Cytological and histochemical methods were used in order to establish that the intercalated cells of the ciliated epithelium of the rabbit's maxillary sinus mucosa performed gland function giving secretion formed in the shape of granules and represented by neutral mucopolysaccharides. Analysis of the morphology of intercalated cells, the amount of secretion in them, the shape and disposition of the Golgi apparatus and the nucleus allowed establishing 4 successive phases in their secretory cycle: the phase of synthesis, accumulation, aquosity and sectering. Under normal conditions of life the secretory activity of intercalated cells was asynchronous. In experiments with synchronization of the secretory activity by administration of pilocarpine it was established that the duration of the intercalated cells secretory cycle was 16-18 hours. Active participation of alkali phosphatase in the synthesis of secretion in the ciliated epithelium was noted.  相似文献

The authors studied morphological and histochemically the mucopolysaccharides and proteins in the gallbladder tubular glands and epithelial cells of the capivara Hydrochoerus hydrochoeris. Based on the results the authors concluded: 1. the gallbladder single columnar epithelium consists of secretory, migrating, and goblet cells; 2. in the lamina propria are single coiled tubular glands; 3. goblet and tubular gland cells show neutral and sulphated mucopolysaccharides and sialic acid; 4. columnar cells show neutral mucopolysaccharides and protein radicals; 5. migrating cells show only protein radicals.  相似文献

Sulfated acid mucopolysaccharides have been shown to be constituents of cortical granules in sea urchin and vertebrate eggs. These observations were made possible by retaining soluble acid mucopolysaccharides in situ within the eggs by precipitation during fixation with cetyltrimethyl-ammonium bromide, a quaternary ammonium salt. The sulfated mucopolysaccharides were then identified by staining with Astrablau at pH 0.2 and also by reaction with sodium rhodizonate. Staining reaction with Alcian blue at pH 2.5 showed that carboxylated mucopolysaccharides may also be present in cortical granules. The natural ionic environment of these eggs would favor the formation of very stable complexes between sulfated mucopolysaccharides and quaternary ammonium salts. Brief exposure of unfertilized sea urchin eggs to several quaternary ammonium compounds produced a residual adverse effect on subsequent fertilization in terms of increased vulnerability to polyspermy and reduced fertilizability. These results suggest that sulfated acid mucopolysaccharides participate in the function of the cortical granules and the establishment of the block to polyspermy at fertilization, and possibly in other cellular secretory processes.  相似文献

The numbers of mucous cells in the epidermis of the head and body and in the surface of the gill arch of the amphibious blenny, Blennius pholis L., were estimated on fish immersed in sea water and after 4 h aerial exposure. During emersion there appeared to be a considerable reduction in the frequency of epithelial mucous cells in the areas studied, although counts for the head epidermis were somewhat variable. A concomitant decline in the number of cells thought to be actively secreting was also recorded in tissue samples from both the head and gills, while in the body epidermis the potential for mucus-secretion was maintained close to the levels observed in immersed fish.
Histochemical studies revealed epidermal mucous cells containing either sialylated acid mucopolysaccharides or neutral mucins, or a mixture of these, in both the head and the body, whereas in the gill arch epithelia there were, in addition, cells containing sulphated acid mucopolysaccharides. After emersion, a disproportionate loss of cells containing neutral and sialylated mucus from the gill epithelia resulted in an increase in the proportion of secreting cells staining positively for sulphated acid mucopolysaccharides.
The results of this study are discussed in relation to nitrogenous excretion during aerial exposure.  相似文献

The end organ of Proteocephalus ambloplitis pleroceroids was studied by light microscopy and histochemistry. The end organ consists of a spherical sac filled with an amorphous, granular secretory product. The lining of the end organ is devoid of an epithelial cell layer. Protease was detected within the end organ by a gelatin-silver film technique. No aminopeptidase activity was detected. The end organ was PAS positive and diastase-fast. A positive test was obtained for neutral mucopolysaccharides and protein. The end organ is thought to assist migration of larvae through host tissue.  相似文献

The ultrastructure and histochemical features of the two types of secretory cells in leech skin are described. Pear-shaped cells secrete mucus containing carboxylated mucosubstances, while tubular cells produce a mucus containing a mixture of neutral, carboxylated, and sulfated mucosubstances. Pear-shaped secretory cells have two types of neuroglandular junctions, one containing dense-core serotonergic vesicles and the other small clear vesicles. Introduction

Shear wave elastography (SWE) has not been studied for diagnosing appendicitis. We postulated that an inflamed appendix would become stiffer than a normal appendix. We evaluated the elastic modulus values (EMV) by SWE in healthy volunteers, patients without appendicitis, and patients with appendicitis. We also evaluated diagnostic ability of SWE for differentiating an inflamed from a normal appendix in patients with suspected appendicitis.

Materials and Methods

Forty-one patients with clinically suspected acute appendicitis and 11 healthy volunteers were prospectively enrolled. Gray-scale ultrasonography (US), SWE and multi-slice computed tomography (CT) were performed. The EMV was measured in the anterior, medial, and posterior appendiceal wall using SWE, and the highest value (kPa) was recorded.

Results

Patients were classified into appendicitis (n = 30) and no appendicitis groups (n = 11). One case of a negative appendectomy was detected. The median EMV was significantly higher in the appendicitis group (25.0 kPa) compared to that in the no appendicitis group (10.4 kPa) or in the healthy controls (8.3 kPa) (p<0.001). Among SWE and other US and CT features, CT was superior to any conventional gray-scale US feature or SWE. Either the CT diameter criterion or combined three CT features predicted true positive in 30 and true negative in 11 cases and yielded 100% sensitivity and 100% specificity. An EMV of 12.5 kPa for the stiffest region of the appendix predicted true positive in 28, true negative in 11, and false negative in two cases. The EMV (≥12.5 kPa) yielded 93% sensitivity and 100% specificity.

Conclusion

Our results suggest that EMV by SWE helps distinguish an inflamed from a normal appendix. Given that SWE has high specificity, quantitative measurement of the elasticity of the appendix may provide complementary information, in addition to morphologic features on gray-scale US, in the diagnosis of appendicitis.  相似文献

The mucus-producing glands of the tropical slug Veronicella floridana Leidy were examined histochemically. The bulk of the dorsal mucus is a carboxylated mucopolysaccharide produced by deeply embedded gland cells which contribute to inflated common ducts. Little mucus is apparent on the dorsal surface of an unirritated slug, giving the dorsal aspect of the animal a dry appearance. Measurements of the rate of water loss of whole animals indicate that water is lost significantly more slowly than from the equivalent area of water.
The foot possesses regular transverse ridges and the pedal mucus is produced largely by a set of gland cells associated with each ridge. This mucus is a mixture of mucopolysaccharides and protein. The suprapedal gland which also contributes to the pedal mucus is extremely small and produces a weakly acidic and neutral mucopolysaccharide and a protein. There is no specialization in the leading edge of the foot and no Semper's organ. The cleft between the foot and the body is unciliated and bears few specialized gland cells.
Compared with the mucus produced by Limax pseudoflavus , the pedal mucus of Veronicella is of comparable function and composition though produced by glands of different derivations. The dorsal mucus is histochemically different from that of Limax though similar in physical appearance. Whilst the dorsal mucus of Limax is freely distributed over exposed surfaces, that of Veronicella is held in reserve in inflated ducts resulting in a dry leathery appearance and a depressed rate of water loss.  相似文献

The wall of the spermathecal ampulla in Tubifex tubifex consists of epithelial, muscular and peritoneal layers. The epithelial surface contains closely microvilli while lateral and basal plasma membranes are extensively convoluted. Epithelial cytoplasm exhibits a vertical zonation of subcellular components. The distal zone contains filiform secretory particles which are orientated perpendicular to the apical surface; extrusion occurs by their fusion with the plasma membrane between the bases of neighbouring microvilli. Mitochondiral and Golgi zones, the latter containing the nucleus, subtend the distal zone. The basal zone, composed of vertical compartments formed by the folded plasma membrane, is rich in α-glycogen rosettes. The distal epithelium and lumen material contain neutral mucopolysaccharides and carboxylated acid mucopolysaccharides in conjunction with neutral protein. The ultrastructure of the spermathecal duct wall is comparable with that of the ampulla but is characterized by extremely long microvilli and a prominent musculature.  相似文献

Using the frog palate as a representative model of human mucociliary epithelium, we analyzed, after quick freezing fixation, the three-dimensional (3-D) respiratory mucus secretory release with high voltage (200-300 kV) transmission electron microscopy (TEM). The 3-D vision of the mucus release from the secretory cells was obtained as stereo-pairs and "bas-relief" images after analysis of stereo-pairs using an image analyzer. After standard glutaraldehyde fixation, the secretory cells showed a typical goblet shape with secretory granules heterogeneous in size and electron-density which often fuse together. On the other hand, quick-frozen secretory cells exhibited a columnar shape and their membrane-bound secretory granules contained a homogeneously dark matrix. The expanded gel mucus layer was preserved and its depth never exceeded 2 microns. When the epithelium was immersed in culture medium in presence of cholinergic agonist, a marked discharge of mucus was observed and the granules swelled at the apex of the secretory cell before being discharged in the lumen. In native cryofixed epithelium, the secretory granules exhibited a marked deformability during the process of their extrusion from the secretory cell. Clusters of secretory granules surrounded by cytoplasmic material were observed in the extracellular lumen, suggesting an apocrine-type secretion. These observations indicate that rapid cryofixation and 3-D stereoscopic imaging enable a unique opportunity to analyze, without artifact, the mucous secretory process. We speculate that, apart from the classical merocrine-type secretion mechanism, the respiratory mucus may be released, at least partly by an apocrine-type secretion.  相似文献

Summary The epidermis of the posterior tentacles of the terrestrial snail Achatina fulica was examined by histological and histochemial methods. There are seven types of unicellular glands in the tentacle skin: three mucocytes containing either acid mucopolysaccharides or neutral mucopolysaccharides, or both; two mucocytes containing glycoproteins; a lipid gland; and a protein gland. The mucocytes are considerably more abundant along the shaft of the tentacle than at the tip, where the olfactory organ is situated. Conversely, the lipid glands and the protein glands are found almost exclusively in the olfactory organ. With minor exceptions, none of the foregoing cell types is present in the skin of the head or the foot. These observations indicate a high degree of local specificity in secretory products, consistent with a ubiquitous and generous endowment of glands in the molluscan skin. Collar cells, described by previous authors in closely related species, were not observed.  相似文献

OBJECTIVE--To clarify poorly understood epidemiological features of appendicitis. DESIGN--Retrospective study of consecutive cases from a defined population and analysis of data from published studies. SETTING--County of Jönköping, Sweden. 3029 patients who underwent operation in 1984-9 and 4717 patients from the county town who underwent operation in 1970-89, all for suspected appendicitis, plus 48,426 cases from six reported studies. MAIN OUTCOME MEASURES--Incidences specific for age and sex and temporal trends of perforating and non-perforating appendicitis and removal of a normal appendix. Associations between diagnostic accuracy, rate of perforation, and incidences of removal of a normal appendix and of perforating and non-perforating appendicitis. RESULTS--The incidence of appendicitis was 116/100,000 inhabitants. Appendicitis was more common in male patients. The incidence of perforating appendicitis was independent of age, stable over time, and uninfluenced by the rate of laparotomy, whereas the incidence of non-perforating appendicitis was age dependent, decreasing over time, and related to the diagnostic accuracy and rate of removal of a normal appendix. CONCLUSIONS--Perforating and non-perforating appendicitis seem to be separate entities, and appendicitis that resolves spontaneously is common. This may have important implications for managing suspected appendicitis.  相似文献

The "utero-vaginal junction" that connects the uterus and vagina of duck oviduct differs histologically and histochemically from the adjacent zones. The region is characterised by low, somewhat longitudinally arranged mucosal folds, lined by tall columner cilliated cells with apical nuclei alternating with mucous secreting goblet cells containing basal nuclei. Within the propria of the mucosa there are numerous tubular glands--the sperm host glands--which are responsible for sperm storage after copulation. The goblet cells of the utero-vaginal junction contain an admixture of neutral, sulfated and nonsulfated acid mucopolysaccharides, while the sperm host glands are devoid of any mucopolysaccharide and secrete complex lipoidal materials and exhibit intense acid phosphatase activity. The functional significance of the secretory materials of the sperm host glands have been discussed in the light of sperm release mechanism.  相似文献

In order to develop a method to quantify the mucus secretion, we observed the mucus epithelium of the rabbit colon under a video-enhanced differential interference contrast microscope. Upon stimulation with muscarinic agonists, secretory granules in individual goblet cells were found to undergo a rapid light intensity change. Simultaneously, the lumen was widened and filled with a cloudy material. In each cell, many of such responses were followed by formation of a large cavity which could be recovered after removal of the stimulant. We infer that the light intensity change of a granule arises from exocytosis. Direct counting of the frequency of these quantal responses would be very useful to monitor the secretory activity of single cell in real time at a high sensitivity.  相似文献

Within 1 1/2 h after settling, an electron-dense proteinaceous cyst layer (the outer layer) is formed from secretions originating at the base of the kineties and from the thick pellicular layer between the kineties. The inner cyst layer, composed primarily of chitin (acidic and neutral polysaccharides are also present), is secreted across the entire cell surface. Cyst wall formation is completed within 6 h. The fine structure of endocyst secretion resembles stages in the secretion of chitin by fungi, yeasts, and arthropods. A proteinaceous attachment peduncle is secreted to anchor the cell to a shrimp host and is formed by the release of electrondense dense secretory bodies from the cell's ventral surface.  相似文献

Because the precise immunopathological events occurring in appendicitis are not completely understood, possible local production of endothelin-1 (ET-1) in human appendix was investigated. We used immunohistochemistry and in situ hybridization to detect the presence, distribution, and phenotype of ET-1-positive cells and prepro-ET-1 (pp-ET-1) mRNA-expressing cells. ET-1-positive stromal cells and pp-ET-1 mRNA-expressing cells were detected with different distributions and relative frequencies in normal control appendix, histologically normal appendix, and inflamed appendix. Six of 20 histologically normal appendixes from patients with a clinical diagnosis of acute appendicitis had many ET-1-positive stromal cells and high pp-ET-1 mRNA expression, similar to inflamed appendix. Forty percent of the pp-ET-1 mRNA-expressing cells were neutrophils, and the other positive cells were mast cells and macrophages. We suggest that local production of ET-1 by neutrophils and other inflammatory cells could be a molecular sign of focal inflammation in histologically normal appendixes and that ET-1 could be implicated, with other cytokines, in the pathogenesis of appendicitis by inducing appendiceal ischemia through vasoconstriction.  相似文献

Résumé Diverses réactions histochimiques (A.P.S. et bleu de toluidine) démontrent que l'épiderme du Gymnarchus niloticus est riche en mucopolysaccharides. Ils se rencontrent à la fois dans le cytoplasme des cellules superficielles de l'épiderme et au niveau des organes récepteurs cutanés de la ligne latérale. Les mucopolysaccharides dans les cellules épidermiques superficielles sont neutres.Les mucopolysaccharides des cavités intraépidermiques des organes de type B et C et du canal de l'organe de type A sont métachromatiques, La métachromasie est due à la présence d'un radical acide carboxylé. Les particularités dans la constitution et la répartition du mucus au niveau de ces trois organes résident dans le fait que l'invagination de la cellule sensorielle de l'organe de type A est occupé par du mucus neutre et que le cytoplasme de la petite cellule de l'organe de type C contient du mucus acide de même nature que celui de la cavité intraépidermique.Il faut admettre que l'origine des mucopolysaccharides réside dans les cellules de soutien qui entourent la cellule sensorielle.
Histochemical investigation of the mucopolysaccharides in the epidermis and lateral line sensory organs of gymnarchus niloticus
Summary The mucopolysaccharide content of different epidermal structures in Gymnarchus niloticus has been demonstrated by two histochemical methods, periodic acid Schiff (P.A.S.) and toluidine blue. Mucopolysaccharides are present in the cytoplasm of the superficial cells and in the three specific cutaneous sensory organs of the lateral line system, type A, B and C.The superficial epidermal cells contain only neutral mucopolysaccharide.The mucopolysaccharides of the intraepidermal cavities in the type B and C organs as well as in the pores of the type A organ are acid; the metachromatic reaction of the toluidine blue demonstrates the presence of an acid carboxylic radicle.Histochemical analysis of the pore contents of the type A organ reveals that the mucus in the pore is acid while that of the sensory cell invagination is neutral.Histochemical differences were determined between the two sensory cells of the organ type C: the cytoplasm of the small sensory cell contains acid mucopolysaccharides while that of the large sensory cell is free of it, although the cytoplasmic invagination (extracellular) of the latter is filled with acid mucus.The results suggest that the mucus in each organ type is produced by the supporting cells surrounding the sensory cells.

Bats have a very high mass-specific energy demand due to small size and active flight. European bat species are mostly insectivorous and the morphology of the gastrointestinal (GI) tract should be adapted accordingly. This study investigated the general anatomy by histology and the function by analysing carbohydrate distribution in particular of the mucus of the GI tract of the insectivorous bat Pipistrellus pipistrellus. The GI tracts of three individuals were dissected, fixed in formaldehyde, and embedded in paraffin wax. The tissues and cells of the GI tract of P. pipistrellus were analysed by classical (acid alizarin blue, haematoxylin-eosin, and Masson Goldner Trichrome), histochemical (periodic acid-Schiff, Alcian blue at pH 2.5) and lectin histochemical (lectins WGA and HPA) staining procedures. The GI tract of P. pipistrellus is organised into the typical mammalian layers. The short, narrow, and thin-walled esophagus is simple with a folded stratified squamous epithelium without glands but mucous surface cells secreting neutral mucus. The stomach is globular shaped without specialisation. Mucous surface cells produced neutral mucus whereas neck and parietal cells secreted a mixture of neutral and acid mucus. Chief cell surface was positive for N-acetylglucosamine and the cytoplasm for N-acetylgalactosamine residues. The intestine lacked a caecum and appendix. The small intestine was divided into duodenum, jejunum-ileum and ileum-colon. The epithelium consisted of columnar enterocytes and goblet cells. The large intestine is short, only represented by the descending colon-rectum. It lacked villi and the mucosa had only crypts of Lieberkühn. Towards the colon-rectum, goblet cells produced mucus with N-acetylglucosamine residues increasing in acidity except in colon-rectum where acidity was highest in the base of crypts. Along the tube the surface of enterocytes was positive for N-acetylglucosamine and N-acetylgalactosamine. All over the mucus filling the lumen of the GI tract was positive for N-acetylglucosamine and increased in acidity in all parts except of the stomach.In conclusion, the simple GI tract showed an anatomical reduction of tissue enabling for a short retention time and a reduction of the load carried during flight: short GI tract, lack of lymphoid tissue, missing of glands in certain regions, and a distinct pattern of mucus distribution, indicating different physiological functions of these areas. The GI tract of P. pipistrellus was typical for an insectivorous species probably representing the ancestral condition.Key words: Chiroptera, esophagus, glycoconjugates, intestine, lectins, stomach  相似文献

Nautiloidea is the oldest group within the cephalopoda, and modern Nautilus differs much in its outer morphology from all other recent species; its external shell and pinhole camera eye are the most prominent distinguishing characters. A further unique feature of Nautilus within the cephalopods is the lack of suckers or hooks on the tentacles. Instead, the animals use adhesive structures present on the digital tentacles. Earlier studies focused on the general tentacle morphology and put little attention on the adhesive gland system. Our results show that the epithelial parts on the oral adhesive ridge contain three secretory cell types (columnar, goblet, and cell type 1) that differ in shape and granule size. In the non-adhesive aboral epithelium, two glandular cell types (cell types 2 and 3) are present; these were not mentioned in any earlier study and differ from the cells in the adhesive area. The secretory material of all glandular cell types consists mainly of neutral mucopolysaccharide units, whereas one cell type in the non-adhesive epithelium also reacts positive for acidic mucopolysaccharides. The present data indicate that the glue in Nautilus consists mainly of neutral mucopolysaccharides. The glue seems to be a viscous carbohydrate gel, as known from another cephalopod species. De-attachment is apparently effectuated mechanically, i.e., by muscle contraction of the adhesive ridges and tentacle retraction.  相似文献
