The alternating current resistance and capacity of suspensions of unfertilized and fertilized eggs of Arbacia punctulata have been measured at frequencies from 103 to 1.64 x 107 cycles per second. The unfertilized egg has a static plasma membrane capacity of 0.73 µf./cm.2 which is practically independent of frequency. The fertilized egg has a static membrane capacity of 3.1 µf./cm.2 at low frequencies which decreases to a value of 0.55 µf./cm.2 at high frequencies. The decrease follows closely the relaxation dispersion of the dielectric constant if the dissipation of such a system is ignored. It is considered more probable that the effect is due to a fertilization membrane of 3.1 µf./cm.2 capacity lifted 1.5 µ. from the plasma membrane, the interspace having the conductivity of sea water. The suspensions show a frequency-dependent capacity at low frequencies which may be attributable to surface conductance. The equivalent low frequency internal specific resistance of both the unfertilized and fertilized egg is about 186 ohm cm. or about 6 times that of sea water, while the high frequency data extrapolate to a value of about 4 times sea water. There is evidence at the highest frequencies that the current is penetrating the nucleus and other materials in the cytoplasm. If this effect were entirely due to the nucleus it would lead to a very approximate value of 0.1 µf./cm.2 for the capacity of the nuclear membrane. The measurements do not indicate any change in this effect on fertilization.  相似文献

Apparatus has been designed and constructed for the measurement of the electric impedance of suspensions of Arbacia eggs in sea water to alternating currents of frequencies from one thousand to fifteen million cycles per second. This apparatus is simple, rugged, compact, accurate, and rapid. The data lead to the conclusions that the specific resistance of the interior of the egg is about 90 ohm cm. or 3.6 times that of sea water, and that the impedance of the surface of the egg is probably similar to that of a "polarization capacity". The characteristics of this surface impedance can best be determined by measurements of the capacity and resistance of suspensions of eggs. No specific change has been found in the interior resistance or the surface impedance which can be related either to membrane formation or to cell division.  相似文献

From the low frequency alternating current impedance and the volume concentrations of suspensions of Arbacia eggs, it is shown that the high resistance membrane is either at or very near the plasma membrane for both unfertilized and fertilized eggs, and that the specific resistances of the perivitelline space and fertilization membrane are not greatly different from that of sea water. The effect of the capacity element which appears after fertilization at intermediate frequencies is considerably less than in the earlier experiments on Arbacia and Hipponoë eggs. These findings indicate that the fertilization membrane does not have the high capacity previously attributed to it and that the increase in membrane capacity takes place at or near the plasma membrane.  相似文献

The changes in the alternating current impedance which occur during activity of cells of the fresh water plant Nitella have been measured with the current flow normal to the cell axis, at eight frequencies from 0.05 to 20 kilocycles per second, and with simultaneous records of the action potential under the impedance electrodes. At each frequency the resting cell was balanced in a Wheatstone bridge with a cathode ray oscillograph, and after electrical stimulation at one end of the cell, the changes in the complex impedance were determined from the bridge unbalance recorded by motion pictures of the oscillograph figure. An extension of the previous technique of interpretation of the transverse impedance shows that the normal membrane capacity of 0.9 µf./cm.2 decreases about 15 per cent without change of phase angle, while the membrane resistance decreases from 105 ohm cm.2 to about 500 ohm cm.2 during the passage of the excitation wave. This membrane change occurs during the latter part of the rising phase of the action potential, and it is shown that the membrane electromotive force remains unchanged until nearly the same time. The part of the action potential preceding these membrane changes is probably a passive fall of potential ahead of a partial short circuit.  相似文献

Summary The chlorophyll deficient tl mutant of Nicotiana tabacum var Samsun expresses green, clear and twin, green and clear somatic variations spontaneously on leaves at a low frequency. This character is maintained after both vegetative multiplication and sexual reproduction. However a very important phenotypic variability in the capacity for somatic variation appears in in vitro bud neoformations from leaf fragments of tl/tl homozygous plants. This variability is observed in the type of variations and the variation pattern, defined as the frequency and size of the variant areas.The present work was aimed at determining both the cell frequencies of the events which lead to the somatic variation and the preferential sequence of leaf initial development during which these frequencies are at a maximum. It was limited to plant populations with very different patterns for green variations, some having a high frequency of large variation, others having a high frequency of small variations. They were compared with a population of control plants having a low frequency.In the case of plants having a high frequency of large green variations, the events leading to somatic variation occurred between the twenty-first and the twelfth cell cycles preceding the end of the initial division phase, the maximum cell frequencies being in the seventeenth and sixteenth cycles. The maximum frequencies appeared extremely high, being on average about 10–2. In plants with a high frequency of small green variations the event occurred between cell cycles nine and one, with mean frequencies of 10–3 but without any clearly marked maximum. In the low frequency control plants the event also took place during the last ten cell cycles but with decreasing frequencies from 10–4 to 10–7.The frequency and the starting period of the cell events leading to somatic variation are closely dependent on the state of the cell. This is, on the one hand, strictly linked to the physiology of the plant and, on the other, closely correlated with the stage of differentiation, which may vary according to the genetic back ground of the leaf initial cells.The results are discussed in relation to comparable observations and the relevant interpretations made on other instability mutants.  相似文献

1. The lateral line of Fundulus heteroclitus and Fundulus majalis is shown to react to tone at an intensity level of 20 dynes per sq. cm. at frequencies up to 200 or 300 cycles per second. 2. Evidence is given that the nerve can reproduce the stimulating tone frequency up to at least 180 cycles per second. 3. The response of the lateral line to the swimming movements of nearby fish is demonstrated. 4. Stem and electrode electric impedance at 14 frequencies were monitored during cold acclimation of alfalfa (Medicago sativa L.) and birdsfoot trefoil (Lotus corniculatus L.). Cold acclimation significantly increased high frequency (e.g. 1.11 megahertz) resistance and reactance but not low frequency (49 hertz) resistance and reactance of both species. High frequency resistance of living stems was equivalent to the average resistance at all frequencies of dead stems and the resistance of dead stems (y) was related to stem water content (x):y = 9.28 − 4.11x + 0.47x2, R = 0.92. The low-high frequency (49 hertz/1.11 megahertz) resistance ratio decreased during cold acclimation. A time constant believed to be a function of membrane resistance and capacitance was not affected by cold acclimation.  相似文献

Antibiotic-resistant (either to erythromycin or chloramphenicol) temperature-sensitive mutants were isolated with about the same frequency in 2 strains of the petite negative yeast K. lactis.The eryR and capR mutants isolated in the strain K. lactis CBS 2359 showed with high frequency both a lethal-conditioned (lc) or a petite temperature-sensitive (pts) phenotype, whereas amongst the many eryR and capR mutants isolated in the strain K. lactis CBS 2360 only lc phenotypes appeared. In the mutants isolated from K. lactis CBS 2360, one growth cycle in the presence of ethidium bromide irreversibly blocked the transmission of antibiotic resistance and temperature sensitivity (lc and pts), whereas at least 2 growth cycles were required to give the same results for the mutants isolated in K. lactis CBS 2359.The spontaneous reversion frequencies for the temperature sensitivity were about the same for the lc mutants isolated in the 2 strains, but the frequencies of co-reversion of the antibiotic resistance were higher in eryRlc and capRlc mutants isolated from K. lactis CBS 2360.The analysis of the effect of the exposure to erythromycin or to the temperature of 36°C on protein synthesis carried out by isolated mitochondria of 2 eryRlc mutants of K. lactis CBS 2360 and CBS 2359 showed that, in these mutants, mitochondrial protein synthesis became resistant to the drug and sensitive to temperature. The exposure at 36°C, before protein synthesis was inactived, determined in these mutants a condition of sensitivity to the antibiotic, suggesting that even though the 2 K. lactis strains differ in some aspects concerning the behaviour of their mitochondrial information they might depend, as to their petite-negative character, on the role that mitochondrial protein synthesis has in cell division.  相似文献

A weeping pulsed radar reflectometer designed for measuring the spatial electron density distribution in the Globus-M spherical tokamak with a minor plasma radius of a=24 cm, a major radius of R=36 cm, a toroidal field of B T=0.5 T, a plasma current of I p=200 kA, and an average density of n=(3–10)×1013 cm?3 is described. The reflectometer operation is based on the reflection of microwaves with a carrier frequency f from a plasma layer with the critical density n=(0.0111f)2, where n is the electron density in units of 1014 cm?3 and f is the microwave frequency in GHz. By simultaneously probing the plasma at different frequencies, it is possible to recover the electron density profile. Microwave pulses with different frequencies are obtained by frequency sweeping. To increase the range of measured densities, channels with fixed frequencies are also used; as a result, the instrument has eleven frequency channels: a 19.5-GHz channel, eight channels in the 26-to 40-GHz frequency range, a 51.5-GHz channel, and a 60-GHz channel, which corresponds to eleven points in the density profile: 0.47×1013 cm?3, eight points in the (0.8–1.95)×1013-cm?3 range, 3.27×1013 cm?3, and 4.5×1013 cm?3. The reflectometer allows detailed measurements of the density profile with a time resolution of several tens of microseconds, which can be useful, in particular, in studying the processes related to the formation of an internal transport barrier in plasma. The first results obtained using this reflectometer in the Globus-M tokamak under various operating conditions are discussed.  相似文献

Electrical impedance measurements were made upon unfertilized and fertilized eggs of the leopard frog, Rana pipiens, over a frequency range of 0.05 to 10 kc. Average values of 170 ohm cm.2 were obtained for the plasma membrane resistance of the egg, 2.0 µf/cm.2 for the plasma membrane capacity, 86° for the phase angle of the membrane, and 570 ohm cm. for the specific resistance of the interior. These values did not change upon fertilization. No spontaneous rhythmical impedance changes such as have been found by Hubbard and Rothschild in the trout egg were found in frog eggs.  相似文献

Tardigrades represent one of the main animal groups with anhydrobiotic capacity at any stage of their life cycle. The ability of tardigrades to survive repeated cycles of anhydrobiosis has rarely been studied but is of interest to understand the factors constraining anhydrobiotic survival. The main objective of this study was to investigate the patterns of survival of the eutardigrade Richtersius coronifer under repeated cycles of desiccation, and the potential effect of repeated desiccation on size, shape and number of storage cells. We also analyzed potential change in body size, gut content and frequency of mitotic storage cells. Specimens were kept under non-cultured conditions and desiccated under controlled relative humidity. After each desiccation cycle 10 specimens were selected for analysis of morphometric characteristics and mitosis. The study demonstrates that tardigrades may survive up to 6 repeated desiccations, with declining survival rates with increased number of desiccations. We found a significantly higher proportion of animals that were unable to contract properly into a tun stage during the desiccation process at the 5th and 6th desiccations. Also total number of storage cells declined at the 5th and 6th desiccations, while no effect on storage cell size was observed. The frequency of mitotic storage cells tended to decline with higher number of desiccation cycles. Our study shows that the number of consecutive cycles of anhydrobiosis that R. coronifer may undergo is limited, with increased inability for tun formation and energetic constraints as possible causal factors.  相似文献

Scrapie eradication efforts cost 18 million dollars annually in the United States and rely heavily upon PRNP genotyping of sheep. Genetic resistance might reduce goat scrapie and limit the risk of goats serving as a scrapie reservoir, so PRNP coding sequences were examined from 446 goats of 10 breeds, 8 of which had not been previously examined at PRNP. The 10 observed alleles were all related to one of two central haplotypes by a single amino acid substitution. At least five of these alleles (M142, R143, S146, H154, and K222) have been associated with increased incubation time or decreased odds of scrapie. To the best of our knowledge, neither S146 nor K222 has been found in any goats with scrapie, though further evaluation will be required to demonstrate true resistance. S146 was more common, present in several breeds at widely varying frequencies, while K222 was observed only in two dairy breeds at low frequency. Overall, this study provides frequency data on PRNP alleles in US goats, shows the pattern of relationships between haplotypes, and demonstrates segregation of multiple scrapieassociated alleles in several breeds not examined before at PRNP.  相似文献

Seven umr mutants of Saccharomyces cerevisiae which had reduced capacity for ultraviolet light (UV)-induced forward mutation from CAN1 to can1 were tested for sensitivity to L-canavanine relative to one wild-type UMR strain and one slightly UV-sensitive but phenotypically umr+ strain (mutant 306). Relative UV mutation resistance was estimated by dividing the UV fluence needed to yeild a particular induced mutation frequency by that needed to reach the same frequency in the genotypic wild-type strain. The umr5 and umr6 strains were especially sensitive to canavanine growth inhibition, while umr1 was no more sensitive than either wild type; umr2, umr3, umr4, a umr7, and α umr7 were equally sensitive to an intermediate degree. Incubation at 30°C of wildtype cells plated on canavanine-selective agar for increasingly longer times before UV irradiation resulted in decreasing UV mutation frequencies (reduced to 50% in 1.6 h). All umr strains tested in this way lost UV mutability faster than wild type, including mutant 306, umr1 (not sensitive to growth inhibition), and umr6 (very sensitive to growth inhibition). Cells were grown to stationary phase in YEDP growth medium and assayed for arginine and tryptophan transport into the cell. The umr6 strain, which had weak UV mutation resistance but high sensitivity to canavanine growth inhibition, transported arginine and tryptophan at essentially wild-type levels. The umr1 strain, however, which had moderate UV mutation resistance and normal canavanine toxicity, transported both amino acids at rates tenfold higher than wild type. The data suggest that increased canavanine toxicity does not necessarily lead to defective mutability at CAN1, and that mutational deficiency cannot result solely from increased canavanine toxicity. Although exposure to canavanine was shown to block mutation fixation and/or expression, it is suggested that the degree of growth inhibition is not strictly correlated with the degree of mutation resistance.  相似文献

Alternating current measurements have been taken on single Nitella cells over a frequency range from 30 to 2,500,000 cycles per second with the current flow perpendicular to the axis of the cell. The measuring cells were so constructed that electrolytes of any desired concentration could be circulated during the course of the measurements. The cellulose wall which surrounds the cell is found to play an important part in the interpretation of the results obtained. In a mature cell, this cellulose has a specific resistance of about 1000 ohm cm. which is independent of the medium in which the cell is suspended. The thickness of the wall is computed to be about 10 µ. The cell membrane is found to be virtually non-conducting, and to have a capacity of 0.94 µf./cm.2 ± 10 per cent and a phase angle of 80° ± 4°. The specific resistances of the sap were difficult to compute from data on living cells and were unsatisfactory because they were very much dependent upon the medium, while measurements on extracted sap gave 58 ohm cm. ± 8 per cent which was independent of the medium. There are indications that the chloroplasts have impedance properties similar to those of living cells.  相似文献

The interaction of low water potential effects on photosynthesis, and leaf K+ levels in wheat (Triticum aestivum L.) plants was studied. Plants were grown at three K+ fertilization levels; 0.2, 2, and 6 millimolar. With well watered plants, 2 millimolar K+ supported maximal photosynthetic rates; 0.2 millimolar K+ was inhibitory, and 6 millimolar K+ was superoptimal (i.e. rates were no greater than at 2 millimolar K+). Photosynthesis was monitored at high (930 parts per million) and low (330 parts per million) external CO2 throughout a series of water stress cycles. Plants subjected to one stress cycle were considered nonacclimated; plants subjected to two successive cycles were considered acclimated during the second cycle. Sensitivity of photosynthesis to declining leaf water potential was affected by K+ status; 6 millimolar K+ plants were less sensitive, and 0.2 millimolar K+ plants were more sensitive than 2 millimolar K+ plants to declining water potential. This occurred with nonacclimated and acclimated plants at both high and low assay CO2. It was concluded that the K+ effect on photosynthesis under stress was not mediated by treatment effects on stomatal resistance. Differences between the K+ treatments were much less pronounced, however, when photosynthesis of nonacclimated and acclimated plants was plotted at a function of declining relative water content during the stress cycles. These results suggest that K+ effects on the relationship between relative water content and water potential in stressed plants was primarily responsible for the bulk of the K+-protective effect on photosynthesis in stressed plants. In vitro experiments with chloroplasts and protoplasts isolated from 2 millimolar K+ and 6 millimolar K+ plants indicated that upon dehydration, K+ efflux from the chloroplast stroma into the cytoplasm is less pronounced in 6 millimolar K+ protoplasts.  相似文献

In situ mating experiments were done in the River Taff, South Wales, United Kingdom, by using a natural mercury resistance plasmid (pQM1) isolated from a mixture of epilithic bacteria in vitro. The river temperature from March to November was found to influence transfer frequencies strongly (6.8 × 10−9 to 1.5 × 10−2 per recipient). A linear relationship existed between log10 transfer frequency and river temperature (6 to 21°C), a 2.6°C change in temperature giving a 10-fold change in transfer frequency. In vitro experiments showed that pQM1 transferred most efficiently between fluorescent pseudomonads and that one epilithic isolate (Pseudomonas fluorescens) was an efficient donor in situ. Experiments with a P. putida recipient showed that intact epilithic bacterial communities could transfer mercury resistance plasmids in situ at frequencies of up to 3.75 × 10−6 per recipient. Nineteen of the large (>250-kilobase) plasmids isolated by transfer into P. putida were studied in detail and grouped into seven types by restriction digests. Mercury resistance and UV resistance were found to be common linked phenotypes in 19 of the 23 plasmids tested.  相似文献
