Transesterification activity and the industrial potential of a novel lipase prepared from Acinetobacter ventiatus RAG-1 were evaluated. Purified lipase samples were dialyzed against pH 9.0 buffer in a single optimization step prior to lyophilization. The enzyme and organic phase were pre-equilibrated (separately) to the same thermodynamic water activities (a w) ranging from a w 0.33 to 0.97. Production of 1-octyl butyrate by lipase-catalyzed transesterification of vinyl butyrate with 1-octanol in hexane was monitored by gas chromatography. Production of 1-octyl butyrate and initial rate of reaction depended on water activity. Product synthesis and rate of transesterification increased sharply with increase from a w 0.33 to 0.55. Highest product concentration (218 mM) and rate of reaction (18.7 μmol h−1 · 10 μg protein) were measured at a w 0.86. Transesterification activity in hexane represented 32% of comparable hydrolytic activity in aqueous buffer.  相似文献

The use cyclodextrins for the intensification of bioremediation by improving the mobility and bioavailability of contaminants has recently been studied. In this work, the role of randomly methylated -cyclodextrin in the bioremediation of soils contaminated with transformer oil was studied both in bench scale bioreactors and through field experiments. The aims of this research were to (a) establish the scientific background of a cyclodextrin-based soil bioremediation technology, (b) demonstrate its feasibility and effectiveness in the field, and (c) develop an integrated methodology, consisting of a combination of physical, chemical, biological and ecotoxicological analytical methods, for efficiently monitoring the technology performances. The stepwise increasing scale of the experiments and the application of the integrated analytical methodology supported the development of a scientifically established new technology and the identification of the advantages and the limitations of its application in the field. At each phase of the study, randomly methylated -cyclodextrin was found to significantly enhance the bioremediation and detoxification of the transformer oil-contaminated soils employed by increasing the bioavailability of the pollutants and the activity of indigenous microorganisms.  相似文献

A bacterial strain was isolated from soil that was contaminated with diesel oil and was used in our experiments. The strain was then phenotypically, biochemically and genetically tested and named as Achromobacter 4(2011). In order to examine the impact of long-term contact with diesel oil of bacterial cells, the strain was stored under different conditions – on standard nutrient agar plates and on agar plates with 50 μl diesel oil as a sole carbon and energy source. The results clearly indicated that longer contact with diesel oil led to changes in both the bacterial surface and biochemical properties, as well as the hexadecane monooxygenase activity. Moreover, the fatty acid profiles also changed, leading to an increased content of saturated fatty acids. In addition, the rates of biodegradation of diesel oil were higher even when supplemented with the surfactants – rhamnolipids and saponins. This work demonstrates that prolonged contact of microorganisms with diesel oil can lead to many changes, not only in biodegradation potential, but also in their surface and genetic properties.  相似文献

An aliphatic ether (1-phytanyl-1-octadecanyl-ether) of high molecular weight was used as a sole carbon source in degradation experiments with different aerobic bacteria. The enriched culture B5, obtained from fuel contaminated soils, was able to degrade the substance for more than 90%. A culture of Rhodococcus ruber was similarly effective. Detailed investigation of the metabolites allowed us to characterize an unusual degradation pathway via a mid-chain oxidation mechanism (`internal oxidative pathway'). Obviously, formation of intermediate alkenes mainly at the unbranched side chain was a prerequisite for bacterial degradation of the added substrate. Degradation proceeded – in spite of the usually preferred terminal oxidation – via oxidation of the internal double bond and was followed by an ester cleavage. In turn, a series of alcohols was formed which were subsequently oxidized to the respective carboxylic acids and were further metabolized via the normal -oxidation pathway.  相似文献

Eccentric exercise has been shown to have a measurable effect on the force-length relationship (FLR), as peak force is shifted to longer muscle lengths following exercise. Recently, this shift in the FLR has been proposed as a "simple, reliable indicator" for assessing contractile element damage following eccentric exercise. However, eccentric exercise causes fatigue and damage, and there is evidence that fatigue alone may also cause a shift in the FLR. The purpose of this paper was to assess the role of fatigue on the FLR (as measured by a torque-joint angle relationship) following isometric and eccentric exercise in the New Zealand white (NZW) rabbit. Six NZW rabbits were divided into two groups for eccentric or isometric contractions of the hindlimb dorsiflexor muscles. Pre- and post-exercise torque-joint angle relationships were measured, and the shift from the pre- to the post-exercise relationship was measured as the change in joint angle at which peak torque was produced. Eccentric exercise resulted in a rightward shift of seven degrees; isometric exercise, which is thought to not cause damage, resulted in a shift of four degrees. Furthermore, torque production was reduced to a greater extent at short compared to long muscle lengths for the eccentric and isometric exercise, resulting in a post-exercise torque-joint angle relationship that was altered in shape. We conclude from these results, that the shift in peak torque may not be a simple and reliable indicator of muscle damage, but is caused by a combination of damage and post-exercise fatigue.  相似文献

To assess whether allantoin levels in serum and urine are influenced by exhaustive and moderate exercise and whether allantoin is a useful indicator of exercise-induced oxidative stress in humans, we made subjects perform exhaustive and moderate (100% and 40% VO2max) cycling exercise and examined the levels of allantoin, thiobarbituric acid reactive substances (TBARS) and urate in serum and urine. Immediately after exercise at 100% VO2max, the serum allantoin/urate ratio was significantly elevated compared with the resting levels while the serum urate levels was significantly elevated 30 min after exercise. The serum TBARS levels did not increase significantly compared with the resting levels. Urinary allantoin excretion significantly increased during 60 min of recovery after exercise, however, urinary urate excretion decreased significantly during the same period. The urinary allantoin/urate ratio also rapidly increased during 60 min of recovery after exercise. Urinary TBARS excretion decreased during the first 60 min of the recovery period and thereafter significantly increased during the latter half of the recovery period. On the contrary, after 40% VO2max of exercise, no significant changes in the levels of urate, allantoin and TBARS in serum or urine were observed. These findings suggest that allantoin levels in serum and urine may reflect the extent of oxidative stress in vivo and that the allantoin which appeared following exercise may have originated not from urate formed as a result of exercise but from urate that previously existed in the body. Furthermore, these findings support the view that allantoin in serum and urine is a more sensitive and reliable indicator of in vivo oxidative stress than lipid peroxidation products measured as TBARS.  相似文献

The constituents of tar oil comprise a wide range of physico-chemically heterogeneous pollutants of environmental concern. Besides the sixteen polycyclic aromatic hydrocarbons defined as priority pollutants by the US-EPA (EPA-PAHs), a wide range of substituted (NSO-PAC) and alkylated (alkyl-PAC) aromatic tar oil compounds are gaining increased attention for their toxic, carcinogenic, mutagenic and/or teratogenic properties. Investigations on tar oil biodegradation in soil are in part hampered by the absence of an efficient analytical tool for the simultaneous analysis of this wide range of compounds with dissimilar analytical properties. Therefore, the present study sets out to explore the applicability of comprehensive two-dimensional gas chromatography (GC2/MS) for the simultaneous measurement of compounds with differing polarity or that are co-eluting in one-dimensional systems. Aerobic tar oil biodegradation in a historically contaminated soil was analyzed over 56 days in lab-scale bioslurry tests. Forty-three aromatic compounds were identified with GC2/MS in one single analysis. The number of alkyl chains on a molecule was found to prime over alkyl chain length in hampering compound biodegradation. In most cases, substitution of carbon with nitrogen and oxygen was related to increased compound degradation in comparison to unalkylated and sulphur- or unsubstituted PAH with a similar ring number.The obtained results indicate that GC2/MS can be employed for the rapid assessment of a large variety of structurally heterogeneous environmental contaminants. Its application can contribute to facilitate site assessment, development and control of microbial cleanup technologies for tar oil contaminated sites.  相似文献

Purpose In 2003 the Dutch Health Care Inspectorate introduced performance indicators to monitor and compare quality of care in Dutch hospitals. In 2007, the new performance indicator ‘one-year mortality after a first visit to a cardiology outpatient clinic’ was introduced. We set out to evaluate this new indicator in three Dutch teaching hospitals. Methods Using electronic medical records, information was collected retrospectively of patients aged ≥70 years who visited the cardiology outpatient clinic of Medical Centre Alkmaar, Meander Medical Centre Amersfoort and Deventer Hospital between 1 January 2006 and 31 January 2006. Diagnoses were based on the diagnosis treatment combination (DBC) coding system. Results 547 patients (mean age 78.0 years, 53% men) were included, 35 (6.4%) of whom had died after one year. Cardiovascular disease was the most frequent cause of death (22/35, 62.9%). The oneyear mortality among the three hospitals varied from 5.0 to 7.3% (NS). Conclusion One-year mortality after the first visit to a cardiology outpatient clinic amounted to 6.4% in patients aged ≥70 years and did not differ significantly between the three Dutch teaching hospitals. The administrative load to obtain the necessary information was considerable. One-year mortality should be regarded as an ‘outcome’ parameter rather than a ‘performance’ indicator. A range of biodiversity metrics are available to assess the ecological integrity of aquatic ecosystems. However, performance varies considerably among different types of metrics and provides different information regarding ecosystem conditions, thus making difficult the selection of appropriate metrics for biomonitoring. The present study evaluated the robustness of six biodiversity metrics to assess environmental change and determine their utility as relevant indicators of ecosystem biodiversity and functionality. Traditional metrics such as species richness and Shannon diversity were considered along with alternative metrics such as functional diversity, size diversity and taxonomic distinctness. To that end, invertebrate assemblages in a river floodplain were used as a case study to evaluate the performance of metrics using Generalized Additive Models (GAM). GAM explained between eight and 49% of the variability in biodiversity. The regression models exhibited differences in the response of biodiversity indicators to environmental factors, suggesting that intermediate levels of turbidity and low salinity are conditions favouring increased biodiversity in the study area. Based on correlations among metrics and responses to primary environmental factors, it is concluded that Shannon and functional diversity, and rarefied species richness generated similar information regarding ecosystem conditions (i.e., the metrics were redundant); while size diversity and distinctness provided useful additional data characterizing ecosystem quality (i.e., the metrics were complementary). Functional diversity indicated not only number and dominance of species, but also each species functional role in the community, and was therefore the most informative biodiversity metric. Nevertheless, the use of a combination of metrics, for example functional and size diversity, and variation in taxonomic distinctness, provides complementary data that will serve to achieve a more thorough understanding of ecosystem structure and function, and response to primary environmental influences.  相似文献

For biofilm studies, artificial models can be very helpful in studying processes in hydrogels of defined composition and structure. Two different types of artificial biofilm models were developed. Homogeneous agarose beads (50–500 μm diameter) and porous beads (260 μm mean diameter) containing pores with diameters from 10 to 80 μm (28 μm on average) allowed the embedding of cells, particles and typical biofilm matrix components such as proteins and polysaccharides. The characterisation of the matrix structures and of the distribution of microorganisms was performed by confocal laser scanning microscopy. The physiological condition of the embedded bacteria was examined by redox activity (CTC-assay) and membrane integrity (Molecular Probes LIVE/DEAD-Kit). Approximately 35% of the immobilised cells (Pseudomonas aeruginosa SG81) were damaged due to the elevated temperature required for the embedding process. It was shown that the surviving cells were able to multiply when provided with nutrients. In the case of homogeneous agarose beads, cell growth only occurred near the bead surface, while substrate limitation prevented growth of more deeply embedded cells. In the porous hydrogel, cell division was observed across the entire matrix due to better mass transport. It could be shown that embedding in the artificial gel matrix provided protection of immobilized cells against toxic substances such as sodium hypochlorite (0.5 mg/l, 30 min) in comparison to suspended cells, as observed in other immobilized systems. Bioimprinting and sol–gel encapsulation of lipases by silane precursors are efficient methods of enhancing lipase performance in non-aqueous medium. The correlation between bioimprinting, the alkyl-chain length of silane precursors, and the catalytic activity of gel-encapsulated lipase was investigated using a series of silane precursors: methyltrimethoxysilane (MTMS), vinyltrimethoxysilane (VTMOS), vinyltriethoxysilane (VTEOS), and n-octyltrimethoxysilane (OTMOS). The optimal parameters for lipase immobilization were also determined. Both bioimprinting and increasing the chain-length of alkyl groups, apparently by increasing hydrophobicity, significantly improved the specific activity and the total activity of the immobilized lipase. Compared to a non-imprinted MTMS/TMOS gel, the specific activity of an imprinted OTMOS/TMOS gel improved 14.4-fold, and the total activity improved 6.8-fold. Nitrogen adsorption–desorption assays and gel matrix surface characterization showed that the bioimprinting molecule and the hydrophobic alkyl groups of silane triggered lipase to change from the closed to the open conformation, and contributed to creating sol–gel matrices that were more porous and with less mass transfer resistance structure, apparently improving the activity of encapsulated lipase.  相似文献

Arbuscular mycorrhizal barley-plants were more susceptible to the obligate biotrophic shoot pathogen Erysiphe graminis f. sp. hordei. In experiments under greenhouse and open-air conditions on leaves of mycorrhizal plants, the sporulation rate of the mildew fungus was more than twice that on control plants. However, mycorrhizal plants suffered less than non-mycorrhizal plants in terms of grain number, ear yield and thousand-grain weight. Disease-yield-relationship analysis showed that the symbiosis neutralised the positive correlation between disease severity and yield loss (up to 25% infected leaf area tested). After mildew infection, nitrogen in ears of non-mycorrhizal barley was higher because of an impaired starch accumulation during grain filling. In mycorrhizal plants, leaf disease did not impair either the quantity or quality of grain yield. This improved compensation in mycorrhizal plants was related to maintained photosynthetic capacity and a delay in pathogen-induced senescence. Thus filling of long-term storage pools (fructans in internodes) and consequently reallocation of these reserves during grain filling was improved. The results suggest that higher availability of energy and material during grain formation, together with longer physiological activity, were the basis of yield maintenance and, therefore, expression of mycorrhiza-induced tolerance towards the pathogen.  相似文献

Kluyveromyces marxianus was grown in submerged culture in a complex medium with several potential inducers of lipolytic activity (triacylglycerols, fatty acids). The highest extracellular lipolytic enzyme production (about 80 U ml–1 in 3 d) was obtained when the medium was supplemented with 2 g urea l–1 plus 5 g tributyrin l–1. Addition of surfactants (1 g l–1) did not improve production. The lipase had a high thermal stability in aqueous solution (73% residual activity after 9 d at 50 °C, 16 min half-life time at 100 °C). It was also stable at acidic pH and showed good tolerance to organic solvents (70% residual activity after 2 d in n-hexane of cyclohexane).  相似文献

Ellenberg indicator values (EIVs) describe the realized niche of species and habitat parameters, and are commonly used for vascular plants, bryophytes, and lichens. We provide a methodology for EIVs and an EIV list for nearly 650 species of macromycetes. We propose a new EIV scale, namely substrate openness (O). We also give the results of two applications and compare EIV values related to the Red List classification with those related to lifestyle classification. Mycorrhizal species on average have higher demands on substrate openness and are less tolerant of high nutrient levels than saprotrophic or parasitic species. Critically endangered species have on average distinctly higher demands on openness of habitat and substrate than not threatened or less strongly threatened species, which in turn have higher demands for nutrient availability. This pattern clearly highlights the points of threat for many macromycete species.  相似文献

Plant extracts and essential oils have been widely studied and used as antimicrobial agents in the last decades. In our study we investigated the antimicrobial activities of Olbas(?) Tropfen (in the following named Olbas), a traditionally used complex essential oil distillate, in comparison to its individual essential oil ingredients. Olbas (10g) consists of three major components such as peppermint oil (5.3g), eucalyptus oil (2.1g), and cajuput oil (2.1g) and of two minor constituents like juniper berry oil (0.3g) and wintergreen oil (0.2g). The composition of Olbas and the five individual essential oils were characterized by GLC-MS. According to GLC-MS analysis 1,8-cineol is the main component of the complex essential oil distillate followed by menthol and menthone. The minimum inhibitory and minimum microbicidal concentrations of Olbas and each of the single essential oils were evaluated in 17 species/strains of bacteria and fungi. Time-kill assay was performed to compare the microbicidal activity of Olbas and peppermint oil during several time intervals. Olbas displayed a high antimicrobial activity against all test strains used in this study, among them antibiotic resistant MRSA (methicillin-resistant Staphylococcus aureus) and VRE (vancomycin-resistant Enterococcus). Its antimicrobial activity was comparable to that of peppermint oil which was the most potent one of all individual essential oils tested. In the time kill assay Olbas as well as peppermint oil demonstrated similar microbicidal activities. Based on its wide antimicrobial properties Olbas can be a useful agent for the treatment of uncomplicated infections of skin and respiratory tract.  相似文献

Polyethylene is considered one of the most durable plastic polymers. Virtually, non-biodegradable polyethylene accumulates in the environment, thus posing an ecological threat to man and wildlife. We have previously isolated a strain of the actinomycete Rhodococcus ruber (designated C208; EC 1.10.3.2.) capable of utilizing and degrading polyethylene. Here, we report the role of the bacterial copper-binding enzyme, laccase, in the oxidation and degradation of polyethylene by this strain. Copper markedly affected the induction and activity of laccase, resulting in polyethylene degradation. mRNA quantification by RT-PCR, revealed a 13-fold increase in laccase mRNA levels, in copper-treated cultures compared with the untreated control. Addition of copper to C208 cultures containing polyethylene enhanced the biodegradation of polyethylene by 75%, as compared with the non-amended control. Furthermore, when an extracellular isoform of laccase collected from the media of copper-induced cells was incubated with polyethylene, reductions of 20% and 15% were obtained in the Average Molecular Weight (Mw) and Average Molecular Number (Mn) with the polymer, respectively. FTIR analysis of similar polyethylene films incubated with the extracellular laccase exhibited an increase in the carbonyl peak, indicating that enzymatic oxidation by laccase plays a major role in the biodegradation of polyethylene.  相似文献

Objective: To review the recent research into human‐mediated dispersal (HMD) in the European rural landscape, and explore the potential positive aspect of HMD for grassland conservation, in contrast to it's common association with the spread of invasive species. Methods: A literature search was undertaken to identify HMD vectors in the rural landscape for discussion regarding dispersal potential past and present, implications for management, and the identification of future research needs. Results: Grazing animals are important propagule dispersers, but the reduced movement of livestock through the landscape has also meant a reduction in seeds dispersed in this way. Other, non‐standard human‐mediated dispersal vectors such as clothing and motor vehicles can also transport seeds of many species, and HMD vectors often transport seeds with a variety of dispersal specialisations. Recommendations: There should be a greater movement of grazing animals throughout the landscape, either within larger grazing areas or between existing grasslands. Where this is not possible, other, more directed dispersal of propagules from species‐rich communities to target sites should be considered. The potential of non‐standard HMD vectors to make a positive contribution to biodiversity should be considered, but more research into all types of HMD vectors is important if we are to fully understand their role in the dispersal of plant species in fragmented landscapes.  相似文献

Recently, research on the use of binary aqueous-organic liquid phase systems for the treatment of polluted air has significantly increased. This paper reports the removal of α-pinene from a waste air stream in a continuous stirred tank bioreactor (CSTB), using either a single-liquid aqueous phase or a mixed aqueous-organic liquid phase. The influence of gas flow rate, load and pollutant concentration was evaluated as well as the effect of the organic to aqueous phase ratio. Continuous experiments were carried out at different inlet α-pinene concentrations, ranging between 0.03 and 25.1 g m?3 and at four different flow rates, corresponding to residence times (RTs) of 120 s, 60 s, 36 s and 26 s. The maximum elimination capacities (ECs) reached in the CSTB were 382 g m?3 h?1 (without silicone oil) and 608 g m?3 h?1 (with 5%v/v silicone oil), corresponding to a 1.6-fold improvement using an aqueous-organic liquid phase. During shock-loads experiments, the performance and stability of the CSTB were enhanced with 5% silicone oil, quickly recovering almost 100% removal efficiency (RE), when pre-shock conditions were restored. The addition of silicone oil acted as a buffer for high α-pinene loads, showing a more stable behaviour in the case of two-liquid-phase systems.  相似文献

During analysis of components of baobab (Adansonia digitata) seed oil, several new fluorescent compounds were detected in HPLC chromatograms that were not found previously in any seed oils investigated so far. After preparative isolation of these compounds, structural analysis by NMR spectroscopy, UHPLC-HR-MS, GC-FID and spectroscopic methods were applied and allowed identification of these substances as series of N-acylserotonins containing saturated C22 to C26 fatty acids with minor contribution of C27 to C30 homologues. The main component was N-lignocerylserotonin and the content of odd carbon-atom-number fatty acids was unusually high among the homologues. The suggested structure of the investigated compounds was additionally confirmed by their chemical synthesis. Synthetic N-acylserotonins showed pronounced inhibition of membrane lipid peroxidation of liposomes prepared from chloroplast lipids, especially when the peroxidation was initiated by a water-soluble azo-initiator, AIPH. Comparative studies of the reaction rate constants of the N-acylserotonins and tocopherols with a stable radical 2,2-diphenyl-1-picrylhydrazyl (DPPH) in solvents of different polarity revealed that N-acylserotonins showed similar activity to δ-tocopherol in this respect. The described compounds have been not reported before either in plants or in animals. This indicates that we have identified a new class of plant lipids with antioxidant properties that could have promising pharmacological activities.  相似文献
