First, second, third, early and late fourth-instar larvae, and pupae of Aedes aegypti were infected with Romanomerrais culicivorax and reared at 20, 25, and 30 C. An increase in the ratio of male to female nematodes was observed with increase in host age at the time of infection at each temperature. The number of pupal and late fourth-instar infections was low, but R. culicivorax continued to develop in adult A. aegypti. Since male nematodes were recovered from both male and female hosts infected as late fourth instars or pupae, the sex of the host did not influence the sex of the nematode.  相似文献

A new nematode, Tripius gyraloura n. sp., is described from the arundo gall midge, Lasioptera donacis Coutin (Diptera: Cecidomyiidae). This gall midge is being considered as a biological control agent for use in North America against the introduced giant reed Arundo donax (L.) (Poaceae: Cyperales). Thus the present study was initiated to investigate a nematode parasite that was unknown at the time studies with L. donacis were initiated. The new species has a rapid development in the fly host and the mature parasitic female nematodes evert their uterine cells in the hosts’ hemolymph. Because large numbers of nematodes sterilise the host, eradication of the parasite from laboratory colonies of the midge may be necessary before populations of the fly are released.  相似文献

Nippostrongylus brasiliensis: effect of host hormones on helminth ingestion in vivo. International Journal for Parasitology16: 77–80. Ingestion of dye in vivo by both sexes of Nippostrongylus brasiliensis was increased in gonadectomized male, but not female, mice that were implanted with testosterone-filled capsules. The nematodes increased their ingestion of dye in both sexes of gonadectomized hosts that were implanted with 17 β-estradiol and decreased their uptake of dye in progesterone-implanted animals. Implantation of intact male mice with capsules of hydrocortisone and cholesterol suggested that ingestion by N. brasiliensis was decreased and increased, respectively, in these animals. Ligation of the bile duct in gonadectomized male mice that were implanted with testosterone capsules reduced the ingestion of dye by both sexes of nematode.  相似文献

Gonadectomy of the host reduced the establishment of Nippostrongylus brasiliensis in male mice, but elevated the recovery of worms from female hosts. Similarly, production of eggs by individual females of N. brasiliensis decreased and increased in gonadectomized male and female animals, respectively. Implantation of testosterone-filled capsules caused a dosage-dependent release of eggs by helminths in gonadectomized hosts of both sexes. Maximal production of eggs by nematodes was found when plasma testosterone in the host exceeded 20 ng/ml. Treatment of gonadectomized mice of both sexes with testosterone implants gave recoveries of worms that were similar to the number of helminths in intact male mice. Ligation of the bile duct of male mice that were implanted with testosterone reduced both establishment and reproduction of N. brasiliensis.  相似文献

The entomogenous nematode Steinernema feltiae was encapsulated in an alginate matrix containing a tomato seed. When these capsules were placed on 0.8% agar for 7 days, the seed germinated and ca. 20% of the nematodes escaped from the capsules, whereas only 0.1% escaped from capsules without seeds. When capsules containing nematodes and a seed were planted into sterilized or nonsterilized soil, nematodes escaped to infect Galleria mellonella larvae. When seed in capsules containing ca. 274 nematodes per capsule were planted in nonsterilized soil, Galleria mortality was 90% 1 week later. Galleria mortality declined to 27%, 23%, and 0% in weeks 2, 4, and 8 postplant, respectively. In sterilized soil, Galleria mortality was 96% and did not differ significantly from the nonsterilized soil in week 1, but was significantly higher in sterilized soil over nonsterilized soil for week 2 (81%) and week 4 (51%). When capsules containing nematodes only were used, Galleria mortality was 71% in sterilized soil 1 week after planting and 58%, 33%, and 12% in weeks 2, 4, and 8 postplant, respectively. In nonsterilized soil, Galleria mortality was 8%, 30%, 21%, and 28% after 1, 2, 4, and 8 weeks, respectively, using only encapsulated nematodes. When the number of nematodes per capsule was increased to ca. 817, Galleria mortality was 92 % or higher in sterilized soil from week 1 to week 4.  相似文献

The Diaprepes root weevil, Diaprepes abbreviatus, is a pest of vegetables, ornamental plants, sugarcane, and citrus in Florida and the Caribbean. The entomopathogenic nematode, Steinernema riobrave, can reduce larval populations of D. abbreviatus substantially. Efficacy of entomopathogenic nematodes, however, may be affected by culture method and formulation. Using D. abbreviatus as the host, we compared the efficacy of two commercial S. riobrave formulations, a liquid and a waterdispersible granule (WDG), with each other and with in vivo produced S. riobrave. Nematodes in the commercial formulations were produced in vitro through liquid fermentation; the in vivo nematodes were cultured in Galleria mellonella and applied in aqueous suspension. Laboratory experiments measured nematode virulence in plastic cups containing soil and seventh-eighth instar D. abbreviatus. One laboratory experiment was conducted using only fresh nematodes (less than 5 days old); another experiment included WDG nematodes that were stored for 25 days at 10 °C. Two field experiments were conducted in which nematodes were applied either to potted citrus (containing D. abbreviatus larvae) placed beneath mature citrus trees or to soil directly beneath the tree. In the latter experiment, efficacy was determined by measuring mortality of caged D. abbreviatus larvae that were buried beneath the soil surface prior to application. Mortality of D. abbreviatus treated with nematodes ranged from 80-98% and 50-75% in laboratory and field experiments, respectively. In all experiments, we did not detect any significant effects of culture method or formulation.  相似文献

The presence and diversity of beetle pathogens associated with different tree species in Bulgaria was investigated. In total, 818 specimens belonging to 22 beetle species were examined. Pathogens occurred in 9 host species. The infections were found in the gut (virus, nematodes, protozoans, microsporidia) and haemolymph (nematodes) of the infected insects. The following pathogen species: ItEPV, Beauveria bassiana, B. brongniartii, Isaria farinosa, Gregarina typographi, Gregarina spp., Chytridiopsis typographi, Chytridiopsis sp., and nematodes were revealed. Insects with mycoses were mummified and filled up with fungal structures, formed pseudosclerotium. Most pathogens were established in bark beetles (Scolytinae). For the first time, the fungus B. bassiana was reported in Tomicus piniperda, Orthotomicus longicollis, O. erosus, X. spinole, Taphrorychus villifrons and Phylobius sp. in Bulgaria. We also present the first records of gregarines in O. longicollis, Acanthocinus aedilis, Rhagium inquisitor, Pyrochroa coccinea, and of the microsporidium Chytridiopsis sp. in O. longicollis.  相似文献

Entomopathogenic nematodes are lethal insect parasites that reproduce exclusively inside their hosts in nature. Infection decisions made by the free-living infective-stage juveniles have an impact on reproductive success, but it is likely that mating decisions are made by adults while inside their host. We investigated sexual communication between male and female adult stages of Steinernema carpocapsae (Rhabditida: Steinernematidae) to assess whether mating is chemically mediated during the adult stage or results from incidental encounters between adults inside the insect host. To assess chemical communication, we measured the behavioral response of adult male S. carpocapsae to several different potential sources of chemical information. Male S. carpocapsae responded to virgin females only and were not influenced by mated conspecific females, conspecific males, or heterospecific females. These results show that species-specific communication takes place between adult entomopathogenic nematodes within the host cadaver just prior to mating.  相似文献

Live adult and juvenile entomopathogenic Steinernema carpocapsae DD136 (P. Nematoda) were not subjected to adhesion by haemocytes of lepidopteran insect larvae of Galleria mellonella or Malacosoma disstriain vitro or in vivo. In vitro freeze-killed nematodes exhibited haemocyte attachment, the intensity increasing with time. Accumulation of haemocytes on the dead nematodes was associated with host phenoloxidase activity; live nematodes and their exudates did not activate the enzyme whereas dead nematodes but not their exudate did activate phenoloxidase. Live-nematode exudate inhibited granular cell and some plasmatocyte adhesion to slides, increased granular cell but not plasmatocyte dissociation from preformed haemocyte monolayers and in vivo elevated total haemocyte counts and changed the floating haemocyte types while impairing bacterial removal from the haemolymph. Dead-nematode exudate did not affect these parameters thus immunosuppressant activity by live nematodes may represent the release of inhibitors not associated with their cuticle. The third stage juveniles released the inhibitors.  相似文献

The many decades during which the cultivation of Cannabis sativa (hemp) was strongly restricted by law resulted in little research on potential pathogenic nematodes of this increasingly important crop. The primary literature was searched for hemp-nematode papers, resulting in citations from 1890 through 2021. Reports were grouped into two categories: (i) nematodes as phytoparasites of hemp, and (ii) hemp and hemp products and extracts for managing nematode pests. Those genera with the most citations as phytoparasites were Meloidogyne (root-knot nematodes, 20 papers), Pratylenchus (lesion nematodes, 7) and Ditylenchus (stem nematodes, 7). Several Meloidogyne spp. were shown to reproduce on hemp and some field damage has been reported. Experiments with Heterodera humuli (hop cyst nematode) were contradictory. Twenty-three papers have been published on the effects of hemp and hemp products on plant-parasitic, animal-parasitic and microbivorous species. The effects of hemp tissue soil incorporation were studied in five papers; laboratory or glasshouse experiments with aqueous or ethanol extracts of hemp leaves accounted for most of the remainder. Many of these treatments had promising results but no evidence was found of large-scale implementation. The primary literature was also searched for chemistry of C. sativa roots. The most abundant chemicals were classified as phytosterols and triterpenoids. Cannabinoid concentration was frequently reported due to the interest in medicinal C. sativa. Literature on the impact of root-associated chemicals on plant parasitic nematodes was also searched; in cases where there were no reports, impacts on free-living or animal parasitic nematodes were discussed.  相似文献

Diethylcarbamazine is a drug that is used for the treatment of filariasis in humans and animals; it also has effects on intestinal nematodes, but its mechanism of action remains unclear. Emodepside is a resistance-busting anthelmintic approved for treating intestinal parasitic nematodes in animals. The novel mode of action and resistance-breaking properties of emodepside has led to its use against intestinal nematodes of animals, and as a candidate drug for treating filarial parasites. We have previously demonstrated effects of emodepside on SLO-1 K+-like currents in Ascaris suum. Here, we demonstrate that diethylcarbamazine, which has been proposed to work through host mediated effects, has direct effects on a nematode parasite, Ascaris suum. It increases activation of SLO-1 K+ currents and potentiates effects of emodepside. Our results suggest consideration of the combination of emodepside and diethylcarbamazine for therapy, which is predicted to be synergistic. The mode of action of diethylcarbamazine may involve effects on parasite signaling pathways (including nitric oxide) as well as effects mediated by host inflammatory mediators.  相似文献

To date, the knowledge of the helminth communities of Antarctic birds is scarce or fragmented. Knowledge about diseases and parasites is crucial for understanding and managing ecosystems, particularly in isolated areas where host species are more sensitive to new diseases or parasite infections. It has been showed that variations in rate of parasitism may occur between populations of host species. Two major non-exclusive hypotheses have been proposed to explain such variability: exposure to parasitism and, perhaps more important, life history strategies of hosts. We studied the helminth community of the Kerguelen Shag Phalacrocorax verrucosus, an endemic seabird species of the Kerguelen Archipelago. We provide new data on the helminths infecting this species from partial or complete digestive tracts of two birds. Two nematodes (Contracaecum rudolphii s.l. and Ingliseria cirrohamata) were found free or attached to the wall of the proventriculus of birds, while the acanthocephalan Corynosoma sp. and the cestode species Tetrabothrius sp. occurred in the intestine of the shags. The genus Tetrabothrius is reported for the first time in Kerguelen Shags and in this area. The analysis of stomach contents from 41 live Kerguelen Shag individuals revealed infection by Contracaecum nematodes. The proportion of infected birds differed between colonies, possibly in relation to differential exposure to infected fish hosts.  相似文献

The host status of 15 commonly occurring pasture species for Helicotylenchus pseudorobustus was tested in a greenhouse trial. Only tall fescue, with and without Neotyphodium endophyte infection, was a good host (Pf/Pi = final/initial population > 1). Inoculation survival was tested in a second trial, which showed that only 10% of the H. pseudorobustus nematodes survived the first 7 days after inoculation. When the Pf/Pi was adjusted to account for a 10% survival, all of the grass and clover hosts tested had a Pf/Pi > 1. Both trials showed a positive correlation between increased numbers of H. pseudorobustus and free-living nematodes.  相似文献

Entomopathogenic nematodes carrying symbiotic bacteria represent one of the best non-chemical strategies for insect control. Infective juveniles of Heterorhabditidae and Steinernematidae nematodes actively seek the host in the soil, penetrating through insect’s openings to reach the hemocoel where symbiotic bacteria in the genera Photorhabdus or Xenorhabdus, respectively, are released. The bacteria replicate and produce virulence factors that rapidly kill the insect host, providing nutrients for the nematodes development and reproduction within the insect cadaver. More studies are necessary to better understand the factors implicated in the nematode-bacteria association, particularly focusing the bacterial symbionts, the final effectors of the insect death. Our group has shown that ureases are lethal to some groups of insects and may contribute to the entomopathogenic properties of the symbiotic bacteria.The fall armyworm Spodoptera frugiperda (Lepidoptera: Noctuidae) is one of the major insect pests in corn (Zea mays) crops in Brazil, with infestations resulting in reduction up to 39% yield and losses amounting US$ 500 million annually. Native strains of entomopathogenic nematodes active against S. frugiperda represent a promising alternative to the intensive use of chemical insecticides to control fall armyworm population in corn plantations.In this study we screened soil nematodes collected in the south region of Brazil for pathogenicity against S. frugiperda. Symbiotic bacteria associated with these nematodes were isolated and characterized. We also evaluated urease production by the symbiotic bacteria in vitro and along the course of infection in S. frugiperda and demonstrated that urease production correlated positively to their entomopathogenicity.  相似文献

The natural history of many entomopathogenic nematode species remains unknown, despite their wide commercial availability as biological control agents. The ambushing entomopathogenic nematode, Steinernema carpocapsae, and the introduced European earwig, Forficula auricularia, forage on the soil surface. Since they likely encounter one another in nature, we hypothesized that earwigs are susceptible to nematode infection. In the laboratory, the LC50 for F. auricularia was 226 S. carpocapsae/earwig and the reproductive potential was 123.5 infective juvenile nematodes/mg tissue. This susceptibility depended on host body size with significantly higher mortality rates seen in larger earwigs. In a study of host recognition behavior, S. carpocapsae infective juveniles responded to earwig cuticle as strongly as they did to Galleria mellonella cuticle. We also found that earwigs exposed to S. carpocapsae cleaned and scratched their front, middle and back legs significantly more than controls. Coupled with previous field data, these findings lead us to suggest that F. auricularia may be a potential host for S. carpocapsae.  相似文献

Studies with last instar larvae of the fall armyworm, Spodoptera frugiperda (J. E. Smith), the black cutworm, Agrotis ipsilon (Hufnagel), and the greater wax moth, Galleria mellonella (L.) were used to quantify the invasive ability of two strains (All and Mexican) of Steinernema carpocapsae and to determine how factors in the bioassay procedure affect both nematode invasion and host mortality. Nematode invasive ability was variable, with 10-50% of nematodes successfully infecting the host. The percentage of infectives invading the host (invasion efficiency) was positively related to increases in length of host exposure time and number of hosts per arena, negatively related to increases in substrate surface area per host, and not affected by nematode concentration. There was a direct relationship between concentration applied and the number of nematodes invading the host. Mortality was less affected than invasion efficiency by bioassay conditions and appears to be a much less sensitive index of nematode activity than invasive ability.  相似文献

Under laboratory conditions, the nematodes Neoaplectana carpocapsae and Heterorhabditis heliothidis were able to infect and develop inside the hemocoel of terrestrial isopods belonging to the genus Porcellio. Armidillidium vulgare was also infected by N. carpocapsae, but this host was less susceptible than Porcellio. Results show that, under suitable ecological conditions, it would be feasible to utilize N. carpocapsae as a biological control agent of sowbugs (Porcellio spp.). This is the first report showing the ability of neoaplectanids and heterorhabditids to invade representatives of the Crustacea.  相似文献

The mermithid parasite Heleidomermis magnapapula was maintained in larvae of the midge Culicoides variipennis for 20 months in enamel pans containing nutrient-rich water and polyester pads as a substrate. Inseminated female mermithids were introduced to the pad surface when the host was in the late second or early third-instar. Host larvae were harvested from the pans 9 days after exposure and held in tap water for nematode emergence. Preparasite yield was positively correlated with female nematode size and averaged 1,267 preparasites/female. Male and female nematodes emerged an average of 12.2 and 13.4 days after host exposure, respectively. Supplemental host food (Panagrellus) during the final days of parasitism did not alter time of emergence. Parasites emerging singly were 64% females, whereas superparasitized hosts yielded males (up to nine/host). Nematode carryover into the adult midge normally occurred at a level of 0.5-2.5%. Parasite load (nematodes/ parasitized individual) in midge adults was lower than that of larvae from the same cohort, and adult midges were more likely to harbor female parasites. Exposure of fourth-instar host larvae resulted in higher levels of adult parasitism (up to 17%).  相似文献

Aims

The use of entomopathogenic nematodes in the biological control of soil insect pests is hampered by the costly and inadequate application techniques. As a possible solution we evaluated a nematode encapsulation approach that offers effective application and may possibly attract the pest by adding attractants to the capsule shell.

Methods

Heterorhabditis bacteriophora nematodes, which show high virulence against the maize root pest Diabrotica virgifera virgifera, were encapsulated in a polysaccharide shell derived from the algae Laminaria ssp. Shells of varying thickness and composition were evaluated.

Results

Nematodes readily survived the encapsulation process and were able, varying with shell thickness and temperature, to break through the shell and subsequently infect hosts. The added attractants and feeding stimulants to the shell attracted the pest larvae as much as maize roots. In field trials, encapsulated H. bacteriophora nematodes were more effective in controlling D. v. virgifera than those sprayed in water over the soil surface, but in these trials the addition of stimulants did not increase the control efficiency.

Conclusions

The study demonstrates that nematodes can be successfully applied in capsules in the field. Further improvements are needed to make the capsules a cost effective alternative to conventional field application of nematodes.  相似文献

The entomogenous nematode Neoaplectana carpocapsae and its associated bacterium, Xenorhabdus nematophilus, could not infect the pupal stage of the tachinid Compsilura concinnata through the puparium. N. carpocapsae had an adverse effect on 1-, 2- and 3-day-old C. concinnata larvae within the armyworm host in petri dish tests. All 1-day-old larvae treated with nematodes died in their hosts, whereas 61% and 69% of 2- and 3-day-old larvae treated with nematodes, respectively, died. However, the survivors developed to adults. Nine to thirty-seven percent of adult tachinids which emerged from nematode-treated soil (50 nematodes/cm²) were infected with N. carpocapsae. The nematode adversely affects C. concinnata directly by the frank infection of the tachinid and indirectly by causing the premature death of the host which results in tachinid death.  相似文献
