Abstract: The characteristics of β-alanine transport at the blood-brain barrier were studied by using primary cultured bovine brain capillary endothelial cells. Kinetic analysis of the β-[3H]alanine transport indicated that the transporter for β-alanine functions with Kt of 25.3 ± 2.5 µ M and J max of 6.90 ± 0.48 nmol/30 min/mg of protein in the brain capillary endothelial cells. β-[3H]Alanine uptake is mediated by an active transporter, because metabolic inhibitors (2,4-dinitrophenol and NaN3) and low temperature reduced the uptake significantly. Furthermore, the uptake of β-[3H]alanine required Na+ and Cl in the external medium. Stoichiometric analysis of the transport demonstrated that two sodium ions and one chloride ion are associated with one β-alanine molecule. The Na+ and Cl-dependent uptake of β-[3H]alanine was stimulated by a valinomycin-induced inside-negative K+-diffusion potential. β-Amino acids (β-alanine, taurine, and hypotaurine) inhibited strongly the uptake of β-[3H]alanine, whereas α- and γ-amino acids had little or no inhibitory effect. In ATP-depleted cells, the uptake of β-[3H]alanine was stimulated by preloading of β-alanine or taurine but not l -leucine. These results show that β-alanine is taken up by brain capillary endothelial cells, via the secondary active transport mechanism that is common to β-amino acids.  相似文献

Abstract An alkylhydroxylamine N -acetyltransferase was partially purified from extracts of Kelbsiella pneumoniae . N 6-Hydroxylysine, N 5-hydroxyornithine and N -methylhydroxylamine were acetylated with decreasing velocity. N6-Acetyl- l -lysine showed noncompetitive inhibition while l -ornithine, l -lysine, their α-acetyl derivatives and N 5-hydroxylysine were without effect at 0.1 M. A sensitive assay using 14C-labelled acetyl coenzyme A was developed.  相似文献

This study investigated the impact of lipid extraction, CaCO3 removal and of both treatments combined on fish tissue δ13C, δ15N and C:N ratio. Furthermore, the suitability of empirical δ13C lipid normalization and correction models was examined. δ15N was affected by lipid extraction (increase of up to 1·65‰) and by the combination of both treatments, while acidification alone showed no effect. The observed shift in δ15N represents a significant bias in trophic level estimates, i.e. lipid-extracted samples are not suitable for δ15N analysis. C:N and δ13C were significantly affected by lipid extraction, proportional to initial tissue lipid content. For both variables, rates of change with lipid content (ΔC:N and Δδ13C) were species specific. All tested lipid normalization and correction models produced biased estimates of fish tissue δ13C, probably due to a non-representative database and incorrect assumptions and generalizations the models were based on. Improved models need a priori more extensive and detailed studies of the relationships between lipid content, C:N and δ13C, as well as of the underlying biochemical processes.  相似文献

Abstract The srnB + gene located on the F plasmid was assayed for its capacity to facilitate the release from infected cells of phage λ lacking the usual lytic activity. The srnB + plasmid pOY54, carrying the 1.4–2.5F fragment in the Eco RI- Bam HI fragment of pBR322, induced bacteriolysis and the release of progeny phage of the λcI 857 susS 7 lysogen in the presence of rifampin at 42°C. An srnB 1 mutant plasmid, pOY541, did not promote bacteriolysis. These results suggest that the srnB + gene of the F plasmid complements the function of the λ S gene in the nonpermissive host strain.  相似文献

SYNOPSIS. A reduction in the growth temperature of Tetrahymena pyriformis strain WH-14 from 35 C to 15 C resulted in distinct alterations in the fatty acid composition of the glycerophospholipids. The proportion of normal saturated acids declined from 26 to 19%; palmitoleic acid increased by 6%, and the composition of the polyunsaturated fatty acids increased in 18:2 Δ6,11(n) and decreased in 18:2 Δ9,12(n) and 18:3 Δ6,9,12(n). The unsaturation index (the average number of double bonds/100 molecules) did not change with a shift in temperature.
Two biosynthetic pathways exist in Tetrahymena for the formation of unsaturated fatty acids. The observed changes in fatty acid composition that accompany a lowering of the environmental temperature can be accounted for by a reduction in the accumulation of products of the fatty acid pathway leading to the formation of γ-linolenic acid [16:0(n) → 18:0(n) → 18:1 Δ9(n) → 18:2 Δ9,12(n) → 18:3 Δ6,9,12(n)] and an increase in the components of the pathway leading to the formation of 18:2 Δ6,11(n) [16:0(n) → 16:1 Δ9(n) → 18:1 Δ11(n) → 18:2 Δ6,11(n)]. The data suggest that the regulatory mechanism in Tetrahymena differs from that found in some bacteria where a simple substitution of unsaturated fatty acids for saturated fatty acids occurs at low culture temperatures.  相似文献

The role of methionine as a precursor in mugineic acid (MA) biosynthesis was studied by feeding 15N-ammonium sulfate, 14C-amino acids, and [1-14C, 15N]-methionine to iron-deficient barley roots ( Hordeum vulgare L. cv. Minorimugi), grown hydroponically. The incorporation of isotopes into amino acids was also examined. Methionine appears to be the most efficient precursor of the mugineic acid family (MAs) of phytosiderophores; homoserine was also incorporated into the MAs, but other amino acids such as glutamate, alanine, and γ-amino butyric acid did not act as precursors of MAs. Carbon-14 and 15N of methionine were incorporated into MAs. This specific incorporation of 14C and 15N indicated that the nitrogen atoms of MAs were derived from two molecules of methionine. It is suggested that deoxymugineic acid (DMA) is probably the first phytosiderophore to be synthesized on the biosynthetic pathway of MAs.  相似文献

Quantitative estimates of soil C input under ambient (35 Pa) and elevated (60 Pa) CO2-partial pressure (pCO2) were determined in a Free-Air Carbon dioxide Enrichment (FACE) experiment. To facilitate 13C-tracing, Trifolium repens L. was grown in a soil with an initial δ13C distinct by at least 5‰ from the δ13C of T. repens grown under ambient or elevated pCO2. A shift in δ13C of the soil organic C was detected after one growing season. Calculated new soil C inputs in soil under ambient and elevated pCO2 were 2 and 3 t ha–1, respectively. Our findings suggest that under elevated CO2 conditions, soil C sequestration may be altered by changes in plant biomass production and quality.  相似文献

Abstract: Methyl 3β-(4-[125I]iodophenyl)tropane-2β-carboxylate ([123I]β-CIT) is a single photon emission computed tomographic radiotracer for in vivo labeling of dopamine (DA) and serotonin (5-HT) transporters. Single photon emission computed tomographic experiments in nonhuman primates showed that [123I]β-CIT in vivo binding to DA transporters had a much slower washout than binding to 5-HT transporters. This observation was not predicted from previously published in vitro studies. These studies, performed at 22°C in nonphysiological buffer, reported similar affinity of [125I]β-CIT for DA and 5-HT transporters. We now report [125I]β-CIT binding parameters to fresh rat membranes at 22°C and 37°C, in a buffer mimicking the composition of cerebrospinal fluid. At both temperatures, binding to DA transporters was best fit by a twosite model, whereas binding to 5-HT transporters was compatible with one population of sites. At 22°C, [125I]β-CIT showed similar affinity to high-affinity DA (0.39 n M ) and 5-HT transporter sites (0.47 n M ). Increasing the incubation temperature from 22°C to 37°C reduced binding to DA transporters by 60%, whereas binding to 5-HT transporters was only marginally affected. In vitro kinetic experiments failed to detect significant differences in on or off rates that could explain the observed in vivo kinetics. These experiments thus failed to explain [123 I]β-CIT in vivo uptake kinetics, suggesting the existence of specific factors affecting the in vivo situation.  相似文献

Water deficit and high temperature often occur simultaneously, but their effects on plants are usually investigated separately. The aim of this study was to test how interactions between water stress and nocturnal warming affect carbon allocation in the perennial grass, Leymus chinensis . Plant biomass, dry mass allocation, 14C partitioning and carbon isotope composition (δ13C) were measured. Severe and extreme water stress during nocturnal warming decreased the allocation of dry mass and 14C partitioning below ground to the roots, but moderate water stress significantly increased the below-ground allocation of dry mass and 14C, especially at the lower night temperature. The δ13C values were more positive at day/night temperatures of 30/20°C than at 30/25°C, and greater in the roots than in the leaves. By plotting the δ13C values of the leaves against the δ13C values of the roots, the slopes of regressions were steeper at low than at high night temperature, also indicating that nocturnal warming reduces carbon allocation below ground to the roots. The results suggest that nocturnal warming may weaken acclimation during water stress in this species by regulating carbon allocation between source and sink organs.  相似文献

In contrast to the light-promoted uptake by mesophyll cells, light slightly inhibited sucrose uptake by stripped leaf disks of Commelina benghalensis L. This phenomenon appeared to result from a light-promoted vein-associated release, as light stimulated photosynthate release from stripped disks and inhibited that from mesophyll cells. In the -presence of the resorption-blocker p -chloromercuriphenylsulfonic acid, (PCMBS) the release of preloaded [14C]-sugars (sucrose, glucose) and [14C]-amino acids (alanine, asparagine, proline, valine, α-aminoisobutyric acid) from stripped disks was doubled in the light. Illumination enhanced by 20 to 60% the release of endogenous leaf cell compounds (sucrose, H2PO-4, K+, Mg2+, Ca2+) from stripped disks in the presence of PCMBS. Light also increased the export of [14C]-assimilates from intact leaves by 20% after pulse-labelling with 14CO2. A model for loading is proposed, based on the differential light sensitivities of the plasma membranes in the mesophyll-to-sieve tube path.  相似文献

The growth and survival of pathogenic and non-pathogenic strains of Escherichia coli was determined in traditionally fermented pasteurized and unpasteurized milk and in Lacto, an industrially fermented milk. Each milk treatment was incubated at 20C for 24 h and then stored at either 20C or 5C for 96 h.
Lacto inhibited all the three E. coli strains. Two strains could not be recovered and the third survived only in very low numbers after 24 h storage of Lacto at both 20C and 5C. All three E. coli strains survived and multiplied to maximum cell numbers in the range 107-109/ml during traditional fermentation of unpasteurized milk. Cell numbers decreased to 103-106 and 102-105 during storage of the fermented product at 20C and 5C respectively. Higher maximum numbers, 109-1010, of the three strains of E. coli were attained during traditional fermentation of pasteurized milk. The numbers decreased to 105-108 and 104-107 during storage of the fermented product at 20C and 5C respectively. Generally, fewer E. coli survived when the fermented milk products were stored at refrigeration temperature.  相似文献

So far, at least eight alleles in the goat CSN2 locus have been associated with the level of β -casein expression in milk. Alleles CSN2 A , CSN2 A 1, CSN2 B , CSN2 C , CSN2 D and CSN2 E have been associated with normal content (allele effects of about 5 g of β -casein per litre), whereas the CSN2 0 and CSN2 01 alleles have been associated with non-detectable levels of β -casein. Most of these alleles have been characterized genetically. Herein, we report the identification of a previously unreported SNP in the goat CSN2 promoter region ( AJ011018 :g.1311T>C), which is associated with the absence of β -casein in the milk. Furthermore, we developed a PCR-based method that allows detection of this mutation.  相似文献

Z. DRICI-CACHON, J.F. CAVIN AND C. DIVIÈS. 1996. This study is concerned with the fatty acid composition of three strains of Leuconostoc oenos grown at different pH. The most abundant fatty acids were C18: 1 w 9, C19: 0 cy( w 9,10) and C16:0, followed by C16: 1 w 9 and C14: 0. The pH considerably modified the fatty acid distribution in Lo107 (an acidophilic strain) and Lo8413 (a moderately acidophilic strain). However, moderate changes occurred for LoATCC 23277 (a less acidophilic strain). At pH 2.9, Lo107 has a remarkably high level of C19: 0 cy-( w 9,10) and C19:0 cy( w 11,12). Proportions of C18: 1 and C19:0 cyclo acids varied mainly with the pH of the medium and also as a function of growth phase. The degree of unsaturation of fatty acids also varied with pH.  相似文献

Abstract: The accumulation and utilization of [35S]3'-phos-phoadenosine 5'-phosphosulfate (PAPS) were studied in slices from rat cerebral cortex incubated in the presence of inorganic [35S]sulfate. [35S]PAPS levels were directly evaluated after either isolation by ion-exchange chromatography or quantitative enzymatic transfer of its active [35S]sulfate group to an acceptor phenol under the action of added phenolsulfotransferase activity. [35S]PAPS formation was also indirectly followed by incubating slices in the presence of β-naphthol and measuring the levels of [35S]β-naphthyl sulfate ([35S]β-NS). Whereas [35S]PAPS levels rapidly reached a plateau, [35S]β-NS formation proceeded linearly with time for at least 1h, an observation indicating that the nucleotide was continuously synthesized and utilized for endogenous sulfation reactions. [35S]PAPS formation in ices was completely and rather potently blocked by 2,6-dichloro-4-nitrophenol (IC50= .10 μM), an inhibitor of the PAPS-synthesizing enzyme system in a cytosolic preparation. [35S]PAPS accumulation and [35S]β-NS'formation were strongly reduced by depolarizing agents such as potassium or veratridine. At millimolar concentrations, various excitatory amino acids (glutamate, aspartate, cysteate, quisqualate, and homocysteate) also elicited similar effects, whereas kainate and N -methyl-D-aspartate were inactive. This suggests that PAPS synthesis is turned off when cerebral cells are strongly depolarized.  相似文献

We used stable carbon (δ13C) and nitrogen (δ15N) isotopes to assess the importance of benthic algae for the zooplankton individual growth in winter in a shallow, clear subarctic lake. The δ13C values of calanoid ( Eudiaptomus graciloides ) and cyclopoid ( Cyclops scutifer ) zooplankton in autumn suggest a food resource of pelagic origin during the ice-free period. The zooplankton δ13C values were high in spring compared to autumn. E. graciloides did not grow over winter and the change in δ13C was attributed to a decrease in lipid content during the winter. In contrast, the increase in δ13C values of C. scutifer over the winter was explained by their growth on organic carbon generated by benthic algae. The δ15N of the C. scutifer food resource during winter was low compared to δ15N of the benthic community, suggesting that organic matter generated by benthic algae was mainly channelled to zooplankton via 15N-depleted heterotrophic bacteria. The results demonstrate that benthic algae can sustain zooplankton metabolic demands and growth during long winters, which, in turn, may promote zooplankton growth on pelagic resources during the summer. Such multi-chain omnivory challenges the view of zooplankton as mainly dependent on internal primary production and stresses the importance of benthic resources for the productivity of plankton food webs in shallow lakes.  相似文献

We evaluated diurnal and seasonal patterns of carbon isotope composition of leaf dark-respired CO2 ( δ 13Cl) in the C3 perennial shrub velvet mesquite ( Prosopis velutina ) across flood plain and upland savanna ecosystems in the south-western USA. δ 13Cl of darkened leaves increased to maximum values late during daytime periods and declined gradually over night-time periods to minimum values at pre-dawn. The magnitude of the diurnal shift in δ 13Cl was strongly influenced by seasonal and habitat-related differences in soil water availability and leaf surface vapour pressure deficit. δ 13Cl and the cumulative flux-weighted δ 13C value of photosynthates were positively correlated, suggesting that progressive 13C enrichment of the CO2 evolved by darkened leaves during the daytime mainly resulted from short-term changes in photosynthetic 13C discrimination and associated shifts in the δ 13C signature of primary respiratory substrates. The 13C enrichment of dark-respired CO2 relative to photosynthates across habitats and seasons was 4 to 6‰ at the end of the daytime period (1800 h), but progressively declined to 0‰ by pre-dawn (0300 h). The origin of night-time and daytime variations in δ 13Cl is discussed in terms of the carbon source(s) feeding respiration and the drought-induced changes in carbon metabolism.  相似文献

The natural abundance of 13C and 15N was measured in basidiocarps of at least 115 species in 88 genera of ectomycorrhizal, wood-decomposing and litter-decomposing fungi from Japan and Malaysia. The natural abundance of 13C and 15N was also measured in leaves, litter, soil and wood from three different sites. 15N and 13C were enriched in ectomycorrhizal and wood-decomposing fungi, respectively, relative to their substrates. Ectomycorrhizal and wood-decomposing fungi could be distinguished on the basis of their δ13C and δ15N signatures. Although there was high variability in the isotopic composition of fungi, the following isotope- enrichment factors (ε, mean±SD) of the fungi relative to substrates were observed:
εectomycorrhizal fungi/litter = 6.1±0.4‰15N
εectomycorrhizal fungi/wood = 1.4±0.8‰13C
εwood-decomposing fungi/wood = −0.6±0.7‰15N
εwood-decomposing fungi/wood = 3.5±0.9‰13C

SUMMARY. The oxygen consumption of shrimps ranging from 1 to 30 mg dry mass was determined at 18, 24 and 30°C using a continuous flow recording respirometer based upon a Clark-type oxygen electrode. Respiration (ascribed to routine metabolism) is described by the power curve: R = a Mb , ( R =μg O2 h−1, M = mg dry mass), which gives values of a = 1.632, 2.564 and 4.181, and b = 0.800, 0.898, and 0.793, at 18, 24 and 30°C respectively. The single expression, R = 0.008 T 1.829 M 0.830 provides a reasonable prediction of respiration as a combined function of shrimp size ( M ) and temperature (T, °C). Using an energy equivalent of 14.14 J mg O2−1 estimates of the energy requirements ( E , J h−1 10−3) of routine metabolism are given by the expression: E = 0.115 T 1.829 M 0.830.
Variability in oxygen consumption values between individuals is discussed and the observations on C. nilotica are compared with other crustacean studies.  相似文献

Ethanol and the γ-Aminobutyric Acid-Benzodiazepine Receptor Complex   总被引:3,自引:2,他引:1  
Abstract: Ethanol appears to enhance γ-aminobutyric acid (GABA)-mediated synaptic transmission. Using radioligand binding techniques, we investigated the possibility that the GABA-benzodiazepine receptor complex is the site responsible for this effect. Ethanol at concentrations up to 100 m M failed to alter binding of [3H]flunitrazepam (FNZ), [3H]Ro 15-1788, or [3H]methyl-γ-carboline-3-carboxylate (MBCC) to benzodiazepine receptors, or of [3H]muscimol to GABA receptors in rat brain membranes. Scatchard analyses of the binding of these radioligands at 4°C and 37°C revealed no significant effects of 100 m M ethanol on receptor affinity or number. A variety of drugs as well as chloride ion increased binding of [3H]FNZ and/or [3H]muscimol, but these influences were not modified by ethanol. These findings indicate that ethanol probably potentiates GABAergic neurotransmission at a signal transduction site beyond the GABA-benzodiazepine receptor complex.  相似文献
