Variants of serum transferrin in Japanese wild pig, Sus scrofa leucomystax, and Ryukyu wild pig, S.s. riukiuanus, of Japan were investigated by using starch gel electrophoresis. Five phenotypes, TfB, BC, C, CX and X, were observed, of which two, TfCX and X, are new variants. Comparison of gene frequency estimates which were calculated for each population showed remarkable geographic differences among several populations of these two subspecies.  相似文献

The polymorphism of five enzyme loci (amylase, alkaline phosphatase, albumin, for 4-week body weight was compared to that of the unselected control line (C). for 4 week body weight was compared to that of the unselected control line (C). Three loci in the C line and two in the P line demonstrated polymorphism. Plasma amylase was separated into six bands and zymograms were classified on the basis of these bands into nine phenotypes. Three of the nine types were of relatively high activity and six were of relatively low activity. All nine types were found in the C line, whereas, all birds of the P line had only the most active type. Two alkaline phosphatase alleles (Akp-2B and Akp-2C) were segregating in the C line. Gene frequencies of alkaline phosphatase for the Akp-2B allele were 0.92 in the C line and 1.00 in the P line. Two albumin alleles (AlbQ1 and AlbQ2) were segregating in both populations. Gene frequencies for the AlbQ1 allele were 0.74 in the C line and 0.81 in the P line. Two red cell esterase-D alleles (Es-DF and Es-Ds) were segregating in both populations. The gene frequency for the Es-Ds allele (0.61) was higher than that of the Es-DF allele in the C line. In the P line the frequency of the Es-DF allele was higher than that of the Es-Ds allele. Heterozygosities of the C and P lines were estimated as 0.2258 and 0.1560 respectively. The relative inbreeding coefficient of the P line, calculated from heterozygosities was 0.31.  相似文献

An unusual variant, Tf Aw, has been found in sheep transferrins. Its position in starch gel electrophoresis is identical with the variant Tf A, but the intensity of corresponding bands is substantially lower. Family analyses prove that the variant Aw is genetically controlled and represents either the product of an unusual allele Tf Aw or the interaction between the allele Tf A and a hypothetical modifying locus.  相似文献

Summary. To provide information on the extent to which domestication has altered the natural levels of genetic variability of the wild quail, electrophoretic variability at 32 loci was estimated in a quail population established by capturing wild quail and keeping them for 15 years in a domestic environment without any artificial selection. The gene frequencies of the domesticated quail population were very similar to those of the commercial and laboratory quail populations and different from those of the wild quail populations previously reported by Kimura et al . A linkage map of the Japanese quail (Coturnix japonica) genome was constructed based upon segregation analysis of 72 microsatellite loci in 433 F(2) progeny of 10 half-sib families obtained from a cross between two quail lines of different genetic origins. One line was selected for long duration of tonic immobility, a behavioural trait related to fearfulness, while the other was selected based on early egg production. Fifty-eight of the markers were resolved into 12 autosomal linkage groups and a Z chromosome-specific linkage group, while the remaining 14 markers were unlinked. The linkage groups range from 8 cM (two markers) to 206 cM (16 markers) and cover a total map distance of 576 cM with an average spacing of 10 cM between loci. Through comparative mapping with chicken (Gallus gallus) using orthologous markers, we were able to assign linkage groups CJA01, CJA02, CJA05, CJA06, CJA14 and CJA27 to chromosomes. This map, which is the first in quail based solely on microsatellites, is a major step towards the development of a quality molecular genetic map for this valuable species. It will provide an important framework for further genetic mapping and the identification of quantitative trait loci controlling egg production and fear-related behavioural traits in quail.  相似文献

The quail is a valuable farm and laboratory animal. Yet molecular information about this species remains scarce. We present here the first genetic linkage map of the Japanese quail. This comprehensive map is based solely on amplified fragment length polymorphism (AFLP) markers. These markers were developed and genotyped in an F2 progeny from a cross between two lines of quail differing in stress reactivity. A total of 432 polymorphic AFLP markers were detected with 24 TaqI/EcoRI primer combinations. On average, 18 markers were produced per primer combination. Two hundred and fifty eight of the polymorphic markers were assigned to 39 autosomal linkage groups plus the ZW sex chromosome linkage groups. The linkage groups range from 2 to 28 markers and from 0.0 to 195.5 cM. The AFLP map covers a total length of 1516 cM, with an average genetic distance between two consecutive markers of 7.6 cM. This AFLP map can be enriched with other marker types, especially mapped chicken genes that will enable to link the maps of both species and make use of the powerful comparative mapping approach. This AFLP map of the Japanese quail already provides an efficient tool for quantitative trait loci (QTL) mapping.  相似文献

As soon as they hatch, gallinaceous chicks follow broody hens. This matriarchal unit presents a temporal organization of activity. The ontogeny of this ultradian rhythm of activity was followed in Japanese quail during their first 3 weeks of life. Under controlled laboratory conditions, 12 groups of four chicks were recorded using an activity monitoring system. They were observed between the ages of 2 and 17 days. Chicks in groups presented an ultradian rhythm of activity, with a period that increased significantly from 14.3 ± 1.4 minutes when chicks were 2 days old to 26.0 ± 1.9 minutes when they were 16 days old. The increase of ultradian periodicity was particularly pronounced during their first and third weeks of life. Finally, the ultradian period was correlated positively with body weight of the chicks. In the research fields of experimental embryology, teratological testing, and developmental engineering in avian species, a knowledge of normal embryonic development is necessary so that research may be performed efficiently and precisely. A series of normal stages based on external appearance has been established in both chicken and quail embryos. Those based on skeletal features, however, have not been elucidated. The present study newly established a series of normal stages for the development of the Japanese quail embryo skeleton. This series is composed of 15 stages determined by observing the timing of chondrification and calcification of the skeleton every 24 h, from 3 to 17 days of incubation. Cartilage and ossified bones were stained blue and red with Alcian blue 8GX and alizarin red S, respectively. These skeletogenous stages of the Japanese quail embryo will be useful as a normal control not only in studies of experimental embryology, teratological testing, and developmental engineering, but also in the analysis of mutant embryos with skeletal abnormalities.  相似文献

Domestic fowl or chicken (Gallus gallus) and Japanese quail (Coturnix japonica) belong to the family Phasianidae. The exchange of marker information between chicken and quail is an important step towards the construction of a high-resolution comparative genetic map in Phasianidae, which includes several poultry species of agricultural importance. We tested chicken microsatellite markers to see if they would be suitable as genetic linkage markers in Japanese quail. Twenty-six per cent (31/120) of chicken primers amplified individual loci in Japanese quail and 65% (20/31) of the amplified loci were found to be polymorphic. Eleven of the polymorphic loci were excluded as uninformative because of the lack of amplification in some individuals or high frequency of nonspecific amplification. The sequence information of the remaining nine loci revealed six of them to contain microsatellites that were nearly identical with those of the orthologous regions in chicken. For these six loci, allele frequencies were estimated in 50 unrelated quails. Although the very few chicken markers that do work well in quail could be used as anchor points for a comparative mapping, most chicken markers are not useful for studies in quail. Therefore, more effort should be committed to developing quail-specific markers rather than attempting to adapt chicken markers for work in quail.  相似文献

Summary A 17-generation selection experiment was conducted to study direct and correlated responses to mass selection under a mating system with alternating generations of full-sib inbreeding and wide outbreeding (population I) as compared with a mass selected, randomly mated population (population II). The selection criterion was an index of total egg mass to 78 days divided by adult female body weight. Estimates of heritabilities and genetic correlations are reported. Estimated heritabilities for the index were 0.38±0.04 and 0.29±0.05 in population I and II, respectively. Realized heritabilites were 0.10±0.05 and 0.12±0.03. For most traits studied the mean phenotypic values in the cyclic mated population decreased for inbred generations. Increased inbreeding levels also caused outbred generation means of population I to decrease through the first six or seven generations. After this period of adaptation to inbreeding selection response was positive for the index and positively correlated traits. Total response to selection under the cyclic inbred-outbred mating system did not exceed selection response made under random mating. However, the rate of response in the cyclically mated population exceeded that in the randomly mated population in later generations when the cyclically mated population had apparently adapted to inbreeding.Published with approval of the Director of the Montana Agricultural Experiment Station, Journal Series No. 1221  相似文献

Summary. An electrophoretic variant of sheep transferrin, TfL, has been described. Transferrin L has been shown to be controlled by a single codominant allele, TfL, at the Tf locus. Transferrin L is electrophoretically distinguishable from the very similar transferrin TfKCzech. The value of gradient polyacrylamide gel electrophoresis for transferrin phenotyping in sheep is discussed.  相似文献

In line with the Gifu University''s initiative to map the Japanese quail genome, a total of 100 Japanese quail microsatellite markers isolated in our laboratory were evaluated in a population of 20 unrelated quails randomly sampled from a colony of wild quail origin. Ninety-eight markers were polymorphic with an average of 3.7 alleles per locus and a mean heterozygosity of 0.423. To determine the utility of these markers for comparative genome mapping in Phasianidae, cross-species amplification of all the markers was tested with chicken and guinea fowl DNA. Amplification products similar in size to the orthologous loci in quail were observed in 42 loci in chicken and 20 loci in guinea fowl. Of the cross-reactive markers, 57.1% in chicken and 55.0% in guinea fowl were polymorphic when tested in 20 birds from their respective populations. Five of 15 markers that could cross-amplify Japanese quail, chicken, and guinea fowl DNA were polymorphic in all three species. Amplification of orthologous loci was confirmed by sequencing 10 loci each from chicken and guinea fowl and comparing with them the corresponding quail sequence. The microsatellite markers reported would serve as a useful resource base for genetic mapping in quail and comparative mapping in Phasianidae.  相似文献

Japanese quail exhibit a robust circadian rhythm in body temperature. This rhythm is readily entrainable by 24 h light-dark (LD) cycles and persists under constant conditions. Because both the pineal organ and the eyes have been implicated as major components of the circadian system of birds, the role of these organs in generating the rhythm of body temperature was investigated. Pinealectomy, when performed alone, had little effect on the body temperature rhythm of quail either under LD or under constant darkness (DD). Most birds subjected to optic nerve section alone remained rhythmic in DD although the robustness of the rhythm was decreased, and 25% became arrhythmic. Birds subjected to both pinealectomy and optic nerve section behaved similarly to birds subjected to optic nerve section alone. However, complete eye removal, when performed alone or in combination with pinealectomy, caused all birds to become arrhythmic in DD. The data support the hypothesis that the eyes are the loci of circadian pacemakers in quail that act, via both neural and hormonal outputs, to preserve the integrity of (self-sustaining or damped) circadian oscillators located elsewhere.  相似文献

Common responses to hypoxia include decreased body temperature (Tb) and decreased energy metabolism. In this study, the effects of hypoxia and hypercapnia on Tb and metabolic oxygen consumption (V.O2) were investigated in Japanese quail (Coturnix japonica). When exposed to hypoxia (15, 13, 11 and 9% O2), Tb decreased only at 11% and 9% O2 compared to normoxia; quail were better able to maintain Tb during acute hypoxia after a one-week acclimation to 10% O2. V.O2 also decreased during hypoxia, but at 9% O2 this was partially offset by increased anaerobic metabolism. Tb and V.O2 responses to 9% O2 were exaggerated at lower ambient temperature (Ta), reflecting a decreased lower critical temperature during hypoxia. Conversely, hypoxia had little effect on Tb or V.O2 at higher Ta (36 °C). We conclude that Japanese quail respond to hypoxia in much the same way as mammals, by reducing both Tb and V.O2. No relationship was found between the magnitudes of decreases in Tb and V.O2 during 9% O2, however. Since metabolism is the source of heat generation, this suggests that Japanese quail increase thermolysis to reduce Tb. During hypercapnia (3, 6 and 9% CO2), Tb was reduced only at 9% CO2 while V.O2 was unchanged.  相似文献

Summary. Phosvitin polymorphism in egg yolk of the Japanese quail was found by horizontal polyacrylamide gradient gel electrophoresis. Six phenotypes of yolk phosvitin designated A, B, C, AB, AC, and BC were observed in a population of 281 birds. Analysis of family data revealed that the phenotypic variation of quail yolk phosvitins was controlled by an autosomal Pv locus with three codominant alleles, Pva, Pvb and Pvc. The gene frequencies of Pva, Pvb and Pvc were 0.064, 0.824 and 0.112, respectively.  相似文献

Plasma transferrin (Tf) types in dogs were studied by a method of horizontal polyacrylamide gel electrophoresis with a 10 % separation gel and a discontinuous buffer system (Tris-citrate-borate, pH 9.0). Samples were analysed from 1127 dogs belonging to 60 different breeds. TfB and Tfc alleles, each in considerably high frequency, were observed in most of the breeds. TfA with relatively lower frequency was observed in Beagle, Cocker Spaniel, Doberman Pinscher, German Shepherd, German Shorthaired Pointer, Old Danish Pointer and Poodle. Two rare Tf alleles, TfD and the other designated as TfE, were observed only in Cocker Spaniel and in Poodle respectively. Basenji (44 samples) and Small Münsterlander (17 samples) showed the highest frequency (0.97) for TfC allele while Carelian Bear Dog (19 samples) showed the highest frequency (0.97) for TfB allele. In the total material, the frequencies of Tf alleles A, B, C, D and E were found to be 0.0182, 0.5075, 0.4716, 0.0009 and 0.0018 respectively. By using the observed and expected numbers of Tf heterozygotes, the average inbreeding coefficient (F) within breeds was estimated to be 0.14. Six samples of wolf (Canis lupus L.) studied seemed to be of Tf B type. The present study, along with earlier reports, showed that some of the plasma proteins and enzymes (albumin, transferrin, eserine resistant esterase) exhibit considerably more polymorphism than that reported for hemoglobin and some of the red cell and tissue enzymes in the domestic dog.  相似文献

Age-related changes occur in histones of the liver and oviduct of the female quail. In the liver an extra histone band, named HCX, between H2A and H4, is observed that increases with age. In the oviduct, a large complex of histones is seen which is tissue-specific. This complex declines with increasing age. The changes in the histones of the oviduct of adult and old birds in response to estradiol and progesterone are age-related. In the adult, the histone-complex increases after administration of either one of the hormones. In old birds, however, it is seen only after progesterone administration. Thus, the alterations in histones in the birds are not only tissue- and age-related, but also vary in response to steroid hormones.  相似文献
