In seedlings of Vigna radiata (L.) R. Wilczek cultivars ADT-1 and CO-5 exposed to acidic showers (H2SO4 : HNO3 : HCl, 4 : 2 : 1, v/v) of different pH (7.0, 5.5, 4.0, and 2.5) for 10 d, net CO2 uptake and stomatal conductance were reduced. The chlorophyll (Chl) a and b contents were reduced but the carotenoid (Car) content increased. In vivo Chl a fluorescence patterns of both the cultivars were altered. No significant change in photosystem (PS) 1 activity was observed except at pH 2.5 where an inhibition was evident. By contrast, PS2 activities declined rapidly with increasing acidity. The room temperature absorption spectra of isolated chloroplasts showed very little changes. SDS-PAGE analysis revealed depletion of 23, 33, and 55 kDa polypeptides. Cultivar CO-5 was more sensitive to acidic rain than cv. ADT-1.  相似文献

Seedlings of green gram (Vigna radiata cv. ADT-1 and CO-5) were exposed to daily showers of simulated acidic rain (H2SO4 : HNO3 : HCl, 4 : 2 : 1, v/v) for 10 d. The effects were analysed after 5 and 10 showers, respectively. Rain of pH 2.5 inhibited seedling growth and biomass accumulation, though in other acidic levels the effects were mostly inconsistent. Both cultivars had high degree of surface wettability indicated by high leaf surface contact angles and water-holding capacity. Treated leaves were thinner with smaller mesophyll cells. Stomatal index and trichome density were lower in contrast to epidermal cell density and stomatal frequency which increased with increasing acidity. Decreases in chlorophyll (Chl), carotenoid (Car), and starch contents in cv. ADT-1 at pH 2.5 were observed after 5 showers, while in cv. CO-5 decreases were noted only after 10 showers. In contrast to total sugar levels, the protein content of cv. CO-5 was augmented significantly after simulated acidic rain (SAR) treatment.  相似文献

以朱砂叶螨的寄主植物三月早茄为对象,研究了不同pH值和不同时间模拟酸雨胁迫下,茄子对酸雨的抵抗、适应能力和营养物质含量变化.结果表明,随酸雨酸度增强,茄叶的过氧化氢酶(CAT)活性、磷和可溶性蛋白含量呈单峰变化趋势;而过氧化物酶(POD)活性、可溶性糖含量呈单谷变化趋势;还原糖含量和超氧化物歧化酶(SOD)活性随酸度增加呈上升趋势;茄叶pH值呈下降趋势,含水量不受酸雨影响.POD对酸雨作用反应迟缓,而SOD、CAT较敏感,其他几项生理指标则对酸雨作用时间反应灵敏.高pH(4.0~5.6)的酸雨刺激茄叶的保护能力增强,促进茄叶的生长发育.由于茄叶内糖含量、磷及可溶性蛋白含量的改变,植物的适口性有所变化,可促进叶螨生长,而在强酸性酸雨(pH<3.0)胁迫下,植物的生长受到抑制,营养物质含量降低,抑制朱砂叶螨的生长.  相似文献

为了探讨酸雨胁迫与毛竹(Phyllostachys pubescens)绿叶挥发物(green leaf volatiles, GLVs)释放规律以及抗氧化酶活性的关系, 通过盆栽试验, 采用不同pH值(5.6、4.0、2.5)的模拟酸雨对毛竹三年生实生苗进行处理, 研究酸雨对毛竹叶片可溶性蛋白质含量、丙二醛(MDA)含量和抗氧化酶活性的影响, 并采用热脱附/气相色谱/质谱联用技术对毛竹释放的GLVs成分和含量进行分析。结果表明: 酸雨胁迫下毛竹叶片MDA含量明显增加, pH 2.5模拟酸雨胁迫处理45天毛竹叶片MDA含量与对照相比增加了43.0% (p < 0.01); pH 4.0处理MDA含量增加缓慢, 处理75天时MDA含量比对照增加了0.36倍(p < 0.01)。pH 4.0和pH 2.5模拟酸雨胁迫处理45天时, 毛竹叶片可溶性蛋白质含量极显著增加, 与对照相比分别增加了32.0%和65.0% (p < 0.01)。在酸雨胁迫下, 毛竹叶片超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和过氧化物酶(POD)的响应时间存在一定差异, 表现为互相协调, pH 2.5模拟酸雨胁迫处理SOD活性和POD活性分别在45天和60天时达到最大值, 分别为对照的1.67倍和1.31倍(p < 0.01), 随后降低。pH 4.0和pH 2.5模拟酸雨胁迫处理, 毛竹叶片GLVs含量比对照分别增加26.4%和132.9% (p < 0.01), 新增GLVs为 (E)-2-辛烯醛、2-乙基己醛、(E)-2-己烯醛和(E)-2-壬烯醛。研究表明: 酸雨胁迫条件下, 毛竹可以通过调节保护酶活性、可溶性蛋白质含量和释放GLVs来提高适应环境的能力。  相似文献

A protease activity was detected in rice (Oryza sativa L. cv. Ratna) leaves that hydrolysed hemoglobin more efficiently than bovine serum albumin. The activity was high when the enzyme was extracted and assayed with tris-maleate buffer [tris (hydroxymethyl) methyl amino-maleate] pH 7.0 rather than with water or with citrate-phosphate buffer pH 7.0. The enzyme had a strong dependence on sulfhydryl groups for its activity without which it was inaotive. The pH optimum was 7.0 and the temperature optimum was 40 °C. Protease activity expressed per unit leaf fresh weight (absolute activity) increased only little during senescence of detached rice leaves while the same activity expressed per unit soluble protein content (specific activity) increased by a greater factor (about 5 times) than absolute activity. Total and soluble protein content decreased during the senescence of detached leaves. Benzimidazole (10-3M) and kinetin (0.5xl0-5M) treatment arrested the increase of the protease activity and the deorease in the protein content during detached leaf senescence. It was indicative that protease protein was more stable than the bulk of other proteins in senescing leaves.  相似文献

Changes in endogenous cytokinin (CK) content and cytokinin oxidase/dehydrogenase activity (CKX) in response to gibberellic acid (GA3) in two pea cultivars with different life span were assessed. The control leaves of cv. Scinado, which developed faster, had higher initial cytokinin content and lower CKX activity, while opposite trend was observed in cv. Manuela with longer life span. Increased CKX and decreased CK content were detected in leaves of cv. Scinado after treatments with 0.5, 1 and 5 μM GA3. Changes in CK content and CKX activity in GA3-treated cv. Manuela leaves were reciprocal to those in cv. Scinado. CK content and CKX activity in roots were not significantly influenced by the application of GA3. The slight repression of CKX activity in some of the root samples was accompanied by increased isopentenyladenine and isopentenyladenine riboside content. Obtained results suggest that CKX was responsible for the changes in endogenous cytokinin pool in GA3-treated plants and most probably this enzyme represents an important link in GA/cytokinin cross talk.  相似文献

The effects of short-term exposure to chilling temperature (10 °C) on sucrose synthesis in leaves of the cold-tolerant sugarcane cultivars Saccharum sinense R. cv. Yomitanzan and Saccharum sp. cv. NiF4, and the cold-sensitive cultivar S. officinarum L. cv. Badila were studied. Plants were grown at day/night temperatures of 30/25 °C, and then shifted to a constant day/night temperature of 10 °C. After 52-h exposure to the chilling temperature, sucrose content in the leaves of NiF4 and Yomitanzan showed a 2.5- to 3.5-fold increase relative to that of the control plants that had been left on day/night temperatures of 30/25 °C. No such increase was observed in Badila leaves. Similarly, starch content in the leaves of NiF4 and Yomitanzan was maintained high, but starch was depleted in Badila leaves after the 52-h exposure. During the chilling temperature, sucrose phosphate synthase (SPS; E.C.2.4.1.14) activity was relatively stable in the leaves of NiF4 and Yomitanzan, whereas in Badila leaves SPS activity significantly decreased. There was no significant change in cytosolic fructose-1,6-bisphosphatase activity for the three cultivars at the chilling temperature. This supports the hypothesis that: (1) on exposure to chilling temperature, sucrose content in sugarcane leaves is determined by the photosynthetic rate in the leaves, and is not related to SPS activity; (2) SPS activity in sugarcane leaves at chilling temperature is to be determined by sugar concentration in the leaves.  相似文献

选择亚热带常绿阔叶林优势树种青冈幼苗为研究对象,设置3个酸雨处理:pH 2.5、pH 4.0、pH 5.6(CK),研究不同强度的模拟酸雨对青冈幼苗光合特性、叶绿素荧光参数和叶绿素含量的影响.结果表明:经过2年酸雨处理,青冈的净光合速率随着酸雨强度的增加而显著上升.pH 2.5、pH 4.0处理的酸雨增大了青冈的气孔导度和蒸腾速率,且对pH 2.5处理的影响更为显著.胞间CO2浓度的大小顺序是pH 2.5>pH 5.6>pH 4.0.pH 2.5、pH 4.0处理青冈的最大净光合速率、光补偿点、光饱和点、暗呼吸速率显著高于对照,表观量子效率对酸雨胁迫不敏感.pH 2.5、pH 4.0处理青冈的PSⅡ原初光能转化效率和PSⅡ的潜在活性显著高于对照.青冈的叶绿素相对含量大小顺序是pH 2.5>pH 5.6>pH 4.0,且pH 2.5与pH 4.0之间有显著差异.说明青冈幼苗的光合参数、叶绿素荧光参数指标在pH 2.5和pH 4.0的酸雨处理下有所增加,且在pH 2.5强度下增加更为明显.  相似文献

研究模拟酸雨对3种农作物种子萌发年和幼苗生长的影响。结果表明:不同pH值(2.5,4.5,5.6)的模拟酸雨对水稻和小麦的种子萌发没有影响,但明显抑制了玉米种子萌发。模拟酸雨条件下,3种农作物幼苗的生长受到抑制,生物量减少,叶绿素和类胡萝卜素含量下降,而叶绿素a/b的变化却不明显。pH4.5和5.6的模拟酸雨对玉米Fv/Fm、光化学猝灭(qP)的影响较小,非光化学猝灭(NPQ)却明显下降,表明酸雨伤害了植物PSⅡ天线对激发能的非辐射耗散能力。  相似文献

The magnitude of the effect of salt stress on proline content, pyrroline-5-carboxylate (P5C) reductase activity and water relations was found to be leaf position dependent in an advance generation (R4) of twoBrassica juncea L. somaclones (SR-2 and SR-3) selected in vitro for NaCl-tolerance and the parent cv. Prakash. Free proline content and P5C reductase activity increased with increase in salt stress in all the lines but at different rates; the maximum increase being in the SR-3 derived somaclonal line. At 100 mM NaCl, SR-3 showed a nearly 19 fold increase in proline content compared to a 4–5 fold increase in the other two genotypes. The proline level and P5C reductase activity of the first (youngest) leaf was higher than in the other leaves and decreased linearly with increase in age of the leaf in all the lines. The relationship between relative water content and osmotic potential of the leaves at different positions also varied. The results indicate that a significant effect of salt may appear non-significant if the position of the leaves is not taken into account while sampling.  相似文献

The effects of Al, Cd and pH on growth, photosynthesis, malondialdehyde (MDA) content, and some antioxidant enzyme activities of the two soybean cultivars with different Al tolerance were determined using a hydroponic culture. There were six treatments as follows: pH 6.5; pH 4.0; pH 6.5 + 1.0 μM Cd; pH 4.0 + 1.0 μM Cd; pH 4.0 + 150 μM Al; pH 4.0 + 1.0 μM Cd + 150 μM Al. The results showed that the low pH (4.0) and Al treatments caused marked reduction in the growth (root and shoot length and dry mass), chlorophyll content (SPAD value) and net photosynthetic rate. Higher malondialdehyde content, superoxide dismutase (SOD) and peroxidase (POD) activities were detected in the plants exposed to both Al and Cd than in those exposed to Al treatment alone. An expressive enhancement of SOD and POD was observed in the plants exposed to 150 μM Al in the comparison with the control plants, especially in Al-sensitive cv. Zhechun 2 which had also significantly higher Al and Cd content than Al tolerant cv. Liao-1. Cd addition increased Al content in the plants exposed to Al + Cd stress, and cv. Zhechun 2 had relatively lower Al content. The present research indicated that Al and Cd are synergistic in their effects on plant growth and some physiological traits.  相似文献

通过比较浙江大盘山不同海拔(A1:550~660 m,A2:700~800 m,A3:900~990 m,A4:1 000~1 015 m,A5:1 020~1 100 m)七子花(Heptacodium miconioides)叶片色素含量、含水量及比叶面积变化,探讨不同海拔对七子花生长的影响。结果表明,随着海拔的升高,叶绿素含量(包括叶绿素a、叶绿素b、叶绿素a+b)先增加后减少,在A3达最高值;叶绿素a/b比值、类胡萝卜素含量变化呈先低后高,在A3降至最低;比叶面积及叶片含水量,在较高海拔A4、A5都明显下降。七子花通过叶片色素、比叶面积、干重和含水量的变化以适应高海拔环境。  相似文献

Excised rice (Oryza sativa L. cv. Ratna) leaves were used to compare the changes in the levels of various biochemical intermediates and enzyme activities during senescence in turgid and water-stressed conditions. Chlorophyll, total protein and soluble protein content decreased but α-amino nitrogen content increased during the senescence of turgid leaves. In the leaves subjected to water stress, these changes were accelerated, the acceleration being greater with higher degree of water stress. Starch, soluble sugars, total carbohydrates and non-reducing sugar content decreased during senescence of turgid leaves. Water stress accelerated the changes in the levels of starch and non-reducing sugar, but the changes in the levels of soluble sugars and total carbohydrates were retarded. Reducing sugar content increased at first and then decreased in the turgid leaves, and water stress accelerated the change. The decline in the catalase activity and the increase in the peroxidase activity with time was faster in the water-stressed leaves than in the turgid leaves. Acid inorganic pyrophosphatase activity increased, but alkaline inorganic pyrophosphatase activity decreased during the senescence of turgid leaves, and such changes were accelerated by water stress. The results suggest that water stress does not accelerate all the processes connected with leaf senescence.  相似文献

The two cultivars of Digitaria eriantha: cv. Sudafricana (a cold-sensitive cultivar) and cv. Mejorada INTA (a cold-resistant cultivar) were exposed to low temperature and compared in terms of the involvement of abscisic acid (ABA) and catabolites, jasmonates, and antioxidant defense in cold tolerance. Cold stress caused a greater ABA increase in cv. Mejorada INTA than in cv. Sudafricana. In both cultivars abscisic acid glucose ester and dihydrophaseic acid were the most abundant catabolites. Cold treatment decreased JA in leaves of both cultivars. In cv. Sudafricana, 12-hydroxyjasmonate (12-OH-JA) decreased and 12-oxo-phytodienoic acid increased. In regard to antioxidant defense, both cultivars increased the non-protein thiols in response to cold stress. However, reduced glutathione (GSH) level was higher in leaves of cv. Mejorada INTA than cv. Sudafricana. Cold-treated leaves of cv. Sudafricana increased thiobarbituric acid-reactive substances (TBARS), but cv. Mejorada INTA leaves showed no such increase. Superoxide dismutase activity decreased and ascorbate peroxidase activity increased in cold-treated leaves of cv. Sudafricana. No significant change of these enzymes was observed for cv. Mejorada INTA. The cold tolerance of cv. Mejorada INTA could be related to JA, 12-OH-JA and GSH high basal contents, ABA increase, and TBARS stability after cold treatment.  相似文献

Alterations in tetraspanin CO-029 expression are associated with the progression and metastasis of cancers in the digestive system. However, how CO-029 promotes cancer metastasis is still poorly understood. To determine the mechanism, we silenced CO-029 expression in HT29 colon cancer cells and found that the CO-029 knockdown significantly reduced cell migratory ability. The diminished cell migration was accompanied by the upregulation of both integrin-dependent cell-matrix adhesion on laminin and calcium-dependent cell-cell adhesion. The cell surface levels of laminin-binding integrin α3β1 and fibronectin-integrin α5β1 were increased while the level of CD44 was decreased upon CO-029 silencing. These changes contribute to the altered cell-matrix adhesion. The deregulated cell-cell adhesion results, at least partially, from increased activity of cadherins and reduced level of MelCAM. In conclusion, CO-029 functions as a regulator of both cell-matrix and cell-cell adhesion. During colon cancer progression, CO-029 promotes cancer cell movement by deregulating cell adhesions.  相似文献

A series of experiments were conducted to characterize the water stress-induced changes in the activities of RuBP carboxylase (RuBPCO) and sucrose phosphate synthase (SPS), photosystem 2 activity, and contents of chlorophylls, carotenoids, starch, sucrose, amino acids, free proline, proteins and nucleic acids in mulberry (Morus alba L. cv. K-2) leaves. Water stress progressively reduced the activities of RuBPCO and SPS in the leaf extracts, the chlorophyll content, and PS2 activity in isolated chloroplasts. Plants exposed to drought showed lower content of starch and sucrose but higher total sugar content than control plants. While the soluble protein content decreased under water stress, the amino acid content increased. Proline accumulation (2.5-fold) was noticed in stressed leaves. A reduction in the contents of DNA and RNA was observed. Reduced nitrogen content was associated with the reduction in nitrate reductase activity. SDS-PAGE protein profile showed few additional proteins (78 and 92 kDa) in the water stressed plants compared to control plants.  相似文献

以‘乌紫’和‘荸荠’两个杨梅品种为试材,测定了干鲜重、糖含量、可滴定酸含量、蔗糖和己糖代谢相关酶活性的动态变化。结果表明,杨梅果实的干鲜重、含糖量的快速增长和可滴定酸含量的快速下降均发生在果实发育后期。成熟‘乌紫’杨梅果实的蔗糖含量约占总糖的2/3以上,而‘荸荠’杨梅仅为总糖的49%。‘荸荠’杨梅的转化酶和蔗糖合酶分解活性随着果实发育呈上升趋势,‘乌紫’杨梅的则变化不大。两个品种的蔗糖磷酸合酶活性随着果实发育呈上升趋势,但蔗糖合酶合成活性到果实发育中期后下降。两个品种的己糖激酶活性变化相似,但果糖激酶活性的变化趋势不同。  相似文献

Salt stress-induced changes in antioxidant enzymes, lipid peroxidation, proline and glycine betaine contents, and proline-metabolizing enzymes were examined in the leaves of two mulberry cultivars (Local and Sujanpuri). With increasing salinity up to 150 mM NaCl, superoxide dismutase, catalase, ascor-bate peroxidase, guaiacol peroxidase, glutathione reductase, and monodehydroascorbate reductase activities were increased in both cultivars as compared to control, but more pronounced increase was observed in cv. Local. Salt stress enhanced the rate of lipid peroxidation (as indicated by increasing MDA content) in both cultivars. Under NaCl stress, cv. Local showed less change in the MDA content than cv. Sujanpuri. Salt stress resulted in a significant accumulation of free proline in mulberry leaves, and more accumulation was detected in cv. Local than cv. Sujanpuri. The leaves of cv. Local showed 9-fold accumulation of glycine betaine in comparision with cv. Sujanpuri after 20 days at 150 mM NaCl. A decrease in proline oxidase activity and an increase in γ-glutamyl kinase activity were observed with increasing NaClconcentration. The relative water content and electrolyte leakage also decreased after increasing the NaCl concentration, but a decrease was more pronounced in cv. Sujanpuri than in cv. Local. The results indicate that oxidative stress may play an important role in salt-stressed mulberry plants and cv. Local have more efficient antioxidant characteristics, which could provide for a better protection against oxidative stress.  相似文献
