Atropinesterase was found to exist in approximately one out of every four rabbits, and no relation could be observed between the incidence of the enzyme and season, sex, color, age, or weight. The occurrence of the enzyme was also shown to be unrelated to that of cholinesterase. The distribution of atropinesterase in the blood and organs of rabbits was studied; the animals devoid of the enzyme in their blood contained no demonstrable activity in any of the organ extracts tested. The presence of atropinesterase in frog liver, and its absence from the serum, has been confirmed. Hydrolysis of homatropine, but not atropine, by guinea pig liver was observed, while the serum was without action on either of the compounds. On this basis the possibility arises that guinea pig liver contains a homatropinesterase enzyme separate from atropinesterase. It was shown that lack of atropinesterase activity in certain rabbits is not likely to be due to the presence of a naturally occurring inhibitor. It has been demonstrated that contrary to previous indications neither fresh egg white nor yolk possess atropinesterase activity. The specificity of tropinesterases was investigated and evidence was presented for the possible existence of two distinct enzymes, cocainesterase and tropacocainesterase.  相似文献

The pollen of three monotypic genera, Circaeaster, Kingdonia, and Sargentodoxa has been examined by light and scanning electron microscopy and in the case of the last genus, also by transmission electron microscopy. The type of tectum found in Circaeaster and Kingdonia, derivations of a compound layer of striae, has a restricted distribution in the Order Ranunculales. Of 64 genera examined in this order only six had species with a similar tectum. They include Achlys, Epimedium, Jeffersonia, and Vancouveria of the Berberidaceae s.l., the controversial Hydrastis, and Trollius of the Ranunculaceae. Circaeaster and Kingdonia have been considered as related since both have rare and primitive vegetative characteristics, the most notable being open dichotomous leaf venation. They are probably best treated as a ditypic family, Circaeasteraceae. The pollen of Sargentodoxa, especially the structure of the exine, closely resembles that of the Lardizabalaceae. However, the fruits of Sargentodoxa have been considered to be distinct from those of the Lardizabalaceae, suggesting that it be treated as a separate, but closely allied, family.  相似文献

Electron microscope and electron diffraction studies of developing embryonic bovine enamel have revealed the organization of the organic matrix and the inorganic crystals. The most recently deposited inorganic crystals located at the ameloblast-enamel junction are thin plates, approximately 1300 A long, 400 A wide, and 19 A thick. During maturation of the enamel, crystal growth occurs primarily by an increase in crystal thickness. Statistical analyses failed to show a significant change in either the width or the length of the crystals during the period of maturation studied. Even in the earliest stages of calcification, the crystals are organized within the prisms so that their long axes (c-axes) are oriented parallel to the long axes of the prisms but randomly distributed about their long axes. With maturation of the enamel, the crystals become more densely packed and more highly oriented within the prisms. The organic matrix in decalcified sections of enamel is strikingly similar in its over-all organization to that of the fully mineralized tissue. When viewed in longitudinal prism profiles, the intraprismatic organic matrix is composed of relatively thin dense lines, approximately 48 A wide, which are relatively parallel to each other and have their fiber axes parallel to the long axes of the prisms within which they are located. Many of these dense lines, which have the appearance of thin filaments, are organized into doublets, the individual 48 A wide filaments of the doublets being separated by approximately 120 A. When observed in oblique prism profiles, the intraprismatic organic matrix is likewise remarkably similar in general orientation and organization to that of the fully mineralized tissue. Moreover, the spaces between adjacent doublets or between single filaments have the appearance of compartments. These compartments, more clearly visualized in cross- or near cross-sectional prism profiles, are oval or near oval in shape. Therefore, the appearance of the intraprismatic organic matrix (in longitudinal, oblique, and cross-sectional prism profiles) indicates that it is organized into tubular sheaths which are oriented with their long axes parallel to the long axes of the prisms in which they are located, but randomly oriented about their own long axes, an orientation again remarkably "blue printing" that of the inorganic crystals. The predominant feature of the walls of the tubular sheaths, when viewed in cross- or near cross-section, is that of continuous sheets, although in many cases closely packed dot-like structures of approximately 48 A were also observed, suggesting that the wall of the sheaths consists of a series of closely packed filaments. The 48 A wide dense lines (filaments) representing the width of the sheath wall were resolved into two dense strands when viewed in longitudinal prism profiles. Each strand was 12 A wide and was separated by a less electron-dense space 17 A wide. The intraprismatic organic matrix is surrounded by a prism sheath which corresponds in mineralized sections to the electron-lucent uncalcified regions separating adjacent prisms. Structurally, the prism sheaths appear to consist of filaments arranged in basket-weave fashion.  相似文献

The structure, assembly, and composition of the extracellular hairs (mastigonemes) of Ochromonas are detailed in this report. These mastigonemes form two lateral unbalanced rows, each row on opposite sides of the long anterior flagellum. Each mastigoneme consists of lateral filaments of two distinct sizes attached to a tubular shaft. The shaft is further differentiated into a basal region at one end and a group of from one to three terminal filaments at the free end. Mastigoneme ontogeny as revealed especially in deflagellated and regenerating cells appears to begin by assembly of the basal region and shaft within the perinuclear continuum. However, addition of lateral filaments to the shaft and extrusion of the mastigonemes to the cell surface is mediated by the Golgi complex. The ultimate distribution of mastigonemes on the flagellar surface seems to be the result of extrusion of mastigonemes near the base of the flagellum, and it is suggested that mastigonemes are then pulled up the flagellum as the axoneme elongates. Efforts to characterize mastigonemes biochemically after isolation and purification on cesium chloride (CsCl) followed by electrophoresis on acrylamide gels have demonstrated what appear to be a single major polypeptide and several differentially migrating carbohydrates. The polypeptide is not homologous with microtuble protein. The functionally anomalous role of mastigonemes in reversing flagellar thrust is discussed in relation to their distribution relative to flagellar anatomy and to the plane of flagellar undulations.  相似文献
