This study assesses the influence of thermal regime on the development, survival rates and early growth of embryos of sea lamprey Petromyzon marinus incubated at five constant temperatures (7, 11, 15, 19 and 23° C). The time from fertilization to 50% hatching and from hatching to 50% burrowing were inversely related to incubation temperature. All the embryos incubated at 7° C died at very early stages, while those maintained at 11° C did not attain the burrowing stage. Survival from fertilization to hatching was 61, 89, 91 and 89% at 11, 15, 19 and 23° C, decreasing to 58, 70 and 70% from hatching to burrowing at 15, 19 and 23° C, respectively. Larvae reared during the first 3 months of exogenous feeding in a common environment at constant 21° C, revealed maximum survival for an incubation temperature of 15° C (43% of burrowed larvae) decreasing strongly at 19° C (16%) and 23° C (one suvivor among 240 larvae). Body length at the burrowing stage was maximum for embryos incubated at 19° C, but body mass increased in the interval 15–23° C. Mean incubation temperatures experienced by 117 broods during the embryonic development in the source river were estimated in 15·3±2·30° C and 16·7±1·76° C (mean±1 s.d .) for the periods fertilization-to-hatching and hatching-to burrowing, respectively.  相似文献

Growth of eight selected potentially pathogenic strains of Bacillus cereus was evaluated in a rich medium at different temperatures. No strain grew at 50°C; maximal growth-permissive temperatures were in the range 46–50°C for six strains and under 46°C for one strain. Faster growth occurred at 42°C. Growth may be delayed at 20°C, where the lag phase can reach 7 h. Furthermore at 20°C, cells generally show an aggregation immediately in the early exponential stage, except for two strains. Owing to this aggregation, growth is more difficult to estimate by turbidimetry at lower temperatures. These data describe the behaviour of type and field strains between 50° and 20°C and can help the prediction of shelf-life of potentially contaminated products.  相似文献

When Arctic charr Salvelinus alpinus from two diVerent stocks were fed live Neomysis integer , the upper thermal limits for feeding and growth were established in the range 21·5–21·8° C. These critical temperatures might have been underestimates, because fish tend to show increased sensitivity to handling at high experimental temperatures. In the second experiment, the proportion of feeding undisturbed charr from four stocks decreased initially as temperature was raised in steps from 18 to 22° C. At the lower temperatures, 18 and 20) C, almost all fish resumed feeding, but the recovery time was longer and more fish ceased to feed at 20) C than at 18° C. When the temperature was increased to 21° C, 50% of the fish ceased feeding permanently, and all fish ceased feeding within 2 days at 22° C. It is concluded that 0+ charr cease to feed and grow at c .21·5) C and that the critical temperatures for feeding and growth coincide.  相似文献

Electric Field-induced Fusion of Sea Urchin Eggs   总被引:2,自引:2,他引:0  
An electrical method is described which permits the fusion of denuded eggs of the sea urchin species Paracentrotus lividus . In a nearly non-conductive medium, containing 1.2 M glucose at pH 6.0–8.4, eggs or fertilized stages are brought into close membrane contact by dielectrophoresis arising from the application of a highly inhomogeneous alternating electric field. During this process the eggs aligne parallel to the field forming "egg-chains". In well pigmented eggs pigmentcapping is observed in the areas of cell contact. After completion of the alignment, the application of an additional single high field pulse of μs duration induces fusion of two or more eggs. The mechanism underlying the fusion process is the reversible electric breakdown of membranes in the zones of cell-to-cell contact. Fusion proceeds within 1–10 min at 10–20°C. Fused eggs have intact nuclei, can be fertilized, but undergo abortive cleavage.  相似文献

Seventy isolates of moulds and blue stain fungi ( Zygomycota and Deuteromycota ), isolated from discoloured outdoor softwood in Sweden, comprising of 27 different species, (the two largest genera Penicillium and Cladosporium ) were investigated for their linear growth at three different start-pH values (5, 7 and 9.5) at two temperatures (2°C and 24°C) on malt extract agar (MEA). At 24°C all isolates showed growth at all three start-pH values except for one isolate which did not grow at initial-pH 9.5. After 21 days at 2°C at the three start pH-values, only six isolates showed no growth indicating that 64 of the isolates were cold-tolerant (psychrotrophic). Of these 64 strains, 58 showed growth at an initial pH of 9.5. Lower pH optima at 2°C than at 24°C were found for most of the isolates. The reduction of the linear growth at initial pH 9.5 in relation to the growth at optimal pH was more pronounced (higher) at the low temperature.  相似文献

Stages of prophase of living primary spermatocytes were determined by use of Rose culture chambers (1). Dissociated primary spermatocytes were cultured at low cell-density in a collagen matrix at 22°C or 27°C and the percentages of cells which had progressed from various stages in prophase through meiosis to various advanced stages were measured. In a standard medium (Leibovitz-15 + 10% fetal bovine serum), more than 70% of the primary spermatocytes at stages beyond the pachytene stage could advance to round spermatids with flagella within a few days at 22°C. The percentages of cells that progressed from stages before the late zygotene stage were less, but at least 13 % of leptotene cells reached metaphase I within a week at 22°C. The percentage of cells that progressed was slightly lower at 27°C than at 22°C: 6.3 and 4.3 days were required for progress from leptotene to metaphase I at 22°C and 27C, respectively. Fetal bovine serum was not indispensable for progression through meiosis. Moreover, 0.5–5.0 μg/ml ovine follicle stimulating hormone (NIAMDD-o-FSH-13), 0.01–1.0 μg/ml 5α-dihydrotestosterone and 1.0 μg/ml testosterone propionate had no significant effect in increasing the percentage of cell progression at 22°C.  相似文献

Temperature-sensitive mutants (TS-1 and TS-7) of a thermotolerant yeast, Hansemula polymorpha CK-1, were isolated. The mutants were unable to grow at 50°C, the maximum growth temperature of the wild type. Mutants TS-1 and TS-7 grown at 20°C showed 33 and 50% viabilities after 6 h of incubation of 50°C, respectively. Mutant TS-1 showed little variation of the degree of fatty acid unsaturation (1.26–1.28/mol) and mutant TS-7 had an almost constant sterol/phospholipid molar ratio (0.31–0.34) at 20, 30 and 40°C, although the wild type had a decrease of the degree of fatty acid unsaturation from 1.56 at 20°C to 1.30 at 40°C and an increase of the sterol/phospholipid molar ratio from 0.26 at 20°C to 0.54 at 40°C.  相似文献

Abstract A total of 84 strains of microaerophiles were isolated from the Edo River mouth, Tokyo Bay, Japan, and their morphological, physiological and biochemical properties and mole percent guanine-plus-cytosine contents (mol%) of DNAs extracted from them were examined. Eighty-eight to 100% of the strains, collected in summer, grew at 15–37°C but only 5% of the strains grew at 5°C. Seventy-seven to 100% of the strains, collected in winter, were able to grow at 10–25°C but 79% of the strains were unable to grow at 37°C. The shift in growth-range of bacteria strongly suggests that the microflora of estuarine microaerophiles changes seasonally. All the strains could grow at 50% seawater while 70 and 42% of the strains could not grow at 0 and 100% seawater, respectively.
On the other hand, 72 strains, out of the 84 strains, clustered into 9 phena using numerical taxonomy methods. The strains belonging to phena 1 through 9 were all Gram-negative, non-spore-forming rod-shaped organisms which were negative for susceptibility to vibrostatic agent O/129, H2S production and acid from dulcitol, and the G + C contents of DNAs ranged from 29.8 to 56.6 mol%. However, almost all strains could not be identified and are suggested to be new species.  相似文献

ABSTRACT. In laboratory-laid eggs of the striped ground cricket, Allonemobius fasciatus DeGeer (Gryllidae), water absorption occurs at an early stage of embryogenesis (stage II) at 30°C but is delayed until later stages at lower temperatures. This is related to the variation in diapause stage at different temperatures. No egg developed beyond stage VII (the end of anatrepsis) without water absorption.
A. fasciatus shows seasonal variation in the stage of water absorption. At 30°C, eggs collected in August absorb water at the early stage while many of those collected in September avert diapause and absorb water at a later stage.
Diapause also influenced the water absorption of eggs in A. socius Scudder. Eggs of short-day females enter diapause and absorb water at stage II, while those of long-day females develop without diapause and absorb water at a later stage (around stage IV).
The susceptibility to desiccation (r.h. 50%) was examined at 20°C with A. fasciatus eggs. The percentage water loss and mortality of eggs varied with the time and duration of exposure to desiccation. Eggs are most sensitive to desiccation during the first several days after being laid and during the period of water absorption.  相似文献

ABSTRACT. Free-living amoebae belonging to the species Naegleria fowleri are known to be the etiological agents for a form of fulminant meningoencephalitis that is generally fatal (primary amoebic meningoencephalitis). In a broad bacterial screening from soil and water we have isolated three strains (M-4, D-13 and A-12) belonging to the species Bacillus licheniformis that have remarkable amoebicidal activity against Naegleria sp. and also against different Gram-positive and Gram-negative bacteria. Physical-chemical characteristics, partial purification and biological activities of a substance produced by the M-4 strain have been investigated. This substance (m-4) is stable at high temperature (up to 100°C) and extremes of pH (2.5-9.5) and also at -20°C for months. Its production is greatly influenced by oxygenation of the cultures and is probably related to the sporulation process of the bacterium. Scanning electron microscope observations reveal that amoebae are lysed after a few minutes contact with m-4.  相似文献

The production and stability of pediocin N5p from Pediococcus pentosaceus , isolated from wine, were examined in grape juice medium. Maximum growth and higher titre (4000 U ml-1) were observed at a initial pH of 7·5 and 30°C. The activity of the inhibitory substance was stable between pH values from 2·0 to 5·0 at 4° and 30°C. At pH 10·0 it was completely inactivated. When submitted to 30 min at 80°, 100° and 115°C, maximal stability was observed at pH 2·0. Ethanol up to 10% did not affect pediocin activity at acid pH, nor did 40–80 mg 1-1 SO2, independently or combined with different ethanol concentrations, affect inhibitory activity.  相似文献

Development of the Paraguayan anuran Lepidobatrachus laevis is unusual in that the larvae are obligate carnivores, facultative cannibals and apparently exist at high environmental temperatures in their natural habitat. In the present study, the effect of environmental temperature on the rate of anuran development was investigated. The larvae have a thermotolerance range of 18°C for normal development between 19 and 37°C. The effect of temperature on the rate of development was dramatic; larvae that were incubated at 36.8°C develop to stage 24 (Gosner) in approximately 9 h compared with 24 h for larvae incubated at 19°C. The ability of larvae to survive heat shock was also examined; larvae did not survive a shock of 45°C for 15 min when it was administered at stages 3, 5, 9, 10 or 20. However, using the same heat shock conditions, 50% survival was observed when larvae were shocked at stage 16. To study protein synthesis during heat shock, larvae were pulsed with [35S]-methionine during heat shock and labeled proteins were analyzed by electrophoresis under reducing and denaturing conditions. Larvae synthesized two sets of heat-shock proteins at doublet molecular weights of 83/78 and 62/59 kDa. These proteins were synthesized independently of the stage of development at which the shock was administered or the magnitude of the heat shock.  相似文献

Albino mouse pups develop homoiothermy during the first 19 days of their life and show three distinct stages during this period. From day 1 to day 6 they are almost completely poikilothermic at low ambient temperatures; from day 7 to day 14 or 15 is a transitional period during which they develop homoiothermy at medium temperatures (e.g. 22°C); from day 15 or 16 to day 19 or 20 is the final stage when homoiothermy at very low ambient temperatures is developed. These changes are very closely correlated with changes in the ultrasounds produced by isolated pups within the 19 day period. Thus their acoustic response to cold is weak from day 1 to day 5 or 6 and very strong from day 6 or 7 to day 12 or 13, on exposure even to medium temperatures. From day 14 onwards the pups progressively stop producing ultrasounds depending on the ambient temperatures, and there is complete cessation of calling after day 19 or 20 even on exposure to temperatures as low as 2°C. In general, decreasing the ambient temperatures increases the rate and intensity of ultrasounds produced but very low ambient temperatures have an inhibiting effect and at 2–3°C breathing and calling cease completely after a time which varies with age of the pups.  相似文献

When sea urchin embryos up to the 2-cell stage are treated with 5 × 10−3 M chloramphenicol for a short period, small blastulae filled with mesenchyme-like cells (type A) are formed. If a homogenate of the embryos up to the 2-cell stage is introduced just after the chloramphenicol treatment, almost all embryos developed to normal plutei. Chloramphenicol treatment, started at the 8 ∽ 32-cell stages, induces vegetalized larvae (type B), and presumptive vegetalized ones develop normally after treatment with a homogenate of the embryos at the 16 ∽ 64-cell stages. If embryos are treated in the same manner at 7 hr after insemination, abnormal embryos which seem to be bipolar ones (type C) are observed at 40 hr after the treatment. These also develop normally, if a homogenate of embryos at the stages from unfertilized egg to gastrula is added to them at the end of the chloramphenicol treatment. The substances having these activities are named morphogenetic substances α, β and γ, respectively. The morphogenetic substance β (anti-vegetalizing substance) is heat stable and its molecular weight is less than 10,000.  相似文献

This investigation was performed in order to analyze the basic relationships between the archenteron roof and the overlying ectoderm in primary induction in the Cynopus (Triturus) pyrrhogaster embryo.
The part of the archenteron roof that is active in inducing capacity extends linearly after invagination at the speed of 0.15 mm per hr at 23°C until stage 13b. The period of contact at each position of the presumptive neuro-ectoderm with the active archenteron roof could be estimated by the formula described in the Discussion.
Pieces of the presumptive neuro-ectoderm were isolated from gastrulae at three developmental stages and cultured separately in Holtfreter solution after being divided caudo-cranially into 4 parts. The result showed that some of them were able to differentiate into neural tissues even in the mid-gastrula stage and that the presumptive neuro-ectoderm acquired the capacity to differentiate into neural tissue along a caudocranial axis from the part adjacent to the blastopore during gastrulation.
It could be estimated that 3 hr of contact with the active archenteron roof is sufficient for the presumptive neuro-ectoderm to differentiate into neural tissue.
The present study also showed that the neuralizing capacity of the whole prospective neuro-ectodermal area has already been determined before the end of stage 13, i.e., within less than 14 hr after first contact of the ectoderm with the active archenteron roof at 23°C.  相似文献

The effects of low temperature on differentiation of Dictyostelium discoideum cells in the vegetative and preaggregation stages were examined immunohistochemically and electronmicroscopically. Ultimately cells in both stages were found to stain with FITC-conjugated anti- Dictyostelium mucoroides spore antibody at low tempeature (5°C) in nutrient-rich medium under submerged conditions. These cells were sensitive to 0.1% Triton X-100. In contrast, cells in the late aggregation stage were converted to detergent-resistant spores. Irrespective of their high stainability with antispore-Ig, these cells did not contain the prespore-specific vacuoles (PSVs) and spore-coats characteristic of stainable cells. From these facts, it is concluded that cells in the vegetative and preaggregation stages are unable to differentiate at low temperature into prespore and spore cells, which have PSVs and spore-coats, respectively. The composition of spore-specific (SS) antigens accumulated by these cells was examined by immunoblotting. Results showed that under these conditions the cells mainly produced one SS-antigen, namely SP70. The effects of cell density and components of the medium on the production of SS-antigens were also examined.  相似文献

The growth and activity of juvenile Japanese eels Anguilla japonica in different pigmentation stages from the glass eel to the elver stage were studied in the laboratory at 15, 20 and 25° C. The growth and activity of the eels were significantly influenced by both temperature and fish size. Growth rate generally declined with increasing fish size, and fish were least active and experienced a low growth during the pigmenting stage at all temperatures. They were nocturnal and spent significantly more time moving (swimming, feeding and moving over the substratum) at 20 and 25° C than at 15° C at night within each pigmentation stage. Accordingly, they grew significantly faster at 20 and 25° C than at 15° C throughout the study. The development of pigmentation appeared to be dependant on water temperature but not on fish size. This study suggested that the growth and activity of juvenile Japanese eels were positively correlated, because fish were least active and grew slowest at low temperature (15° C) or during the pigmenting stage at all temperatures.  相似文献
