An analysis of milk constituents during various stages of lactation in the southern elephant seal Mirounga leonina was carried out. Forty-six milk samples were taken from 30 females throughout lactation during 1985, 1987, 1990 and 1991 on Stranger Point, King George Island, South Shetland Islands. Total nitrogen (TN), non-protein nitrogen (NPN), sugar, fat, ash and water were measured, and from some of these data true protein and energy content were calculated. The results showed a high degree of variation in water and fat concentrations among samples at different stages of lactation. During the first 20 days the fat content of milk increased from about 12 to approximately 52%, while water content fell from 70 to 33%. The composition of milk changes rapidly during the first days post-partum. Protein, minerals and sugar appear to remain stable after the fourth day of lactation. Milk samples contain significant levels of sugars; thin layer chromatography indicates the presence of lactose and glucose together with other unidentified components. There is evidence of a striking change in composition of the milk in the later part of lactation; the progressive increase in the fat:water ratio is abruptly reversed just prior to weaning.  相似文献

Blood and milk samples from foremilk during afternoon milking, were simultaneously collected from 285 dairy goats. In experiment 1, fiva cyclic goats were sampled daily for 21 days. In experiment 2, 280 females from 9 flocks were submitted to sampling 21 days after insemination. In addition, some milk samples were divided in two parts, after which one was frozen and the other kept at +4 degrees C until assay. Progesterone concentrations were measured in blood, whole milk and milk fat by radioimmunoassay. No difference in whole milk or fat progesterone levels was found between frozen and refrigerated milk samples. Milk butterfat progesterone concentrations paralleled those in plasma or whole milk throughout the estrous cycle and ranged from about 20 ng/ml at estrus to about 400 ng/ml in mid-luteal phase. The ratio of mid-luteal phase progesterone levels to those seen in the estrous period was over 20 in fat and in blood. This ratio was very much lower in whole milk. Consequently the determination of pregnant and non-pregnant goats from the samples collected 21 days after service was very much easier and accuracy was better when the progesterone content was assayed from milk fat than from whole milk. It was concluded that early pregnancy diagnosis in goats can be done routinely by determination of progesterone levels in milk fat.  相似文献

The fertility of high-performance (high milk yield) dairy breeds such as the Holstein within the Australian dairy herd has been on the decline for the past two decades. The 12-month calving interval for pasture-based farming practices results in oocyte maturation coinciding with peak lactation, periods of negative energy balance, and energy partitioning for lactation, causing energy deficiency in some organ systems, including the reproductive system. Oocyte developmental competence (the ability to undergo successful fertilization, embryo development, and establishment of pregnancy) is intrinsically linked with the composition of follicular fluid (FF). The aim of this study was to determine if there was a relationship between the fat and carbohydrate levels in plasma and FF and the ability to support in vitro oocyte maturation (IVM). Plasma and FF were collected in vivo from eight Holstein cows between 52 and 151 days post-partum. Plasma glucose trended (P = 0.072) higher and triglyceride levels were significantly higher than in FF (P < 0.05), but there were no relationships between FF and plasma composition. Glucose FF concentration was negatively related to follicular lactate and nonesterified fatty acid (NEFA) levels and days post-partum. Conversely, FF triglyceride concentrations were positively related to FF NEFA levels and negatively related to milk fat and protein composition. Abattoir-derived cumulus–oocyte complexes were cultured in either 50% FF (FF-IVM) or 50% plasma (plasma-IVM), with on-time embryo development then assessed. Although there were no differences between animals, the blastocyst rates after FF-IVM were negatively related to plasma glucose and days post-partum and positively related to body condition score and plasma NEFA levels. In comparison to the previous studies, total NEFA levels in FF were not related to animal parameters and did not influence oocyte developmental competence in vitro. Results from this study suggest that days post-partum and body condition score influence carbohydrate metabolism within the follicular environment, and this may be attributed to the pasture-based feed system applied in the Australian dairy industry.  相似文献

The effects of colchicine on release of milk lipids from mammary tissue were evaluated by biochemical analysis of milk and morphological study of the tissue following intramammary infusions of the alkaloid into lactating goats. Colchicine produces a reversible drop in milk yield. As the flow of milk resumes, 36–48 h after infusion, the fat content of the milk increases, phospholipid per g of total globule lipid falls, mean size of milk fat globules increased and diameters of fat droplets (presecretory milk fat globules) within lactating cells approximately double. These observations are consistent with the conclusion that colchicine suppresses milk fat globule secretion but that globules continue to grow in size within cells during the suppression period. These findings indicate that secretion of milk fat globules and the skim milk phase are coupled.  相似文献

Conjugated and unconjugated oestrone, oestradiol and oestriol were measured in simultaneous milk and plasma samples obtained from 21 women in the early post-partum period. Conjugated oestrogens comprised more than 90% of the total oestrogen content of both milk and plasma. Oestrone glucosiduronate was the major oestrogen metabolite in milk (33%), the levels being significantly higher (P less than 0.01) than in plasma. Oestriol glucosiduronates were the predominant oestrogen metabolites (63%) in plasma.  相似文献

Purified plasma membrane fractions from lactating bovine mammary glands and membranes of milk fat globules from the same source were similar in distribution and fatty acid composition of phospholipids. The sphingomyelin content of the phospholipid fraction of both membranes was higher than in these fractions from other cell components, β-carotene, a constituent characteristic of milk fat, was present in the lipid fraction of the plasma membrane. Cholesterol esters of plasma membrane were similar in fatty acid composition to those of milk fat globule membranes. Disc electrophoresis of either membrane preparation on polyacrylamide gels revealed a single major protein component characteristic of plasma membrane from other sources. Distinct morphological differences between plasma membrane and milk fat globule membranes were observed in both thin sections and in negatively stained material. Plasma membrane was vesicular in appearance while milk fat globule membranes had a platelike aspect. These observations are consistent with derivation of fat globule membrane from plasma membrane accompanied by structural rearrangement of membrane constituents.  相似文献

Summary The lactating mammary gland is one of the most highly differentiated and metabolically active organs in the body. Membranes of the lactating mammary cell have important roles in transmitting from one membrane to another of hormonal information and in milk secretion, which is the final event. During milk secretion, the projection of the surface membrane into the alveolar lumen by enveloping intracellular lipid droplets with the apical plasma membrane is one of the most remarkable aspects of biological membrane action throughout nature.This review focuses on current knowledge about membranes in the lactating mammary gland. (1) Advances in the isolation and properties of membranes, especially the plasma membrane and Golgi-derived secretory vesicles, concerned with milk secretion from the lactating mammary gland are described. (2) Milk serum components are secreted by fusing the membranes of secretory vesicles that condense milk secretions with the plasma membrane in the apical regions. This occurs through the formation of a tubular-shaped projection and vesicular depression in a ball-and-socket configuration, as well as by simple fusion. (3) Intracellular lipid droplets are directly extruded from the mammary epithelial cells by progressive envelopment of the plasma membranes in the apical regions. (4) The balance between the surface volume lost in enveloping lipid droplets and that provided by fusion of the secretory vesicle and other vesicles with the apical plasma membrane is discussed. (5) The membrane surrounding a milk fat globule, which is referred to as the milk fat globule membrane (MFGM), is composed of at least the coating membrane of an intracellular lipid droplet, of the apical plasma membrane and secretory vesicle membrane, and of a coat material. Consequently, MFGM is molecularly different from the plasma membrane in composition. (6) MFGM of bovine milk is structurally composed of an inner coating membrane and outer plasma membrane just after segregation. These two membranes are fused and reorganized through a process of vesiculation and fragmentation to stabilize the fat globules. Delay in the onset of lactogenesis has been shown to occur in women who are obese, however the mechanism altered within the mammary gland causing the delay remains unknown. Consumption of high fat diets (HFD) has been previously determined to result decreased litters and litter numbers in rodent models due to a decrease in fertility. We examined the effects of feeding a HFD (60% kcal from fat) diet versus a low-fat diet (LFD; 10% kcal from fat) to female Wistar rats on lactation outcomes. Feeding of HFD diet resulted in increased pup weights compared to pups from LFD fed animals for 4 d post-partum. Lactation was delayed in mothers on HFD but they began to produce copious milk volumes beginning 2 d post-partum, and milk yield was similar to LFD by day 3. Mammary glands collected from lactating animals on HFD diet, displayed a disrupted morphologies, with very few and small alveoli. Consistently, there was a significant decrease in the mRNA expression of milk protein genes, glucose transporter 1 (GLUT1) and keratin 5 (K5), a luminobasal cell marker in the mammary glands of HFD lactating animals. Expression of tryptophan hydroxylase 1 (TPH1), the rate-limiting enzyme in serotonin (5-HT) biosynthesis, and the 5-HT(7) receptor (HTR7), which regulates mammary gland involution, were significantly increased in mammary glands of HFD animals. Additionally, we saw elevation of the inflammatory markers interleukin-6 (IL-6) and tumor necrosis factor-α (TNF- α). These results indicate that consumption of HFD impairs mammary parenchymal tissue and impedes its ability to synthesize and secrete milk, possibly through an increase in 5-HT production within the mammary gland leading to an inflammatory process.  相似文献

The potential to modify milk fatty acid composition and milk production by dietary administration of marine oils rich in n-3 PUFAs in goats diets is reviewed. Moreover animal and human health implications are considered. Role of nutrition in dairy goats for enhancing content of CLA in milk fat is also discussed. At last, rumen protected choline supplementation is evaluated to improve productive performance and metabolic health. While the effects of n-3 PUFAs administration on goat productive performance seem to depend on many factors, fish oil administration has been extensively shown to lower average concentration of C18:0 and saturated fatty acids, with a relative increase of C16:1, C18:3 n-3 and very long-chain n-3 PUFAs. Positive results have been evidenced in animals health following administration of EPA and DHA from fish oil, leading to increased phagocytic activity with no effects on extracellular ROS production in incubated goats cells. The nutritional and health properties of goat's milk could be further improved by increasing the content of CLA in milk fat. Provision of PUFAs from fresh pasture and plant lipids, mainly C18:2 n-6 and C18-3 n3 which serve as precursor for trans C18:1 formation in the rumen, have proved to enhance content of CLA in goat milk fat. Marine oils rich in n-3 PUFAs have been shown to be very effective at increasing CLA content in bovine milk, but very scarce data are available on dairy goats.Rumen protected choline has been show to increase productive performance, particularly milk production, fat percentage, and fat and protein yield without detrimental effects on methyl groups, thus reducing BHBA plasma content and hepatocellular lipid accumulation around transition.However the magnitude of the production response seems to be affected by the composition of the diet, and other factors as already reported for n-3 PUFAs administration.  相似文献

More (P less than 0.05) post-partum acyclic ewes (8/9) showed evidence of pulsatile LH release than did seasonally anoestrous ewes (2/8). Mean plasma prolactin concentrations were higher (P less than 0.05) in the post-partum ewes than in the seasonally anoestrous ewes. Hypothalamic and pituitary cytosol oestrogen receptor number, median eminence GnRH content and pituitary LH, FSH and prolactin contents were similar in the two groups of ewes. The number of nuclear-bound oestrogen receptors was greater (P less than 0.01) in pituitaries from seasonally anoestrous ewes than in pituitaries from post-partum ewes. These data suggest that the basis of acyclicity in seasonally anoestrous ewes and in post-partum ewes is probably due to their inability to generate LH pulse frequencies similar to that in the follicular phase of the oestrous cycle. The higher LH pulse frequency in post-partum ewes, compared to that in seasonally anoestrous ewes, suggests that pregnancy may partly negate the reduction in LH secretion that is associated with photoperiod-induced acyclicity. The lower number of nuclear-bound oestrogen receptors in post-partum ewes suggests that there may be lower plasma oestrogen levels and less ovarian follicular growth than in non-suckling anoestrous ewes.  相似文献

After intravenous administration of alloxan (50 mg kg-1 liveweight) to lactating ewes, there were triphasic changes in plasma glucose and insulin. Almost immediately, plasma insulin decreased and hyperglycaemia occurred, then, between c. 5-12 h, insulin increased and ewes became hypoglycaemic. Thereafter, insulin decreased and glucose increased from c. 20 h after alloxan and the diabetic state was established. Changes in glucose production and utilization correlated with changes in plasma glucose. Exogenous insulin was administered from 30 h after alloxan, and it took some 2 weeks to stabilize ewes. During this period, when mild hyperglycaemia persisted, milk yields and feed intakes were decreased but milk fat content was elevated. Once ewes were stabilized, plasma glucose, milk yield, feed intake and milk fat content returned to levels prior to alloxan. These observations are consistent with insulin playing a role in the aetiology of the 'low milk fat syndrome' in the ruminant. It appears that the alloxan-treated, insulin-stabilized ewe would be a useful model for studying the role of insulin during lactation, but it is necessary to allow time for animals to overcome effects of administration of alloxan.  相似文献

There is very little information on the composition of lion milk. Milk was obtained from all four teats of two African lionesses (Panthera leo) while they were temporarily immobilized; the first lioness was 40 hours postpartum and the second lioness was 10 days postpartum. The lion's milk obtained from the two lionesses in this study contained less solids than reported in literature. The milk of the two females contained, respectively, 192.2 and 166.9 g dry matter/kg milk; 60.2 and 84.6 g protein/kg milk; 113.6 and 136.9 g fat/kg milk; and 68.6 and 77.9 g fat free dry matter/kg milk. Carbohydrate content, analysed only for the first lioness, was 26.5 g lactose/kg milk. These values are much lower than those cited in literature. Electrophoresis and identification of protein bands showed a similar pattern of proteins as seen in cow's milk, but with caseins of less negative charge and whey proteins of smaller molecular size. The milk fat content of the second lioness was higher and varied considerably more among teats. The lipid fraction of lion's milk is characterized by a high content of saturated (palmitic and stearic) and mono-unsaturated (palmitoleic and oleic) fatty acids. Regarding fatty acid composition, lion milk closely resembles human milk. Animal fat plays an important role in the nutrition of African lions. The high fat content of lion milk suggests that the dependency by large predators such as lions on dietary fat is present from birth.  相似文献

A solid-phase radioimmunoassay using antibody-coated tubes and 125I-progesterone label was developed to provide an alternative to the conventional aqueous method of assaying progesterone in milk for early pregnancy diagnosis. The accuracy of diagnoses made using the solid-phase assay of progesterone in milk was assessed by comparison with diagnoses made using an aqueous assay of serum progesterone. Both methods agreed in the thirteen cows that were diagnosed. When “fat-free” milk was assayed by both aqueous (x) and solid-phase (y) methods, the progesterone values which resulted showed a high correlation (r = .94) and a linear relationship of y = 1.67x ? 0.68. Milk samples, in which the fat concentration ranged from 0.20 to 4.04%, were assayed by the solid-phase method and no relationship between milk fat and progesterone concentration was observed. Pregnancy diagnosis from solid-phase assay of milk samples collected on the day of breeding, 21 and 23 days following breeding was performed on 62 Holstein cattle. The accuracy of “non-pregnant” diagnoses was 95% to 100% and the accuracy of “pregnant” diagnoses was 80% if breeding day values were used and 72% if these values were excluded.The accuracy of this assay in diagnosing pregnancy was equal to that of previously published assays and provided the advantages of requiring less technical time and equipment. In addition, foremilk, composite or stripping samples can be accomodated in this assay since the estimates of progesterone are not affected by the concentration of milk fat.  相似文献

The objective was to determine performance and milk fatty acid changes of high producing dairy cows in early lactation, under summer heat, by adding a supplemental rumen inert fat in the form of a saturated free fatty acid (856 g/kg C16:0/kg of total fatty acids) to the total mixed ration (TMR). Early lactation multiparous Holstein cows in two similar pens of 99 and 115 cows were used in a 2 × 2 Latin Square design experiment with 35 d periods during a period when daily high and low temperatures averaged 34.3 and 15.9 °C, the relative humidity averaged 51% and there were no rain events. The TMR was the same for both groups, consisting of approximately 435 g/kg forage and 565 g/kg concentrate, except that the vitamin/mineral premix had no added fat (control, C) or added fat (C16:0) at a level designed to deliver approximately 450 g/cow/d of supplemental fat if cows consumed 26.5 kg/d of dry matter (DM). The two TMR averaged 905 g/kg organic matter (OM), 318 g/kg neutral detergent fiber (aNDF), and 186 g/kg crude protein (CP). The ‘C’ TMR had 58 g/kg total fatty acids with an estimated net energy for lactation (NEl) of 7.3 MJ/kg (DM), while the C16:0 TMR had 72 g/kg total fatty acids and 7.5 MJ/kg NEl (DM). Whole tract digestibility of DM, OM, aNDF and CP tended (P<0.10) to increase, and that of fatty acids increased substantially (P<0.01), with C16:0 feeding, whereas, DM intake was not affected. Milk fat content decreased (P<0.01) with C16:0 feeding (37.5 versus 36.0 g/kg), whereas, true protein content tended (P=0.09) to increase. There was a tendency (P=0.07) for increased milk yield (36.69 versus 38.04 kg/d), while milk protein yield increased (P=0.03) with C16:0 supplementation (1.08 versus 1.13 kg/d). Milk fat yield was unaffected by treatment. Concentrations of short and medium chain milk fatty acids (C6:0–C15:0), decreased, or tended to decrease, with C16:0 addition (C13:0 and C15:0, P<0.10; all others, P≤0.05). The concentration of C16:0 increased (P<0.001) in milk triglycerides from cows fed C16:0 (27.10 versus 31.57 g/kg), the longer chain saturated fatty acids C17:0 and C18:0 decreased (P≤0.05) and other long chain unsaturated fatty acids were unaffected. Benefits of C16:0 feeding on cow productivity must be balanced against negative effects on the nutritive value of the milk (i.e., increased C16:0 in milk fatty acids) produced for human consumption. However, relatively low amounts of supplemental C16:0 (27.10 versus 31.57 g/kg in milk triglycerides for C and C16:0 supplemented cows, respectively) were actually secreted in milk, in spite of them being essentially fully digested in the digestive tract. Strategies to divide cows into production groups based on milk yield and/or milk fat proportions could further limit C16:0 secretion in milk. Supplemental dietary C16:0 may have positive effects on milk production that outweigh the negative health effects of the increased C16:0 content in the milk fat.  相似文献

Ultrastructural observations were made of the non-lactating and lactating mammary gland of the Weddell seal, and measurements of several cell components were compared with those in other species to determine whether there are any morphological modifications within the gland to explain the unusual milk composition and very rapid growth of sucking young in this species.
The mammary parenchyma in non-lactating and lactating Weddell seals is almost identical morphologically to that found in other mammals. Although the relative volumes of most organelles are similar to those in other eutherians, the relative volume of rough endoplasmic reticulum (RER) is reduced in the Weddell seal. In addition, the absolute and relative volumes of Golgi vacuoles are smaller in the Weddell seal, probably associated with the low lactose and water content of the milk. The synthesis and secretion of milkspecific proteins and fat droplets by mammary secretory cells in the Weddell seal appear to be identical to other eutherians. Relatively small numbers of fat droplets less than 1 μm diameter are present in epithelial cells, alveolar lumina and milk samples, and there is a far greater contribution by large fat droplets to the total fat volume of Weddell seal milk than milk from other mammals.  相似文献

The supplementation of conjugated linoleic acids (CLA) to the rations of dairy cows represents an opportunity to reduce the content of milk fat. Therefore, CLA have the potential beneficial effect of reducing energy requirements of the early lactating cow. The present study aimed at the examination of long-term and posttreatment effects of dietary CLA intake on performance, variables of energy metabolism-like plasma levels of non esterified fatty acids (NEFA) and beta-hydroxybutyrate (BHB), and fatty acid profile in milk fat. Forty-six pregnant German Holstein cows were assigned to one of three dietary treatments: (1) 100 g/ d of control fat supplement (CON), (2) 50 g/d of control fat supplement and 50 g/ d of CLA supplement (CLA-1) and (3) 100 g/d of CLA supplement (CLA-2). The lipid-encapsulated CLA supplement consisted of approximately 10% of trans-10, cis-12 CLA and cis-9, trans-11 CLA each. The experiment started 1 d after calving and continued for about 38 weeks, divided into a supplementation (26 weeks) and a depletion period (12 weeks). Over the first 7 weeks of treatment, 11 and 16% reductions in dry matter intake compared to control were observed for the cows fed CLA-1 and CLA-2 supplements respectively. Consequently, the calculated energy balance for these two CLA groups was lower compared to the control. Plasma levels of NEFA and BHB remained unaffected. Later in lactation the highest CLA supplementation resulted in a reduction of milk fat content of 0.7%. However, no reduction in milk fat yield, and accordingly no milk fat depression (MFD), could be shown. The trans-10, cis-12 CLA in milk fat increased with increasing dietary CLA supplementation in a dose-dependent manner. The proportion of C16 in milk fat was decreased by the highest CLA supplementation. With the exception of an increase in plasma glucose level in the CLA-2 group, no post-treatment effects were observed. Overall, under the conditions of the present study no improvement in the calculated energy balance by CLA supplementation could be shown for the entire evaluation period.  相似文献

Since many dairy cows calve during late summer, the objective was to determine if heat stress immediately post-partum would (1) alter metabolism, thus, increasing susceptibility to metabolic disorders, (2) affect lactation and/or (3) affect reproduction. Forty four cows, calving during late summer, were paired with one member of each pair stressed (HS) for the first 10 post-partum days in a hot barn. Controls (CC) were kept in a cooled section of the barn. Plasma drawn weekly for 7 weeks was analyzed in an autoanalyzer for calcium, inor. phosphorus, protein, glucose and cholesterol and by radioimmunoassay for cortisol and progesterone. Ovaries and uteri were palpated weekly. Rectal temperatures were significant higher for HS during the first 10 post-partum days. No significant effects on plasma constituents were observed during the 10-day treatment period. For the 7-week period, glucose and cholesterol were lower in HS, as were cyclic peaks of progesterone and cortisol. Both calcium and inorganic phosphorus remained clinically low for the 7 weeks, but no treatment effects were seen. Uteri of HS involuted more rapidly than the CC. Treatment did not affect reproductive efficiency. Lactation milk yields did not differ, but milk fat percent was lower in HS. In this study we document growth, milk intake and energy consumption in nursing pups of icebreeding grey seals (Halichoerus grypus). Change in body composition of the pups, change in milk composition as lactation progresses, and mass transfer efficiency between nursing mothers and pups are also measured. Mass transfer efficiency between mother-pup pairs (n=8) was 42.5±8.4%. Pups were gaining a daily average of 2.0±0.7 kg (n=12), of which 75% was fat, 3% protein and 22% water. The total water influx was measured to be 43.23±8.07 ml·kg-1·day-1. Average CO2 production was 0.85±0.20 ml·g-1·h-1, which corresponds to a field metabolic rate of 0.55±0.13 MJ·kg-1·day-1, or 4.5±0.9 times the predicted basal metabolic rate based on body size (Kleiber 1975). Water and fat content in the milk changed dramatically as lacation progressed. At day 2 of nursing, fat and water content were 39.5±1.9% and 47.3±1.5%, respectively, while the corresponding figures for day 15 were 59.6±3.6% fat and 28.4±2.6% water. Protein content of the milk remained relatively stable during the lactation period with a value of 11.0±0.8% at day 2 and 10.4±0.3% at day 15. Pups drank an average of 3.5±0.9 kg of milk daily, corresponding to a milk intake of 1.75 kg per kg body mass gained. The average daily energy intake of pups was 82.58±19.80 MJ, while the energy built up daily in the tissue averaged 61.72±22.22 MJ. Thus, pups assimilated 74.7% of the energy they received via milk into body tissue. Two lactating cows were given tritiated hay containing organically bound tritium (OBT) only for about 4 weeks. Tritium activity was determined in milk fat, casein, lactose, milk water, and whole milk. In one cow, milk was sampled for approximately 450 days, covering two lactation periods. At steady state, specific tritium activities in casein, lactose, and milk water were 58, 10, and 11%, respectively, of those in milk fat. Some OBT was converted into THO during catabolism and entered the body water pool. This 3H source accounted for nearly 40% of tritium in lactose, but in casein and milk fat about 97% of tritium was derived from ingested OBT. Comparison of the specific activity of milk constituents with the specific activity of ingested hay showed the following values: 0.84 for milk fat, 0.49 for casein, 0.05 for lactose, 0.10 for milk water. About one-half of the tritium transferred to milk was found in organic milk constituents and the other half in milk water. Decrease of tritium activity with time could be represented by three components with different half-lives for the organic milk constituents. Those for milk fat and casein were quite similar, with a slow component of nearly 3 months.  相似文献

During secretion of milk fat globules, triacylglycerol (TAG) droplets are enveloped by a phospholipid (PL) trilayer. Globule size has been found to be related to polar lipid composition and fat content, and milk fat content and fatty acid composition have been associated with the diacylglycerol acyltransferase 1 (DGAT1) K232A polymorphism; however, the association between the DGAT1 polymorphism and fat globule size and polar lipid composition has not been studied. The ratio between polar and neutral lipids as well as the composition of the polar lipids in milk has industrial as well as nutritional and health implications. Understanding phenotypic and genotypic factors influencing these parameters could contribute to improving milk lipid composition for dairy products. The focus of the present study was to determine the effect of both fat content and DGAT1 polymorphism on PL/TAG ratio, as a marker for milk fat globule size, and detailed PL composition. Milk samples were selected from 200 cows such that there were equal numbers of samples for the different fat contents as well as per DGAT1 genotype. Samples were analyzed for neutral and polar lipid concentration and composition. PL/TAG ratio was significantly associated with both fat content and DGAT1 genotype. Phosphatidylinositol and phosphatidylserine concentrations were associated with fat content*DGAT1 genotype with a stronger association for the AA than the KK genotype. Sphingomyelin concentration tended to interact with fat content*DGAT1 genotype. Phosphatidylethanolamine (PE) concentration showed a biphasic response to fat content, suggesting that multiple biological processes influence its concentration. These results provide a new direction for controlling polar lipid concentration and composition in milk through selective breeding of cows.  相似文献
