A computer-controlled force gauge designed to measure the adhesive strength of barnacles on test substrata is described. The instrument was evaluated with adult barnacles grown in situ on Silastic T2®-coated microscope slides and epoxy replicas adhered to the same substratum with synthetic adhesive. The force per unit area required to detach the barnacles (critical removal stress) using the new automated system was comparable to that obtained with ASTM D5618 (1994 ASTM D5618. 1994. A long-term investigation of the shell shape and the basal morphology of barnacles grown on tough, double-network (DN) hydrogels and polydimethylsiloxane (PDMS) elastomer was conducted in a laboratory environment. The elastic modulus of these soft substrata varied between 0.01 and 0.47 MPa. Polystyrene (PS) (elastic modulus, 3 GPa) was used as a hard substratum control. It was found that the shell shape and the basal plate morphology of barnacles were different on the rigid PS substratum compared to the soft substrata of PDMS and DN hydrogels. Barnacles on the PS substratum had a truncated cone shape with a flat basal plate while on soft PDMS and DN gels, barnacles had a pseudo-cylindrical shape and their basal plates showed curvature. In addition, a large adhesive layer was observed under barnacles on PDMS, but not on DN gels. The effect of substratum stiffness is discussed in terms of barnacle muscle contraction, whereby the relative stiffness of the substratum compared to that of the muscle is considered as the key parameter.  相似文献

A number of factors affect the adhesion strength of organisms to fouling-release coatings, and except for a few studies focussing on black or white surfaces none have dealt specifically with the effect of coating colour. The aim was to test the effect of colour on the adhesion strength of the barnacle Elminius modestus. Panels coated in six commercial colours of Intersleek 700® were submerged at two field sites and barnacles were pushed-off using a standard assay procedure. The strength of adhesion (SOA) varied between and within sites for colour and by barnacle basal area, SOA per unit area being higher for smaller barnacles. Higher SOA with a small basal area may be because of size-specific predation, differential hydrodynamic effects or adhesive failure with age. The complex effect of colour on barnacle adhesion may be because of physico-chemical surface characteristics varying with pigments, and their interactions with local environmental conditions, as well as interactions with the settling barnacle larvae.  相似文献

Release mechanisms of barnacles (Amphibalanus amphitrite or Balanus amphitrite) reattached to platinum-cured silicone coatings were studied as a function of coating thickness (210-770 microm), elastic modulus (0.08-1.3 MPa), and shear rate (2-22 microm s(-1)). It was found that the shear stress of the reattached, live barnacles necessary to remove from the silicone coatings was controlled by the combined term (E/t)(0.5) of the elastic modulus (E) and thickness (t). As the ratio of the elastic modulus to coating thickness decreased, the barnacles were more readily removed from the silicone coatings, showing a similar release behavior to pseudobarnacles (epoxy glue). The barnacle mean shear stress ranged from 0.017 to 0.055 MPa whereas the pseudobarnacle mean shear stress ranged from 0.022 to 0.095 MPa.  相似文献

The microbiological diagnosis of respiratory tract infections requires serial manual dilutions of the clinical specimen before agar plate inoculation, disrupting the workflow in bacteriology clinical laboratories. Automated plating instrument systems have been designed to increase the speed, reproducibility and safety of this inoculating step; nevertheless, data concerning respiratory specimens are lacking. We tested a specific procedure that uses the Previ Isola® (bioMérieux, Craponne, France) to inoculate with broncho-pulmonary specimens (BPS). A total of 350 BPS from a university-affiliated hospital were managed in parallel using the manual reference and the automated methods (expectoration: 75; broncho-alveolar lavage: 68; tracheal aspiration: 17; protected distal sample: 190). A specific enumeration reading grid, a pre-liquefaction step and a fluidity test, performed before the inoculation, were designed for the automated method. The qualitative (i.e., the number of specimens yielding a bacterial count greater than the clinical threshold) and quantitative (i.e., the discrepancy within a 0.5 log value) concordances were 100% and 98.2%, respectively. The slimmest subgroup of expectorations could not be managed by the automated method (8%, 6/75). The technical time and cost savings (i.e., number of consumed plates) reached 50%. Additional studies are required for specific populations, such as cystic fibrosis specimens and associated bacterial variants. An automated decapper should be implemented to increase the biosafety of the process. The PREVI Isola® adapted procedure is a time- and cost-saving method for broncho-pulmonary specimen processing.  相似文献

Principal stresses acting in the midshaft cortices of the tibia and metatarsus of the horse were determined from in vivo rosette strain gauge recordings for overground locomotion at different gaits, as well as for jumping and acceleration. Bone stresses were correlated with limb kinematics and ground reaction forces. The results for these two hind limb bones were compared to earlier determinations of locomotor stress in the forelimb radius and metacarpus (Biewener, Thomason & Lanyon, 1983b). Peak stresses generally increased with increasing speed; however, because of greater bending, stresses in the tibia were substantially higher (45%) than in the metatarsus over the range of steady state speeds. Bending of the tibia resulted from significant off-axis loading by the ground reaction force. In contrast, the metatarsus was loaded in compression due to its close alignment with the ground reaction force. Peak stresses as high as - 53 MPa (caudal cortex) in the tibia and -38 MPa (plantar cortex) in the metatarsus acted at a canter. Increased skeletal stress was matched by a corresponding increase in ground reaction force and a decrease in hind limb duty factor. In both bones, peak stresses were significantly greater and differed in their distribution during jumping and acceleration, compared to peak stresses during steady speed locomotion. Maximal values of - 126 MPa (cranial cortex) in the tibia and - 117 MPa (dorsal cortex) in the metatarsus were developed during jumping. These stresses are similar in magnitude to those reported for a range of different sized mammals during strenuous activity and correspond to a safety factor to yield failure of 1.5 to 3. Though generally consistent within an individual bone, the distribution and magnitude of stresses varied about 20% among individuals. This variation was greater for the metatarsus because of its lesser curvature, which diminishes the bone's ability to control for bending in a fixed direction.  相似文献

One of the central controversies regarding the evolution of adhesion concerns how adhesive force scales as animals change in size, either among or within species. A widely held view is that as animals become larger, the primary mechanism that enables them to climb is increasing pad area. However, prior studies show that much of the variation in maximum adhesive force remains unexplained, even when area is accounted for. We tested the hypothesis that maximum adhesive force among pad-bearing gecko species is not solely dictated by toepad area, but also depends on the ratio of toepad area to gecko adhesive system compliance in the loading direction, where compliance (C) is the change in extension (Δ) relative to a change in force (F) while loading a gecko’s adhesive system (C = dΔ/dF). Geckos are well-known for their ability to climb on a range of vertical and overhanging surfaces, and range in mass from several grams to over 300 grams, yet little is understood of the factors that enable adhesion to scale with body size. We examined the maximum adhesive force of six gecko species that vary in body size (~2–100 g). We also examined changes between juveniles and adults within a single species (Phelsuma grandis). We found that maximum adhesive force and toepad area increased with increasing gecko size, and that as gecko species become larger, their adhesive systems become significantly less compliant. Additionally, our hypothesis was supported, as the best predictor of maximum adhesive force was not toepad area or compliance alone, but the ratio of toepad area to compliance. We verified this result using a synthetic “model gecko” system comprised of synthetic adhesive pads attached to a glass substrate and a synthetic tendon (mechanical spring) of finite stiffness. Our data indicate that increases in toepad area as geckos become larger cannot fully account for increased adhesive abilities, and decreased compliance must be included to explain the scaling of adhesion in animals with dry adhesion systems.  相似文献

An automated hemagglutination-inhibition (HI) test for the "shipping fever" strain (SF-4) of parainfluenza 3 antibody in bovine sera was developed and compared to manual tube and microtiter test procedures. The automated system operating at 60 samples per hr provided the most test results per specified time period, and the manual tube test provided the least. The manual microtiter test and the automated system at 40 samples per hr, falling between the two above procedures, were comparable in the number of sera that could be titrated in 1 day by one technician. There was little difference between automated and manual test reproducibility when measured at the twofold titer one-dilution difference level. However, the automated system titrated a higher number of sera at the same titer on repeat runs than either of the manual test procedures. The automated one-quartile difference reproducibility (each twofold dilution subdivided into 4 units-"quartiles") was equal to the manual test one-dilution difference reproducibility. The standard deviation of the per cent variation from the mean of paired serum titers for 40-sample-per-hr runs ranged from +/-3.49 to +/-5.36%. The manual and automated systems were of comparable sensitivity in their detection of negative sera.  相似文献

Cyprids are the final planktonic stage in the larval dispersal of barnacles and are responsible for surface exploration and attachment to appropriate substrata. The nanomechanical properties of barnacle (Balanus amphitrite) cyprid permanent cement were studied in situ using atomic force microscopy (AFM). Force curves were recorded from the cement disc continually over the course of its curing and these were subsequently analysed using custom software. Results showed a narrowing of the pull-off force distribution with time, as well as a reduction in molecular stretch length over time. In addition, there was a strong correlation between maximum pull-off force and molecular stretch length for the cement, suggesting 'curing' of the adhesive; some force curves also contained a 'fingerprint' of modular protein unfolding. This study provides the first direct experimental evidence in support of a putative 'tanning' mechanism in barnacle cyprid cement.  相似文献

Fouling-release coatings prevent fouling of ships' hulls through hydrodynamic forces generated as the ship moves through the water. The effectiveness of such coatings may be evaluated by measuring the adhesion strength of settled organisms, e.g. barnacles. The influence of desiccation of the barnacle adhesive on such measurements was investigated. Shear forces required to remove barnacles of the genus Balanus increased during the course of desiccation up to the point when the barnacles suddenly self-detached. The increase was thought to be due to the rising cohesive strength of the adhesive. Growing tensile forces within the weakly cross-linked adhesive, however, are suggested to have led to self-detachment. The shear forces required to remove barnacles of the genus Elminius were generally low and did not differ significantly during the course of desiccation. The different results may be attributed to specific base morphologies. It was concluded that measuring the adhesion strength of members of the Balanidae on non-stick surfaces in air could produce flawed results due to the influence of desiccation of the barnacle adhesive. The investigations have also provided new insights into the characteristics of barnacle adhesive.  相似文献

Cancer diseases are a common problem of the population caused by age and increased harmful environmental influences. Herein, new therapeutic strategies and compound screenings are necessary. The regular 2D cultivation has to be replaced by three dimensional cell culturing (3D) for better simulation of in vivo conditions. The 3D cultivation with alginate matrix is an appropriate method for encapsulate cells to form cancer constructs. The automated manufacturing of alginate beads might be an ultimate method for large-scaled manufacturing constructs similar to cancer tissue. The aim of this study was the integration of full automated systems for the production, cultivation and screening of 3D cell cultures. We compared the automated methods with the regular manual processes. Furthermore, we investigated the influence of antibiotics on these 3D cell culture systems. The alginate beads were formed by automated and manual procedures. The automated steps were processes by the Biomek® Cell Workstation (celisca, Rostock, Germany). The proliferation and toxicity were manually and automatically evaluated at day 14 and 35 of cultivation. The results visualized an accumulation and expansion of cell aggregates over the period of incubation. However, the proliferation and toxicity were faintly and partly significantly decreased on day 35 compared to day 14. The comparison of the manual and automated methods displayed similar results. We conclude that the manual production process could be replaced by the automation. Using automation, 3D cell cultures can be produced in industrial scale and improve the drug development and screening to treat serious illnesses like cancer.  相似文献

The purpose of this study was to investigate the consistency of commonly reported rapid force characteristics utilizing both automated and manual muscle contraction onset detection methods. Twenty-four healthy volunteers performed isometric strength testing of the plantar flexor muscle group on two nonconsecutive days. Test–retest reliability was evaluated using intraclass correlation coefficients (ICCs), standard errors of measurement (SEM), and the SEM as a percentage of the mean (SEM%) for rate of force development (RFD), relative RFD, contractile impulse, and absolute force–time values at various epoch durations using automated and manual onset detection methods. For all rapid force variables, ICC and SEM% values ranged from 0.52 to 0.96 and 7.56% to 37.56%, respectively. For the majority of these variables (20 of 23), the automated onset detection method resulted in higher ICC and lower SEM% values compared to the manual onset detection method. Regardless of onset detection methodology, the consistency of relative RFD values declined following 50% of MVC. Collectively, these findings indicated that commonly evaluated rapid muscle force variables demonstrated acceptable relative and absolute consistency values. However, these values were generally superior for the automated onset detection methodology. Additionally, the consistency of relative RFD values declines following 50% MVC and therefore should be evaluated with caution.  相似文献

A quantitative genetics approach was used to examine variation in the characteristics of the adhesive plaque of the barnacle Balanus amphitrite Darwin attached to two silicone substrata. Barnacles settled on silicone polymer films occasionally form thick, soft adhesive plaques, in contrast to the thin, hard plaques characteristic of attachment to other surfaces. The proportion of barnacles producing a thick adhesive plaque was 0.31 for Veridian, a commercially available silicone fouling-release coating, and 0.18 for Silastic T-2, a silicone rubber used for mold-making. For both materials, significant variation among maternal families in the proportion of barnacles producing a thick adhesive plaque was observed, which suggests the presence of genetic variation, or maternal environmental effects, for this plaque characteristic. For the Veridian coating, barnacles expressing the thick adhesive plaque also exhibited significantly reduced tenacity. This represents the first reported case for potential genetic control of intraspecific phenotypic variation in the physical characteristics and tenacity of the adhesive of a fouling invertebrate.  相似文献

A commercially available instrument that automatically makes serial dilutions and delivers reagents was used for the determination of antistreptolysin O titers in serum. The automated method was compared with tube-dilution and manual microtitration techniques. It gave higher reproducibility of results and was quicker to perform than both the other tests; and it was much more economical in reagents than the tube test. The automated technique is considered to be the best of the three methods when more than a small number of specimens are examined at one time. It is now in routine use in our laboratory.  相似文献

The imprecision, inaccuracy, and clinical sensitivity of an automated differential counter based on pattern recognition technology (HEMATRAK model 590) were evaluated using the NCCLS standard H20-T. The data show that the instrument is capable of performing assays in a manner comparable with the reference method proposed by NCCLS. In addition, data are presented from a multicenter study involving 174 HEMATRAK instruments which showed that the automated systems produce results equivalent to manual procedures (based on approximately 350 individual assays).  相似文献

We investigated whether measurements of barnacle microtopography could be used as covariates to increase precision in randomised field experiments with intertidal Littorina populations. We used image analysis to quantify the microtopography of the barnacles within 100 cm2 quadrats in July 1994 using as variables: number of barnacles, number of dead empty barnacles, mean distance to nearest neighbouring barnacle, total area covered by barnacles, and number of pockets of different size-classes [extra-small (< 0.15 cm2), small (0.15 and < 0.30 cm2), medium ( 0.30 and < 0.45 cm2), large ( 0.45 and < 0.60 cm2), and extra-large (0.60 cm2)] at least 75% surrounded by barnacles. We then used these variables to predict the abundance of Littorina spp. in the same quadrat. Two variables: the number of extra-small pockets and the number of small pockets accounted for 50% of the variation in total littorinid snail abundance among quadrats for January 1995 and for 47% of the variation for August 1994 but for only 16% of the variation for June 1994. However the single variable, the number of barnacles, accounted for 29–36% of the variation in snail abundance on these dates and can be measured without special equipment. The best combination of covariates was the number of extra-small pockets alone. The number of extra-small pockets between barnacles may be influencing the snail abundance by providing a refuge from wave shock and heat stress/desiccation. We suggest that the availability of these refuges among the barnacles could act as a density-dependent regulator of the total population size of a particular species of Littorina.  相似文献
