Conditions in vitro producing high rates of evagination of the cysticercus of Taenia hydatigena were obtained with an enzyme mixture of either 100 nig trypsin or 100 mg pancreatin and 5 ml whole dog bile made up to 100 ml with Hanks' balanced salt solution (BSS). Single enzyme solutions, or whole bile, in Hanks' BSS were not efficient. A pretreatment with an acid pepsin solution had little effect, on the rate of evagination. Evagination in vivo took place in the small intestine where a large portion of the larval tissue was shed almost immediately. There was no significant relationship between the mean weight of cysticerci fed to dogs AND the mean weight of scoleces recovered after 24 hr or 20 days.  相似文献

The complex permittivity of sonicated aqueous solutions of purified dimyristoylphosphatidylcholine has been measured as a function of frequency between 3 kHz and 40 GHz. The dielectric spectrum of the samples shows two dispersion/absorption regions, one centered at about 80 MHz the other at about 20.GHz (30°C). Otherwise than in previous studies no additional dispersion/absorption process has been found at frequencies below 10 MHz.The complex dielectric spectrum of the samples is discussed with respect to the dynamical state of solvent water in solutions of single-bilayer vesicles. The main relaxation time of the solvent water, τ1 ((2πτ1)?1 ≈ 20 GHz), is smaller than that of pure water, τW, at the same temperature. This effect results from the action of internal depolarizing fields which obviously overcompensate and enhancement of τ1 due to specific solute/solvent interactions (hydration) as had been previously found with micellar solutions of lysolecithins.It cannot be excluded, that some solvent water shows unusual dynamical behaviour. If there exists a substantial amount of such motionally perturbed water, however, it must be characterized by a relaxation time close to that of the phosphorylcholine zwitterions, τ2 ((2πτ2)?1 ≈ 80 MHz).  相似文献

It has previously been demonstrated that far-red irradiation of dry Lactuca sativa L. seeds results in inhibition of subsequent germination. Although red has no effect on dry seeds, a red irradiation following a farred irradiation reverses the effect of far-red. This phenomenon is most noticeable in seeds with artificially raised levels of phytochrome in the far-red absorbing form. Qualitatively similar results have been found for the seeds of Plantago major L., Sinapis arvensis L., and Bromus sterilis L. Action spectra studies on Plantago seeds show that the action peaks for promotion and inhibition of germination of hydrated seeds are at 660 and 730 nanometers, respectively. The action spectrum for inhibition of subsequent germination following irradiation of dry seeds is qualitatively and quantitatively similar to that for hydrated seeds, with an action peak at 730 nanometers, indicating absorption by phytochrome in the far-red absorbing form. However, the action spectrum for the reversal of this far-red effect on dry seeds has a broad peak at 680 nanometers and subsidiary peaks at 650 and 600 nanometers. It is proposed that this effect is due to light absorption by the phytochrome intermediate complex meta-Fa, and that the action spectrum reflects the in vivo absorption properties of this intermediate.  相似文献

The mitochondrial cytochrome spectrum of the poky strain of Neurospora crassa and its variation during the life cycle have been analyzed. Two factors are taken into account in addition to those usually considered; absorption in the near-ultraviolet Soret region and the physical nature of the electronic transitions which give rise to the absorption bands. From this extended analysis, a hypothesis based on the chemical nature of the axial environment of the heme groups has been constructed to account for spectroscopic observations. A model has been developed that provides a biochemical mechanism by which a genetic defect in mitochondrial DNA can lead to structural defects in the axial environment of the hemes and thus to an altered cytochrome spectrum. Previously reported absences or deficiencies of cytochromes based on visible absorption spectroscopy may in some cases indicate the absence or deficiency of only the particular polypeptide subunits of a complex which are synthesized within the mitochondria. This interpretation is consistent with the various manifestations of the poky phenotype which have been observed.  相似文献

1. The pH-activity relationship of invertase has been studied in vivo and in vitro under identical external environmental conditions. 2. The effect of changing (H+) upon the sucroclastic activity of living cells of S. cerevisiae and of invertase solutions obtained therefrom has been found, within experimental error, to be identical. 3. The region of living yeast cells in which invertase exerts its physiological activity changes its pH freely and to the same extent as that of the suspending medium. It is suggested that this may indicate that this intracellular enzyme may perform its work somewhere in the outer region of the cell. 4. In using live cells containing maltase, no evidence of increased sucroclastic activity around pH 6.9, due to the action of Weidenhagen''s α-glucosidase (maltase), was found.  相似文献

In current feed evaluation systems, the nutritional value of protein sources in diets for pigs is based on the ileal digestibility of protein and amino acids, which does not account for the kinetics of protein digestion along the gastrointestinal tract. The objective of the present study was to determine the in vitro protein digestion kinetics of different protein sources (soya bean meal (SBM), wheat gluten (WG), rapeseed meal (RSM), whey powder (WP), dried porcine plasma protein, yellow meal worm larvae and black soldier fly larvae (BSF)). Protein sources were incubated with pepsin at pH 3.5 for 0 to 90 min and subsequently with pancreatin at pH 6.8 for 0 to 210 min at 39°C. The in vitro protein digestion kinetics were described as the kinetics of nitrogen (N) solubilisation and the release of low molecular weight peptides (LMW) (<500 Da). The N solubilisation rate ranged from 0.025 min−1 for BSF to 0.685 min−1 for WP during the incubation with pepsin, and from 0.027 min−1 for RSM to 0.343 min−1 for WP during the incubation with pancreatin. The release rate of LMW peptides ranged from 0.027 min−1 for WG to 0.093 min−1 for WP during the incubation with pepsin, and from 0.029 min−1 for SBM to 0.385 min−1 for WP. Black soldier fly larvae showed a similar release rate of LMW peptides as WP during the incubation with pancreatin. At the end of the sequential incubation with pepsin (90 min) and pancreatin (210 min), WG and WP showed the highest percentage of N present in LMW peptides relative to total N (78% and 79%, respectively), whereas SBM showed the lowest (35%). In conclusion, protein sources for pig diets show substantial differences in in vitro protein digestion kinetics as measured by the kinetics of N solubilisation and the release of LMW peptides. The rate of release of LMW peptides was not correlated to the rate of N solubilisation for each of the protein sources evaluated.  相似文献

Utilisation of microbial enzymes may represent an alternative strategy to the use of conventional pancreatin obtained from pig pancreas for the treatment of severe pancreatic insufficiency. In this study, we focused on the capacity of two microbial preparations for their capacity to digest alimentary proteins (caseins and soya proteins) in comparison with pancreatin. These microbial enzymatic preparations were found to be able to generate small, medium-size and larger polypeptides from caseins and soya proteins but were inactivated at pH 3.0. As determined by Liquid Chromatography–Mass Spectrometry analysis, microbial enzymes generated very different peptides from caseins when compared with peptides generated through pancreatin action. These microbial preparations were characterised by relatively low trypsin- and low carboxypeptidase-like activities but high chymotrypsin-like activities and strong capacity for cleavage of caseins at the methionine sites. Although the efficiency of these microbial preparations to increase the rate of absorption of nitrogen-containing compounds in severe pancreatic insufficiency remains to be tested in vivo, our in vitro data indicate proteolytic capacities of such preparations for alimentary protein digestion.  相似文献

1. The ions of Ca and K condition general luminescence, and are therefore necessary to the conduction of the impulse. 2. In van''t Hoff''s solution from which Mg is omitted, Berœ shows hyperirritability with respect to luminescence. This is the result of the action of Ca and K ions unantagonized by Mg. 3. The luminescent material spread on filter paper does not show luminescence in sea water, NaCl, MgCl2, or saccharose solutions isotonic with sea water. In solutions of CaCl2, SrCl2, BaCl2, KCl, and K2SO4 the indicator paper glows with a bright luminescence. 4. In dark adapted Berœ, luminescence is inhibited by a certain quantity of light. This quantity has an average value of 57,285 meter-candle-minutes, which is twelve times the value given by Mnemiopsis.  相似文献

The ability to absorb solutions has been examined in the coxal vesicles of the ventral tube in two sympatric surface dwelling Collembola (Tomocerus sp. and Orchesella villosa) from a beech forest. The net influx of distilled water and different sodium chloride solutions was measured, followed by examination of the effective surface of the vesicles which contacts the medium. The transport rate decreased with increasing salinity of the medium. Orchesella always showed higher absorption rates than Tomocerus, if the values were related to unit surface area. However, considering that the effective surface area of the vesicles of Tomocerus is larger, the total absorption rate by the ventral tube of Tomocerus exceeded that of Orchesella. Calculating the increase in the total water content. Orchesella compensated for its deficit faster than Tomocerus, because Orchesella is smaller in total weight and water content. In most cases the efficiency of the absorptive epithelium decreased during an absorption cycle; this also occurred before moulting. Some hours after moulting, the absorption rates increased to their former level. Comparing the rates of transpiration, absorption by the ventral-tube vesicles, and drinking, confirmed the dominant role of the ventral tube in the water balance of Collembola. It is an important factor in the strategy of adaptation from the hypogaic to the epigaic life.  相似文献

A polarographic oxygen determination, with tissue in direct contact with a stationary platinum electrode, has been used to measure the photosynthetic response of marine algae. These were exposed to monochromatic light, of equal energy, at some 35 points through the visible spectrum (derived from a monochromator). Ulva and Monostroma (green algae) show action spectra which correspond very closely to their absorption spectra. Coilodesme (a brown alga) shows almost as good correspondence, including the spectral region absorbed by the carotenoid, fucoxanthin. In green and brown algae, light absorbed by both chlorophyll and carotenoids seems photosynthetically effective, although some inactive absorption by carotenoids is indicated. Action spectra for a wide variety of red algae, however, show marked deviations from their corresponding absorption spectra. The photosynthetic rates are high in the spectral regions absorbed by the water-soluble "phycobilin" pigments (phycoerythrin and phycocyanin), while the light absorbed by chlorophyll and carotenoids is poorly utilized for oxygen production. In red algae containing chiefly phycoerythrin, the action spectrum closely resembles that of the water-extracted pigment, with peaks corresponding to its absorption maxima (495, 540, and 565 mµ). Such algae include Delesseria, Schizymenia, and Porphyrella. In the genus Porphyra, there is a series P. nereocystis, P. naiadum, and P. perforata, with increasingly more phycocyanin and less phycoerythrin: the action spectra reflect this, with increasing activity in the orange-red region (600 to 640 mµ) where phycocyanin absorbs. In all these red algae, photosynthesis is almost minimal at 435 mµ and 675 mµ, where chlorophyll shows maximum absorption. Although the chlorophylls (and carotenoids) are present in quantities comparable to the green algae, their function is apparently not that of a primary light absorber; this role is taken over by the phycobilins. In this respect the red algae (Rhodophyta) appear unique among photosynthetic plants.  相似文献

Action spectra derived from dose-response curves measured for various processes associated with chloroplast development in Euglena gracilis var. bacillaris are presented. The action spectrum for chlorophyll synthesis during the first 36 hours of continuous illumination of dark-grown resting cells resembles the absorption spectrum of protochlorophyll(ide). The action spectrum for the preillumination phase of potentiation, during which preillumination followed by a dark period brings about lag elimination in chlorophyll synthesis when the cells are subsequently exposed to postilluminating light, shows a high peak in the blue region (at about 433 nm) with a small peak in the yellow-orange region (at about 597 nm); the postillumination phase yields an action spectrum very similar to that obtained for chlorophyll synthesis in continuous light in normal, unpotentiated cells, with peaks at 433 and 631 nm. Alkaline DNase and TPN-linked triose phosphate dehydrogenase, two plastid enzymes which are synthesized outside the chloroplast, yield action spectra which are consistent with protochlorophyll(ide) being the major light receptor. The action spectra which implicate pigments resembling protochlorophyll(ide) holochrome have blue to red peak ratios in the vicinity of 5:1 as does the absorption spectrum of the protochlorophyllide holochrome from beans; the action spectrum is not identical with the holochrome spectrum indicating that the Euglena holochrome may differ from the bean pigment in details of its absorption spectrum. The action spectrum for preillumination, shows a ratio of the blue peak to the red effectiveness of about 24:1. This suggests that preillumination is controlled by a photoreceptor different from the protochlorophyll(ide) holochrome.  相似文献

1. The deposition of casein on a platinum anode which takes place on the passage of a direct current through solutions of alkali caseinates was quantitatively studied, and it was found that: (a) the amount of casein which is deposited is directly proportional to the current, i.e. it obeys Faraday''s law; (b) the amount of casein deposited is inversely proportional (within the limits studied) to the amount of alkali which is combined with the casein. 2. A method of determining the transport numbers of proteins insoluble at their isoelectric point has been developed. 3. A titration method for determining the amount of alkali in a casein solution is given. 4. Data from the results of transference experiments on sodium caseinate, potassium caseinate, cesium caseinate, and rubidium caseinate solutions are given. It is shown that the data are best explained on the assumption that in these solutions the carriers of the current are alkali metal cations and casein anions. 5. On the basis of our transference results an explanation is given of the results which were obtained by Robertson and by Haas in their migration experiments.  相似文献

1. A study has been made of the equilibrium existing between trypsin and the substances formed in the digestion of proteins which inhibit its action. 2. This substance could not be obtained by the hydrolysis of the proteins by acid or alkali. It is dialyzable. 3. The equilibrium between this substance (inhibitor) and trypsin is found to agree with the equation, trypsin + inhibitor ⇌ trypsin-inhibitor The equilibrium is reached instantaneously and is independent of the substrate concentration. If it be further assumed that the rate of hydrolysis is proportional to the concentration of the free trypsin and that the equilibrium conforms to the law of mass action, it is possible to calculate the experimental results by the application of the law of mass action. 4. The equilibrium has been studied by varying (a) the concentration of the inhibiting substance, (b) the concentration of trypsin, (c) the concentration of gelatin, and (d) the concentration of trypsin and inhibitor (the relative concentration of the two remaining the same). In all cases the results agree quantitatively with those predicted by the law of mass action. 5. It was found that the percentage retarding effect of the inhibiting substance on the rate of hydrolysis is independent of the hydrogen ion concentration between pH 6.3 and 10.0. 6. The fact that the experimental results agree with the mechanism outlined under 3, is contrary to the assumption that any appreciable amount of trypsin is combined with the gelatin at any one time; i.e., the velocity of the hydrolysis must depend on the time required for such a compound to form rather than for it to decompose. 7. The experiments may be considered as experimental proof of the validity of Arrhenius'' explanation of Schütz''s rule as applied to trypsin digestion. 8. Inactivated trypsin does not enter into the equilibrium.  相似文献

Action spectra for the assimilation of nitrate and nitrite have been obtained for several blue-green algae (cyanobacteria) with different accessory pigment composition. The action spectra for both nitrate and nitrite utilization by nitrate-grown Anacystis nidulans L-1402-1 cells exhibited a clear peak at about 620 nanometers, corresponding to photosystem II (PSII) C-phycocyanin absorption, the contribution of chlorophyll a (Chl a) being barely detectable. The action spectrum for nitrate reduction by a nitrite reductase mutant of A. nidulans R2 was very similar. All these action spectra resemble the fluorescence excitation spectrum of cell suspensions of the microalgae monitored at 685 nanometers—the fluorescence band of Chl a in PSII. In contrast, the action spectrum for nitrite utilization by nitrogen-starved A. nidulans cells, which are depleted of C-phycocyanin, showed a maximum near 680 nanometers, attributable to Chl a absorption. The action spectrum for nitrite utilization by Calothrix sp. PCC 7601 cells, which contain both C-phycoerythrin and C-phycocyanin as PSII accessory pigments, presented a plateau in the region from 550 to 630 nanometers. In this case, there was also a clear parallelism between the action spectrum and the fluorescence excitation spectrum, which showed two overlapped peaks with maxima at 562 and 633 nanometers. The correlation observed between the action spectra for both nitrate and nitrite assimilation and the light-harvesting pigment content of the blue-green algae studied strongly suggests that phycobiliproteins perform a direct and active role in these photosynthetic processes.  相似文献

1. The irradiation of bacteria in liquid suspension has been made possible through: (a) the use of a specially balanced physiological salt solution which is practically non-absorbing for the wave lengths used, and which is of such composition that subsequent dilution of the bacterial suspension gives the proper number of organisms; (b) special design of the exposure cell and a very thorough method of stirring which subjects each organism equally to the radiation; (c) practically complete absorption of the incident radiation, through the use of very dense suspensions, thus eliminating the necessity for a separate determination of the absorption coefficients of the bacteria for the wave lengths used. 2. The method also provides a means for determining the effects of sub-lethal doses. 3. A formula is given for calculating from observed survival ratios the energy required to inactivate bacteria with ultraviolet radiation. The formula corrects for the protective action of non-viable organisms. 4. Data are given for the inactivation of 15 hour and 240 hour cultures of E. coli, washed and unwashed) and for 6–7 hour cultures, unwashed. These data are compared with those of other investigators. 5. A possible explanation for the differences in energy required to inactivate old, young, and standard cultures of bacteria is suggested. 6. The possible mechanism of the action of ultraviolet radiation on microorganisms is discussed.  相似文献

Alzheimer's disease is a tauopathy characterized by pathological fibrillization of tau protein to form the paired helical filaments (PHFs), which constitute neurofibrillary tangles. The methylthioninium (MT) moiety reverses the proteolytic stability of the PHF core and is in clinical development for treatment of Alzheimer's disease in a stable reduced form as leuco-MT. It has been hypothesized that MT acts via oxidation of cysteine residues, which is incompatible with activity in the predominantly reducing environment of living cells. We have shown recently that the PHF-core tau unit assembles spontaneously in vitro to form PHF-like filaments. Here we describe studies using circular dichroism, SDS-PAGE, transmission electron microscopy and site-directed mutagenesis to elucidate the mechanism of action of the MT moiety. We show that MT inhibitory activity is optimal in reducing conditions, that the active moiety is the reduced leuco-MT form of the molecule and that its mechanism of action is cysteine independent.  相似文献

The bacterial virus, ΦX174, which contains a single strand of DNA has been inactivated by different wave lengths of monochromatic ultraviolet light at pH 7, 2, and 12. The action spectra for inactivation at these three pH's all showed minima at 2400 A rather than at 2300 A, which is the characteristic absorption minimum of DNA. The shapes of the action spectra have been analyzed in terms of the effects of absorbed light on the pyrimidines and purines rather than the effect on nucleoprotein. In this interpretation the pyrimidines are at least 2 to 3 times more sensitive than the purines. The quantum yield for inactivation of the virus at 2650 A and pH 7 is 0.006. The quantum efficiency for quanta absorbed in the pyrimidines is 0.0085 and for the purines 0.0035. It is pointed out that action spectra for single- and double-stranded polynucleotides should have minima at different wave lengths, and that this difference may be used to distinguish between these two configurations in vivo.  相似文献

Studies have been carried out on the movement of salt and water across the small intestine of the rat. Segments of the ileum of anesthetized rats have been perfused in vivo with unbuffered NaCl solutions or isotonic solutions of NaCl and mannitol. Kinetic analysis of movements of Na24 and Cl36 has permitted determination of the efflux and influx of Na and Cl. Net water absorption has been measured using hemoglobin as a reference substance. Water was found to move freely in response to gradients of osmotic pressure. Net water flux from isotonic solutions with varying NaCl concentration was directly dependent on net solute flux. The amount of water absorbed was equivalent to the amount required to maintain the absorbed solute at isotonic concentration. These results have been interpreted as indicating that water movement is a passive process depending on gradients of water activity and on the rate of absorption of solute. The effluxes of Na and Cl are linear functions of concentration in the lumen, but both ions are actively transported by the ileum according to the criterion of Ussing (Acta Physiol. Scand., 1949, 19, 43). The electrical potential difference between the lumen and plasma has been interpreted as a diffusion potential slightly modified by the excess of active Cl flux over active Na flux. The physical properties of the epithelial membrane indicate that it is equivalent to a membrane having negatively charged uniform right circular pores of 36 Å radius occupying 0.001 per cent of the surface area.  相似文献
