Recent studies indicate that toll-like receptors (TLRs) are expressed on T cells and that these receptors directly or indirectly activate the adaptive immune system. We have shown previously that acute alcohol/ethanol (EtOH) intoxication combined with burn injury suppresses mesenteric lymph node (MLN) T-cell interleukin-2 (IL-2) and interferon γ (IFN-γ) production. We examined whether direct stimulation of T cells with TLR2, 4, 5 and 7 agonists modulates CD3-mediated T-cell IL-2/IFN-γ release following EtOH and burn injury. Male mice were gavaged with EtOH (2.9 gm/kg) 4 h prior to receiving an ~12.5% total body surface area sham or full-thickness burn injury. Animals were killed on d 1 after injury and T cells were purified from MLN and spleens. T cells were cultured with plate-bound anti-CD3 in the presence or absence of various TLR ligands. Although TLR2, 4 and 5 agonists potentiate anti-CD3-dependent IFN-γ by T cells, the TLR2 agonist alone induced IFN-γ production independent of CD3 stimulation. Furthermore, T cells were treated with inhibitors of myeloid differentiation primary response protein 88 (MyD88), TIR domain-containing adaptor protein (TIRAP), p38 and/or extracellular signal-regulated kinase (ERK) to determine the mechanism by which TLR2 mediates IL-2/IFN-γ production. IL-2 was not influenced by TLR agonists. MyD88 and TIRAP inhibitory peptides dose-dependently diminished the ability of T cells to release IFN-γ. p38 and ERK inhibitors also abolished TLR2-mediated T-cell IFN-γ. Together, our findings suggest that TLR2 directly modulates T-cell IFN-γ production following EtOH and burn injury, independent of antigen-presenting cells. Furthermore, we demonstrated that MyD88/TIRAP-dependent p38/ERK activation is critical to TLR2-mediated T-cell IFN-γ release following EtOH and burn injury.  相似文献

Lipid droplets (LDs) and peroxisomes are central players in cellular lipid homeostasis: some of their main functions are to control the metabolic flux and availability of fatty acids (LDs and peroxisomes) as well as of sterols (LDs). Both fatty acids and sterols serve multiple functions in the cell—as membrane stabilizers affecting membrane fluidity, as crucial structural elements of membrane-forming phospholipids and sphingolipids, as protein modifiers and signaling molecules, and last but not least, as a rich carbon and energy source. In addition, peroxisomes harbor enzymes of the malic acid shunt, which is indispensable to regenerate oxaloacetate for gluconeogenesis, thus allowing yeast cells to generate sugars from fatty acids or nonfermentable carbon sources. Therefore, failure of LD and peroxisome biogenesis and function are likely to lead to deregulated lipid fluxes and disrupted energy homeostasis with detrimental consequences for the cell. These pathological consequences of LD and peroxisome failure have indeed sparked great biomedical interest in understanding the biogenesis of these organelles, their functional roles in lipid homeostasis, interaction with cellular metabolism and other organelles, as well as their regulation, turnover, and inheritance. These questions are particularly burning in view of the pandemic development of lipid-associated disorders worldwide.WORK for the past five decades on the yeast Saccharomyces cerevisiae has contributed fundamental insight into peroxisome biogenesis and function that is also relevant for mammalian cells. While LD research in yeast is still in its infancy and looks back to a much shorter history—the previous edition of YeastBook did not even mention LDs as an “organelle”—combined biochemical, cell biological, lipidomic, and proteomic studies in recent years have already contributed significant insight into LD biogenesis and function.  相似文献

Inflammation, proliferation, and tissue remodeling are essential steps for wound healing. The hypoxic wound microenvironment promotes cell migration through a hypoxia—heat shock protein 90 alpha (Hsp90α)—low density lipoprotein receptor-related protein-1 (LRP-1) autocrine loop. To elucidate the role of this autocrine loop on burn wound healing, we investigated the expression profile of Hsp90α at the edge of burn wounds and found a transient increase in both mRNA and protein levels. Experiments performed with a human keratinocyte cell line—HaCaT also confirmed above results. 17-dimethylaminoethylamino-17demethoxygeldanamycin hydrochloride (17-DMAG), an Hsp90α inhibitor, was used to further evaluate the function of Hsp90α in wound healing. Consistently, topical application of Hsp90α in the early stage of deep second-degree burn wounds led to reduced inflammation and increased tissue granulation, with a concomitant reduction in the size of the wound at each time point tested (p<0.05). Consequently, epidermal cells at the wound margin progressed more rapidly causing an expedited healing process. In conclusion, these results provided a rationale for the therapeutic effect of Hsp90α on the burn wound management.  相似文献

This study extends existing life cycle assessment (LCA) literature by assessing seven environmental burdens and an overall monetized environmental score for eight recycle, bury, or burn options to manage clean wood wastes generated at construction and demolition activity sites. The study assesses direct environmental impacts along with substitution effects from displacing fossil fuels and managed forest wood sourcing activities. Follow‐on effects on forest carbon stocks, land use, and fuel markets are not assessed. Sensitivity analysis addresses landfill carbon storage and biodegradation rates, atmospheric emissions controls, displaced fuel types, and two alternative carbon accounting methods commonly used for waste management LCAs. Base‐case carbon accounting considers emissions and uptakes of all biogenic and fossil carbon compounds, including biogenic carbon dioxide. Base‐case results show that recycling options (recycling into reconstituted wood products or into wood pulp for papermaking) rank better than all burning or burying options for overall monetized score as well as for climate impacts, except that wood substitution for coal in industrial boilers is slightly better than recycling for the climate. Wood substitution for natural gas boiler fuel has the highest environmental impacts. Sensitivity analysis shows the overall monetized score rankings for recycling options to be robust except for the carbon accounting method, for which all options are highly sensitive. Under one of the alternative methods, wood substitution for coal boiler fuel and landfill options with high methane capture efficiency are the best for the overall score; recycling options are next to the worst. Under the other accounting alternative, wood substitution for coal and waste‐to‐energy are the best, followed by recycling options.  相似文献
