运用PCR直接测序法和石蜡切片法,测定5种安徽产大戟属植物的nrDNA的内转录间隔区(ITS)序列包括5.8SrDNA及其叶片的解剖。结果表明(1)5种大戟属植物的ITS的长度范围为641~662bp,运用Maga软件构件ITS树把5种植物归为两大支大戟、月腺大戟与乳浆大戟聚为一支;地锦和斑地锦聚为另一支。结果表明地锦和斑地锦亲缘关系较近,而大戟、月腺大戟和乳浆大戟亲缘关系较近。(2)5种大戟属植物的叶片结构中,除地锦和斑地锦外,其余3种有明显的栅栏组织和海绵组织之分,但二者在叶肉中的厚度比在种间有一定的差别;而地锦和斑地锦叶的栅栏组织与海绵组织分化不明显,且皆有乳汁管,其他3种植物叶片内未见此结构,叶片结构分析表明大戟、月腺大戟和乳浆大戟叶片结构相近,而地锦草和斑地锦叶片结构也相近。(3)分析表明nrDNA的ITS序列及叶的比较解剖特征具有明显的相关性。以上研究结果为大戟属植物的分类和药用开发提供了实验证据。  相似文献

用光学显微镜和扫描电镜,对石竹属12个种的植物叶表皮特征进行观察,统计并测量叶表皮气孔大小、气孔密度及气孔指数。结果表明,石竹属植物叶表皮细胞形态(表面观)为长方形和不规则多边形。乳突只存在于针叶石竹、高石竹和细茎石竹中。按气孔形状可将其分为了3个类型:椭圆形、卵圆形和长方形。研究结果对石竹属的系统分类和种间亲缘关系研究具有一定的参考价值。  相似文献

利用光镜和扫描电镜对安徽7种大戟属(Euphorbia)植物叶表皮进行了观察。发现该属7种植物叶表皮细胞为不规则形或多边形,垂周壁一般为平直、浅波状、深波状;气孔器通常为无规则型,有的种类叶表皮具单细胞毛或多细胞毛,表皮毛在电镜下具瘤状突起的纹饰。在扫描电镜下,角质层纹饰多具鳞片或颗粒等特征,种间有细微差别。结果表明该7种植物叶表皮微形态有明显差别,这些表皮特征有助于识别一些在外部形态较相近的种类。  相似文献

采用光学显微镜,对华北地区分布的繁缕属植物16种2变种,其它4属4种植物的叶表皮微形态进行了观察。结果表明:繁缕属植物叶片的下表皮均存在气孔器,而上表皮仅少数种类缺乏气孔器。气孔器类型比较多样,有不等细胞型、无规则型、无规则四细胞型;表皮细胞主要为不规则型,垂周壁从平直、弓形、浅波状、波状、到深波状;上表皮气孔指数在5.23~31.65之间;下表皮气孔指数在10.02~29.44。叶表皮被毛或无,毛被分为星状毛、腺柔毛、柔毛、绢毛。该研究说明叶表皮的微形态特征在种间差异明显,可以为分种及属间亲缘关系的探讨提供依据,但是对组的划分存在一定的局限性。  相似文献

利用整体透明法、石蜡制片法和半薄切片法,对金丝桃属9组43种l亚种1变种的植物叶分泌 结构的类型、形态、结构和分布进行了比较研究。结果表明,分泌结构是金丝桃属植物叶片普遍的结构特征,根据其分泌结构的特征,可划分为分泌细胞团、分泌囊(道)和韧皮部中分泌小管道等3种分泌结构。其中,分泌细胞团按其在叶片的分布可分为叶缘型和散生型;分泌囊按其在叶横切面中的位置可分为栅栏组织型、海绵组织型、居中型(位于栅栏组织型与海绵组织型之间)和横跨叶肉组织型。根据分泌囊和分泌细胞团在该属植物叶中的分布可划分为3种类型:(1)只有分泌囊的类型;(2)只有分泌细胞团的类型;(3)具分泌囊和分泌细胞团的类型。两种主要分泌结构的类型、分布密度、分布位置及其形态等方面在组间和种间均存在—定的差异,对金丝桃属属以下等级的区分具有一定意义。在此基础上,对该属分泌结构的形态演化以及金丝桃属各组间的亲缘关系进行了探讨。  相似文献

利用扫描电镜技术、叶片叶片离析方法和石蜡切片法对暗罗属12种植物叶和形态结构进行了比较研究。结果表明,叶表皮细胞形状、气孔器形态、表皮毛类型、表皮细胞中晶体类型、叶肉中油细胞分布位置、栅栏组织和海绵组织厚度的比值,以及主脉维管组织的结构特征等具有明显的种间差异。可以利用这些叶的解剖特征将暗罗属植物相互区别开来。  相似文献

以中国北方生长的鸢尾属9种植物为研究对象,利用光学显微镜对叶表皮细胞形状及大小、气孔大小、气孔分布及气孔密度、气孔指数等叶表皮构造特征进行观察,并探讨了叶表皮特征在鸢尾属内各个亚属、亚组间的分类学价值,结果表明:表皮细胞的形状和大小显示了同亚属或同亚组植物之间较为明显的亲缘关系,在区分鸢尾属下不同亚属或亚组植物上也有一定的分类学价值。气孔密度和气孔指数在鸢尾属内分类学价值不大,但却能很好地反映不同鸢尾属植物与生境之间的密切关系。并从表皮构造特征上证实了野鸢尾与射干有很强的亲缘关系,且近于鸢尾属内的其他植物的结论;此外,通过对鸢尾属植物叶横切解剖结构的观察:从维管束、横切面形状、表皮细胞与角质层、乳突、气孔、气室大小、叶肉细胞的分化这7个较为明显的特征上的相关性和差异性,探讨了由于生境的不同而产生的植物形态结构的差异。  相似文献

为探讨叶表皮微形态特征在变豆菜属植物中的系统及分类学意义,我们利用扫描电镜对变豆菜属11个种(13个居群)的叶表皮微形态特征进行了研究。结果表明,11种变豆菜属植物叶表皮微形态特征较稳定,上表皮细胞轮廓大多不清晰,初级蜡质纹饰均存在条状纹饰,表皮上均粘附着颗粒物;下表皮均存在气孔,气孔周围存在均匀分布的纹饰,气孔外拱盖表面大多有颗粒状纹饰,并且其二级纹饰类型、气孔周围纹饰、气孔外拱盖内缘纹饰等叶表皮微形态特征上存在一定差异。上述研究结果可为探讨变豆菜属类群界定及种间关系提供重要依据。  相似文献

利用ISSR标记对9种大戟属植物的亲缘关系进行了分析.结果表明:筛选出的24条ISSR引物共扩增251条带,其中多态性条带248条,多态率为98.8%.9种大戟属材料的遗传相似系数在0.51~0.69之间,平均为0.62.利用UPGMA法将9种大戟属植物分别聚为3个类群,第一类群包含地锦亚属与一品红亚属的5个种及大戟亚属的绿玉树,第二、第三类群分别包含大戟亚属中的3个种.ISSR标记获得的聚类结果与根据形态特征获得的分类结果有很大的差异.  相似文献

The tropical Indo-Pacific genus Excoecaria L. (Euphorbiaceae) has several closely related species in Australia whose taxonomic relationships are unclear. The most widely reported species in Australia is the mangrove species Excoecaria agallocha L. (type species), whose taxonomic and geographic limits are difficult to define from its closely related species or sub-species. Two additional taxa have also been described but not clearly differentiated from the type species: Excoecaria dallachyana Baillon and Excoecaria ovalis Endl. This project aimed to determine the taxonomic relationships of the Australian Excoecaria species using both leaf morphological data and DNA sequence data from the internal transcribed spacer (ITS) region of ribosomal genes. The nucleotide differences in the examined ITS1 region show that E. agallocha from eastern Australia and E. ovalis from Western Australia respectively, are genetically uniform within species but differ from each other consistently, thus supporting species status. The leaf morphological data also support this view: single factor analysis of variance consistently separated E. ovalis from E. agallocha on the basis of leaf width, leaf length and length of petiole. In contrast, E. ovalis from the Gulf of Carpentaria differs only slightly from E. ovalis in Western Australia, but no evidence was found to suggest any leaf morphological differentiation within this species. The analysis also suggests that E. dallachyana is not closely related to either mangrove species E. agallocha or E. ovalis, despite superficial morphological similarities.  相似文献

This study is focused on the genus Euphorbia L. in a part of northeast Iran, viz. the three Khorassan provinces. Since there are many taxa of Euphorbia in Iran which are used in different industries and have significant effects on human and non‐human life it is important to revise their taxonomy. With about 90 species, following Turkey with 91 species, Iran is the second richest country for Euphorbia in Asia. Of these, 30 species are distributed in the Khorassan provinces. This is the first comprehensive work on the genus in this region. According to ‘Flora Iranica’, there are 17 species of Euphorbia in northeast Iran, while according to our results, there are 30 species of Euphorbia in the Khorassan provinces alone. In addition to various new taxonomic and biogeographic results, a new species, viz. E. chamanbidensis, is described. Euphorbia chamanbidensis is closely related to E. aucheri, but seed micro‐morphological characters differentiate them. Two identification keys to the Euphorbia species of the studied area are provided, one based on macro‐morphological characters and another based on seed micro‐morphological characters. Phytogeographic analysis and distribution maps for all species are also presented.  相似文献

Seed coat surfaces of 127 species, representing 23 sections of the genusEuphorbia L. (Euphorbiaceae) have been examined with the scanning electron microscope (SEM). Various surface patterns and cell wall structures are described. In some groups the seed coat is rather uniform (e.g. in sect.Euphorbium), in other sections even closely related species can be separated by seed coat morphology (e.g.Tithymalus). The taxonomic application of testa micromorphology and the possible systematic position of certain taxa are briefly discussed. The seed coat cells of all species in sect.Tithymalus show remarkable intercellulares filled with small particles, which are described in detail for the first time.

该研究以森林草莓(Fragaria vesca)为试验材料,利用RT-PCR技术克隆草莓FvARF10、FvARF18基因全长cDNA序列。生物信息学分析表明,FvARF10、FvARF18基因开放阅读框(ORF)分别为2 056bp和2 100bp,编码685和699个氨基酸,预测其分子量和等电点分别为75.73、76.78kD和8.12、6.55。系统进化树分析表明,FvARF10与苎麻BnARF10聚为一个分支,FvARF18与桑树MnARF18聚集在一个分支上,进化关系较密切。亚细胞定位预测分析表明,FvARF10、FvARF18蛋白均定位于细胞核或细胞质中。启动子分析显示,FvARF10、FvARF18基因均具有多样化的激素应答元件。荧光定量PCR结果显示,FvARF10和FvARF18基因在草莓不同组织中均有表达,FvARF10在果实中表达较高,且在半熟果中表达量最高,而FvARF18在茎中相对表达量最高;外源激素IAA处理1h后,FvARF10和FvARF18基因受到强烈诱导均呈上调表达,而后渐恢复正常水平。研究结果表明,FvARF10和FvARF18基因可能参与草莓的营养生殖及果实的生长发育调控,且与生长素信号转导过程相关。  相似文献

Cactus and succulent Euphorbia plants have green stems that are exposed to sunlight over long periods. Previous research demonstrated that barking injuries occur to stem surfaces of 20 species of cacti in the Americas. For these 20 cactus species the amounts of injuries were related to annual amounts of sunlight exposure on surfaces. Cacti with many injuries had high mortality rates. Data herein show that stem surfaces of 20 species of Euphorbia in South Africa have identical surface injuries as for cacti. Euphorbia plants at 30°S show a mean ratio of scale and bark injuries of 3.5:1 of equatorial-facing to polar-facing surfaces, a ratio is similar to the 4:1 of annual sunlight exposure ratio at that latitude. Results show that Euphorbia stems have injuries on younger tissues than cactus species. Thus, Euphorbia stems are more sensitive than cactus stems. Euphorbia species with most stem injuries had smooth (non-undulate) surfaces, a one-celled epidermal cell layer only, cortex cells that abutted the epidermis, and initial injuries that involved only epidermal cells. This is the first report of identical stem surface injuries to Euphorbia species that are identical to surface injuries to cactus species worldwide. Moreover, the amounts on stem injuries on Euphorbia and long-lived, columnar cactus species are highly correlated with relative amounts of sunlight on stem surfaces over the annual cycle. These results suggest that these species are good bio-indicators of annual sunlight exposure.  相似文献

为了解柿属(Diospyros)植物的叶表皮特征,在光学显微镜下观察了46种2变种植物的叶表皮微形态特征,并进行了主成分分析。结果表明,柿属植物的叶表皮微形态可分为2组:A组的表皮细胞为不规则形,垂周壁浅波状或深波状;B组的表皮细胞为多边形,垂周壁平直或弓形。气孔全部位于下表皮,以无规则型为主,但同时有辐射型、环列型和十字型气孔存在。大多数种具有表皮毛,表皮毛有非腺毛和头状腺毛的区别。柿属植物的叶表皮微形态特征类型多样,能够为部分类群的分类提供较好的形态学证据。  相似文献

New specimens of the early Oligocene (32 million years ago) stem group hummingbird Eurotrochilus inexpectatus Mayr, 2004, are described from the type locality Frauenweiler in Southern Germany. One of these is the second slab of the holotype, whose existence has been hitherto unknown. The new fossils add significantly to our knowledge of the morphology of E. inexpectatus and allow more detailed comparisons with other stem and crown group Trochilidae. A new apomorphy of the clade (Eurotrochilus + crown group Trochilidae) is described. With species successively more closely related to the crown group, the temporal sequence of stem group hummingbirds in Europe indicates an Old World origin of Pan-Trochilidae. E. inexpectatus is the earliest long-beaked stem group hummingbird with hovering capabilities, and absence of modern-type hummingbirds before the early Oligocene may provide a maximum age for the origin of hummingbird-pollinated plants and the colonization of the New World by modern-type stem group hummingbirds.  相似文献

Reexamining the classification proposed by Hurusawa, the phylogeny of Far EasternEuphorbia subgenusEsula was analyzed using thirteen morphological and seventeen phenolic compound data. These data were analyzed independently and in combination using PAUP under the assumptions of Fitch parsimony. Ten species, comprised of three sections and five subsections within Far EasternEuphorbia subg.Esula and one outgroup from subg.Chamaesyce, were used as terminal taxa. The phylogenetic results did not support the sectional classifications within subg.Esula proposed by Hurusawa. SectionDecussatae was nested in the paraphyletic sectEsula in all of the analyses, and the relationship of sectHelioscopiae was equivocal among data sets. The disagreement of data sets over the placement ofEuphorbia ebracteolata is probably due to a hybrid origin of the species and missing phenolic data forE. pallasii. A sister-group relationship of the Korean endemicE. fauriei with the widespreadE. pekinensis was strongly supported by the morphological and phenolic data.  相似文献

The tricolporate pollen grains of 38 Mediterranean and Macaronesian species ofEuphorbia L. andChamaesyce S. F. Gray have a special apertural sporoderm not found in the other taxa of theEuphorbiaceae. At the apertural margo the ectexine is thinner because of shorter columellae and the thin, fragmented or even absent foot-layer. Ectexinous granules, mixed with endexinous material, are present near the ora. The endexine is homogeneous and thickened under the colpi (at the end and at the proximity of the end of colpus). Around the ora, the endexine is granulate and lamellar with irregular cavities. The apertural intine presents a characteristic structure with thickenings running along both sides of the colpi. The arrangement and structure of these intinous thickenings depend on the distance from the ora. This special morphology of the intine is present in all taxa studied here. The genusEuphorbia is considered to be the most evolved taxon of this family. The characteristic apertural sporoderm may be an adaptative modification to different physiological conditions, so it may present an apertural mechanism which is more adapted to harmomegathic changes and thus facilitate the germination and the formation of the pollen tube.  相似文献

Human activities can promote increased hybridization in the genus Eucalyptus with potentially detrimental consequences for the persistence of rare species. However, many hybrid combinations have not been investigated with combined use of genetic markers and morphology. We assessed the efficiency of the STRUCTURE program and morphological intermediacy for identifying hybrids between the uncommon tree, Eucalyptus aggregata, which putatively hybridizes with the common congeners, E. rubida and E. viminalis in south-eastern Australia. We sampled 1,005 seedlings across 27 populations, all seedlings were genotyped at 6 allozyme loci and scored for 22 stem and leaf characters. Both marker sets confirmed that E. aggregata is hybridizing with both E. rubida and E. viminalis. Allozymes revealed hybrids from E. aggregata trees in 88% of populations and hybrids comprised 7.3% of all seedlings. Both genetics and morphology indicated that ~50% were likely to be F1 hybrids, and both simulations and morphological characteristics indicated that the remainder were mostly backcrosses. Morphological analysis correctly distinguished 71% of F1 hybrids from parentals and was least accurate when dealing with potential backcrosses (50% success). Hence, techniques using genetic data (no prior information) and the assessment of appropriate admixture thresholds through simulations provided the most accurate estimates of hybrid frequency. In this study, potential introgression and the high frequency of hybrids in small populations (~30%), suggests that hybridization should be considered in the management and conservation of E. aggregata.  相似文献
