (ATCC 204093). 3-Oxoartemisinic acid was the most active (4 to 16 times more potent than artemisinic acid) with MIC ranges from 125 to 500?µg/mL among all tested compounds. This study suggested that the artemisinic acid molecule has a great potential to be exploited for further biotransformation by the different fungi and can produce chemically diverse molecules with better biological activity.  相似文献

To obtain new bioactive natural products, the effect of acidic stress on the metabolites of an aciduric fungus was investigated. This fungus, Penicillium sp. OUCMDZ-4736, which was isolated from the sediment around roots of mangrove (Acanthus ilicifolius), produced different compounds and higher yields under pH 2.5 than under neutral conditions. Using spectroscopic analyses and calculations, three new anthraquinone derivatives (13) were isolated and identified from the acidic fermentation broth (pH 2.5) of OUCMDZ-4736. Compound 1 showed much stronger anti-hepatitis B virus activity than that of the positive control, lamivudine, strongly inhibiting HBsAg and HBeAg secretion from HepG2.2.15 cells. These results show that extremophiles are a valuable resource of bioactive compounds, and that pH regulation is an effective strategy to induce metabolite production in aciduric fungi.

The filamentous fungus, Lambertella corni-maris (L. corni-maris), a mycoparasite on Monilinia fructigena, produces the antibiotics, lambertellols A (1), B (2), and lambertellin (3), in a substantial amounts under acidic conditions, whereas these antibiotics were hardly detected when the fungus was cultured on a potato-sucrose (PS) medium without added acids. Our investigations also revealed that the host, M. fructigena, changed its surroundings into acidic conditions, suggesting that the acidic conditions acted as kairomones that stimulated the production of 13.  相似文献

An endophytic fungus was isolated from the root of the medicinal plant Moringa oleifera Lam. Based on analyzing the rDNA sequence, the fungus was identified as Nigrospora sp. This is the first report of the isolation of endophytic Nigrospora from M. oleifera. By bioassay-guided fractionation, four antifungal secondary metabolites were isolated from liquid cultures of the fungus Nigrospora sp. LLGLM003, and their chemical structures were determined to be griseofulvin (1), dechlorogriseofulvin (2), 8-dihydroramulosin (3) and mellein (4) on the basis of spectroscopic analyses. Compound 2, 3 and 4 were isolated from Nigrospora sp. for the first time. In vitro antifungal assay showed that griseofulvin displayed clear inhibition of the growth of 8 plant pathogenic fungi. Dechlorogriseofulvin and mellein exhibited only weak antifungal activities, whereas 8-dihydroramulosin displayed no antifungal activities.  相似文献

药用植物内生放线菌具有合成天然活性化合物的潜力,放线菌新种是寻找新型抗生素先导化合物的一个重要来源。【目的】挖掘药用植物地黄内生放线菌资源,并对地黄轮纹病拮抗菌株leaf-16进行新种鉴定。【方法】本研究采用五步消毒法分离河南道地药材地黄的内生放线菌,以地黄轮纹病原真菌草茎点霉(Phoma herbarum)为指示菌,采用平板对峙法筛选对该病菌有抑制作用的菌株,16S rRNA基因测序发现一株抗地黄轮纹病的放线菌新种leaf-16。通过形态、生理生化、细胞壁化学组分和分子生物学等特征对菌株leaf-16进行多相分类学鉴定。【结果】经平板对峙实验得到8株抗地黄轮纹病的放线菌,其中菌株leaf-16经16S rRNA基因测序、形态比较、生理生化、化学组分和分子生物学以及DNA-DNA杂交分析,确定菌株leaf-16为1株链霉菌新种,并命名为Streptomyces folium。【结论】菌株leaf-16为1株链霉菌新种,具有抑制地黄轮纹病原真菌的活性,为进一步分离新型抗地黄轮纹病的生物制剂奠定物质基础。  相似文献

The virulence of two strains of the entomopathogenic fungus Verticillium lecanii to the aphids Myzus persicae, Aphis gossypii and Brevicoryne brassicae was bioassayed. The strain (V24 and V18) tested were capable of infecting all three aphids species, but their virulence determined by LC50 and LT50 varied. The strain (V24) originally isolated from M. persicae showed the highest virulence against the homologous aphid species, but the whitefly (Trialeurodes vaporariorum) derived strain (V18) showed also the highest virulence against an aphid species. The variability of the bioassay results and the impossibility of defining clearly traits associated with the virulence of a fungus strain toward a specific insect species are discussed. Effect of additives (glycerin, soyflour and rape oil) was studied for strain improvement. Considerable improvement of the strains virulence was not observed with glycerin and soyflour. The results are discussed in conjunction with the non-convincing results published earlier.  相似文献

We investigated the climate response of three Siberian taiga species, Larix cajanderi, Picea obovata, and Pinus sylvestris, across a latitudinal gradient in central Siberia. We hypothesized that warming is more frequently associated with increased growth for evergreen conifers (P. obovata and P. sylvestris) than for L. cajanderi, and for northern than for southern sites; we also hypothesized that increased growth is associated with a positive trend in normalized difference vegetation index (NDVI). In mixed stands, growth of L. cajanderi and P. obovata increased over time, but the larger growth increases in P. obovata may presage a shift in competitive balance between these species. Climate response varied among and within populations of all species, and positive responses to temperature prevailed at northern sites, where trees grew faster in years with warm early summers. Negative responses to warming declined along the south to north latitudinal gradient. We observed considerable variability in climate response within populations which even exceeded that among species or sites. Tree response to climate was correlated with NDVI trends, indicating that patterns of tree‐growth response to climate were indicative of a coherent landscape‐scale response to warming. Our findings suggest that increased productivity with warming is likely only in the northern reaches of the Siberian taiga. An increased prevalence of evergreen conifers in areas currently dominated by deciduous Larix species also seems likely.  相似文献

Abstract

A new die back symptoms in many avocado orchards had been reported in Palestine. The disease is associated with the Ambrosia beetle Euwallacea fornicatus. Stem samples from infected avocado trees with obvious symptoms were collected from different regions in Palestine. Stem cuttings and dissected adult and galleries of the insect were placed on potato dextrose agar media and incubated for 5–7?days at 25?°C. PCR amplification using EF1/2 specific primers was performed to identify the isolated fungus. The resulting PCR products were sequenced. BLASTn search showed 99% similarity with Fusarium euwallaceae (Accession Nos. JX891785.1, JQ723763.1, JQ723762.1 and JQ723761.1). The isolated fungus was identified as F. euwallaceae (Genbank accession no. MK054177).  相似文献

【背景】小麦/玉米轮作是中国粮食作物主要种植模式之一,目前对小麦/玉米轮作田根际土壤微生物差异变化缺乏全面的了解。【目的】明确小麦/玉米根际土壤微生物差异变化并了解其潜在功能。【方法】以小麦/玉米根际土壤为材料,运用细菌16S rRNA基因和真菌rDNA ITS基因测序,分析小麦/玉米根际土壤微生物多样性。【结果】玉米季微生物丰富度高于小麦季,而多样性无明显差异。放线菌门(Actinobacteria)、变形菌门(Proteobacteria)、酸杆菌门(Acidobacteria)和绿弯菌门(Chloroflexi)为小麦季和玉米季根际土壤的优势细菌门,优势真菌门为子囊菌门(Ascomycota)。小麦季和玉米季共有细菌和真菌分别是631个和261个,小麦季特有细菌和真菌分别是38个和58个,玉米季特有细菌和真菌分别是25个和39个。LEfSe分析(LDA阈值为2)细菌和真菌表明,放线菌纲(Actinobacteria)和微囊菌目(Microascales)在小麦季富集,鞘脂单胞菌目(Sphingomonadales)和银耳纲(Tremellomycetes)在玉米季富集。小麦季、玉...  相似文献
