Germination of whole barley seeds for 4 and 6 days followed by measurement of lysophospholipase (lysolecithin acyl hydrolase, LAH) in the embryo-containing and embryo-free halves revealed a gradient of activity between the two halves of the seed. Most of the activity appeared in the embryo-containing half. This gradient decreased slightly in the aleurone and dramatically in the starchy endosperm during the 2 day germination interval. Embryo-containing and embryo-free half seeds of surface sterilized barley were placed separately on sterile agar plates. After 4 and 6 days LAH was observed in both the aleurone and starchy endosperm of the embryo-containing halves. In the embryo-free halves, LAH appeared at low levels in the aleurone and was virtually absent in the starchy endosperm. The scutellum of germinating seeds contains LAH activity. Exposure of embryo-free half seeds to GA3 for 24 hr showed enhancement of acidic and alkaline LAH activities in the aleurone fraction and in the GA3-medium in which the half seeds were treated. The LAH activity of the starchy endosperm of these half seeds was little changed by GA3 treatment. Exposure of isolated aleurones to GA3 for 24 hr resulted in substantial enhancement of acidic and alkaline LAH activities in the bathing medium and in fractions prepared from the aleurone. The physiological significance of the influence of GA3 on LAH activity during barley germination is discussed.  相似文献

Lysophospholipase was measured in extracts of germinating barley by determining the amount of free [14C]palmitate released from [1-14C] 1-palmitoyl-lysophosphatidylcholine (LPC). Soluble and particulate lysophospholipase activity was measured at 1-day intervals in extracts from the aleurone and endosperm of barley seeds germinated for 8 days. The soluble and particulate activities of the aleurone increase approximately in parallel with one another and after 8 days of germination have 20–30 times more activity than at day 1. The activity profiles and the distribution of the activity between the soluble and particulate forms of lysophospholipase in the endosperm are markedly different. With the exception of the first 2 days when the aleurone activity is low, the endosperm activity is less than that associated with the aleurone. The soluble activity increases during the first 3 days and is more active than that of the aleurone. Thereafter it diminishes and remains low. The particulate enzyme, however, increases dramatically between days 4 and 5 and remains moderately high. The fourth and fifth day represent that stage of germination when starch-bound LPC is released in concert with the increase in amylase activity. It is proposed that it is this particulate form of the endosperm activity which may be responsible for maintaining the level of free LPC low in the endosperm of the germinating seed.  相似文献

Endosperms of quiescent barley grains contained, on average, 54.5 μg of neutral glyceride-glycerol, equivalent to ca 480 μg glyceride. Of this probably 90% was located in the aleurone layer. During germination the level of glyceride-glycerol declined. It also declined in degermed grains and aleurone layers incubated in vitro. The fall was accelerated by GA3, but indoleacetic acid, kinetin and glutamine were without effect. Increases in the levels of malate synthase and isocitrate lyase from very low initial values, and the results of incorporation studies with [14C]-labelled substrates, indicate that the glyoxylate cycle functions to convert glycerides to sucrose in germinating grains and degermed grains incubated with GA3, but not in degermed grains without the hormone. In the absence of GA3 the glyceride could be a respiratory substrate in degermed grains. The aleurone layers converted exogenous glucose to sucrose. Little label from [14C]-amino acids appeared in sucrose but in some cases considerable incorporation occurred into glutamine.  相似文献

Onion (Allium cepa) cell walls were fractionated by successive extraction with oxalate-citrate buffer and with alkali. The substantial oxalate-citrate extracted fraction comprised a range of pectic polysaccharides with varying proportions of neutral side-chains. Methylation analysis of the alkali extract indicated that (1,4′)-linked galactans and a substituted xyloglucan were probably major components. Onions thus resemble dicotyledonous plants more than the Gramineae in their cell wall composition.  相似文献

Abstract. Immature cereal embryo development can be controlled by in vitro culture on media containing ABA, or by media of low osmotic potential. To assess the possible in vivo roles of these factors, endogenous ABA levels and water relations of embryos and grains of wheat ( Triticum aestivum L.) and barley ( Hordeum vulgare L.) were determined during development. ABA concentrations remained consistent with those required to inhibit precocious germination in vitro of early stage embryos but not of more mature embryos. With increasing maturity, a difference in water potential developed between grain and embryo, suggestive of an in vivo role for water status in controlling the development of the embryo.  相似文献

In excised stem segments of floating rice (Oryza sativa L.), as well as in intact plants, submergence greatly stimulates the elongation of internodes. The differences in the composition of cell wall polysaccharides along the highest internodes of submerged and air-grown stem segments were examined. The newly elongated parts of internodes that had been submerged for two days contained considerably less cellulosic and noncellulosic polysaccharides than air-grown internodes, an indication that the cell walls of the newly elongated parts of submerged internodes are extremely thin. In the young parts of both air-grown and submerged internodes, the relative amounts of noncellulosic polysaccharides were equal to those of -cellulose, whereas the relative amounts of -cellulose were higher than those of noncellulosic polysaccharides in the upper, old parts. In the cell-elongation zones of both air-grown and submerged internodes, glucose was predominant among the noncellulosic neutral sugars of cell wall. The relative amount of glucose in noncellulosic neutral sugars decreased toward the upper, old parts of internodes, whereas that of xylose increased.  相似文献

Cell walls of petrol-defatted non-waxy IR32 rice bran and germ were prepared by protein removal with 0.5% SDS—0.6% β-mercaptoethanol, heating the residue to 80°, and destarching with Bacillus licheniformis α-amylase. A waxy rice, IR29, had a similar cell wall composition as IR32. Principal wall sugars were arabinose, xylose, and glucose. The 0.5 M sodium or potassium hydroxide and 8 M urea preferentially extracted arabinose-, xylose- and uronic acid-rich polysaccharides but 6 M sodium hydroxide—0.81 M boric acid extracted mannose-rich polysaccharides. DEAE-cellulose BO33? chromatography of the 0.5 M sodium hydroxide extracts gave fractions of similar arabinose— xylose ratios. Proteins in the cell wall preparations had only 0.4–1.6% hydroxyproline, and were bound mainly to polysaccharides, based on disc gel electrophoresis. The preparations were autofluorescent in UV and rich in phenols, mainly ferulic acid. The cell wall preparations and their 8 M urea fractions had a softening effect on defatted waxy starch aqueous gel at 0.2–2% of the starch.  相似文献

The feasibility of producing plant cell wall polysaccharide-hydrolysing feed enzymes in the endosperm of barley grain was investigated. The coding region of a modified xylanase gene (xynA) from the rumen fungus, Neocallimastix patriciarum, linked with an endosperm-specific promoter from cereal storage protein genes was introduced into barley by Agrobacterium-mediated transformation. Twenty-four independently transformed barley lines with the xylanase gene were produced and analysed. The fungal xylanase was produced in the developing endosperm under the control of either the rice glutelin B-1 (GluB-1) or barley B1 hordein (Hor2-4) promoter. The rice GluB-1 promoter provided an apparently higher expression level of recombinant proteins in barley grain than the barley Hor2-4 promoter in both transient and stable expression experiments. In particular, the mean value for the fungal xylanase activity driven by the GluB-1 promoter in the mature grains of transgenic barley was more than twice that with the Hor2-4 promoter. Expression of the xylanase transgene under these endosperm-specific promoters was not observed in the leaf, stem and root tissues. Accumulation of the fungal xylanase in the developing grains of transgenic barley followed the pattern of storage protein deposition. The xylanase was stably maintained in the grain during grain maturation and desiccation and post-harvest storage. These results indicate that the cereal grain expression system may provide an economic means for large scale production of feed enzymes in the future.  相似文献

Free and starch-bound lysophosphatidylcholine (LPC) in germinating barley was isolated and quantified during the first 8 days of germination. During the first 4 days the starch-bound LPC remained at a relatively constant level (ca 0.4,μmol/seed) and then declined during the next 2 days to ca 0.1 μmol/seed. There appeared to be no further loss of this starch-bound lipid on further germination. The decrease in the content of starch-bound LPC is not due to the action of phospholipase C and/or D on the starch lipid because there was no corresponding accumulation of starchbound lysophosphatidic acid or monoacylglycerol. The free LPC remained relatively constant at 0.02 to 0.04 μmol/seed during the entire germination period indicating that the LPC released from the starch during days 5 and 6 is further metabolized. Amylase activity was also measured in the germinating seed and increased 20-fold between days 2 and 4 which just precedes the rapid decline in starch-bound LPC. The starch content of the seed however declined to ca one third of the original value by day 5. LPC represents 65–70 % of the starch-bound lipid phosphorus in the dry seed. Through days 5 and 6 when the loss of LPC is most rapid there is no marked change in this percentage. After 8 days, however, the LPC is only ca 30%. of the starch-bound lipid phosphorus. Ofthe two major populations of starch-bound LPC, the one bearing a linoleyl group appears to decline more rapidly during days 4–6 than does that carrying a palmitoyl group. The role of starch-bound LPC in barley development and germination is discussed.  相似文献

In order to study changes in the cell-wall composition of growing poplar cambium during the seasonal cycle, cell walls were isolated from the cambium and newly formed vascular tissues of poplar branches at three times during the year (winter, beginning of spring, end of summer). Polysaccharide material was isolated by sequential extraction of the cell walls and analysed. The principal polysaccharides identified were pectins and xylose- and glucose-containing polysaccharides, possibly xylans and (xylo)glucans. Our results indicate changes in the relative quantities of these polysaccharides during the seasonal cycle.  相似文献

Pectin lyase was superior to polygalacturonase for the extraction of onion cell wall pectic polysaccharides. Exhaustive treatment of onion tissue with pectin lyase solubilized 89% of the total uronides of the tissue. The galacturonides released from the tissue were separated into three fractions (10.7, 5.3 and 84%, in order of MW) by gel filtration on Sephadex G-100. The low MW fraction was a mixture of oligogalacturonides. High and intermediate MW fractions were purified by DEAE-Sephadex column chromatography. The intermediate MW fraction was a rhamnogalacturonan II type component which contained 3- and 3,4-linked rhamnose. Methylation analysis showed that the pectic polysaccharides of onion resembled those of potato tuber.  相似文献

We have identified the major endo-beta-1,4-xylanase (XYN-1) in the aleurone of germinating barley grain, and show that it is expressed as a precursor of Mr 61 500 with both N- and C-terminal propeptides. XYN-1 is synthesized as an inactive enzyme in the cytoplasm, and only becomes active at a late stage of germination when the aleurone ceases to secrete hydrolases. A series of processing steps, mediated in part by aleurone cysteine endoproteases, yields a mature active enzyme of Mr 34 000. Processing and extracellular release of the mature enzyme coincide with the programmed cell death (PCD)-regulated disintegration of aleurone cells. We discuss the significance of delayed aleurone cell-wall degradation by endoxylanases in relation to the secretory capacity of the aleurone, and propose a novel role for aleurone PCD in facilitating the export of hydrolases.  相似文献

The objective of this study was to examine the composition and molecular structure of the endosperm cell walls (CW) derived from barley grain grown in three environments in Canada, and differing in grain hardness, protein, and total β-glucan contents. The endosperm CW were isolated from barley, cv. Metcalfe, grown in Davidson, SK (Sample A), Hythe, AB (sample B), and Hamiota, MB (sample C). The CW were sequentially extracted with water at 65 oC, saturated Ba(OH)2, again with water at 25 oC, and 1 M NaOH, resulting in fractions designated WE65, BaE, Ba/WE, and NaE, respectively. The monosaccharide analysis indicated the presence of β-glucans, arabinoxylans, and small amounts of arabinogalactans, glucomannans, and xyloglucans. Cellulose was detected in the CW remnants. The CW of sample A, exhibiting a lower grain hardness than sample B, contained the lowest amount of β-glucans, but the highest amount of arabinoxylans and the mannose-containing polysaccharides. The CW of sample C, characterized by very high protein content in the grain, contained the highest amount of β-glucans and the lowest amount of other polysaccharides. Polysaccharides in the CW of sample B, exhibiting the highest grain hardness, were characterized by the highest weight average molecular weights (Mw). β-Glucans in the CW of Sample B showed the highest ratio of DP3/DP4 and the longest cellulosic fragments in the polymeric chains. Of the three barley samples, arabinoxylans in the endosperm CW of sample A exhibited the lowest degree of branching, the highest amount of unsubstituted Xyl residues, and the highest ratio of singly to doubly substituted Xylp. The highest water solubility of the CW of sample C was associated with the highest concentration of β-glucans, the lowest DP3/DP4 ratio, and the lowest Mw of the polymeric constituents. Arabinoxylans with the lowest amount of doubly substituted but the highest amount of unsubstituted xylose residues and long sequences of unsubstituted xylan regions were found in the NaE fractions. The NaE fractions showed a high ratio of →4)-Glcp-(1→ to →3)-Glcp-(1→ linkages and some →4)-Manp-(1→ linkages, indicating a high level of long cellulosic regions in β-glucan chains and the presence of glucomannans.  相似文献

Isogenic dormant and non-dormant barley grains provide a useful system to study the molecular mechanisms of grain dormancy and the role of plant hormones in this process. As ion fluxes are associated with dormancy-related plant hormone responses, we compared the properties of the inward rectifying potassium conductance in aleurone protoplasts isolated from dormant and non-dormant Triumph grains and in germinating Himalaya grains. Maximal conductance, voltage dependency of steady-state activation, activation and deactivation kinetics were studied in the whole-cell patch-clamp configuration. Activation and deactivation time courses were single exponential. No differences in the above described properties were found between the protoplasts isolated from non-dormant Triumph and Himalaya grains. However, the maximal conductance (corrected for cell size) in protoplasts from dormant Triumph grains was much smaller (65%), and activation time constants were much larger as compared to protoplasts from non-dormant grains. No differences were found in the deactivation kinetics in the three different types of protoplasts. The half-maximal activation potential was slightly more negative in protoplasts from dormant grains than from non-dormant grains.  相似文献
