Measurements of chlorophyll fluorescence and hyperspectral reflectance were used to detect salinity stress in Suaeda salsa L., beach of Dongtai, Jiangsu Province, China. Three experimental sites were used in our study, which belong to low salinity, middle salinity and high salinity. The results showed that leaf chlorophyll fluorescence changed along salinity gradient. To select the sensitive hyperspectral ranges of leaf chlorophyll fluorescence, the correlationship between leaf chlorophyll fluorescence and hyperspectral reflectance was regressed and analyzed. Statistical results indicated that the 680 and 935 nm were the most sensitive hyperspectral bands for estimating leaf chlorophyll fluorescence. Then, 11 relative hyperspectral indices were selected based on the sensitive bands and previous literature. (R 680 − R 935)/(R 680 + R 935) and R 680/R 935 have higher correlationship coefficient (R) and lower root mean square error, may be used for detecting chlorophyll fluorescence, such as F o, F m, F v/F m, qP, and ΦPSII, while NPQ may be detected by (R 780 − R 710)/(R 780 − R 680). These results suggest that chlorophyll fluorescence of halophyte response to salinity stress could be identified by remote sensing.  相似文献

We studied the dominant environmental factors that affect the gas-exchange characteristics and water potential (ψ) of broad-ovate leaves of Populus euphratica Oliv. in extreme arid area of Tarim River, China, and their correlation to water status of P. euphratica by analyzing on-field monitored meteorological data, soil moisture and salinity, P. euphratica leaf gas exchange and ψ, and revealed the indicative threshold of environmental factors for P. euphratica leaf water changes and growth. The results indicated that meteorological factors such as air relative humidity (RH), air temperature (T air), etc. are the dominant factors. The threshold value of RH is (48.19 ± 1.06)% for P. euphratica growth, i.e. RH from 10.69% to 48.19% is suitable for P. euphratica growth in extremely arid region of Tarim River. This study provides a theoretical basis for reducing drought damage to P. euphratica and maintaining normal growth of P. euphratica by in-time watering.  相似文献

In vitro regeneration of black nightshade (Solanum nigrum L.) plants was achieved through callus-mediated shoot organogenesis followed by 30 d indoor ex vitro adaptation to nutritional stress under environmental ambience and thereafter 6-d outdoor acclimatization in pots prior to field establishment. Relevant physiological parameters including pigment content, chlorophyll a fluorescence, net photosynthetic rate (P N), transpiration rate (E), and stomatal conductance (g s) of in vitro-regenerated plants were investigated during the course of ex vitro adaptation. During the first 4 d of indoor transplantation to potting substrate, there was a marginal reduction in the leaf chlorophyll and carotenoid contents but P N and E were strongly reduced. The stomatal conductance and E/P N ratio were significantly higher in plants up to 20 d of indoor adaptation than those of comparable age grown naturally from seeds. The shape of the OJIP fluorescence transient varied significantly with acclimatization, and the maximum change was observed at 2.0 ms. The 2.0 ms variable fluorescence (V j), 30 ms relative fluorescence (M 0), photon trapping probability (TR0/Abs), and photosystem II (PSII) trapping rate (TR0/RC) showed initial disturbance and subsequent stabilization during 30 d of indoor acclimatization. Energy dissipation (DI0/RC) and electron transport probability (ET0/TR0) showed an initial phase of increase during the 4 d after plants were transplanted outdoors. During the 6-d outdoor acclimatization after transfer of plants to soil, no significant change in total chlorophylls and carotenoids, E, and g s were observed, but P N improved after reduction on the first d. The OJIP-derived parameters experienced change on the first d but were stabilized quickly thereafter. There was no significant difference between outdoor acclimatized plants and those of the seed-grown plants of comparable age with respect to photosynthetic and fluorescence parameters. Direct transfer of plants without indoor acclimatization, however, showed a completely different trend with respect to P N, E, and OJIP fluorescence transients. The bearing of this study on optimizing micropropagation is discussed.  相似文献

Direct genetic transformation of mesophyll protoplasts was studied in Pelargonium × hortorum. Calcein and green-fluorescent protein (GFP) gene were used to set up the process. Electroporation (three electric pulses from a 33-μF capacitor in a 250-V cm−1 electric field) was more efficient than PEG 6000 for membrane permeation, protoplast survival and cell division. Transient expression of GFP was detected in 33–36% of electroporated protoplasts after 2 days and further in colonies. A protoplast suspension conductivity of >1,500 μS cm−1 allowed high colony formation and plant regeneration. Stable transformation was obtained using the plasmid FAJ3000 containing uidA and nptII genes. When selection (50 mg l−1 kanamycin) was achieved 6 weeks after electroporation, regenerated shoots were able to grow and root on 100 mg l−1 kanamycin. The maximum transformation efficiency was 4.5%, based on the number of colonies producing kanamycin-resistant rooted plants or 0.7% based on the number of cultured protoplasts. Polymerase chain reaction (PCR) analysis on in vitro micropropagated plants showed that 18 clones out of 20 contained the nptII gene, while the uidA gene was absent. These results were confirmed after PCR analyses of five glasshouse-acclimatized clones.  相似文献

For centuries in Asia, Rhodiola coccinea has been used as a valuable adaptogen medicinal plant known for its remarkable resistance to various stress factors. Physiological tolerance in chloroplasts is believed to be an important factor, which affects the geographic distribution of plant species. However, there is currently no report on how the physiological tolerance in the chloroplasts of R. coccinea varies along an altitudinal gradient in a semi-arid mountain region. In this work, we investigated the physiological variations of the chloroplasts of R. coccinea plants along an altitudinal gradient in the Tianshan Mountains, which are located in northwest China. It was found that the physiological parameters of chloroplasts in R. coccinea plants, including superoxide generation, fatty acid compositions, thylakoid membrane fluidity, chlorophyll a/b ratios and photosynthetic electron transport rates (ETRs), varied nonlinearly with altitude. Indeed, an inflection point was observed at approximately 3,800 m. For altitudes were higher or lower than 3,800 m, the ETR, index of unsaturated fatty acids in the thylakoid membrane and Chl a/b ratios decreased with increasing altitude, whereas the superoxide generation and DPH polarisation of thylakoid membranes increased. In addition, variations in the AsA and GSH contents of chloroplasts could be divided into two distinct phases along the altitudinal gradient. The AsA content slowly decreased with increasing altitudes up to 3,800 m and rapidly decreased above 3,800 m. However, there was a gradual increase in the GSH content between 3,570 and 3,750 m, followed by an abrupt rise to a plateau level. These results demonstrate that the physiological tolerance of chloroplasts widely varies with altitude and have a tight relationship with the range of growth altitude of R. coccinea in Tianshan Mountains.  相似文献

Pineapple is one of the most important tropical fruits, but the availability of planting material is insufficient to agricultural demands. Therefore, several pineapple micropropagation protocols have been developed. However, acclimatization of in vitro plants continues to take a prolonged period. Biofertilizers have been found as safe alternatives to improve the agricultural performances of many crops. This study highlights some of the effects of the application of Azotobacter chroococcum (INIFAT5 strain) on in vitro pineapple plants during acclimatization. The bacteria were sprayed immediately after transplanting to the ex vitro environment; the plants were then sprayed every 4 wk. A control group of plants was established. Subsequently, after 5 mo, the evaluated variables included fresh and dry plant weight, plant height (cm), and root length (cm). The anatomy of middle-aged leaves and roots was also studied: transversal thickness and width of cuticle, epidermis, hypodermis, aquiferous parenchyma, and photosynthetic parenchyma. Thickness of root exoderm, external cortex, internal cortex, and stele were also evaluated. In general, the INIFAT5 strain improved the plant development. Results showed that the bacteria significantly provoked changes in the plant fresh weight, the thickness of the leaf abaxial and adaxial cuticles, and the root exoderm width. Contrastingly, A. chroococcum did not affect the thickness of the leaf photosynthetic parenchyma.  相似文献

The hydrozoan Cordylophora caspia, with its origin in the Caspian and Black Seas, is considered an invasive biofouling species to power stations and water treatment plants in Brazil, causing economic and ecological impacts. Tolerance of the species to variation in pH was tested to evaluate its resistance to acidic and basic pH levels. Values of pH were tested over a range from 4.0 to 10.0. The colonies were exposed for 6 h to each treatment, using phosphoric acid and NaOH to set water pH to the desired acidic or basic levels, respectively. Colonies of C. caspia displayed 100% mortality only at pH 4 and 10, and showed high survival rates (>50%) within the pH range of 5.0–8.5. The species demonstrated tolerance to a wide range of pH levels, and the results will help in the development of techniques to reduce biofouling by this species. Treatments that keep the pH below 5 or above 8.5 for at least 6 h are suggested to control and eradicate C. caspia where fouling causes significant economic impacts.  相似文献

FBP21 is one of the SOC1-like genes isolated from Petunia hybrida. Based on sequence analysis, FPB21 is suggested to have a role in promoting flowering. In this study, FBP21 was expressed in a tobacco host plant under the control of the CaMV 35S promoter. Our results showed that the transgene accelerated flowering, i.e. the transgenic plants flowered just 3 months after germination, in comparison to the wild-type tobacco which flowered after 5 months. Plant morphology was also affected, with the transgenic tobacco plants developing at least five robust lateral branches, while the control plants generally had just three. Total leaf area was significantly reduced in the transgenic tobacco compared to wild-type tobacco. By contrast, there was no significant difference between transgenic and control plants for the total number of flowers or fruits. Thus, the flower or fruit yield expressed per unit leaf area was higher in transgenic tobacco than in wild-type plants. Semi-quantitative RT-PCR analysis indicated that overexpression of FBP21 in tobacco resulted in the up-regulation of some flowering-related genes. The results of this study in tobacco indicate that the Petunia FBP21 gene may permit the engineering of early-flowering and short-growth habits without compromising flower or fruit yields.  相似文献

This study was conducted to evaluate the load of three food- and water-borne parasites, namely, Giardia, Entamoeba, and Cryptosporidium on food items that are consumed either raw or in an unprocessed state in the northern parts of India. A two-step diagnostic method was employed to assess the presence of Giardia and Cryptosporidium, the immunofluorescence assay (IFA) combined with the polymerase chain reactiion (PCR assay), whereas a Tech lab diagnostic kit in combination with PCR assay was used for accurate detection of Entamoeba histolytica in the samples. The methods for isolation and enrichment of cysts/oocysts from the various food items were tested and discussed here. Our results showed that the overall spectrum of incidence of the three parasites on food items in decreasing order were Giardia Entamoeba Cryptosporidium. When data were subjected to the chi-square test, the prevalence of all three parasites was found to be independent of the food items. To determine whether the presence of two types of parasites in a food item is uniform, a Poisson distribution test was conducted. On comparing the intensity of occurrence of the different parasites in various food items, it was observed that the occurrence of Giardia and Entamoeba was not of the same order at 5% level of significance only in case of samples of raw meat and milk. This confirmed that a high number of raw or unpasteurized milk and meat samples are more likely to be contaminated with Giardia than with Entamoeba. Therefore, our observations point to the unhygienic practices of food handlers being a major contributor in the transmission of parasites to unprocessed food products.  相似文献

Despite species in the Rubus fruticosus complex (wild blackberry) being among the most invasive plants globally in regions with large annual fluctuations in water availability, little is known about their water relations. We compared water relations of a prominent member of the complex, R. armeniacus (Himalayan blackberry), with species native to the Pacific Northwest of North America (PNW), R. spectabilis (salmonberry) and R. parviflorus (thimbleberry). In eight stands of each species located near Portland, Oregon, USA, we measured mid-day hydraulic resistance (R plant), and daily time series of stomatal conductance (g s), leaf water potential (Ψlf), and environmental conditions at four time periods spanning the 2007 growing season. Although all species maintained Ψlf above −0.5 MPa in spring, R. armeniacus maintained less negative Ψlf (≥−1.0 MPa) than the natives in summer, a factor attributable to advantages in both its root and shoot systems. R plant of R. armeniacus was ≤0.1 MPa mmol−1 m2 s for the duration of the study, and approximately 25–50% of R plant for the native species in summer. R. armeniacus had higher g s compared to the native species throughout the spring and summer, with approximately twice their rates in summer. Our R plant and g s results show that R. armeniacus has access to more water during PNW summers than congeneric natives, allowing it to maintain high water-use, and potentially helping it achieve higher growth and reproductive rates. Water relations may therefore be a critical component of the competitive and invasive success of R. armeniacus and other R. fruticosus species worldwide.  相似文献

Argas neghmei Kohls & Hoogstraal is a Neotropical tick species parasite of poultry and man in the arid western of the southern cone of America in Argentina and Chile. Males, females and nymphs of an argasid were collected from 20 to 22 of June, 2007 in nests of Creamy-breasted Canastero, Asthenes dorbignyi (Reinchenbach) (Passeriformes: Furnariidae) at about 7 km S of La Poma (24°46′S, 66°12′W) and a male tick at 24°48′S, 66°10′W on Nov 27, 2008, Salta province, Argentina. Scanning electron microscope micrographies and sequences of 16S rDNA confirmed that the ticks found were A. neghmei. Development in breeding areas of A. dorbignyi is probably a source of infestation for animal premises and houses. However, the role of other unknown wild birds may also contribute to the invasion of A. neghmei in chicken houses or residences.  相似文献

A new species, endemic to the Flora Iranica area, Gagea robusta (Liliaceae) is described and illustrated. It differs from G. reticulata (Pall.) Schult. & Schult. f. in having a puberulent indumentum on the basal leaf and from G. setifolia Baker ex Aitch. in having a long neck (up to 12 cm long). Pseudocercospora griseola is the causal agent of angular leaf spot of common bean (ALS). It has undergone parallel coevolution with its host and two major groups have been defined, “Andean” (P. griseola f. griseola) and “Mesoamerican” (P. griseola f. mesoamericana). The aim of this study was to analyze the nature and the level of the dark pigment synthesized by the representatives of each group. After 21 days of incubation on potato dextrose agar medium, P. griseola f. griseola isolate S3b developed colonies with diameters of 17.5 ± 1.3 mm and concentric rings of pigmentation. Isolate T4 of P. griseola f. mesoamericana presented smaller colonies (9.9 ± 0.3 mm) with a uniform dark-gray color. Both isolates, S3b and T4, produced the same pigment, a 1,8-dihydroxynaphthalene-melanin, although different in quantity and structural features as suggested by the IR spectrum. The P. griseola f. griseola isolate S3b had a higher growth rate and melanin content as well as smaller sensitivity to melanin synthesis inhibitors compared to the isolate T4 of P. griseola f. mesoamericana. These results suggest a possible link between melanin and growth in P. griseola.  相似文献

Callus cultures derived from leaf segments of chrysanthemum cultivar ‘Snow Ball’ which was susceptible to Septoria obesa were successfully used for in vitro selection for resistance to this pathogenic fungus. Resistant cell lines were selected by culturing callus on growth medium containing various concentrations of S. obesa filtrate. Resistant calluses obtained after two cycles (30 d each cycle) of selection were used for plant regeneration. About 30% of the plants regenerated from the resistant calluses and 70–80% of the plants raised from cuttings had acquired considerable resistance against the pathogen in the field. No phenotypic variation was observed in the selected regenerates.  相似文献

We performed comparative analyses of four cross-sections of the distal radius and tibia in two species of macaque to clarify the relationships between bone morphology and locomotor type. The lengths of bones and five bone geometric properties in each section were examined and compared separately in both female and male Macaca mulatta and Macaca fascicularis. In M. mulatta, there were no significant gender-specific differences in either the radius or the tibia. In contrast, the radius and tibia of male M. fascicularis had greater geometric parameters in the 20% and 40% positions relative to the 5% and 10% positions from the distal end than those of their female counterparts. The radius and tibia of M. mulatta were relatively longer than those of M. fascicularis, and the sectional parameters of the tibia of M. mulatta were relatively larger than those of M. fascicularis. Standardization of the log-transformed bone length between the species revealed larger radial cortical bone areas in M. fascicularis. In contrast, there were minimal differences in the tibial cortical bone areas between the two species. This study suggests that the observed distinctions in bone geometry in female and male M. fascicularis may be due to gender-specific differences in the muscle weights of the forearm and calf, which may underlie the divergence in the leaping abilities of females and males of this species. Taken together, these results of interspecies comparisons may be related to the fact that arboreal primates such as M. fascicularis undergo compressive mechanical stress due to the forelimb lead that occurs as the animal descends a sloping trunk or bridges a tree gap downward, while terrestrial primates such as M. mulatta move on nearly flat substrates. Differences in fore- and hind-limb bone properties between the two species are discussed with regard to functional morphology and locomotor type.  相似文献

The effect of p-nitrophenol (PNP) concentration with or without glucose and yeast extract on the growth and biodegradative capacity of Ralstonia eutropha was examined. The chemical constituents of the culture medium were modeled using a response surface methodology. The experiments were performed according to the central composite design arrangement considering PNP, glucose and yeast extract as the selected variables whose influences on the degradation was evaluated (shaking in reciprocal mode, temperature of 30°C, pH 7 and test time of about 9 h). Quadratic polynomial regression equations were used to quantitatively explain variations between and within the models (responses: the biodegradation capacity and the biomass formation). The coefficient of determination was high (R adjusted2 = 0.9783), indicating the constructed polynomial model for PNP biodegradative capacity explains the variation between the regressors fairly well. A PNP removal efficiency of 74.5% occurred within 9 h (15 mg/L as the initial concentration of PNP with use of yeast extract at 0.5 g/L).  相似文献

Lactic acid bacteria (LAB) are generally sensitive to hydrogen peroxide (H2O2), Lactobacillus sakei YSI8 is one of the very few LAB strains able to degrade H2O2 through the action of a heme-dependent catalase. Lactobacillus rhamnosus strains are very important probiotic starter cultures in meat product fermentation, but they are deficient in catalase. In this study, the effect of heterologous expression of L. sakei catalase gene katA in L. rhamnosus on its oxidative stress resistance was tested. The recombinant L. rhamnosus AS 1.2466 was able to decompose H2O2 and the catalase activity reached 2.85 μmol H2O2/min/108 c.f.u. Furthermore, the expression of the katA gene in L. rhamnosus conferred enhanced oxidative resistance on the host. The survival ratios after short-term H2O2 challenge were increased 600 and 104-fold at exponential and stationary phase, respectively. Further, viable cells were 100-fold higher in long-term aerated cultures. Simulation experiment demonstrated that both growth and catalase activity of recombinant L. rhamnosus displayed high stability under environmental conditions similar to those encountered during sausage fermentation.  相似文献
