The influence of Ca homoionic clay minerals (montmorillonite, illite, and kaolinite) on the activity,K m , andV m values of acid phosphatase was examined in model experiments. At each substrate (p-nitrophenyl phosphate) level tested, the addition of increasing amounts of clays (50, 100, and 150 mg, respectively) decreased the activity and increased theK m value from 1.43×10–3 m PNP (in the soluble state) to 82.3×10–3M (montmorillonite), 8.02×10–3 m (kaolinite), and 7.65×10–3 m (illite) at the 150 mg level. The maximum enzyme reaction velocity (V m ) remained nearly constant at different amounts of kaolinite and illite, but increased remarkably with rising quantities of montmorillonite. Apparently, the substrate affinity of sorbed acid phosphatase is significantly lower with montmorillonite than with kaolinite or illite. This may be ascribed to an intensive sorption of both substrate and enzyme to the surface as well as to interlattice sites of montmorillonite.  相似文献

Summary The uptake and release of tylosin by Wyoming bentonite, Ca++ montmorillonite, illite (Fithian Illinois) and kaolin (china clay) was studied. The adsorption capacities of bentonite and montmorillonite for tylosin were found to be 190 and 65 g mg–1 respectively, while the adsorption capacities of illite and kaolinite were found to be 22 and 6.5 g mg–1. 7.6 and 7.4% respectively of the tylosin adsorbed by bentonite and montmorillonite, and 30 and 25% respectively of the tylosin adsorbed by illite and kaolinite, was released in phosphate buffer.The presence of tylosin in two different soils amended with tylosin fermentation waste was also studied, and the effect of temperature on tylosin release was examined in one of the soils. The concentration of tylosin in the soil water was found to be related more to the concentration of tylosin fermentation waste than to the incubation temperature. The amounts of tylosin present in the initial leachates of the two soils, a modified John Innes compost and a limestone waste, were found to be in excess of the minimum inhibitory concentrations for susceptible bacteria. It is hypothesised that in these particular soils if the tylosin is not broken down by microbial action it could affect susceptible populations of bacteria.  相似文献

Abstract— High concentrations of dopamine were found in the nucleus accumbens and olfactory tubercle of the rat brain using a radiochemical enzymatic assay technique. An active uptake system for [3H]dopamine that is temperature sensitive and dependent on external sodium ions is present in synaptosome-rich homogenates of these two brain areas. This uptake process is potently inhibited by benztropine (IC50= 2.0 × 10-7m ). Dextroamphetamine d was 4.5 times more potent than 1-amphetamine in inhibiting dopamine uptake in the nucleus accumbens and six times more potent in the olfactory tubercle and corpus striatum. Low concentrations of dopamine caused an increase in adenosine 3′5′-monophosphate (cyclic AMP) formation in homogenates of both the nucleus accembens and olfactory tubercle. This effect was potently blocked by chlorpromazine. The α-adrenoceptor antagonist phentolamine weakly antagonized the stimulation of this adenylate cyclase by dopamine, but the β-adrenoceptor antagonist propranolol did not.  相似文献

Accumulation of cobalt and cesium by the microalga Scenedesmus obliquus and the cyanobacterium Synechocystis PCC 6803 has been characterized at metal concentrations ranging from 1–100 µM in the presence of three clay minerals, montmorillonite, illite, and kaolinite. The majority of metal uptake over a 4 h period consisted of rapid binding to the clay mineral-cell aggregates, and was unaffected by incubation in the dark or by the presence of the metabolic inhibitor carbonyl cyanide-3-chlorophenyl hydrazone (CCCP). This was followed by a slower, energy-dependent uptake of metal by the cell components of the mixtures, which was inhibited by incubation in the dark or in the presence of CCCP. The initial phase of uptake by the clay mineral-cell mixtures and mixture components alone conformed to a Freundlich adsorption isotherm, the order of uptake for both cobalt and cesium being montmorillonite-cells > illite-cells > kaolinite-cells. S. obliquus-clay mineral mixtures accumulated more cobalt and cesium than Synechocystis PCC 6803-clay mineral mixtures. On a dry weight basis, clay minerals alone accumulated greater amounts of metals than clay mineral-cell mixtures, which accumulated more than the cells alone. However, when the same data was expressed as amount of metal adsorbed per unit surface area, S. obliquus, in most cases, adsorbed greater amounts of cobalt and cesium than the clay minerals or Synechocystis PCC 6803. As the proportion of clay in a cell-clay mineral mixture was increased, the amount of metal accumulated also increased. Reduced accumulation of cobalt and cesium by cell-clay mineral mixtures, exhibited by equal amounts of the individual components added together, indicated that the formation of clay-cell aggregates had masked some of the binding sites normally available to metal ions. Accumulation of cobalt and cesium by all clay mineral-cell mixtures was dependent on the external pH and NaCl concentration, and decreased with decreasing pH and increasing external NaCl concentration. Investigation of the weathering of silicate minerals is helpful to understand the process of soil development, cycling of nutrient elements, and potential applications in fixation of carbon dioxide from the atmosphere through carbonate precipitation. In this study, weathering experiments of calcium-montmorillonite were conducted using Paenibacillus sp. strain SB-6 for 70 days. The results indicated that the Si4+, Al3+, Ca2+ and Na+ concentrations in the medium of the biotic experiments were evidently higher than those of the abiotic experiments, and that Paenibacillus sp. could help the transformation of partial montmorillonite into an illite–montmorillonite mixed-layer. In the process of illitization, K+ went into the interlayer of montmorillonite and hydrated Ca2+ and Na+ released from it. In the late stage of the experiments, the Ca2+ released from montmorillonite combined with carbonate ions generated by the bacterial metabolism, forming calcite.  相似文献

Abstract

Effect of light on uptake by photosynthetic tissues in the absence of co2. — The effect of light and of CO2 on the uptake of 86Rb and -32P by isolated leaves of Elodea canadensis has been studied. The uptake of both these ions is strongly stimulated by light both in the presence and in the absence of CO2. In the case of phosphate, light stimulates uptake and inhibits the leakage of the ion from the tissues.

These results which confirm and extend the results of other Authors, support the conclusion that the stimulation by light of active uptake of solutes by green tissues is physiologically relevant. Furthermore, they suggest that the utilization of light energy for active uptake does not depend on CO2 fixation and assimilation.  相似文献

The effect of calcium and zinc ions on superoxide dismutase (SOD) from four plant species (Taxus baccata, Pinus sylvestris, Medicago rigidula, and Zea mays) was followed at three temperatures: optimal (20 °C), increased (50 °C), and high, inhibiting temperature (70 – 80 °C). At 20 and 50 °C in vitro added calcium increases SOD activity, but the degree was different for the plants investigated. The effect of zinc ions at the same temperatures varied in the investigated plants from activation to inhibition. An inhibiting effect of high temperature on SOD activity was diminished in the presence of calcium or zinc ions. It was shown that calcium and zinc ions can increase activity and thermostabilize different SOD isoforms.  相似文献

锌作为一种结构、催化和信号的成分,在许多生理过程中起着关键的作用。它也是病原微生物生长所必需的,不但参与病原微生物代谢和各种毒力因子的调控,而且是病原微生物在宿主中感染和定殖所必需的。铜绿假单胞菌侵染宿主发挥毒力时,宿主会采取营养免疫的策略来限制体内环境中游离的锌离子浓度而抑制该病原菌的感染和定殖。反过来,铜绿假单胞菌则通过自身的锌离子摄取系统克服宿主的营养免疫防御。本综述重点介绍了铜绿假单胞菌中已知的3种锌离子摄取系统(ZnuABC摄取系统、HmtA摄取系统和CntRLMN摄取系统)和锌摄取调控蛋白Zur,同时分析了其他潜在的锌离子摄取途径,并进一步阐述了铜绿假单胞菌锌离子摄取系统在其侵染宿主发挥毒力时和抵御宿主营养免疫时发挥的重要作用。系统总结铜绿假单胞菌对锌离子的摄取过程,旨在为靶向锌离子摄取系统的新型抗铜绿假单胞菌药物的开发提供指导。  相似文献

SUMMARY

This study was designed to investigate the apparent loss from the water column in Lake McIlwaine This study of significant quantities of reactive phosphorus. The Total Reactive Phosphorus mass-balance for the lake for 1977/78 was calculated, and both in situ and laboratory experiments were carried out to determine the effect of the lake sediments on the phosphorus loading of the lake. The experimental results showed that both uptake and release of phosphorus occurs in the lake, but that uptake of phosphorus by the sediments was by far the dominant process, thus accounting for the observed loss of phosphorus from the water column. The availability of the bound phosphorus for algal growth was also studied and it is suggested that algal uptake of bound phosphorus is possible. Various factors affecting phosphorus uptake by the sediments are discussed.  相似文献

A series of iron intercalated montmorillonites (Fe‐Monts) were prepared using (i) ion exchange of native sodium and calcium ions with iron ions, (ii) base hydrolysis of inserted iron ions in montmorillonite suspension, and (iii) insertion of pre‐hydrolyzed iron colloid in montmorillonite. The materials were characterized by X‐ray diffraction and gas adsorption‐desorption techniques. The basal d(001)‐spacing and BET specific surface area increased after the intercalation of iron species in montmorillonite. Local iron structure studied by X‐ray absorption fine structure (XAFS) spectroscopy showed an unsaturation of the Fe···Fe coordination number (N 2.5) of the intercalated iron species as compared to the bulk iron oxyhydroxides (N 6). The Fe‐Monts were employed for arsenic removal from aqueous solutions at low concentration (0.2–16 mg/L). Among the Fe‐Monts, the one prepared by the hydrolysis of inserted iron ions, was the best in performance. The saturation adsorption amount of the optimized iron‐montmorillonite was 4 and 28 times higher for the removal of arsenite and arsenate, respectively, as compared to bulk iron oxyhydroxide (goethite). Compared with bulk iron oxyhydroxide, the Fe‐Monts were superior for arsenate uptake and comparable for arsenite. In addition, arsenite adsorbed on the Fe‐Monts was found to be oxidized to arsenate based on XAFS spectroscopy.  相似文献

—The uptake of [3H]5HT, [3H]dopamine, [3H]noradrenaline and [3H]octopamine into the auricle of Helix pomatia was studied. When tissues were incubated at 25°C in media containing radioactive amines, tissue:medium ratios of about 49:1, 14:1 and 5:1 for 5-HT, dopamine, noradrenaline, and octopamine respectively were obtained after a 20–30 min incubation time. Tissues incubated at 25°C in media containing radioactive amines for 20–30 mins showed that almost all (96%) the radioactivity was present as unchanged [3H]5-HT, [3H]dopamine, [3H]octopamine or [3H]noradrenaline. The high tissue:medium ratios for 5-HT and dopamine, but not for noradrenaline and octopamine, showed saturation kinetics which were dependent upon temperature and sodium ions. From the Lineweaver–Burk plots, two uptake mechanisms for 5-HT at 25°C were resolved; the high affinity uptake process having a Km1 value of 6.0 ± 10?8m and a Vm1 value of 0.115 nmol/g/min while the lower affinity process had a Km2 value of 1.04 ± 10?6m and a Vm2 value of 0.66nmol/g/min. At 0°C a single uptake mechanism for 5-HT occurred which gave a Km value of 5.02 ± 10?8m and a Vm value of 0.0165 nmol/g/min. In the case of dopamine, the Lineweaver–Burk plot at 25°C showed a single uptake process with values for Km and Vm of 1.55 ± 10?7m and 0.086 nmol/g/min respectively. This process did not function at 0°C. The effect of various agents and ions upon the accumulation processes for all amines was also studied, and the data indicate that the same neurons probably accumulate more than one amine type. It is concluded that 5-HT and dopamine uptake in the auricle is a mechanism for inactivating these substances at 25°C and that an uptake mechanism for 5-HT also functions at 0°C. The results are discussed from the point of view of 5-HT's being the cardioexcitatory substance in the snail heart.  相似文献

The acrodermatitis enteropathica (AE) mutation affects intestinal zinc absorption. Our goal was to determine whether the AE mutation affects zinc uptake in human fibroblasts. Zinc uptake was determined during initial rates of uptake (10 min) following incubation in HEPES/saline buffer. Zinc uptake (from 0.25 to 1 μM) into normal fibroblasts was significantly greater than into the AE fibroblasts (p<0.05). In order to identify factors that may alter cellular zinc uptake and be affected by the AE mutation, zinc uptake in the presence of albumin or bicarbonate was measured. Albumin restricted zinc uptake in both normal and AE fibroblasts, whereas bicarbonate stimulated zinc uptake in the normal fibroblasts. The effect of bicarbonate on zinc uptake in the AE fibroblasts was significantly reduced in both the Pronase-sensitive and Pronase-resistant compartments. Following loading of the fibroblasts with 1 μM zinc for 60 min, zinc efflux and retention were measured. The AE mutation did not affect zinc retention compared to normal fibroblasts. We conclude that the AE mutation affects both zinc binding to the cell surface and its translocation across the plasma membrane into the cell, possibly mediated through a defective anionic exchange mechanism.  相似文献

Abstract

A high copper accumulation is induced in the yeast Saccharomyces cerevisiae either by menadione at the level of 200 μM in the growth medium, or by elevated concentrations of copper. While the uptake, as well as the toxicity of copper, strongly depend on the zinc concentration in the medium, there is no influence of zinc on copper intake induced by menadione. The copper binding ligand d-penicillamine suppresses the accumulation of copper in the menadione systen, whereas it has virtually no effect in the copper system. Within the range of non-toxic concentrations, copper is predominantly taken up by an energy-dependent mechanism. In contrast, the accumulation in the menadione system is clearly energy-independent. Thus there exist at least two different mechanisms for the uptake of copper in the yeast Saccharomyces cerevisiae.  相似文献

Abstract

We have determined solution structure of r(GGAGGUUUUGGAGG) (R14) by NMR; the RNA 14-mer forms an intra-strand parallel quadruplex with a G-tetrad and a hexad, in which a G-tetrad core is augmented by association of two A residues. The quadruplex further forms a dimer through stacking interaction between the hexads. In order to obtain insight into the difference between RNA and DNA quadruplexes, we synthesized the corresponding DNA 14-mer, d(GGAGGTTTTGGAGG) (D14), and examined its properties and structure by CD, gel electrophoresis, and NMR. K+ ions increased the thermal stability of both R14 and D14 structures. The binding affinity of K+ ions to R14 was much higher than that to D14. The CD and gel electrophoretic studies suggest that D14 forms a quadruplex entirely different from that of R14 in the presence of K+ ions; two molecules of D14 form a quadruplex with both antiparallel and parallel strand alignments and with diagonal loops at both ends of the stacked G-tetrads. The NMR study also gave results that are consistent with such structure: alternate glycosidic conformation, 5′G(syn)-G(anti)3′, and characteristic chemical shift data observed for many quadruplexes containing diagonal TTTT loops.  相似文献

Abstract– (1) The uptake and release of glutamic acid by guinea-pig cerebral cortex slices and rat synaptosomal fractions were studied, comparing the naturally occurring l - and non-natural d -isomers. Negligible metabolism of d -glutamic acid was observed in the slices. (2) Whereas in the cerebral slices the accumulation of glutamic acid was almost the same for the two isomers, d -glutamic acid was accumulated into the synaptosomal fraction at a markedly lower rate than was the L-isomer. (3) The uptake systems for d -isomer into the slices and synaptosomal fraction were found to be of single component, in contrast with the two component systems, high and low affinity components, for the uptake of l -glutamic acid. The apparent Km values for the uptake of d -glutamic acid into the slices and synaptosomal fraction were comparable with those reported for the low affinity components for l -isomer. The uptake systems for d -glutamic acid were dependent on the presence of Na+ ions in the medium, like those for l -glutamic acid and GABA. (4) The evoked release of radioactive preloaded d -glutamic acid was observed both from the slices and synaptosomal fraction following stimulation by high K+ ions in the medium. From these observations, it is evident that the evoked release of an amino acid by depolarization in vitro is not necessarily accompanied by a high affinity uptake process. (5) The uptake of l -glutamic acid, expecially into the synaptosomal fraction, was highly resistant to ouabain. On the other hand, the uptake rate of d -glutamic acid and GABA into the synaptosomal fraction was inhibited by varying concentrations of ouabain in accordance with the inhibition for brain Na-K ATPase. (6) The uptake of l -glutamic acid into subfractions of the P2 fraction was studied in relation to the distribution of the ‘synaptosomal marker enzymes’. An attempt to correlate the activities of enzymes of glutamic acid metabolism with the uptake of l -glutamic acid into the synaptosomal fraction from various parts of brain was unsuccessful. The high affinity uptake of l -glutamic acid was found to be very active in the synaptosomal fraction from any part of brain examined.  相似文献

Zinc toxicity on photosynthetic activity in cells of Synechocystis aquatilis f. aquatilis Sauvageau was investigated by monitoring Hill activity and fluorescence. The oxygen‐evolving activity decreased to about 80% of the initial value after exposure to 0.1 mM ZnSO4 for 1 h. The PSII activity was inhibited by 40% in the presence of zinc concentrations ranging from 0.5 to 5.0 mM, suggesting that the metal effect is limited by zinc uptake. The fluorescence capacity (Fmax–F/Fmax) decreased from 0.57 to 0.35 and 0.20 in Zn‐treated cells for 15 and 60 min, respectively, thus providing evidence for rapid inactivation of electron transport at PSII. Zinc treatment promoted a rapid increase in PSII fluorescence that was counteracted by addition of 1,4‐benzoquinone, indicating that electron transfer at the reducing side of the PSII reaction center is arrested by zinc. Furthermore, a decline in the fluorescence yield could be observed after 1 h of zinc treatment as well as when Zn‐treated cells were excited in presence of 3‐(3′,4′‐dichlorophenyl)‐1,1‐dimethylurea. Under these conditions, zinc did not affect energy transfer from phycobilisomes to PSII, and the gradual quenching of PSII fluorescence may be due to a decrease in electron flow on the donor side of PSII. However, the 20% increase in the minimal fluorescence intensity (Fo) in parallel to the absence of changes in the maximal fluorescence intensity (Fmax), observed in the first hour of zinc treatment, could also suggest a metal‐induced decline in the energy transfer from PSII‐chl a antenna to the PSII reaction center.  相似文献

—A rapid accumulation of [3H]GABA occurs in slices of rat cerebral cortex incubated at 25° or 37° in a medium containing [3H]GABA. Tissue medium ratios of almost 100:1 are attained after a 60 min incubation at 25°. At the same temperature no labelled metabolites of GABA were found in the tissue or the medium. The process responsible for [3H]GABA uptake has many of the properties of an active transport mechanism: it is temperature sensitive, requires the presence of sodium ions in the external medium, is inhibited by dinitrophenol and ouabain, and shows saturation kinetics. The estimated Km value for GABA is 2·2 × 10?5m , and Vmax is 0·115 μmoles/min/g cortex. There is only negligible efflux of the accumulated [3H]GABA when cortical slices are exposed to a GABA-free medium. [3H]GABA uptake was not affected by the presence of large molar excesses of glycine, l -glutamic acid, l -aspartic acid, or β-aminobutyrate, but was inhibited in the presence of l -alanine, l -histidine, β-hydroxy-GABA and β-guanidinopropionate. It is suggested that the GABA uptake system may represent a possible mechanism for the inactivation of GABA or some related substance at inhibitory synapses in the cortex.  相似文献

The COT1 and ZRC1 genes of Saccharomyces cerevisiae are structurally related dosage-dependent suppressors of metal toxicity. COT1 confers increased tolerance to high levels of cobalt; ZRC1 confers increased tolerance to high levels of zinc. The two genes are not linked and have been mapped; COT1 to chromosome XV and ZRC1 to chromosome XIII. Phenotypes related to metal homeostasis have been examined in strains with varied COT1 and ZRC1 gene doses. Overexpression of COT1 confers tolerance to moderately toxic levels of zinc and ZRC1 confers tolerance to moderately toxic levels of cobalt. Strains that carry null alleles at both loci are viable. The metal-hypersensitive phenotypes of mutations in either gene are largely unaffected by changes in dosage of the other. COT1 and ZRCI function independently in conferring tolerance to their respective metals, yet the uptake of cobalt ions by yeast cells is dependent on the gene dosage of ZRC1 as well as of COT1 Strains that overexpress ZRC1 have increased uptake of cobalt ions, while ZRCI null mutants exhibit decreased cobalt uptake. The defects in cobalt uptake due to mutations at COT1 and ZRC1 are additive, suggesting that the two genes are responsible for the majority of cobalt and zinc uptake in yeast cells. The function of either gene product seems to be more important in metal homeostasis than is the GRR1 gene product, which is also involved in metal metabolism. Mutations in the GRR1 gene have no effect on the cobalt-related phenotypes of strains that have altered gene dosage of either COT1 or ZRC1.  相似文献

Effects of various ionic conditions on the development of the cellular slime molds D. discoideum and D. mucoroides were studied. A certain concentration of lithium ions (7 mM) promoted differentiation of the stalk cells and conversely inhibited formation of the spores. The presence of calcium and magnesium ions was needed for Li to manifest its specific effect. A high concentration of Ca (100-120 mM) also facilitated differentiation of the stalk cells. On the other hand, fluoride ions stimulated considerable formation of spores at 15 mM. In the absence of divalent cations, sodium ions inhibited morphogenesis and cell differentiation proportionately with its concentration, and complete inhibition was obtained at 20 mM. The inhibitory effect of Na was nullified by addition of small amounts of Ca. Possible mechanisms by which these ions exert their influences on development of this organism were discussed.  相似文献

The mechanism(s) by which zinc is transported into cells has not been identified. Since zinc uptake is inhibited by reducing the temperature, zinc uptake may depend on the movement of plasma membrane micoenvironments, such as endocytosis or potocytosis. We investigated the potential role of potocytosis in cellular zinc uptake by incubating normal and acrodermatitis enteropathica fibroblasts with nystatin, a sterol-binding drug previously shown to inhibit potocytosis. Zinc uptake was determined during initial rates of uptake (10 min) following incubation of the fibroblasts in 50 μg nystatin/mL or 0.1% dimethyl-sulfoxide for 10 min at 37°C. The cells were then incubated with 1 to 30 μM 65zinc. Michaelis-Menten kinetics were observed for zinc uptake. Nystatin inhibited zinc uptake in both the normal and AE fibroblasts. Reduced cellular uptake of zinc was associated with its internalization, not its external binding. In normal fibroblasts, nystatin significantly reduced theK m 56% and theV max 69%. In the AE fibroblasts, nystatin treatment significantly reduced theV max 59%, but did not significantly affect theK m. The AE mutation alone affected theV max for cellular zinc uptake. The control AE fibroblasts exhibited a 40% reduction inV max compared to control normal fibroblasts. We conclude that nystatin exerts its effect on zinc uptake by reducing the velocity at which zinc traverses the cell membrane, possibly through potocytosis. Furthermore, the AE mutation also effects zinc transport by reducing zinc transport.  相似文献
