The lengths of the A, B, and D genomes of common wheat,Triticum aestivum, were measured from the karyotype. Relative to the B genome, standardized as length 1.000, the lengths of the A and D genomes were 0.835 and 0.722, respectively. The lengths of the chromosome arms in the A and D genomes were then multiplied by the appropriate constants so that the total lengths of each genome also equalled 1.000. These calculations revealed that homoeologous chromosomes in wheat, with a few exceptions, have similar sizes and arm ratios. The arm lengths of the three homoeologues in each homoeologous group were then averaged. These average chromosomes turned out to be remarkably similar, in size and arm ratio, to their homoeologues in the E genome ofElytrigia elongata. This evidence and data on cross-compatibility and morphological characteristics suggested that the genusTriticum is a result of adaptive radiation from the perennial genusElytrigia, specifically from the complex of species possessing the E genome or one closely related to it.  相似文献

A modification of the general system of classification ofCalliphoridae is proposed in this report. Information about the chromosome complements of about 90 species is provided including references to the earlier literature on chromosomes of this family. With one exception all species studied by us or by earlier investigators have 2n=12. The total complement length of 556 complements from 83 species averaged 66.4 μ. Most species have a short heteromorphic sex pair but the size and morphology vary considerably and one species,Hemipyrellia fernandica, seems to have a quadrivalent sex-chromosome complex in a 2n=14 complement. The autosomal pairs vary in both length and arm ratios between the species but seem to be more stable than the sex chromosomes. Only minor variations were found between different collections of the same species except for two collections ofCalliphora croceipalpis. Although presently available data on the chromosomes of Diptera species suggest that increases in total complement length accompany evolutionary progress, no such general trends were obvious in either the total complement lengths or chromosome morphology, either within or between the tribes ofCalliphoridae. Thus the karyotypical reorganizations in the family have apparently not been directly associated with changes in the general morphological features of the adults which may have responded in these respects more extensively to genetic mutations.  相似文献

Numerous studies on Oenothera species have been investigated for the physiological and ecological characteristics. However, no such an information based on molecular cytogenetic has yet been introduced, in turn, is very essential for identifying sequence polymorphisms of rRNA genes with their loci on mitotic phases for further biological researches. In this study, sequence variations of rRNA genes in Oenothera odorata and O. laciniata were examined to identify informative factors as unique or repeat sequences in intra- and inter-specific variations. Intra-specific variation revealed that the sequences of the rRNA genes including the spacer regions were highly conserved revealing only a few variations. From the inter-specific variation, spacer regions of species were completely different as (1) non-homologous sequences in NTS and (2) different type repeat sequences in ITS 1, 2 and 5.8S rRNA, whereas the remaining coding regions were highly conserved. FISH was carried out on mitotic phases using the 5S rDNA of the analyzed sequences. From the interphase and metaphase chromosomes of the examined species, two loci of 5S rDNA in O. odorata and four loci in O. laciniata were confirmed on the telomeric region of the short arm. Due to the small size and unclear centromere of the chromosomes, karyotype could not be completed. However, we confirmed that the chromosomes are organized by meta- and acrocentric chromosomes and the chromosomes with identified loci were assumed to be paired by the location of loci at the telomeric region on the short arm with relative lengths.  相似文献

The morphology of the pachytene chromosomes ofPennisetum purpureum was studied. On the basis of relative lengths and arm ratios it was possible to identify them individually. They are numbered 1 to 14 in the order of their decreasing lengths and diagnostic characters are given for each chromosome. The first and the fourteenth chromosomes, the longest and the shortest of the complement respectively, are the nucleolus organizing chromosome. In all the fourteen chromosomes the centromeres are flanked by deep staining regions. This feature is common to the two species.P. typhoides andP. purpureum. Seven out of the fourteen chromosomes have terminal knobs in their long arms. This is a feature in which it differs from the other seven chromosome species.Pennisetum typhoides. These knobs, however, are not present uniformly in all populations; material obtained from Ghana did not show these knobs.  相似文献

Silene latifolia is a key plant model in the study of sex determination and sex chromosome evolution. Current studies have been based on genetic mapping of the sequences linked to sex chromosomes with analysis of their characters and relative positions on the X and Y chromosomes. Until recently, very few DNA sequences have been physically mapped to the sex chromosomes of S. latifolia. We have carried out multicolor fluorescent in situ hybridization (FISH) analysis of S. latifolia chromosomes based on the presence and intensity of FISH signals on individual chromosomes. We have generated new markers by constructing and screening a sample bacterial artificial chromosome (BAC) library for appropriate FISH probes. Five newly isolated BAC clones yielded discrete signals on the chromosomes: two were specific for one autosome pair and three hybridized preferentially to the sex chromosomes. We present the FISH hybridization patterns of these five BAC inserts together with previously described repetitive sequences (X-43.1, 25S rDNA and 5S rDNA) and use them to analyze the S. latifolia karyotype. The autosomes of S. latifolia are difficult to distinguish based on their relative arm lengths. Using one BAC insert and the three repetitive sequences, we have constructed a standard FISH karyotype that can be used to distinguish all autosome pairs. We also analyze the hybridization patterns of these sequences on the sex chromosomes and discuss the utility of the karyotype mapping strategy presented to study sex chromosome evolution and Y chromosome degeneration.Communicated by J.S. Two cytologically distinguishable species have been found in material ofChironomus australis. These species have been calledCh. australis andCh. duplex. Both belong to thepseudothummi-cytological grouping because they have the chromosome arm combinations AE, BF, CD, G.Ch. duplex shows a modified arm pattern due to a tandem fusion of arm G to arm E.—The banding patterns of the polytene chromosomes of the two species are compared to each other and to the Australian standard species,Ch. oppositus. Ch. australis is very close cytologically toCh. oppositus, whileCh. duplex, which is considered a derived species because of the tandem fusion, shows a number of inversion differences from the morphologically similarCh. australis. Ch. duplex is polymorphic for six inversions, four of which are simple inversions, the other two are complex involving also the transposition of some bands.Ch. australis appears to be monomorphic.  相似文献

Summary The chromosome complements of eighty brain cells ofHylemya antiqua have been studied. The eighty cells were found in thirty-three larvae. Total complement length (TCL) is not randomly distributed among the larvae. Because there is an inverse correlation between chromosome length and width, it appears that in the cells studied the different chromosome lengths are partly expressions of different stages of metaphase contraction. It is suggested that synchronous division of cells still occurs in late larvae.The length of each chromosome arm is highly correlated with that of every other arm. It is possible that the correlations are complete but that inadequate technique causes the departures from completeness which are observed. The chromosome lengths are corrected slightly for distortions, but the corrections make very little difference in the correlation coefficients. There is a high value for the correlation between the correlation of two arm lengths and the sum of the two arm lengths. This is to be expected if the perfect correlation between all arm lengths is being obscured by errors of drawing and measurement.The autosomal arms have very similar coefficients of variation. The arm ratios (length of long arm divided by short arm) are not correlated with TCL or with each other, and arm ratio is randomly distributed among the larvae. The sex chromosomes have a smaller coefficient of variation than the autosomes, so that they are relatively large in small cells and relatively small in large cells.Twenty-two cells inHylemya fugax were measured. The autosomes also showed a high correlation between arm lengths. An entirely heterochromatic autosomal arm showed the same phenomenon of a low coefficient of variation which was shown by the heteropycnotic sex chromosomes inH. antiqua. The low variability of heterochromatic regions accompanied by an apparently non-random distribution of the TCL may produce an erroneous picture of the species complement when dealing with small numbers.It is suggested that for simplicity in using cytological observations of this sort for taxonomic purposes, the technique of measuring the percent TCL of a chromosome plus its arm ratio be replaced by the percent TCL of each arm plus the average length difference between the arms of each chromosome pair in units of percent TCL.  相似文献

Studying the genome structure of Epimedium has been hindered by the large genomes and uniform karyotypes. Consequently our understanding of the genome organization and evolutionary changes of Epimedium is extremely limited. In the present study, the 45S and 5S rDNA loci of 14 Epimedium species were physically mapped by double-probe FISH for the first time. Results showed the following: (1) Chromosomes I and II of all 14 species examined, except for E. shuichengense, hosted one pair of 45S rDNA sites, respectively. Most of the 45S rDNA sites gave clear signals and were positioned in the distal regions of the short arms. (2) All species studied of section Diphyllon were found to have one pair of 5S rDNA sites localized in the interstitial regions of the long arm of chromosome IV, and the two species of section Epimedium, E. alpinum and E. pubigerum, had two pairs of 5S rDNA sites localized in the interstitial regions of the long arm of chromosomes IV and V, respectively. (3) In section Diphyllon, all species of small flower taxa, except E. shuichengense, had three pairs of 45S rDNA sites, clearly more than species of big flower taxa, except E. davidii, with two pairs of 45S rDNA sites. Based on the 45S and 5S rDNA distribution patterns and other chromosomal morphological characteristics, six pairs of chromosomes can be unambiguously identified in all 14 Epimedium species. The stable differentiation in 45S and 5S rDNA FISH patterns between the two sections suggests that chromosomal rearrangements and transpositional events played a role in the splitting of the two sections, and section Diphyllon may be more primitive than section Epimedium. In the same way, big flower taxa may be more primitive than small flower taxa in section Diphyllon.  相似文献

Dual-color fluorescence in situ hybridization (FISH) analysis of three Cucurbitaceae species from different genera was conducted using 5S and 45S rDNA probes. In Benincasa hispida (Thunb.) Cogn. (2n=24), the 45S rDNA probe hybridized on two chromosomes, one in the short arm of a medium-sized metacentric chromosome and another at the satellite of a chromosome. The 5S rDNA hybridized at a site proximal to the centromere of the same short arm of the 45S rRNA gene locus that occupied almost the entire short arm. For Citrullus lanatus (Thunb.) Matsum & Nakai (2n=22), the 45S rDNA probe hybridized at sites in the short arms of two chromosomes and the 5S rDNA probe was co-localized with the 45S rRNA locus at the region proximal to the centromere in one chromosome. The 45S rRNA loci occupied almost all of the short arms in both chromosomes. In Cucurbita moschata Duch. (2n=40), the 45S rDNA probe hybridized in five chromosomes in which the 45S rRNA genes occupied almost two-thirds of the chromosomes in two large chromosomes and the entire short arm of a medium-sized chromosome. Two other loci were present in two medium-sized chromosomes, one in the proximal region in the short arm of a chromosome and another at the tip of the long arm of a chromosome. Chromosomes of B. hispida were relatively larger than those of the other two species. The karyotype of B. hispida is composed of two metacentrics and 10 submetacentrics, while that of C. lanatus is composed of seven metacentrics and four submetacentrics and that of C. moschata is composed of 18 metacentrics and two submetacentrics. Comparative chromosome evolution among the three Cucurbitaceae species was attempted using the karyotypes and the chromosomal distribution patterns of the 5S and 45S rDNAs. The results presented herein will be useful in elucidating the phylogenetic relationships among Cucurbitaceae species, and will provide basic data for their breeding programs.  相似文献

Mitotic chromosomes of 13 species included in Solanum sect. Lasiocarpa were studied. All species have 2n = 24. The chromosome numbers of S. stagnale, S. felinum, and S. repandum are reported for the first time. Statistical analyses of chromosomes, genome length, and centromere position yielded estimates of karyotype composition and asymmetry. A generalized karyotype of the section shows that most of the chromosomes are metacentric (73%) or submetacentric (25.6%). Two pairs of subtelocentric chromosomes are present only in S. sessiliflorum. Satellites are quite common, characterizing chromosomes for ten species and are attached to the short arms of either m or sm chromosomes. They could be seen in 10% to 70% of the cells. Only S. pseudolulo bears two satellited chromosomes, one of them attached to a long chromosome arm. Although the section is chromosomally quite homogeneous, species can be distinguished by karyotype formula, presence of satellites in a particular chromosome pair, and total chromosome length. There are no indications of major chromosomal rearrangements within the section. Using cytological data exclusively, cluster analysis indicates S. sessiliflorum is isolated in the section. Solanum candidum and S. vestissimum are somewhat isolated as well and have unique karyological traits. Solanum pectinatum has a peculiar karyotype, but in the phenogram it is not particularly separated. Karyotype features suggest that morphological differentiation was not always followed by chromosomal divergence. The origin of the domesticated S. quitoense is no further elucidated by our data.  相似文献

Modern sugar cane varieties are derived from interspecific crosses involving as many as four species. Because a chromosome increase accompanies certain crosses and backcrosses, modern varieties have very high aneuploid chromosome numbers and complicated genetics. Despite this complexity, the chromosome behavior of some modern varieties approaches that of allopolyploids. In achieving homozygosity, therefore, such varieties should respond to inbreeding almost like diploids. The meiotic chromosome behavior of F1 hybrids and modern varieties indicates little or no genetic exchange between chromosomes ofSaccharum officinarum andS. spontaneum. Irradiation may break linkages between desirable and undesirableS. spontaneum genes not ordinarily broken by crossing-over between the chromosomes of the two species. The quick success of nobilizingS. spontaneum (recurrently back-crossing to “noble canes”) depends on a peculiar increase of the chromosomes ofS. officinarum. Experience with nobilizingS. spontaneum should not make breeders impatient when they turn to interspecific crosses unaccompanied by chromosome increases.  相似文献

Karyotypes of 24 diploid (2n=12)Tragopogon species are similar with one long pair of chromosomes (A), two medium-length pairs (B and C), and three short pairs (D, E, and F). These species may be divided into three karyotypic groups: 1) seven species with a satellite on A and on D; 2) 14 species with a satellite on A only; and 3) three species with a satellite on D only. Most species within karyotype groups may be separated from each other either by distinctive features of certain chromosomes or by statistical differences in length of chromosome arms or long arm: short arm ratios of chromosome A. Three tetraploid (2n=24) species had two long pairs (A,A′), four medium-length pairs (B,B′;C,C′), and six short pairs (D,D′;E,E′;F,F′). Three new unusual compound chromosomes have been synthesized in Drosophila melanogaster. They consist of two homologous autosomes joined together in the new order: right arm, left arm, centromere, left arm, right arm, for each of the two major autosomes, and one in which chromosomes 2 and 3 have been combined in the order: right arm of 2, left arm of 2, centromere, left arm of 3, right arm of 3. The attachments of the autosomal arms were accomplished by obtaining chromosome breaks at or very close to the ends of the left arms of the autosomes such that no essential chromosome material has been removed; the compounds derived from them are therefore referred to as entire compounds. These large chromosomes are recovered in progeny with frequencies lower than expectation partly because of zygote mortality associated with these chromosomes, and partly because of a failure of spermiogenesis.  相似文献

Sex chromosomes have been studied in many plant and animal species. However, few species are suitable as models to study the evolutionary histories of sex chromosomes. We previously demonstrated that papaya (Carica papaya) (2n = 2x = 18), a fruit tree in the family Caricaceae, contains recently emerged but cytologically heteromorphic X/Y chromosomes. We have been intrigued by the possible presence and evolution of sex chromosomes in other dioecious Caricaceae species. We selected a set of 22 bacterial artificial chromosome (BAC) clones that are distributed along the papaya X/Y chromosomes. These BACs were mapped to the meiotic pachytene chromosomes of Vasconcellea parviflora (2n = 2x = 18), a species that diverged from papaya ∼27 million years ago. We demonstrate that V. parviflora contains a pair of heteromorphic X/Y chromosomes that are homologous to the papaya X/Y chromosomes. The comparative mapping results revealed that the male-specific regions of the Y chromosomes (MSYs) probably initiated near the centromere of the Y chromosomes in both species. The two MSYs, however, shared only a small chromosomal domain near the centromere in otherwise rearranged chromosomes. The V. parviflora MSY expanded toward the short arm of the chromosome, whereas the papaya MSY expanded in the opposite direction. Most BACs mapped to papaya MSY were not located in V. parviflora MSY, revealing different DNA compositions in the two MSYs. These results suggest that mutation of gene(s) in the centromeric region may have triggered sex chromosome evolution in these plant species.  相似文献

All extant birds share the same sex-chromosome system: ZZ males and ZW females with striking differences in the stages of sex-chromosome differentiation between the primitive palaeognathus ratites and the large majority of avian species grouped within neognaths. Evolutionarily close to ratites is the neotropical order Tinamiformes that has been scarcely explored regarding their ZW pair morphology and constitution. Tinamous, when compared to ratites, constitute a large group among Palaeognathae, therefore, exploring the extent of homology between the Z and W chromosomes in this group might reveal key features on the evolution of the avian sex chromosomes. We mapped MLH1 foci that are crossover markers on pachytene bivalents to determine the size and localization of the homologous region shared by the Z and W chromosomes in two tinamous: Eudromia elegans and Crypturellus tataupa. We found that the homologous (pseudoautosomal) region differ significantly in size between these two species. They both have a single recombination event on the long arm of the acrocentric Z and W chromosomes. However, in E. elegans the pseudoautosomal region occupies one-fourth of the W chromosome, while in C. tataupa it is restricted to the tip of the long arm of the W. The W chromosomes in these two species differ in their heterochromatin content: in E. elegans it shows a terminal euchromatic segment and in C. tataupa is completely heterochromatic. These results show that tinamous have ZW pairs with more diversified stages of differentiation compared to ratites. Finally, the idea that the avian proto-sex chromosomes started to diverge from the end of the long arm towards the centromere of an acrocentric pair is discussed.  相似文献

A comparative analysis of mitotic chromosomes of Theobroma cacao (cacao) and T. grandiflorum (cupuaçu) was performed aiming to identify cytological differences between the two most important species of this genus. Both species have symmetric karyotypes, with 2n = 20 metacentric chromosomes ranging in size from 2.00 to 1.19 μm (cacao) and from 2.21 to 1.15 μm (cupuaçu). The interphase nuclei of both species were of the arreticulate type, displaying up to 20 chromocentres, which were more regularly shaped in cacao than in cupuaçu. Prophase chromosomes of both species were more condensed in the proximal region, sometimes including the whole short arm. Both species exhibited only one pair of terminal heterochromatic bands, positively stained with chromomycin A 3 , which co-localized with the single 45S rDNA site. Each karyotype displayed a single 5S rDNA site in the proximal region of another chromosome pair. Heterochromatic bands were also observed on the centromeric/pericentromeric regions of all 20 chromosomes of cacao after C-banding followed by Giemsa or DAPI staining, whereas in cupuaçu they were never detected. These data suggest that the chromosomes of both species have been largely conserved and their pericentromeric chromatin is the only citologically differentiated region.  相似文献

Region 20 of the polytene X chromosome of Drosophila melanogaster was studied in salivary glands (SG) and pseudonurse cells (PNC) of otu mutants. In SG chromosomes the morphology of the region strongly depends on two modifiers of position effect variegation: temperature and amount of heterochromatin. It is banded in XYY males at 25°?C and β-heterochromatic in X0 males at 14°?C, i.e. it shows dynamic transitions. In PNC chromosomes region 20 is not heterochromatic, but demonstrates a clear banding pattern. Some molecular markers of mitotic heterochromatin were localized by means of in situ hybridization on PNC chromosomes: DNA of the gene su(f) in section 20C, the nucleolar organizer and 359-bp satellite in 20F. The 359-bp satellite, which has been considered to be specific for heterochromatin of the mitotic X chromosome, was found at two additional sites on chromosome 3L, proximally to 80C. The right arm of the X chromosome in SG chromosomes was localized in the inversion In(1LR)pn2b: the telomeric HeT-A DNA and AAGAG satellite from the right arm are polytenized, having been relocated from heterochromatin to euchromatin.  相似文献
