In two experiments of 3 × 3 Latin square with growing pigs, the effect was investigated of supplementation of 5 % or 10 % (2.5 % vs. 5 % DM) polyol mixture and 2.5 % or 5 % of xylitol on digestibility of diet, N-balance, blood clinical-chemical parameters and insulin level in serum. Apparent digestibility of crude protein was lower for the diet with 10 % polyol mixture compared to the control. Sugar alcohols were not found in faeces. Arabinitol, mannitol and rhamnitol were excreted in the urine 25–67 %. Little sorbitol and xylitol were found in urine on diets with polyol mixture 5–10 %. On xylitol diets the pigs did not excrete xylitol in urine. Plasma glucose rose in pigs fed xylitol. Blood total protein and albumin decreased in pigs fed polyol mixture. ALAT-activities were higher for xylitol diets than for the controls. Serum insulin tended to increase in pigs fed polyol mixture 10 % one hour after feeding, and in xylitol feeding two hours after feeding; these values were higher with increasing xylitol inclusion in diet.  相似文献

Dietary supplementations with L-glutamine and/or nucleotides were screened for their effects on intestinal mucosa in 16 female weaning piglets. The animals were transported to the university's facilities 24 hours after weaning. They were grouped four to a pen in controlled environmental conditions and fed one of the following four diets for 28 days: control diet (C); C+0.5% L-glutamine (G); C+0.05% "nucleotides" (N); and C+0.5 % L-glutamine+0.05% "nucleotides" (GN). Individual body weights and feed intake per group were recorded at the beginning and the end of the study as well as weekly during it. There were no significant performance differences among the groups. After 28 days the animals were slaughtered and the distal ileum and liver were examined histologically. Anti-proliferating cell nuclear antigen (PCNA) as well as anti-human macrophage immunostaining, and a modified TdT-mediated dUTP nick-end labeling technique (TUNEL) were performed, and intraepithelial lymphocyte percentage was evaluated to assess morpho-functional aspects of the ileum. Histometry was performed by assessing cell indices and counts of immuno-reactive structures. Feeding G and/or N resulted in an increase in villi (V) height, crypt (C) depth, and a decrease in V:C ratio (P<0.01). In addition, feeding G and/or N resulted in an increase in mitotic mucosal cells (M), and a decrease in apoptotic mucosal cells (A), thus decreasing the A:M index (P<0.01). The percentages of mucosal macrophages were greater in G and/or N groups (P<0.001) than in control piglets, and similarly among the groups the percentages of intraepithelial lymphocytes varied (P<0.01). Our data showed that the diet supplementation with G and/or N had positive effects on some morpho-functional characteristics of piglet ileal mucosa. These ameliorative effects may potentially be linked to a good responsiveness of piglets to a stressful period, like a precocious weaning is in this species.  相似文献

Among substances intended to replace growth promoting antibiotics in pig nutrition, non-digestible oligosaccharides or polysaccharides could be potential alternative compounds. Therefore, the influence of β-1,3-1,6 glucans on bacteriological, biochemical and morphological aspects of the small intestine in weaned piglets was investigated. As sources of β-glucans, Lentinan (extract of Lentinus edodes mycelium) or dried L. edodes mycelium were added to the diet. Four homogenous groups of 5 newly weaned piglets (4 weeks of age) received one of four diets: control diet (C), C supplemented with Avilamycin (50?mg/kg, positive control), C supplemented with 0.1% of Lentinan and C supplemented with 5% of dried L. edodes mycelium powder. A first group of 10 piglets was euthanized after 11 days and the remaining 10 on day 12 of the experiment. The gastrointestinal tract was divided in segments and samples taken from digesta (stomach, proximal and distal jejunum, caecum), mucosal scrapings (jejunum) and ring shaped tissue samples (1?cm) of proximal and distal jejunum. Bacterial counts were made with digesta and mucosal samples, and short-chain fatty acids (SCFA), lactic acid and ammonia concentrations were determined. Tissue samples of both jejunal sites were embedded in paraffin wax for morphometrical (villus length, crypt depth) and histological observations (numbers of intraepithelial lymphocytes (IEL), goblet cells, apoptotic enterocytes on villi, mitotic cells in crypts). Only the diet containing 5% of dried L. edodes consistently resulted in lower viable counts (ca. 1?–?2 log10 CFU) of total bacteria, E. coli, streptococci and lactic acid bacteria, and luminal and mucosal effects agreed very well. With this diet, acetate and butyrate concentrations in the distal jejunum were doubled, which is favourable in view of the trophic effect on enterocytes and colonocytes. Villus length (V) was increased with both diets containing β-glucans while crypt depth (C) was not altered, but V/C was higher. IEL counts were decreased by both diets although bacterial numbers, which is only one parameter of bacterial load, were only diminished with the L. edodes feed. The three supplemented feeds lowered the number of apoptotic enterocytes on the villi, but these numbers were very low (control diet?:?44 cells per 100 villi), making clear interpretation difficult. The mitotic index was slightly lower with the L. edodes feed, although not statistically significant. Decreased viable counts observed with the latter diet is a favourable effect as it is accepted that a lower bacterial load causes lower turnover rates of the intestinal epithelial cells, while there is also less competition for specific substrates. A higher V/C ratio, a smaller number of IEL in the epithelium and a lower apoptotic index also indicate slower turnover rate of the mucosa when Lentinan and L. edodes diets were fed. The inconsistent effects observed with Lentinan were probably due to the low amount added to the diet. It should be taken into account that the influence of L. edodes mycelium powder was more likely due to the presence of antibacterial compounds (eg. lenthionine, lentinamycin, terpenoids, polyphenols), rather than to an immunostimulating action of β-glucans with increased release of IgA onto the mucosa surface.  相似文献

This study investigated the gender differences in the kidney function of magnesium (Mg)-deficient rats. Male and female rats were fed a control diet or a Mg-deficient diet for 21 d. Mg-deficient diet had no significant effect on kidney calcium (Ca) or phosphorus (P) concentration in male rats, while Ca and P concentrations in female rats were significantly higher in Mg-deficient rats than in the control rats. With regard to indicators of kidney function, no significant differences in creatinine clearance and serum urea nitrogen concentration were observed among the groups. Serum albumin concentrations were significantly lower in rats fed the Mg-deficient diet than in rats fed the control diet. In both sexes, urinary albumin excretion was significantly higher in rats fed the Mg-deficient diet than in rats fed the control diet. Gender differences had no significant influence on creatinine clearance, serum urea nitrogen concentration, serum albumin concentration and urinary albumin excretion. These results suggest that gender differences have no effect on kidney function in Mg-deficient rats under the condition used.  相似文献

Upper jejunum and terminal ileum were examined in specified-pathogen-free (SPF), conventional and conventional after SPF rearing (ex-SPF) rats. The effect of 2 differential diets on the last 2 groups was examined. Ex-SPF rats had taller villi and deeper crypts than SPF rats, but similar crypt to villus ratios and cell production rates. Ex-SPF rats had similar crypt depth and jejunal villus height to conventional rats on the same diet, but taller ileal villi and a lower cell production rate. Even after 6-8 weeks, in a conventional environment, ex-SPF rat intestine was still not identical with conventional rat intestine. Diet had a significant effect on mucosal architecture, and a smaller effect on cell production rate. It is concluded that diet, microbiological status of colony of origin, and environment after weaning, can all affect mucosal architecture and epithelial cell production, and should be properly controlled in experimental studies.  相似文献

We investigated the effect of chronic alcohol ingestion on buccal mucosal ulcer healing by analyzing the interplay between mucosal expression of tumor necrosis factor-alpha (TNF-alpha), endothelin-1 (ET-1), and interleukin-4 (IL-4). Chronic ulceration was induced in rats maintained for 5 weeks on alcohol-containing or control liquid diet. In both groups, the ulcer onset was characterized by a massive increase (6.5-8.9-fold) in TNF-alpha and ET-1 (1.6-4.0-fold), and a decrease (1.4-1.5-fold) in IL-4. However, the group on the alcohol diet exhibited a 38.3% higher mucosal expression of TNF-alpha, a 26. 2% higher ET-1 level, and a 6.5% lower content of IL-4. While in both groups the ulcer healing was accompanied by an increase in buccal mucosal expression of IL-4, and a decline in ET-1 and TNF-alpha, the changes were significantly slower in the alcohol diet group and manifested by a 4 day delay in ulcer healing. The results suggest that chronic alcohol ingestion exerts detrimental effects on the buccal mucosal IL-4 expression, causing dysregulation of ET-1 production, induction of TNF-alpha, and triggering apoptotic events that delay the mucosal repair.  相似文献

The effects of age, breed, and diet on fecal chemistry, enzyme activity, and bacterial populations of dogs were studied. Eighteen dogs from two age groups (young: 2.5 +/- 0.5 years, old: 10.9 +/-0.7 years) and three different breeds (German shepherds, miniature schnauzers, and English setters) were rotated through a Latin Square design such that every dog was fed each of the diets. The test diets included a low-fiber (control) diet and a 10% fiber diet which contained 5% soybean hulls and 5% beet pulp. Inclusion of 10% fiber in the diet decreased the fecal concentration of ammonia, sulfide, and indole. Fiber inclusion significantly increased acetic, propionic, and butyric acid concentrations, while fecal pH decreased by 0.4 units. Fresh fecal samples were plated on selected aerobic and anaerobic culture media and DNA extracted for denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S ribosomal DNA fragments. Plate counts showed significant effects of breed (p < or = 0.05) and age (p < or = 0.01) on selected aerobic and anaerobic bacterial counts, while no significant effect of diet was found. Analysis of PCR-DGGE banding patterns showed there was a tendency for individual dogs to cluster together according to age (young or old dogs) and also for size (large or small dogs). However, the outstanding conclusion obtained from the DGGE analysis of fecal bacterial profiles was that individual dogs had their own characteristic banding pattern which was unique and stable. The relative stability and individuality of the patterns indicates that each individual harbored a characteristic fecal bacterial community which was independent of diet.  相似文献

Abstract. A group of thirty-two Merino sheep infested with itch mites (Psorergates ovis) and fed a maintenance diet which imposed moderate nutritional stress had a significantly higher mite population, significantly more skin scurf, and significantly more fleece damage or derangement (P < 0.05) than a second group of thirty-two infested sheep fed a diet designed for unrestricted body weight gain and wool growth. Histologically there were no significant differences between the groups in the numbers of mast cells, neutrophils or eosinophils observed in skin sections, but sheep that had high mite counts (>10 per 200 cm2 of skin area) in both groups, had more dermal mast cells than sheep with fewer mites irrespective of the plane of nutrition. Skin thickness and greasy fleece weight in die group maintained on the low plane of nutrition were significantly less (P < 0.05) than in die well-nourished group, reflecting the difference in protein and energy content of the two diets. Within the nutritionally stressed group, the sheep with low mite counts had a significantly lower (P < 0.05) greasy fleece weight and a shorter mean staple length than the sheep with high mite counts. There was no significant difference in greasy fleece weight between sheep with low or high mite counts in the group fed on the high plane of nutrition.  相似文献

This study was designed to evaluate the effects of algal and yeast β-glucans on the porcine gastrointestinal microbiota, specifically the community of Lactobacillus, Bifidobacterium and coliforms. A total of 48 pigs were fed four diets over a 28-day period to determine the effect that each had on these communities. The control diet consisted of wheat and soya bean meal. The remaining three diets contained wheat and soya bean meal supplemented with β-glucan at 250 g/tonne from Laminaria digitata, Laminaria hyperborea or Saccharomyces cerevisiae. Faecal samples were collected from animals before feeding each diet and after the feeding period. The animals were slaughtered the following day and samples were collected from the stomach, ileum, caecum, proximal colon and distal colon. Alterations in Lactobacillus in the gastrointestinal tract (GIT) were analysed using denaturing gradient gel electrophoresis (DGGE) profiles generated by group-specific 16S rRNA gene PCR amplicons. Plate count analysis was also performed to quantify total coliforms. DGGE profiles indicated that all β-glucan diets provoked the emergence of a richer community of Lactobacillus. The richest community of lactobacilli emerged after feeding L. digitata (LD β-glucan). Plate count analysis revealed that the L. hyperborea (LH β-glucan) diet had a statistically significant effect on the coliform counts in the proximal colon in comparison with the control diet. β-glucan from L. digitata and S. cerevisiae also generally reduced coliforms but to a lesser extent. Nevertheless, the β-glucan diets did not significantly reduce levels of Lactobacillus or Bifidobacterium. DGGE analysis of GIT samples indicated that the three β-glucan diets generally promoted the establishment of a more varied range of Lactobacillus species in the caecum, proximal and distal colon. The LH β-glucan had the most profound reducing effect on coliform counts when compared with the control diet and diets supplemented with L. digitata and S. cerevisiae β-glucans.  相似文献

We investigated the effects of n-3 polyunsaturated fatty acids (PUFAs) on non-steroidal anti-inflammatory drug (NSAID)-induced changes in microcirculation and eicosanoid production in the gastrointestinal mucosa. We measured gastric mucosal blood flow using laser Doppler flowmetry, assessed the fatty acid composition in the mucosal phospholipids, and quantified the production of prostaglandin E2 (PGE2), leukotriene B4, and leukotriene C4 (LTB4 and C4) from the mucosa with the stimulation of calcium ionophore 20 min after an injection of indomethacin or vehicle in rats fed a diet containing different compositions of alpha-linolenic acid. Four weeks after the initiation of the test diet the arachidonic acid level in gastric mucosal phospholipids was significantly lower in the perilla group than in the other three groups. Conversely, alpha-linolenic acid and eicosapentaenoic acid (EPA) were significantly higher in the perilla group than in the other three groups. The percent of gastric mucosal blood flow in the three groups administered indomethacin were significantly lower than that in the control group injected with vehicle alone. The percent of gastric mucosal blood flow in the perilla group was significantly higher than that in the corn group. LTB4 and LTC4 production from the gastric mucosa in the soybean and corn groups were significantly higher than those in the control group, and the LTC4 production in the perilla group was significantly lower than that in the corn group. There were no significant differences in PGE2 production among the four groups. Our results suggest that alpha-linolenic acid affectively suppressed the indomethacin-induced decreases in gastric mucosal blood flow by increasing EPA and decreasing the levels of arachidonic acid and LTC4 in the gastric mucosa.  相似文献

We studied the effects of xylitol on biofilms containing xylitol-resistant (Xr) and xylitol-sensitive (Xs) Streptococcus mutans, Actinomyces naeslundii and S. sanguinis. The biofilms were grown for 8 and 24 h on hydroxyapatite discs. The viable microorganisms were determined by plate culturing techniques and fluorescence in situ hybridization (FISH) was performed using a S. mutans-specific probe. Extracellular cell-bound polysaccharides (EPS) were determined by spectrofluorometry from single-species S. mutans biofilms. In the presence of 5 % xylitol, the counts of the Xs S. mutans decreased tenfold in the young (8 h) biofilm (p < 0.05) but no effect was seen in the mature (24 h) biofilm. No decrease was observed for the Xr strains, and FISH confirmed these results. No differences were detected in the EPS production of the Xs S. mutans grown with or without xylitol, nor between Xr and Xs S. mutans strains. Thus, it seems that xylitol did not affect the EPS synthesis of the S. mutans strains. Since the Xr S. mutans strains, not inhibited by xylitol, showed no xylitol-induced decrease in the biofilms, we conclude that growth inhibition could be responsible for the decrease of the counts of the Xs S. mutans strains in the clinically relevant young biofilms.  相似文献

The aim was to determine the effect of orally administered ovine serum immunoglobulin (Ig) on growth performance, organ weight, gut morphology and mucin production in the Salmonella enteritidis--gavaged growing rat. Four groups consisted of non-gavaged rats fed a casein-based control basal diet (BD) and three groups of rats gavaged with 1×10(7) CFU S. enteritidis and fed a casein-based diet, a diet containing freeze-dried ovine Ig (FDOI) or a casein-based diet containing inactivated ovine Ig (IOI). The rats were randomly allocated to one of the four groups (n=15/group) and received their respective diets for an 18-day experimental study. Gavaging took place on day 15. Average daily gain and body gain : feed ratio (post-gavage, 3 days) were significantly (P<0.05) higher for the Salmonella-challenged rats fed the FDOI diet compared to those fed the BD and IOI diets. At the end of the study, the small intestine and colon were significantly (P<0.05) heavier for the gavaged rats fed the FDOI diet compared to the gavaged rats fed either the BD or IOI diet. Moreover, the relative weights of the caecum, liver and spleen of the gavaged rats fed the BD or IOI diet were significantly (P<0.05) heavier compared to the gavaged rats fed the FDOI diet. Generally, the gavaged rats fed the FDOI diet had significantly (P<0.05) higher goblet cell counts and luminal mucin protein contents than the gavaged rats fed either the BD or IOI diet and had a more functional gut morphology. Overall, the FDOI fraction prevented the acute effects of S. enteritidis.  相似文献

Atlantic salmon (Salmo salar L.) were fed on a control diet or experimental diets containing betaine (15 mg g-1) or dimethylglycine (DMG, I mg g-1 or 5 mg g-1). After 10 weeks of feeding, resistance to infection was assessed following inoculation with Vibrio anguillarum. Total blood and differential leucocyte counts were made, and plasma lysozyme and ceruloplasmin were assayed as non-specific humoral factors. The mortality during the bacterial exposure was of the same magnitude in all feeding groups. Betaine or DMG had no effect on the 'basal' levels of plasma total protein, lysozyme or ceruloplasmin, but 3 days postinjection the lysozyme and ceruloplasmin levels were higher in the control group compared with the experimental groups. In both DMG groups, the lymphocyte response took place 1-2 weeks earlier than in the control or betaine supplemented group indicating that DMG has an immunomodulating effect on salmon.  相似文献

The effects of three-month dietary xylitol supplementation on the amounts and hexose contents of acid-soluble collagen as well as on the amounts and fluorescence of collagenase-soluble collagen were studied in healthy and streptozotocin-diabetic male rats. Collagen was extracted from skin samples. In the healthy rats, supplementation with xylitol (10%) increased the hydroxyproline content of the acid-soluble fraction and skin thickness. In diabetic rats receiving and not receiving xylitol, the acid-soluble collagen fraction was markedly lower than in healthy rats. However, its amount was significantly elevated when xylitol had been added to the diet. Supplementation with xylitol caused no changes in the amounts of collagenase-soluble fraction in either healthy or diabetic rats. Supplementation with xylitol (10%) significantly decreased the hexose content of acid-soluble collagen and the fluorescence of the collagenase-soluble fraction in both healthy and diabetic rats. The results indicate that dietary xylitol affects collagen synthesis and collagen glycosylation.  相似文献

S ummary . Quantitative methods were used to study the microflora of faeces produced by a group of baboons receiving fresh fruit and vegetables (natural diet), and 3 groups fed a synthetic diet deficient in either pyridoxine or riboflavin, or which contained both vitamins (pair fed). There was no significant difference between the microbial counts of any of the groups of organisms studied in the pyridoxine or riboflavin deficient and pair fed baboons. Compared with baboons on natural diet, faecal counts from baboons fed the synthetic diet showed highly significant increases in Clostridium welchii and in lactose-fermenting enterobacteria, together with a considerable decrease in the number of lactobacilli. Small increases in the numbers of micrococci, staphylococci and faecal streptococci were noted, whilst the numbers of yeasts were reduced slightly. Whilst the change from the natural to the synthetic diet caused a great alteration in the flora, the synthetic diet flora appeared to be extremely stable and generally unaffected by drastic modifications of the synthetic diet. However, marked alteration did occur on re-feeding with the natural diet. Data on the faecal microflora of baboons are compared briefly with published data for man and other animals, and the effects of dietary changes on the composition of the faecal flora are discussed.  相似文献

The experiment was conducted to evaluate the effect of copper-loaded chitosan nanoparticles on the small intestinal morphology and activities of digestive enzyme and mucosal disaccharase in rats. Forty male Sprague–Dawley rats, with average body weight of 82 g, were randomly allotted to five groups (n = 8). All rats were received a basal diet (control) or the same basal diet added with 80 mg/kg BW CuSO4, 80 mg/kg BW chitosan (CS-I), 80 mg/kg BW copper-loaded chitosan nanoparticles (CSN-I), 160 mg/kg BW copper-loaded chitosan nanoparticles (CSN-II), respectively. The experiment lasted 21 days. The results showed that the villus heights of the small intestinal mucosa in groups CSN-I and CSN-II were higher than those of the control, group CuSO4 or CS-I. The crypt depth of duodenum and ileum mucosa in group CSN-I or CSN-II was depressed. Compared with the control, there were no significant effects of CuSO4 or CS-I on the villus height and crypt depth of small intestinal mucosa. Supplementation with CSN improved the activities of trypsin, amylase and lipase in the small intestinal contents and maltase, sucrase and lactase of duodenum, jejunum, and ileum mucosa while there were no significant effects of CuSO4 on the digestive enzyme activities of the small content compared with the control. The results indicated that intestinal morphology, activities of digestive enzyme in digesta and mucosal disaccharase were beneficially changed by treatment of copper-loaded chitosan nanoparticles.  相似文献

目的 探讨复合微生态制剂对断奶仔猪生长性能、粪便乳酸菌和大肠埃希菌数、血液红胞数以及血红蛋白含量及其对免疫水平的影响.方法 选择25日龄的断奶仔猪共108头,平均分为两组:Ⅰ组和Ⅱ组,每组3个重复,每个重复18只,Ⅰ组饲喂全价料+2.0‰复合微生态制剂;Ⅱ组为对照组,饲喂全价料,实验期为42 d.结果 至实验结束时,复合微生态制剂组的料重比显著低于对照组(P <0.05);14、28和42 d时复合微生态制剂组粪便中的大肠埃希菌数比对照组低7.94%,2.90%和4.89%,与对照组相比差异有统计学意义(P<0.05);14d时,复合微生态制剂组粪便乳酸菌数与对照组相比差异有统计学意义(P<0.05),28和42 d时,两组粪便乳酸菌数差异无统计学意义(P>0.05).14、28和42 d时复合微生态制剂组血液中红细胞数和血红蛋白浓度均显著高于对照组.两组血清尿素氮水平差异无统计学意义(P>0.05),复合微生态制剂组血清总蛋白和IgG的含量与对照组相比有升高的趋势,但差异无统计学意义(P>0.05),复合微生态制剂组血清IgA和IgM的浓度显著高于对照组(P<0.05).结论 复合微生态制剂可以显著降低断奶仔猪的料重比,降低粪便中大肠埃希菌数量,促进血液红细胞和血红蛋白的含量,提高断奶仔猪的免疫水平.  相似文献

Bait sprays containing the toxicant spinosad (GF-120) were applied to citrus groves in the Rio Grande Valley of Texas where Mexican fruit flies were detected in surveillance traps. The sprays were applied as a supplement to a continuous sterile insect release program. Sterile fly captures were 47-63% lower in the treated groves compared with control groves. Eight of 10 secondary pest populations declined in the test groves subsequent to spray applications, but they also declined in the control groves, suggesting that the decline was a seasonal phenomenon rather than a result of the bait sprays. Citrus whitefly, Dialeurodes citri (Ashmead), populations increased modestly and citrus blackfly, Aleurocanthus woglumi (Ashby), populations remained unchanged compared with pretreatment levels. Thus, no outbreaks of secondary pests occurred as a result of the spinosad bait sprays in this instance, as has been reported for malathion bait sprays in citrus. The bait sprays had no detectable effect on populations of specific indicator species of parasitoids (including Aphytis spp. and Comperiella bifasciata Howard), or on numbers of beneficial insects in general, in the treated groves.  相似文献
