The triterpene alcohol fraction was isolated from the seed oil of horse chestnut; it amounted to 42.2% of the unsaponifiable matter, i.e. 0.93% of the oil. The components were identified as taraxerol, β-amyrin, butyrospermol, parkeol, 5α-tirucalla-8,23-dien-3β-ol, α-amyrin and 24-methylenecycloartenol. Butyrospermol and 5α-tirucalla-8,23- dien-3β-ol were the major components and amounted to 60% of the fraction. The latter has not been identified in any vegetable oils yet.  相似文献

As a plant-derived pentacyclic triterpenoid, β-amyrin has been heterogeneously synthesized in Saccharomyces cerevisiae. However, β-amyrin is intracellularly produced in a lower gram scale using recombinant S. cerevisiae, which limits the industrial applications. Although many strategies have been proven to be effective to improve the production of β-amyrin, the intracellularly accumulation is still a challenge in reaching higher titer and simplifying the extraction process. To solve this problem, the amphiphilic β-cyclodextrin (β-CD) has been previously employed to aid the efflux of β-amyrin out of the cells. Nevertheless, the supplemented β-CD in the medium is not consistent with β-amyrin synthesis and has the disadvantage of rather high cost. Therefore, an aided-efflux system based on in situ synthesis of β-CD was developed in this study to enhance the biosynthesis of β-amyrin and its efflux. The in situ synthesis of β-CD was started from starch by the surface displayed cyclodextrin glycosyltransferase (CGTase) on yeast cells. As a result, the synthesized β-CD could capture 16% of the intracellular β-amyrin and improve the total production by 77%. Furthermore, more strategies including inducing system remodeling, precursor supply enhancement, two-phase fermentation and lipid synthesis regulation were employed. Finally, the production of β-amyrin was increased to 73 mg/L in shake flask, 31 folds higher than the original strain, containing 31 mg/L of extracellular β-amyrin. Overall, this work provides novel strategies for the aided-efflux of natural products with high hydrophobicity in engineered S. cerevisiae.  相似文献

From the rhizome of Iris germanica one new hexaoxygenated isoflavone, two new polyoxygenated isoflavone glucosides, the known isoflavonoids irisolidone, irigenin and iridin and acetovanillone, sitosterol, α-amyrin and β-amyrin were isolated. From the neutral fraction of the n-hexane extract of stem bark of Mangifera indica, (var./cv Banganpalli) six new tetracyclic triterpenoids, cycloart-24-ene-3β,26-diol, the C-24 epimers of cycloart-25-ene-3β,24,27-triol, the C-24 epimers of cycloartane-3β,24,25-triol and 3-ketodammar-24E-ene-20S,26-diol together with the known compounds cycloartenol, α-amyrin, β-amyrin, sitosterol 3β-hydroxycycloart-25-en-26-al, dammarenediol II, the C-24 epimers of cycloart-25-ene-3β,24-diol, 24-methylenecycloartane-3β,26-diol, ψ-taraxastane-3β,20-diol and ocotillol II have been isolated. The acidic fraction of the same extract, on esterification with diazomethane followed by chromatography, yielded methyl mangiferonate, methyl isomangiferolate, methyl mangiferolate and the diazomethane adduct of methyl mangiferolate. The structures were elucidated by spectroscopic and chemical methods.  相似文献

3-Oxo-friedelan-20α-oic acid, named as maytenonic (polpunonic) acid, has been isolated along with β-amyrin and sitosterol from Gymnosporia emarginata. 1H and 13C NMR signals have been assigned for the structure. X-ray analysis of the single crystal confirmed that the carboxylic acid is α-oriented at C-20 and the D/E rings of this D:A-friedo-oleanane are in chair-chair conformation. 13C NMR data of the present compound also enabled the assignment of the C-20α- and β-methyl carbons in friedelin.  相似文献

A new tropane alkaloid, named the 7β-acetoxy-3β,6β-dibenzoyloxytropane (1), was isolated from a methanol extract of Erythroxylum rimosum O.E. Schulz leaves. Other known compounds were detected, including quercetin, kaempferol-3-O-α-l-arabinofuranoside, (+)-catechin, epicatechin, quercetin-3-O-α-arabinofuranoside, quercetin-3-O-α-arabinopyranoside, quercetin-3-O-β-arabinopyranoside, quercetin-3-β-glucopyranoside, kaempferol, quercetin-3-O-β-galactopyranoside, β-sitosterol, α-amyrin, β-amyrin, and the ester derivatives of these two amyrins. Compound 1 exhibited weak inhibition of acetylcholinesterase. Structural identification was performed using IR, ESIHRMS and one- and two-dimensional NMR data analyses and confirmed by comparison with literature data.  相似文献

In Calendula officinalis leaves the cyclization of squalene to β-amyrin and its further oxidation to oleanolic acid as well as the biosynthesis of all derivatives of oleanolic acid 3-glucoside and some derivatives of oleanolic acid 3-glucuronoside occur in the microsomal fraction. The final metabolites of oleanolic acid 3-glucoside series i.e. pentaglycosides, are translocated from this fraction, one to the cell wall and plasmalemma fraction and the other to the cytosol. The derivatives of oleanolic acid 3-glucoronoside are synthesized partially in other fractions and accumulate in the different membraneous structures of the cell.  相似文献

A new triterpenoid, calaminthadiol, 3,4-seco-4(23),12-ursadien-3,28-diol, was isolated from the leaves of Satureia calamintha and Satureia graeca. Its structure was elucidated by spectroscopic methods and partial synthesis. The natural occurrence of this compound can be conclusive for the chemotaxonomic characterization of the genus because of the rarity of the 3,4-fission in the α- and β-amyrin skeletons. A biogenetic mechanism for its formation is proposed. Sitosterol, stigmasterol and campesterol were also isolated.  相似文献

Abstract: Antibodies specific for α-N-acetyl-β-endorphins have been prepared by injecting into rabbits either α-N-acetyl-β-endorphin(1-31) or [α-N-acetyl, ε-acetyl-Lys9]-β-endorphin(1-9) linked by carbodiimide to bovine thyroglobulin. Both antisera were used to develop specific radioimmunoassays for α-N-acetyl-β-endorphins. The radioimmunoassays were used to measure α-N-acetylated β-endorphins in extracts of pituitary regions from different species. By comparison of the amounts of total β-endorphin and α-N-acetyl-β-endorphin immunoreactivity, a relative ratio of β-endorphin acetylation was obtained. The relative acetylation of β-endorphin was highest in rat posterior-intermediate lobe extracts (>90%). Beef and monkey intermediate lobes had a lower degree of acetylation (53 and 31%, respectively). Anterior lobe extracts from all three species contained low amounts of acetylated β-endorphin. Human pituitary extracts did not contain acetylated β-endorphins. By the use of cation exchange and high performance liquid chromatography, six different acetylated derivatives and fragments of β-endorphin were resolved in extracts of rat posterior-intermediate pituitaries. Two of these peptides corresponded to α-N-acetyl-β-endorphin(1-31) and -(1-27). One acetylated β-endorphin fragment had the same size as α-N-acetyl-β-endorphin(1-27) but was eluted earlier from the cation exchange column. This peptide had full cross-reactivity with antibodies directed against the middle and amino-terminal parts of β-endorphin. Compared with α-N-acetyl-β-endorphin(1-27), it had much less cross-reactivity with antibodies directed against the COOH-terminal part of β-endorphin, suggesting that it was a COOH-terminally modified derivative of β-endorphin(1-27). The remaining N-acetylated β-endorphin derivatives were eluted even earlier from the cation exchange column. The majority of these fragments were slightly larger in size than y-endorphin, i.e., β-endorphin(1-17), but smaller than β-endorphin(1-27). They had full cross-reactivity in an amino-terminally directed β-endorphin radioimmunoassay and a greatly diminished cross-reactivity with antibodies to the middle region of β-endorphin.  相似文献

α-Amyrin is a plant-originated high-valued triterpene that is highly effective against several pathological ailments. α-Amyrin production by engineered Saccharomyces cerevisiae has been achieved by introducing α-amyrin synthase (αAS). However, the low yield of α-amyrin highly limits its industrial application; the low catalytic activity of αAS and the toxic effect of α-amyrin have been considered key elements. In this study, the highest yield of α-amyrin was obtained in engineered S. cerevisiae by remodeling α-amyrin synthase MdOSC1 and expanding the storage pool. The yield of α-amyrin was increased to 11-fold higher than that of the control by the triple mutant MdOSC1N11T/P250H/P373A obtained based on the modeling analysis. Furthermore, key genes of MVA pathway were overexpressed to provide sufficient precursors, and DGA1 (Diacylglycerol acyltransferase) was overexpressed to expand the intracellular storage capacity. Finally, the as-constructed aAM12 strain produced 213.7 ± 12.4 mg/L α-amyrin in the shake flask and 1107.9 ± 76.8 mg/L in fed-batch fermentation; the fermentation yield was 106-fold higher than that of the original aAM1 strain under the same conditions, representing the highest α-amyrin yield in yeast reported to date. Microbial production of α-amyrin with over 1 g/L will be suitable for commercialization and can accelerate the industrial production of α-amyrin in yeast.  相似文献

During the last decades, different natural compounds have been demonstrated to modulate inflammatory pathways. In this study, methanolic extract of Echinophora tenuifolia L. inflorescences was investigated for its chemical composition and in vitro antioxidant and anti-inflammatory properties. Phytochemical profile was investigated by means of high-performance thin layer chromatography (HPTLC) and gas chromatography–mass spectrometry analyses. Myristic acid and palmitic acid were found to be the major constituents. Six terpenes were identified: α-phellandrene, o-cymene, dihydroactinidiolide, neophytadiene, phytol and β-amyrin, and two phenolic compounds: carvacrol and ferulic acid. HPTLC analysis of the ethyl acetate fraction highlighted the presence of the flavonoid glycosides rutin and quercitrin. This sample showed the best diphenylpicrylhydrazyl scavenging capacity, with an IC50 value equal to 40.39 μg/ml, and the strongest capacity to protect linoleic acid from peroxidation, as assessed by the β-carotene bleaching test (IC50 = 16.31 μg/ml). All samples inhibited nitric oxide production in cell supernatants in a dose-dependent manner, being the two apolar fractions (n-hexane and dichloromethane) even more active than the positive control indomethacin. A relevant biological activity was observed for dichloromethane fraction (IC50 value equal to 39.97 μg/ml). Obtained results indicate that this sample could be an excellent candidate for further investigations aimed at the development of new antioxidant and anti-inflammatory drugs.  相似文献

Propolis or bee glue is commonly named as a natural resinous mixture produced by honeybees (Apis mellifera) from substances collected from parts of plants, buds, and exudate. The result of the ethyl acetate - methanol (3 : 2) volume by volume fraction yielded a total of two compounds namely betulinic acid and β-amyrin isolated from Bodji Dirmaji and Fincha'a district propolis, respectively. The crude ethanolic extract was portioned with the different solvent systems by increasing the polarities in the following order of hexane, ethyl acetate, and methanol. Column chromatographic method on normal silica gel was used to isolate the compounds. The structures of the compounds were characterized using 1D NMR techniques. The study revealed that western Ethiopian propolis was rich in saponins, tannins, flavonoids, steroids, triterpenes, and glycosides. The antibacterial activity for the isolated compound (betulinic acid) showed the highest inhibition for S. aureus (11.2±1.6), E. coli (17.7±1.1), and A. niger (12.6±1.2) mm.  相似文献

The inflammatory disorders represent a serious health issue. Certain Cissus species possess anti-inflammatory effect. Cissus rhombifolia Vahl. leaves’ anti-inflammatory activities and phytoconstituents are poorly characterized. In this study, 38 constituents were tentatively characterized in Cissus rhombifolia Vahl. leaves’ aqueous methanolic extract (CRLE) using high-performance liquid chromatography combined with mass spectrometry (HPLC/MS) and Proton Nuclear Magnetic Resonance (1H-NMR). Myricetin, β-amyrin, and alliospiroside A, were isolated from CRLE using column chromatography. The anti-inflammatory effect of CRLE and its isolated compounds were studied in lipopolysaccharide (LPS)-induced RAW 264.7 cells. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT assay) was used to assess how CRLE and its isolated compounds affected cell viability. Further, its effects on the production of intracellular NO, and inflammatory cytokines cyclooxygenase-2 (COX-2), tumor necrosis factor alpha (TNF-α), and interleukin 6 (IL-6) were assessed by the Griess test, and cytokine enzyme-linked immunosorbent assays, respectively. CRLE and its isolated compounds, myricetin, β-amyrin, and alliospiroside A decreased the NO production. Western blotting was performed to assess the protein expression levels of the inflammatory cytokines inducible nitric oxide synthase (iNOS). Alliospiroside A downregulated IL-6, TNF-α, and COX-2 and inhibited the expression of iNOS. CRLE and its compounds represent effective alternative candidate to treat inflammatory diseases.  相似文献

Some glycosidases in light-grown cucumber (Cucumis sativus L. cv. Aonaga-jibae) hypocotyl sections were examined with respect to their localization and relation to endogenous and IAA-induced growth. Frozen-thawed sections were used directly for measurement of enzyme activities, and β-glucosidase, α- and β-galactosidases and β-xylosidase were assayed by using p- or o-nitro-phenylglycopyranosides as substrates. The order of the activity of these enzymes were β -glucosidase > β -galactosidase =α-galactosidase > β-xylosidase. No activity of α-glucosidase was detected. High glycosidase activities were found in the youngest region of the hypocotyl, where the endogenous growth rate was highest. However, there was no significant difference in the activities of this region between seedlings at different growth stages. Among the enzymes tested, β -glucosidase showed a high correlation with the endogenous growth rate. β-glucosidase was found to be mostly associated with the cell wall fraction, while β-galactosidase was rather found in the soluble fraction of the cell. Separation of the epidermis from the section showed that a very high activity of β-glucosidase was associated with the epidermis. In both whole sections and isolated cell wall fractions, IAA was shown to have no effect on the activities of β-glucosidase and β-galactosidase.  相似文献
