The differentiation of sieve and tracheary elements was studied in callus culture of Daucus carota L., Syringa vulgaris L., Glycine max (L.) Merr., Helianthus annuus L., Hibiscus cannabinus L. and Pisum sativum L. By the lacmoid clearing technique it was found that development of the phloem commenced before that of the xylem. In not one of the calluses was differentiation of tracheary elements observed in the absence of sieve elements. The influence of indole-3-acetic acid (IAA) and sucrose was evaluated quantitatively in callus of Syringa, Daucus and Glycine. Low IAA levels resulted in the differentiation of sieve elements with no tracheary cells. High levels resulted in that of both phloem and xylem. IAA thus controlled the number of sieve and tracheary elements, increase in auxin concentration boosting the number of both cell types. Changes in sucrose concentration, while the IAA concentration was kept constant, did not have a specific effect on either sieve element differentiation, or on the ratio between phloem and xylem. Sucrose did, however, affect the quantity of callose deposited on the sieve plates, because increase in the sucrose concentration resulted in an increase in the amount of callose. It is proposed that phloem is formed in response to auxin, while xylem is formed in response to auxin together with some added factor which reaches it from the phloem.  相似文献

Summary A study of the ultrastructure of cultured tobacco callus phloem shows that it is very similar to that found in the intact stem.Fibrils similar to the P protein, characteristic of angiosperm phloem, are found initially within the rough endoplasmic reticulum. This has not been shown previously.Preincubation of the callus at 0 or 50° C or in solutions of cholchicine prior to fixation have no effect on the relative proportions or the individual structures of the two forms of P protein found in tobacco callus phloem sieve elements. However 1% colchicine does reduce the number of microtubules found in callus cells by up to 80%.The morphological similarity between P 1 protein tubules and microtubules therefore appears coincidental and not due to a different arrangement of a common basic unit.The function of the P protein is considered and reasons for the comparative stability of plant microtubules compared with their animal equivalents are discussed.  相似文献

Illus. 1963.—Callus tissues in established maintenance culture lack morphological and physiological organization. Such callus consists of homogeneous parenchyma. Movement of auxin and sugar, therefore, must be along diffusion gradients. The only vascular tissues occurring in callus are induced. Experimental induction of vascular tissues has been successful in callus of 3 sp. of the Oleaceae: a tree, Fraxinus americana, and 2 shrubs, Syringa vulgaris and Ligustrum vulgare; another tree, Salix purpurea, var. lambertiana; a vine, Parthenocissus tricuspidata; and an herb, Helianthus tuberosus. In each of these species, an auxin (IAA or NAA in these studies) and a sugar (sucrose or glucose in these studies) prove necessary for the induction and complete differentiation of xylem and phloem in callus tissues. Varying concentrations of sugar alter the proportions of xylem to phloem: low concentrations, 1.5%–2.5%, favor xylem formation; high, 3%–4%, favor phloem. Middle concentrations, 2.5%–3.5%, favor the presence of xylem and phloem, usually with a cambium between. The almost universal association of xylem and phloem may have its explanation in this middle concentration of sugar. Grafting of apices into callus or direct application of appropriate concentrations of an auxin and a sugar in agar to the surface of callus causes nodules of vascular tissue to be formed, mostly in a circular pattern when seen in section transverse to the axis of orientation of the callus in the medium. The diameter of this circle varies directly with the auxin concentration at the place of application, 0.05 mg/liter giving a narrow, and 1 mg/liter, a wide pith. In individual nodules, xylem is characteristically oriented towards the center of the callus and the phloem towards the outside. Variable cross-sectional views of nodule distribution in calli under different treatments suggest experimental approaches to understanding stelar patterns. The induction and differentiation in callus of xylem and phloem tissues has no relation to conduction. Any use of vascular tissues can occur only after their induction.  相似文献

该研究以侧柏一年生硬枝插穗为实验材料,利用连续组织切片技术观察插穗不定根发生发育过程中的组织结构变化,分析插穗外部形态变化、不定根原基起源和不定根的形成过程,探讨侧柏插穗不定根发生模式和不定根的组织学起源。结果显示:侧柏扦插后可由愈伤组织、皮部诱导产生不定根,出现皮部生根、愈伤组织生根、愈伤组织兼具皮部生根3种类型;侧柏插穗中存在少量潜伏根原基,但插穗生根类型以诱导生根为主;不定根原基诱导产生于愈伤组织、木质部、形成层及次生韧皮部等部位。研究认为侧柏扦插生根属于多位点发生模式,不定根原基的组织学起源是愈伤组织、髓射线、射线原始细胞、尚未分化成熟的木质部细胞,通过人工诱导同时激活这些不定根起源位点能够显著提高生根率和生根质量。  相似文献

The development of adventitious shoot formation from cultured somatic tissues of Solanum carolinense was studied using light microscopy. For purposes of comparison, callus initiation and proliferation were also followed. When stem segments of this plant were cultured on medium supplemented with 6 mg/l benzyladenine (BA), cell division was first observed after 48 hr in the external phloem and inner cortex of the segments. This division activity gave rise to meristematic zones which subsequently formed shoot primordia within 6 days of culture. While organogenic potential appears to be limited to specific regions, all tissues of the explant were capable of callus formation when cultured on medium containing 3 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D). In conjunction with a previous study, it appears that unlike somatic embryogenesis in this species, organogenesis does not require an intermediate callus phase for differentiation to occur.  相似文献

为探究小檗科植物八角莲组织培养的器官发生方式,该研究以八角莲离体叶片、叶柄在MS培养基上诱导产生的愈伤组织、不定芽、不定根为对象,用连续石蜡切片技术分析八角莲组织培养的器官发生途径。结果表明:八角莲愈伤组织形成的解剖学特征是靠近表皮的薄壁细胞经激素刺激恢复分裂能力,继续培养形成拟分生组织。拟分生组织可形成许多分化中心。通过对八角莲组织培养产生的不定芽细胞组织学观察发现芽原基起源于愈伤组织外侧的几层薄壁细胞,芽原基背离愈伤组织中央生长形成不定芽,故八角莲脱分化形成的芽起源方式为外起源。而八角莲的根原基起源于组织深处髓部薄壁细胞和部分维管形成层细胞,进而形成类似球形或楔形并朝韧皮部突起的根原基轮廓,根原基继续发育会突破表皮生成不定根,起源方式为内起源。八角莲离体再生途径为器官发生型,在组培苗生长过程中先诱导形成不定芽,再诱导形成不定根,在愈伤组织上形成维管组织将不定芽和不定根连接成完整植株。  相似文献

利用徒手切片、石蜡切片和超薄切片及显微摄像的方法,对当归根的显微结构及根腐病致病真菌的分布进行了研究。结果表明:当归的根由周皮和次生维管组织两部分组成,周皮由外向内依次分为木栓层、木栓形成层、栓内层;次生韧皮部占根径的比例在60%以上,主要成分包括筛胞、韧皮薄壁细胞、韧皮纤维和分泌道,薄壁细胞富含淀粉粒等营养物质;次生木质部由导管、木薄壁细胞和木射线组成,木质部呈多元形,木射线和韧皮射线明显。在根的周皮细胞和中柱中均有真菌分布,说明真菌由木栓层、木栓形成层、栓内层依次向里侵入到韧皮薄壁细胞,在薄壁细胞内定殖并形成菌丝结或团块状结构,进而扩展成一定的侵染区域;真菌不仅侵染周皮和韧皮部,而且还进一步侵染木质部并破坏导管。此外,研究还发现,淀粉粒是真菌定殖的主要场所,真菌穿透或缠绕在淀粉粒上,并利用其营养不断地生长与繁殖。  相似文献

The fine structure of Pinus radiata D. Don callus before andafter differentiation into stem-like tissues has been examinedwith the electron microscope. In callus prior to differentiation(here called parenchymatous callus) the cells accumulate tanninsas they age and are quite distinct from the cells of differentiatedcallus. In the latter, cambium, phloem and xylem cells may beidentified by their general morphology and by their ultrastructuralfeatures. Differentiation into a true stem-like structure is,however, incomplete in that the tissues are not uniformly oriented,and parenchyma cells of the rays and phloem contain chloroplasts.The tracheids also show unusual differentiation in that borderedpits form over their entire surface and may be of two types.The reasons for these variations are discussed.  相似文献

The origin and development of zygotic and somatic embryos of Trifolium rubens L. was studied with the aid of paraffin sections and light microscopy. Zygotic embryos were collected, fixed and prepared daily from one to ten days after cross-pollination. Somatic embryos were obtained by plating petiole sections on modified L2 medium with 0.015 mgl-1 picloram and 0.1 mgl-1 6-BAP. Cultured petioles were collected and fixed daily from one to 25 days after plating. Two regions in the vascular bundle sheath of cultured petioles gave rise to callus. The first region was adjacent to the phloem fibers and produced friable callus. The second region gave rise to compact callus that was connected to the fascicular cambium. Somatic embryos originated from single cells in the cortex directly without intervening callus formation and from single cells in the friable callus. In addition, embryos arose from meristematic regions in compact callus. Many early stages of embryogenesis (one, two and four-celled stages) were observed in the cortex and friable callus. Zygotic embryogenesis in Trifolium differs from other legumes in that the suspensor is short and has a broad attachment. This arrangement was observed in zygotic embryos of T. rubens and in many somatic embryos. However, a continuum of somatic embryogenesis was observed where some young embryos had a Trifolium suspensor-like arrangement while others were attached to a long narrow suspensor-like structure more characteristic of Medicago.  相似文献

Summary Only one or two layers of sieve cells of the previous year's phloem in lateral branches of Larix decidua persist as fully mature cells. Immature sieve cells or cambial derivatives that have not completed differentiation may also over-winter. Periclinal cell divisions of the vascular cambium were first observed by mid-April. During the short period of greatest cambium activity (mid-April to mid-May), the early phloem is laid down. Late phloem is formed over a much longer period, from mid-May to late September. Microautoradiography revealed that only mature sieve cells of the early phloem are involved in translocation of 14C assimilates in June. The fine structure of actively translocating sieve cells is described. The impact of structure on long-distance transport of assimilates is discussed.  相似文献

Summary Seeds of theArabidopsis thaliana mutant primordia timing (pt) were germinated in 2,4-dichlorophenoxyacetic acidcontaining liquid medium. The seedlings formed somatic embryos and nonembryogenic and embryogenic callus in vitro in a time period of approximately two to three weeks. Embryogenesis and callus formation were monitored with respect to origin, structure, and development. Ten days after germination globular structures appeared in close vicinity of and on the shoot apical meristem (SAM). Somatic embryos formed either directly on the SAM region of the seedling or indirectly on embryogenic callus that developed at the SAM zone. Globular structures developed along the vascular tissue of the cotyledons as well, but only incidentally they formed embryos. Upon deterioration, the cotyledons formed callus. Regular subculture of the embryogenic callus gave rise to high numbers of somatic embryos. Such primary somatic embryos, grown on callus, originated from meristematic cell clusters located under the surface of the callus. Embryos at the globular and heart-shape stage were mostly hidden within the callus. Embryos at torpedo stage appeared at the surface of the callus because their axis elongated. Secondary somatic embryos frequently formed directly on primary ones. They preferentially emerged from the SAM region of the primary somatic embryos, from the edge of the cotyledons, and from the hypocotyl. A close sheathing of the plastids by endoplasmic reticulum has been observed. This is restricted to the companion cells and developing sieve tubes of the phloem of Acer pseudoplatanus and the resin canal cells and leaf callus cells from Pinus pinea. The sheathing is transitory in callus and sieve tubes but is a permanent feature of the companion and the resin canal cells. Most research involving phloem proteins is done with phloem exudates, which are not easily obtained from many plants. We report here on the use of tissue cultures to study phloem proteins. Monoclonal antibodies against the filamentous phloem protein, P-protein, were made by injecting mice with a phloem-enriched fraction isolated from Streptanthus tortuosus callus grown on a medium that stimulates the differentiation of xylem and phloem (phloem[+] cultures). Monoclonal antibodies specific for P-protein were identified by incubating free-hand stem sections of S. tortuosus in hybridoma supernatants, then in a goat anti-mouse antibody conjugated to fluorescein isothiocyanate (FITC), and observing the FITC under an epifluorescence microscope. Antibodies specific for P-protein in stem sections were used to probe nitrocellulose blots of polyacrylamide gels separating proteins isolated from both phloem(+) and phloem(-) tissue cultures. Immunoblots were incubated overnight in hybridoma supernatants followed by a secondary antibody conjugated to alkaline phosphatase. Three monoclonal antibodies—RS21, RS22, and RS23—bound to an 89-kD band in the phloem(+) lanes but failed to bind to any proteins in the phloem(—) lanes. In leaf sections of Arabidopsis thaliana processed by freeze-substitution, a mixture of RS21 and RS22 bound to the P-protein filaments in sieve elements, but not to any proteins in adjacent cells. A control antibody specific for tubulin did not bind to the P-protein filaments.  相似文献

Phytolacca dioica L., an evergreen tree of the Phytolaccaceae, is one of the species of Phytolacca which shows anomalous secondary thickening in its stem. This mode of thickening has been regarded as successive cambial activity or alternatively, in some more recent interpretations, as thickening by unidirectional activity of a cambial zone. The stem thickening of P. dioica is of the former type. The cambium produces fascicular strands, showing centrifugal differentiation of xylem and centripetal differentiation of phloem on opposite sides of the cambial layer, and rays are produced between the fascicular areas. In both xylem and phloem the younger elements are closer to the cambium than the older elements. Succeeding cambia arise periodically by periclinal divisions in a layer of parenchyma cells two or three cells beyond the outermost intact phloem derived from the current cambium. Each cambium forms a few parenchyma cells on both sides before it forms derivatives which mature into lignified xylem elements or conductive elements of the phloem. The parenchyma thus formed toward the outside later becomes the site of the origin of the succeeding cambium. Only one or two layers of this phloem parenchyma go on to form the new cambium; the remaining cells accumulate between the outermost phloem and the cortex. P. weberbaueri shows stem structure similar to P. dioica. P. meziana, a shrub, shows normal stem structure.  相似文献

Summary The twolined chestnut borer, Agrilus bilineatus (Weber) (Coleoptera: Buprestidae), attacks oaks (Quercus spp.) that have been weakened by prior environmental or biotic stress. Our earlier work showed that trees with relatively low winter starch reserves are more likely to be attacked by A. bilineatus the following summer. We hypothesized that such trees may have less energy available for defense (Callus formation and allelo-chemical synthesis) in tissues wounded by borer larvae. However, wounding experiments showed little or no relationship between winter or summer carbohydrate reserves, callus formation, radial growth, or concentrations of tannins and phenolics in wounded or nonwounded phloem tissues. Trees with relatively low winter carbohydrate reserves were again found to be attractive to adult A. bilineatus, although not all low starch trees were attacked or successfully colonized by borers. There was a trend for carpenterworm larvae, Prinoxystus robiniae (Lepidoptera: Cossidae), a generalist bark and wood borer, to be more successful in establishing galleries on low starch trees. Carpenterworms gained significantly more weight when fed phloem from trees attractive to A. bilineatus. Oaks that attracted large numbers of A. bilineatus or that were successfully colonized by the borer produced significantly less callus than did non-attacked trees when experimentally wounded at about the time of Agrilus egg hatch. Callus formation may limit the establishment of small larvae that feed slowly in the cambial region. These results indicate that current theory regarding relationships between increased tree stress and decreased allocation of energy reserves to radial growth and defense against phloem borers may be an oversimplification. We suggest that tree growth and the defensive response of phloem tissues may be limited more by the rate of carbohydrate utilization or by changes in source-sink relationships than by storage levels. Callus formation and synthesis of allelochemicals in wounded phloem may be under the same control as cambial activation, which is mediated by plant growth regulators and can be influenced by environmental conditions.  相似文献

Modification of external morphology and internal structure of plants is a key feature of their successful survival in extreme habitats. They adapt to arid habitats not only by modifying their leaves, but also show several modifications in their conducting system. Therefore, the present study is aimed to investigate the pattern of secondary growth in Leptadenia pyrotechnica (Forssk.) Decne., (Asclepiadaceae), one such species growing in Kachchh district, an arid region of Gujarat State. A single ring of vascular cambium, responsible for radial growth, divided bidirectionally and formed the secondary xylem centripetally and the phloem centrifugally. After a short period of secondary xylem differentiation, small arcs of cambium began to form secondary phloem centripetally instead of secondary xylem. After a short duration of such secondary phloem formation, these segments of cambium resumed their normal function to produce secondary xylem internally. Thus, the phloem strands became embedded within the secondary xylem and formed interxylary phloem islands. Such a recurrent behavior of the vascular cambium resulted in the formation of several patches of interxylary phloem islands. In thick stems the earlier formed non-conducting interxylary phloem showed heavy accumulation of callose on the sieve plates followed by their crushing in response to the addition of new sieve elements. Development of intraxylary phloem is also observed from the cells situated on the pith margin. As secondary growth progresses further, small arcs of internal cambium get initiated between the protoxylem and intraxylary phloem. In the secondary xylem, some of the vessels are exceptionally thick-walled, which may be associated with dry habitats in order to protect the vessel from collapsing during the dryer part of the year. The inter- and intraxylary phloem may also be an adaptive feature to prevent the sieve elements to become non-conducting during summer when the temperature is much higher.  相似文献

Water sorption by powdered human callus was studied using a vacuum microbalance, X-ray powder diffraction and NMR relaxation. The sorption data were fitted to theoretical isotherms. At high relative vapour pressures an increase in the monolayer value was found which is probably related to the swelling of the material. Adsorption/desorption hysteresis is present below 0.75' relative vapour pressure (r.v.p.). Only a small increase in protein chain separation was observed on water uptake, indicating that the primary level of organization of the keratin is hardly affected. The effect of solvent extraction is to reduce the strength of water binding but to leave the monolayer values constant. Isosteric heats confirm the reduction in affinity for water on solvent extraction and show that totally extracted callus behaves as a mildly hydrophobic material. The results are consistent with a model in which the water binding properties of callus are determined by the presence of water soluble components which allow a monomolecular layer of water to be formed at low r.v.p. followed by physical multilayer formation.Adsorption of sodium dodecyl sulphate was found to increase the monolayer value while dodecyl trimethyl ammonium bromide left the monolayer value unchanged.The NMR relaxation behaviour of water in callus is very similar to that of water on human hair and wool fibres. An activation enthalpy of 48 kJ · mol−1 was found for the T2 relaxation and a very broad T1 minimum was observed at about −20°C at an operating frequency of 45 MHz corresponding to a correlation time of 2.2 ns.  相似文献

本文对翅果油树大宫灯型茎段培养在MS附加6-BA较高、NAA较低浓度的培养基上培养0~30d的组织学变化进行了研究。创伤对其愈伤组织的形成有明显的刺激作用,培养3~4d切口处的皮层细胞、形成层细胞、韧皮部薄壁细胞以及髓组织细胞,甚至表皮细胞均脱分化开始分裂;培养8~11d,切口明显膨大,起源于髓及维管组织周围薄壁细胞的愈伤组织突起大;培养12~20d愈伤组织块中出现了分生组织和维管组织结节;培养21~30d,愈伤组织表层和近表层细胞分化出芽原基,但与维管组织结节无直接联系。  相似文献

采用常规石蜡切片法,观察油茶插穗不定根发育的解剖学特征。结果表明:油茶当年生插穗茎内无潜伏根原基, 插条的皮部存在连续排列成环状的厚壁细胞,这些结构特征可能与油茶插条生根时间较长有关。不定根由诱生根原基发育形成, 诱生根源于形成层、韧皮部及愈伤组织等部位,不定根属于诱导根原始体型、混合生根型。只有极少数插条的不定根由愈伤组织长出,不定根的发生与愈伤组织没有直接关系。  相似文献

The anthers of three genotypes ofLycopersicon esculentum, viz. cv. HS-101, cv. HS-102 and an F1 hybrid (Montfavet 63-4xHS-101) in different stages of development were cultured in various defined nutritive media. Only anthers containing microspores in the early uninucleate stage were found to respond with the culture medium in the formation of androgenic callus. The DGII medium with 2 mg l−1 NAA and 1 mg 1−1 kinetin was found to be best for callus induction but MS medium supplemented with 2 mg l−1 2,4-D and 0.1 mg 1−1 BAP favoured proliferation and growth of the callus. The androgenic microspores followed the ‘B’ type pathway of androgenesis in the formation of callus. Induction of tracheids in the callus could be achieved by supplementing the basal medium with NAA and kinetin or 2,4-D and BAP. Initiation of vessel elements and cambium were favoured by addition of NAA and kinetin and that of the phloem in the presence of 2,4-D and BAP in the basal medium, suggesting that the hormonal requirements for production of different elements of the vascular system in androgenic callus are different. Although roots could be induced from the callus, shoot differentiation could not be achieved under cultural conditions.  相似文献
