Concentrations of volatile fatty acids (VFA) normally found in bovine rumen fluid inhibited growth of Escherichia coli in Antibiotic Medium 3. Acetic, propionic, and butyric acids each produced growth inhibition which was markedly pH-dependent. Little inhibition was observed at pH 7.0, and inhibition increased with decreasing pH. A combination of 60 mumoles of acetate, 20 mumoles of propionate, and 15 mumoles of butyrate per ml gave 96, 69, and 2% inhibition at pH 6.0, 6.5, and 7.0, respectively. Rumen fluid (50%) gave 89 and 48% inhibition at pH 6.0 and 6.5, respectively, and growth stimulation (22%) at pH 7.0. Rumen fluid inhibitory activity was heat-stable, was not precipitated by 63% ethyl alcohol, and was lost by dialysis and by treatment with anion-exchange resins but not with cation-exchange resins. These results are consistent with the idea that VFA are the inhibitory substances in rumen fluid. Previous results which indicated that rumen fluid VFA did not inhibit E. coli growth were due to lack of careful control of the final pH of the growth medium. The E. coli strain used does not grow in rumen fluid alone at pH 7.0.  相似文献

The fermentation of uniformly labeled glucose-C14, glucose-1-C14, -2-C14, and -6-C14, xylose-1-C14, cellulose-1-C14, -2-C14, and -6-C14, and lactate-2-C14 by rumen fluids from cows fed all-hay, hay and concentrate (50:50), and all-concentrate diets was investigated. The results obtained suggested that the Embden-Meyerhof glycolytic pathway is the major pathway of hexose utilization, that the major pathway of xylose fermentation involves hexose synthesis, and that the contributions of the nonrandomizing (acrylate) pathway of propionate formation during glucose, xylose, and cellulose fermentations are 4.5, 8.0, and 10.5%, and 24.6, 25.8, and 17.2%, respectively, by rumen fluids from the cows fed all-hay and all-concentrate rations.  相似文献

Volatile Fatty Acid Requirements of Cellulolytic Rumen Bacteria   总被引:11,自引:1,他引:11  
A gas chromatographic method was developed which could separate the isomers isovaleric and 2-methylbutyric acid. Subsequent analyses revealed that most commercially available samples of these acids were cross-contaminated; however, one sample of each acid was found to be pure by this criterion. The growth response of seven strains of cellulolytic rumen bacteria (three strains of Bacteroides succinogenes, three strains of Ruminococcus flavefaciens, and one strain of R. albus) to additions of isobutyric, isovaleric, 2-methylbutyric, valeric, and combinations of valeric and a branched-chain acid was determined. Strains of B. succinogenes required a combination of valeric plus either isobutyric or 2-methylbutyric acid. Isovaleric acid was completely inactive. Either isobutyric or 2-methylbutyric acid was required for the growth of R. albus 7. Strain C-94 of R. flavefaciens grew slowly in the presence of any one of the three branched-chain acids, but a combination of isobutyric and 2-methylbutyric acids appeared to satisfy this organism's growth requirements. None of the individual acids or mixtures of straight- and branched-chain acids allowed growth of R. flavefaciens strain C1a which would approach the response obtained from the total mixture of acids. Further work indicated that all three branched-chain acids were required for optimal growth by this strain, although isovaleric acid only influenced the rate of maximal growth. Either 2-methylbutyric or isovaleric acid allowed growth of nearly the same magnitude as that of the positive control for R. flavefaciens B34b. The presence of acetic acid had little influence on the rate or extent of growth of any of the strains except R. albus 7, for which the extent of growth was markedly increased. Determination of the quantitative fatty acid requirements for the three B. succinogenes strains indicated that 0.1 μmole of valeric per ml and 0.05 μmole of 2-methylbutyric per ml permitted maximal growth. However, with isobutyric acid as the branched-chain component, strains A3c and B21a required 0.1 μmole/ml in contrast to S-85 which exhibited optimal growth at the 0.05 μmole/ml level. By use of mixtures of isobutyric and 2-methylbutyric acids, good growth of C-94 was obtained at concentrations of 0.1 and 0.01 μmole/ml, respectively. About 0.3 μmole/ml of each acid was required for satisfactory growth of C1a.  相似文献

The composition of the rumen microflora and the volatile fatty acids were examined in cattle free-grazing on grass or stall-fed on hay, grass pellets, oats or dried beet pulp with molasses. Total and viable counts of anaerobic bacteria were highest on the grass feeding, but viable counts as a percentage of total counts were highest when oats or beet pulp with molasses were fed. Counts of cellulolytic bacteria were lowest on these latter 2 diets, and highest on grass or grass pellet diets. Studies of the anaerobic flora showed that the composition in animals fed on grass pellets resembled more that found in animals free-grazing on grass than in those fed on hay. Counts of aerotolerant bacteria were only a small percentage of the total count, but were highest on the hay diet. On this latter diet and on grass-feeding the streptococci (identified as Streptococcus bovis) were predominant, but contrary to expectation, streptococci were found only in small numbers on the oats diet, where coryneform rods were the major type present. Although a period of 4–6 weeks was allowed for the animals to adapt to the feeds, the 2 periods of feeding on oats and dried beet pulp with molasses markedly affected the composition of the rumen flora in the subsequent periods of feeding grass pellets and hay. Ruinen volatile fatty acid analysis showed a propionogenic effect of oats and the highest percentage of butyric acid when beet pulp with molasses was fed. The expected propionogenic effect of grass pellets was not observed.  相似文献

The major volatile acid produced by Haemophilus vaginalis was acetic acid. The absence of propionate, butyrate, and cellular branched-chain fatty acids indicates that H. vaginalis is not related to Propionibacterium or Butyribacterium.  相似文献

The effects of low concentrations of long-chain fatty acids (palmitic, stearic, oleic, and vaccenic) on the growth of seven species (13 strains) of rumen bacteria were investigated. Except for Bacteroides ruminicola and several strains of Butyrivibrio fibrisolvens, bacterial growth was not greatly affected by either palmitic or stearic acids. In contrast, growth of Selenomonas ruminantium, B. ruminicola, and one strain of B. fibrisolvens was stimulated by oleic acid, whereas the cellulolytic species were markedly inhibited by this acid. Vaccenic acid (trans Δ11 18:1) had far less inhibitory effect on the cellulolytic species than oleic acid (cis Δ9 18:1). Inclusion of powdered cellulose in the medium appeared to reverse both inhibitory and stimulatory effects of added fatty acids. However, there was little carry-over effect observed when cells were transferred from a medium with fatty acids to one without. Considerable variation in response to added fatty acids was noted among five strains of B. fibrisolvens. In general, exogenous long-chain fatty acids appear to have little, if any, energy-sparing effect on the growth of rumen bacteria.  相似文献

In our searching program for novel sorbicillin related compounds, three novel compounds, spirosorbicillinols A (1), B (2), and C (3), were isolated from the fermentation broth of the USF-4860 strain isolated from a soil sample. The planar structures of compounds 13 were determined from spectroscopic evidence and degradation reaction, and that of 1 was the same as that of 2. The relative stereochemistries of compounds 13 were determined by 1H-1H coupling constants, the elucidation of HMBC and NOESY spectra in detail. 1 and 2 were stereoisomers at C8 position, each other. We propose that compounds 1 and 2 were formed by exo and endo intermolecular Diels-Alder reaction between sorbicillinol as a diene and scytolide (proposed precursor-1) as a dienophile, respectively. Similarly, we propose that compound 3 was formed by an endo intermolecular Diels-Alder reaction between sorbicillinol and proposed precursor-2.  相似文献

A microscale biosensor for acetate, propionate, isobutyrate, and lactate is described. The sensor is based on the bacterial respiration of low-molecular-weight, negatively charged species with a concomitant reduction of NO3 to N2O. A culture of denitrifying bacteria deficient in N2O reductase was immobilized in front of the tip of an electrochemical N2O microsensor. The bacteria were separated from the outside environment by an ion-permeable membrane and supplied with nutrients (except for electron donors) from a medium reservoir behind the N2O sensor. The signal of the sensor, which corresponded to the rate of N2O production, was proportional to the supply of the electron donor to the bacterial mass. The selectivity for volatile fatty acids compared to other organic compounds was increased by selectively enhancing the transport of negatively charged compounds into the sensor by electrophoretic migration (electrophoretic sensitivity control). The sensor was susceptible to interference from O2, N2O, NO2, H2S, and NO3. Interference from NO3 was low and could be quantified and accounted for. The detection limit was equivalent to about 1 μM acetate, and the 90% response time was 30 to 90 s. The response of the sensor was not affected by changes in pH between 5.5 and 9 and was also unaffected by changes in salinity in the range of 2 to 32‰. The functioning of the sensor over a temperature span of 7 to 30°C was investigated. The concentration range for a linear response was increased five times by increasing the temperature from 7 to 19.5°C. The life span of the biosensor varied between 1 and 3 weeks after manufacturing.  相似文献

Deoxyribonuclease I (DNase I) is known to be a glycoprotein, and two potential N-linked glycosylation sites (N18 and N106) are known for mammalian enzymes. In the present study, N18 and N106 were mutated in order to investigate the biological role of N-linked glycosylation in three mammalian (human, bovine, and equine) DNases I. The enzyme activities of N18Q and N106Q were lower than that of the wild type, and that of the double mutant (N18Q/N106Q) was lower than those of the single mutants, in accord with the sugar moiety contents in the three mammals. In addition, all mutant enzymes were unstable to heat, suggesting that both sites are required for heat stability. Moreover, in human and equine enzymes, the N18Q and N106Q mutant enzymes were less resistant to trypsin, while N18Q/N106Q was the most sensitive to trypsin. As for bovine DNase I, the trypsin resistance of N18Q and N106Q was similar to that of the wild type, but that of N18Q/N106Q decreased in a time-dependent manner. On the other hand, N-linked glycosylation was not related to pH sensitivity. The results of the present study suggest that N18 and N106 are both necessary for (i) full enzymatic activity, (ii) heat-stability, and (iii) trypsin resistance.  相似文献

应用毛细管气相色谱-质谱联用技术对红树植物木榄胚轴中挥发性成分和脂肪酸成分进行了分析检测,检测到挥发性成分30个,鉴定了其中的27种成分,占所检测挥发性成分总量的97.01%,以烷烃类为主,占鉴定成分的67.43%,还有酸、酯及萜类化合物等;检测到脂肪酸组分23种,确定了其中的19种,脂肪酸检出量为97.81%,其中饱和脂肪酸占64.87%,不饱和脂肪酸占35.13%。  相似文献

亚油酸、亚麻酸是哺乳动物体内的必需脂肪酸,但哺乳动物由于缺乏△12和ω-3脂肪酸脱氢酶而自身不能合成.△12和ω-3脂肪酸脱氢酶存在于真菌、植物和一些低等动物中.为了实现哺乳动物细胞亚油酸的自身合成,克隆了线虫编码△12脂肪酸脱氢酶的FAT-2基因eDNA序列,通过优化密码子,构建真核表达载体,稳定转染细胞,经抗生素筛选获得稳定整合FAT-2基因的CHO细胞.PCR和RNA印迹(Northern blot)验证了基因的整合和表达.气相色谱分析细胞的脂肪酸含量表明,FAT-2基因的表达显著提高了转基因细胞中亚油酸的含量,亚油酸含量为阴性对照细胞的2.4倍.研究结果表明,低等动物△12脂肪酸脱氢酶可以重建哺乳动物多不饱和脂肪酸合成途径,并利用细胞中的油酸合成亚油酸.上述研究为进一步利用转基因技术促进农业动物合成多不饱和脂肪酸从而提高食品营养价值奠定基础.  相似文献

S ummary . Suspensions of mixed rumen bacteria were incubated with long-chain fatty acids and their corresponding triglycerides in the presence and absence of the food particles present in the rumen. The uptake of free fatty acids and triglycerides by the bacteria was due largely to physical adsorption. With increasing unsaturation of the free fatty acids, less adsorption occurred and in all instances the presence of food particles reduced greatly the extent to which the fatty acids were adsorbed on the bacteria. The triglycerides were adsorbed to the bacteria to a much less extent than their corresponding free fatty acids, more of the substrate remaining in the supernatant fraction. The presence of food particles had little effect on the extent to which the triglycerides became adsorbed to the bacteria, but reduced greatly the amount of triglyceride present in the supernatant fraction. The decrease in amount of free fatty acid associated with the bacteria and the decrease in the amount of triglyceride in the supernatant liquid when food particles were present, was wholly accounted for by the increase in amount of these substrates associated with the food particles. The implications of these findings with respect to the metabolism of lipids in the rumen is discussed.  相似文献

Two experiments were conducted to examine the effect of zinc (Zn) source on the performance, Zn status, immune response, and rumen fermentation of lactating cows to find the most available Zn source for dairy production. In Experiment 1, a total of 30 multiparous Holstein cows were randomly allocated by body weight and milk yield to one of five treatments in a completely randomized design. Cows were fed a total mixed ration (TMR) with no Zn addition (containing 37.60 mg?Zn/kg TMR by analysis), and the basal TMR supplemented with 40 mg?Zn/kg TMR from either Zn sulfate or one of three organic Zn chelates with weak (Zn-AA W), moderate (Zn-Pro M), or strong (Zn-Pro S) chelation strengths, respectively for 55 days. In Experiment 2, the in vitro rumen fermentation method was used in a completely randomized design involving a 4?×?3 factorial arrangement of treatments. The four Zn sources were the same as those used in Experiment 1, and the three supplemental Zn levels in the rumen fluid were 0, 10, and 20 μg/mL, respectively. The feed intake, milk composition, and somatic cell count (SCC) were unaffected (P?>?0.05) by treatments. However, the milk yield was increased (P?<?0.05) by addition of Zn from both the Zn-AA W and Zn-Pro S. Plasma Zn level at the end of the experiment was increased (P?<?0.05) by addition of Zn from all three organic sources. Serum antibody titers on day 21 after vaccination with foot and mouth disease (FMD) vaccine were increased (P?<?0.05) by both supplemental Zn-AA W and Zn-Pro S. The organic Zn sources with different chelation strengths supplemented at the added Zn level of 10 μg/mL were more effective (P?<?0.05) in improving the rumen fermentation than Zn sulfate, with the most effective being Zn-AA W. In conclusion, Zn source had no influence on the feed intake, milk composition, and SCC; however, both the Zn-AA W and Zn-Pro S were more effective than Zn-Pro M and Zn sulfate in enhancing the rumen fermentation, Zn status, and humoral immune response as well as improving milk yield of lactating cows. The improved milk production might be attributed to the improved rumen fermentation, Zn status, and immune function.  相似文献

Acetate dominated the extracellular pool of volatile fatty acids (VFAs) in the hindgut fluid of Reticulitermes flavipes, Zootermopsis angusticollis, and Incisitermes schwarzi, where it occurred at concentrations of 57.9 to 80.6 mM and accounted for 94 to 98 mol% of all VFAs. Small amounts of C3 to C5 VFAs were also observed. Acetate was also the major VFA in hindgut homogenates of Schedorhinotermes lamanianus, Prorhinotermes simplex, Coptotermes formosanus, and Nasutitermes corniger. Estimates of in situ acetogenesis by the hindgut microbiota of R. flavipes (20.2 to 43.3 nmol · termite−1 · h−1) revealed that this activity could support 77 to 100% of the respiratory requirements of the termite (51.6 to 63.6 nmol of O2 · termite−1 · h−1). This conclusion was buttressed by the demonstration of acetate in R. flavipes hemolymph (at 9.0 to 11.6 mM), but not in feces, and by the ability of termite tissues to readily oxidize acetate to CO2. About 85% of the acetate produced by the hindgut microbiota was derived from cellulose C; the remainder was derived from hemicellulose C. Selective removal of major groups of microbes from the hindgut of R. flavipes indicated that protozoa were primarily responsible for acetogenesis but that bacteria also functioned in this capacity. H2 and CH4 were evolved by R. flavipes (usually about 0.4 nmol · termite−1 · h−1), but these compounds represented a minor fate of electrons derived from wood dissimilation within R. flavipes. A working model is proposed for symbiotic wood polysaccharide degradation in R. flavipes, and the possible roles of individual gut microbes, including CO2-reducing acetogenic bacteria, are discussed.  相似文献

A diffusion chamber is described which concentrates short-chain, volatile fatty acids from seawater while simultaneously separating them from interfering salts. The procedure relies on the passive diffusion of volatile compounds from acidified seawater samples and their subsequent absorption onto a base-impregnated filter. As part of our research program assessing the use of liquid hog manure (LHM) to control root-lesion nematodes, Pratylenchus penetrans, a series of acute toxicity tests was conducted to: (i) examine if non-ionized forms of volatile fatty acids (VFA) are responsible for the mortality of P. penetrans exposed to LHM under acidic conditions, (ii) determine if Caenorhabditis elegans can be a surrogate for P. penetrans in screening tests by comparing their sensitivities to VFA, (iii) characterize the nematicidal effect of individual VFA in LHM to P. penetrans, and (iv) determine whether individual VFA in LHM interact in their toxicity to P. penetrans. LHM was significantly (P ≤ 0.05) more toxic to P. penetrans than a mixture of its main VFA components at concentrations of 5% and 10% (vol. VFA or LHM /vol. in buffer). Pratylenchus penetrans was more sensitive to acetic acid than C. elegans, whereas the sensitivity of both nematode species to n-caproic acid was similar. Individual VFA vary in their lethality to P. penetrans. n-valeric acid was the most toxic (LC95= 6.8 mM), while isobutyric acid was the least toxic (LC95 = 45.7 mM). Individual VFA did not interact in their toxicity to P. penetrans, and their effects were considered additive. VFA account for the majority of the lethal effect of LHM to P. penetrans under acidic conditions. Caenorhabditis elegans cannot be used as a surrogate to P. penetrans in toxicity studies using VFA. The efficacy of LHM to control P. penetrans can be evaluated by assessing its VFA content prior to application, and this evaluation is facilitated by the fact that the interaction of individual VFA appears to be simply additive.  相似文献
