Although roots of species in the Pinaceae are usually colonized by ectomycorrhizal (EM) fungi, there are increasing reports of the presence of arbuscular mycorrhizal (AM) and dark septate endophytic (DSE) fungi in these species. The objective of this study was to determine the colonization patterns in seedlings of three Pinus (pine) species (Pinus banksiana, Pinus strobus, Pinus contorta) and Picea glauca x Picea engelmannii (hybrid spruce) grown in soil collected from a disturbed forest site. Seedlings of all three pine species and hybrid spruce became colonized by EM, AM, and DSE fungi. The dominant EM morphotype belonged to the E-strain category; limited colonization by a Tuber sp. was found on roots of Pinus strobus and an unknown morphotype (cf. SuillusRhizopogon group) with thick, cottony white mycelium was present on short roots of all species. The three fungal categories tended to occupy different niches in a single root system. No correlation was found between the percent root colonized by EM and percent colonization by either AM or DSE, although there was a positive correlation between percent root length colonized by AM and DSE. Hyphae and vesicles were the only AM intracellular structures found in roots of all species; arbuscules were not observed in any roots.

The last lowland locality ofGentiana verna in the Czech Republic is a calcareous grassland near Rovná at Strakonice in South Bohemia. This locality was the subject of a recovery programme that included support of the remaining population by micropropagation. The plants were inoculated with arbuscular mycorrhizal fungi (AMF) after their transfer toex vitro and the effect of AMF on their establishment and survival was studied. Although the conventional method of inoculation ofG. verna using spores or colonized root segments as an inoculum source resulted in no or negligible root colonization, the transplantation of gentians to the locality Rovná was successful and the plants became colonized with AMF very rapidly in the field. Successful mycorrhization of gentians under experimental conditions occurred only via the extraradical mycelial network established by neighbouring mycorrhizal plant species (nurse plant effect). Different nurse plant species formed different morphological types of mycorrhiza when inoculated with the same fungal isolate. Gentians always had theParis type of root colonization with intracellular hyphal loops and swellings. Intercellular hyphae, arbuscules and vesicles were not observed. No evidence for a positive growth response was found inG. verna.

Hyphae and vesicles of arbuscular mycorrhizal fungi (AMF) were found within the decomposing leaves of Myrica parvifolia, M. pubescens and Paepalanthus sp. at three montane sites in Colombia. Hyphae, vesicles, and arbuscule-like structures were also found within scale-like leaves of the rhizomes of Paepalanthus sp. The litter found in the vicinity of the roots was divided into three decomposition layers. The highest AMF colonization occurred in the most decomposed leaves, which were in close association with roots. In contrast, there were no differences in AMF colonization of roots present in the different decomposition layers. Colonization of decomposing leaves by AMF did not differ between the two closely related species M. parvifolia and M. pubescens, nor between two sites (Guatavita and Zipacón, Colombia) differing in soil fertility. Occurrence of vesicles in decomposing leaves was correlated with abundant AMF extraradical hyphae among the leaves. We propose that AMF enter decomposing leaves mechanically through vascular tissue. As a consequence, AMF are well positioned to obtain and efficiently recycle mineral nutrients released by decomposer microorganisms before their loss by leaching or immobilization in soil.

Thlaspi praecox (Brassicaceae) is a recently discovered metal hyperaccumulating plant species colonized by arbuscular mycorrhizal fungi (AMF). The identity and diversity of the AMF colonizing its roots have not been determined so far. Therefore, T. praecox was inoculated with an indigenous fungal mixture from a metal polluted site and grown in original polluted soil/ commercial substrate mixtures (i.e., 100%, 50%, and 25%). Low to moderate mycorrhizal frequencies (F = 33–68%) and only rare arbuscules were observed. Densities of vesicles and microsclerotia, typical structures of dark septate endophytes (DSE), were greater in pots with 100% original polluted soil. In contrast, the highest diversity of fungal genotypes was observed in the roots from 25% polluted soil/ commercial substrate mixture, with the lowest soil concentrations of Cd, Zn, and Pb. The sequences obtained corresponded to Glomus species (Glomeromycota), to putative DSE Phialophora verrucosa and Rhizoctonia sp. and to some other fungi from Asco- and Basidiomycota, that are known to associate with plants, namely Capnobotryella sp., Penicillium brevicompactum, Rodotorula aurantiaca and Rodotorula slooffiae. This is the first report of DSE occurrence in roots of hyperaccumulating T. praecox, a promising candidate for phytoextraction.

Herbivory, competition and soil fertility interactively shape plant communities and exhibit an important role in modifying conditions for host-dependent fungal symbionts. However, field studies on the combined impacts of natural herbivory, competition and soil fertility on root fungal symbionts are rare. We asked how mammalian herbivory, fertilization, liming and plant–plant competition affect the root colonization of arbuscular mycorrhizal fungi (AMF) and dark septate endophytic (DSE) fungi of the dicot herb, Solidago virgaurea. The 2-year full-factorial experiment was conducted in two contrasting habitats: non-acidic and acidic mountain tundra. We found that herbivory increased arbuscular colonization (i.e. the site of resource exchange) at fertile non-acidic sites, where vegetation was rich in species having AMF symbionts, whereas at infertile acidic sites, where plants having AMF symbiont are scarce, the response was the opposite. Herbivory of the host plant negatively affected DSE hyphal and sclerotial colonization in unfertilized plots, possibly due to reduced carbon flow from the host plant while there was no effect of herbivory in fertilized plots. DSE colonization was highest in unfertilized exclosures where soil nutrient concentrations were also lowest. Liming had a negative effect on DSE hyphal colonization, and its effect also interacted with herbivory and the habitat. Biomass removal of the neighboring plants did not affect the root colonization percent of either arbuscules or DSE. Our results show that the impacts of aboveground mammalian herbivory, soil nutrient availability and specific habitat conditions on belowground root fungal symbionts are highly dependent on each other. Arbuscule response to herbivory appeared to be regulated by specific habitat conditions possibly caused by differences in the AMF availability in the soil while DSE response was associated with availability of host-derived carbon. Our result of the relationship between herbivory and soil nutrients suggests an important role of DSE in ecosystem processes.

Here, we examined the colonization by fungal root symbionts in the cultivated Andean grain Chenopodium quinoa and in 12 species that dominate plant communities in the Bolivian Altiplano above 3,700 m elevation and explore for the possible relationships between fungal colonization and fine root proportion. The 12 most abundant species in the study area were consistently colonized by AMF and DSE. In contrast, the annual Andean grain C. quinoa showed negligible or absence of mycorrhizal fungi colonizing roots. On the other hand, C. quinoa, Junelia seriphioides and Chersodoma jodopappa were infected to a varying degree by the root pathogen Olpidium sp. We observed no relationship between AMF and DSE colonization and proportion of fine roots in the root system, but instead, the ratio between DSE and AMF colonization (ratio DSE/AMF) negatively related with proportion of fine roots. Our findings support the hypothesis regarding the importance of DSE at high altitudes and suggest a functional relationship between the rate of DSE/AMF and proportion of fine roots. The colonization by the root pathogen Olpidium sp. in C. quinoa deserves further study since this Andean grain is increasingly important for the local economy in these marginal areas.

As an initial step towards evaluating whether mycorrhizas influence composition and diversity in calcareous fen plant communities, we surveyed root colonization by arbuscular mycorrhizal fungi (AMF) and dark septate endophytic fungi (DSE) in 67 plant species in three different fens in central New York State (USA). We found colonization by AMF and DSE in most plant species at all three sites, with the type and extent of colonization differing between monocots and dicots. On average, AMF colonization was higher in dicots (58±3%, mean±SE) than in monocots (13±4%) but DSE colonization followed the opposite trend (24±3% in monocots and 9±1% in dicots). In sedges and cattails, two monocot families that are often abundant in fens and other wetlands, AMF colonization was usually very low (<10%) in five species and completely absent in seven others. However, DSE colonization in these species was frequently observed. Responses of wetland plants to AMF and DSE are poorly understood, but in the fen communities surveyed, dicots appear to be in a better position to respond to AMF than many of these more abundant monocots (e.g., sedges and cattails). In contrast, these monocots may be more likely to respond to DSE. Future work directed towards understanding the response of these wetland plants to AMF and DSE should provide insight into the roles these fungal symbionts play in influencing diversity in fen plant communities.

A survey was made of the arbuscular mycorrhizal (AM) status of five dominant wild plants Tamarix chinensis, Phragmites communis, Suaeda glauca, Aeluropus littoralis var. sinensis and Cirsium setosum in saline-alkaline soils of the Yellow River Delta that show low plant diversity. All of the species were colonized and showed typical AM structures (arbuscules, vesicles). The colonization percentage ranged from 0.2% to 9.5%, where C. setosum was the highest. The species richness of AMF at the different sites ranged from 2.00 to 2.40 per 50 ml soil, with an average of 2.16. Species diversity ranged from 1.99 to 2.22 per 50 ml soil, with an average of 2.13. Spore density ranged from 3 to 30 per 50 ml soil, with an average of 12. Glomus was the dominant genus, with a frequency and relative abundance of 88.1% and 68.4%, respectively. G. caledonium, with a frequency and relative abundance of 15.0% and 4.6%, respectively, was the dominant species. Differences were also observed in the distribution of AMF in different soil layers. Although there were still AM fungal spores in the layer 40 cm below the surface, most spores were found at a depth of 0–40 cm.

Arbuscular mycorrhizal fungi (AMF) form symbiotic relationships with most vascular plants including some gymnosperm species. Although species in the gymnosperm family Pinaceae normally develop ectomycorrhizal associations, AMF hyphae and vesicles, typical of members of the Suborder Glomineae, have been reported in the roots of some Pinaceae species. However, it is not known whether AMF belonging to various species and suborders are able to colonize roots of Pinaceae species and to what extent this influences the performance of Pinaceae seedlings. We tested in each of the Glomaceae, Acaulosporaceae and Gigasporineae AMF families two species for their ability to colonize and affect the growth of Pinus strobus (eastern white pine) in the presence or absence of an AMF host plant (Trifolium pretense—red clover). Glomus intraradices was the only AMF that colonized pine roots, predominantly in the presence of clover, forming intracellular hyphae and vesicles but not arbuscules. Colonization, however, did not relate to increased pine biomass and the overall presence of AMF, regardless of colonization abilities, resulted in a biomass reduction. This effect on pine seedling biomass was explained by the AMF family to which the AMF belonged, indicating that the effects of AMF on the non-host pine may be related to phylogeny. Acaulosporaceae species reduced pine biomass the most whereas, Gigasporineae species had the smallest effect on biomass. These preliminary results suggest that AMF may affect the soil microflora differently among AMF families in previously unsuspected ways with potential consequences for non-AMF host growth.

Rising atmospheric carbon dioxide partial pressure (pCO2) and nitrogen (N) deposition are important components of global environmental change. In the Swiss free air carbon dioxide enrichment (FACE) experiment, the effect of altered atmospheric pCO2 (35 vs. 60 Pa) and the influence of two different N‐fertilization regimes (14 vs. 56 g N m?2 a?1) on root colonization by arbuscular mycorrhizal fungi (AMF) and other fungi (non‐AMF) of Lolium perenne and Trifolium repens were studied. Plants were grown in permanent monoculture plots, and fumigated during the growth period for 7 years. At elevated pCO2 AMF and non‐AMF root colonization was generally increased in both plant species, with significant effects on colonization intensity and on hyphal and non‐AMF colonization. The CO2 effect on arbuscules was marginally significant (P=0.076). Moreover, the number of small AMF spores (≤100 μm) in the soils of monocultures (at low‐N fertilization) of both plant species was significantly increased, whereas that of large spores (>100 μm) was increased only in L. perenne plots. N fertilization resulted in a significant decrease of root colonization in L. perenne, including the AMF parameters, hyphae, arbuscules, vesicles and intensity, but not in T. repens. This phenomenon was probably caused by different C‐sink limitations of grass and legume. Lacking effects of CO2 fumigation on intraradical AMF structures (under high‐N fertilization) and no response to N fertilization of arbuscules, vesicles and colonization intensity suggest that the function of AMF in T. repens was non‐nutritional. In L. perenne, however, AM symbiosis may have amended N nutrition, because all root colonization parameters were significantly increased under low‐N fertilization, whereas under high‐N fertilization only vesicle colonization was increased. Commonly observed P‐nutritional benefits from AMF appeared to be absent under the phosphorus‐rich soil conditions of our field experiment. We hypothesize that in well‐fertilized agricultural ecosystems, grasses benefit from improved N nutrition and legumes benefit from increased protection against pathogens and/or herbivores. This is different from what is expected in nutritionally limited plant communities.

AMF (arbuscular mycorrhizal fungi) colonization of the grass chalk false-brome (Brachypodium pinnatum (L.) P. B.) was studied in selected habitats under spatially different light regimes: (a) shade condition under oak trees, (b) half shade in a shrubby area and (c) full-sun conditions on unshaded grassland. This study assessed the variations in AMF colonization of the grass dependent on the light supply in field habitats. Soil, root and shoot samples were collected four times during the vegetation period (in June, July, September and October). Root colonization, root and shoot biomass as well as soil water content were determined. The highest rate of AMF colonization was detected in June under half-sun and full-sun conditions, where about 50% of the roots were colonized. The average amount of arbuscules was less than 20% in the roots at the three sites, with the highest number of arbuscules in June, under half-sun and full-sun conditions, however, not under the trees. Overall, best mycorrhizal colonization occurred during summer, and its rate decreased in autumn. This tendency inversely correlated with the amount of precipitation, and thus with the water content of soils. The high colonization rate of the examined root samples, and also its seasonal fluctuation, might reflect the importance of the symbiosis where inorganic nutrients and water are the growth-limiting factors. The marginal AMF colonization of chalk false-brome under shade conditions indicates that plants do not use AMF under all stress conditions. When low light limits photosynthesis and thus growth of the plants, they dispense with the colonization of AMF in order to save the expenditure of organic carbon.

Anaerobic microbial activity in northern peat soils most often results in more carbon dioxide (CO 2 ) production than methane (CH4) production. This study examined why methanogenic conditions (i.e., equal molar amounts of CH4 production and CO2 production) prevail so infrequently. We used peat soils from two ombrotrophic bogs and from two rheotrophic fens. The former two represented a relatively dry bog hummock and a wet bog hollow, and the latter two represented a forested fen and a sedge-dominated fen. We quantified gas production rates in soil samples incubated in vitro with and without added metabolic substrates (glucose, ethanol, H2/CO2). None of the peat soils exhibited methanogenic conditions when incubated in vitro for a short time (< 5 days) and without added substrates. Incubating some samples > 50 days without added substrates led to methanogenic conditions in only one of four experiments. The anaerobic CO2:CH4 production ratio ranged from 5:1 to 40:1 in peat soil without additions and was larger in samples from the dry bog hummock and forested fen than the wet bog hollow and sedge fen. Adding ethanol or glucose separately to peat soils led to methanogenic conditions within 5 days after the addition by stimulating rates of CH4 production, suggesting CH4 production from both hydrogenotrophic and acetoclastic methanogenesis. Our results suggest that methanogenic conditions in peat soils rely on a constant supply of easily decomposable metabolic substrates. Sample handling and incubation procedures might obscure methanogenic conditions in peat soil incubated in vitro.

Arbuscular mycorrhizal and dark septate endophyte associations of 31 medicinal plant species collected from the Garden of Medicinal Plants of the Faculty of Pharmacy, Jagiellonian University, Collegium Medicum in Kraków were investigated. Arbuscular mycorrhiza (AM) was found in 30 species; 23 were of the Arum-type, 5—Paris and 2 taxa revealed intermediate morphology. Many plants were strongly colonized by arbuscular mycorrhizal fungi (AMF). The mycelium of dark septate endophytes (DSE) was observed in 21 taxa. However, the percentage of root colonization by these fungi was low. Spores of 15 species of AMF (Glomeromycota) were found in the rhizosphere of the investigated plants. Our results are the first detailed report of both AMF and DSE associations of these plant species. The use of AMF and DSE during the process of medicinal plant cultivation for pharmaceutical purposes is discussed.

Human alteration of symbiont genetics among aboveground endophytic Epichloë coenophiala strains within tall fescue (Schedonorus arundinaceus) has led to widespread deployment of novel grass-endophyte combinations, yet little is known about their ecological consequences. In this study, clone pairs (endophyte-infected, endophyte-free) of two tall fescue genotypes received factorial combinations of increased temperature (+3 °C) and precipitation (+30% long-term annual mean) for 2 yr. We measured root arbuscular mycorrhizal fungi (AMF), dark septate endophyte (DSE) colonization, and soil AMF extraradical hyphae (ERH) length. We hypothesized that genetically distinct grass-E. coenophiala associations would differentially affect belowground fungi, and that these relationships would be climate-sensitive. Tall fescue genotype, endophyte presence, and climate treatment interactions affected AMF arbuscules, vesicles, and ERH. DSE decreased with E. coenophiala presence but increased with warming. Genetically distinct tall fescue-E. coenophiala associations may have divergent long-term impacts on other host-symbiont interactions, potentially impacting ecosystem function and response to climate change.

Most plant roots are associated with glomalean fungi forming arbuscular mycorrhizas (AM) and a wide range are also colonized by ascomycetous dark septate endophytes (DSE). Bromeliaceae species can be epiphytic, rupicolous or terrestrial but their mycorrhizal status is poorly studied. We examined the AM and DSE status of 5 epiphytic and 4 terrestrial Bromeliaceae from an arid area of Central Argentina. The terrestrial species were either dually associated (AM and DSE) or non-associated whereas the epiphytes were only DSE colonized. Terrestrial Bromeliaceae that formed AM-DSE associations were likely responding to the arid conditions of the area and the availability of AM fungal (AMF) spores in the soil. The terrestrialBromelia ubaniana was not colonized either by AMF or DSE. This could reflect its root morphology and high number of root hairs. DSE are endosymbiotic in the stressful ecosystems experienced by canopy epiphytes in the studied environment. The different fungal associations are discussed in relation to the three Bromeliaceae subfamiles and we suggest that environmental features determinethe type of association formed by species in this plant family.

In the past few decades, it has been widely accepted that forest loss due to human actions alter the interactions between organisms. We studied the relationship between forest fragment size and arbuscular mycorrhizal fungi (AMF) and dark septate endophytes (DSE) colonization, and the AMF spore communities in the rhizosphere of two congeneric Euphorbia species (native and exotic/invasive). We hypothesized that these fungal variables will differ with fragment size and species status, and predicted that (a) AMF and DSE colonization together with AMF spore abundance and diversity would be positively related to forest fragment size; (b) these relationships will differ between the exotic and the native species; and (c) there will be a negative relationship between forest fragment size and the availability of soil nutrients (NH4 +, NO3 , and phosphorus). This study was performed in the eight randomly selected forest fragments (0.86–1000 ha), immersed in an agricultural matrix from the Chaquean region in central Argentina. AMF root colonization in the native and exotic species was similar, and was positively related with forest fragment size. Likewise, AMF spore diversity and spore abundance were higher in the larger fragments. While DSE root colonization in the native host was positively related with forest fragment size, DSE colonization in the exotic host showed no relationship. Soil nutrients contents were negatively related with forest fragment size. In addition, NH4 + and NO3 were negatively correlated with AMF spores abundance and root colonization and with DSE colonization in the native species. The results observed in this study show how habitat fragmentation might affect the interaction between key soil components, such as rhizospheric plant-fungal symbiosis and nutrient availability. These environmental changes may have important consequences on plant community composition and nutrient dynamics in this fragmented landscape.

Rates of methanogenesis vary widely in peat soils, yet the reasons are poorly known. We examined rates of methanogenesis and methanogen diversity in relation to soil chemical and biological characteristics in 2 peatlands in New York State. One was an acidic (pH < 4.5) bog dominated by Sphagnum mosses and ericaceous shrubs, although deeper peat was derived from sedges. The other was a fen dominated by Carex lacustris sedges with near-neutral pH soil. At both sites, the most active rates of methanogenesis occurred in the top 20 cm of the peat profile, even when using a substrate-induced methanogenesis technique with added glucose that stimulated rates up to 2 μ mol g ? 1 day ?1 in the bog and 6 μ mol g ?1 day ?1 in the fen. Rates of anaerobic CO 2 production were greater in the bog (0–36 μ mol g ?1 day ?1 ) than in the fen (0–5 μ mol g ?1 day ?1 ), and added glucose induced greater rates in the sedge-derived peat from the bog than the fen. The peat soil was much more decomposed throughout the profile in the fen. Analysis of chemical elements in the peat profile revealed a striking anomaly: a very high concentration of Pb in surface peat of the bog, which might have constrained methanogenesis. Application of T-RFLP analysis to methanogens revealed dominance by a Methanomicrobiales E2 clade of H 2 /CO 2 users in the acidic peat soil of the bog, whereas deeper peat had a different Methanomicrobiales E1 clade, uncultured euryarchaeal rice cluster (RC)-I and RC-II groups, marine benthic group D (MBD) and a new cluster called subaqueous cluster (SC). In contrast, T-RFLP analysis of peat from the fen revealed co-dominance by Methanosaetaceae and Methanomicrobiales E1. The results showed complex relationships between rates of methanogenesis, methanogen populations and metabolic substrate availability with idiosyncratic interactions of trace chemical elements.
