Summary A comparative study was made of the photoperiodic and thermoperiodic induction of diapause in the phytoseiid mite Amblyseius potentillae. Sensitivity to thermoperiod was found to be highest during the protonymphal and deutonymphal stages, with some sensitivity still being present in the young adult. Summation of both photoperiodic and thermoperiodic cycles was shown to take place, which demonstrated the presence of a photoperiodic counter as well as a thermoperiodic counter in these mites. Vitamin A appeared to be necessary for some early step in the physiological mechanism of diapause induction and not just for the expression of the diapause response. The light sensitivity threshold for photoperiodic induction of diapause was found to be extremely low, viz. less than 0.02 W/cm2. Moreover, the light sensitivity threshold appeared to be strongly temperature dependent in A. potentillae. Experiments in which the mites experienced various sequences of short-day photoperiods and short-day thermoperiods, applied either concurrently or in succession, showed that the information collected by the photoperiodic counter and the thermoperiodic counter is integrated into one induction sum. Psychiatric practice involves writing as much as it involves talking. This study examines the interpretive processes of reading, writing and interviewing which are central to the clinical interaction. It is part of a broader ethnographic study of an Australian psychiatric hospital (which specializes in the treatment of patients with a diagnosis of schizophrenia). The paper examines two major types of written assessment of patients — the admission assessment and the complete work-up. Writing is analyzed as performance, thereby focusing on the transformations that are effected in patients, their perceptions of their schizophrenia, and their total identity. One crucial transformation is from person suffering from schizophrenia to schizophrenic. The paper aims to show that as much as psychiatry is a talking cure it is also a writing cure.  相似文献

Embryogenic avocado cultures were exposed to ionizing irradiation in order to determine its effect on proliferation and subsequent somatic embryo development. The approximate PD50 as determined by linear regression is 35 Gy 2 weeks after irradiation for Fuerte 2.11.1 and 4 weeks after irradiation for T362 2.11.1. Irradiation of embryogenic cultures did not significantly affect the number of early stage Fuerte 2.11.1 somatic embryos that developed directly from irradiated cultures; however, 10–50 Gy inhibited somatic embryo development. Irradiation of T362 2.11.1 embryogenic cultures at 25–50 Gy inhibited the number of intermediate and mature stages of somatic embryos that developed directly from irradiated cultures, and 50 Gy inhibited somatic embryo maturation. Inhibition of somatic embryo development could be partially offset by proliferation of irradiated embryogenic cultures as suspensions. Irradiation up to 10 Gy significantly increased the number of mature Fuerte 2.11.1 somatic embryos that developed from suspension cultures. Irradiation with doses up to 25 Gy stimulated development of heart stage T362 2.11.1 somatic embryos; however, mature somatic embryo development was suppressed at dosages of 10 Gy and greater.  相似文献

Bragg and Cobb soybean [Glycine max (L.) Merr.] seeds were germinated in sand at temperatures ranging from 25 to 40°C. Emergence decreased with increasing temperature above 37°C, with virtually no emergence at 40°C. Emergence of 12 other cultivars at 38°C ranged from 25 to 95%. We report the phylogenetic and physiological characterization of a mesophilic and halophilic member of the filamentous anoxygenic phototrophic (FAP) bacteria, provisionally named Candidatus Chorothrix halophila gen. nov. sp. nov., that has been maintained in a highly enriched culture in our laboratory for over a decade. Phylogenetic analysis of small-subunit RNA-encoding sequences places Candidatus Chlorothrix halophila in a clade that includes cultivated members of the genera Chloroflexus and Oscillochloris. Physiological studies demonstrated sulfide-dependent photosynthetic uptake of 14C-labeled bicarbonate. Enzymatic assays for the activity of propionyl-coenzyme A synthase indicated that Candidatus Chlorothrix halophila does not use the 3-hydroxypropionate cycle of Chloroflexus aurantiacus OK-70-fl for autotrophic carbon assimilation. Summary Ten hairy-cell leukemia patients were treated with interferon (IFN-) at a dose rate of 2 × 106 IU/m2 × 5 days for 4 weeks (induction therapy) and, thereafter, at the same dose three times a week for 11 months (maintenance therapy). The effect of this treatment on serum neopterin, 2-microglobulin, (2–5)oligoadenylate [(2–5)A n ] levels, intracellular (2–5)A n values and human Mx protein synthesis was analysed. There were significant rises in serum neopterin and (2–5)A n levels during both induction and maintenance, whereas 2-microglobulin levels rose only during induction. Rises in intracellular (2–5)A n were documented mainly during induction, but they were not significantly higher than pretherapy values. IFN provoked an increase in human Mx protein synthesis over the entire induction — maintenance period, but was only significantly higher than baseline during induction. All markers proved useful for monitoring the effects of IFN dose schedules, but were not predictive of clinical outcome. Natural killer activity and IFN production, which were initially defective, followed a different trend from that of the other factors studied, in that increases were documented only late in the course of therapy when the disease was already in remission.  相似文献

Rhizopus is a zygomycetous genus. Several species of this taxon may infect humans and lower animals. Seventeen isolates ofRhizopus species in three distinct morphological groups were studied: the stolonifer group (sporangiophores greater than 1 mm in height, sporangial diameters of 100–275 µm, branched rhizoids); the arrhizus group (sporangiophores greater than 1 mm in height, branched rhizoids, sporangial diameters of 100–240 µm); and the microsporus group (sporangiophores less than 0.8 mm in height, sporangial diameters less than 100 µm, simple rhizoids). Maximal growth temperatures were characteristic: the stolonifer group grew at 30°C, the arrhizus group grew at 36°C, and the microsporus group grew at 45°C. The DNA mol% G + C base composition of all isolates ranged from 34.9 to 40.2% Species within the three groups were grouped by DNA differences. The arrhizus group was most distinctive with a value of 34.9–36.3%; the stolonifer and microsporus groups had G + C values of 37.0–39.3% and 37.8–40.2%, respectively. Our research clarifies and defines the G-C values of the three important groups ofRhizopus species.  相似文献

There might be an undetermined order in the interaction of the substrates with the ribosomes in the reaction of CACCA-Leu-Ac with puromycin to form Ac-Leu-puromycin and CACCA (fragment reaction). Km 0°=6×10-4 M for the puromycin · ribosome interaction. Chloramphenicol totally blocks the fragment reaction as a consequence of a single interaction with the ribosome of Kd 0°=2.2×10-6 M. The inhibition by chloramphenicol of the fragment reaction is mixed competitive for puromycin.  相似文献

Summary An exotic Zea mays L. population (Tuxpeno) was adapted to North Carolina conditions by first introducing genes for adaptability from two North Carolina varieties ([(Jarvis X Indian Chief)Tuxpeno]Tuxpeno) including four generations of intermating, and then selecting for adaptability using maturity as the primary measure. The study evaluated selection for adaptability and the diversity available between adapted Tuxpeno and the local varieties, Jarvis and Indian Chief. Analytical procedures were developed to quantify the diversity between populations and the complementation of local varieties by introduced germ plasms. The analyses utilized the specific effects available from the diallel mating design.Three replicate selections responded similarly under simple recurrent mass selection (1/10) for the earliest disease-free plants initially and additionally for plant types (primarily height) in the final generation. The 1/4 local germ plasm permitted rapid adaptation of Tuxpeno gene pool to local conditions. The adapted Tuxpeno populations yielded similarly to the local populations with an average heterosis for grain yield of 28% when crossed to the local populations used as source of genes for adaptability. The diversity found between adapted Tuxpeno lines and these local varieties based on genes affecting grain yield was 1.5 to 2.5 times that measured between the local varieties (Jarvis and Indian Chief). Diversity lost through intergradation with local material was a reasonable investment. Yield genes introduced from Tuxpeno complemented local gene pools through nonadditive, primarily dominance-associated, gene effects. Reassortment of major gene blocks apparently occurred leading to significant divergence among replicate selections involving both additive-associated and dominance-associated gene effects.Paper No. 6355 of the North Carolina Agri. An Arctic clone ofPhaeocystis pouchetii LAGERHEIM was compared toPhaeocystis globosa SCHERFFEL isolated from the southern North Sea with regard to temperature tolerance and colony shapes. Already youngP.pouchetii colonies (<100 m) show the typical distribution of the cells in groups, separated from each other by wide zones of cell-free mucilage; the maximum colony size is ca 2 mm in diameter.P.pouchetii colonies form clouds with bubble-like vesicles, spherical colony-shapes are seldom found.P.globosa colonies are spherical up to a size of 2 mm; the cells are distributed homogeneously over the periphery of the colonies. A pouchetii-like distribution of cells never occurs either in the spherical young colonies or in the pear-shaped old colonies (size up to 8 mm). A development from the colony shape of the globosa-type to the pouchetii-type or vice versa was never found. Therefore the colony shape has to be considered a constant distinctive character. Single cells ofP.pouchetii andP.globosa cannot be separated from each other by using the light microscope; this also holds for the flagellates and the non-motile cells.P.pouchetii grows well between 0°C and 14°C,P.globosa between 4°C and 22°C, respectively. Because of the distinctive differences in the morphology of the colonies and the differences in temperature tolerances we propose thatPhaeocystis globosa should no longer be considered conspecific withPhaeocystis pouchetii.  相似文献

Zusammenfassung 1. In den Wintern 1960/61, 1961/62 und 1962/63 wurde mit künstlicher Kälte (–13 bis –25 °C), die ein fahrbares Kälteaggregat erzeugte, die Frostresistenz von Pflaumenzuchtmaterial und einer Reihe von Pflaumenunterlagen im Freiland geprüft. Besondere Berücksichtigung fand dabei der natürliche Temperaturverlauf.2. Von den geprüften Pflaumengehölzen zeigte die geringste Frostresistenz eine Auslese von Prunus insititia und die Auslesen von Prunus cerasifera, besonders der tetraploide Klon B IV, 19,13, die diploiden Klone B IV, 20,6 und B IV, 17,15. Diese Typen erfroren nach einer zweitägigen, natürlichen Abhärtung im Dezember bei –20 °C; erst nach einer längeren Abhärtungszeit von 4 Tagen und mehr war ihre Frostresistenz gesteigert worden, so daß sie –20 °C schadlos überlebten. Nach kurzfristigen, witterungsbedingten Erwärmungen im Mittwinter wurden diese Formen derart schnell enthärtet, so daß sie bei Temperaturen um –15 °C erfroren. Nach längerem Tauwetter im Mittwinter erlangten sie nur sehr langsam eine erneute Frostresistenz.3. Insgesamt etwas besser zu beurteilen waren einige hexaploide Unterlagensorten, wie Ackermann, Brünker und Schwamborn 103. Sie tolerierten nach ztägiger Abhärtung –20 °C; Naundorf 102 und Brompton jedoch erst nach 4 Tagen. Ebenso stellt sich nach einer kurzfristig erfolgten Erwärmung nach ztägiger, erneuter Abhärtung wieder eine hohe Frostresistenz ein. Die relative Frostempfindlichkeit der geprüften Unterlagen kam darin zum Ausdruck, daß sie nach einer eintägigen Erwärmung im Mittwinter bei –24 °C total erfroren.4. Die beste Frostresistenz zeigten Vertreter der nordamerikanischen Pflaumen, wie Prunus americana und Assiniboine. Bereits nach einer zweitägigen Abkühlung Anfang Dezember überlebten sie schadlos –20 °C; nach 4 Tagen –25 °C. Ihre erreichte Frostresistenz war auch bedeutend stabiler als die der geprüften Auslesen von Prunus cerasifera und der hexaploiden Unterlagen.5. Es wurde der Versuch unternommen, einige der geprüften Pflaumentypen nach ihrer Frostverträglichkeit zu gruppieren, wobei die Merkmale Abhärtung und Enthärtung Berücksichtigung fanden.6. Zur Verbesserung der Frostresistenz der Pflaumen werden der Züchtung zwei Wege vorgeschlagen: 1. Herstellung von Nachkommenschaften aus genügend frostresistenten hexaploiden Formen; 2. Artbastardierungen mit bekannt frostresistenten Arten nordamerikanischer Herkunft.7. Neue Wege müssen auch in der Unterlagenzüchtung beschritten werden. Es wäre sinnvoll, dafür besonders diploide frostharte Arten als Kreuzungspartner zu berücksichtigen.
Contributions to breeding research on plumsIV. Experiments on frost resistance of plum clones and rootstocks
Summary 1. During the winters of 1960/61, 1961/62 and 1962/63 a number of plum clones and stocks were tested outdoors for frost resistance. Artificial cold (–13 to –25 °C) was produced by means of a portable cooling system, taking natural temperature conditions into account.2. Of the plum woods tested, the lowest frost resistance was found in Prunus insititia and Prunus cerasifera, particularly in the tetraploid clone B IV, 19.13 and the diploid clones B IV, 20.6 and B IV, 17.15. These types froze in December after 2 days of hardening at –20 °C; only after 4 or more days of hardening was their frost resistance increased sufficiently to allow survival at –20 °C. After short warm spells in mid winter they became less hardy rapidly and froze at –15 °C. Prolonged thaw delayed their ability to acquire renewed frost resistance.3. The hexaploid plum rootstocks Ackermann, Brünker and Schwamborn 103 survived a temperature of –20 °C after a two day period of hardening; Naundorf 102 and Brompton required four days. A short warm spell followed by two days of hardening reinstated their high frost resistance, but the relatively high sensitivity of these plums to frost made them lose this resistance again after only one day's exposure to warm weather: they froze at 24 °C.4. Representatives of North American plum varieties such as Prunus americana and Assiniboine showed the highest resistance to frost. After two days of hardening they survived –20 °C undamaged, a temperature of –25 °C after four days. Their resistance to cold was more stable than that of Prunus cerasifera and of the hexaploid strains.5. An attempt was made to group the plums tested, using the characteristics hardening and loss of hardening as criteria.6. Two methods for improving frost resistance in plums are suggested to breeders: 1. The production of progeny from hexaploid forms that show adequate resistance to frost; 2. Interspecific hybridization with North American species known to be frost resistant.7. New means for the breeding of plum rootstocks must be used, and hardy diploid species are particularly suitable for this purpose.

Delicious apple (Malus domestica Borkh.) and several of its strains, which have been difficult to root in vitro, were successfully propagated with rooting percentages up to 100%. The combination of treatments used to achieve this result included placing the shoots on rooting medium in the dark at 30°C for the first week of the rooting stage, then moving them to a regime of 16 hr light-8 hr dark at 25°C. The rooting medium contained half strength Murashige and Skoog salts plus 1.2 M thiamine HCl, 0.56 mM myo-inositol, 1 mM phloroglucinol (PG), 1.4 M indolebutyric acid (IBA), 1.3 M gibberellic acid (GA3), 87.6 mM sucrose, and 7 g l–1 Difco Bacto agar. Dark treatment applied during the proliferation stage (etiolation) was less effective than one applied at the beginning of the rooting stage. The optimum length of dark treatment during rooting was 4 to 7 days. Increasing the temperature from 25°C to 30°C improved rooting of Delicious, Royal Red Delicious, and Vermont Spur Delicious in the absence of PG but generally had less effect in the presence of PG. Further increase in temperature to 35°C stimulated rooting of Royal Red Delicious but reduced rooting of Vermont Spur Delicious. Transfer of the cuttings to auxin-free medium after 1 week had no effect on percentage rooting and increased the number of roots per cutting for only 1 of 4 cultivars tested and then only in the presence of PG. In general PG stimulated rooting of Delicious and its strains, but had no effect on Golden Delicious.  相似文献

Two types of lagoon which occur along the West Coast of Africa described as open and closed, were studied using oceanographic methods. The parameters were temperature, salinity, dissolved oxygen and productivity. It was observed that rainfall, evaporation and the tides have a combined effect on the variations in the temperature and the salinity. The opening of the lagoons into the sea affects the level of the productivity and the rate at which salinity increases with time.Minimum temperatures at the heads of the lagoons were found to be lower than those near the ocean, while the maximum temperatures were higher at the heads. During the dry period, due to continuous evaporation of water, the salinity in the closed lagoon usually rises steeply, while those in the open lagoon rise gently.The levels of dissolved oxygen concentration in the open lagoon are higher than those in the closed lagoon. This is the result of the fact that phytoplankton count in the open lagoon is higher than that in the closed lagoon. There is also a greater diversity of phytoplankton and more live phytoplankton cells per cubic centimetre of water in the open lagoon than in the closed lagoon.As a result of all this, the main organic productivity in the open lagoon is higher than those in the closed lagoon.  相似文献

Effects of the combination of low temperature and silicone treatment on the storage of grape callus (Vitis vinifera L. x V. labrusca L. cv. Kyoho; V. vinifera L. cv. Koshusanjaku) were examined. In Kyoho, the calli were stored at 10°C successfully for up to 360 days. Embryogenic calli of Koshusanjaku stored at 10°C retained the ability of embryogenesis after 360 days of storage. However, the color of both calli became brownish. This was improved by the combination of low temperature and silicone treatment. The calli of Kyoho survived by the storage under the combination of 15°C and silicone. Embryogenic calli stored at 10 and 15°C in combination with silicone survived for 360 days, and regenerated only after transfer onto a regeneration medium. Thus the combination of low temperature and silicone affects the longevity of the grape callus.  相似文献

The response of effective quantum yield of photosystem 2 (F/Fm) to temperature was investigated under field conditions (1 950 m a.s.l.) in three alpine plant species with contrasting leaf temperature climates. The in situ temperature response did not follow an optimum curve but under saturating irradiances [PPFD >800 µìmol(photon) m–2s–1] highest F/Fm occurred at leaf temperatures below 10°C. This was comparable to the temperature response of antarctic vascular plants. Leaf temperatures between 0 and 15°C were the most frequently (41 to 56%) experienced by the investigated species. At these temperatures, F/Fm was highest in all species (data from all irradiation classes included) but the species differed in the temperature at which F/Fm dropped below 50% (Soldanella pusilla >20°C, Loiseleuria procumbens >25°C, and Saxifraga paniculata >40°C). The in situ response of F/Fm showed significantly higher F/Fm values at saturating PPFD for the species growing in full sunlight (S. paniculata and L. procumbens) than for S. pusilla growing under more moderate PPFD. The effect of increasing PPFD on F/Fm, for a given leaf temperature, was most pronounced in S. pusilla. Despite the broad diurnal leaf temperature amplitude of alpine environments, only in S. paniculata did saturating PPFD occur over a broad range of leaf temperatures (43 K). In the other two species it was half of that (around 20 K). This indicates that the setting of environmental scenarios (leaf temperature×PPFD) in laboratory experiments often likely exceeds the actual environmental demand in the field.This revised version was published online in March 2005 with corrections to the page numbers.  相似文献

Thidiazuron [(TDZ)N-phenyl-N-1,2,3-thidiazol-5-ylurea] at 750 M was applied to buds of apple trees to determine if it could substitute for the chilling requirement to induce bud break. Shoots of cv. Anna (low chill), Delicious cv. Redchief (medium chill), and Northern Spy (high chill) were untreated, treated with TDZ prior to chilling (before-chill), and treated with TDZ at various intervals after the accumulation of specific amounts of chilling (after-chill). Shoots were stored in a cold room at 4°C. TDZ applied prior to chilling reduced the chill unit (CU) requirement (1 CU = 1 h at 4°C) for the promotion of bud break on 1-year-old shoots of Anna and Northern Spy and 1- and 2-year-old wood of Delicious. TDZ applied after-chill promoted bud break only for Anna and buds on 2-year-old wood of Delicious. While accumulating CUs, untreated buds or buds treated with TDZ on 1-year Delicious and Northern Spy did not respond to the cold treatment even after 1848 h of CU accumulation. For all three cultivars, TDZ treatment was more effective in promoting bud break when applied before the initiation of chilling.The use of a company or product name does not constitute an endorsement by USDA or the University of Maryland nor imply approval to the exclusion of other suitable products.  相似文献

Chlorine, chlorine dioxide (ClO2), and a commercial raw fruit and vegetable sanitizer (Fit powder) were evaluated for their effectiveness in killing vegetative cells and spores of Bacillus cereus and spores of Bacillus thuringiensis. The ultimate goal was to use one or both species as a potential surrogate(s) for Bacillus anthracis in studies that focus on determining the efficacy of sanitizers in killing the pathogen on food contact surfaces and foods. Treatment with alkaline (pH 10.5–11.0) ClO2 (200 mg/mL) produced by electrochemical technologies reduced populations of a five-strain mixture of vegetative cells and a five-strain mixture of spores of B. cereus by more than 5.4 and more than 6.4 log cfu/mL, respectively, within 5 min. This finding compares with respective reductions of 4.5 and 1.8 log cfu/mL resulting from treatment with 200 mg/mL chlorine. Treatment with a 1.5% acidified (pH 3.0) solution of Fit powder product was less effective, causing 2.5-log and 0.4-log cfu/mL reductions in the number of B. cereus cells and spores, respectively. Treatment with alkaline ClO2 (85 mg/mL), acidified (pH 3.4) ClO2 (85 mg/mL), and a mixture of ClO2 (85 mg/mL) and Fit powder product (0.5%) (pH 3.5) caused reductions in vegetative cell/spore populations of more than 5.3/5.6, 5.3/5.7, and 5.3/6.0 log cfu/mL, respectively. Treatment of B. cereus and B. thuringiensis spores in a medium (3.4 mg/mL organic and inorganic solids) in which cells had grown and produced spores with an equal volume of alkaline (pH 12.1) ClO2 (400 mg/mL) for 30 min reduced populations by 4.6 and 5.2 log cfu/mL, respectively, indicating high lethality in the presence of materials other than spores that would potentially react with and neutralize the sporicidal activity of ClO2.Published by permission of the International Association for Food Protection: Journal of Food Protection (2004) 60:1702–1708This revised version was published online in April 2005 with corrections to the text and the section heading.In section Preparation of treatment solutions the phrase 22-28°C was replaced by 22±2°C.  相似文献

Summary Detached pistils of the clonal variety, Lilium longiflorum Aral No. 5, were submerged before pollination in 50°C water for 0, 1, 2, 3, 4, 5, 6 or 7 min and then immediately compatibly and incompatibly pollinated. Incompatibility, as indicated by pollen tube length after 48 h at 23.5°C, was eliminated by a 1–2 min submersion while compatibility was removed by a 4–5 min one. The window of incubation temperatures at which incompatible and compatible pollen tubes are clearly differentiated occurred between 15 and 30°C.  相似文献

Cryopreservation has been recognized as a practical and efficient tool for long-term storage of vegetatively propagated plants. This study was conducted to investigate effects of modified vitrification techniques on cryopreservation of potato. In vitro plants of potato cultivars Superior and Atlantic were cold acclimated, and axillary buds were precultured, osmoprotected, exposed to PVS-2 solution, plunged into liquid nitrogen, thawed, and finally planted in the regeneration medium. In the modified vitrification technique an ice-blocking agent, Supercool X1000, was added with PVS-2 solution. Cold acclimation affected survival of cryopreserved shoot tips, and the highest survival (46.7%) was obtained after 3 weeks of acclimation at 10°C. Shoot tips exposed to 2M glycerol plus 0.6M sucrose for 40 min gave 51.5% and 11.7% survival in Atlantic and Superior at 10°C, respectively. Cold acclimated and osmoprotected shoot tips were dehydrated with PVS-2 containing different concentrations of Supercool X1000 prior to a plunge into liquid nitrogen. Treatments with 0.1% and 1% of Supercool X1000 significantly improved survival by 55% in Superior and 71.3% in Atlantic, respectively. After cryopreservation, vitrified shoot tips resumed growth within a week in a medium (1 mg l–1 GA3, 0.5 mg l–1 zeatin, and 0.1 mg l–1 IAA) with a low level of Pluronic F-68 (0.005%) and survival was 33.7% higher in Atlantic and 14.7% higher in Superior than the control (without Pluronic F-68).  相似文献

Summary The ability of a piscivorous bird, the western reef heron,Egretta gularis schistacea, to cope with light refraction at the air/water interface was investigated. The heron's capture rate of small stationary underwater prey from a variety of angles was high, indicating an ability to correct for refraction. Two distinct phases were described during head movement: i) Pre-strike (mean path angle 60° to the vertical, mean velocity 52 cm/s). ii) Strike (mean path angle 33°, mean velocity 270 cm/s). When prey was unsubmerged the two phases were less distinct (pre-strike 47°, 62 cm/s; strike 42°, 154 cm/s). The point of change between phases (STR) was assumed to be the point at which corrections for refraction were performed. Calculated disparities between real and apparent prey positions at STR may reach 10 cm. At STR, highly significant correlations were found between i) the heron's eye height above the water, and prey depth, ii) the apparent prey depth and real prey depth.
