The crystal–Stellate system is one of the most known example of interaction between heterochromatin and euchromatin: a heterochromatic locus on the Y chromosome (crystal) 'represses a euchromatic locus (Stellate) on the X chromosome in Drosophila melanogaster. The molecular mechanism regulating this interaction is not completely understood. It is becoming clear that an RNA interference (RNAi) mechanism could be responsible for the silencing carried out by crystal on the Stellate sequences. Here, a detailed structural analysis of all the sequences involved in the system is reported, demonstrating a their 'puzzling structure. In addition three autosomal mutations: sting, scratch and sirio are described that interfere with the system. All of them are male sterile mutations and exhibit crystals made by the STELLATE protein in their primary spermatocytes. They are requested during oogenesis and early in embryogenesis as well. Hypothesis on the involvement of these genes in activating the Stellate sequences are discussed.  相似文献

Some organisms that experience subzero temperatures, such as insects, fish, bacteria, and plants, synthesize antifreeze proteins (AFPs) that adsorb to surfaces of nascent ice crystals and inhibit their growth. Although some AFPs are globular and nonrepetitive, the majority are repetitive in both sequence and structure. In addition, they are frequently encoded by tandemly arrayed, multigene families. AFP isoforms from the mealworm beetle, Tenebrio molitor, are extremely potent and inhibit ice growth at temperatures below −5°C. They contain a 12-amino acid repeat with the sequence TCTxSxxCxxAx, each of which makes up one coil of the β-helix structure. TxT motifs are arrayed to form the ice-binding surface in all three known insect AFPs: the homologous AFPs from the two beetles, T. molitor and Dendroides canadensis, and the nonhomologous AFP from the spruce budworm, Choristoneura fumiferana. In this study, we have obtained the cDNA and genomic sequences of additional T. molitor isoforms. They show variation in the number of repeats (from 6 to 10) which can largely be explained by recombination at various TCT motifs. In addition, phylogenetic comparison of the AFPs from the two beetles suggests that gene loss and amplification may have occurred after the divergence of these species. In contrast to a previous study suggesting that T. molitor genes have undergone positive Darwinian selection (selection for heterogeneity), we propose that the higher than expected ratio of nonsynonymous-to-synonymous substitutions might result from selection for higher AT content in the third codon position. 为探讨香樟(Cinnamomum camphora)叶肉含晶细胞超微结构的季节变化,阐明香樟叶肉中草酸钙晶体在春夏秋冬的变化规律。该研究以多年生香樟(C. camphora)叶片为材料,分别于春夏秋冬四个季节露地取样,制作超薄切片,用透射电子显微镜(TEM)观察叶肉含晶细胞超微结构的变化。结果表明:春季时香樟叶肉中只有少数细胞有草酸钙晶体,数量较少,晶体结构多为柱状晶、方晶; 夏季时香樟叶肉细胞中随机分布于液泡的草酸钙晶体明显比春季的数量多、体积大、形态丰富,晶体多为柱状晶、方晶、针晶、簇晶; 秋季时香樟叶肉细胞草酸钙晶体和夏季的类似,数量较多,形态多样,以方晶和柱状晶针晶为主,伴有晶簇; 冬季时香樟叶肉含晶细胞晶体形态为柱状晶、方晶、针晶,数量比夏季和秋季的数量略有减少。该研究结果表明在一年四季中香樟叶肉细胞液泡中均有草酸钙晶体结构存在。  相似文献

Summary The red alga,Antithamnion kylinii Gardner, was found to have needle-shaped inclusions about 10 m long and less than 0.4 m thick. They ranged in abundance from one or a few in young cells to hundreds in fully enlarged cells. Under polarized light, the inclusions were birefringent, indicating crystallinity. Solubility tests suggested that the inclusions were composed of calcium oxalate: they dissolved in 1 N hydrochloric acid and in a saturated solution of aqueous cupric acetate, but they were not soluble in 10 N acetic acid or 5.25% sodium hypochlorite. X-ray microanalysis confirmed the presence of calcium. Calcium oxalate crystals were present in cells of indeterminate axes, but cells of determinate lateral filaments lacked them. Light and electron microscopic study demonstrated that the crystals were associated with the parietal cytoplasm. Calcium oxalate crystals were also present inA. defectum Kylin, but they were not found in ten more distantly related taxa.  相似文献

The freshwater eels of the genus AnguillaSchrank are widely distributed. They have beenconsidered for a long time to all have acatadromous spawning strategy. However, in arecent work Tsukamoto et al. (2002)considered that catadromy should be seen asfacultative. They all have a long oceaniclarval stage known as the leptophalus stage. Alarge number of studies on their ecology,biology, and physiology exist but littleattention has been focused on their systematicsand species-relationships. Ege (1939)described 19 species and sub-species based onmorphometric data. Castle and Williamson (1974)made A. ancestralis a synonym of A.celebesensis. Morphological characters,however, have limitations as taxonomiccharacters because they overlapped in mostspecies. Biochemical characters, such asmt-DNA, are more informative. Dijkstra andJellyman (1999) found no genetic differencesbetween A. australis australis and A. a. schmidtti, and now 15 species arewidely recognized. Phylogenetic studies(Aoyama and Tsukamoto, 1997) suggestdescendant-ancestor relationships betweenAtlantic and Pacific eels, respectively. Themost likely dispersal route for the Atlanticeels from the Pacific appears to have beenthrough the Tethys Sea. Two species are foundin the Atlantic: A. anguilla and A.rostrata. They both spawn in the Sargasso Sea,but differ in morphometric and genetic traits,and are considered as two separate species witha relatively recent evolutionary divergence.The purpose of this paper is to presentknowledge and questions about the life history,taxonomy, and evolution of this mysteriousgenus.  相似文献

The cell wall lipopolysaccharides from three phototrophic species of the alpha1-group of Proteobacteria, Rhodospirillum rubrum, Rhodospirillum molischianum, and Rhodopila globiformis were isolated and chemically characterized. Sodium deoxycholate polyacrylamide gel electrophoresis patterns revealed that the lipopolysaccharides of all three species possess O-chains. They are composed of repeating units only in R. molischianum and R. globiformis. The presence of l-glycero-d-mannoheptose and 2-keto-3-deoxyoctonate indicated core structures in all three lipopolysaccharides. Glucosamine was found as backbone amino sugar in lipid A of R. molischianum and R. rubrum, while R. globiformis has 2,3-diaminoglucose as backbone amino sugar. The latter species also differed from the two former ones in its content of hydroxy fatty acids (3-OH-14:0, 3-OH-16:0 in R. rubrum and R. molischianum and 3-OH-14:0, 3-OH-18:0 and 3-OH-19:0 (possibly iso- or anteisobranched) in R. globiformis).Abbreviations DOC-PAGE sodium deoxycholate polyacrylamide gel electrophoresis - GC/MS combined gas-liquid chromatography/mass spectrometry - KDO 2-keto-3-deoxyoctonate  相似文献

Proteinaceous intranuclear crystals are found in the fern Scolopendrium vulgare. During mitosis these crystals are eliminated from the nucleus into the cytoplasm, where they are dissolved. New crystals appear in the nucleus. The site of synthesis of intranuclear crystal proteins was investigated using quantitative ultrastructural autoradiography after incubation with tritiated lysine. The results suggest a migration of cytoplasmic proteins to the nucleus, part of which would then be incorporated into the intranuclear crystals.  相似文献

Monocotyledons are distinguishable from dicotyledons by their subtype P2 sieve-element plastids containing cuneate protein crystals, a synapomorphic character uniformly present from basal groups through Lilioids to Commelinoids. The dicotyledon generaAsarum andSaruma (Aristolochiaceae-Asaroideae) are the only other taxa with cuneate crystals, but their sieveelement plastids include an additional large polygonal crystal, as is typical of many eumagnoliids. New investigations in Melanthiaceae s.l. revealed the same pattern (polygonal plus cuneate crystals) in the sieve-element plastids ofJaponolirion osense (Japonoliriaceae/Petrosaviaceae), ofHarperocallis flava, Pleea tenuifolia, andTofleldia (all: Tofieldiaceae). InNarthecium ossifragum a large crystal, present in addition to cuneate ones, usually breaks up into several small crystals, whereas inAletris glabra andLophiola americana (Nartheciaceae) and in all of the 15 species studied and belonging to Melanthiaceae s.str. only cuneate crystals are found. Highresolution TEM pictures reveal a crystal substructure that is densely packed in both cuneate and polygonal forms, but in Tofieldiaceae the polygonal crystals stain less densely, probably as a result of the slightly wider spacing of their subunits. The small crystals ofNarthecium are “loose”; that is, much more widely spaced. Such “loose” crystals are commonly found in sieve-element plastids of Velloziaceae, present there in addition to angular crystals, and together with cuneate crystals in a few Lilioids and many taxa of Poales (Commelinoids). Ontogenetic studies of the sieve elements ofSaruma, Aristolochia, and several monocotyledons have shown that in their plastids cuneate crystals develop very early and independent from a polygonal one present in some taxa. Therefore, a conceivable particulation of polygonal into cuneate crystals is excluded. Consequently, mutations of some monocotyledons that contain a lone, large, polygonal crystal in their sieve-element plastids are explained as the result of a complex genetic block. The total result of all studies in sieve-element plastids suggests thatJaponolirion and Tofieldiaceae are the most basal monocotyledons and that Aristolochiaceae are their dicotyledon sister group.  相似文献

Summary The otoliths of embryos and young animals of the lizard Podarcis s. sicula were studied by X-ray diffraction and scanning electron microscopy. Two types of crystal that give different X-ray diffraction patterns were found in the membranous labyrinth of Podarcis. The crystals consist of calcite or aragonite and are easily distinguished by scanning electron microscopy because of their different morphology. The two calcium carbonate crystal forms are not mixed at random but are present in the embryo from the very beginning in specific sites. The endolymphatic sac contains aragonite crystals while the saccule contains calcite crystals adjacent to the wall, in addition to a preponderance of aragonite crystals. The utricle and lagena contain only calcite crystals. The presence of two crystal forms of calcium carbonate in the membranous labyrinth are discussed in terms of differing genetic and functional significance.  相似文献

Examination of leaf cells of Dianthus barbatus with the election microscope revealed the presence of crystals in the nuclei, but not in the cytoplasm or in other organelles. They were at first thought to be viral crystalline inclusions, but no evidence could be adduced to support this possibility. The crystals were found in the nuclei of cells from all parts of the leaf, including parenchyma cells of the vascular elements. They were usually 0.5–2 μ long, and occasionally up to 5 μ. In longitudinal section they appeared as electron-dense striations, about 10–15 mμ wide, alternating with electron-transparent bands, 6–8 mμ wide. In cross section they had a honeycomb appearance, resulting from closely packed hexagonal elements, with electron-transparent centers. Similar crystals were found in nuclei of D. chinensis leaf cells, but not in other members of the Caryophyllaceae, including two varieties of D. caryophyllus, D. deltoides, D. arenarius, Silene acaulis, Lychnis chalcedonica, Saponaria calibrica, and Stellaria media. This appears be the first fine structural report of a lattice inclusion in nuclei of cells of healthy higher plants.  相似文献

The changes in the levels of proline, sugar and soluble protein during leaf rolling and its relationship to osmotic adjustment were studied in laboratory conditions. Upon irrigation of plants which have rolled leaves, many sugar crystals occurred on the abaxial surface of the leaves in Ctenanthe setosa (Rosc.) Eichler. The sugar crystals were determined to have sucrose, glucose and fructose. The levels of reducing sugars and proline are higher in rolled leaves while soluble protein levels in rolled leaves are lower than those of unrolled leaves. It was found 1–3, 9–13, 16–21 and 24–28 crystals at degree of leaf rolling 23, 28, 47 and 52%, respectively. Finally, we found a significant correlation between the crystal number and degree of leaf rolling in Ctenanthe setosa. In addition, soluble sugars are found predominant accumulating solute in the plant and are of major importance as a contributor to osmotic adjustment during leaf rolling.  相似文献

Several extreme thermophilic Gram negative bacteria found in a thermally polluted river in Belgium have been compared with Thermus strains isolated from widely distant geographical areas. This analysis has become possible after the design of a new culture medium (162).All strains examined (including the isolate successively denominated Flavobacterium thermophilum and Thermus thermophilus) were found to be morphologically identical with strain YT-1 of Thermus aquaticus. The cells are immotile, rod-like, strictly aerobic, catalase and oxidase positive. They produce amylase, hydrolyze gelatin and are confirmed to be highly sensitive towards penicillin.The nutritional pattern of all strains has been analysed extensively, by testing a broad spectrum of possible substrates.The strains display a uniform response to the microbiological tests applied and most probably belong to the same species: Thermus aquaticus.Abbreviations GC guanosine cytosine - ATCC American Type Culture Collection - DSM Deutsche Sammlung von Mikroorganismen  相似文献

The ultrastructure of β-chitin microfibrils from a centric diatom, Thalassiosira, and a tubeworm, Lamellibrachia, was studied using electron diffraction and high-resolution electron microscopy. Electron microdiffraction diagrams corresponding to each projection of the β-chitin crystals were obtained, and all the data support the structure model of anhydrous β-chitin crystals proposed by X-ray diffraction experiments. From high-resolution electron microscopy on ultrathin sections, the cross-sectional shapes of the microfibrils from Thalassiosira and Lamellibrachia were observed as a rectangular and parallelogram, respectively. The lattice fringes corresponding to the (0 1 0) plane of anhydrous β-chitin crystals were clearly observed in both cross-sections. Based on these observations, we have constructed a molecular packing model for β-chitin microfibrils.  相似文献

For experimental induction of crystal cells (=crystal idioblasts) containing calcium-oxalate crystals, the lower epidermis was peeled from seedling leaflets of Gleditsia triacanthos L., exposing the crystal-free mesophyll and minor veins to the experimental solutions on which leaflets were floated for up to 10 d under continous light. On 0.3–2.0 mM Ca-acetate, increasing numbers of crystals, appearing 96 h after peeling, were induced. The pattern of crystal distribution changed with Ca2+-concentration ([Ca2+]): at low [Ca2+], crystals formed only in the non-green bundlesheath cells surrounding the veins, believed to have a relatively low Ca2+-extrusion capacity; at higher [Ca2+], crystals developed in up to 90% of the mesophyll cells, and at supraoptimal [Ca2+], large extracellular crystals formed on the tissue surface. By sequential treatments with solutions of different [Ca2+], the following three phases were identified in the induction of crystal cells: (1) during the initial 24-h period (adaptive aging), Ca2+ is not required and crystal induction is not possible; (2) during the following 48 h (induction period), exposure to 1–2 mM Ca-acetate induces the differentiation of mesophyll cells into crystal cells; (3) crystal growth begins 72 h after the start of induction. In intact leaflets of Albizia julibrissin Durazz., calcium-oxalate crystals are found exclusively in the bundle-sheath cells of the veins, but crystals were induced in the mesophyll of peeled leaflets floating on 1 mM Ca-acetate. Exposure to inductive [Ca2+] will thus trigger the differentiation of mature leaf cells into crystal cells; the spatial distribution of crystals is determined by the external [Ca2+] and by the structural and functional properties of the cells in the tissue.  相似文献

Summary Many gynandromorphs were obtained from the natural population ofVollenhovia emeryi (microgyna form) in Gifu, Japan. They were primarily male: most had the thorax and gaster of males, and the head contained tissues partially feminized to varying degrees. These gynandromorphs were found in 27 of 45 colonies studied (60.0%). Their proportion to total males in each colony varied from 3.7–47.7%, with a mean of 21.4% (n = 21). The gynandromorphs were found in all study areas and in every study year, suggesting that gynandromorphism in this species is not a rare phenomenon. Moreover, this observation suggests that gynandromorphs may occur more frequently in micraners than in macraners.  相似文献

Summary The development and distribution of calcium oxalate crystals, stomates and hairs were studied in the first trifoliolate leaf ofRhynchosia caribaea (Leguminosae: Papilionoideae, Phaseoleae). Using light and transmission electron microscopy, the crystals were shown to occur in both bundle sheath and mesophyll cells. Crystal distribution and shapes are characteristic forRhynchosia. Crystals develop late in leaf development in contrast to the stomates and hairs. As these latter two structures decrease in number per unit area with leaf age, crystal number increases.  相似文献

The biological activity of Bacillus thuringiensis (Bt) strain PS86Q3 against five Hymenopteran species was determined by means of bioassays adapted to each species. Four species of sawfly that are important pests of conifers (Diprion pini, Gilpinia hercyniae and Pristiphora abietina) or ornamental plants (Arge rosae), as well as the non-target honeybee, Apis mellifera, were studied. Two out of the four sawfly species tested were found to be sensitive to PS86Q3 crystals or spore/crystal suspensions. A sporulated culture of this strain was moderately active on D. pini, and a complete bioassay with solubilized crystals was performed to estimate the LC50 of 4.9 mg/ml. Pristiphora abietina was also found to be sensitive to PS86Q3, with an LC50 of 1.6 mg/ml. By contrast, at the concentrations tested, PS86Q3 did not prove active on the remaining sawflies, G. hercyniae and A. rosae. The strain was administered orally to check its effects on honeybees which were fed sucrose solutions supplemented with a PS86Q3 sporulated suspension, in a field assay using commercial beehives. No significant differences in larval mortality (as deduced by comparing the number of larvae, pupae and empty cells) were found between the Bt and control treatments. On the basis of the results presented here, the suitability of PS86Q3 for the control of Hymenopteran pests, particularly sawflies, in terms of both potency and environmental safety, is discussed.  相似文献

Unicellular autofluorescent picoplankton ranging from 0.4 to 1.5 µm in diameter were found to be a significant component of phytoplankton in the North Basin of Lake Biwa during early summer in 1989 and 1990. The abundance of these picoplankton varied seasonally by about three orders of magnitude with one maximum of up to 106 cells ml–1. Bloom-forming picoplankton were isolated by dilution and further cultivated in liquid medium. Three clones were found to be representative species of the bloom. Using epifluorescence and electron microscopy as well as absorption and fluorescence emission spectroscopy, we examined these clones according to shape and pigment composition. They have ringlike thylakoids, are photosynthetically active and have no nuclear envelope. The cyanobacterial clones isolated represent three types containing phycobilisomes with either phycocyanin or phycoerthrin as the dominant accessory pigment. They are described here as three new species, two phycoerythrin-rich types and one phycocyanin-rich type, all of them belonging to the Synechococcus group. The differences found by fluorescence emission of isolated clones are discussed with respect to in situ strain identification.  相似文献

Bacteria belonging to the genus Acetobacter and Gluconobacter, and enzymes isolated from them, have been extensively used for biosensor construction in the last decade. Bacteria used as a biocatalyst are easy to prepare and use in amperometric biosensors. They contain multiple enzyme activities otherwise not available commercially. The range of compounds analyzable by Gluconobacter biosensors includes: mono- and poly-alcohols, multiple aldoses and ketoses, several disaccharides, triacylglycerols, and complex parameters like utilizable saccharides or biological O2 demand. Early reports of the presence of calciumoxalate crystals in the cortices ofPhaseolus vulgaris root nodules have been confirmed. Crystals were found in all six genera examined (Cajanus, Desmodium, Glycine, Lespedeza, Phaseolus, Vigna) that have determinate nodules and export ureides. They were absent from six genera examined that have indeterminate nodules and export amides. The possible physiological significance of these structures is discussed.  相似文献
