The ratios between the indices of relative abundance for different trophic life forms have been used to characterize bionomical types of a rocky intertidal zone. It has been shown that the distribution of life forms is determined by the geomorphological peculiarities of the surveyed intertidal areas. A brief critical historical review has been provided of the terms bionomy and bionomical type.  相似文献

Osmotically fragile forms of Streptococcus faecalis 9790 were grown in 0.5 m sucrose- or 0.5 m NH(4)Cl-stabilized medium. The "protoplast" cultures exhibit an average growth rate constant of 0.66 to 0.94 mass doublings/hr. In a variety of experiments, turbidity and the net content of protein, ribonucleic acid (RNA) and deoxyribonucleic acid increase at the same rate, indicating balanced macromolecular biosynthesis. A total of two to three mass doublings was obtained, with no evidence of cell division. After osmotic shock, "protoplast" cultures released 93 to 94% of their RNA content in a form not sedimentable at 12,800 x g for 15 min, in contrast to streptococci, which released 7% of their RNA content after the same treatment.  相似文献

Soluble immune response suppressor (SIRS) isolated from the T cell hybrid 393D2.6 was originally reported to exist as at least two m.w. forms and to migrate on reverse-phase high-performance liquid chromatography columns as three separate species. In experiments presented here, a further analysis of the different chromatographic forms of SIRS has been carried out. SIRS-alpha elutes from C-18 reverse-phase columns in 20% propanol. When SIRS-alpha is subjected to isoelectric focusing, three biologically active species are isolated at approximately pH7, approximately pH6, and approximately pH5 (SIRS-alpha 7, SIRS-alpha 6, and SIRS-alpha 5, respectively). SIRS-beta elutes in 30% propanol, and on isoelectric focusing the biologic activity is found only at approximately pH7 (SIRS-beta 7). Both the alpha and beta forms of SIRS have nearly identical m.w. when subjected to molecular sieve chromatography and migrate with a m.w. of 11,000. The molecular basis for these isoforms is not yet clear but is consistent with earlier studies showing two separate messenger RNA species coding for SIRS.  相似文献

A histamine-releasing factor from activated human mononuclear cells   总被引:10,自引:0,他引:10  
Human mononuclear cells activated by streptokinase-streptodornase have been shown to elaborate a factor capable of releasing histamine from human basophils. We have developed reproducible methods for its production in large quantities by using cells obtained from leukapheresis packs, by detection utilizing donor basophils known to release well with anti IgE, and by quantitation of histamine by the radioenzyme method. Human histamine-releasing factor (HRF) gave a single peak upon gel filtration with an estimated m.w. of 32,000; SDS gel electrophoresis revealed a single major band as seen at m.w. 30,000. HRF can be resolved into at least two forms separable by ion-exchange chromatography on QAE Sephadex, and two peaks of activity were obtained by chromatofocusing or isoelectric focusing in gels at pH 6.9 and between 7.4 and 8.3. This factor represents an important potential link between cellular immunity and immediate hypersensitivity.  相似文献

Whole-cell syntheses of representative modified purine and pyrimidine 2-deoxy--D-ribonucleosides are described. The transglycosylation reactions were carried out at 55¡C using the thermostable bacteria, Bacillus stearothermophilus. These transformations were efficient and gave yields close to or greater than 50% (conversions >70%).  相似文献

A clone of the continuous human T cell line HUT-102, termed YM 1.2, can spontaneously release alpha-LT in vitro. However, when stimulated with phorbol myristic acetate, these cells release other LT forms. These LT forms were purified to homogeneity by DEAE chromatography, column isoelectric focusing, and polyacrylamide gel electrophoresis. One LT form, termed LT-2, is a 79,000 m.w. component in aqueous solution and composed of 21,000 m.w. subunits. This form is immunologically related to macrophage-derived TNF and has a lytic capacity in vitro on K-562, Molt-4F, and Raji cells similar to that described for cytotoxins derived from NK effector cells, termed NK-CF. A second LT form, termed LT-3, is a single 69,000 m.w. peptide which could not be reduced into the smaller subunits. This form expresses antigens in common with both alpha-LT and TNF, because both anti-LT and anti-TNF were required to completely neutralize cell lytic activity in vitro. Functional testing revealed that the LT-3 form is lytic on all continuous cells tested in vitro, including NK-resistant target cells. The LT-3 component appears similar by immunologic, biochemical, and functional criteria to the LT form derived from primary human cytolytic T cells in vitro. At the levels tested, none of these LT-TNF forms had measurable effects on primary fibroblasts in vitro.  相似文献

The milk-clotting and proteolytic activities of extracellular enzyme preparations from Nocardiopsis sp. were investigated under different culture conditions. A soybean flour medium was used, with concentrations of soybean flour and of glucose varying from 0.25 to 1% w/v and from 0 to 1% w/v, respectively. Growth was monitored with 2ml samples withdrawn from the culture medium at 8-h intervals, for determination of total protein, proteolytic activity, milk-clotting activity and sugar reduction. The best milk-clotting protease production, with a specific activity of 24.49U/mg at 40h, was obtained in the glucose-free medium containing soybean flour 1% w/v.  相似文献

Three multiple phycoerythrin-545 forms were purified from crude extracts of Cryptomonas maculata by preparative isoelectric focusing. The phycoerythrin forms are charge isomers with isoelectric points at 7.83, 5.05 and 4.84. The multiple pigment forms have similar molecular weights of 44500 daltons and are composed of subunits of unequal size in a 1:1 stoichiometry with molecular weights of () 9900 and () 15700 daltons, twice. The proposed quarternary structure of the native pigments is ()2()2.The charge differences of the phycoerthrins are caused by a charge heterogeneity of the light subunits, as revealed by urea gel electrophoresis. The chains of pigment form pI 7.83 had a greater electrophoretic mobility than those subunits of the acidic pigment forms pI 5.05 and pI 4.84.The phycoerythrin forms have an absorption spectrum with similar absorption maxima at 544 nm, but differ in the position of the long wavelength shoulders lying at 555 and 557 nm in the negatively charged pigment forms and at 560 nm for the phycoerythrin form with a pI at 7.83.The fluorescence emission spectra coincide in their asymmetrical shape with shoulders at about 620 nm; they slightly differ int he position of the emission maxima at 586 nm for the phycoerythrins with pIs at 4.84 and 5.05 and at 584 nm for phycoerythrin with pI at 7.83.Abbreviations PC phycocyanin - PE phycoerythrin - pI isoelectric point - SDS sodium dodecyl sulphate  相似文献

Protoplasts isolated from cotyledons of Brassica carinata, underwent sustained division when cultured at 5.0 × 104 ml-1 in modified 8p medium (KM8P) with 1.0% (w/v) Seaplaque agarose. Cell colonies produced callus when agarose droplets, in which the protoplasts were embedded, were transferred to K8 medium with 0.6% (w/v) Sigma Type I or Type VII agarose at day 16, giving a plating efficiency of 1.6%. Seventy percent of the protoplast derived-tissues produced shoot buds after subculture to MS medium containing 3.0% (w/v) sucrose, 1.125 mgl-1 BAP, 0.035 mgl-1 GA and 0.6% (w/v) Type I agarose, resulting in shoot formation from 1.1% of the protoplasts originally plated. Protoplast-derived colonies transferred to hormone-free MS medium with 1.0% (w/v) sucrose and 0.6% (w/v) Type I agarose produced roots. The latter gave rise to shoots after excision from the parent callus and culture on MS medium with 3.0% sucrose, 0.225 mgl-1 BAP, and 0.6% (w/v) Type I agarose. Waxy maize starch (100% amylopectin) granules were modified by reaction of the granules with glucoamylase in a minimum amount of water to give 29% (w/w) d-glucose inside the granules [Kim, Y.-K.; Robyt, J. F. Carbohydr. Res.1999, 318, 129−134]. These granules were made into beads by dropping an ethanol slurry of starch and different amounts of Eudragit L100-55 in a constant ratio of 100:1 from a pipette onto Whatman 3MM filter paper. The starch beads were air dried and then repeatedly sprayed 0-12 times with 2.0% (w/v) Eudragit L100-55 in ethanol, with drying between each spraying, to coat the surface of the starch beads, giving different amounts of Eudragit L100-55 coating. Seven different kinds of beads, with different amounts of Eudragit L100-55 binding and coating agent, were obtained. The rates of release of d-glucose into water from the seven kinds of beads were inversely proportional to the amount of binding and coating agent. Bead type I, which was without any binding and coating gave a fast 100% release of d-glucose in 30 min. Beads II and III also gave a fast 100% release in 60 min and 90 min, respectively. Bead IV gave a near linear release of 97% d-glucose in 150 min; Bead V gave a 50% release in 120 min followed by the remaining 50% in 60 min; and Beads VI and VII gave a slow release of 10% and 4%, respectively, from 0 to 120 min, followed by a rapid 100% release from 120 to 180 min.  相似文献

The action of the lysoamidase bacteriolytic complex on Staphylococcus aureus VKM B-209P cells has been studied to obtain protoplasts. The cells in the midlogarithmic phase were the most sensitive to lysoamidase action. It led to local destruction of cell wall due to hydrolysis of the peptidoglycan. Protoplast formation occurred in two steps in the presence of 1 M sucrose. First, osmotically fragile spheroplasts were formed. Then, the protoplasts were released from the destructed cell wall. The protoplast yield was about 80%. The protoplasts preserved the intact ultrastructure and were able to synthesize peptidoglycan fibrillae. Mainly the spheroplasts that maintained the cell-wall residues reversed into bacterial forms. The protoplasts had respiratory activity similar to cells. Respiration of cells and protoplasts was stimulated by various substrates. High rates of oxygen consumption were observed with -glycerophosphate and ethanol as substrates.  相似文献

The conformation of brain proteolipid apoprotein (PLA) has been investigated using infrared spectroscopy and freeze-fracture electron microscopy. For this purpose, spectroscopic samples consisting of a mixture of liquid paraffin and wet protein have been prepared. These systems have allowed us to record the infrared spectra of PLA at neutral pH. The amide I and III regions reveal the existence of a predominantly -helical structure, as well as the presence of minor -strands and random coil forms. The effect of sonication and a non-denaturing detergent, (n-octyl--d-glucopyranoside), on the structure of the protein have also been investigated. Sonication produces an increase of the and unordered structures at the expense of the -helical conformation. These structural changes are enhanced in the presence of the non-ionic detergent n-octyl--d-glucopyranoside. Lipids protect the native protein structure from the effects of sonication. The aforementioned detergent changes the PLA conformation by increasing the -helical content at the expense of -sheet and random coil forms. Therefore the PLA structure seems to be similar to the structures of other proteins intrinsic to non-neural membranes. The effects investigated also suggest that PLA behaves in a conformationally flexible manner.  相似文献

Membrane boundl-glutamate decarboxylase (GAD) has been solubilized and partially purified from hog brain. The solubilized GAD appears to exist in two forms, and , differing in their size and electrophoretic mobility. The form has similar mobility as that of the soluble GAD in 7.5% and 5–25% gradient polyacrylamide gel electrophoresis suggesting that they are similar in size and charge. In addition, gene encoding for mouse brain GAD has been cloned and characterized. Mouse brain GAD cDNA consists of two DNA fragments with 1.6 and 1.0 Kb. The 1.6 and 1.0 Kb fragments contain 1657 and 974 bP, respectively. The significance of multiple forms of GAD is also discussed.Special issue dedicated to Dr. Eugene Roberts.  相似文献

"Biosynthetic" L-threonine dehydratase (EC 4.2.1.16) was purified to a homogeneous state with 29% yield of total activity from Escherichia coli K-12. The homogeneity of the enzyme was shown by polyacrylamide gel disc electrophoresis in the presence of dodecyl sulphate. The enzyme consisted of equal subunits having a molecular weight of about 57 000. The polyacrylamide gel disc electrophoresis has shown that the native enzyme consisted of a set of oligomeric forms. The multiplicity of molecular organization of the enzyme was reflected in complicated kinetic behaviour: at pH greater than 9 on the plots of initial reaction rate (v) versus initial substrate concentration ([S]o) there were four inflexion points (two intermediate plateaux), the position and deepness of which depended on enzyme concentration. At pH 8.3 on the v versus [S]o plots appeared two inflexion points (one intermediate plateu), the position of which practically did not depend on enzyme concentration in the reaction mixture, but strongly depended on the enzyme concentration in the stock solution. Repeated polyacrylamide gel disc electrophoresis of several oligomeric forms, isolated by the first electrophoresis, has shown that the oligomeric forms underwent a slow polymerization. It was suggested that "biosynthetic" L-threonine dehydratase from E. coli K-12 is a set of multiple oligomeric forms, having different kinetic parameters. Probably, each form of the enzyme has a "simple" kinetics characterized by hyperbolic or sigmoidal shape of v versus [S]o plots. The rate of equilibrium installation between the oligomeric forms was small in comparison with the enzyme reaction velocity, that lead to the complex kinetic curves, appearing as a result of summing up of the kinetics inherent to theindividual forms.  相似文献

The monoclonal antibody A2B4-2 has been shown to bind to the antigen receptor on the cloned pigeon cytochrome c-specific T cell hybrid, 2B4. Initial immunoprecipitation and SDS-PAGE analysis with this clonotypic antibody demonstrated that the antigen receptor on this cell had a m.w. of 85,000 to 90,000. Under reducing conditions, the receptor protein appeared as two bands of 45,000 to 50,000 and 40,000 to 44,000 on an SDS-PAGE gel. In this paper the antigen receptor on this T cell hybrid is further characterized. The molecule is shown to be a heterodimer that exists in two different forms on the cell surface. Receptor molecules with an apparent m.w. of 84,000 and 86,000 were isolated by immunoprecipitation and separation on polyacrylamide gradient gels. After reduction, the individual alpha- and beta-chains were separated by isoelectric focusing. In both forms of the receptor, the acidic alpha-chain had an apparent m.w. of 42,000 to 44,000. This alpha-chain associated with one of two forms of beta-chain. One beta-chain had a m.w. of 42,000 to 44,000, with a pI range of 7.5 to 7.9, and the alternate form of the beta-chain, beta', had a m.w. of 46,000 to 48,000 and a more acidic pI range of 6.5 to 7.5. The results of this investigation indicate that under reducing conditions on SDS-PAGE gels, the original upper 45,000 to 50,000 m.w. band represented beta'-chains alone, whereas the lower 40,000 to 44,000 m.w. band represented a mixture of alpha- and beta-chains. Additional data are presented to indicate that this heterodimeric protein has intrachain as well as interchain disulfide bonds. This conclusion was reached from the characteristic pattern of protein migration on SDS-PAGE gels in the presence of a reducing agent concentration gradient. Thus, both chains of the antigen receptor must have intrachain disulfide bonds and may have similar domain structures.  相似文献

IgM and IgD genes of the Japanese flounder were cloned and characterized from a genomic bacterial artificial chromosome (BAC) library. The gene contained four constant region exons (C1–C4), and two transmembrane exons (TM1 and TM2), which conforms to the organizational pattern of all other vertebrate -chain genes examined so far. In the same BAC clone, the gene, which is homologous to the IgD gene in mammals and teleost fish, was found immediately (0.9 kb) downstream of the IgM gene. This gene encoded seven exons (C1–C7) and two TM exons (TM1 and TM2) and had no duplication of C1-C2, as found in Atlantic cod, or C2-C3-C4, as found in Atlantic salmon and channel catfish. Phylogenetic and sequence analyses strongly suggest that teleost is more closely related to non-IgM isotypes than IgM isotypes. The heavy chain (IgH) locus of Japanese flounder, which encodes m, s and m, was found to be fully functional.  相似文献

Acetylcholinesterase (AChE) from housefly heads was purified by affinity chromatography. Three different native forms were separated by electrophoresis on polyacrylamide gradient gels. Two hydrophilic forms presented apparent molecular weights of 75,000 (AChE1) and 150,000 (AChE2). A third component (AChE3) had a migration that depended on the nature and concentration of detergents. In the presence of sodium deoxycholate in the gel, AChE3 showed an apparent molecular weight very close to that of AChE2. Among the three forms, AChE3 was the only one found in purified membranes. The relationships among the various forms were investigated using reduction with 2-mercaptoethanol or proteolytic treatments. Such digestion converted purified AChE3 into AChE2 and AChE1, and reduction of AChE3 and AChE2 by 2-mercaptoethanol gave AChE1, in both cases with a significant loss of activity. These data indicate that the three forms of purified AChE may be classified as an active hydrophilic monomeric unit (G1) plus hydrophilic and amphiphilic dimers. These two components were termed G2s and G2m, where "s" refers to soluble and "m" to membrane bound.  相似文献

American ginseng, Panax quinquefolius L., is one of the most heavily traded medicinal plants in North America. The effect of harvest on genetic diversity in ginseng was measured with a single generation culling simulation program. Culling scenarios included random harvest at varying levels, legal limit random harvest and legal limit mature plant harvest. The legal limit was determined by the proportion of legally harvestable plants per population (% mature plants per population). Random harvest at varying levels resulted in significant loss of genetic diversity, especially allelic richness. Relative to initial levels, average within-population genetic diversity (H e) was significantly lower when plants were culled randomly at the legal limit (Mann–Whitney U=430, p<0.001) or when only mature plants were culled (Mann–Whitney U=394, p<0.01). Within-population genetic diversity was significantly higher with legal limit mature plant harvest (H e=0.068) than when plants were culled randomly at the legal limit (H e=0.064; U=202, p<0.01). Based on these simulations of harvest over one generation, we recommend that harvesting fewer than the proportion of mature plants could reduce the negative genetic effects of harvest on ginseng populations.  相似文献

The aim of the study was to investigate theAmbrosia pollen concentrations inselected Polish cities and for Kraków torelate it to some meteorological factors.Sampling was carried out in Kraków in1982–1997 and in Rabka in 1992–1996 with theuse of the gravimetric method. In Zakopane,Kraków, Ostrowiec witokrzyski,Warszawa and Pozna in 1995–1996 both thegravimetric and volumetric methods (Burkardtrap) were employed. In Kraków themonitoring has been performed since 1994 usingthe volumetric method. The results show theragweed pollen presence in August and Septemberwith the tendency to appear more frequently inAugust in some years. In Kraków (1994–1999)Ambrosia pollen was found either in thelast two weeks of August or in the first twoweeks of September which seems to be a regularand repeatable pattern every two years.Seasonal fluctuations of Ambrosia pollenconcentration do not show a clear increasingtendency except at Warszawa and Ostrowiecwitokrzyski in 1996 and at Krakówin 1999. Percentage of Ambrosia pollen inannual sums of total pollen is very low anddoes not exceed 1% except at Ostrowiecwitokrzyski in 1996 (1.2%) and atKraków in 1999 (2%). For Kraków theanalysis of some meteorological factors (Tmax,Tmin, precipitation, wind direction) wasperformed. High temperature and lack of rain orlow precipitation correlate well with ragweedpollen concentrations. During the Ambrosia pollen seasons ESE, E, S, SE, WSW, SWwind directions prevailed which could suggest along-distance transport from Ukraine, the CzechRepublic, Slovakia and also from Hungary, one ofthe most ragweed-polluted countries.  相似文献
