Summary Genetic studies on radiation-induced chlorina and variegated mutants of okra (Abelmoschus esculentus (L.) Moench) revealed the existence of an unstable gene. The normal green color of the leaves is controlled by duplicate genes C1 and C2, either of which produces the green colour. The chlorina plants are C 1 C 1 C 2 C 2. The allele c 1 v is dominant to both C 1 and C 2 but is unstable. The homozygote c 1 v c 1 v c 2 c 2 is a normal green while the heterozygote c i v c 1 c 2 c 2 has a variegated phenotype as a result of the mutation of c 1 v to c 1 during development. In green plants with a c 1 v c{sh1/v}c 2 c 2 genotype, the autonomous mutation of one of the c 1 v alleles to c 1 may take place at the pre-meiotic stage. In the variegated genotype (c 1 v c 1 c 2 c 2), the mutation of c 1 to c 1 v may take place in early ontogeny, thus producing green plants. The allele C 1, when associated with c 1 v in a heterozygous condition, mutates to c 1 at the pre-meiotic stage even in the presence of the allele C 2.  相似文献

以沙质草地优势物种猪毛蒿、胡枝子和糙隐子草为研究对象,利用开顶式生长室(OTC)模拟增温,研究降水减少20%、40%和60%与增温的交互作用对3种典型植物叶片光合气体交换特征及叶绿素荧光特征的影响,以揭示沙质草地3种优势植物对气候变化的响应规律。结果显示:(1)与自然温度相比,OTC模拟增温增加了猪毛蒿C_(i),显著降低了胡枝子G_(s)、P_(n)和T_(r)、糙隐子草G_(s)和P_(n)、猪毛蒿WUE和L_(s),也显著降低了猪毛蒿和胡枝子F_(v)/F_(m)和F_(v)/F_(o)。(2)无论增温与否,随着降水减少幅度的增加,猪毛蒿G_(s)和P_(n)呈下降趋势,且中度以上的干旱胁迫下(降水减少>40%)胡枝子和糙隐子草P_(n)显著低于对照。(3)在自然温度条件下,轻度干旱胁迫时(降水减少20%)猪毛蒿T_(r)显著低于对照,重度干旱胁迫时(降水减少60%)其WUE、F_(v)/F_(m)和F_(v)/F_(o)显著低于对照;重度干旱胁迫时,胡枝子C_(i)显著高于对照,差异幅度达10.7%,L_(s)显著低于对照,轻度干旱胁迫时(降水减少20%)其F_(v)/F_(m)和F_(v)/F_(o)显著高于中度以上的干旱胁迫;中度以上的干旱胁迫下糙隐子草T_(r)和G_(s)显著低于对照,重度干旱胁迫时,其C_(i)、F_(v)/F_(m)和F_(v)/F_(o)显著低于对照,WUE和L_(s)显著高于对照。(4)增温与降水减少交互作用下,所有处理猪毛蒿C_(i)均高于对照,差异幅度分别达4.5%,6.0%和8.4%;胡枝子T_(r)均显著低于对照,差异幅度达57.8%;重度干旱胁迫时猪毛蒿L_(s)和WUE显著低于对照,糙隐子草F_(v)/F_(m)和F_(v)/F_(o)随降水减少而降低,中度以上的干旱胁迫时其值显著低于对照。(5)相关性分析表明,3个优势物种的P_(n)与F_(v)/F_(m)和F_(v)/F_(o)均呈显著正相关关系,其中猪毛蒿和糙隐子草的P_(n)—F_(v)/F_(m)和P_(n)—F_(v)/F_(o)斜率明显高于胡枝子。研究表明,气候变暖会在一定程度上加剧降水减少对沙质草地3种群落优势物种光合作用的抑制。  相似文献

The parameters estimated from traditional A/C i curve analysis are dependent upon some underlying assumptions that substomatal CO2 concentration (C i) equals the chloroplast CO2 concentration (C c) and the C i value at which the A/C i curve switches between Rubisco- and electron transport-limited portions of the curve (C i-t) is set to a constant. However, the assumptions reduced the accuracy of parameter estimation significantly without taking the influence of C i-t value and mesophyll conductance (g m) on parameters into account. Based on the analysis of Larix gmelinii’s A/C i curves, it showed the C i-t value varied significantly, ranging from 24 Pa to 72 Pa and averaging 38 Pa. t-test demonstrated there were significant differences in parameters respectively estimated from A/C i and A/C c curve analysis (p<0.01). Compared with the maximum ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) carboxylation rate (Vcmax), the maximum electron transport rate (Jmax) and Jmax/Vcmax estimated from A/C c curve analysis which considers the effects of g m limit and simultaneously fits parameters with the whole A/C c curve, mean Vcmax estimated from A/C i curve analysis (Vcmax-C i) was underestimated by 37.49%; mean Jmax estimated from A/C i curve analysis (Jmax-C i) was overestimated by 17.8% and (Jmax-C i)/(Vcmax-C i) was overestimated by 24.2%. However, there was a significant linear relationship between Vcmax estimated from A/C i curve analysis and Vcmax estimated from A/C c curve analysis, so was it Jmax (p<0.05).  相似文献

Genetics of tolerance to iron chlorosis was investigated in eight crosses involving parents distinctly different in their level of tolerance. The segregating populations with parents and F1s were screened under actual stress conditions in the field. Also, selected crosses were studied for Fe3+ uptake capacity. Tolerance/moderate tolerance to Fe chlorosis was dominant over susceptibility and it was controlled by two sets of nonallelic genes with complementary interaction. Gene Ic 1 has been found to be basic and in complementation with Ic 3 it confers tolerance. Likewise, Ic 2 with Ic 4 confers tolerance. The basic genes Ic 1 and Ic 2 are nonallelic and, in the absence of their respective complementary genes Ic 3 and 4 , ineffective, which results in susceptibility. Of tolerant cultivars, ARC 10372 and Cauvery have been tentatively assigned the genotype of Ic 1 , Ic 2 , Ic 3 , Ic 4 , and moderately tolerant IET 7613, Prasanna and Akashi Ic 1 , 2 Ic 3 Ic 4 . The susceptible ARC 5723 has been assigned Ic 1 , 2 , Ic 3 , Ic 4 , and IET 9829, Ic 1 , 2 Ic 3 Ic 4 . IET 7614 is susceptible, due to the presence of inhibitory genes I-Ic 1 , I-Ic 2 together with ic 1 pt>, ic 2 , Ic 3 , Ic 4 . Further, the gene Pc for purple coleoptile shows linkage with one of the complementary genes with a crossover value of 15.26%, while the gene(s) for seedling height Ts with Ic 1 with a crossover value of 1.7%. It is possible that the gene(s) for iron chlorosis tolerance might belong to the second linkage group, where genes for purple leaf were located.  相似文献

Summary The hybrid produced between a Carbondale haploid strain (-methyl-glucoside rapid fermenter) and a haploid strain (non-fermenter), derived from a hybrid between a homothallic and a heterothallicSaccharomyces, showed an irregular segregation pattern with regard to the fermentation of this sugar.To explain this irregularity, three pairs of alleles,MG 1/mg 1,MG 2/mg 2 andMG 3/mg 3, were assumed to be in quantitative control of the fermetation. Haploid cultures carrying the genotypes (1)mg 1 mg 2 mg 3, (2)MG 1 mg 2 mg 3, (3)mg 1 MG 2 mg 3, (4)mg 1 mg 2 MG 3, (5)MG 1 MG 2 mg 3, (6)MG 1 mg 2 MG 3, (7)mg 1 MG 2 MG 3, and (8)MG 1 MG 2 MG 3, were actually recovered. Strains equipped with: either (1) or (2); either (4) or (6); (3); (5); (7); or (8) are non-fermenters, extremely-slow-fermenters, slow-fermenters, medium-fermenters, semi-rapid-fermenters and rapid-fermenters respectively.The role of these genes in sugar fermentation and the identity or nonidentity of some of these genes with maltose and sucrose genes was discussed.With 2 Figures in the Text  相似文献

This paper explores the consequences of the theoretical forward activation enzymatic pathway A 0 A 1 A 2 A 3 where E 1 convents A 0 to A 1, E 2 converts A 1 to A 2 and E 3 converts A 2 to A 3. A 0, which is environmentally determined, also serves to activate (or modulate) the activity of E 3 in such a way as to keep the concentration of A 2 ([A 2]) constant at a particular set-point value. For mathematical simplicity, first order rate kinetics are used where k 1, k 2 and k 3 are the rate constants for E 1, E 2, and E 3 respectively. It is shown that if k 3 is modulated appropriately so as keep [A 2] at the setpoint value with a changing upstream [A 0], then the modulation of k 3 must be anticipatory of the dynamics of the biochemical pathway. In other words, the rate of change of k 3, will be a function of [A 0], k 1, k 2, k 3, and the set-point value of [A 2]. If the modulation of k 3 does not perfectly model (anticipate) the reaction pathway of which it is a part, then the actual [A 2] will deviate from the set-point value. This type of anticipatory feed-forward activation may represent an important aspect of biological organization.  相似文献

Estimating effective population size (Ne) using linkage disequilibrium (LD) information (Ne(LD)) has the operational advantage of using a single sample. However, Ne(LD) estimates assume discrete generations and its performance are constrained by demographic issues. However, such concerns have received little empirical attention so far. The pedigree of the endangered Gochu Asturcelta pig breed includes individuals classified into discrete filial generations and individuals with generations overlap. Up to 780 individuals were typed with a set of 17 microsatellites. Performance of Ne(LD) was compared with Ne estimates obtained using genealogical information, molecular coancestry (Ne(M)) and a temporal (two‐sample) method (Ne(JR)). Molecular‐based estimates of Ne exceeded those obtained using pedigree data. Estimates of Ne(LD) for filial generations F3 and F4 (17.0 and 17.3, respectively) were lower and steadier than those obtained using yearly or biannual samplings. Ne(LD) estimated for samples including generations overlap could only be compared with those obtained for the discrete filial generations when sampling span approached a generation interval and demographic correction for bias was applied. Single‐sample Ne(M) estimates were lower than their Ne(LD) counterparts. Ne(M) estimates are likely to partially reflect the number of founders rather than population size. In any case, estimates of LD and molecular coancestry tend to covary and, therefore, Ne(M) and Ne(LD) can hardly be considered independent. Demographically adjusted estimates of Ne(JR) and Ne(LD) took comparable values when: (1) the two samples used for the former were separated by one equivalent to discrete generations in the pedigree and (2) sampling span used for the latter approached a generation interval. Overall, the empirical evidence given in this study suggested that the advantage of using single‐sample methods to obtain molecular‐based estimates of Ne is not clear in operational terms. Estimates of Ne obtained using methods based in molecular information should be interpreted with caution.  相似文献

Gas-residence time distribution (RTD) response curves measured in a 23 m high pilot plant airlift tower loop reactor, which consisted of a riser, a special downcomer construction and at the top of the riser a large head. C3 photosynthesis is often limited by CO2 diffusivity or stomatal (gs) and mesophyll (gm) conductances. To characterize effects of stomatal closure induced by either high CO2 or abscisic acid (ABA) application on gm, we examined gs and gm in the wild type (Col‐0) and ost1 and slac1‐2 mutants of Arabidopsis thaliana grown at 390 or 780 μmol mol?1 CO2. Stomata of these mutants were reported to be insensitive to both high CO2 and ABA. When the ambient CO2 increased instantaneously, gm decreased in all these plants, whereas gs in ost1 and slac1‐2 was unchanged. Therefore, the decrease in gm in response to high CO2 occurred irrespective of the responses of gs. gm was mainly determined by the instantaneous CO2 concentration during the measurement and not markedly by the CO2 concentration during the growth. Exogenous application of ABA to Col‐0 caused the decrease in the intercellular CO2 concentration (Ci). With the decrease in Ci, gm did not increase but decreased, indicating that the response of gm to CO2 and that to ABA are differently regulated and that ABA content in the leaves plays an important role in the regulation of gm.  相似文献

Abstract. The behaviour of five statistics (extensions of Pielou's, Clark and Evansapos;, Pollard's, Johnson & Zimmer's, and Eberhardt's statistics, which are denoted as Pi, Ce, Po, Jz and Eb respectively) that involve the distance from a random point to its jth nearest neighbour were examined against several alternative patterns (lattice‐based regular, inhomogeneous random, and Poisson cluster patterns) through Monte Carlo simulation to test their powers to detect patterns. The powers of all the five statistics increase as distance order j increases against inhomogeneous random pattern. They decrease for Pi and Ce and increase for Po, Jz, and Eb against regular and Poisson cluster patterns. Po, Jz, and Eb can reach high powers with the third or higher order distances in most cases. However, Po is recommended because no extra information is needed, it can reach a high power with the second or third distance even though the sample size is not large in most cases, and the test can be performed with an approximate χ2 distribution associated with it. When a regular pattern is expected, Jz is recommended because it is more sensitive to lattice‐based regular pattern than Po and Eb, especially for the first distance. However, simulation tests should be used because the speed of convergence of Jz to normal distribution is very slow.  相似文献

A review of the literature revealed that a variety of methods are currently used for fitting net assimilation of CO2–chloroplastic CO2 concentration (A–Cc) curves, resulting in considerable differences in estimating the A–Cc parameters [including maximum ribulose 1·5‐bisphosphate carboxylase/oxygenase (Rubisco) carboxylation rate (Vcmax), potential light saturated electron transport rate (Jmax), leaf dark respiration in the light (Rd), mesophyll conductance (gm) and triose‐phosphate utilization (TPU)]. In this paper, we examined the impacts of fitting methods on the estimations of Vcmax, Jmax, TPU, Rd and gm using grid search and non‐linear fitting techniques. Our results suggested that the fitting methods significantly affected the predictions of Rubisco‐limited (Ac), ribulose 1,5‐bisphosphate‐limited (Aj) and TPU‐limited (Ap) curves and leaf photosynthesis velocities because of the inconsistent estimate of Vcmax, Jmax, TPU, Rd and gm, but they barely influenced the Jmax : Vcmax, Vcmax : Rd and Jmax : TPU ratio. In terms of fitting accuracy, simplicity of fitting procedures and sample size requirement, we recommend to combine grid search and non‐linear techniques to directly and simultaneously fit Vcmax, Jmax, TPU, Rd and gm with the whole A–Cc curve in contrast to the conventional method, which fits Vcmax, Rd or gm first and then solves for Vcmax, Jmax and/or TPU with Vcmax, Rd and/or gm held as constants.  相似文献

Leaf CO2 uptake (A) in C4 photosynthesis is limited by the maximum apparent rate of PEPc carboxylation (Vpmax) at low intercellular [CO2] (ci) with a sharp transition to a ci-saturated rate (Vmax) due to co-limitation by ribulose-1:5-bisphosphate carboxylase/oxygenase (Rubisco) and regeneration of PEP. The response of A to ci has been widely used to determine these two parameters. Vmax and Vpmax depend on different enzymes but draw on a shared pool of leaf resources, such that resource distribution is optimized, and A maximized, when Vmax and Vpmax are co-limiting. We collected published A/ci curves in 49 C4 species and assessed variation in photosynthetic traits between phylogenetic groups, and as a function of atmospheric [CO2]. The balance of Vmax-Vpmax varied among evolutionary lineages and C4 subtypes. Operating A was strongly Vmax-limited, such that re-allocation of resources from Vpmax towards Vmax was predicted to improve A by 12% in C4 crops. This would not require additional inputs but rather altered partitioning of existing leaf nutrients, resulting in increased water and nutrient-use efficiency. Optimal partitioning was achieved only in plants grown at pre-industrial atmospheric [CO2], suggesting C4 crops have not adjusted to the rapid increase in atmospheric [CO2] of the past few decades.  相似文献

The growth and energy budget for F2‘all‐fish’ growth hormone gene transgenic common carp Cyprinus carpio of two body sizes were investigated at 29·2° C for 21 days. Specific growth rate, feed intake, feed efficiency, digestibility coefficients of dry matter and protein, gross energy intake (IE), and the proportion of IE utilized for heat production (HE) were significantly higher in the transgenics than in the controls. The proportion of IE directed to waste products [faecal energy (FE) and excretory energy loss (ZE+UE) where ZE is through the gills and UE through the kidney], and the proportion of metabolizable energy (ME) for recovered energy (RE) were significantly lower in the transgenics than in the controls. The average energy budget equation of transgenic fish was as follows: 100 IE= 19·3 FE+ 6·0 (ZE+UE) + 45·2 HE+ 29·5 RE or 100 ME= 60·5 HE+ 39·5 RE. The average energy budget equation of the controls was: 100 IE= 25·2 FE+ 7·4 (ZE+UE) + 35·5 HE+ 31·9 RE or 100 ME= 52·7 HE+ 47·3 RE. These findings indicate that the high growth rate of ‘all‐fish’ transgenic common carp relative to their non‐transgenic counterparts was due to their increased feed intake, reduced lose of waste productions and improved feed efficiency. The benefit of the increased energy intake by transgenic fish, however, was diminished by their increased metabolism.  相似文献

Self-incompatibility has been studied extensively at the molecular level in Solanaceae, Rosaceae, and Scrophulariaceae, all of which exhibit gametophytic self-incompatibility. In the present study, we successfully isolated nine S-RNase alleles from cultivars of Chinese cherry by PCR amplification from genomic DNA and stylar cDNA combining with cleaved amplified polymorphic sequence marker. Analysis of amino acid sequences revealed five novel S-alleles, S 2 , S 4 , S 6 , S 8 , and S 9 , with respective accession numbers in the NCBI database of EF541168, EF541173, EF541172, FJ628598, and FJ628599. Results showed that “Dongtang” and “Yinzhu” contained six S-alleles (S 1 , S 3 , S 5 , S 7 , S 8 , and S 9 ); “Taishanganying” contained four S-alleles (S 1 , S 2 , S 4 , and S 6 ); “Daiba”, “Dayingzui”, and “Xiaomizi” contained four S-alleles (S 1 , S 2 , S 5 , and S 8 ); “Laiyangduanzhi”, “Shuangquanchangba”, and “Daqingye” contained three S-alleles (S 1 , S 2 , and S 8 ). It is interesting that different cultivars collected from the same place hold the same S-genotypes. Moreover, pollination tests and pollen tube growth assays showed that nine cultivars were self-compatible. Chinese cherry presented in this article are naturally polyploidy, which is a very useful material for the study of self-compatibility, and much of this information will be valuable for further work on self-(in)compatibility of fruit tree in Rosaceae.  相似文献

The effect of NH4Cl addition on intracellular pH (pH i ) was determined by flow cytometric measurements of the fluorescence of a pH-sensitive dye. The effects of NH4Cl on growth were determined for batch growth of cells in flasks in an incubator. The addition of NH4Cl caused a cytoplasmic acidification. A new lower steady-state value of pH i was attained within 20–40 min of NH4Cl addition. A correlation was found between the effects of NH4Cl on growth and on pH i : whereas 3 mM NH4Cl had little effect on growth and on pH i , 10 mM NH4Cl caused a substantial growth inhibition and a pH i decrease of 0.2–0.3 units. The effects of NH4Cl on growth and on pH i were found to be independent of the external pH value (pH e over the range 6.8 to 7.6, except that 10 mM NH4Cl was more toxic at pH e 7.6. The addition of NH4Cl caused an increase in the average cell volume at pH e 7.6, but had no effect on the average cell volume at pH e 's 6.8 and 7.2. For comparison, the effects of pH e alone on growth and on pH i were determined. There was little difference in cell growth at pH e 's 6.8, 7.2 and 7.6. At pH e 6.6, there was a substantial growth inhibition. Some measurements of the effects of pH e on pH i were made, although the steady-state value of pH i as a function of pH e was not determined due to limitations in the pH i -measuring technique. These measurements showed that pH i remained constant from pH e 7.6 to 6.8, but fell by 0.2 units at pH e 6.6, in agreement with the growth results.  相似文献

We previously identified both self-incompatible and self-compatible plants in a natural population of self-incompatible Petunia axillaris subsp. axillaris, and found that all the self-compatible plants studied carried either SC1- or SC2-haplotype. Genetic crosses showed that SC2 was identical to S17 identified from another natural population of P. axillaris, except that its pollen function was defective, and that the pollen-part mutation in SC2 was tightly linked to the S-locus. Recent identification of the S-locus F-box gene (SLF) as the gene that controls pollen specificity in S-RNase-based self-incompatibility has prompted us to examine the molecular basis of this pollen-part mutation. We cloned and sequenced the S17-allele of SLF of P.axillaris, named PaSLF17, and found that SC2 SC2 plants contained extra restriction fragments that hybridized to PaSLF17 in addition to all of those observed in S17 S17 plants. Moreover, these additional fragments co-segregated with SC2. We used the SC2-specific restriction fragments as templates to clone an allele of PaSLF by PCR. To determine the identity of this allele, named PaSLFx, primers based on its sequence were used to amplify PaSLFalleles from genomic DNA of 40 S-homozygotes of P. axillaris, S1 S1 through S40 S40. Sequence comparison revealed that PaSLFx was completely identical with PaSLF19 obtained from S19 S19. We conclude that the S-locus of SC2 contained both S17-allele and the duplicated S19-allele of PaSLF. SC2 is the first naturally occurring pollen-part mutation of a solanaceous species that was shown to be associated with duplication of the pollen S. This finding lends support to the proposal, based on studies of irradiation-generated pollen-part mutants of solanaceous species, that duplication, but not deletion, of the pollen S, causes breakdown of pollen function.  相似文献

Cross-compatibility relationships in almond are controlled by a gametophytically expressed incompatibility system partly mediated by stylar RNases, of which 29 have been reported. To resolve possible synonyms and to provide data for phylogenetic analysis, 21 almond S-RNase alleles were cloned and sequenced from SP (signal peptide region) or C1 (first conserved region) to C5, except for the S 29 allele, which could be cloned only from SP to C1. Nineteen sequences (S 4 , S 6 , S 11 S 22 , S 25 S 29 ) were potentially new whereas S 10 and S 24 had previously been published but with different labels. The sequences for S 16 and S 17 were identical to that for S 1 , published previously; likewise, S 15 was identical to S 5 . In addition, S 4 and S 20 were identical, as were S 13 and S 19 . A revised version of the standard table of almond incompatibility genotypes is presented. Several alleles had AT or GA tandem repeats in their introns. Sequences of the 23 distinct newly cloned or already published alleles were aligned. Sliding windows analysis of Ka/Ks identified regions where positive selection may operate; in contrast to the Maloideae, most of the region from the beginning of C3 to the beginning of RC4 appeared not to be under positive selection. Phylogenetic analysis indicated four pairs of alleles had ‘bootstrap’ support > 80%: S 5 /S 10 , S 4 /S 8, S 11 /S 24 , and S 3 /S 6 . Various motifs up to 19 residues long occurred in at least two alleles, and their distributions were consistent with intragenic recombination, as were separate phylogenetic analyses of the 5′ and 3′ sections. 目的:检测老年住院患者分离的鲍曼不动杆菌的主要耐药基因,并研究不同耐药基因型与耐药表型之间的对应关系。方法:用PCR方法检测分离自老年住院患者的不同标本来源的170例非重复鲍曼不动杆菌的耐药基因。检测的耐药基因包括D类碳青霉烯酶:bla_(OXA-51),bla_(OXA-23),bla_(OXA-24),bla_(OXA-58),B类金属碳青霉烯酶:bla_(VIM),bla_(IMP),bla_(SIM),bla_(GIM),bla_(DIM),bla_(NDM-1),以及A类超广谱β-内酰胺酶:blaKPC,共计11种。根据检测结果对菌株进行基因分型,并研究不同基因型与CRAB和CSAB这两种耐药表型之间的对应关系。结果:170株鲍曼不动杆菌的固有基因bla_(OXA-51)均为阳性,此外,主要检出基因为bla_(OXA-23),共124株。另外检测出blaKPC12株,bla OXA-58 6株,bla_(NDM-1)3株,bla_(SIM)2株,bla_(OXA-24)、bla_(VIM)和bla_(DIM)各1株,IMP和GIM未检出。根据检出耐药基因的不同组合,分为bla OXA-51+bla_(OXA-23)阳性为基础的A型(124株)及bla_(OXA-23)阴性为基础的B型(bla_(OXA-51),39株)、C型(bla_(OXA-51)+bla_(OXA-58),6株)、D型(bla_(OXA-51)+bla_(OXA-24),1株)共计四类基因型。从耐药表型来看,128株碳青霉烯耐药菌中有122株bla_(OXA-23)为阳性,在CRAB中占95.3%(122/128),42株碳青霉烯敏感株中,有40株bla_(OXA-23)为阴性,在CSAB中占95.2%(40/42)。结论:老年病房流行的耐碳青霉烯鲍曼不动杆菌的耐药基因型以bla_(OXA-23)阳性为主。其与鲍曼不动杆菌CRAB耐药表型、bla_(OXA-23)阴性与CSAB耐药表型之间有良好的对应关系。  相似文献

Using promoters expressed in nonendosperm tissues to activate target genes in specific plant tissues or organs with very limited expression in the endosperm is an attractive approach in crop transgenic engineering. In this article, five putative nonendosperm tissue‐expressed promoters were cloned from the rice genome and designated POsNETE1, POsNETE2, POsNETE3, POsNETE4 and POsNETE5. By qualitatively and quantitatively examining GUSplus reporter gene expression in transgenic rice plants, POsNETE1‐POsNETE5 were all found to be active in the roots, leaves, stems, sheaths and panicles but not in the endosperm of plants at different developmental stages. In addition, POsNETE2, POsNETE4 and POsNETE5 were also inactive in rice embryos. Among these promoters, POsNETE4 and POsNETE5 exhibited higher activities in all of the tested tissues, and their activities in stems, leaves, roots and sheaths were higher than or comparable to those of the rice Actin1 promoter. We also progressively monitored the activities of POsNETE1‐POsNETE5 in two generations of single‐copy lines and found that these promoters were stably expressed between generations. Transgenic rice was produced using POsNETE4 and POsNETE5 to drive a modified Bt gene, mCry1Ab. Bt protein expressed in the tested plants ranged from 1769.4 to 4428.8 ng/g fresh leaves, whereas Bt protein was barely detected in the endosperm. Overall, our study identified five novel nonendosperm tissue‐expressed promoters that might be suitable for rice genetic engineering and might reduce potential social concern regarding the safety of GMO crops.  相似文献
