Quantitative and qualitative variations of free-amino-acids contents (FAA) of ovaries, hepatopancreas and haemolymph have been studied in the course of ovarian maturation in Penaeus schmitti. In the ovary, FAA content, expressed as microM/g ovary fresh weight, decreases from 56.2 to 23.6 microM between gonadosomatic index (GSI) 0.8 and GSI 7.1. There is an increase of hepatopancreas FAA content of 33.4% till GSI 4. Haemolymph FAA content increases from 0.32 to 1.76 microM/ml, with maximal values of 3.74 microM/ml when GSI are between 3 and 4. FAA content of ovaries, expressed as microM/g of animal, is 3.2 fold higher at the end of maturation, with a main increase in the first stages of ovogenesis. Maturation process has a quantitative effect on all FAA, but, on a qualitative point of view, it concerns mainly lysine, arginine and glycine. The main variations of AA composition (expressed as percentage of total FAA) are observed in haemolymph.  相似文献

Gravid females of Penaeus semisulcatus were spawned in the laboratory by natural means. The embryos were documented and the larvae were reared from hatching to postlarval stage at 28.2–30.0 °C and 33.5–34.5 g kg−1 salinity for about 10 days (223 h 55 min). Six naupliar stages, three protozoea stages, three mysis stages and the first postlarval stage were described and illustrated. The larvae were fed only with microalgae Tetraselmis tetrathele and Chaetoceros gracilis from first protozoea until the second mysis, with about 90% survival rate; from the third mysis until the first postlarva they were fed with similar microalgae coupled with rotifer Brachionus plicatilis and Artemia nauplii. The embryonic and larval stages of P. semisulcatus are generally similar to those of other closely related species in the family Penaeidae, such as Melicertus canaliculatus, Fenneropenaeus merguiensis, and Marsupenaeus japonicus, except for the size and structure of diagnostic characters, setation of appendages and duration of metamorphoses. The change in the feeding habit during ontogeny was related to morphological transformation of the feeding apparatus of larvae and postlarvae. This paper is the first comprehensive and complete account of the early developmental stages of P. semisulcatus.  相似文献

P. semisulcatus is a commercially important species of the ArabianSea. In the present studies the mature females were broughtfrom the sea to the laboratory for spawning. The spawned eggswere reared in laboratory conditions up to the post larval stage.Five naupliar, three protozoeal, three mysis and first postlarval stages were described. Ontogeny of the gut in Penaeus setiferus was investigated by reconstruction of serial sections examined by light microscopy. Development of the gut into the adult form is protracted over several weeks beyond metamorphosis in steps that may be directly related to the unique postlarval life history of Penaeus. The gastric mill is lacking in larval stages of P. setiferus. In protozoeal stages Z1-Z3, the pyloric ampullae are blind sacs that do not communicate with the midgut. The gland filter first appears in mysis stage M2. The gastric mill in early postlarval (PL) stages consists of poorly chitinized lobes with flexible setae. By PL21 the ossicles of the gastric mill are rigid and setae are replaced by spine-like denticles, but even by PL35 the gastric mill is neither as massive nor heavily chitinized as in adults. During the mysis stages and early PL stages, the hepatopancreas communicates freely with both the foregut and the midgut trunk. By PL35 the hepatopancreatic ducts are essentially isolated from the remainder of the midgut by foregut ossicles. The midgut in Z1 consists of two pairs of simple caeca and the midgut trunk. During larval growth, each of the lateral midgut caeca develops into a number of lobes. After metamorphosis these lobes begin to ramify into small-diameter tubules, and by PL35 have completely ramified into the hepatopancreas of adults. From M1 to PL4, the anterior midgut caeca decrease in absolute size and become a single anterior diverticulum. The posterior midgut diverticulum first appears in PL21 as a simple sac and thereafter increases in size and complexity.  相似文献

Summary An ultrastructural comparison between the unistellate spermatozoa of the shrimps Penaeus kerathurus and P. japonicus reveals a number of similarities that are common among dendrobranchiates, but also some marked differences which would confirm the validity of a potential use of sperm structure in systematic and phylogenetic studies. Typical morphological features shared by the spermatozoa of P. kerathurus and P. japonicus are: a membrane-bound acrosomal vesicle consisting of a cap and spike; non-membrane-bound filamentous chromatin; a perinuclear cytoplasmic band including degenerative membranous organelles (mostly mitochondria), small vesicles with a dense core and parallel membrane lamellae. Discordant spermatozoal characteristics between both species involve a significantly different size (ca. 5 m in length by ca. 3 m in diameter in P. kerathurus; ca. 8 m in length by ca. 4.7 m in diameter in P. japonicus), the occurrence of intranuclear lipid droplets only in P. kerathurus and the presence of a deeper subacrosomal space in this species as compared to P. japonicus. It is very likely that the most significant difference between both species is, however, the appearance of microtubule bundles in the spermatozoon of P. japonicus. So far, the occurrence of spermatozoal microtubules in decapod crustaceans appears to be restricted to reptantian species, whereby the finding of such elements in sperm of a dendrobranchiate shrimp could be of phylogenetic interest.  相似文献

Two cortical rod proteins having molecular weights of 28.6 kDa and 30.5 kDa were isolated from the mature ovary of Marsupenaeus japonicus using gel filtration and reversed-phase HPLC. Analysis of the N-terminal amino acid sequence of the 28.6 kDa molecule revealed that amino acid residues 1-21 corresponded to residues 9-29 of the 30.5 kDa molecule. Examination of homology using BLAST showed that 21 amino acids out of 29 residues of the 28.6 kDa molecule, and 14 out of 29 residues of the 30.5 kDa molecule were identical to that of the ovarian cortical rod proteins of Penaeus semisulcatus. Positive immunohistochemical reaction to antiserum raised against the 28.6 kDa protein was observed on cortical rods forming around the periphery of oocytes at the maturation stages. Western blotting analysis revealed that both the 28.6 kDa and 30.5 kDa molecules stained with the anti-28.6 kDa antiserum. Furthermore, the 28.6 kDa and 30.5 kDa proteins were both glycosylated, as evidenced by positive carbohydrate staining using Concanavalin A and production of positive PAS reaction. These results indicate that the cortical rods are comprised of the 28.6 kDa and 30.5 kDa molecules. We subsequently cloned two full-length cDNAs based on the N-terminal sequences of the 28.6 kDa and 30.5 kDa molecules. The open reading frame of 28.6 kDa and 30.5 kDa encoded 276 amino acid residues. Comparison analysis of the two cDNAs revealed that the location of the processing site and sequence of signal peptides differed, indicating that the two cDNAs are products of two separate genes and encode the 28.6 kDa molecule and 30.5 kDa molecule, respectively. Both proteins possessed one potential N-linked glycosylation site. It is considered that both molecules are components of the cortical rods, forming a jelly layer after fertilization.  相似文献

Free fatty acids extracted from the abdominal muscle of Palaemon serratus (Crustacea, Decapoda) maintained on a L-D: 19-5 photoperiod show important circadian variations, characterised by two diurnal maximums and one nocturnal by the end of dark period. 1. A heat-stable alkaline phosphatase was purified from Penaeus japonicus, with a final specific activity of 21,280 U/mg of protein. 2. In polyacrylamide-gel electrophoresis under non-denaturing conditions, the purified shrimp alkaline phosphatase was found to have an identical molecular size and surface charge as the human placental enzyme. 3. By using SDS-PAGE, the monomers of shrimp alkaline phosphatase were discovered to have a Mr 55,000 but those of human placental enzyme with a Mr 70,000. Deglycosylation decreases the Mr values of the subunits to 33,000 for shrimp alkaline phosphatase. 4. The purified alkaline phosphatase from shrimp was recovered with both the attachment sites for sialic acids and phosphatidylinositol. 5. The shrimp alkaline phosphatase has an isoelectric point (pI) of 7.6 and the human placental enzyme has a pI of 4.8.  相似文献

The cleavage pattern of the black tiger shrimp Penaeus monodon was analyzed from the first division until gastrulation. Observations were based on microscopy combined with the use of fluorescent dyes, histological techniques, and computer based three-dimensional reconstructions. Early cleavage is holoblastic and follows a stereotypic pattern, which largely corresponds to what is known from other dendrobranchiate decapods. However, for the first time in this group, we report the presence of an intracellular structure throughout early development. This intracellular body (icb) marks the lineage of one of the two enlarged and division-delayed mesendoderm cells that initiate gastrulation. The identity of the icb as a natural marker and putative determinant of the germ line and its implications on the establishment of the body axes are discussed. The icb as a landmark reveals that the same stereotypic cell division pattern can lead to different fates of individual cells. Hence, the results of this study permit an additional approach to study the relation between cell lineage pattern and the identity of cell lineages.  相似文献

Effects of bilateral eyestalks ablation and injections of eyestalks, (Es) brain (Br) and thoracic ganglia (ThG) extract separately on the androgenic gland and testis development of Es-ablated (Experiment 1) and normal (Experiment 2) P. hardwickii, were investigated. The androgenic gland activity increased significantly in prawns having Es-ablated (Group II), and eyestalkless (Groups VI & VII) and normal (Groups D & E) prawns received Br and ThG extracts, separately, as compared to their respective controls (Groups I & A). Injection of unboiled Es extract in eyestalkless (Group V) and normal (Group C) prawns arrested the androgenic gland activity. Simultaneously, there was a significant (p<0.05) increase in the diameter of testicular follicles, testis weight, testis index and number of mature spermatocytes per follicle in the prawns of groups II, VI & VII of experiment 1 and of groups D & E of experiment 2 when compared to their respective controls. Injection of Es extract into the eyestalkless (Group V) and normal (Group C) prawns ceased testicular development. A significant (<0.05) decrease in testicular protein and midgut gland glycogen and lipid was found in groups II, VI & VII of experiment 1 and in groups D & E of experiment 2, but the glycogen and lipid content of the testis increased. These results indicate that the hormones released from the neuroendocrine centres regulate androgenic gland activity and spermatogenesis.  相似文献

Energy metabolism in early life stages of the shrimp Farfantepenaeus paulensis subjected to temperature reduction (26 and 20 °C) was determined using the activities of citrate synthase (CS) and pyruvate kinase (PK). At both temperatures, weight-specific activity of CS decreased throughout the ontogenetic development from protozoea II (PZ II) to postlarva XII–XIV (PL XII–XIV). PK activity reached a pronounced peak in PL V–VI, followed by a further decrease in PL XII–XIV. Temperature reduction produced variation in oxygen consumption rates (QO2), ammonia–N excretion and in enzyme activities. Ammonia–N excretion was higher at 20 °C in mysis III (M III), PL V–VI and PL XII–XIV, resulting in substantially lower O:N ratios in these stages. QO2 was increased in protozoea II (PZ II) and mysis I (M I) at 26 °C, while no difference in QO2 was detected in the subsequent stages at either temperature. This fact coincided with higher CS and PK activities in M III, PL V–VI and PL XII–XIV at 20 °C compared with 26 °C. Regressions between individual enzyme activities and dry weight exhibited slope values of 0.85–0.92 for CS and 1.1–1.2 for PK and temperature reduction was reflected by higher slope values at 20 than at 26 °C for both enzymes. Weight-specific CS activity was positively correlated with QO2 at 20 and 26 °C, and may thus be used as an indicator of aerobic metabolic rate throughout the early stages of F. paulensis. The variation in enzyme activities is discussed in relation to possible metabolic adaptations during specific ontogenetic events of the F. paulensis life cycle. Here, the catalytic efficiency of energy-metabolism enzymes was reflected in ontogenetic shifts in behaviour such as larval settlement and the adoption of a benthic existence in early postlarvae. In most cases, enhanced enzyme activities appeared to counteract negative effects of reduced temperature.  相似文献

The effects of neuropeptide Y (NPY) administered intramuscularly or orally on postlarvae (PLs) of two penaeid species were investigated in this study. In experiment 1, food intake (FI) of Marsupenaeus japonicus PLs (0.96 g), injected with NPY at 0.6 microg per g BW, was investigated within 48 h posttreatment. In experiment 2, oral administration of NPY (at doses of 0, 0.125, 0.25, 0.5 microg g(-1) food) on feed intake and growth performance of Penaeus semisulcatus PLs (0.27 g) was examined for 6 weeks. In experiment 1, NPY injection significantly increased average daily FI of M. japonicus PLs within the first 24 h compared to the control (P<0.05), but its stimulatory effect decreased on the second day (P>0.05). The increase in FI was 33% during the first 24 h and 17% during the next 24 h. In experiment 2, significant (P<0.05) differences were found among the groups in terms of weight gain and food utilization (P<0.05). Mean FI significantly increased (as much as 1.3-fold over the control) when NPY was orally administered at doses from 0.125 to 0.5 microg g(-1) feed. There was a positive relationship between FI and final weight (y=-0.972+2.098x, R(2)=0.81) and between FCE and NPY doses in the diets (y=45.37+3.46x, R(2)=0.91). The present findings indicated for the first time that NPY is a potent stimulator of food intake when administered either intramuscularly or orally to penaeid shrimps.  相似文献

1. The sialidase purified from the hepatopancreas of Penaeus japonicus is able to bind the acidic beta-galactosidase in vitro. No protective protein, Mr 32,000, was detected in either purified enzyme preparation. 2. The specific activity of the isolated sialidase is 55.0 mU/mg of protein. After polyacrylamide gel electrophoresis under denaturing conditions, the purified shrimp enzyme was found to consist of monomers of Mr 32,000. 3. The sialidase from shrimp has an isoelectric point (pI) of 4.6 +/- 0.1. 4. The shrimp enzyme has the pH optimum at 5.0 and its Km was 5.5 microM with 2'-(4-methylumbelliferyl)-alpha-D-N-acetylneuraminic acid as substrate. Pyloric pattern-generating neurons that control the pyloric region of the foregut were identified in the stomatogastric ganglion of the most primitive decapod genus Penaeus. Five types of motor neurons and one interneuron are involved in generation of pyloric motor pattern. One cell type of motor neurons innervates muscles of both the gastric mill and the pylorus like the gastric motor neurons in Cancer, but unlike those in Panulirus. These identified neurons are connected to each other either by electrical or inhibitory chemical synapses to construct the neural circuit. This pyloric circuit is similar to the homologous circuit of other crustacean species though some differences are seen in synaptic connections, supporting the hypothesis that the basic design of the neural circuit has been conserved during evolution of the Malacostraca, and that differences have occurred in the synaptic connectivity as the foregut structure has become complex. The motor neurons use either acetylcholine or glutamate as a neurotransmitter like in reptantians. The foregut structure, the number of the pyloric cells, muscle innervation, neurotransmitters, and circuitry are compared among malacostracan crustaceans to provide insight into how the neural circuits change and evolve to produce the motor patterns mediating behaviour. Metapenaeopsis sibogae inhabits the South Pacific and Southeast Asia and is an emerging fisheries resource. However, studies on the population biology of M. sibogae are scant, and their morphometric relationships have not been examined. Thus, we studied the relative growth of M. sibogae, hypothesizing that its morphological sexual maturity can be estimated based on abrupt shifts in allometry. The relationships of body length (BL) and body weight (BW) with carapace length (CL) were determined by the least squares method. Data for each sex were repeatedly partitioned into two size-delimited subsets (early and late phases), considering a CL value as a hypothesized transition point. Analysis of the sum of squared residuals indicated that two separate linear BL/CL models fit the data better than a single linear model showing clear transition points in both males (13.2 mm CL) and females (15.0 mm CL). The changes in allometry in the BL/CL relationships between early- and late-phased males and females were indicated by a 20.5 and 21.5% decrease in the slope, respectively. These transition points could be associated with sexual maturity. The growth rate of BW vs. CL slowed significantly after morphological sexual maturity was reached in both sexes, demonstrating the existence of dimorphic growth.  相似文献

In the present study on female Parapenaeopsis hardwickii information is presented concerning the variation in blood glucose concentration during the different stages (I to V) of ovarian growth. A direct proportionality was found between blood glucose level and ovary growth. There was an incessant hike in blood glucose level as ovary development proceeds from stage I to IV but a drastic downfall was noticed in stage V, i.e. in the spawned prawns. Penaeoidean shrimp pleonal muscle is a valuable economic resource worldwide, but little is known of its development during larval stages. The development of pleonal muscle in Penaeus (Litopenaeus) vannamei was studied by rhodamine-phalloidin staining and laser-scanning confocal microscopy. Dorsal pleonal muscle was first evident at the protozoea I stage while ventral pleonal muscle was present by the protozoea II stage. Identifiable ventral pleonal muscles were evident by the protozoea III stage and all ventral muscle types were present in the mysis I. The tail flex response began at the mysis stage and growth of existing pleonal muscles continued. The pleopods formed during the mysis stages, with coxal and basis muscles developed by mysis III. The pleopods became functional beginning with the first post-larval stage. We conclude that the pleonal muscle pattern of P. vannamei larvae is similar to that of adult Penaeus setiferus, and that homologous muscles are present. The major formation of dorsal pleonal muscles occurs during the protozoea II stage, while significant development of ventral pleonal muscles occurs during the protozoea III stage.  相似文献

Variations of protein concentrations in the haemolymph, hepatopancreas and ovaries were studied during the ovarian maturation of the shrimp Penaeus schmitti. The main variations observed during this physiological process are the increase of protein and the decrease of water contents in the ovary, more pronounced at the first stages of gonadosomatic index (GSI:0-4). During ovarian maturation, water content of the ovaries and hepatopancreas decrease from 10.7 and 3.9% respectively. In relative values, protein content increases from about 40.0% in the ovary, 47.5% in the haemolymph and 15.4% in the hepatopancreas. In absolute values, the protein concentrations of the hepatopancreas showed no significant variation. In the opposite, ovary protein content is 14.7 fold higher at the end than the early stages of vitellogenesis. Relations between haemolymph, hepatopancreas and ovary with eventual transfers of protein material are discussed.  相似文献
