The lipophilic triphenylmethylphosphonium cation (TPMP+) has been employed to measure delta psi m, the electrical potential across the inner membrane of the mitochondria of intact hepatocytes. The present studies have examined the validity of this technique in hepatocytes exposed to graded concentrations of inhibitors of mitochondrial energy transduction. Under these conditions, TPMP+ uptake allows a reliable measure of delta psi m in intracellular mitochondria, provided that the ratio [TPMP+]i/[TPMP+]e is greater than 50:1 and that at the end of the incubation more than 80% of the hepatocytes exclude Trypan blue. Hepatocytes, staining with Trypan blue, incubated in the presence of Ca2+, do not concentrate TPMP+. The relationships between delta psi m and two other indicators of cellular energy state, delta GPc and Eh, or between delta psi m and J0, were examined in hepatocytes from fasted rats by titration with graded concentrations of inhibitors of mitochondrial energy transduction. Linear relationships were generally observed between delta psi m and delta GPc, Eh or J0 over the delta psi m range of 120-160 mV, except in the presence of carboxyatractyloside or oligomycin, where delta psi m remained constant. Both the magnitude and the direction of the slope of the observed relationships depended upon the nature of the inhibitor. Hepatocytes from fasted rats synthesized glucose from lactate or fructose, and urea from ammonia, at rates which were generally linear functions of the magnitude of delta psi m, except in the presence of oligomycin or carboxyatractyloside. Linear relationships were also observed between delta psi m and the rate of formation of lactate in cells incubated with fructose and in hepatocytes from fed rats. The linear property of these force-flow relationships is taken as evidence for the operation of thermodynamic regulatory mechanisms within hepatocytes.  相似文献

Magnetic field-dependent recombination measurements together with magnetic field-dependent triplet lifetimes (Chidsey, E.D., Takiff, L., Goldstein, R.A. and Boxer, S.G. (1985) Proc. Natl. Acad. Sci USA 82, 6850–6854) yield a free energy change ΔG(P+H3P*) = 0.165 eV ±0.008 at 290 K. This does not depend on whether nuclear spin relaxation in the state 3P* is assumed to be fast or slow compared to the lifetime of this state. This value, being (almost) temperature independent, indicates ΔG(P+H3P*) ΔH(P+H3P*) and is consistent with ΔG(1P* − P+H) and ΔH(1P* − 3P*) from previous delayed fluorescence and phosphorescence data, implying ΔG ΔH for all combinations of these states.  相似文献

The relationship between the respiration rate and the magnitude of the electrochemical proton potential (ΔμH+) in rat liver mitochondria was investigated. (1) Under the active-state conditions, the action of inhibitors of either phosphorylation (oligomycin) or respiration (rotenone, malonate) on the respiration and ΔμH+ was measured. Both inhibitors diminished the respiration, whereas rotenone resulted in a decrease of ΔμH+, and oligomycin produced an increase of this potential. The effect of the inhibitors was much more pronounced on the respiration rate than on ΔμH+; for example, the excess of oligomycin produced a 90% inhibition of the respiration while ΔμH+ was changed only by 9%. (2) Under the resting-state conditions, small concentrations of the uncoupler stimulated the respiration while changing ΔμH+ to a relatively small extent. The uncoupler concentrations which doubled and tripled the respiration rate produced only 5 and 9% decrease of ΔμH+, respectively. (3) The present results enabled us to propose a model describing the interrelationship between respiration and ΔμH+.  相似文献

The distribution of glutamate and aspartate and the mitochondrial membrane potential (Δψ) were studied in isolated rat heart mitochondria and in the intact perfused rat heart. The diffusion potential imposed by the glutamate-aspartate exchange through mediation of the electrogenic glutamate-aspartate translocator attained a value close to the mitochondrial Δψ measured from the distribution of triphenylmethylphosphonium ion (TPMP+) both in isolated mitochondria and in intact myocardium. Distributions of the Δψ probe and metabolites were determined by subcellular fractionation of the heart muscle in a non-aqueous medium. The results indicate that the glutamate-aspartate translocator is in near equilibrium in the myocardium. The diffusion potential of the glutamate-aspartate exchange, and the mitochondrial/cytosolic difference in the redox potentials of the free NAD+/NADH pools are equal allowing for experimental error. These data obtained from intact tissue can therefore be interpreted as supporting the notion of the transmembrane uphill transport of reducing equivalent from the cytosolic free NAD+/NADH pool being driven by the malate-aspartate cycle energized by the mitochondrial Δψ.  相似文献

We have previously shown that crystals of calcium oxalate (COM) elicit a superoxide (O2) response from mitochondria. We have now investigated: (i) if other microparticles can elicit the same response, (ii) if processing of crystals is involved, and (iii) at what level of mitochondrial function oxalate acts. O2 was measured in digitonin-permeabilized MDCK cells by lucigenin (10 μM) chemiluminescence. [14C]-COM dissociation was examined with or without EDTA and employing alternative chelators. Whereas mitochondrial O2 in COM-treated cells was three- to fourfold enhanced compared to controls, other particulates (uric acid, zymosan, and latex beads) either did not increase O2 or were much less effective (hydroxyapatite +50%, p < 0.01), with all at 28 μg/cm2. Free oxalate (750 μM), at the level released from COM with EDTA (1 mM), increased O2 (+50%, p < 0.01). Omitting EDTA abrogated this signal, which was restored completely by EGTA and partially by ascorbate, but not by desferrioxamine or citrate. Omission of phosphate abrogated O2, implicating phosphate-dependent mitochondrial dicarboxylate transport. COM caused a time-related increase in the mitochondrial membrane potential (Δψm) measured using TMRM fluorescence and confocal microscopy. Application of COM to Fura 2-loaded cells induced rapid, large-amplitude cytosolic Ca2+ transients, which were inhibited by thapsigargin, indicating that COM induces release of Ca2+ from internal stores. Thus, COM-induced mitochondrial O2 requires the release of free oxalate and contributes to a synergistic response. Intracellular dissociation of COM and the mitochondrial dicarboxylate transporter are important in O2 production, which is probably regulated by Δψm.  相似文献

Neutrophils die rapidly via apoptosis and their survival is contingent upon rescue from constitutive programmed cell death by signals from the microenvironment. In these experiments, we investigated whether prevention of K+ efflux could affect the apoptotic machinery in human neutrophils. Disruption of the natural K+ electrochemical gradient suppressed neutrophil apoptosis (assessed by annexin V binding, nuclear DNA content and nucleosomal DNA fragmentation) and prolonged cell survival within 24–48 h of culture. High extracellular K+ (10–100 mM) did not activate extracellular signal-regulated kinase (ERK) and Akt, nor affected phosphorylation of p38 MAPK associated with constitutive apoptosis. Consistently, pharmacological blockade of ERK kinase or phosphatidylinositol 3-kinase (PI 3-kinase) did not affect the anti-apoptotic action of KCl. Inhibition of K+ efflux effectively reduced, though never completely inhibited, decreases in mitochondrial transmembrane potential (ΔΨm) that preceded development of apoptotic morphology. Changes in ΔΨm resulted in attenuation of cytochrome c release from mitochondria into the cytosol and decreases in caspase-3 activity. Culture of neutrophils in medium containing 80 mM KCl with the pan-caspase inhibitor Z-VAD-FMK resulted in slightly greater suppression of apoptosis than KCl alone. High extracellular KCl also attenuated translocation of apoptosis-inducing factor (AIF) and endonuclease G (EndoG) from mitochondria to nuclei. The DNase inhibitor, aurintricarboxylic acid (ATA) partially inhibited nucleosomal DNA fragmentation, and the effects of ATA and 80 mM KCl were not additive. These results show that prevention of K+ efflux promotes neutrophil survival by suppressing apoptosis through preventing mitochondrial dysfunction and release of the pro-apoptotic proteins cytochrome c, AIF and EndoG independent of ERK, PI 3-kinase and p38 MAPK. Thus, K+ released locally from damaged cells may function as a survival signal for neutrophils.  相似文献

Density functional theory (DFT) computations at the B3LYP/Lanl2DZ level were used to elucidate the oxygen atom transfer (OAT) and coupled electron proton transfer (CEPT) reaction steps involved in the biomimetic catalytic cycle performed by polymer-supported MoVIO2(NN′)2 complexes [NN′ = phenyl-(pyrrolato-2-ylmethylene)-amine] with water as oxygen source, trimethyl-phosphane as oxygen acceptor and one-electron oxidising agents. The DFT method employed has been validated against experimental data [X-ray crystal structures of a NN′ ligand and a MoVIO2(NN′)2 complex as well as kinetic data]. The rate-limiting step in the forward-OAT from [MoVIO2] to PMe3 is the attack of PMe3 at an oxo ligand with ΔG (298 K) = 64.6 kJ mol−1. Dissociation of the product OPMe3 is facile with ΔG (298 K) = 26.3 kJ mol−1 giving a mono-oxo [MoIVO] complex which fills its coordination sphere with a further PMe3 substrate with ΔG (298 K) = 39.2 kJ mol−1. One-electron oxidation to a Mo(V) phosphane complex precedes the coordination of water/hydroxide. Additionally, the comproportionation of [MoVIO2] and [MoIVO] to dinuclear oxo-bridged [OMoV–O–MoVO] species has been calculated as the thermodynamic sink in this system and the back-OAT from dmso to mono-oxo [MoIVO] to give [MoVIO2] has been shown to involve an equilibrium between stereoisomeric [MoVIO2] complexes with an activation barrier of ΔG (298 K) = 113.1 kJ mol−1.  相似文献

Cp#2Yb (Cp#=C5H4(CH2)2NMe2) has been obtained by reaction of YbI2(THF)2 with 2 equiv. of C5H4(CH2CH2NMe2)K in THF. The X-ray structure analysis shows a bent structure with intramolecular coordination of both nitrogen atoms to ytterbium. The reaction of C60-fullerene with Cp#2Yb leads to the formation of the fullerenide derivative [Cp#2Yb]2C60, which shows an ESR signal in the solid state and in THF solution at room temperature (solid: ΔH = 50 G, G = 1.9992; solution: ΔH = 10 G, G = 2.0001) and a magnetic moment of 3.6 BM. The lutetium fullerenides CpLu(C60)(DME) (3) and Cp*Lu(C60)(DME)(C6H5CH3) (4), (Cp = η5−C5H5, Cp* = η5−C5Me5), were obtained by reaction of C60 with CpLu(C10H8) (DME) and Cp*Lu(C10H8) (DME) in toluene. Both complexes are paramagnetic (μeff = 1.4 and 0.9 BM) and exhibit temperature-dependent ESR signals (293 K: g = 1.992 and 2.0002 respectively).  相似文献

Steady-state current-voltage relationships (SSCVRs) of the plasma membrane of human T-lymphocytes were studied at the physiological temperature of 37°C by using the whole-cell patch-clamp technique. SSCVRs displayed a characteristic N-like shape with a negative resistance region (NRR) in a voltage range of −45 to −35 mV. The majority of cells assayed revealed SSCVR patterns crossing the V-axis at three points (in mV): V1 = −55 to −45, V2 = −40 to −35, V3 = −30 to −10. SSCVRs of T-cells activated by phytohaemagglutinin (48–96 h) also displayed NRR, but crossed the V-axis at one point only (V1 = −55 to −60 mV). It implies the possibility of two stable levels of membrane potential (V1 and V3) for the resting T-cells, but only one (V1) for activated T-cells. These data thus account for the triggering property of T-cell membrane potential previously reported. The NRR can be explained on the basis of the Hodgkin-Huxley type n4j model of K+ channel kinetics. According to the model the possibility for a membrane to have on or two stable levels of membrane potential depends on the ratio of selective K+ conductance to non-selective leaky conductance (Gk/Gleak). The steady-state level of K+ conductance in resting T-lymphocytes proved to be sensitive to Ca2+. Buffering Ca2+ ions from either external or internal solution resulted in an appreciable increase in K+ conductance. The possibility for membrane potential have two stable levels of membrane potential in connection with the Ca2+ dependence of K+ conductance was supposed to be important for Ca2+-signalling during T-cell activation.  相似文献

σ-Methyl-(η5-indenyl) chromium tricarbonyl (III) rearranges quantitatively into η6-1-endo-methylindene) chromium tricarbonyl (IV) in C6D6 solution at 30–60°C. Methyl group attachment to the positions 2 or 3 of indenyl ligand in (III) has no influence on the activation parameters of this ricochet inter-ring haptotropic rearrangement (ΔG#=23.6 kcal mol−1; ΔH#=18.9±0.2 kcal mol−1; ΔS#=−18.6±0.2 cal K−1 mol−1). (IV) undergoes further irreversible isomerization at 60–120° into (ν6-3-methylindene) chromium tricarbonyl (V) with a higher activation barrier (ΔG#=28.5±0.1 kcal mol−1) via two consecutive [1,5]-sigmatropic hydrogen shifts. The mechanisms of both rearrangements have been studied in detail using density functional theory (DFT) calculations with extended basis sets. Calculations show that the rearrangement (III) → (IV) proceeds in two steps. Methyl group migration from chromium into position 1 of the indenyl ligand is the rate-determining step leading to the formation of the 16-electron intermediate (VII). The calculated activation barrier (Ea=19.6 kcal mol−1) is in good agreement with the experimental one. Further rearrangement (VII) → (V) proceeds via a trimethylenemethane-type transition state (XVIII) with an activation barrier 11.8 kcal mol−1. The coordination of the chromium tricarbonyl group at the six-membered ring has only minor influence on the kinetic parameters of the hydrogen [1,5]-sigmatropic shift in indene.  相似文献

The equilibria and dynamics of the disorder-to-order transition of the anionic polysaccharide iota-carrageenan have been studied in the presence of tetramethyl-ammonium salts. By the use of a stopped-flow polarimeter, the rate equation and temperature dependence of the observed forward rate-constant were found to accord with a co-operative dimerisation process. Activation parameters for helix nucleation were shown to be independent of the anion for solutions containing tetramethylammonium chloride and bromide, i.e., ΔH = 1 ±3 kJ.mol−1, ΔS = −178 ±10 J.mol−1.K−1, ΔG298K = 54 ±2 kJ.mol−1, and knuc,298K = 1880 ±80 dm3.mol−1.s−1. The temperature dependence of optical rotation was also shown to be independent of the anion present.  相似文献

Complexes of type A4[VO(tart)]2·nH2O, where A = Rb or Cs and tart =d,l-tartrate(4−) (n = 2) or d,d-tartrate(4−) (n = 2 for Rb and n = 3 for Cs), were prepared from an aqueous mixture of V2O5, AOH and H4tart. These complexes were studied by single-crystal X-ray diffraction methods: Rb4[VO(d,l-tart)]2·2H2O, space group P1 with a = 8.156(1),b = 8.246(1),c = 8.719(1)Å, = 66.09(1)°, β = 65.07(1)°, γ = 82.40(1)°,Z = 2, 1917 observed reflections, and final Rw = 0.035; Cs4[VO(d,l-tart)]2·2H2O, space group P21/c with a = 9.350(1),b = 13.728(2),c = 8.479(1)Å, β = 106.77(1)°,Z = 4, 2235 observed reflections, and final Rw = 0.054; Rb4[VO(d,d-tart)]2·2H2O, space group P4122 with a = 8.072(1),c = 32.006(3)Å,Z = 8, 1014 observed reflections and final Rw = 0.038; Cs4[VO(d,d-tart)]2·3H2O, space group P122 with a = 8.184(1),c = 33.680(5)Å,Z = 8, 1310 observed reflections, and final Rw = 0.063. Bulk magnetic susceptibility data (1.5–300 K) for these compounds and A4[VOl,l-tart)]2·nH2O (A = Rb, Cs) were obtained on polycrystalline samples. These data were analyzed in terms of a Van Vleck exchange coupled S = 1/2 model which was modified to include an interdimer exchange parameters Θ. Analysis of the low-temperature (1.5–20 K) susceptibility data gave 2J = +1.30 cm−1 and Θ = −1.86 K for Rb4[VO(d,l-tart)]2·2H2O, 2J = +1.16 cm−1 and Θ = −1.69 K for Cs4[VO(d,l-tart)]2·2H2O, 2J = +1.90 cm−1 and Θ = −0.82 K for Rb4[VO(d,d-tart)]2·2H2O, 2J = +2.04 cm−1 and Θ = −0.80 K for Rb4[VO(l,l-tart)]2·2H2O, 2J = +1.52 cm−1 and Θ = −0.25 K for Cs4[VO(d,d-tart)]2·3H2O, and 2J = +1.64 cm−1 and Θ = −0.31 K for Cs4[VO(l,l-tart)]2·3H2O. These results suggest the magnitudes of intradimer (ferromagnetic and interdimer (antiferromagnetic) exchange interactions are similar in these complexes, as observed for the analogous Na salts.  相似文献

The H+-ATPase from chloroplasts, CF0F1, was isolated, purified and reconstituted into asolectin liposomes. The enzyme was brought either into the oxidized state or into the reduced state, and the rate of ATP synthesis was measured after energisation of the proteoliposomes with an acid—base transition ΔpH (pHin = 5.0, pHout = 8.5) and a K+/valinomycin diffusion potential, Δφ (K+in = 0.6 mM, K+out = 60 mM). A rate of 250 s−1 was observed with the reduced enzyme (85 s−1 in the absence of Δφ). A rate of 50 s−1 was observed with the oxidized enzyme under the same conditions (15 s−1 in the absence of Δφ). The reconstituted enzyme contained 2 ATPbound per CF0F1 and 1 ADPbound per CF0F1. Upon energisation the enzyme was activated and 0.9 ADP per CF0F1, was released. Binding of ADP to the active reduced enzyme was observed under different conditions. In the absence of phosphate the rate constant for ADP binding was 105 M−1·s−1 under energized and de-energized conditions. In the presence of phosphate the rate of ADP binding drastically increased under energized conditions, and strongly decreased under de-energized conditions.  相似文献

An analysis of the complete strain field within Flexercell membranes   总被引:4,自引:0,他引:4  
The purpose of the current investigation was to use finite element analysis to quantify the complete strain field for the membranes of the FlexercellTM apparatus, a device extensively used to study the effects of mechanical loading on cultured cells. Four vacuum pressure simulations were run for the membrane for both the uniaxial and biaxial loading post, yielding the distribution of longitudinal (Exx) and transverse (Eyy) strain for the uniaxial post, and the radial (Err) and the circumferential (Eθθ) strain for the biaxial post. The discrete values of each strain were evaluated at the center of the loading post as well as the region off of the post. Experimental measurements were made for both types of loading posts in order to validate our simulations. The biaxial post simulation was found to provide a central circular region of equal and constant Err and Eθθ in the membrane on the post. Likewise, the uniaxial post simulation provided a definitive region of constant Exx for a central rectangular region on the post. For the uniaxial simulation, the region on the post resulted in small compressive Eyy, while the region off the post resulted in tensile Eyy. The biaxial simulation resulted in large tensile Err and Eθθ on the post, while the region off the post resulted in large Err and smaller Eθθ. Our simulations were reasonably consistent with the experimental measurements made for both types of loading posts. We believe that the results of this study will allow scientists to more accurately describe the response of cells to known strains on all portions of the membrane, thus increasing the range of known strain regions for investigation in the FlexercellTM apparatus.  相似文献

In this paper a number of experiments with the purple bacteria Rhodospirillum rubrum and Rhodopseudomonas capsulata is described in which the total fluorescence yield and/or the total fraction of reaction centers closed after a picosecond laser pulse were measured as a function of the pulse intensity. The conditions were such that the reaction centers were either all in the open or all in the closed state before the pulse arrived. These experiments are analysed using the theoretical formalism discussed in the preceding paper (Den Hollander, W.T.F., Bakker J.G.C., and Van Grondelle, R., Biochim. Biophys. Acta 725, 492–507). From the experimental results the number of connected photosynthetic units, λ, the rate of energy transfer between neighboring antenna molecules, kh, and the rate of trapping by an open reaction center, kot, can be estimated. For R. rubrum it is found that λ = 14−17, kh = (1−2)·1012 s−1 and kot = (4−6)·1011 s−1, for Rps. capsulata λ ≈ 30, kh ≈ 4·1011 s−1 and kot ≈ 3·1011 s−1. The findings are discussed in terms of current models for the structure of the antenna and the kinetic properties of the decay processes occurring in these purple bacteria.  相似文献

Respiration, membrane potential generation and motility of the marine alkalotolerant Vibrio alginolyticus were studied. Subbacterial vesicles competent in NADH oxidation and Δψ generation were obtained. The rate of NADH oxidation by the vesicles was stimulated by Na+ in a fashion specifically sensitive to submicromolar HQNO (2-heptyl-4-hydroxyquinoline N-oxide) concentrations. The same amounts of HQNO completely suppressed the Δψ generation. Δψ was also inhibited by cyanide, gramicidin D and by CCCP + monensin. CCCP (carbonyl cyanide m-chlorophenylhydrazone) added without monensin exerted a much weaker effect on Δψ. Na+ was required to couple NADH oxidation with Δψ generation. These findings are in agreement with the data of Tokuda and Unemoto on Na+-motive NADH oxidase in V. alginolyticus. Motility of V. alginolyticus cells was shown to be (i) Na+-dependent, (ii) sensitive to CCCP + monensin combination, whereas CCCP and monensin, added separately, failed to paralyze the cells, (iii) sensitive to combined treatment by HQNO, cyanide or anaerobiosis and arsenate, whereas inhibition of respiration without arsenate resulted only in a partial suppression of motility. Artificially imposed ΔpNa, i.e., addition of NaCl to the K+-loaded cells paralyzed by HQNO + arsenate, was shown to initiate motility which persisted for several minutes. Monensin completely abolished the NaCl effect. Under the same conditions, respiration-supported motility was only slightly lowered by monensin. The artificially-imposed ΔpH, i.e., acidification of the medium from pH 8.6 to 6.5 failed to activate motility. It is concluded that Δ Na+ produced by (i) the respiratory chain and (ii) an arsenate-sensitive anaerobic mechanism (presumably by glycolysis + Na+ ATPase) can be consumed by an Na+-motor responsible for motility of V. alginolyticus.  相似文献

The effect ot Cu2+ and Ca2+ ions, on the ultraviolet differential (UVD) spectra of single-stranded poly I was studied and the coordination (Δεb) and conformation (Δεc) conponents of the spectra calculated The comparison of Δεb and the UVD spectrum of protonated IMP leads to the conclusion that N(7) ot inosine-5'-monophosphate (IMP) is a coordinating site tor Ca2+ and Cu2+ ions on the polymer bases. The binding ot Ca2+ and Cu2+ ions causes differently directed displacements of the four absorption bands of poly I, which are observed in the wavenumber range (50-34) × 103 cm−1 The calculation of concentration dependencies tor the association constants (K“) ot Ca2+ and Cu2+ ions binding to poly I bases shows that the binding is cooperative The K“ values for the poly I + Ca2+ complex are two orders of magnitude lower than those for the poly 1 + Cu2+ complex At low ion concentrations, binding to the poly I phosphates predominates and increases the degree of the polynucleotide helicity. At higher concentrations the spectra are mainly affected by the ion binding to bases, which results in melting of the helical parts of poly I At Ca2+ concentrations exceeding 10−3 M light-scattering aggregates are formed. Rates of stepwise anation of cis-Cr(ox)2(H2O2) with SCN/N3, Cr(acac)2(H2O)2+ with SCN and Cr(atda)(H2O)2 with SCN have been investigated in weakly acidic aqueous solutions. Rate constants, kI and kII for the two steps in each system, are composite as kx = kx0+kxX[X] (x = I, II; X = SCN, N3). These rate constants have been evaluated also as the corresponding ΔH and ΔS values. The results obtained and the plausible Id mechanism seem to suggest Cr---OOC bond dissociation (hence a strongly negative ΔS) generating the transition state in each system with outer-sphere association forming the precursor complex in the X dependent paths.  相似文献

The action of rotameric probes introduced either in position 7 or 8 in the sequence of substance P (SP) was investigated, i.e. -tetrahydroisoquinoleic acid (Tic), -fluorenylglycine (Flg), -diphenylalanine (Dip), the diastereoisomers of -1-indanylglycine (Ing) and -benz[ƒ]indanylglycine (Bfi), the Z- and E-isomers of dehydrophenylalanine and dehydronaphthylalanine (ΔZPhe, ΔEPhe, ΔZNal, ΔENal) and (Dmp). The aim of this study was the topographical characterization of the binding subsites of human NK-1 receptor expressed in CHO cells, especially the S7 and S8 subsites, corresponding to residues Phe7 and Phe8 of substance P. According to the binding potencies of these substituted-SP analogues, the S7 binding subsite is smaller than the S8 subsite: the S7 subsite accepts only one aromatic nucleus, while the S8 can accommodate three coplanar nuclei altogether. These findings are compatible with the idea that the S8 binding subsite may reside in the extracellular loops of the hNK-1 receptor. NK-1 agonists bind to human NK-1 receptor and activate the production of both inositol phosphates and cyclic AMP. As already quoted for septide, [pGlu6, Pro9]SP(6–11), discrepancies are observed between affinity (Ki) and activity (EC50) values for IPs production. While a weak correlation between Ki and EC50 values for IPs production could be found (r = 0.70), an excellent correlation could be demonstrated between their affinities (Ki) and their potencies (EC50) for cAMP production (r = 0.97). The high potency (EC50) observed for ‘septide-like’ molecules on PI hydrolysis, compared to their affinity is not an artefact related to the high level of NK-1 receptors expressed on CHO cells since a good correlation was found between EC50 values obtained for PI hydrolysis and those measured for spasmogenic activity in guinea pig ileum bioassay (r = 0.94).

According to the binding potencies of constrained analogues of phenylalanine, the S7 binding subsite of human NK-1 receptor is small, whereas the S8, which can accommodate three coplanar nuclei, might probably reside in the extracellular loop. The discrepancies observed between affinity (Ki) and activity (EC50) values for IPs production are not an artefact of CHO cells since a good correlation was found between EC50 for PI hydrolysis and those measured in guinea pig ileum bioassay.  相似文献

The role of Na+ in Vibrio alginolyticus oxidative phosphorylation has been studied. It has been found that the addition of a respiratory substrate, lactate, to bacterial cells exhausted in endogenous pools of substrates and ATP has a strong stimulating effect on oxygen consumption and ATP synthesis. Phosphorylation is found to be sensitive to anaerobiosis as well as to HQNO, an agent inhibiting the Na+-motive respiratory chain of V. alginolyticus. Na+ loaded cells incubated in a K+ or Li+ medium fail to synthesize ATP in response to lactate addition. The addition of Na+ at a concentration comparable to that inside the cell is shown to abolish the inhibiting effect of the high intracellular Na+ level. Neither lactate oxidation nor Δω generation coupled with this oxidation is increased by external Na+ in the Na+-loaded cells. It is concluded that oxidative ATP synthesis in V. alginolyticus cells is inhibited by the artificially imposed reverse ΔPNa, i.e., [Na+]in > [Na+]out. Oxidative phosphorylation is resistant to a protonophorous uncoupler (0.1 mM CCCP) in the K+-loaded cells incubated in a high Na+ medium, i.e., when ΔpNa of the proper direction ([Na+]in < [Na+]out) is present. The addition of monensin in the presence of CCCP completely arrests the ATP synthesis. Monensin without CCCP is ineffective. Oxidative phosphorylation in the same cells incubated in a high K+ medium (ΔpNa is low) is decreased by CCCP even without monensin. Artificial formation of ΔpNa by adding 0.25 M NaCl to the K+-loaded cells (Na+ pulse) results in a temporary increase in the ATP level which spontaneously decreases again within a few minutes. Na+ pulse-induced ATP synthesis is completely abolished by monensin and is resistant to CCCP, valinomycin and HQNO. 0.05 M NaCl increases the ATP level only slightly. Thus, V. alginolyticus cells at alkaline pH represent the first example of an oxidative phosphorylation system which uses Na+ instead of H+ as the coupling ion.  相似文献
