The continuous production of large numbers of high quality gametes is essential for aquaculture, particularly in candidate species, such as pikeperch, Sander lucioperca (L.). The common practice of year‐round reproduction is under suspicion of inflicting adverse effects on the quality of the gametes through the disturbance of endogenous rhythms. We hypothesized that such perturbation does not affect RAS‐reared F‐generation broodstock. Reproductive performance (number of eggs) and gamete quality (fertilization and hatching rate) were assessed over the course of 3 years covering six independent, photothermal shifted spawning seasons in a commercial pikeperch hatchery (n = 31 egg batches of F‐generation fish in total). No substantial differences in fertilization or hatching rates could be detected between the individual spawning seasons. Fecundity varied, but there are indications for a size effect on female fecundity with intermediate sized females producing higher number of eggs (~65–70 cm). Low egg quality could be detected in batches of very large fish. In conclusion, size‐specific broodstock composition, but not year‐round reproduction of F‐generation pikeperch spawners affects the reproductive performance.  相似文献

This study examines the metabolic fate of total and individual yolk fatty acids (FA) during the embryonic development of the king penguin, a seabird characterized by prolonged incubation (53 days) and hatching (3 days) periods, and a high n-3/n-6 polyunsaturated FA ratio in the egg. Of the approximately 15 g of total FA initially present in the egg lipid, 87% was transferred to the embryo by the time of hatching, the remaining 13% being present in the internalized yolk sac of the chick. During the whole incubation, 83% of the transferred FA was oxidized for energy, with only 17% incorporated into embryo lipids. Prehatching (days 0-49), the fat stores (triacylglycerol) accounted for 58% of the total FA incorporated into embryo lipid. During hatching (days 49-53), 40% of the FA of the fat stores was mobilized, the mobilization of individual FA being nonselective. At hatch, 53% of the arachidonic acid (20:4n-6) of the initial yolk had been incorporated into embryo lipid compared with only 15% of the total FA and 17-24% of the various n-3 polyunsaturated FA. Similarly, only 32% of the yolk's initial content of 20:4n-6 was oxidized for energy during development compared with 72% of the total FA and 58-66% of the n-3 polyunsaturated FA. The high partitioning of yolk FA toward oxidization and the intense mobilization of fat store FA during hatching most likely reflect the high energy cost of the long incubation and hatching periods of the king penguin. The preferential partitioning of 20:4n-6 into the structural lipid of the embryo in the face of its low content in the yolk may reflect the important roles of this FA in tissue function.  相似文献

Although the fatty acid (FA) requirements of marine teleosts not only are a function of the amount of each FA individually, but also of the relative proportions of the FAs; mostly, the interactions have been ignored and merely limited to a few interrelations of 20:4[n-6], 20:5[n-3], and 22:6[n-3]. To address this shortcoming, viable eggs of Dentex dentex were obtained from broodfish in captivity. Nine viability parameters (VP) (i.e., floating rate (FR), hatching rate (HR), and survival rate (SR) from day 0 to 5 posthatch (dph)) that are currently used in mariculture systems were determined. Egg FAs were characterized and quantified. One hundred and twenty ratios were made based on the FA contents estimated. Sixty-four ratios were significantly and strongly correlated to embryo/larva success through 201 simple regression models (r(2) = 0.640-0.948; P = 0.006-P < 0.001). Of the 201 significant relationships found, -12, -5, -2, -3, -22, -23, ~21, and -13% show the relations of the egg FA ratios with FR, HR, and SR at 0, 1, 2, 3, 4, and 5?dph, respectively. All the FAs characterized in this study were significantly correlated to VPs through either their individual relative proportions or relative proportion of their combinations. This study, for the first time, presents a series of new FA interrelations and uncovers their biological meanings under both basic and applied aspects through correlating them to embryo/larva success.  相似文献

Morphology and physiology of fish embryos undergo dramatic changes during their development until the onset of feeding, supplied only by endogenous yolk reserves. For obtaining an insight how these restructuring processes are reflected by body mass related parameters, dry weights (dw), contents of the elements carbon and nitrogen and lipid and protein levels were quantified in different stages within the first four days of embryo development of the zebrafish (Danio rerio). The data show age dependent changes in tissue composition. Dry weights decreased significantly from 79μgdw/egg at 0hours post fertilization (hpf) to 61 μgdw/egg after 96 hpf. The amounts of total carbon fluctuated between 460 mg g-1 and 540 mg g-1 dw, nitrogen was at about 100 mg g-1 dw and total fatty acids were between 48–73 mg g-1 dw. In contrast to these parameters that remained relatively constant, the protein content, which was 240 mg g-1 at 0 hpf, showed an overall increase of about 40%. Comparisons of intact eggs and dechorionated embryos at stages prior to hatching (24, 30, 48 hpf) showed that the differences seen for dry weight and for carbon and nitrogen contents became smaller at more advanced stages, consistent with transition of material from the chorion to embryo tissue. Further, we determined the effect of 2,4-dinitrophenol at a subacutely toxic concentration (14 μM, LC10) as a model chemical challenge on the examined body mass related parameters. The compound caused significant decreases in phospholipid and glycolipid fatty acid contents along with a decrease in the phospholipid fatty acid unsaturation index. No major changes were observed for the other examined parameters. Lipidomic studies as performed here may thus be useful for determining subacute effects of lipophilic organic compounds on lipid metabolism and on cellular membranes of zebrafish embryos.  相似文献

An association between dietary fish oil and decreased yolk weight and reduced sensory quality of eggs has been reported when eggs are enriched with n-3 FA from fish oil. Seaweeds are an important source of compounds that seem to increase egg weight when included in the laying hen diet. The objectives of this study were to determine the influence of the dietary seaweeds Macrocystis pyrifera, Sargassum sinicola and Enteromorpha sp. on the physical quality, lipid composition and consumer acceptability of n-3 FA enriched eggs. One-hundred and forty-four 35-week-old Leghorn hens were randomly distributed in four treatments that consisted of the inclusion of 2% of sardine oil (SO) and 10% of each marine alga (MA) in laying hens' diets; a control diet (C) was also prepared. The study lasted 8 weeks and egg physical quality, egg lipids and sensory attributes were evaluated. The results showed that incorporation of 10% M. pyrifera in the diets is an effective way of increasing the n-3 FA content, the albumen height and yolk color, but not the egg weight, when these are enriched with n-3 FA from fish oil. The egg flavor was also not affected.  相似文献

This study aims to determine the production characteristics and performance of rainbow trout hatcheries in Seydikemer, Mu?la, Turkey where over half of the total production of eyed eggs and juveniles take place. For this purpose six hatcheries with different production scales were selected and coded A, B, C, D, E and F (with capacities of 60, 15, 5.6, 4, 0.95 and 0.6 million juveniles per year, respectively). From each hatchery, 20 females were selected and their egg qualities (egg number, diameter and weight, fertilization, eyed egg, hatching, swim‐up fry and survival rates at 120 days after hatching) and juvenile growth rates were monitored until the 120th day after hatching under each farms own conditions. The sperm characteristics of 10 broodstock males from each hatchery were also examined. Although fertilization and eyed egg rates were similar among the farms, hatching, swim‐up fry, and survival rates at the 120th day after hatching differed significantly. The average eyed egg, hatching and survival rates (calculated from selected 20 females) were 72, 55, and 32%, respectively. While sperm characteristics except duration of motility (s), were significantly different among the farms, average sperm concentration (per ml), motility (%) and duration of motility (s) in the region were 11.8 × 109, 55.3, and 56.4, respectively. The best hatchery performance was observed in Farm A because of better records and management applications than in the other farms.  相似文献

To enable cryopreservation of fish semen to become an efficient, routine technique, much more detailed information is required. Therefore, the present study was conducted to investigate various fertilization techniques and media, straw volumes as well as optimal semen volume for cryopreservation. The bleak (Chalcalburnus chalcalburnus) was used as the main model for investigation. Using frozen-thawed semen the fertilization rate was similar up to the morula stage, independent of the fertilization technique. Thereafter, in egg batches fertilized using the wet technique most embryo development stopped. In egg batches fertilized using the dry technique, embryonic development proceeded normally. For cryopreserved semen full activation of sperm motility was obtained at ratios of fertilization media (hatchery water and all tested types of saline solutions) to semen of 10:1. Sperm motility rate was much higher in the saline solutions than in water. In contrast hatching rates were higher when water was used as fertilization medium. Therefore, the requirements necessary for optimal sperm motility and optimal sperm-egg contact were different and so for these parameters optimal levels could not be achieved. When adjusting the freezing and thawing conditions 0.5ml straws as well as larger straws (1.2ml) proved suitable for cryopreservation of cyprinid semen. The highest fertilization rates were obtained with sperm to egg ratios of (1.3-2.5) x 10(6):1 and were 77-92% of fresh semen control. This was also similar for Ch. nasus, R. meidingerii, B. barbatus and C. carpio and suggests that the cryopreservation requirements of spermatozoa are not species specific.  相似文献

Yolk fatty acid (FA) concentrations and egg quality were evaluated with respect to the inclusion of different levels of marine fish oil (MFO) and linseed oil (LO) in a basal diet. Ten diets were arranged factorially with two levels of MFO addition (15 and 17 g/kg) and five levels of linseed oil (1, 2, 3, 4 and 5 g/kg). Two commercial feeds with no n-3 FA sources added, or containing 15 g MFO/kg served as controls for egg sensorial quality traits. Type of diet did not affect egg production traits, nor total fat content in yolk or saturated and monounsaturated FA concentration in yolk fat. An increase in the inclusion of MFO from 15 to 17 g/kg increased (P<0.05) egg yolk content of total n-3 FA, C20:5 n-3 and C22:6 n-3 by 3.3, 9.8 and 3.5%, respectively. Linseed oil addition increased linearly (P<0.001) the level of total n-3 FA and the proportion of C18:3 n-3, C20:5 n-3, C22:5 n-3 and C22:6 n-3 retained in yolk fat by respectively 131, 12.0, 6.9 and 20.7% between extreme diets. Efficiency of retention of n-3 FA decreased (P<0.001) with dietary level of n-3 FA, and with the dietary ratio of MFO to LO. Both MFO and LO addition decreased (P<0.001) C20:4 n-6 to C18:2 n-6 ratio in yolk. Sensorial quality of eggs was not affected by treatments and did not differ with respect of that obtained in commercial n-3 FA enriched eggs. No interaction was found between LO and MFO addition for any of the traits studied. Regression equations have been calculated in order to predict efficiency of retention and yolk fat content of LC n-3 and n-6 FA from dietary characteristics.  相似文献

目的1-苯基-2-硫脲(PTU)可抑制斑马鱼胚胎黑色素的产生,保持斑马鱼透明,便于形态观察和信号检测。本文研究了PTU对斑马鱼胚胎发育的影响和抑制斑马鱼胚胎黑色素生成,保持斑马鱼透明性的最佳浓度。方法用不同浓度PTU处理23hpf(受精后,hourspostfertilization,hpf)斑马鱼胚胎,作用57h后观察80hpf斑马鱼的形态学、生理学改变,计算死亡率和孵化率,测量心率和静脉窦-动脉球之间的距离。结果浓度为0.197mmo]/L、0.296mmol/LPTU可以有效抑制黑色素生成,保持斑马鱼整体透明,对斑马鱼心血管系统结构和生理功能无影响,且不影响斑马鱼正常孵化过程。随着PTU浓度的增加,斑马鱼死亡率增加,孵化率下降,出现心包水肿,心脏畸形等改变,心率下降,静脉窦-动脉球之间的距离增大。结论浓度不高于0.296mmol/L的PTU溶液能有效抑制斑马鱼黑色素生成,对斑马鱼心血管毒性研究无影响。  相似文献

The mechanisms controlling egg diapause and circadian rhythms of hatching activity have been extensively studied in insects. However, relatively little attention has been paid to the mechanisms controlling synchronized hatching from an egg mass. In this study, we examined the possible involvement of embryo–embryo interaction in controlling hatching time in Halyomorpha halys (Stål). Eggs tended to hatch earlier as the egg mass size increased. Egg separation and clumping of separated eggs at various times showed that hatching synchrony was largely determined shortly before hatching. However, whether eggs were kept in a mass or separated until several hours before hatching also influenced the hatching time, indicating the presence of embryo–embryo interactions. Eggs derived from different masses and kept in physical contact with one another hatched synchronously if their ages were within approximately 8 h. In this case, both younger and older eggs advanced only in hatching time, in contrast to a case of locusts reported by others. Eggs separated by more than 7 mm hatched as synchronously as those kept in a mass when glued to the same substrate, suggesting an important role of the egg substrate in transmitting the vibrational hatching signals to neighboring sibling eggs to synchronize hatching.  相似文献

ABSTRACT. 1. We investigated the influence of larval density on egg hatching in the treehole mosquito, Aedes triseriatus Say (Diptera: Culicidae) by submerging laboratory-produced egg batches in natural habitats stocked with fixed densities of larvae.
2. We found a significant decrease in hatching rate as larval density increased. The results suggest that inhibited eggs refrain from hatching into environments in which competitive effects of large larvae are potentially severe, and in which cannibalism may occur.
3. The amount of time the egg batches were exposed to experimental conditions (between 2 and 16 days) had no significant influence on hatch rate.  相似文献

The quality of day old chicks is crucial for profitable broiler production, but a difficult trait to define. In research, both qualitative and quantitative measures are used with variable predictive value for subsequent performance. In hatchery practice, chick quality is judged on a binomial scale, as chicks are divided into first grade (Q1-saleable) and second grade (Q2) chicks right after hatch. Incidences and reasons for classifying chicks as Q2, and potential of these chicks for survival and post-hatch performance have hardly been investigated, but may provide information for flock performance. We conducted an experiment to investigate (1) the quality of a broiler flock and the relation with post-hatch flock performance based on a qualitative score (Pasgar©score) of Q1 chicks and based on the incidence of Q2 chicks and (2) the reasons for classifying chicks as Q2, and the potential of these chicks for survival and post-hatch growth. The performance was followed of Q1 and Q2 chicks obtained from two breeder flocks that hatched in two different hatching systems (a traditional hatcher or a combined hatching and brooding system, named Patio). Eggs were incubated until embryo day 18, when they were transferred to one of the two hatching systems. At embryo day 21/post-hatch day 0, all chicks from the hatcher (including Q2 chicks) were brought to Patio, where the hatchery manager marked the Q2 chicks from both flocks and hatching systems and registered apparent reasons for classifying these chicks as Q2. Chick quality was assessed of 100 Q1 chicks from each flock and hatching system. Weights of all chicks were determined at days 0, 7, 21 and 42. There were no correlations between mean Pasgar©score and post-hatch growth or mortality, and suboptimal navel quality was the only quality trait associated with lower post-hatch growth. Growth was clearly affected by breeder flock and hatching system, which could not be linked to mean Pasgar©score or incidence of Q2 chicks. Q2 chicks showed lower post-hatch growth compared to Q1 chicks but effects on flock performance at slaughter weight were limited because early mortality in Q2 chicks was high (62.50% at 7 days). We concluded that chick qualitative scores and the incidence of Q2 chicks may be informative for the quality of incubation, but are not predictive for post-hatch flock performance. Culling Q2 chicks after hatch is well-founded in terms of both animal welfare and profitability.  相似文献

Yolk fat fatty acid (FA) concentrations, sensory quality and firmness of eggs and laying hen performance were evaluated with respect to the combined inclusion in the diet of conjugated linoleic acid (CLA), high n-3 oil sources and high-oleic sunflower oil (HOSO). Nine diets were arranged factorially, with three levels of n-3 FA supplementation (2.9, 3.7 and 4.5 g/kg) from three different sources (two fish oils highly concentrated in eicosapentanoic (EPA) or docosahexanoic acid (DHA) and one algae oil with a very high-DHA content) in diets added with fixed amounts of CLA (2.5 g/kg) and HOSO (30 g/kg). A commercial feed with no CLA, n-3 or HOSO added, and another one containing 4.5 g/kg of high-DHA fish oil but not CLA or HOSO were also formulated. An increase in n-3 FA supplementation had little effect on proportions of CLA, monounsaturated FA, saturated FA or total polyunsaturated FA in yolk fat, but increased (P<0.005) long-chain n-3 FA and decreased (P<0.001) long-chain n-6 FA. An increment of dietary n-3 FA also impaired linearly (P<0.001) egg acceptability by consumers. An increment in the proportion of DHA with respect to total n-3 FA from 0.28 to 0.96 increased yolk concentrations of DHA (P<0.001) and total n-3 FA (P<0.01), but decreased (P<0.001) concentrations of EPA and docosapentanoic acid FA. Current data indicate that addition of HOSO to diets supplemented with moderate amounts of CLA and n-3 FA allows the production of double enriched eggs while maintaining sensory quality for consumers at acceptable levels.  相似文献

The aim of the study was to improve the elimination of egg stickiness in tench as an attempt to increase the egg hatching rate. The usual desticking procedure involving treatment with milk solution/clay suspension resulted in a hatching rate of 74.1%. The highest hatching rate of 87.1% was found with a 10.0 ml L?1 enzyme treatment; hatching rates of 85.2 and 85.1% were recorded at 15.0 and 5.0 ml L?1, but the hatching rate decreased to 80.0% at 20.0 ml L?1. anova showed a significant effect of differing concentrations of enzyme and milk solution/clay suspension control (P < 0.0015, P < 0.0003) on the hatching rate and duration of egg incubation until hatching, respectively. Application of proteolytic alcalase enzyme, Merck EC 3.4.21.14, for desticking of tench eggs also decreased the time of egg handling from almost 1 h, necessary for traditional milk/clay desticking, to just 2 min.  相似文献

Two fitness components, development rate and egg size, were examined in six hatchery strains of rainbow trout, Oncorhynchus mykiss (syn. Salmo gairdneri Richardson), with different amounts of enzyme heterozygosity. The average expected heterozygosities per strain ranged from 4 to 8%, based upon an electrophoretic analysis of the protein products of 42 loci. Strains with higher heterozygosities had faster development rates, as measured by hatching time, than strains with lower heterozygosity. Concordance between hatching time and another measure of development rate, degree of yolk-sac resorption, suggests that hatching time is a valid measure of embryonic development rate in salmonid fishes. Earlier-hatching embryos were longer and heavier at the time of yolk-sac resorption than later-hatching fish. Females from more-heterozygous strains also had a tendency to have larger eggs. These data suggest that genetic variation is an important biological resource to be conserved in hatchery stocks.  相似文献

Avidin, an exceptionally stable protein in egg white, binds the vitamin biotin with very high affinity and can induce biotin deficiency when fed to animals. To determine if biotin bound to avidin is available to the chicken embryo, the fate of [3H]biotin complexed to avidin was monitored during embryonic development. The majority (greater than 85%) of the [3H]biotin was extraembryonic until the day before hatching, when embryos swallow egg white and withdraw the yolk sac into their abdomen. Thus, biotin in the egg white of chicken eggs contributes little to the biotin status of the chick prior to hatching. After hatching, much of the [3H]biotin was assimilated. About 30% of the total was found in the liver and kidneys by 4 days of age. The biotin in liver was associated with large proteins and not with avidin. In a separate experiment, biotin injected into the egg white of biotin-deficient eggs failed to increase embryonic development or hatchability. Both experiments suggest that biotin in egg yolk is the primary and virtually sole source of biotin for the chicken embryo.  相似文献

In the wild, water temperature cycles daily: it warms up after sunrise, and cools rapidly after sunset. Surprisingly, the impact of such daily thermocycles during the early development of fish remains neglected. We investigated the influence of constant vs daily thermocycles in zebrafish, from embryo development to sexual differentiation, by applying four temperature regimens: two constant (24°C and 28°C) and two daily thermocycles: 28:24°C, TC (thermophase coinciding with daytime, and cryophase coinciding with night-time) and 24:28°C, CT (opposite to TC) in a 12:12 h light:dark cycle (LD). Embryo development was temperature-dependent but enhanced at 28°C and TC. Hatching rhythms were diurnal (around 4 h after lights on), but temperature- and cycle-sensitive, since hatching occurred sooner at 28°C (48 hours post fertilization; hpf) while it was delayed at 24°C (96 hpf). Under TC, hatching occurred at 72 hpf, while under CT hatching displayed two peaks (at 70 hpf and 94 hpf). In constant light (LL) or darkness (DD), hatching rhythms persisted with tau close to 24 h, suggesting a clock-controlled “gating” mechanism. Under 28°C or TC, larvae showed the best performance (high growth and survival, and low malformations). The sex ratio was strongly influenced by temperature, as the proportion of females was higher in CT and TC (79 and 83% respectively), contrasting with 28°C and 24°C, which led to more males (83 and 76%). Ovarian aromatase (cyp19a) expression in females was highest in TC and CT (6.5 and 4.6 fold higher than at 28°C, respectively); while anti-müllerian hormone (amh) expression in males increased in testis at 24°C (3.6 fold higher compared to TC) and particularly at 28°C (14.3 fold increase). Taken together, these findings highlight the key role of environmental cycles during early development, which shaped the daily rhythms in fish embryo and larvae, and ultimately influenced sex differentiation.  相似文献

A statistical method for generating seasonal egg hatching profiles is applied to the brachyuran crabs Uca pugnax (Smith) and U. minax (LeConte) in New Jersey. Calibration experiments quantified the time course of egg development, using serial egg sampling at 1- to 2- day intervals from marked ovigerous females maintained in cages in the field. Egg stage was ranked from 1 to 10, based on morphological changes during development. Equations for predicting the number of days remaining until hatching from egg developmental stage were obtained from the calibration experiments, using stepwise polynomial regression. To cover the reproductive season, three consecutive calibration experiments using 15 or more females were run for U. pugnax; two for U. minax. Significant seasonal differences in the time course of egg development were detected. Weekly collections of females for each species were made; the date of larval release for each ovigerous female was predicted from the proximate calibration equation, yielding weekly hatching profiles. Weekly hatching profiles were summed to obtain seasonal hatching profiles. The average number of broods produced per female over the reproductive season was 1.9 for U. pugnax and 1.6 for U. minax. Hatching peaks for both species were associated with spring tides. The merits of this and other methods used to estimate daily variation in egg hatching of crabs are discussed.  相似文献

Disturbances in fatty acid (FA) metabolism may link chronic psychological stress, endocrine responsiveness, and psychopathology. Therefore, lipid metabolome-wide responses and their relationships with endocrine (cortisol, insulin, and adiponectin) responsiveness to acute stress (AS) were assessed in a primate model of chronic social stress (CS). Compared to controls (not exposed to CS), CS increased (P≤0.05) circulating triacylglycerol (TG) and phosphatidylethanolamine (PE) n-6/n-3 and reduced (P≤0.05) cholesterol ester (CE) 16:1n7 and phosphatidylcholine (PC) 18:1n7, suggesting lower omega-3 FA status and stearoyl-CoA desaturase activity, respectively. Cortisol responses to AS positively correlated with TG n-6/n-3 (r=0.93; P=0.007), but only in CS monkeys. The adiponectin response to AS inversely correlated with CE n-6/n3 (r=-0.89; P=0.045) and positively with TG 16:1n7 (r=0.98; P=0.004), only in CS monkeys. Our results are consistent with previously reported FA profiles in stress-related psychopathology and suggest that compositional changes of specific lipid FAs may form new functional markers of chronic psychological stress.  相似文献
