A foliar blight disease of onion (Allium cepa L.) and garlic (Allium sativum L.) hitherto unreported from India has been described from Varanasi, U. P. The infection is confined to the leaves and inflorescence stalks. The conidial stage of the pathogen is more injurious to the plants, whereas the perithecial stage occurs on the inflorescence stalks in isolated locations. The conidiophores and conidia typical of the genus Stemphylium Wallroth are formed during the early development of the disease followed by perithecia predominantly developing on the peduncles. The perithecia were also induced to develop in artificial culture and were identical in diagnostic characters to those from the field. The conidia and ascospores from both host and artificial culture were reciprocally pathogenic to onion and garlic. Morphology of the fruiting structures indicated identity of the pathogen in its conidial stage with Stemphylium vesicarium (Wallroth) Simmons and in the perithecial stage with Pleospora allii (Rabenh.) Ces. and de Not., to which they are referred respectively.  相似文献

The present investigation aimed to isolate the causative agents of onion purple blotch and Stemphylium blight diseases and evaluate the efficacy of certain plant extracts against the two identified pathogens, in vitro and under greenhouse condition. Fourteen isolates of S. vesicarium and two isolates of Alternaria porri were tested for pathogenicity. The results indicated that all isolates were able to produce the symptoms of Stemphylium blight and onion purple blotch diseases with different degrees of severity ranging from 10.42 to 81.25%. A. porri No. 6022 caused the highest disease severity (81.25%), while S. vesicarium No. 6003 was the best one out of the tested 14 isolates (37.5%). Antifungal activity of some aqueous plant extracts (Azadirachta indica, Cydonia oblonga, Datura stramonium, Eucalyptus globulus, Foeniculum vulgare, Ocimum basilicum, Rosmarinus officinalis and Salix mucronata) was assayed in vitro by dry weight technique. The data indicated that there were significant differences between these extracts in their effect on fungal growth of A. porri and S. vesicarium and the best were A. indica and D. stramonium. Under greenhouse conditions, application of the aqueous extract of A. indica either before or after 48?h A. porri inoculation produced the highest reduction in disease severity comprising 70 and 74.7%, respectively. On the other hand, the highest percentage of disease reduction before and after 48?h S. vesicarium inoculation was produced by Ridomil gold plus reached to 84.4 and 95.8% respectively, followed by the aqueous extract of A. indica (74.1 and 89.7, respectively). According to our results, it can be concluded that plant extracts of A. indica and D. stramonium can be used for the biocontrol of purple blotch and Stemphylium blight diseases instead of fungicides to minimise the risks and hazards of using toxic fungicides.  相似文献

Recently, a new disease known as ‘brown spot of European pear’ caused by Stemphylium sp. appeared on the leaves, twigs and fruits of the cultivar Le Lectier in Niigata Prefecture, Japan. Because Stemphylium vesicarium (teleomorph: Pleospora allii), which causes a similar disease in Europe, has been shown to produce host‐selective SV‐toxins in culture filtrates (CFs), SV‐toxin production by Stemphylium sp. in Japan was investigated. In pathogenicity tests, the pathogen induced severe necrotic spots on the leaves of the European pear cultivar Le Lectier, slight spots on cultivar La France and slight or no spots on cultivar Bartlett. The Japanese pear cultivar Nijisseiki was not affected by the pathogen. Culture filtrates of the pathogen were tested for phytotoxicity on cultivars by a leaf necrosis assay. The sensitivity of cultivars to the CFs was consistent with the susceptibility of cultivars to the pathogen infection, indicating the presence of host‐selective toxins. The toxins were purified from the CFs according to the procedure reported for SV‐toxin purification in S. vesicarium. The results indicated that Stemphylium sp. in Japan produces the same SV‐toxins as S. vesicarium in Europe.  相似文献

Seven-day volumetric spore samplers were installed in pear orchards of northern Italy, in the years between 1993 and 2002, and operated continuously during the development of brown spot epidemics (mid-April–mid-August), caused by Stemphylium vesicarium. Aerial concentration of conidia was recorded at 2 h intervals to study their diurnal and seasonal patterns and the influence of weather conditions. The diurnal periodicity of aerial conidia showed a peak around midday and low counts in the dark. The increase in spore concentration was significantly correlated with the reduction of relative humidity and wetness in early morning, and the increase of wind in late morning and afternoon. Conidia of S. vesicarium became easily airborne to form a regular component of the air-spora in pear orchards, while ascospores were caught only sporadically. Differences between years concerned total spore counts and numbers of peaks (defined as days with more than 30 conidia/m3 air per day). Periods with highest spore counts occurred in late-May to early-June (in 2 years), mid to end of June (5 years), or after mid-July (3 years). There was a significant correlation between spore peaks and days with favourable weather conditions, defined as days with air temperature between 15 and 25°C and high humidity, particularly a wet period longer than 10 h. Occurrence of one or more consecutive days with favourable weather conditions determined an increase in the airborne concentration of conidia, which usually lasted some days and then decreased.  相似文献

The conidia of Colletotrichum gloeosporioides were found to be dispersed during rainfall by wash-off and splash mechanisms. The initiation and development of onion anthracnose was found to depend on the frequency of rainfall and the movement of conidial inoculum during rainfall. Experiments conducted under controlled conditions in the laboratory employing splash and wash-off assemblies showed that impacting incident water drops (splash) and flowing water (wash-off) liberated the conidia from the anthracnose lesions of the onion leaf/peduncle. Peak liberation of conidia occurred with 3 to 5 water drops and most of the conidia were removed from the source within 90 seconds. A possibility of the dispersal of conidia of C. gloeosporioides from soil to lower leaf by splash mechanisms and then from the leaves to the neck of the onion bulb and to the bulb by wash-off mechanisms is indicated.  相似文献

Development of insecticide resistance in onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae), populations in onion (Allium spp.) fields and the incidence of the T. tabaci transmitted Iris yellow spot virus have stimulated interest in evaluating alternative management tactics. Effects of straw mulch applied in commercial onion fields in muck areas of western New York were assessed in 2006 and 2007 as a possible onion thrips management strategy. In trials in which no insecticides were applied for thrips control, straw mulch-treated plots supported significantly lower T. tabaci populations compared with control plots. In both years, the action thresholds of one or three larvae per leaf were reached in straw mulch treatments between 7 and 14 d later than in the control. Ground predatory fauna, as evaluated by pitfall trapping, was not increased by straw mulch in 2006; however, populations of the common predatory thrips Aeolothrips fasciatus (L.) (Thysanoptera: Aeolothripidae) were significantly lower in straw mulch plots in both years. Interference of straw mulch in the pupation and emergence of T. tabaci was investigated in the lab and their emergence was reduced by 54% compared with bare soil. In the field the overall yield of onions was not affected by the straw mulch treatment; however, the presence of jumbo grade onions (>77 mm) was increased in 2006, but not in 2007. These results indicate that populations of T. tabaci adults and larvae can be significantly reduced by the use of straw mulch without compromising overall onion yield. The use of this cultural practice in an onion integrated pest management program seems promising.  相似文献

Abstract

The onion thrips Thrips tabaci is one of the most important pests of greenhouse and open-field broccoli, onion and other crops. However, the current strategy of using synthetic pesticides for its control is inadequate and unsustainable, leading to a growing interest in novel and effective biological control alternatives such as entomopathogenic fungi. Among 20 isolates of Beauveria bassiana tested for virulence against T. tabaci in laboratory bioassays, we found strain SZ-26 as the most potent, causing 83–100% mortality in adults at 1×107 mL?1conidia after 4–7 days. Further experiments in greenhouses showed the strain SZ-26 significantly lowered the numbers of adult and larval stages.  相似文献

An outbreak of grey leaf spot caused by Stemphylium solani was observed on tomato in Shandong Province of China in recent years and brought huge economical losses. Fludioxonil is a phenylpyrrole fungicide with strong antifungal activity against S. solani. To evaluate the risk of S. solani developing fludioxonil resistance, a total of 145 field isolates were examined for sensitivity to fludioxonil by measuring mycelial growth. The baseline sensitivity was distributed as a unimodal curve with a mean EC50 value of 0.0659 (±0.0170) µg mL?1. Five mutants with high resistance to fludioxonil (RF > 1000) were obtained by successively selecting on fludioxonil‐amended plates in the laboratory. All the resistant mutants associated with strongly reduced fitness in mycelial growth, sporulation and pathogenicity. Fludioxonil had positive cross‐resistance with procymidone and iprodione, but there was no cross‐resistance with other fungicides including boscalid, fluazinam, azoxystrobin and flusilazole. Based on the current results, resistance risk of S. solani to fludioxonil could be moderate. This is the first report of baseline sensitivity of S. solani to fludioxonil and its risk assessment. In order to delay the resistance development, it is recommended that fludioxonil can be used as one component of the mixture or fungicides with different modes of action should be alternatively used for this disease management.  相似文献

An impact of the term of one-fold insecticide application to reduce the damage of the onion thrips in early white cabbage was established. During the growing season plants were treated with insecticide (abamectin) in three different terms (treatment 1: 9 June, treatment 2: 16 June, treatment 3: 23 June, and treatment 4: untreated). On the exterior leaves of cabbage heads, treated with the insecticide, statistically significantly lower mean index of damage was determined as compared to the untreated plants. No significant differences were found between three different terms of application. The majority of the economically important damages in all of the treatments was found between the 3rd and 6th exterior leaf in the head. The highest mean weight of heads (1517.3 g) and mean net weight of heads I = mean weight of heads - weight of damaged and removed leavesl (1166.3 g) were established in plants which were treated the last. These parameters were the lowest in untreated plants (1083.3 g / 805.6 g). The yield loss due to damaged leaves removal amounted from 22.9% (treatment 2) to 25.6% (treatment 4). Based on the results obtained in this research we concluded that yield loss due to onion thrips attack in plants with one-fold insecticide application is not substantially lower as in untreated plants (though statistically significant differences between them were established), because insecticide cannot reach the interior leaves in the head. Substantial differences in the total and net weight of heads between the treatments and especially between the treated and untreated plants are explained by the fact that feeding of numerous thrips populations in the heads and on the exterior cabbage leaves negatively affects plant physiology and yield. The highest average yield in plants which were treated the last indicates a possibility that insecticide also inhibits plant growth and development to a certain extent.  相似文献

Brown spot is one of the most serious fungal diseases that can affect pear fruits and leaves in the Po valley (Italy). Stemphylium vesicarium is the causal agent of this disease, and several antifungal treatments, repeated throughout the period between bloom and harvest, are needed to control its spread. Many of the most important pear cultivars (such as ‘Abbé Fétel’) are very susceptible to this fungus, while others (such as ‘Bartlett’ and its mutated sports) are known to be resistant. Our research aimed to develop molecular markers linked to this trait. To this end, 92 seedlings derived from an ‘Abbé Fétel’?×?‘Max Red Bartlett’ cross were evaluated for resistance to S. vesicarium for two consecutive years by artificial inoculation with conidia on detached leaves and fruits under controlled conditions in greenhouse. The extent of the lesions was recorded at different time points. A major QTL for susceptibility was located at the lower end of linkage group 15 of ‘Abbé Fétel’. This region was saturated with three SSR markers, and the putative position of a susceptibility gene was also estimated by the single gene mapping approach. This putative gene was located at 2 cM far from the lower end of the linkage group. Molecular markers tightly associated to this locus represent a first step towards the development of MAS (marker-assisted selection) to support the selection of new pear genotypes more resistant to brown spot.  相似文献

The polycyclic phase of Stemphylium vesicarium is the key factor for the forecast and integrated control of purple spot on asparagus. The annual dynamics of airborne conidia were determined under field conditions by conidia traps. From 2013 to 2015, conidia became airborne at the earliest at mid‐July, but the number trapped was considerably enhanced only after mid‐August, early September. The cumulative percentage of trapped conidia was best described using a logistic function depending on the daily temperature sum (base 0°C) accumulated only on days with >0.2 mm of rainfall (R= .81). The germination of conidia was modelled by a generalized beta‐modified Chapman Richards function, and the germ tube length was modelled by a generalized beta‐power function. Conidia germinated in a wide temperature range, with an optimum at 23.3°C, whereas germ tube length had a narrow nearly optimum temperature range around 28.7°C, which indicates that infection by conidia is more restricted by germ tube growth than by germination. The effect of temperature on the number of lesions produced by two strains on green asparagus spears had the narrowest optimum range (optimum at 21.9°C) of all parts of the polycyclic phase. In plant tissue, the spread of the fungus depends on the mycelium growth. The mycelium growth of the four strains, which was modelled with data from a petri dish experiment, had an optimum temperature at 24.7°C.  相似文献

We compared the efficacy of four plant essential oils to repel onion thrips, Thrips tabaci (Lindeman) (Thysanoptera: Thripidae), in the presence of an attractive odour, ethyl iso‐nicotinate in a pasture field. Four horizontal white sticky plates were placed adjacent to (directions: N, S, E, W) a central horizontal white plate (C). After 24 h, in the treatment combination where the four plates were sprayed with essential oil surrounding a central sticky plate sprayed with ethyl iso‐nicotinate, fewer onion thrips were found on the plates treated with sweet marjoram [Origanum majorana L. (Labiatae)] or clove basil [Ocimum gratissimum L. (Labiatae)] (87 and 71% less, respectively) compared to the control treatment of four water‐sprayed plates surrounding a central plate with ethyl iso‐nicotinate. We also compared the distribution of onion thrips on the plates. Relative thrips numbers on each plate were compared with similar (N, S, E, W, and C) plates in the control treatment. There were relatively lower thrips numbers on the south (23% reduction) and west (26% reduction) O. majorana‐treated plates and higher numbers (37% increase) on the central attractant‐treated plate indicating a short‐distance push–pull effect. When four plates were sprayed with the thrips attractant surrounding a central sticky plate sprayed with an essential oil or water (control), only O. majorana reduced the number of thrips on the attractant‐sprayed plates (62% reduction). The distribution of thrips on the different plates within this treatment combination did not change substantially when compared to the distribution in the water‐control treatment. Other essential oils tested (wormwood [Artemisia arborescens L. (Compositae)]) and tea tree [Melaleuca alternifolia (Maiden. & Betche.) Cheel. (Myrtaceae)]) were not effective in any of the treatments. It appears that O. majorana is a promising thrips repellent which could be used for further testing in a push–pull system with the attractant ethyl iso‐nicotinate. The field setup used also proved to be a valuable tool for evaluating the potential of repellents to control onion thrips.  相似文献

It is often assumed that efficient application of a mycoinsecticide involves hitting the target pest insect directly with a lethal dose of conidia. However, secondary pick‐up of conidia from surrounding vegetation may be a more important source of inoculum. We have investigated ways of increasing conidia acquisition by enhancing host movement. The aphid alarm pheromone, E‐β‐farnesene, significantly increased mortality among peach potato aphids. Myzus persicae Sulzer, that were exposed for 24 h to discs of green pepper leaf sprayed with conidia of Verticillium lecanii (Zimmerman) Viegas then transferred to fresh untreated discs to allow disease development. A more practical approach to increasing conidia pick‐up appears to be the use of sub‐lethal doses of the chloronicotinyl insecticide imidacloprid. One percent of the recommended dose, applied systemically, dramatically increased aphid movement; quantified by image analysis of videotaped aphid behaviour. This resulted in greater mortality from mycosis in experiments where aphids were exposed to insecticide‐treated leaf discs that had been sprayed with fungal conidia. A comparison with results from an experiment where conidia were sprayed directly onto aphids which were feeding on insecticide‐infused pepper discs established that synergy was due to an indirect effect of the insecticide, i.e. through increased movement, rather than a direct effect viz. predisposition of insecticide‐weakened insects to disease.  相似文献

Pepino (Solanum muricatum var. pepino) plants were found affected by an extensive leaf spot caused by plant pathogenic fungi during a survey in the Cameron highlands, Pahang state, Malaysia. Symptomatic leaf samples were collected from infected pepino plants and cultivated on PDA medium, and the pathogen was isolated and purified; then, consequently, all isolates were identified as Stemphylium lycopersici on the basis of their cultural and morphological characteristics and combined sequences of the internal transcribed spacer (ITS) and glyceraldehyde‐3‐phosphate dehydrogenase (gpd) regions. A pathogenicity assay on detached leaves further confirmed that S. lycopersici causes leaf spot disease. To the best of our knowledge, this is the first report of S. lycopersici causing leaf spot on pepino in Malaysia and worldwide.  相似文献

Purple blotch (Alternaria porri), having symptom similar to stemphylium blight (Stemphylium vesicarium), is one of the important diseases that significantly lowered shallot yield in Indonesia. However, stemphylium blight has never been reported. Purple blotch pathogens from farmers’ crops in Bantul and experimental crop in Centre of Innovative Agricultural Technology Universitas Gadjah Mada (CIAT-UGM) Sleman Regencies were identified to observe whether Stemphylium sp. was found with A. porri. Thirteen isolates obtained showed variability in colony texture, colour and pigmentation. DNA fingerprinting through Polymerase Chain Reaction of BOX and Enterobacterial Repetitive Intergenic Consensus region dispersed those isolates into two main groups. Identification using Alternaria-specific primers, Dir1ITSAlt/Inv1ITSAlt revealed that 10 isolates were Stemphylium sp. and 3 isolates were Alternaria sp. A. porri was only identified from CIAT-UGM. This finding emphasise that the Alternaria-specific primer was able to amplify Stemphylium sp. Besides, stemphylium blight disease may have been occurred in farmers’ crops in Bantul.  相似文献

Background  

The western flower thrips (Frankliniella occidentalis [Pergande]) is one of the most important insect herbivores of cultivated plants. However, no pesticide provides complete control of this species, and insecticide resistance has emerged around the world. We previously reported the important role of jasmonate (JA) in the plant's immediate response to thrips feeding by using an Arabidopsis leaf disc system. In this study, as the first step toward practical use of JA in thrips control, we analyzed the effect of JA-regulated Arabidopsis defense at the whole plant level on thrips behavior and life cycle at the population level over an extended period. We also studied the effectiveness of JA-regulated plant defense on thrips damage in Chinese cabbage (Brassica rapa subsp. pekinensis).  相似文献

In insect reverse genetics, dietary delivery of interfering RNAs is a practical approach in nonmodel species, such as thrips, whose small size, and feeding behavior restricts the use of other delivery methods. In a laboratory context, an unsuitable diet could confound the interpretation of an RNA interference (RNAi) phenotype, however well-formulated artificial diets can minimize experimental variability, reduce the need for insect handling, and can further be used for roles, such as delivering double-strand RNA (dsRNA)-expressing recombinant bacteria. In this study, artificial diets for oral delivery of dsRNA were developed for two important pest thrips species, western flower thrips (Frankliniella occidentalis) and onion thrips (Thrips tabaci), with the goal of (a) stimulating feeding behavior, (b) supporting optimal growth rates of dsRNA-expressing symbiotic bacteria, and (c) nutritionally supporting the thrips for sufficient periods to observe RNAi phenotypes. The efficacy of artificial diets for ingesting “naked” dsRNA or dsRNA-expressing symbionts and dsRNA delivery via host plant uptake was evaluated. Compared with previously published diet formulations, new combinations based on tryptone, yeast, and soy were superior for enhancing feeding and longevity. However, simply adding “naked” dsRNA to an artificial diet was an unreliable form of RNAi delivery in our hands due to dsRNA degradation. Delivery via host plants was more successful, and the new diet formulation was suitable for symbiont-mediated dsRNA delivery, which we believe is the most convenient approach for large-scale knockdown experiments. This study, therefore, provides alternative methodologies for thrips rearing, dietary RNAi delivery, and insights into the challenges of performing dietary RNAi in nonmodel insects.  相似文献

The herb Stevia rebaudiana is a potential source of low-calorie sweeteners. In 1995, a severe leaf spot and blight was observed in stevia production fields and research plots in the provinces of Ontario and British Columbia, Canada. The disease was characterized by angular, shiny, olive-grey lesions that rapidly coalesced and were often surrounded by a chlorotic halo. Leaves quickly became necrotic and often dropped off the plant. The disease progressed upwards in the foliage during the growing season. A Septoria sp. was isolated from diseased leaves. Ten isolates (five from each of the two provinces) of the Septoria sp. were compared with respect to conidial size. Across isolates, conidia lengths and widths overlapped (grand means for length and width were 71.4 μm and 1.4 μm, respectively). Conidiogenesis was holoblastic. Morphological characteristics and disease symptoms were similar to those of Septoria steviae, previously reported only from Japan. It was concluded that the Canadian isolates belonged to S. steviae. Isolates from Canada did not differ significantly from one another with respect to effects of temperature on colony growth or germination of conidia. Optimum temperatures for these parameters were between 20 and 25°C. In field trials, the pathogen was shown to successfully over-winter in diseased leaf tissue. In order to develop procedures for identification of resistant germplasm and greenhouse screening of candidate fungicides, effects of leaf wetness period, inoculum concentration, and plant age on disease development were determined. Thirty-six hours of leaf wetness were required for consistent development of leaf spots. Inoculum concentrations of 5 × 105 conidia/ml or more were required to produce high disease severities; 6-week-old plants were more susceptible than older plants. In the growth chamber, greenhouse, and field trials, germplasm selections with high levels of resistance to S. steviae were identified. This is the first report of resistance to this disease in S. rebaudiana.  相似文献

When given a choice of feeding sites in laboratory arenas, Delia antiqua (Meigen) larvae exhibited a 3: 1 bias for internal vs outer sections of onion bulbs and distributed themselves non-randomly among identical preferred onion sections. Larval clustering did not appear to be correlated with increased fitness in the laboratory; larval development was identical across the range of densities from 1 to 50 larvae per onion. Larvae feeding on preferred internal sections developed 14% faster, were 38% heavier upon pupation, and were two times more likely to survive to the adult stage than larvae feeding on outer sections. D. antiqua larval food preference was therefore positively correlated with increased fitness.  相似文献
