A high level of genetic variability was detected in North American fall armyworm (FA W), Spodoptera frugiperda (J. E. Smith), populations by restriction fragment length polymorphism (RFLP) analysis of genomic DNA. In nearly all cases individual larvae could be differentiated using 22 probe-enzyme combinations. Laboratory colonies formed distinct groups based on RFLP analysis. Individuals from a colony of the previously determined ‘rice strain’ formed a group showing obvious deviation in RFLP patterns from the other five populations, which included a population from a ‘com strain’ colony. The results were consistent with previous studies using allozymes which indicated that there are two genetically differentiated forms of the fall armyworm. Diagnostic markers were also found that distinguish these two groups.  相似文献

Excessive use of chemical insecticides to control Spodoptera frugiperda in maize (Zea mays L.) crops in Colombia has resulted in pest resistance and environmental contamination. A Colombian multicapsid nucleopolyhedrovirus (SfMNPV) was produced in laboratory reared larvae and formulated as a wettable powder by microencapsulating the viral occlusion bodies (OBs) with a methacrylic acid polymer. Microencapsulated OBs were tested for insecticidal activity in laboratory, greenhouse and field bioassays, and were then sprayed onto a commercial crop where their effect on arthropod populations was estimated. In laboratory bioassay, microencapsulated and non-microencapsulated OBs caused a larval mortality of 98 and 96%, respectively. The percent of plants showing recent feeding damage in a greenhouse experiment oscillated between 2.5 and 7.5% when OBs were applied compared to 70% observed in the control. In two field trials, the highest dose of 1.5×1012 OBs/ha (1500 g/ha) was selected as the recommended rate for field application in maize crops, with efficacies higher than 70%. Total arthropod populations were negatively affected by chemical pesticides; whereas the OBs formulation did not affect the diversity of these organisms. Our results indicate that microencapsulated SfMNPV has the potential to be commercialised as an alternative to substitute or complement chemical insecticides in fall armyworm integrated management programmes.  相似文献

The fall armyworm (FAW), Spodoptera frugiperda (Lepidoptera: Noctuidae), is the most important pest of maize in many countries. Entomopathogenic viruses mainly Baculoviruses family are excellent biological control agents and therefore a viable alternative for managing this pest. The aim of this study was to determine the biological activity of eight native nucleopolyhedrovirus (NPVs) against FAW larvae. Additionally, two of the most virulent isolates (SfCH32 and SfCH15) were characterized biologically by bioassays to estimate their median lethal dose (LD50) and median lethal time (LT50), morphologically by scanning electron microscopy (SEM) and transmission electron microscopy (TEM) and molecularly by restriction enzymes. Three (SfCH15, SfCH18 and SfCH32) of the eight tested native nucleopolyhedrovirus isolates caused mortalities ˃98% at 168-hr post-inoculation (hpi) with a dose of 9.2 × 104 OBs/larva. SfCH15 and SfCH32 isolates showed occlusion bodies (OBs) of irregular shape and size (1.02–2.24 μm). The SfCH15 and SfCH32 isolates showed similar median lethal dose (5.6 × 102–6.4 × 102 OBs/larva). The lowest median lethal time (114.5 hpi) was observed with the SfCH15 isolate at the highest concentration (2.5 × 106 OBs/larva). The DNA restriction profiles for SfCH15 and SfCH32 were different, with their genome size being ~128,000 bp and 132,000 bp, respectively. SfCH15 and SfCH32 isolates showed similar morphological characteristics and the highest virulence against fall armyworm. This study showed that native isolates were highly virulent against S. frugiperda larvae, being similar to other reported strains; however, field studies are required to confirm their insecticidal effect.  相似文献

We evaluated the consequences of parasitism by the solitary ichneumonid endoparasitoid Campoletis sonorensis(Cameron) towards the replication, genetic composition and virulence of a nucleopolyhedrovirus (Baculoviridae) originating from Spodoptera frugiperda(J. E. Smith) larvae. Parasitism by C. sonorensisand viral infection of third and fourth instar S. frugiperdalarvae resulted in reduced growth compared with nonparasitized control larvae. A positive correlation was observed between virus yield and larval instar at the moment of infection. When larvae were virus-inoculated in the fourth instar, parasitism resulted in a significant reduction in mean per capita virus yield compared to the virus yield from nonparasitized larvae. In an experiment involving 10 serial passages of virus in both parasitized and nonparasitized larvae, restriction endonuclease analysis of viral DNA amplified in nonparasitized larvae revealed the presence of the wild-type virus as well as three additional variants (A, B, and C) diagnosed by the presence of novel submolar PstI fragments of different sizes. In contrast, analysis of viral DNA from parasitized larvae showed the presence of the wild-type virus and two other variants (E and F), each characterized by a different submolar BglII fragment. Southern blot analysis indicated that the submolar fragments of variants E and F contained sequences originating from the viral genome. Bioassay of the different virus variants in S. frugiperdalarvae indicated that their virulence was equal or less than that of the wild-type virus. We conclude that parasitism can affect the quantity of virus produced in dually infected and parasitized larvae, but no adverse effects were detected in terms of the biological activity of the virus.  相似文献

We investigated the consequences of cannibalism for some correlatesof fitness in the fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae).The benefits gained by cannibals were ascertained by comparing survival,development rate, and pupal weight of larvae that had the opportunityto cannibalize with those that did not, at two levels of food availability.Larvae in the cannibalism treatments were provided with a conspecificone instar younger than themselves on five dates throughoutlarval development. Cannibalism was frequent; given the opportunityall larvae predated at least one younger conspecific. The frequencyof cannibalism was not affected by the sex of the cannibal orby the availability of alternative food. However, cannibalssuffered a significant reduction in survival at both high andlow food availability. Furthermore, cannibals had a lower pupal weightin the high food treatment and a reduced development rate inthe low food treatment. The only detectable fitness benefitassociated with cannibalism was a positive correlation betweenthe number of victims consumed and development rate under conditionsof low food availability. On balance, cannibalism appears tobe costly; alternative explanations for its occurrence in thisspecies are discussed.  相似文献

The effect of an optical brightener on the insecticidal activity of a Spodoptera littoralis (Boisduval) (Lepidoptera: Noctuidae) nucleopolyhedrovirus (SpliNPV) were examined in three instars of S. littoralis. LD50 values of the SpliNPV were reduced from 33 to <5, 73 to 5.7 and 342 321 to 288 412 occlusion bodies for second, third and fourth instars, respectively, by the addition of 1% Tinopal UNPA-GX. Relative potencies were >66, 12.8, and 1.2 for second-. third- and fourth-instar S. littoralis larvae, respectively. Relative mortality between the treatments with and without the brightener decreased from third- to fourth-stage larvae. In terms of speed of kill, the ST50 values of the baculovirus-infected larva were reduced from 210 to 159, 213 to 147, and 207 to 165 h for second-, third- and fourth-instar larvae, respectively, by the addition of the optical brightener at biologically equivalent doses.  相似文献

The incorporation of stilbene-derived optical brighteners into baculovirus formulations can substantially enhance virus infectivity and persistence of inoculum in the field. We evaluated the effect of the optical brightener Tinopal UNPA-GX (Sigma Chemical Co.) on the weight, development time, adult emergence and sex ratio of the principal pest of maize in the Americas, Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae). Tinopal UNPA-GX was incorporated into semi-synthetic diet at a final concentration of 0.1 or 0.05% (w/v) and larvae of S. frugiperda were individually reared on the Tinopal-treated or an untreated control diet. In both Tinopal treatments, the weights of larvae and pupae were significantly reduced by up to 36 and 13%, respectively. The duration of larval development was increased by up to 30% compared to the controls. Adult emergence was high in all treatments (≈90%) and was not affected by the presence of Tinopal in the diet. Significant differences in the proportion of adult females were observed in the 0.05% Tinopal treatment but not in the control or 0.1% Tinopal treatments. By affecting the integrity of the peritrophic membrane and the rate of sloughing of intestinal epithelial cells, natural pest populations exposed to optical brightener residues from bioinsecticide applications are likely to suffer a reduction in growth rate and other correlates of insect fitness.  相似文献

1. Antagonistic interactions in herbivorous insects are often density‐dependent, so rates are predicted to vary dynamically over time as density changes. Fatal intraspecific interactions, especially cannibalism, occur between equal‐aged larvae in young first‐ and second‐instar Spodoptera frugiperda (J.E. Smith). 2. A cannibalism experiment was conducted, starting with seven different densities of neonate S. frugiperda larvae, each replicated 50 times. Larvae were examined daily for the duration of the first and second instars (7 days). Seven‐day mortality was density‐dependent. 3. A stochastic mathematical model was developed in which per‐capita mortality from antagonistic interactions among equal‐aged larvae varies dynamically as density changes. A maximum likelihood method was developed to estimate the conditional per‐capita mortality rate from antagonistic interactions given an intraspecific encounter. An alternative model with mean‐mortality from antagonistic interactions that depends only on the initial larval density was also developed. 4. The models were fitted to the experimental data, and compared using log‐likelihood. The dynamic model fitted the cannibalism data significantly better than the time‐averaged mortality model for all starting densities for the experimental data, implying that density‐dependent mortality varied dynamically over time even within short 7‐day periods. 5. The conditional per‐capita mortality rate from antagonistic interactions was also density‐dependent, possibly because encounters became more aggregated at higher density, or because the probability that a larva died from the interaction was higher at higher density, or both.  相似文献

1. Experiments were carried out to investigate the incidence of cannibalism throughout the larval development of the noctuid moth Spodoptera frugiperda, and to examine the risk of infection from consuming conspecifics infected with a nuclear polyhedrosis virus (SfNPV). 2. Cannibalism was observed commonly even when food was not limiting, but occurred more frequently at low food quantities and/or high rearing densities. The sex of the larvae had no effect on the incidence of cannibalistic behaviour, however the probability of cannibalism occurring was affected by larval stage. The frequency of cannibalism was significantly higher among fifth- and sixth-instar larvae than among earlier instars, and larvae were more likely to consume younger conspecifics than larvae of the same stage. 3. Fifth-instar larvae offered fourth-instar victims fed equally on healthy larvae, virus-infected larvae (2 days post-infection), uninfected corpses, and virus-killed corpses (6 days post-infection). Horizontal transmission of SfNPV was only recorded in larvae offered virus-killed corpses, however, and total mortality in this treatment was only 32%. 4. In a similar experiment, fourth-instar larvae avoided cannibalising virus-killed corpses. Horizontal transmission of SfNPV was recorded in fourth-instar larvae that consumed 2-day post-infected larvae. The low incidence of cannibalism observed in fourth-instar larvae, however, suggests that this is unlikely to provide an important route for the transmission of SfNPV.  相似文献

Abstract Caterpillars of the noctuid moth, Spodoptera frugiperda (J. E. Smith), reared on artificial diets diluted with cellulose and water, increased fresh weight (fw) consumption 2.2–2.5-fold over those on the undiluted diet. At moderate levels of water- or cellulose-dilution, the increased consumption, combined with increased digestion and absorption of nutrients (ADNU), sufficiently compensated for the reduced nutrient intake to achieve pupal biomass equivalent to that of pupae from the undiluted diet. At higher levels of water- and cellulose-dilution, fw consumption and ADNU increased further but pupal dry weight declined on the water-diluted diets. At each level of dilution fw consumption and ADNU increased similarly on the water- and cellulose-diluted diets, but biomass gain was reduced on the water- compared with the cellulose-diluted diets. This was due in part to lowered food conversion efficiency on the water-diluted diets, possibly caused by increased costs of metabolizing the wetter diets. Our data support the hypothesis that consumption rates are regulated by a nutrient feedback mechanism. Our data also suggest that digestive enzyme activity is correlated with consumption. Furthermore, the cost of increased consumption rates on diets of reduced energetic value may constitute a substantially greater energy expenditure than previously believed. However, this cost was insufficient to reduce relative growth rates but was apparently manifested in lowered lipid accumulation.  相似文献

The behaviour of two abundant predators in Mesoamerican maize crops, Chrysoperla rufilabris larvae and Doru taeniatum adults, towards healthy and nucleopolyhedrovirus-infected Spodoptera frugiperda larvae was compared. C. rufilabris did not discriminate between healthy and virus-infected prey, although the mean search time was approximately two times longer towards virus-infected larvae. In contrast, D. taeniatum directed a greater proportion of their attacks towards virus-infected prey but there was no significant difference in the search time. Prey consumption time did not differ significantly for each type of prey by either predator, although prey consumption was much faster in D. taeniatum. Viable virus was detected in D. taeniatum faeces up to 3 d after feeding on infected S. frugiperda larvae, whereas virus was inactivated in the gut of C. rufilabris. Both predators were shown to have acidic guts. A field experiment demonstrated that D. taeniatum that had fed on infected prey could contaminate foliage resulting in the transmission of the disease at a low prevalence (4.7%) to S. frugiperda larvae in a field maize crop.  相似文献

The incorporation of certain stilbene optical brighteners into virus-based formulations has been demonstrated to increase viral pathogenicity (as indicated by reduced LD/LC50 values) but their effect on Bacillus thuringiensis activity has been scarcely investigated. We determined the effect of nine optical brighteners on the insecticidal activity of B. thuringiensis ser. kurstaki HD-1 strain (Bt HD-1) on Helicoverpa armigera and also compared the effect of two optical brighteners on the insecticidal activity of Bt HD-1 and occlusion bodies (OBs) of a Spanish isolate of H. armigera single nucleocapsid nucleopolyhedrovirus (HearNPV-SP1). Blankophor CLE, Blankophor DRS, Blankophor ER, and Leucophor SAC significantly increased the pathogenicity of Bt HD-1. In contrast, Tinopal UNPA-GX, Tinopal CBS, Blankophor BA, Leucophor AP, and Leucophor UO had an adverse or no effect on its insecticidal activity. Mixtures of HearNPV-SP1 OBs with Tinopal UNPA-GX or Leucophor UO resulted in 31.4- and 11.4-fold increases in pathogenicity, respectively, at 1%, and 11.4- and 6.3-fold increases in pathogenicity, respectively, at 0.1%, compared to the OBs alone. However, none of these brighteners increased Bt HD-1 activity. These results appear consistent with the hypothesis that the enhancement of HearNPV-SP1 pathogenicity and the null or antagonistic effects observed in Bt HD-1 against H. armigera were due to optical brightener-mediated degradation of the peritrophic membrane, but additional systematic studies involving a broad range of brighteners and electron microscope observations are required to confirm this premise.  相似文献

Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae) is an important parasitoid of lepidopteran pests. Its parasitism success is related to its foraging behavior, mediated by kairomones from its hosts. We aimed to evaluate the chemotactic responses and parasitism of T. pretiosum from Ephestia kuehniella (Zeller) (Lepidoptera: Pyralidae) eggs as well as the parasitoid maintained for three generations in Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae) eggs. Chemotactic responses of T. pretiosum from both hosts were evaluated in a dual choice olfactometer. Females were submitted to a choice between washed vs. unwashed eggs of E. kuehniella or S. frugiperda, as well as to unwashed eggs of both hosts. Egg extracts of both hosts were also contrasted with hexane and with each other. Parasitism of T. pretiosum exposed to eggs of the two species was also evaluated. Females from E. kuehniella were more attracted to odors from eggs of this host whenever it was present. Third generation parasitoids from S. frugiperda eggs were more attracted to eggs and egg extract of S. frugiperda than to those from E. kuehniella. The highest number of parasitized eggs and emerged parasitoids in S. frugiperda was observed in parasitoids coming from the third generation in this host. Therefore, imaginal conditioning could be an alternative to be considered for T. pretiosum learning before releasing, possibly optimizing its search and parasitism process in S. frugiperda eggs.  相似文献

Experiments were conducted to determine the influence of nicotine (at a range of concentrations) in the food of an herbivorous host on the development, size and survival of its parasitoid. Fall armyworms, Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae) were reared on diets of 0, 0.025, 0.050 and 0.075% nicotine and exposed to parasitism by Hyposoter annulipes (Hymenoptera: Ichneumonidae). As nicotine concentration increased parasitoid mortality and development time increased and adult weight decreased. Development time, pupal weight and survival were recorded for unparasitized armyworms. Unparasitized fall armyworms showed lengthened development and higher mortality but pupal weights were greatest at intermediate nicotine concentrations.
Résumé Des quantités croissantes de nicotine dans l'alimentation ont prolongé la durée du développement des chenilles saines, bien que l'effet ait chuté aux concentrations les plus élevées. Le sexe de la noctuelle n'a pas eu d'effet sur la durée du développement ou l'action de la nicotine. L'influence de la nicotine sur le poids des chrysalides est inhabituel, en ce sens que les chrysalides les plus lourdes ont été obtenues aux concentrations moyennes. Des hypothèses sont proposés sur l'origine de ce phénomène. La mortalité et la durée de développement de H. annulipes ont augmenté et le poids des adultes a diminué quand la concentration de l'aliment de l'hôte en nicotine s'est élevée. L'effet différente de la nicotine sur des générations successives a pu provenir de modifications de la toxicité de la nicotine en fonction de changements dans la qualité de l'aliment. Quoi qu'il en soit, l'augmentation de la concentration de la nicotine dans l'alimentation de l'hôte a eu un effet négatif cohérent sur la valeur adaptative de H. annulipes.

Understanding the behavior of pests targeted with Bacillus thuringiensis Berliner (Bt) crops is important to define resistance management strategies. Particularly the study of larval movement between plants is important to determine the feasibility of refuge configurations. Exposure to Bt maize, Zea mays L. (Poaceae), has been suggested to increase larval movement in lepidopteran species but few studies have examined the potential for resistance to interact with behavioral responses to Bt toxins. Choice and no‐choice experiments were conducted with Spodoptera frugiperda (JE Smith) (Lepidoptera: Noctuidae) and Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae) to determine whether Cry1F resistance influences neonate movement. Leaf discs of Cry1F maize and the corresponding isoline were used to characterize behavioral responses. In both experiments, the location (on or off of plant tissues) and mortality of susceptible and Cry1F resistant neonates was recorded for 5 days, but the analysis of larvae location was performed until 7 h. Our results indicated no strong difference between resistant and susceptible phenotypes in S. frugiperda and O. nubilalis, although a small percentage of susceptible neonates in both species abandoned maize tissue expressing Cry1F. However, significant behavioral differences were observed between species. Ostrinia nubilalis exhibited increased movement between leaf discs, whereas S. frugiperda selected plant tissue within the first 30 min and remained on the chosen plant regardless of the presence of Cry1F. Spodoptera frugiperda reduced larval movement may have implications to refuge configuration. This study represents the first step toward understanding the effects of Cry1F resistance on Lepidoptera larval behavior. Information regarding behavioral differences between species could aid in developing better and more flexible resistance management strategies.  相似文献

Efficient production is essential for developing baculoviruses into marketable bioinsecticides. In this study, we evaluated how juvenile hormone (JH) III and the JH analog pyriproxyfen affected the production efficiency of Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) Nucleopolyhedrovirus (SpltNPV) in SpltNPV‐inoculated early sixth instars. Both the JH III and pyriproxyfen treatments significantly enhanced the body size and weight of S. litura, which resulted in a 1.5‐ to 2‐fold increase in the production of occlusion bodies (OBs). The OBs obtained from JH‐ and pyriproxyfen‐treated larvae were as pathogenic as those produced in untreated larvae. More importantly, JH‐ or pyriproxy‐fen‐treated larvae consumed less than twice the amount of food consumed by two untreated larvae. These results indicate that JH and pyriproxyfen treatments improve the production efficiency of SpltNPV in S. litura. SpltNPV infection affected the development of S. litura not only as larvae, but also during subsequent growth stages and generations.  相似文献

The effect of oscillating dissolved oxygen (DO) concentration on the metabolism of a clonal isolate of the Spodoptera frugiperda IPLB-Sf21-AE insect cell line was investigated. Specifically, the effect on cell growth, re- combinant protein synthesis, glucose and glutamine consumption, and lactate accumulation was determined. Prior to conducting the oscillating DO experiments, it was found that the DO concentration could be reduced to 15% air saturation without adversely affecting the growth rate. Under these conditions, glucose and glutamine became depleted as the maximum cell density was reached. The introduction of DO oscillations, that is, cycles consisting of 30 min at 15% DO followed by 30 min of anoxia, significantly altered cell metabolism, including inhibition of cell growth and recombinant protein synthesis. The effect of DO oscillations on glucose consumption was dependent on the experimental conditions. Glucose exhaustion occurred when the DO oscillations contained either an "apparent" anoxia period (nitrogen sparging discontinued upon reaching 0% DO) without pH control or a "true" anoxia period (nitrogen sparging continued throughout anoxia period) with pH control. Glucose consumption was significantly decreased, however, when the cells were exposed to a "true" anoxia period without pH control, that is, low pH inhibited glucose utilization. Glutamine uptake was not significantly affected by DO oscillations. Lactate only accumulated in the oscillating DO runs, a finding consistent with previous results demonstrating that significant lactate accumulation only occurs under DO-limited conditions.
