In tobacco leaves inoculated with tobacco mosaic virus (TMV), changes in chlorophyll (Chl) and carotenoid contents, parameters of slow Chl fluorescence kinetics, i.e. the maximum quantum yield of photosystem (PS2) photochemistry Fv/Fm, the effective quantum yield of photochemical energy conversion in PS2 Φ2, ratio of quantum yields of photochemical and concurrent non-photochemical processes in PS2 Fv/F0, non-photochemical quenching (NPQ), and photochemical activities of isolated chloroplasts from systemically infected tobacco leaves were investigated. We compared two successive stages of infection, the first in the stage of vein clearing at 9th day post inoculation (dpi) and the second at 22nd dpi when two different regions, i.e. light- (LGI) or dark-green (DGI) islands in the infected leaf were apparent and symptoms were fully developed. These two different regions were measured separately. The Chl and carotenoid contents in infected leaves decreased with a progression of infection and were lowest in LGI in the second stage. Also the ratio of Chl a/b declined in similar manner. The maximum quantum yield of PS2 photochemistry Fv/Fm, was decreased in the following order: first stage, DGI, and LGI. The same is true for the ratio Fv/F0. The decrease of Φ2 in infected leaves declined as compared to their controls. On the contrary, NPQ increased in infected leaves, the highest value was found in the first infection stage. Photochemical activities of the whole electron transport chain in isolated chloroplasts dramatically declined with the progression of symptoms, the lowest value was in LGI. Similarly, but to a lesser extent, the activity of PS2 in isolated chloroplasts decreased in infected leaves. Generally, the most marked impairment of the photosynthetic apparatus was manifested in the LGI of infected leaves.  相似文献

Seeds of Suaeda salsa were cultured in dark for 3 d and betacyanin accumulation in seedlings was promoted significantly. Then the seedlings with accumulated betacyanin (C+B) were transferred to 14/10 h light/dark and used for chilling treatment 15 d later. Photosystem 2 (PS2) photochemistry, D1 protein content, and xanthophyll cycle during the chilling-induced photoinhibition (exposed to 5 °C at a moderate photon flux density of 500 μmol m−2 s−1 for 3 h) and the subsequent restoration were compared between the C+B seedlings and the control (C) ones. The maximal efficiency of PS2 photochemistry (Fv/Fm), the efficiency of excitation energy capture by open PS2 centres (Fv′/Fm′), and the yield of PS2 electron transport (ΦPS2) of the C+B and C leaves both decreased during photoinhibition. However, smaller decreases in Fv/Fm, Fv′/Fm′, and ΦPS2 were observed in the C+B leaves than in C ones. At the same time, the deepoxidation state of xanthophyll cycle, indicated by (A+Z)/(V+A+Z) ratio, increased rapidly but the D1 protein content decreased considerably during the photoinhibition. The increase in rate of (A+Z)/(V+A+Z) was higher but the D1 protein turnover was slower in C+B than C leaves. After photoinhibition treatment, the plants were transferred to a dim irradiation (10 μmol m−2 s−1) at 25 °C for restoration. During restoration, the chlorophyll (Chl) fluorescence parameters, D1 protein content, and xanthophyll cycle components relaxed gradually, but the rate and level of restoration in the C+B leaves was greater than those in the C leaves. The addition of betacyanins to the thylakoid solution in vitro resulted in similar changes of Fv/Fm, D1 protein content, and (A+Z)/(V+A+Z) ratio during the chilling process. Therefore, betacyanin accumulation in S. salsa seedlings may result in higher resistance to photoinhibition, larger slowing down of D1 protein turnover, and enhancement of non-radiative energy dissipation associated with xanthophyll cycle, as well as in greater restoration after photoinhibition than in the control when subjected to chilling at moderate irradiance.  相似文献

Diurnal cycle of chlorophyll fluorescence parameters was done in Colocasia esculenta L. (swamp taro) grown in marshy land under sun or under shade. The sun leaves maintained higher electron transport rate (ETR) and steady state to initial fluorescence ratio (Fs/F0) than shade leaves. In spite of lower ETR, higher photochemical quenching (PQ), and effective quantum yield of photosystem 2 (ΦPS2) was evident in shade plants compared to plants exposed to higher irradiance. ETR increased linearly with increase in irradiance more under low irradiance (r 2 = 0.84) compared to higher irradiance (r 2 = 0.62). The maximum quantum yield of PS 2 (Fv/Fm) did not differ much in sun and shade leaves with the exception of midday when excess of light energy absorbed by plants under sun was thermally dissipated. Hence swamp taro plants adopted different strategies to utilize radiation under different irradiances. At higher irradiance, there was faster decline in proportion of open PS 2 centers (PQ) and excess light energy was dissipated through non-photochemical quenching (NPQ). Under shade, absorbed energy was effectively utilized resulting in higher ΦPS2.  相似文献

Photosynthesis, photorespiration, and chlorophyll (Chl) fluorescence in green and red Berberis thunbergii leaves were studied with two different measuring radiations, red (RR) and “white” (WR). The photosynthetic and photorespiration rates responded differently to the different radiation qualities, which indicate that the carboxylase and oxygenase activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) were affected. Differences in photosynthetic rate between the two color leaves were less under RR than under WR. However, this reduced difference in photosynthetic rate was not correlated with the stomatal response to the measuring radiation qualities. Compared with the WR, the RR reduced the differences in dark-adapted minimum and maximum fluorescence, steady-state fluorescence, light-adapted maximum fluorescence, and actual photochemical efficiency (ΦPS2) of photosystem 2 (PS2), but enlarged the difference in non-photochemical quenching between the two color leaves. Differences in both maximum quantum yield of PS2 and ratio of ΦPS2 to quantum yield of CO2 fixation between the two color leaves were similar under the two measuring radiations. To exclude disturbance of radiation attenuation caused by anthocyanins, it is better to use RR to compare the photosynthesis and Chl fluorescence in green versus red leaves.  相似文献

When dark-acclimated cotton (Gossypium hirsutum L. cv. Coker 312) leaves, pre-treated with lincomycin to inhibit chloroplast protein repair processes, were exposed to 10 °C and a PPFD of 500 mol m-2 s-1, the proportion of excitation energy entering photochemistry (P) increased, but only to 5 % of the total energy absorbed at steady state levels of P, which were reached at 40 min of irradiation. Thermal dissipation (D) of absorbed energy increased throughout the 360 min irradiation period and accounted for the greatest portion of absorbed energy at 10 °C. When D was partitioned into constitutive (DCON), regulated (DREG), and photoinhibitory (DPI) components, it was primarily composed of DREG, the readily reversible portion of D. However, the induction of D was slow at 10 °C. Sixty minutes were required for D to reach 70 % of the energy absorbed. Considerable absorption of energy in excess of that utilized in photochemistry or dissipated thermally (designated as E) occurred, especially during induction of P and D. Over the irradiation period, the time-dependent averaged E exhibited an inverse, linear relationship with the ratio of variable (Fv) to maximum (Fm) fluorescence (PS2 efficiency) and a linear relationship with DPI. We propose that time-dependent averaged E may be useful for estimating the potential for damage to PS2 under stressful environmental conditions.  相似文献

Chlorophyll (Chl) a fluorescence transient and 820-nm transmission kinetic were investigated to explore the development of photosynthetic apparatus in grapevine leaves from emergence to full expansion. In this study, all leaves at various developing stages exhibited typical Chl a fluorescence transient. In newly initiating leaves, the maximum quantum yield of primary photochemistry (ϕP0) was slightly lower (<10 %) than that in fully expanded leaves. Nevertheless, the fluorescence rise from O to J step was clearly speeded up in young leaves compared with that in fully expanded leaves. Additionally, a distinct K step appeared in young leaves at high irradiances. With leaf development, the efficiency that a trapped exciton can move an electron into the electron transport chain further than QA 0), the quantum yield of electron transport beyond QAE0), electron transport flux per excited cross section (ET0/CS0), the amount of active photosystem (PS) 2 reaction centres per excited cross section (RC/CS0), and the performance index on cross section basis (PICS) increased gradually and rapidly. Young leaves had strikingly lower amplitude of transmission at 820 nm. A linear relationship between Ψ0 and the transmission at 820 nm (I30/I0) was evident. Based on these data, we suggest that (1) the primary photochemistry of PS2 may be not the limiting step of the photosynthetic capacity during leaf growth under natural irradiance; (2) oxygen evolving complex (OEC) might be not fully connected to PS2 at the beginning of leaf growth; (3) though there are a few functional PS1 and PS2 at the early stages of leaf development, they match perfectly.  相似文献

The light-harvesting chlorophyll a/b proteins associated with PS II (LHC II) are often considered to have a regulatory role in photosynthesis. The photosynthetic responses of four chlorina mutants of barley, which are deficient in LHC II to varying degrees, are examined to evaluate whether LHC II plays a regulatory role in photosynthesis. The efficiencies of light use for PS I and PS II photochemistry and for CO2 assimilation in leaves of the mutants were monitored simultaneously over a wide range of photon flux densities of white light in the presence and absence of supplementary red light. It is demonstrated that the depletions of LHC II in these mutants results in a severe imbalance in the relative rates of excitation of PS I and PS II in favour of PS I, which cannot be alleviated by preferential excitation of PS II. Analyses of xanthophyll cycle pigments and fluorescence quenching in leaves of the mutants indicated that the major LHC II components are not required to facilitate the light-induced quenching associated with zeaxanthin formation. It is concluded that LHC II is important to balance the distribution of excitation energy between PS I and PS II populations over a wide range of photon flux densities. It appears that LHC II may also be important in determining the quantum efficiency of PS II photochemistry by reducing the rate of quenching of excitation energy in the PS II primary antennae.Abbreviations Fm, Fv maximal and variable fluorescence yields in a light adapted state - LHC II light harvesting chlorophyll a/b protein complex associated with PS II - qp photochemical quenching - A820 light-induced absorbance change at 820 nm - øPSI, øPSII relative quantum efficiencies of PS I and PS II photochemistry - øCO2 quantum yield of CO2 assimilation  相似文献

Photosynthesis in iron-deficient soybean and maize leaves decreased drastically. The quantum yield of photosystem 2 (PS2) electron transport (ΦPS2), the efficiency of excitation energy capture by open PS2 reaction centres (Fv′/Fm′), and photochemical quenching coefficient (qP) under high irradiance were lowered significantly by iron deficiency, but non-photochemical quenching (NPQ) increased markedly. The analysis of the polyphasic rise of fluorescence transient showed that iron depletion induced a pronounced K step both in soybean and maize leaves. The maximal quantum yield of PS2 photochemistry (Φpo) decreased only slightly, however, the efficiency with which a trapped exciton can move an electron into the electron transport chain further than QA0) and the quantum yield of electron transport beyond QAEo) in iron deficient leaves decreased more significantly compared with that in control. Thus not only the donor side but also the acceptor of PS2 was probably damaged in iron deficient soybean and maize leaves. Spraying low concentrated (0.5–5.0 mM) solutions of NaHSO3 on Satsuma mandarin (Citrus unshiu Marc.) leaves resulted in enhancement (maximal about 15 % at 1 mM NaHSO3) of net photosynthetic rate (P N) for 6 d. The potential photochemical efficiency of photosystem 2 (PS2, Fv/Fm) and the quantum yield of PS2 electron transport (ΦPS2) were increased under strong photon flux density (PFD). The slow phase of millisecond delayed light emission (ms-DLE) was increased, showing that the transmembrane proton motive force related to photophosphorylation was enhanced. We also observed that low concentrations of NaHSO3 promoted the production of ATP in irradiated leaves. We suggest that the increase in P N in Satsuma mandarin leaves caused by low concentrations of NaHSO3 solution may have been due to the stimulation of photophosphorylation and, hence, the increase in photochemical efficiency through speeding-up of PS2 electron transport. Photoinhibition of photosynthesis in leaves was modified by NaHSO3 treatment under high PFD. Hence the increase in leaf dry mass seems to be associated with the mitigation of photoinhibition caused by strong PFD.  相似文献

The light harvesting and photosynthetic characteristics of a chlorophyll-deficient mutant of cowpea (Vigna unguilata), resulting from a single nuclear gene mutation, are examined. The 40% reduction in total chlorophyll content per leaf area in the mutant is associated with a 55% reduction in pigment-proteins of the light harvesting complex associated with Photosystem II (LHC II), and to a lesser extent (35%) in the light harvesting complex associated with Photosystem I (LHC I). No significant differences were found in the Photosystem I (PS I) and Photosystem II (PS II) contents per leaf area of the mutant compared to the wildtype parent. The decreases in the PS I and PS II antennae sizes in the mutant were not accompanied by any major changes in quantum efficiencies of PS I and PS II in leaves at non-saturating light levels for CO2 assimilation. Although the chlorophyll deficiency resulted in an 11% decrease in light absorption by mutant leaves, their maximum quantum yield and light saturated rate of CO2 assimilation were similar to those of wildtype leaves. Consequently, the large and different decreases in the antennae of PS II and PS I in the mutant are not associated with any loss of light use efficiency in photosynthesis.Abbreviations LHC I, LHC II light harvesting chlorophyll a/b protein complexes associated with PS I and PS II - A820 light-induced absorbance change at 820 nm - øPS I, øPS II relative quantum efficiencies of PS I and PS II photochemistry  相似文献

The effect of four different NaCl concentrations (from 0 to 102 mM NaCl) on seedlings leaves of two corn (Zea mays L.) varieties (Aristo and Arper) was investigated through chlorophyll (Chl) a fluorescence parameters, photosynthesis, stomatal conductance, photosynthetic pigments concentration, tissue hydration and ionic accumulation. Salinity treatments showed a decrease in maximal efficiency of PSII photochemistry (Fv/Fm) in dark-adapted leaves. Moreover, the actual PSII efficiency (ϕPSII), photochemical quenching coefficient (qp), proportion of PSII centers effectively reoxidized, and the fraction of light used in PSII photochemistry (%P) were also dropped with increasing salinity in light-adapted leaves. Reductions in these parameters were greater in Aristo than in Arper. The tissue hydration decreased in salt-treated leaves as did the photosynthesis, stomatal conductance (g s) and photosynthetic pigments concentration essentially at 68 and 102 mM NaCl. In both varieties the reduction of photosynthesis was mainly due to stomatal closure and partially to PSII photoinhibition. The differences between the two varieties indicate that Aristo was more susceptible to salt-stress damage than Arper which revealed a moderate regulation of the leaf ionic accumulation.  相似文献

A yellow leaf colouration mutant (named ycm) generated from rice T-DNA insertion lines was identified with less grana lamellae and low thylakoid membrane protein contents. At weak irradiance [50 μmol(photon) m−2 s−1], chlorophyll (Chl) contents of ycm were ≈20 % of those of WT and Chl a/b ratios were 3-fold that of wild type (WT). The leaf of ycm showed lower values in the actual photosystem 2 (PS2) efficiency (ΦPS2), photochemical quenching (qP), and the efficiency of excitation capture by open PS2 centres 1 (Fv′/Fm′) than those of WT, except no difference in the maximal efficiency of PS2 photochemistry (Fv/Fm). With progress in irradiance [100 and 200 μmol(photon) m−2 s−1], there was a change in the photosynthetic pigment stoichiometry. In ycm, the increase of total Chl contents and the decrease in Chl a/b ratio were observed. ΦPS2, qP, and Fv′/Fm′ of ycm increased gradually along with the increase of irradiance but still much less than in WT. The increase of xanthophyll ratio [(Z+A)/(V+A+Z)] associated with non-photochemical quenching (qN) was found in ycm which suggested that ycm dissipated excess energy through the turnover of xanthophylls. No significant differences in pigment composition were observed in WT under various irradiances, except Chl a/b ratio that gradually decreased. Hence the ycm mutant developed much more tardily than WT, which was caused by low photon energy utilization independent of irradiance.  相似文献

The effects of Mn-deficiency on CO2 assimilation and excitation energy distribution were studied using Mn-starved maize leaves. Mn-deficiency caused about 70 % loss in the photon-saturated net photosynthetic rate (P N) compared to control leaves. The loss of P N was associated with a strong decrease in the activity of oxygen evolution complex (OEC) and the linear electron transport driven by photosystem 2 (PS2) in Mn-deficienct leaves. The photochemical quenching of PS2 (qP) and the maximum efficiency of PS2 photochemistry (Fv/Fm) decreased significantly in Mn-starved leaves under high irradiance, implicating that serious photoinhibition took place. However, the high-energy fluorescence quenching (qE) decreased, which was associated with xanthophyll cycle. The results showed that the pool of de-epoxidation components of the xanthophyll cycle was lowered markedly owing to Mn deficiency. Linear electron transport driven by PS2 de-creased significantly and was approximately 70 % lower in Mn-deficient leaves than that in control, indicating less trans-thylakoid pH gradient was built in Mn deficient leaves. We suggest that the decrease of non-radiative dissipation depending on xanthophyll cycle in Mn-starved leaves is a result of the deficiency of trans-thylakoid pH gradient.  相似文献

Photosynthetic rate (PN) and chlorophyll (Chl) fluorescence induction of source leaves in response to a low sink demand created by girdling the branch (GB) between the root-tuber-system and the leaves were studied in Dahlia pinnata L. cv. Rigolet during the stage of rapid tuber growth in the greenhouse. GB resulted in significantly lower values of PN, stomatal conductance (gs), and transpiration rate (E), but in higher leaf temperature (Tl) compared with those of controls. With exception of maximum quantum yield of photosystem 2 (PS 2) photochemistry (Fv/Fm) and maximum ratio of quantum yields of photochemical and concurrent non-photochemical processes in PS 2 (Fv/F0), no significant differences were observed in Chl fluorescence parameters between girdled and control leaves on days 1 and 2 after GB, indicating no apparent damage in the photosynthetic apparatus. However, longer girdling duration resulted in higher non-photochemical Chl fluorescence quenching (NPQ), but lower Fv/F0, actual efficiency of energy conversion in PS 2 under steady-state conditions (ΦPS2), and photochemical quenching coefficient (qP) in comparison with controls from 10:00 to 16:00 or 15:00 on days 4 and 5, respectively, indicating reversible injury in the photosynthetic apparatus.  相似文献

To investigate how excess excitation energy is dissipated in a ribulose-1,5-bisphospate carboxylase/oxygenase activase antisense transgenic rice with net photosynthetic rate (P N) half of that of wild type parent, we measured the response curve of P N to intercellular CO2 concentration (C i), electron transport rate (ETR), quantum yield of open photosystem 2 (PS2) reaction centres under irradiation (Fv′/Fm′), efficiency of total PS2 centres (ΦPS2), photochemical (qP) and non-photochemical quenching (NPQ), post-irradiation transient increase in chlorophyll (Chl) fluorescence (PITICF), and P700+ re-reduction. Carboxylation efficiency dependence on C i, ETR at saturation irradiance, and Fv′/Fm′, ΦPS2, and qP under the irradiation were significantly lower in the mutant. However, NPQ, energy-dependent quenching (qE), PITICF, and P700+ re-reduction were significantly higher in the mutant. Hence the mutant down-regulates linear ETR and stimulates cyclic electron flow around PS1, which may generate the ΔpH to support NPQ and qE for dissipation of excess excitation energy.  相似文献

This study investigated the effects of cinnamic acid (CA) on growth, biochemical and physiological responses of Lactuca sativa L. CA (0.1, 0.5, 1.0 and 1.5 mM) treatments decreased plant height, root length, leaf and root fresh weight, but it did not affect the leaf water status. CA treatment (1.5 mM) significantly reduced Fv, Fm, photochemical efficiency of PSII (Fv/Fm) and quantum yield of PSII (ΦPSII) photochemistry in L. sativa. The photochemical fluorescence quenching (qP) and non-photochemical quenching (NPQ) were reduced after treatment with 1.5 mM CA. Fraction of photon energy absorbed by PS II antennae trapped by “open” PS II reaction centers (P) was reduced by CA (1.5 mM) while, portion of absorbed photon energy thermally dissipated (D) and photon energy absorbed by PSII antennae and trapped by “closed” PSII reaction centers (E) was increased. Carbon isotope composition ratios (δ13C) was less negative (−27.10) in CA (1.5 mM) treated plants as compared to control (−27.61). Carbon isotope discrimination (Δ13C) and ratio of intercellular CO2 concentration (ci/ca) from leaf to air were also less in CA treated plants. CA (1.5 mM) also decreased the leaf protein contents of L. sativa as compared to control.  相似文献

The effect of high irradiance (HI, photosynthetically active photon flux density of 1 300 μmol m−2 s−1) on net photosynthetic rate (P N), chlorophyll fluorescence parameters, and xanthophyll cycle components were studied in fruit tree bayberry leaves. HI induced the photoinhibition and inactivation of photosystem 2 (PS2) reaction centres (RCs), which was characterized by decreased P N, maximum yield of fluorescence after dark adaptation (Fm), photochemical efficiency of PS2 (Fv/Fm) and quantum yield of PS2 (ΦPS2), and increased reduction state of QA (1-qP) and non-photochemical quenching (NPQ). Initial fluorescence (F0) showed a decrease after the first 2 h, and subsequently increased from the third hour exposure to HI. Furthermore, a greater increase in the ratio (Fi-F0)/(Fp-F0) which is an expression of the proportion of the QB non-reducing PS2 centres, whereas a remarked decrease in the slope of Fi to Fp which represents the rate of QA reduction was observed in leaves after HI exposure. Additionally, HI caused an increase in the pool size of the xanthophyll cycle pigments and sustained elevated contents of zeaxanthin (Z), antheraxanthin (A), and de-epoxidation state (DES) at the end of the irradiation period. During HI, decreased Fm, Fv/Fm, ΦPS2, NPQ, slope of Fi to Fp, V+A+Z, and DES, and increased F0, 1-qP, ratio (Fi-F0)/(Fp-F0), and V were observed in dithiothreitol (DTT)-fed leaves compared to control ones under the same conditions. Hence photoinhibition caused by HI in bayberry was probably attributed to inactivation of PS2 RCs, and photoprotection from photodamage were mainly related to the xanthophyll cycle-dependent heat dissipation in excess photons.  相似文献

Citrus volkameriana (L.) plants were grown for 43 d in nutrient solutions containing 0, 2, 14, 98, or 686 μM Mn (Mn0, Mn2, Mn14, Mn98, and Mn686, respectively). To adequately investigate the combined effects of Mn nutrition and irradiance on photosystem 2 (PS2) activity, irradiance response curves for electron transport rate (ETR), nonphotochemical quenching (qN), photochemical quenching (qP), and real photochemical efficiency of PS2 (ΦPS2) were recorded under 10 different irradiances (66, 96, 136, 226, 336, 536, 811, 1 211, 1 911, and 3 111 μmol m−2 s−1, I66 to I3111, respectively) generated with the PAM-2000 fluorometer. Leaf chlorophyll content was significantly lower under Mn excess (Mn686) compared to Mn0; its highest values were recorded in the treatments Mn2-Mn98. However, ETR and ΦPS2 values were significantly lower under Mn0 compared to the other Mn treatments, when plants were exposed to irradiances ≥96 μmol m−2 s−1. Furthermore, Mn0 plants had significantly higher values of qN and lower values of qP at irradiances ≤226 and ≥336 μmol m−2 s−1, respectively, than those grown under Mn2-Mn686. Irrespective of Mn treatment, the values of ΦPS2 and qN decreased, while those of qP increased progressively by increasing irradiance from I136 to I3111. Finally, Mn2-Mn98 plants were less sensitive to photoinhibition of photosynthesis (≥811 μmol m−2 s−1) than the Mn686 (≥536 μmol m−2 s−1) and Mn0 (≥336 μmol m−2 s−1) ones.  相似文献

The photoprotection of energy dissipation and water-water cycle were investigated by comparing chilling sensitivity of photosystems 2 (PS2) and 1 (PS1) in two chilling-sensitive plants, cucumber and sweet pepper, upon exposure to 4 °C under low irradiance (100 μmol m−2 s−1) for 6 h. During chilling stress, the maximum photochemical efficiency of PS2 (Fv/Fm) decreased only slightly in both plants, but the oxidisable P700 decreased markedly, which indicated that PS1 was more sensitive to chilling treatment under low irradiance than PS2. Sweet pepper leaves had lower Fv/Fm, higher non-photochemical quenching (NPQ), and higher oxidisable P700 during chilling stress. Activity of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in cucumber leaves was higher, but APX activity decreased apparently compared to that at room temperature. The productions of active oxygen species (H2O2, O2 ) increased in both plants, faster in cucumber leaves than in sweet pepper leaves. In sweet pepper leaves, a stronger de-epoxidation of the xanthophyll cycle pigments, a higher NPQ could act as a major protective mechanism to reduce the formation of active oxygen species during stress. Thus sensitivity of both plants to chilling under low irradiance was dominated by the protective mechanisms between PS1 and PS2, especially the energy dissipation and the water-water cycle. Photoinhibition of photosynthesis was investigated in control (C) and chilling night (CN) leaves of grapevine under natural photoperiod at different sampling time in a day. The degree of photoinhibition was determined by means of the ratio of variable to maximum chlorophyll fluorescence (Fv/Fm) and photosynthetic electron transport measurements. When the potential efficiency of photosystem (PS) 2, Fv/Fm was measured at midday, it markedly declined with significant increase of F0 in CN leaves. In isolated thylakoids, the rate of whole chain and PS2 activity were markedly decreased in CN leaves than control leaves at midday. A smaller inhibition of PS1 activity was also observed in both leaf types. Later, the leaves reached maximum PS2 efficiencies similar to those observed in the morning during sampling at evening. The artificial exogenous electron donors diphenyl carbazide, NH2OH, and Mn2+ failed to restore the PS2 activity in both leaf types at midday. Thus CN enhanced inactivation on the acceptor side of PS2 in grapevine leaves. Quantification of the PS2 reaction centre protein D1 following midday exposure of leaves showed pronounced differences between C and CN leaves. The marked loss of PS2 activity in CN leaves noticed in midday samples was mainly due to the marked loss of D1 protein of the PS2 reaction centre.  相似文献
