The enzymatic procedures for the resolution of dl-lysine such as asymmetric synthesis of acyl l-lysinc anilide and acyl dl-lysines have been studied. As a result, the procedure consisting in the enzymatic asymmetric hydrolysis of ε-benzoyl-α-acctyl-dl-lysine was found to be the most advantageous for the resolution of dl-lysine.  相似文献

As a part of the studies intending to clarify biological significance of the presence of acylase, enzymatic activity which hydrolyzes acyl amino acids, its activity in yeast was investigated. As a results, the occurrence in yeast of acylase activity was confirmed for the first time.

In order to study the enzymatic properties of this acylase activity, experiments were carried out with the enzyme preparation from brewer’s yeast. As a result of the investigation, yeast acylase was found to be able to hydrolyze a number of acyl amino acids, of these chloroacetyl derivatives especially readily, as in the case of previously studied acylase activity in other sources such as mold acylase. Several observations on the influence of metal ions and inhibitors, optical specificity and others were also presented.  相似文献

The solbilities of l-basic amino acids in the type of free, monohydro-chloride and dihydrochloride in water were determined, and the results were formulated as follows.

l-Arginine monohydrochloride: log S = 1.6532+0.01301t (t is from 0°~70°C)

l-Lysine monohydrochloride dihydrate: log S = 1.6990+0.01294t (t is from 0°~55°C)

l-Lysine monohydrochloride monohydrate: log S = 1.7404+0.01256t (t is from 55°~70°C)

l-Lysine dihydrochloride: log S = 2.2138+0.00409t (t is from 0°~70°C)

l-Histidine dihydrochloride: log S = 1.9085+0.00265t (t is from 0°~70°C)

Three component systems (basic amino acid, hydrochloric acid, water) were studied and the solubilities in mixed solution system of alcohol-water were also investigated.  相似文献

Fundamental studies on the cultural conditions for the amino acids production from pentoses and hexoses employing a strain of our new isolates named Brevibacterium pentoso-aminoacidicum nov. sp. were carried out.

As a result of these experiments, it became possible to obtain about 30% of alanine and 10% of L-glutamic acid based on xylose, and alanine from glucose with a yield of about 40% in the proper conditions.  相似文献

This work was undertaken to determine whether the degradation of sugars by Brevibacterium pentoso-aminoacidicum nov. sp., a bacterium capable of producing amino acids from pentoses and hexoses, was due to constitutive or inducible enzymes. It was also intended to clarify the reason for the substrate specificity in the fermentation of sugars by this bacterium. After a series of experiments using washed resting cells grown on various kinds of sugars or their cell-free extracts, it was found that the enzymes involved in the degradation of pentoses were inducible, while those of hexose metabolism were constitutive. The activities of several enzymes related to the pathways of pentose metabolism were demonstrated and the substrate specificity of sugar degradation by this strain was explained satisfactorily by the inducer specificity of these enzymes.  相似文献

Decarboxylation of about twenty kinds of α, β and γ-amino acids in the reaction with glyoxal or ninhydrin was investigated. The decarboxylation rate of amino acids proved that steric and polar effects had important roles in the reaction.

From the data of pK2 values and decarboxylation rates of amino acids, it can be concluded that under a similar steric environment, the decarboxylation rate depends on the anion concentration of amino acids.

Besides carbon dioxide, acetaldehyde, 2-propanone and propionaldehyde were respectively detected from the reaction of β-alanine, β and γ-amino-n-butyric acids with glyoxal or ninhydrin. The decarboxylation mechanism of these amino acids seemed to take place through the corresponding β- or γ-keto acid.

Oxygen absorption was also observed from the reaction of amino acids with dicarbonyl compounds.  相似文献

本文采用热分析方法(TG,DTA)和红外光谱等手段,对四种天然氨基酸(盐酸赖氨基酸,色氨酸,天门冬氨酸,苯丙氨酸)的热稳定性和热分解机理进行了研究,发现这四种天然氨基酸热分解的共同特点是首先失去羧基,形成胺的中间产物,然后是中间产物分解,且脱羧温度与β位碳原子上所连的基团密切相关,从电荷密度等因素出发对此进行了分析和讨论。  相似文献

Two substances besides rubiginol, of positive ferric chloride reaction were isolated from a shaking culture of Gluconoacetobacter liquefaciens, using glucose as the substrate. One of them was found to be a new γ-pyrone derivative and the composition was concluded to be 3,5-dihydroxy-l,4-pyrone-2-carboxylic acid. The other substance was found to be comenic acid. Assumptions were made with regard to the mechanism of the formation of these γ-pyrone derivatives from glucose.  相似文献

An electro-energizing fermentation (E-E F) method has been developed. In this method, a direct electrical current is applied to a microbial culture to accelerate the reductive metabolism of microorganisms or to impart profitable effects to microbial cells. This E-E F method was applied to l-glutamic acid fermentation by Brevibacterium flavum No. 2247. When glucose was used as a substrate, the addition of 0.01 mm neutral red (NR), redox dye (electron carrier), to the fermentation broth at the beginning of cultivation was effective for l-glutamate (l-Glu) production. A direct current of 200~300 μA/cm2 at 1.5 V was applied through out the cultivation of this bacterium. This resulted in about a 10% increase in yield of l-Glu.  相似文献

Evidence from isotope competition studies and enzymic studies indicates that n-acetyl glutamic semialdehyde, alpha-n-acetyl-l-ornithine, l-ornithine and l-citrulline are intermediates between glucose and arginine in cells of Paul's Scarlet Rose. 观察了支链氨基酸(BCAA)对大鼠运动能力和血清游离氨基酸代谢的影响。实验用21只雄性wistar大鼠,随机分为3组:正常组、游泳对照组和游泳补充BCAA组。2个运动组每天游泳训练1h,10天后游泳6h,观察补充BCAA对大鼠游泳运动能力和血清游离氨基酸水平的影响。实验结果表明,补充BCAA可明显提高大鼠游泳存活率,抑制血清中必需氨基酸、非必需氨基酸和总氨基酸水平升高,游泳运动后大鼠的血清中乳酸和LDH的升高幅度有所降低,抑制骨骼肌LDH活力的下降。说明补充BCAA可明显提高大鼠的运动能力,减少运动造成的蛋白质分解  相似文献

The adsorption of amino acids on minerals and their condensation under conditions that resemble those of prebiotic earth is a well studied subject. However, which amino acids should be used in these experiments is still an open question. The main goal of this review is to attempt to answer this question. There were two sources of amino acids for the prebiotic earth: (1) exogenous -- meaning that the amino acids were synthesized outside the earth and delivered to our planet by interplanetary dust particles (IDPs), meteorites, comets, etc. and (2) endogenous -- meaning that they were synthesized on earth in atmospheric mixtures, hydrothermal vents, etc. For prebiotic chemistry studies, the use of a mixture of amino acids from both endogenous and exogenous sources is suggested. The exogenous contribution of amino acids to this mixture is very different from the average composition of proteins, and contains several non-protein amino acids. On the other hand, the mixture of amino acids from endogenous sources is seems to more closely resemble the amino acid composition of terrestrial proteins.  相似文献

在纯乙酸溶剂中,以乙酸酐酰化消旋酪氨酸,消旋温度为90℃,L-酪氨酸和乙酸酐的摩尔比为12,每消旋1gL-酪氨酸时,溶剂乙酸的用量为5mL,效果较好。同样的方法应用于L-脯氨酸,L-苏氨酸,L-缬氨酸,L-赖氨酸,L-半胱氨酸等氨基酸,消旋率均可达到100%。  相似文献

BACKGROUND: The composition and sequence of amino acids in a protein may serve the underlying needs of the nucleic acids that encode the protein (the genome phenotype). In extreme form, amino acids become mere placeholders inserted between functional segments or domains, and--apart from increasing protein length--playing no role in the specific function or structure of a protein (the conventional phenotype). METHODS: We studied the genomes of two malarial parasites and 521 prokaryotes (144 complete) that differ widely in GC% and optimum growth temperature, comparing the base compositions of the protein coding regions and corresponding lengths (kilobases). RESULTS: Malarial parasites show distinctive responses to base-compositional pressures that increase as protein lengths increase. A low-GC% species (Plasmodium falciparum) is likely to have more placeholder amino acids than an intermediate-GC% species (P. vivax), so that homologous proteins are longer. In prokaryotes, GC% is generally greater and AG% is generally less in open reading frames (ORFs) encoding long proteins. The increased GC% in long ORFs increases as species' GC% increases, and decreases as species' AG% increases. In low- and intermediate-GC% prokaryotic species, increases in ORF GC% as encoded proteins increase in length are largely accounted for by the base compositions of first and second (amino acid-determining) codon positions. In high-GC% prokaryotic species, first and third (non-amino acid-determining) codon positions play this role. CONCLUSION: In low- and intermediate-GC% prokaryotes, placeholder amino acids are likely to be well defined, corresponding to codons enriched in G and/or C at first and second positions. In high-GC% prokaryotes, placeholder amino acids are likely to be less well defined. Increases in ORF GC% as encoded proteins increase in length are greater in mesophiles than in thermophiles, which are constrained from increasing protein lengths in response to base-composition pressures.  相似文献
