An experiment was conducted to test the hypothesis that Escherichia coli (E. coli ) endotoxin is readily absorbed from uteri of early postpartum cows and that the absorbed endotoxin provokes systemic relcase of prostaglandins. Eleven postpartum Holstein dairy cows (aged 3 to 7 yr) with normal puerperium were selected and divided into a treatment group (n=7), which received intrauterine infusions of E. coli endotoxin, and a control group (n=4), which received intrauterine infusions of 10 ml of saline on Days 5 and 20 post partum. Blood samples were collected once every 30 min for 6 h starting from the time of infusion. Harvested sera samples were analyzed for concentrations of stable metabolites of prostacyclin (PCM), prostaglandin F(2alpha) (PGFM), and thromboxane A(2) (TXB(2)). Plasma samples were qualitatively tested for the presence of endotoxin. Endotoxin was detected in the plasma samples of cows that received endotoxin on Day 5 post partum 4 h after the infusion. Endotoxin was not detected in any of the samples from control cows on Days 5 and 20 post partum or from treatment group cows on Day 20 post partum. Cows treated on Day 5 post partum showed increases in serum PGFM concentrations from 710 +/-64pg/ml to peak concentrations of 1223 +/- 47 pg/ml within 2 h, followed by a decline to baseline concentrations within 4 h. The amount of PGFM released in treated cows on Day 5 post partum was higher (P < 0.05) than in control cows on Day 5 or in treated and control cows on Day 20 post partum. Serum PCM concentrations increased from 156+/-24 pg/ml to peak concentrations of 1348+/-127 pg/ml within 1 h. The amount of PCM released in treated cows on Day 5 postpartum was higher (P< 0.05) than in control cows on Day 5 or in treated and control cows on Day 20 post partum. The TXB(2) concentrations increased from 315+/-38 pg/ml to peak concentrations of 5043 +/- 242 pg/ml within 1 h and fell to baseline concentrations within 5 h. The amount of TXB(2) concentrations released in treated cows on Day 5 post partum was significant (P < 0.05) compared with those of cows in the other groups. The results support the hypothesis that uteri of early postpartum cows are capable of absorbing endotoxin, and the absorbed endotoxin provokes changes in the serum concentrations of prostanoids.  相似文献

The objective of this study was to determine the value of using plasma concentrations of PGFM to diagnose subclinical endometritis in the dairy cow, and its relationship to subsequent fertility. A total of 274 cows between 24 to 29 d post partum was divided into 4 groups on the basis of clinical features of the uterus and ovary. Cows in Group 1 (n = 74) had a normal, involuting uterus and a CL on the ovary; cows in Group 2 (n = 51) had a normal, involuting uterus but no CL on the ovary; cows in Group 3 (n = 83) did not have a normal, involuting uterus but had a CL on the ovary; and cows in Group 4 (n = 66) did not have a normal, involuting uterus or a CL on the ovary. A blood sample was obtained from each cow on the day they were placed on the study, and plasma concentrations of PGFM and P4 were determined using RIA. Cows were artificially inseminated (AI) at the first observed estrus after Day 60 post partum, and pregnancy was determined by palpation of the uterus per rectum between 45 and 50 d postAI. Reproductive responses evaluated were conception rate to first service, days open, and percentage of cows pregnant by 90, 120, 150 and 180 d post partum. Data were analyzed using GLM procedures of SAS and a 2 x 2 factorial with contrast procedures. Polynomial regression analysis was used to determine the shape of the PGFM, P4 and fertility curves. There was no difference among mean PGFM concentrations of cows in each group. The rate of decline of plasma PGFM concentrations was lower in cows with an abnormal uterus and a CL on the ovary compared with those without a CL. A lower percentage of cows with abnormal uteri was pregnant by 90 d post partum compared with cows with normal uteri. From the results of this study, it was concluded that plasma PGFM concentrations between Days 24 to 29 post partum were not effective in identifying cows with subclinical endometritis.  相似文献

The objective of this study was to investigate the effect of different body condition score (BCS) at 30 days before calving (-30 days) induced by a differential nutritional management from -100 days until -30 days on productive parameters, the interval to first ovulation and blood parameters in primiparous and multiparous Holstein cows under grazing conditions until 60 days post partum. The experimental arrangement was a randomized complete block design, where cows were blocked according to BW and expected calving date and then randomly assigned to different nutritional treatments from -100 to -30 days relative to calving to induce different BCS. As the assignment of cows to treatments was random, cows had to lose, maintain or gain BCS; thus, different planes of nutrition were offered with approximately 7, 14 or 20 kg dry matter per day. The BCS score was assessed every 15 days and animals were reassigned in order to achieve the desired BCS at -30 days. Only animals that responded to nutritional treatment were considered and this was defined as follows: primiparous and multiparous high cows (PH and MH) had to gain 0.5 points of BCS, primiparous low (PL) had to lose 0.5 points of BCS and multiparous low (ML) had to maintain BCS at least in two subsequent observations from -100 to -30 days. From -30 days to calving, primiparous and multiparous cows (P and M cows) were managed separately and cows were offered a diet once a day. From calving to 60 days post partum, cows of different groups grazed in separate plots a second year pasture. Cows were also supplemented individually with whole-plant maize silage and commercial concentrate. Cows had similar BCS at -100 days and differed after the nutritional treatment; however, all groups presented similar BCS at 21 days post partum. The daily milk production and milk yield at 60 days post partum was higher in M than P cows. The percentage of milk fat was higher in PH cows compared with PL cows. Concentrations of non-esterified fatty acids (NEFA) were affected by the BCS at -30 days within parity, and in PH cows the concentration of NEFA was higher than in PL cows. The concentrations of total protein were higher in M cows. A lower probability of cycling was found in PL than in PH cows (P < 0.05) and in ML than in MH cows (P < 0.05). Treatment affected various endocrine/metabolic profiles according to parity, suggesting that the metabolic reserves signal the productive/reproductive axis so as to induce a differential nutrient partitioning in adult v. first-calving cows.  相似文献

The objective of this study was to determine the effect of GnRH treatment in dairy cows differing in body condition score. A total of 2437 primiparous and multiparous cows was allocated randomly to receive either GnRH (100 mug) or placebo at the post partum exam (25 to 35 d) and at the first breeding. Complete records were available from 1906 cows: Group 1 (n = 535) placebo and placebo; Group 2 (n = 489) GnRH and placebo; Group 3 (n = 438) placebo and GnRH; and Group 4 (n = 444) GnRH and GnRH. The change of body condition score during early lactation was determined on a 1 to 5 scale (1 = emaciated to 5 = obese). Cows with a high body condition score (>/=3.0) at the first breeding had 8.8 fewer days to first service and 6.4 fewer days open but more services per conception (0.16) than cows with low body condition score (<3.0). Conception rate improved when GnRH was administered at the first breeding to cows with a body condition score of <3.0 at the first breeding regardless of parity. The administration of GnRH at the post partum exam decreased the conception rate in first lactation cows but was beneficial for cows in second and greater lactations. Thus the efficacy of GnRH may not be consistent in all parities and body condition groups.  相似文献

Five acyclic spring-calving beef cows (20-40 days post partum) were bled every 15 min for 60 h and thereafter every 6 h for 5 days. Gn-RH (5 micrograms) was injected every 2 h for 48 h, starting 12 h after sampling began. Pulsatile patterns of LH release occurred synchronously in response to injection and 4 of the 5 treated animals subsequently ovulated and completed at least one full ovarian cycle. Four of 6 similar control cows were bled every 10 min for 8-h periods at equivalent times post partum. Pulses of LH were seen after approximately Day 25 post partum with a mean pulse frequency of 0.5 per h. There was little evidence of a discrete pulsatile mode of FSH release in any of the treated or control cows. The time to the first significant progesterone rise in the 4 treated and ovulating cows (34.5 +/- 5.6 days post partum) was significantly shorter (P less than 0.05) than in the 6 control animals (66.3 +/- 11.4 days).  相似文献

The relationship between accumulation of triacylglycerols in the liver and first ovulation post partum was studied in an experiment with 32 dairy cows. During the dry period, 16 treated cows were fed a high energy diet to induce overconditioning, while 16 control cows were fed a recommended diet. After parturition, the treated cows entered deeper negative energy balance state, required more days to reach the nadir of negative energy balance, required more days to regain positive value of energy balance, and lost more body weight than the control cows. The treated cows also accumulated greater amounts of triacylglycerols in the liver and had higher concentrations of nonesterified fatty acids and 3-hydroxybutyrate post partum than the control cows. Although the number of days to first ovulation did not differ significantly between the 2 groups, the results of pooled data demonstrated that the liver triacylglycerol concentration was positively correlated with the number of days to first ovulation. In conclusion, negative energy balance caused the body fat of cows to mobilize. As a consequence, the cows accumulated greater amounts of triacylglycerols in the liver, and this accumulation was associated with an increase in the interval from parturition to first ovulation.  相似文献

Two experiments were performed to determine the effects of repeated administration of PGF2alpha in the immediate post partum period on the prevalence of clinical endometritis at 22 and 58 days post partum, and the probability of pregnancy at first insemination, in post partum dairy cows. In Experiment 1, 228 cows on day 7 post partum were used. Cows in Group 1 (n = 114) were treated twice with PGF2alpha (25 mg, im) 8 h apart on days 7 and 14 post partum, and only once on days 22 and 35 post partum. Cows in Group 2 (n = 114) served as untreated controls. Vaginoscopy and transrectal palpation of the genital tract were done on days 22 and 58 post partum. Cows in both groups were inseminated at estrus after a voluntary waiting period of 100 days. Pregnancy was determined by transrectal palpation between 45 and 50 days after insemination. Repeated administration of PGF2alpha in the immediate post partum period did not reduce the prevalence of clinical endometritis on days 22 or 58 post partum (65 versus 62% on day 22 and 28.3 versus 35.2% on day 58 in Groups 1 and 2, respectively). There was no significant difference in the probability of pregnancy at first insemination between cows in both groups. In Experiment 2, 418 cows on day 7 post partum were used. Cows in Group 1 (n = 209) were treated twice with PGF2alpha (25 mg, im) 8h apart on days 7 and 14 post partum, and only once on days 22 and 35 post partum. Cows in Group 2 (n = 209) served as untreated controls. Cows in both groups were subjected to the Presynch and Ovsynch protocols on days 49 and 75 post partum, respectively. Pregnancy was determined by transrectal ultrasonography between days 29 and 32 after timed insemination. There was no significant difference in the probability of pregnancy at first insemination between cows in both groups. In conclusion, repeated administration of PGF2alpha to cows in this herd had no effect on the prevalence of clinical endometritis at 22 and 58 days post partum, and that there was no effect on the probability of pregnancy after insemination at estrus among cows with a voluntary waiting period of > 100 days, or at timed AI at 85 days in milk when Presynch was employed.  相似文献

Beef cows (n = 294) calving between November and April in six states were used to evaluate the effects of postpartum diet and calf separation on body weight, body condition score (BCS), reproductive performance, and weaning weight of calves. In each state, half of the 48 cows that calved during 60 d were group fed an additional 4.5 kg of a 20% crude protein supplement daily for 28 d starting an average of 30 d post partum (flush). Calves were separated from half of the flush and half of the nonflush cows for 48 h at 14 and 28 d after the beginning of flush. Progesterone was quantified in plasma samples obtained weekly during a 56-d breeding period to assess ovarian luteal activity. The breeding period started at the first calf separation. BCS ranged from 3.3 to 5.6 among states (on a scale of 1 to 9) at the start of the flush but was similar for treatments within a state. There was a state x flush (P < 0.008) effect on body weight at the end of the flush period. Weaning weights were influenced by state x separation x flush (P < 0.06) and were greatest for flush nonseparated calves in five of six states. There were state x flush (P < 0.08) and separation (P < 0.04) effects on ovarian luteal activity at the start of the breeding period. Flush and separation tended to increase ovarian luteal activity. During the breeding period, ovarian luteal activity was influenced only by state but there was a state x separation x flush effect (P < 0.001) on the number of weeks post partum to onset of ovarian luteal activity. Conception rate and days postpartum to conception were not influenced by either separation or flush but were affected by state (P < 0.001). These data indicate that flushing may increase weaning weights of calves and calf separation may hasten the onset of postpartum ovarian luteal activity, but conception rate and days postpartum to conception for cows in thin to moderate body condition were not influenced by the calf separation or flushing treatments.  相似文献

Doses of 125, 250 or 500 micrograms LH-RH were injected i.m. into suckled beef cows on approximately Day 11 of an oestrous cycle synchronized by prostaglandin treatment. There was a positive linear relationship between dose of LH-RH and the area under the measured LH peak. Administration of 500 micrograms LH-RH as a single injection to suckled cows 13-32 days post partum resulted in LH release but failed to induce normal ovarian activity. A small transient rise in plasma progesterone for 6--9 days occurred at the expected time after injection in 50% of animals. Administration of 500 micrograms LH-RH to suckled beef cows approximately 20--30 days post partum and a second injection approximately 10 days later at the time when the resulting transient rise in plasma progesterone had returned to basal values induced normal cyclic activity (as shown by progesterone concentrations and observed oestrus) at 35 days compared with 70 days for untreated controls. Pituitary responsiveness to LH-RH, as assessed by LH levels, was found to increase up to 20 days post partum.  相似文献

The effects of the stress of dystocia on the adrenocortical function post partum in cows (n = 6) requiring a cesarean section were assessed by an adrenocorticotropic hormone (ACTH) stimulation test. Nine cows that calved normally were used as controls. The plasma glucocorticoid levels, before and 60 min after an intramuscular injection of 25 IU ACTH, were 4.4 +/- 0.5 (mean +/- SD) and 21.5 +/- 2.4 ng/ml 1 d post partum, 1.9 +/- 0.3 and 18.4 +/- 2.9 ng/ml 4 d post partum and 2.7 +/- 1.1 and 14.6 +/- 3.3 ng/ml 8 d post partum, respectively. Corresponding values of glucocorticoids in cows with normal calving were 4.5 +/- 3.6 and 18.1 +/- 5.2 ng/ml 1 d post partum, 1.7 +/- 1.3 and 13.2 +/- 5.5 ng/ml 4 d post partum and 1.9 +/- 1.7 and 14.6 +/- 3.3 ng/ml 8 d post partum. There were no statistically significant differences of the values between cows with dystocia requiring a cesarean section and cows with normal calving. The results indicate that dystocia requiring a cesarean section like normal calving does not lead to significant depression of the adrenocortical function post partum.  相似文献

Three newly delivered dairy cows were given prostaglandin F2α during the immediate postpartum period. PGF2α was administered from day 3–13 post partum in doses of 25 mg twice daily. Endogenous release of PGF2α and progesterone was studied in blood plasma during the experimental period. Rectal examination of the uterus was performed every second day in order to establish the end of uterine involution. Uterine involution in the three cows was completed days 16, 23 and 20, respectively. These figures are to be compared with earlier investigations of uterine involution times, which show about 27 days. It was concluded that PGF2α had a positive effect on the uterine muscular tone.  相似文献

We have used milk progesterone analysis to monitor reproductive function in lactating dairy cows and have then related this reproductive function to a variety of metabolic variables. Monitoring of cows (n = 41) during the period of onset of luteal function (first milk progesterone reading>5 ng/ml) revealed that delayed onset was associated with increased milk yield and greater loss of body weight and body condition but was not related to plasma metabolite or leptin concentrations. Further monitoring of reproductive function in these 41 cows and an additional 33 cows (total n = 74) during the mating period (from weeks 6 to 14 post partum) identified reproductive cycle abnormalities in 29 (39.2% of animals). The occurrence of cycle abnormalities was associated with increased milk yield (P < 0.05), elevated plasma beta hydroxybutyrate (P < 0.05) and reduced plasma leptin (P < 0.01) concentrations as well as a lower (P < 0.05) rate of gain of body weight and condition score but was not associated with plasma urea or glucose concentrations. Furthermore, cows exhibiting cycle abnormalities had a longer (P < 0.01) interval to first service and a smaller percentage had conceived by 100 days post partum (34.5% versus 66.7%; P < 0.01). These results provide further evidence that impaired reproductive function during the post partum period in dairy cows is caused by a poor energy status and not elevated urea concentrations. Reduced plasma leptin concentrations in animals suffering reproductive dysfunction further supports this view.  相似文献

GnRH (250 μg) was administered intravenously in a total of 121 experiments carried out on 21 cows during the period from 180 days ante (ap) to 50 days post partum (pp). Additionally in one group of animals prolactin secretion was inhibited after parturition by means of 3 intramuscular injections of 150 mg Bromocryptine (CB-154) on days 1, 4 and 7 pp. LH response (peak height, area under the dose response curve) was about the same from 150 to 60 days ap, then decreased significantly towards parturition and was lowest during the first 6 days post partum. At a later time the Lh response was more pronounced than during pregnancy. The FSH response decreased significantly during the last 9 days ap, remained low during the frist 6 days pp and increased thereafter. There was no significant influence of prolactin inhibition on LH and FSH values (except for the total FSH released on day 50 pp). Whereas in all GnRH treated animals pronounced pituitary gonadotropin responsiveness was measurable (except during the period around parturition), the variation of the LH response pp was much higher than ap. The LH results gave some indication of the wide range of response pattern for this hormone after parturition which might be one reason for the individuality in the initiation of a new estrous cycle post partum in cattle.  相似文献

Mature beef cows were actively immunized pre partum (N = 5) or post partum (N = 10) against a PGF-2 alpha-ovalbumin conjugate or against ovalbumin alone (control; N = 5). All cows in the control group exhibited first oestrous cycles which were of short duration (less than or equal to 12 days). Mean specific serum binding to [3H]PGF-2 alpha in the control group was consistently less than 1%. In the pre-partum PGF-2 alpha-immunized cows, lifespan and progesterone secretion of the first corpus luteum formed post partum was maintained for greater than 39 days. Specific serum binding to [3H]PGF-2 alpha in pre-partum and post-partum PGF-2 alpha-immunized cows was elevated. Lifespan of the first corpus luteum formed in post-partum PGF-2 alpha-immunized cows was short (less than 10 days; N = 1), normal (mean = 22 days; N = 4) or maintained (greater than 31 days; N = 5). Luteal lifespan was dependent upon serum PGF-2 alpha antibody titres, with cows exhibiting higher titres frequently having prolonged luteal lifespans after first ovulation. We conclude that active immunization of beef cows against PGF-2 alpha extends the lifespan and progesterone secretion of corpora lutea anticipated to be short-lived. These results support the concept that the shorter lifespan of some corpora lutea in post-partum cows is due to a premature release of PGF-2 alpha from the uterus.  相似文献

During a 13-month period, 64 lactating dairy cows of 2 genetic lines, Holstein and crossbred, housed indoors year-round were subjected to 2 superovulations and embryo collections within 112 days post partum. Half of the follicle stimulating hormone (FSH) treatments were given in a descending dosage regimen (Treatment A; 6.5 mg, 5.5 mg, 4.5 mg, and 3.5 mg, twice a day, total 40 mg) over 4 days; the remaining half of the treatments were administered in a constant dosage regimen (Treatment B) of 5 mg twice a day over 4 days. There were no significant differences due to treatment in the number of cows stimulated (more than 2 corpora lutea) nor in the number of ova/embryos collected. However, embryos were obtained from more cows (P<0.05) when treated with the descending dosage regimen. More cows (P<0.05) were stimulated by the superovulatory treatment during the first period than during the second period regardless of the regiment used, treatment A or B. More embryos (P<0.05) were obtained from the Holstein line than from the crossbred line. Fifty-two cows were inseminated at least once after the second embryo collection. Overall, 41 cows (79%) became pregnant after the second collection, requiring up to 4 services. These results suggest that the reproduction of dairy cattle housed indoors year-round is not adversely affected by 2 superovulation treatments and embryo collections within 112 days post partum. The question as to whether the administration of FSH is more efficacious in a descending dosage regimen or a constant dosage regimen was not resolved.  相似文献

This study was conducted to determine if Escherichia coli endotoxin was absorbed from the equine uterus and if exogenous progesterone and estrogen affected the absorption of intrauterine endotoxin. Six mature anestrous pony mares were used in three consecutive crossover experiments (Periods) with a 14 day recovery between each period. Mares were randomly assigned to one of two treatment groups (three mares per group) and received an intrauterine infusion of either saline or endotoxin. Treatment groups were reversed and readministered after 14 days completing a crossover design (Period 1). During Periods 2 and 3 the mares were treated with estradiol-17beta (4.0 mg/day) or progesterone (400 mg/day), respectively, for 5 days prior to receiving intrauterine infusions. On the day of intrauterine infusions rectal temperatures and jugular blood samples were obtained at 30 to 60 minute intervals. Blood samples were analyzed for total white blood cell counts and by Limulus Amebocyte Lysate Assays. There were no significant alterations in the observed parameters among the various treatment groups. These results indicated that intrauterine E. coli endotoxin was not absorbed from the uterus of anestrous pony mares in large enough amounts to produce a morbid condition and that prior exposure to exogenous progesterone or estradiol-17beta did not affect the absorption of endotoxin from the equine uterus.  相似文献

Endometritis in breeding cattle occurs during the postpartum period, and is associated primarily with contamination of the reproductive tract involving Arcanobacter pyogenes (formerly Actinomyces pyogenes) together with Gram-negative anaerobes. Polymorphonuclear inflammatory cells (PMNs) contribute partly to the defense mechanisms against micro-organisms contaminating the vagina and uterine lumen, whose phagocytic activity depends on bacterial opsonisation by humoral antibodies; significant numbers of lymphocytes are also present. Whilst leukocyte numbers in the uterine lumen are relatively high during metoestrus and dioestrus compared to other phases of the oestrous cycle, their functional activity is unaffected. Humoral antibody concentrations in the reproductive tract are stimulated following exposure to local antigen, and the response is site dependent; of the several different classes of immunoglobulins, IgG predominates in the uterus and IgA the vagina. Only a portion of the total IgG1 found on the uterine lumen is synthesised locally in the endometrium, the remainder and all of the IgG2 is derived from the local uterine blood supply. Generally, concentrations of immunosuppressant proteins present in the uterine lumen increase under progesterone dominance, and these inhibit lymphocyte proliferation, making the uterus more susceptible to infection. The relationship between uterine susceptibility to micro-organism contamination and the luteal phase of the oestrous cycle is still unclear. Intrauterine infusion of immunomodulators such as E. coli lipopolysaccharides (LPS) or oyster glycogen, in healthy cows and those with endometritis, stimulates leukocytes to migrate into the uterine lumen. At a dosage rate of 100 microg, lipopolysaccharides are not absorbed by the healthy endometrium and do not alter the oestrous cycle length. It is unknown, whether a similar dose can be absorbed through an inflamed endometrium in naturally occurring cases of endometritis to cause systemic illness. Currently, prostaglandin F2alpha is recommended for treating endometritis in both cycling and non-cycling cows, but its mode of action in non-cycling cows is not fully understood. The efficacy of endometritis treatment using an intrauterine infusion of an immunomodulator in cases occurring naturally has not been determined on a large scale.  相似文献

The aim of this study was to determine the time taken for complete uterine involution in Bulgarian Murrah buffaloes following normal parturition and oxytocin stimulated milking; and to establish the time course of the change in size of the uterine horns, the cervix and caruncles between parturition and involution by means of ultrasonography. There were 17 animals in the study aged 3-6 years and average parity of 2.17 ± 0.18. They were administered 20 IU oxytocin 15 min before each milking. Rectal palpation and transrectal ultrasonography were performed at 3 d intervals from Days 1 to 34 post partum. The involution of the non-gravid and gravid uterine horns, and the cervix was complete by Days 22 and 25 post partum when their diameters were 2.7 ± 0.4 cm, 2.8 ± 0.3 cm and 3.12 ± 0.4 cm, respectively. Caruncles underwent rapid regression until Day 10 post partum. It was not possible to determine the dimensions of the caruncles after that time. The cumulative percentage of animals whose uterus was located in the pelvic cavity increased from 24% at Day 10 post partum to 100% at Day 34 post partum. The combination of rectal palpation and transrectal ultrasonography provided a reliable method of evaluating changes in the uterus over time and determining the time of uterine involution. The present study showed that complete uterine involution, with the uterus located in the pelvic cavity, was achieved by Day 34 after parturition in all 17 Bulgarian Murrah buffaloes treated with oxytocin before milking.  相似文献

Antimony dextran glycoside (RL-712, manufactured by Rosco A/S Pharmaceutical Taastrup, Denmark) produced a remarkable decrease in Leishmania donovani densities in hamster spleen and liver when administered in a single intraperitoneal (ip) dose of 500 mg Sb/kg body weight or in 600 mg Sb/kg body weight divided into two or three equal parts injected at weekly intervals beginning 15 days after intracardial inoculation of 10 million amastigotes, and also in a single ip dose of 300 mg Sb/kg body weight 14, 7, and 0 days before intracardial inoculation of 10 million amastigotes. No parasites were detected on using a single dose of 500 mg Sb/kg body weight followed by 100 mg Sb/kg body weight one week later.When the drug was administered in a dose of 500 mg Sb/kg body weight either 10 days before subcutaneous inoculation of 60 million promastigotes or eight days before intracardial inoculation of five million promastigotes, no parasites were recovered by smear or culture from the spleen and liver.Results show the prophylactic and therapeutic actions of this drug against L. donovani in the hamster.  相似文献

Embryos were collected non-surgically from the tip of one uterine horn of 23 lactating dairy cows on Day 7 of pregnancy. Embryos were classified on the basis of morphological criteria as normal (n = 12) or abnormal (n = 13). Abnormal embryos were further classified as cleavage stage (n = 9) or morula/blastocyst (n = 4). Cows producing an abnormal embryo did not differ in days post partum at oestrus, age or parity from cows producing a normal embryo. Cows with an abnormal morula/blastocyst also did not differ with respect to days post partum at oestrus from cows with abnormal cleavage-stage embryos but cows with an abnormal morula/blastocyst were significantly older and of greater parity than cows with an abnormal cleavage-stage embryo. Hepes-saline-PVP solution (30 ml) was initially infused into the uterine tip, mixed and then withdrawn with a syringe. Analysis of this fluid revealed that the concentrations of glucose, total protein, calcium, magnesium and potassium were significantly higher in the flushings from the uterus of cows with abnormal embryos than from cows with normal embryos and zinc and phosphorus tended to be higher in the uterine flushings of cows with abnormal embryos. Phosphorus, total protein, calcium and magnesium tended to be higher in the flushings from cows with abnormal morulae/blastocysts than from cows with abnormal cleavage-stage embryos. Plasma progesterone did not differ between cows with normal or abnormal embryos or in cows with abnormal morulae/blastocysts or abnormal cleavage-stage embryos.
