The author studied the effect of different nickel concentrations (0, 0.4, 40 and 80 μM Ni) on the nitrate reductase (NR) activity of New Zealand spinach (Tetragonia expansa Murr.) and lettuce (Lactuca sativa L. cv. Justyna) plants supplied with different nitrogen forms (NO3 –N, NH4 +–N, NH4NO3). A low concentration of Ni (0.4 μM) did not cause statistically significant changes of the nitrate reductase activity in lettuce plants supplied with nitrate nitrogen (NO3 –N) or mixed (NH4NO3) nitrogen form, but in New Zealand spinach leaves the enzyme activity decreased and increased, respectively. The introduction of 0.4 μM Ni in the medium containing ammonium ions as a sole source of nitrogen resulted in significantly increased NR activity in lettuce roots, and did not cause statistically significant changes of the enzyme activity in New Zealand spinach plants. At a high nickel level (Ni 40 or 80 μM), a significant decrease in the NR activity was observed in New Zealand spinach plants treated with nitrate or mixed nitrogen form, but it was much more marked in leaves than in roots. An exception was lack of significant changes of the enzyme activity in spinach leaves when plants were treated with 40 μM Ni and supplied with mixed nitrogen form, which resulted in the stronger reduction of the enzyme activity in roots than in leaves. The statistically significant drop in the NR activity was recorded in the aboveground parts of nickel-stressed lettuce plants supplied with NO3 –N or NH4NO3. At the same time, there were no statistically significant changes recorded in lettuce roots, except for the drop of the enzyme activity in the roots of NO3 -fed plants grown in the nutrient solution containing 80 μM Ni. An addition of high nickel doses to the nutrient solution contained ammonium nitrogen (NH4 +–N) did not affect the NR activity in New Zealand spinach plants and caused a high increase of this enzyme in lettuce organs, especially in roots. It should be stressed that, independently of nickel dose in New Zealand spinach plants supplied with ammonium form, NR activity in roots was dramatically higher than that in leaves. Moreover, in New Zealand spinach plants treated with NH4 +–N the enzyme activity in roots was even higher than in those supplied with NO3 –N.  相似文献

Growth, chemical composition, and nitrate reductase activity (NRA) of hydroponically cultured Rumex crispus, R. palustris, R. acetosa, and R. maritimus were studied in relation to form (NH4 +, NO3 -, or both) and level of N supply (4 mM N, and zero-N following a period of 4mM N). A distinct preference for either NH4 + or NO3 - could not be established. All species were characterized by a very efficient uptake and utilization of N, irrespective of N source, as evident from high concentrations of organic N in the tissues and concurrent excessive accumulations of free NO3 - and free NH4 +. Especially the accumulation of free NH4 + was unusually large. Generally, relative growth rate (RGR) was highest with a combination of NH4 + and NO3 -. Compared to mixed N supply, RGR of NO3 -- and NH4 +-grown plants declined on average 3% and 9%, respectively. Lowest RGR with NH4 + supply probably resulted from direct or indirect toxicity effects associated with high NH4 + and/or low Ca2+ contents of tissues. NRA in NO3 - and NH4NO3 plants was very similar with maxima in the leaves of ca 40 μmol NO2 - g-1 DW h-1. ‘Basal’ NRA levels in shoot tissues of NH4 + plants appeared relatively high with maxima in the leaves of ca 20 μmol NO2 - g-1 DW h-1. Carboxylate to organic N ratios, (C-A)/Norg, on a whole plant basis varied from 0.2 in NH4 + plants to 0.9 in NO3 - plants. After withdrawal of N, all accumulated NO3 - and NH4 + was assimilated into organic N and the organic N redistributed on a large scale. NRA rapidly declined to similar low levels, irrespective of previous N source. Shoot/root ratios of -N plants were 50–80% lower than those from +N plants. In comparison with +N, RGR of -N plants did not decline to a large extent, decreasing by only 15% in -NH4 + plants due to very high initial organic-N contents. N-deprived plants all exhibited an excess cation over anion uptake (net proton efflux), and whole-plant (C-A)/Norg ratios increased to values around unity. Possible difficulties in interpreting the (C-A)/Norg ratio and NRA of plants in their natural habitats are briefly discussed.  相似文献

Supplying both N forms (NH4 ++NO3 ) to the maize (Zea mays L.) plant can optimize productivity by enhancing reproductive development. However, the physiological factors responsible for this enhancement have not been elucidated, and may include the supply of cytokinin, a growth-regulating substance. Therefore, field and gravel hydroponic studies were conducted to examine the effect of N form (NH4 ++NO3 versus predominantly NO3 ) and exogenous cytokinin treatment (six foliar applications of 22 μM 6-benzylaminopurine (BAP) during vegetative growth versus untreated) on productivity and yield of maize. For untreated plants, NH4 ++NO3 nutrition increased grain yield by 11% and whole shoot N content by 6% compared with predominantly NO3 . Cytokinin application to NO3 -grown field plants increased grain yield to that of NH4 ++NO3 -grown plants, which was the result of enhanced dry matter partitioning to the grain and decreased kernel abortion. Likewise, hydroponically grown maize supplied with NH4 ++NO3 doubled anthesis earshoot weight, and enhanced the partitioning of dry matter to the shoot. NH4 ++NO3 nutrition also increased earshoot N content by 200%, and whole shoot N accumulation by 25%. During vegetative growth, NH4 ++NO3 plants had higher concentrations of endogenous cytokinins zeatin and zeatin riboside in root tips than NO3 -grown plants. Based on these data, we suggest that the enhanced earshoot and grain production of plants supplied with NH4 ++NO3 may be partly associated with an increased endogenous cytokinin supply.  相似文献

The kinetics of NH4 + and NO3 uptake in young Douglas fir trees (Pseudotsuga menziesii [Mirb.] Franco) were studied in solutions, containing either one or both N species. Using solutions containing a single N species, the Vmax of NH4 + uptake was higher than that of NO3 uptake. The Km of NH4 + uptake and Km of NO3 uptake differed not significantly. When both NH4 + and NO3 were present, the Vmax for NH4 + uptake became slightly higher, and the Km for NH4 + uptake remained in the same order. Under these conditions the NO3 uptake was almost totally inhibited over the whole range of concentrations used (10–1000 μM total N). Nitrogen (N) limits plant productivity and its uptake and assimilation may be regulated by N source, N availability, and nitrate reductase activity (NRA). Knowledge of how these factors interact to affect N uptake and assimilation processes in woody angiosperms is limited. We fertilized 1-year-old, half-sib black walnut (Juglans nigra L.) seedlings with ammonium (NH4 +) [as (NH4)2SO4], nitrate (NO3 ) (as NaNO3), or a mixed N source (NH4NO3) at 0, 800, or 1,600 mg N plant−1 season−1. Two months following final fertilization, growth, in vivo NRA, plant N status, and xylem exudate N composition were assessed. Specific leaf NRA was higher in NO3 -fed and NH4NO3-fed plants compared to observed responses in NH4 +-fed seedlings. Regardless of N source, N addition increased the proportion of amino acids (AA) in xylem exudate, inferring greater NRA in roots, which suggests higher energy cost to plants. Root total NRA was 37% higher in NO3 -fed than in NH4 +-fed plants. Exogenous NO3 was assimilated in roots or stored, so no difference was observed in NO3 levels transported in xylem. Black walnut seedling growth and physiology were generally favored by the mixed N source over NO3 or NH4 + alone, suggesting NH4NO3 is required to maximize productivity in black walnut. Our findings indicate that black walnut seedling responses to N source and level contrast markedly with results noted for woody gymnosperms or herbaceous angiosperms.  相似文献

 The ability of four ericoid mycorrhizal endophytes isolated from roots of Woollsia pungens (Cav.) F. Muell. (Epacridaceae) to utilise organic forms of nitrogen and phosphorus during growth in axenic culture was assessed. All isolates were able to utilise glutamine, arginine and bovine serum albumin (BSA), along with NH4 + or NO3 , in most cases yielding at least as much biomass as the ericoid mycorrhizal endophyte Hymenoscyphus ericae (Read) Korf & Kernan. All isolated endophytes were able to utilise BSA, arginine and glutamine as sole sources of N and C. With the exception of a single isolate (C40), which showed little growth on glutamine, biomass yields on glutamine as the sole N and C source was significantly greater for all isolates than on either of the other two organic N sources. Two isolates from W. pungens (C40 and A43) utilised DNA and sodium inositol hexaphosphate as sole P sources, in each case yielding significantly more biomass than H. ericae. The results suggest that mycorrhizal endophytes from epacrid plant hosts and those from ericaceous hosts have similar abilities to utilise organic forms of N and P. Although many studies support the importance of the external mycelium for nutrient acquisition of ectomycorrhizal plants, direct evidence for a significant contribution to host nitrogen nutrition is still scarce. We grew nonmycorrhizal seedlings and seedlings mycorrhizal with Paxillus involutus (Batsch) Fr. in a sand culture system with two compartments separated by a 45-m Nylon mesh. Hyphae, but not roots, can penetrate this net. Nutrient solutions were designed to limit seedling growth by nitrogen. Hyphal density in the hyphal compartment, host N status and shoot growth of mycorrhizal seedlings significantly increased in response to NH4 + addition to the hyphal compartment. Labeling the compartment only accessible to hyphae with 15NH4 + showed that the increase in N uptake in the mycorrhizal seedlings was a result of hyphal N acquisition from the hyphal compartment. These results indicate that hyphae of P. involutus may actively forage into N-rich patches and improve host N status and growth. In the mycorrhizal seedlings, which received additional NH4 + via their external mycelium, the increase in NH4 + supply less negatively affected Ca and Mg uptake than in nonmycorrhizal seedlings, where the additional NH4 + was directly supplied to the roots. This was most likely due to the close link of NH4 + uptake and H+ extrusion, which, in the nonmycorrhizal seedlings, lead to a strong acidification in the root compartment, and subsequently reduced Ca and Mg uptake, whereas in the mycorrhizal seedlings the site of intensive NH4 + uptake and acidification was in the hyphal and not in the root compartment. Our data support the idea that the ectomycorrhizal mycelium connected to an N-deficient host may actively forage for N. The mycelium may also be important as a biological buffer system ameliorating negative influence of high NH4 + supply on cation uptake.  相似文献

Bi-directional translocation and degradation of Arginine (Arg) along the arbuscular mycorrhizal (AM) fungal mycelium were testified through 15N and/or 13C isotopic labeling. In vitro mycorrhizas of Glomus intraradices and Ri T-DNA-transformed carrot roots were grown in dual compartment Petri dishes. [15N- and/or13C]Arg was supplied to either the fungal compartment or the mycorrhizal compartment or separate dishes containing the uncolonized roots. The levels and labeling of free amino acids (AAs) in the mycorrhizal roots and in the extraradical mycelia(ERM) were measured by gas chromatography/mass spectrometry (GC-MS) and high-performance liquid chromatography (HPLC). The ERM of AM fungi exposed in either NH4 + or urea as sole external nitrogen source had much higher 15N enrichment of Arg, compared with those in nitrate or exogenous Arg; however, glycerol supplied as an external carbon source to the ERM had no significant effect on the level of Arg in the ERM. Meanwhile, Arg biosynthesized in the ERM could be translocated intact to the mycorrhizal roots and thereby the level of Arg in the mycorrhizal roots increased to about 20% after culture of ERM in 4 mmol/L NH4 + for 6 weeks. Also Arg was found to be bi-directionally transported along the AM fungal mycelium through [U-13C]Arg labeling either in the mycorrhizal compartment or in the fungal compartment. Once Arg was translocated to the potential N-limited sites, it would be further degraded into ornithine (Orn) and urea since either [U-13C] or [U-15N/U-13C]Orn was apparently shown up in the mycorrhizal root tissues when [U-13C] or [U-15N/U-13C]Arg was labeled in the fungal compartment, respectively. Evidently Orn formation indicated the ongoing activities of Arg translocation and degradation through the urea cycle in AM fungal mycelium. We investigated the effect of mineral nitrogen forms on transfer of nitrogen (N) and zinc (Zn) from attached compartments to rhodes grass (Chloris gayana) colonised with arbuscular mycorrhizal fungi (AMF). After being pre-cultivated in substrates with adequate nutrient supply and either AMF inoculated (+AM) or left non-inoculated (?AM), rhodes grass was positioned adjacent to an outer compartment holding a similar substrate but applied with labelled nitrogen (15N) either as ammonium (NH4 +) or nitrate (NO3 ?), and a high supply of Zn (150 mg kg?1 DS). Plant roots together with fungal mycelium were either allowed to explore the outer compartment (with root access) or only mycorrhizal hyphae were allowed (without root access). Within each access treatment, biomasses of rhodes grass were not significantly affected by AMF inoculation or N form. AMF contribution to plant 15N uptake was about double in NH4 + compared with NO3 ?-supplied treatments while the mycorrhizal influence on plant Zn uptake was insignificant. Without root access, the shoot 15N/Zn concentration ratio was up to ten-fold higher in +AM than –AM treatments and this ratio increase was clearly more pronounced in NH4 + than NO3 ?-supplied treatments. In conclusion, rhodes grass in symbiosis with the tested AMF acquired more N when supplied with ammonium. Moreover, there is clear indication that although the AMF have transported both nutrients (N and Zn), N was preferentially transferred as compared to Zn. We confirmed that, while rhodes grass is not able to prevent excessive Zn uptake via roots under conditions of high Zn, mycorrhiza is able to avoid excessive Zn supply to the host plant when the fungus alone has access to contaminated patches.  相似文献

Nitrite (NO2 ) is an intermediate in a variety of soil N cycling processes. However, NO2 dynamics are often not included in studies that explore the N cycle in soil. Within the presented study, nitrite dynamics were investigated in a Nothofagus betuloides forest on an Andisol in southern Chile. We carried out a 15N tracing study with six 15N labeling treatments, including combinations of NO3 , NH4 + and NO2 . Gross N transformation rates were quantified with a 15N tracing model in combination with a Markov chain Monte Carlo optimization routine. Our results indicate the occurrence of functional links between (1) NH4 + oxidation, the main process for NO2 production (nitritation), and NO2 reduction, and (2) oxidation of soil organic N, the dominant NO3 production process in this soil, and dissimilatory NO3 reduction to NH4 + (DNRA). The production of NH4 + via DNRA was approximately ten times higher than direct mineralization from recalcitrant soil organic matter. Moreover, the rate of DNRA was several magnitudes higher than the rate of other NO3 reducing processes, indicating that DNRA is able to outcompete denitrification, which is most likely not an important process in this ecosystem. These functional links are most likely adaptations of the microbial community to the prevailing pedo-climatic conditions of this Nothofagus ecosystem.  相似文献

The aim of this research was to test whether NH4 + and NO3 affect the growth, P demand, cell composition and N2 fixation of Cylindrospermopsis raciborskii under P limitation. Experiments were carried out in P-limited (200 μg l−1 PO4-P) chemostat cultures of C. raciborskii using an inflowing medium containing either 4,000 μg l−1 NH4-N, 4,000 μg l−1 NO3-N or no combined N. The results showed the cellular N:P and C:P ratios of C. raciborskii decreased towards the Redfield ratio with increasing dilution rate (D) due to the alleviation of P limitation. The cellular C:N and carotenoids:chlorophyll-a ratios also decreased with D, predominantly as a result of an increase in the chlorophyll-a and N content. The NH4 + and NO3 supply reduced the P maintenance cell quota of C. raciborskii. Consequently, the biomass yield of the N2-grown culture was significantly lower. The maximum specific growth rate of N2-grown culture was also the lowest observed. It is suggested that these differences in growth parameters were caused by the P and energy requirement for heterocyte formation, nitrogenase synthesis and N2 fixation. N2 fixation was partially inhibited by NO3 and completely inhibited by NH4 +. It was probably repressed through the high N content of cells at high dissolved N concentrations. These results indicate that C. raciborskii is able to grow faster and maintain a higher biomass under P limitation where a sufficient supply of NH4 + or NO3 is maintained. Information gained about the species-specific nutrient and pigment stoichiometry of C. raciborskii could help to access the degree of nutrient limitation in water bodies. Spatial variability of soil total nitrogen (N), available N (KCl extractable NH4+ and NO3), and spatial patterns of N mineralization and nitrification at a stand scale were characterized with geostatistical and univariate analysis. Two extensive soil spatial samplings were conducted in an evergreen broadleaf forest in Sichuan province, southwestern China in June and August 2000. In a study area of 90 × 105 m2, three soil samples were collected from each 5 × 5 m2 plot (n = 378) in June and August, and were analyzed for total N and available N contents. Net N mineralization and nitrification were measured by in situ core incubation and the rates were estimated based on the difference of NH4+ and NO3 contents between the two sampling dates. Total N, NH4+, and NO3 were all spatially structured with different semivariogram ranges (from high to low: NH4+, NO3, and total N). The semivariograms of mineralization and nitrification were not as spatially structured as available N. NH4+ was the dominant soil inorganic N form in the system. Both NH4+ and NO3 affected spatial patterns of soil available N, but their relative importance switched in August, probably due to high nitrification as indicated by greatly increased soil NO3 content. High spatial auto-correlations (>0.7) were found between available N and NH4+, available N and NO3 on both sampling dates, as well as total N measurements between both sampling dates. Although significant, the spatial auto-correlation between NH4+ and NO3 were generally low. Topography had significant but low correlations with mineralization (r = −0.16) and nitrification (r = −0.14), while soil moisture did not. The large nugget values of the calculated semivariograms and high-semivariance values, particularly for mineralization and nitrification, indicate that some fine scale (<5 m) variability may lie below the threshold for detection in this study.  相似文献

Physiology, regulation and biochemical aspects of the nitrogen assimilation are well known in Prokarya or Eukarya but they are poorly described in Archaea domain. The haloarchaeon Haloferax mediterranei can use different nitrogen inorganic sources (NO3, NO2 or NH4+) for growth. Different approaches were considered to study the effect of NH4+ on nitrogen assimilation in Hfx. mediterranei cells grown in KNO3 medium. The NH4+ addition to KNO3 medium caused a decrease of assimilatory nitrate (Nas) and nitrite reductases (NiR) activities. Similar effects were observed when nitrate-growing cells were transferred to NH4+ media. Both activities increased when NH4+ was removed from culture, showing that the negative effect of NH4+ on this pathway is reversible. These results suggest that ammonium causes the inhibition of the assimilatory nitrate pathway, while nitrate exerts a positive effect. This pattern has been confirmed by RT-PCR. In the presence of both NO3 and NH4+, NH4+ was preferentially consumed, but NO3 uptake was not completely inhibited by NH4+ at prolonged time scale. The addition of MSX to NH4+ or NO3 cultures results in an increase of Nas and NiR activities, suggesting that NH4+ assimilation, rather than NH4+ per se, has a negative effect on assimilatory nitrate reduction in Hfx. mediterranei.  Oxygen and CO2 fluxes were measured in hydroponically grown mycorrhizal and non-mycorrhizal Triticum aestivum L. cv. Hano roots. The NO3 uptake of the plants was used to estimate the amount of root respiration attributable to ion uptake. Plants were grown at 4 mM N and 10 μM P, where a total and viable mycorrhizal root colonisation of 48% and 18%, respectively, by Glomus mosseae (Nicol. and Gerd.) Gerd. and Trappe (BEG 107) was observed. The O2 consumption and NO3 uptake rates were similar and the CO2 release was higher in mycorrhizal than in non-mycorrhizal wheat. This resulted in a significantly higher respiratory quotient (RQ, mol CO2 mol–1 O2) in mycorrhizal (1.27±0.13) than in non-mycorrhizal (0.79±0.05) wheat. As the biomass and N and P concentrations in mycorrhizal and non-mycorrhizal wheat were the same, the higher RQ resulted from the mycorrhizal colonisation and not differences in nutrition per se. The impacts of various nitrogen sources, i.e. NO 3, NH4 + or NH4NO3 in combination with gaseous NH3, on nitrogen-, carbon- and water-use efficiency and 13C discrimination (δ13C) by plants of the C3 species Triticum aestivum L. (wheat) and the C4 species Zea mays L. (maize) were studied. Triticum aestivum and Z. mays were hydroponically grown with 2 mol · m−3 of N supplied as NO 3, NH4 + or NH4NO3 for 21 and 18 d, respectively, and thereafter exposed to gaseous NH3 at 320 μg · m−3 or to ambient air for 7 d. In T. aestivum and Z. mays over a 7-d growth period, nitrogen-use efficiency (NUE) values were influenced by N-sources in the decreasing order NH4NO3-N > NO 3-N > NH4 +-N and NO 3-N > NH4NO3-N > NH4 +-N, respectively. Fumigation with NH3 decreased the NUE values of plants grown with any of the N-forms. During 28- and 7-d growth periods, N-sources affected water-use efficiency (WUE) values in the decreasing order of NH4 +-N > NO 3-N≈NH4NO3-N in non-fumigated T. aestivum, while fumigation with NH3 increased the WUE of NO 3-grown plants. There were insignificant effects of N-sources on WUE values of Z. mays over 25- and 7-d growth periods. Furthermore, δ13C values in plant tissues (leaves, stubble and roots) were higher (less negative) in NH4 +-grown plants of T. aestivum and Z. mays than in those supplied with NH4NO3 or NO 3. Regardless of the N-form supplied to the roots of the plant species, exposure to NH3 caused more-positive δ13C values in the plant tissues. These results indicate that the variations in N-source were associated with small but significant variations in δ13C values in plants of T. aestivum and Z. mays. These differences in δ13C values are in the direction expected from differences in WUE values over long or short growth periods and with differences in the extent of non-Rubisco (ribulose-1,5-bisphosphate carboxylase-oxygenase, EC 4.1.1.39) carboxylate contribution to net C acquisition, as a function of N-source. Knowledge of the preferred source of N for Eucalyptus nitens will lead to improved fertiliser management practices in plantations. Ion selective microelectrodes were used non-invasively to measure simultaneously net fluxes of NH4 +, NO3 and H+ along the tap root of solution-cultured E. nitens. Measurements were conducted in solutions containing 100 m NH4NO3. The pattern of fluxes was such that there was a large influx of NH4 +, a smaller influx of NO3 and large H+ efflux. The ratio of these fluxes was constant, according to the ratio 3:1:–6 (NH4 +:NO3 :H+). Within the region 20–60 mm from the root apex of E. nitens seedlings there was spatial and temporal variation in fluxes but flux patterns remained constant. Root hair density did not affect fluxes nor did proximity to lateral roots. Variation was less than that found in previous studies of localised root fluxes using similar high-resolution measurement techniques. It was concluded that small-scale spatial variation in fluxes may have confounded previous studies. There were associations between fluxes of all three ions, the strongest associations being between NH4 + and H+, and NH4 + and NO3 . Overall, these results are consistent with NH4 + being the preferred source N for E. nitens.  相似文献

Rooting of Eucalyptus globulus shoots was influenced by the concentration of the indole butyric acid (IBA) and NH4 + in the root-induction medium. Optimum plantlet vigor and survival were achieved using low concentrations (1 – 2.5 μM) of IBA and when NH4NO3 was removed. Removal of NH4 + also had a significant effect on medium pH, its presence caused a decrease in pH as the culture period proceeded. When different nitrate compounds (excluding NH4NO3) were used as the nitrogen source, the medium pH was more stable and this was associated with higher root production. The higher root production, in association with appropriate IBA concentrations, produced plantlets with higher survival and better growth on transfer to soil. Here, we characterized nitrogen (N) uptake of beech (Fagus sylvatica) and their associated ectomycorrhizal (EM) communities from NH4+ and NO3?. We hypothesized that a proportional fraction of ectomycorrhizal N uptake is transferred to the host, thereby resulting in the same uptake patterns of plants and their associated mycorrhizal communities. 15N uptake was studied under various field conditions after short‐term and long‐term exposure to a pulse of equimolar NH4+ and NO3? concentrations, where one compound was replaced by 15N. In native EM assemblages, long‐term and short‐term 15N uptake from NH4+ was higher than that from NO3?, regardless of season, water availability and site exposure, whereas in beech long‐term 15N uptake from NO3? was higher than that from NH4+. The transfer rates from the EM to beech were lower for 15N from NH4+ than from NO3?. 15N content in EM was correlated with 15N uptake of the host for 15NH4+, but not for 15NO3?‐derived N. These findings suggest stronger control of the EM assemblage on N provision to the host from NH4+ than from NO3?. Different host and EM accumulation patterns for inorganic N will result in complementary resource use, which might be advantageous in forest ecosystems with limited N availability.  相似文献
