Several species in Aspergillus section Nigri have been reported to produce sclerotia on well-known growth media, such as Czapek yeast autolysate (CYA) agar, with sclerotia considered to be an important prerequisite for sexual development. However Aspergillus niger sensu stricto has not been reported to produce sclerotia, and is thought to be a purely asexual organism. Here we report, for the first time, the production of sclerotia by certain strains of Aspergillus niger when grown on CYA agar with raisins, or on other fruits or on rice. Up to 11 apolar indoloterpenes of the aflavinine type were detected by liquid chromatography and diode array and mass spectrometric detection where sclerotia were formed, including 10,23-dihydro-24,25-dehydroaflavinine. Sclerotium induction can thus be a way of inducing the production of new secondary metabolites from previously silent gene clusters. Cultivation of other species of the black aspergilli showed that raisins induced sclerotium formation by A. brasiliensis, A. floridensis A. ibericus, A. luchuensis, A. neoniger, A. trinidadensis and A. saccharolyticus for the first time.  相似文献

Stenocarpella maydis is a fungal pathogen of major importance that causes a dry-rot of maize ears and is associated with a neuromycotoxicosis in cattle grazing harvested maize fields in southern Africa and Argentina. In an effort to investigate the potential roles of S.?maydis metabolites in the fungal disease cycle, ethyl acetate extracts of solid-substrate fermentations of several S. maydis isolates from maize grown in the United States were found to exhibit significant phytotoxic, antifungal, and antiinsectan activity. Chemical investigations of extracts of S. maydis isolates from Illinois and Nebraska led to the isolation or detection of the known metabolites diplodiatoxin, chaetoglobosins K and L, and (all-E)-trideca-4,6,10,12-tetraene-2,8-diol as major components. A culture of Stenocarpella macrospora from maize grown in Zambia produced diplosporin and chaetoglobosins K and L as major components that were isolated. Diplodiatoxin produced significant lesions in a maize leaf puncture wound assay. Diplosporin and chaetoglobosin K displayed moderate antiinsectan activity in dietary assays against the fall armyworm Spodoptera frugiperda, while chaetoglobosin K exhibited significant antifungal activity against Aspergillus flavus and Fusarium verticillioides. Using LC-ESIMS and (1)H NMR data, diplodiatoxin was detected as a major component in S. maydis-rotted grain, stalks, and stalk residues. This constitutes the first report of chaetoglobosins K and L from S. maydis, of (all-E)-trideca-4,6,10,12-tetraene-2,8-diol from Stenocarpella, and the first reported detection of diplodiatoxin, or any other Stenocarpella metabolite, in diseased maize seeds and stalk tissues.  相似文献

A new species, Eupenicillium bovifimosum, was isolated from dry cow manure collected in Wyoming, USA. The outstanding morphological characteristics of this species are its robust, dense penicilli bearing long, broad columns of conidia, and its smooth-walled, unflanged ascospores which are produced within 3 wk of inoculation onto MEA. The new species produces penicillic acid, a fumagillin-like molecule, patulodin-like compounds CK2108A and CK2108B, and a compound resembling 2-[(2-hydroxypropionyl)amino]benzamide. Phylogenetic analysis using 1100 bases of the ITS and LSU DNA regions indicates that E. bovifimosum is most closely related to E. baarnense, and also to E. crustaceum, E. egyptiacum, and E. tularense.  相似文献

Various fractions were extracted from cell-wall material of Eupenicillium crustaceum, Penicillium brevi-compactum, P. decumbens, Aspergillus flavipes and A. ochraceus. The most characteristic fractions, which may have chemotaxonomic relevance, were F1I, an alpha-(1-3) glucan (alkalisoluble, water-insoluble), which amounted to 16.2-32.5% of the cell-wall material, and F1S (alkali and water-soluble) which represented 2.5-6.2% of the cell-wall material and was identified as a beta-(1-5) galactan. 13C-NMR spectra of the F1S fractions showed the same pattern for all the fungal species, characteristic of beta-(1-5) linked galactofuranose.  相似文献

研究了耐冷皮壳正青霉Eupenicillium crustaceum一种木聚糖酶的纯化和酶学性质。采用硫酸铵沉淀和阴离子交换层析的方法,从耐冷皮壳正青霉液体发酵液中分离纯化出一种亚基分子量35kDa的木聚糖酶。酶学性质研究表明,酶的最适pH值为5.5,在pH4.5-6.5范围内具有较高的催化活性。最适温度为50℃,20℃下酶活为最高酶活的40%。Ag+和Fe2+大幅度提高木聚糖酶的酶活,而Mn2+和Hg2+强烈抑制木聚糖酶的活性。同时,该木聚糖酶具有严格的底物特异性。  相似文献

Abstract

There are many diseases linked to oxidative stress, including cancer. Importantly, endogenous antioxidants are insufficient to protect against this process. Peptides derived from food proteins produced by hydrolysis have been investigated as exogenous antioxidants. The present study aimed to identify novel peptides with antioxidant potential produced from egg and milk proteins hydrolysis with two new fungal proteases isolated from Eupenicillium javanicum and Myceliophthora thermophila. The degree of hydrolysis at several time points was calculated and correlated to DPPH scavenging and metal chelating assays, all hydrolysates presented antioxidant activity. Casein hydrolyzed by the M. thermophila protease showed the best antioxidant activity. The identified sequences showed that the proportions of amino acids that influence antioxidant activity support the antioxidant assay. Our data reveal the conditions necessary for the successful generation of antioxidant peptides using two novel fungal proteases. This opens a potential new avenue for the design and manufacture of antioxidant molecules.  相似文献

Forty-three strains of Eupenicillium tropicum sp. nov. were isolated from soils collected in India, Costa Rica and Galapagos, Ecuador. The species is characterized by biverticillate penicilli, slightly rough, subglobose to ovate conidia, brownish cleistothecia that become brown-gray with age, and ascospores with two equatorial flanges and slightly roughened valves. All strains produced a large number of indole alkaloids, and many types of unknown secondary metabolites with characteristic chromophores were produced by a majority of strains. Eupenicillium tropicum is morphologically most similar to E. shearii, but based on ITS-LSU sequences, is most closely related to Penicillium citrinum, P. sartoryi and P. westlingii. Eupenicillium shearii strains consistently produce paxillin, paspalinine and shearinins, while the latter three penicillia all produce citrinin consistently.  相似文献

Eupenicillium parvum was recorded for first time during isolation of phosphate-solubilizing microorganisms from the tea rhizosphere. The fungus developed a phosphate solubilization zone on modified Pikovskaya agar, supplemented with tricalcium phosphate. Quantitative estimation of phosphate solubilization in Pikovskaya broth showed high solubilization of tricalcium phosphate and aluminium phosphate. The fungus also solubilized North Carolina rock phosphate and Mussoorie rock phosphate, and exhibited high levels of tolerance against desiccation, acidity, salinity, aluminium, and iron. Solubilization of inorganic phosphates by the fungus was also observed under high stress levels of aluminium, iron, and desiccation, though the significant decline in phosphate solubilization was marked in the presence of aluminium than iron. The fungal isolate showed 100 % identity with E. parvum strain NRRL 2095 ITS 1, 5.8S rRNA gene and ITS 2, complete sequence; and 28S rRNA gene, partial sequence.  相似文献

Enterobacter cloacae S11: T:07 (NRRL B-21050) is a promising biological control agent that has significantly reduced both fungal dry rot disease and sprouting in laboratory and pilot potato storages. The metabolites phenylacetic acid (PAA), indole-3-acetic acid (IAA), and tyrosol (TSL) were isolated from S11:T:07 liquid cultures provided with three different growth media. The bioactivities of these metabolites were investigated via thin-layer chromatography bioautography of antifungal activity, wounded potato assays of dry rot suppressiveness, and cored potato eye assays of sprout inhibition. Relative accumulations of PAA, IAA, and TSL in cultures were nutrient dependent. For the first time, IAA, TSL, and PAA were shown to have antifungal activity against the dry rot causative pathogen Gibberella pulicaris, and to suppress dry rot infection of wounded potatoes. Disease suppression was optimal when all three metabolites were applied in combination. Dosages of IAA that resulted in disease suppression also resulted in sprout inhibition. These results suggest the potential for designing culture production and formulation conditions to achieve a dual purpose biological control agent able to suppress both dry rot and sprouting of stored potatoes.  相似文献

Kidney homogenates from chicks fed a vitamin D-deficient diet for 10 days and supplemented with 6.5 nmol of vitamin D3 48 hr prior to sacrifice metabolized invitro [3H]-25-hydroxyvitamin D3 (25-OH-D3) to 24,25-dihydroxyvitamin D3 [24,25-(OH)2-D3] and 3 other metabolites (peaks A, C and E). When the homogenates were incubated with purified [3H]-24,25-(OH)2-D3, 3 similar metabolites (peaks A′, C′ and E′) were produced. On high pressure liquid chromatography, peaks A, C and E migrated to exactly the same respective positions as peaks A′, C′ and E′. Kidney homogenates from D-deficient chicks failed to produce these metabolites from [3H]-25-OH-D3 or [3H]-24,25-(OH)2-D3. These results strongly suggest that the new metabolites reported here are synthesized via 24,25-(OH)2-D3 in the kidney of chicks supplemented with vitamin D3.  相似文献

Rhizopus oryzae strain NBRC 4707 produced lactic acid and ethanol more efficiently than strain NRRL 395 in potato pulp, an agricultural by-product of the starch industry. The two strains developed comparable activities of xylanase, cellulase, -amylase, and glucoamylase, while the polygalacturonase activity of strain NBRC 4707 was double that of strain NRRL 395. The addition of commercial pectinase enhanced the formation of metabolites, suggesting that the degradation of pectic substances determines the fermentation of potato pulp by R. oryzae. Orange and apple peel were more effective in the induction of polygalacturonase activity than potato pulp, sugarbeet pulp, or wheat bran when used as a principal carbon source for fungal growth in a solid-state culture. The fungal cells in both types of fruit peel stimulated the fermentation of potato pulp and increased the quantity of lactic acid and ethanol to higher levels than those in other agricultural by-products.  相似文献

The effects of beauverolide L and cyclosporin A, cyclic peptidic metabolites, produced by several genera of entomopathogenic fungi on immune responses of last instar larvae of the greater wax moth Galleria mellonella have been examined. Intrahemocoelic injection of either metabolite-coated silica particles or dissolved metabolites in a concentrations ranging between 10 and 30 μg per larva caused no mortality but activated humoral responses in G. mellonella larvae. The challenge induced a significant release of lysozyme and cecropin-like activity into the hemolymph, suggesting stimulatory activity on humoral immune responses. Injected metabolite-coated particles were rapidly surrounded by hemocytes which subsequently accomplished formation of melanized nodules, which increased in size and number compared with controls. In vitro assays with dissolved metabolites indicated no adverse effects of beauverolide L or cyclosporin A on attachment or spreading of isolated plasmatocytes but dose-dependent inhibition of their phagocytic activity. Isolated plasmatocytes incubated with cyclosporin A or beauverolide L exhibited cytoskeleton alterations that differed from those observed in plasmatocytes from infected G. mellonella larvae or reported from other fungal secondary metabolites. The experiments provided further data to elucidate the role of fungal secondary metabolites in development of mycoses in insects.  相似文献

Colletotrichum truncatum (Schwein.) Andrus and Moore NRRL 13737 (= NRRL 18434) is a fungal plant pathogen which shows promise as a bioherbicide against the troublesome weed Sesbania exaltata (Raf.) Rydb. ex A. W. Hill. Previous studies showed similar amounts of spores were produced/ml of medium in liquid and solid-state fermentations. In this study, Colletotrichum truncatum spores were produced in liquid (LC), solid/vermiculite (SV), and solid/perlite-cornmeal-agar (SP). After drying at room temperature with flowing air, SV and SP retained the most viability. Each product was then stored at 4 degrees, 15 degrees, and 25 degrees C. All three products stored at 4 degrees C and SP stored at 15 degrees C retained highest viability. Efficacy based upon assays utilizing equal numbers of viable spores showed SV and SP spores incited more severe disease symptoms than LC spores.  相似文献

Phanerochaete chrysosporium NRRL 6361 and Pleurotus pulmonarius CBS 664.97 were tested for their ability to grow under nonsterile conditions and to degrade various aromatic hydrocarbons in an aged contaminated soil that also contained high concentrations of heavy metals. After 24 days fungal incubation, carbon-CO2 liberated, an indicator of microbial activity, reached a plateau. At the end of the incubation time (30 days), fungal colonization was clearly visible and was confirmed by ergosterol and cell organic carbon determinations. In spite of unfavorable pH (around 7.4) and the presence of heavy metals, both fungi produced Mn-peroxidase activity. In contrast, laccase and aryl-alcohol oxidase were detected only in the soil treated with P. pulmonarius CBS 664.97 and lignin-peroxidase in that with P. chrysosporium NRRL 6361. No lignin-modifying enzyme activities were present in non-inoculated soil incubated for 30 days (control microcosm). Regardless of the fungus employed, a total removal of naphtalene, tetrachlorobenzene, and dichloroaniline isomers, diphenylether and N-phenyl-1-naphtalenamine, was observed. Significant release of chloride ions was also observed in fungal-treated soil, in comparison with that recorded in the control microcosm. Both fungi led to a significant decrease in soil toxicity, as assessed using two different soil contact assays, including the Lepidium sativum L. germination test and the Collembola mortality test.  相似文献

Peanuts and other seed and grain crops are commonly contaminated with carcinogenic aflatoxins, secondary metabolites produced by Aspergillus flavus and A. parasiticus. Aflatoxin contamination of peanuts in the field can be reduced by 77–98% with biological control through the application of nontoxigenic strains of these species, which competitively exclude native aflatoxin-producing strains from developing peanuts. In this study, viable peanut seeds were artificially wounded and inoculated with field soil containing natural fungal populations that were supplemented with conidia of nontoxigenic A. flavus NRRL 21882 (niaD nitrate-nonutilizing mutant) and A. parasiticus NRRL 21369 (conidial color mutant). Increasing soil densities of applied nontoxigenic strains generally resulted in an increase in the incidence of seed colonization by applied nontoxigenic strains, a decrease in seed colonization by native A. flavus and A. parasiticus, and a decrease in aflatoxin concentration in seeds. Reduction of aflatoxins in peanut seeds depended on both the density and the aflatoxin-producing potential of native populations and on the fungal strain used for biological control. Wild-type strain A. flavus NRRL 21882 and its niaD mutant were equally effective in reducing aflatoxins in peanuts, indicating that nitrate-nonutilizing mutants, which are easily monitored in the field, can be used for evaluating the efficacy of biocontrol strains.  相似文献

The needles of Picea glauca (white spruce) and Pinus strobus (white pine) trees infected with toxigenic fungal endophytes contain varying concentrations of their secondary metabolites that are toxic to either insect pests or needle pathogens. In the present study, liquid chromatography-mass spectrometric methods to determine needle concentrations of metabolites of four endophyte species were developed. The endophytes considered were a Phialocephala sp. (vermiculine) and Phialocephala scopiformis (rugulosin) from white spruce, as well as a Xylaria sp. (griseofulvin) and Lophodermium nitens (pyrenophorol) from white pine needles. To ensure that needles were infected with the associated fungal endophyte, suitable qPCR-based methods were also developed. There was a high degree of concordance between the qPCR analysis of the fungal mycelium and the LC-MS/MS quantification of the associated metabolites. Concentrations of the antifungal compounds griseofulvin and pyrenophorol were present in amounts that affect conifer needle diseases including white pine blister rust caused by Cronartium ribicola. Similarly, concentrations of the antiinsectan compounds vermiculine and rugulosin were in the range known to reduce the growth of Choristoneura fumiferana and mitigate foliage damage.  相似文献

Abstract Electroantennograms were recorded from the antennae of adult male and female corn earworms, Heliothis zea (Boddie). A total of seventeen female moth sex pheromone components from several species were tested. Of these, two components elicited significantly greater responses than the other fifteen. These were (Z)-11-hexadecenal, a conspecific component, and (Z)-9-tetradecenal, a component found in the pheromone blend of a sympatric species H.virescens (F.) that inhibits attraction of H.zea males. The results from dose-response and selective adaptation studies indicate that there are separate populations of receptors for these two chemical signals on the antenna of male H.zea. The more sensitive population is selective for (Z)-11-hexadecenal, while the less sensitive one responds to (Z)-9-tetradecenal. These findings provide a physiological basis by which H.zea males can distinguish the interspecific repellent from the conspecific pheromone blend. It is likely that this discrimination contributes to reproductive isolation between these two species.  相似文献

The mycoparasite Humicola fuscoatra NRRL 22980 was isolated from a sclerotium of Aspergillus flavus that had been buried in a cornfield near Tifton, Ga. When grown on autoclaved rice, this fungus produced the antifungal metabolites monorden, monocillin IV, and a new monorden analog. Each metabolite produced a clear zone of inhibition surrounding paper assay disks on agar plates seeded with conidia of A. flavus. Monorden was twice as inhibitory to A. flavus mycelium extension (MIC > 28 μg/ml) as monocillin IV (MIC > 56 μg/ml). Cerebrosides C and D, metabolites known to potentiate the activity of cell wall-active antibiotics, were separated from the ethyl acetate extract but were not inhibitory to A. flavus when tested as pure compounds. This is the first report of natural products from H. fuscoatra.  相似文献
