An acid phosphatase (EC 3 1 3 2)isolated from the seed-coatsof developing pea seeds was estimated to have MW 30000 by gelfiltration on Sephadex G-75 It was shown to act on a broad spectrumof physiological substrates, the most preferred being ß-glycerophosphate,3-phosphoglycerate and ADP, wich all showed rates of about halfthe maximum rate shown with p-nitrophenyl phosphate (p-NPP)Another model substrate frequently used in enzyme localizationstudies, -naphthyl acid phosphate, was hydrolysed at about 30% of the rate shown with p-NPP This acid phosphatase was enhancedor stabilized by the chelators EDTA and 1, 10-phenanthrolme,unaffected by Mg2+ and N-ethyl maleimide, but strongly inhibitedby Zn2+ and F Both oxidized and reduced glutathione werewithout effect at low concentration and slightly inhibitoryat high concentration (15 mm) Thiol groups are clearly not involvedin regulating the activity of this acid phosphatase, a featurewhich distinguishes it from acid phosphatases from several otherplant species. Mechanism of Photosynthate Efflux from Vicia faba L. Seed Coats   总被引:2,自引:0,他引:2  
In order to develop a tentative model of the mechanism of photosynthateefflux from the vascular region of Vicia faba L. seed coats,wash-out experiments were performed after removal of the embryo. The sulphydryl group modifiers, pCMBS and NEM, reduced 14C-photosynthateefflux by 40% and 50%, respectively. Their inhibitory effectcould be prevented or reduced (in the latter case) by includingDTT in the bathing solution. Maltose competed with sucrose forefflux; a concentration of 300 mol m–3 inhibited 14C-photosynthaterelease by 35%. The cations K+ , Na+ Mg2+ and TPP+ enhancedefflux significantly, whereas the countenon Cl had noeffect. The presence of the protonophore CCCP (0·1 molm–3) led to a reduction of efflux by 50% net proton extrusiondropped by 34%. To a lesser extent, an efflux inhibition wasalso achieved by decreasing the cytoplasmic pH with the weakacid DM0. In contrast, alterations in the external pH causedonly a feeble response. The ATPase inhibitor, EB, decreasedphotosynthate efflux and H+ extrusion. DES reduced efflux slightly,presumably by affecting ATPase activity as well as energy metabolism. Based on these findings, it is proposed that a sucrose/protonantiport mechanism could be responsible for photosynthate effluxfrom Vicia faba seed coats. After removal of the embryo from developing ovules of Viciafaba L. and Pisum sativum L., seed-coat exudates were collectedand the amino acid fraction of the exudate was analyzed. InV. faba, alanine was the most important compound of the aminoacid fraction. In P. sativum, alanine and glutamine were thetwo most important components, whereas only small amounts ofasparagine were present. Comparison with published data suggeststhat seed-coat exudates may differ from phloem sap in the relativeimportance of these amino acids. After removal of the embryo from developing seeds of Pisum sativum,the ‘empty’ ovules (seed coats without enclosedembryo) were filled with a solution (pH 5.5) containing mannitol(usually 400 mM) to which various salts were added. A solutioncontaining two isotopes ((a) [2H]-sucrose/[–14C]aminoisobutyricacid (AIB) or (b) [3H]valine/[14C]asparagine mixture) was administeredto the plant via the petiole subtending the fruiting node, and[2H]solute and [14C]solute unloading from the seed coat wasmeasured, in pulse-labelling experiments of about 5 h. The presenceof 25 or 50 mM K+ in the ‘empty’ ovule enhancedthe release of sucrose from the seed coat particularly duringthe first hours of the experiment, but the stimulating effectof K+ on the release of labelled solutes derived from aminoacids was much smaller. The presence of 25 mM CaCl2 did notaffect the release of sucrose or amino acids from the seed coat.The effect of K+ on sucrose and amino acid release is explainedas an inhibition of sucrose and amino acid resorption from theseed coat apoplast into seed coat cells, after unloading fromthe seed coat unloading sites. It is suggested that amino acidrelease is much less affected by K+ than sucrose release, becausefar less resorption of amino acids by seed coat parenchyma cellstakes place during amino acid transport into the seed coat cavity. Sugar efflux (sucrose + glucose) from attached seed coats ofGlycine max (L.) Merr. was measured at high sampling rates toimprove the kinetic characterization of seed coat exudation.This study confirms that sugar efflux in seed coats has at leasttwo components, and demonstrates that the concentration of mannitolosmoticum bathing the seed coat may influence one or both ofthese components. High leaf irradiance increased sugar effluxrelative to a low leaf irradiance at the same mannitol concentration.A high concentration of mannitol (500 mol m3) enhanced sugarefflux relative to a medium concentration (100 mol m3) underboth high and low leaf irradiance. A low mannitol concentration(10 mol m3) stimulated sugar efflux (relative to 100 mol m3)to a greater extent when leaf irradiance was high. Rapid changesin mannitol concentration produced immediate stimulations ofsugar efflux. Effects of osmoticum on sugar efflux are explainedby simultaneous turgor-mediated effects on import of sucroseby the phloem and retrieval of apoplastic sucrose, presumablyby seed coat parenchyma.  相似文献

Glutamine release from astrocytes is an essential part of the glutamate-glutamine cycle in the brain. Uptake of glutamine into cultured rat astrocytes occurs by at least four different routes. In agreement with earlier studies, a significant contribution of amino acid transport systems ASC, A, L, and N was detected. It has not been determined whether these systems are also involved in glutamine efflux or whether specific efflux transporters exist. We show here that ASCT2, a variant of transport system ASC, is strongly expressed in rat astroglia-rich primary cultures but not in neuron-rich primary cultures. The amino acid sequence of rat astroglial ASCT2 is 83% identical to that of mouse ASCT2. In Xenopus laevis oocytes expressing rat ASCT2, we observed high-affinity uptake of [U-14C]glutamine (Km = 70 microM) that was Na(+)-dependent, concentrative, and unaffected by membrane depolarization. When oocytes were preloaded with [U-14C]glutamine, no glutamine efflux was detected in the absence of extracellular amino acids. Neither lowering intracellular pH nor raising the temperature elicited efflux. However, addition of 0.1 mM unlabeled alanine, serine, cysteine, threonine, glutamine, or leucine to the extracellular solution resulted in a rapid release of glutamine from the ASCT2-expressing oocytes. Amino acids that are not recognized as substrates by ASCT2 were ineffective in this role. Extracellular glutamate stimulated glutamine release weakly at pH 7.5 but was more effective on lowering pH to 5.5, consistent with the pH dependence of ASCT2 affinity for glutamate. Our findings suggest a significant role of ASCT2 in glutamine efflux from astrocytes by obligatory exchange with extracellular amino acids. However, the relative contribution of this pathway to glutamine release from cells in vivo or in vitro remains to be determined.  相似文献

Seed coat exudatesof both species were collected during relatively long experiments(up to about 12 h) and the concentration of sugar (mainly sucrose),amino acids and phosphate in the exudate measured. A discussionis presented on the amino acid/sugar ratio and the phosphate/sugarratio in the seed coat exudate. A pretreatment (15 min) withp-chloromercuribenzenesulphonic acid (PCMBS) reduced the releaseof sugar, amino acids and phosphate from broad bean seed coats.After excision of ‘empty’ ovules of Vicia faba andPisum sativum from the maternal plant, 2–4 h after thistreatment a strong difference became visible between sucroserelease from excised seed coats and sucrose release from attachedseed coats. Similarly, when the rate of phloem transport ofsucrose into an ‘empty’ ovule of Vicia faba or Pisumsativum was reduced by a sub-optimal mannitol concentrationin the solution, a reduced rate of sugar release from the seedcoat could be observed. Excision and treatment with a sub-optimalmannitol concentration reduced the release of amino acids toa lesser extent than for sucrose. The inhibition of macromolecular synthesis in Escherichia coli by ionizing radiation has been investigated. The survival of the ability to incorporate arginine, leucine, isoleucine, histidine, uracil, and glucose after various doses of gamma radiation, deuteron and alpha particle bombardment has been measured. All amino acids are incorporated by processes which show the same radiation sensitivity. The sensitivity of uracil corresponds to a volume which is roughly spherical, of radius about 160A, whereas the amino acids possess sensitive regions which are long and thin in character. The uptake of glucose is concerned with a smaller, roughly spherical unit. The possible identification of the radiation-sensitive targets with cellular constituents is discussed. The long thin character observed for amino acids suggests that the sensitive region affected by radiation is an unfolded form of a ribosome, or alternatively a long nucleic acid molecule. For uracil the sensitive region fits with a 70S ribosome, while for glucose a smaller particle would fit the data.  相似文献

Energetics of Amino Acid Uptake by Vicia faba Leaf Tissues   总被引:2,自引:5,他引:2  
The uptake of [U-14C]threonine and of (α-14C]aminoisobutyrate (α-AIB) by Vicia faba leaf discs is strongly pH dependent (optimum: pH 4.0) and exhibits biphasic saturation kinetics. Kinetics of α-AIB uptake at different pH values indicate that acidic pH values decrease the Km of the carriers while the maximal velocity remains nearly unaffected. Similar results were obtained for both system 1 (from 0.5 to 5 millimolar) and system 2 (from 20 to 100 millimolar).

After addition of amino acids to a medium containing leaf fragments, alkalinizations depending both on the amino acid added and on its concentration have been recorded.

The effects of compounds which increase (fusicoccin) or decrease (uncouplers, ATPase inhibitors, high KCl concentrations) the protonmotive force were studied both on the acidification of the medium and on amino acid uptake by the tissues. There is a close relationship between the time required for the effect of these compounds on the acidification and that needed for inhibition of uptake.

Studies with thiol inhibitors show that 0.1 millimolar N-ethylmaleimide preferentially inhibits uptake by the mesophyll whereas 0.1 millimolar parachloromercuribenzenesulfonate affects rather uptake by the veins.

New evidence was found which added to the electrophysiological data already supporting the occurrence of proton amino acid symport in leaf tissues, particularly in the veins.

Excised seed-coat halves and cotyledons of developing seedsof Pisum sativum L. were incubated in a bathing medium (pH 5·5),in order to measure the release or uptake of sucrose and aminoacids. Net efflux of sucrose and amino acids was reduced bya 250 mol m –3 mannitol solution and a 400 mol m –3solution, in comparison with a 100 mol m–3 control. Thiseffect could not be observed in the case of the amino acid analogue-aminoisobutyric acid (AIB). Net uptake of labelled sucroseor valine by cotyledons and seed coats was enhanced by a highosmolality of the bathing medium. The data on AIB and the datafrom uptake experiments support the view that net efflux ofassimilates is reduced by a high solute concentration in theapoplast (e.g. 400 mol m–3 mannitol), via a stimulationof carrier-mediated sucrose and amino acid uptake into cotyledonaryand seed coat tissues. In experiments with attached empty ovulesof pea in a very early stage of development, sugar release fromthe seed coat was enhanced by a low osmolality of the apoplastsolution (e.g. 100 mol m–3 mannitol, in comparison witha 400 mol m –3 control). This paradoxical effect may beobserved when the stimulatory effect on net assimilate effluxfrom seed coat tissues is exceeding the inhibitory effect onassimilate import into the seed coat. A fast uptake of the preservative benzoic acid was observed in Saccharomyces cerevisiae, reaching saturation in about two min and then remaining constant at this level. The strong dependence of benzoic acid uptake on pH was due to the relative distribution of molecular and ionic forms in solution and not to the pH itself. The molecular form was the only one taken up by the cells. The specificity of the uptake mechanism was evidenced by the pattern of irreversible heat inactivation of the uptake system resembling protein denaturation by heat. Furthermore, the effect of temperature on the uptake was similar to that observed in enzymic reactions, whereas the kinetic data of uptake conformed to the Michaelis-Menten curve of saturation with a Km of 1.54 X 10(-2) M and Vmax of 3 X 10(-3) M/10s. The evidence presented in this paper indicates that compounds of protein nature are involved in the uptake of this preservative.  相似文献

Intact seeds of P. sativum and P. elatius leaked electrolytes,sugars and proteins for as long as 48 h after the beginningof imbibition. Initial leakage was higher at 25 °C thanat 5 °C, but its extent was much greater at the lower temperature.Transfer of seed from 5 °C to 25 °C after 5 h imbibitionresulted in leakage for 48 h at the initial rate at 5 °C.The transfer from 25 °C to 5 °C gave leakage at a rateequivalent to the initial rate at 25 °C. The results arediscussed in relation to behaviour of membranes and to the sensingof the initial temperature. Apical shoot of pea seedling contains two immunochemically distinctspecies of phytochrome, type I (PI) and type II (PII) [Abe etal. (1985) Plant Cell Physiol. 26: 1387-1399]. A PII samplewas prepared from crude extracts from apical shoot of 7-day-old,light-grown pea (Pisum sativum L. cv. Alaska) seedlings, usingbrushite, DEAE-agarose and an agarose linked monoclonal anti-peaPII antibody (mAPll) column chromatography. More than 75% ofthe PII sample consisted of one major 115 kDa chromopeptide,and less than 25% of PII was composed of two minor non-chromophoric49 and 48 kDa fragments, as judged by SDS-polyacrylamide gelelectrophoresis and zinc-induced chromophore fluorescence analysis.Limited proteolysis of PII was performed with either trypsinor Staphylococcus aureus V8 protease, and the resulting fragmentswere separated by SDS-polyacrylamide gel electrophoresis andblotted electrophoretically onto a polyvinylidene difluoridemembrane. Amino-terminal amino acid sequences of the four fragmentsof PII cut from the membrane were determined by a gas-phasesequencer. The sequence of the determined regions (76 aminoacid residues in total), reveals that PII was 63% homologouswith PI. Hence the primary structure of PII is distinct fromthat of PI. Treatment ofNeurospora crassamycelia with cupric ion has been shown to permeabilize the plasma and mitochondrial membranes. Permeabilized mycelia were shown to take up arginine into the vacuoles. Uptake was ATP-independent and appeared to be driven by an existing K+-gradient. The kinetic characteristics of the observed uptake were similar to those observed using vacuolar membrane vesicles: theKmfor arginine uptake was found to be 4.2–4.5 mM. Permeabilized mycelia were used to study the regulation of arginine uptake into vacuoles. The results suggest that uptake is relatively indifferent to the contents of the vacuoles and is not affected by growth of mycelia in amino acid-supplemented medium. Efflux of arginine, lysine, and ornithine from vacuoles was also measured using mycelia permeabilized with cupric ion. Arginine release was shown to be specifically enhanced by cytosolic ornithine and/or increases in the vacuolar pool of arginine or ornithine. Lysine efflux was shown be indifferent to the presence of other amino acids. These observations emphasize the importance of vacuolar compartmentation in controlling arginine and ornithine metabolism and suggest that vacuolar compartmentation may play an important role in nitrogen homeostasis of filamentous fungi.  相似文献

Large doses of γ-irradiation, given to air-dried pea seeds, inhibit the endomitotic DNA synthesis in pea epicotyls during germination in darkness. The cortex cells of the etiolated epicotyls reach only the 4 C DNA level, whereas cortex cells of unirradiated seeds reach the 8 C DNA level. The distribution and degradation of I-14C-ABA in apple seedsduring stratification was investigated. It was found that totalradioactivity of labelled ABA present in the apple seed after48 h of soaking in I-14C-ABA solution decreased gradually duringthe stratification period. The decrease of radioactivity wasobserved in seed coats and endosperm with the simultaneous considerableincrease in the cotyledon and embryo axis. Decarboxylation ofI-14C-ABA was demonstrated and it is postulated as a mode ofABA degradation in apple seed.  相似文献

The embryo-transplantation technique has been used to studythe behaviour of Triticum embryos and endosperm during the courseof ageing of 1-, 2-, 3-, 4-, and 5-year-old seeds. The resultsshow that the ageing process in Triticum seeds is a progressivephenomenon. It affects both embryo and endosperm as shown by(a) the behaviour of old embryos transplanted on young endosperms,and (b) the deleterious effects of aged endosperms on youngembryos.  相似文献
