Elapid, crotalid and viperid venoms were screened in vitro and in vivo for cytotoxicity towards B16F10 melanoma and chondrosarcoma cell lines. The cytotoxic activity of elapid venoms was considerably higher than that of viperid or crotalid venoms. Elapid venoms disrupted the cell membrane within the first hour, leading to cell death. The strongest activity was found in the venom of Naja nigricollis. The venoms of some Viperidae and of all Crotalidae examined caused the cells to become rounded, without loss in their original volume, and to form aggregates. These changes were reversible when cells were changed to fresh medium. In vivo experiments with the venom of Naja nigricollis were in total agreement with the results achieved in vitro with melanoma cells and the venom exhibited similar cytotoxic activity on chondrosarcoma, inhibiting its development in vivo.  相似文献

Increase in the extracellular Ca2+ concentration from low (≤ 10−7 M) to normal (10−3 M) caused morphological changes of cultured myocardial cells obtained from fetal mouse heart. The extracellular Na+ and K+ concentrations of the normal medium (10−3 M Ca2+) did not significantly affect the genesis of these morphological changes. Like Ca2+, Ba2+ and Sr2+, but not Mg2+, Co2+ or Ni2+, could induce morphological changes. Increase in the extracellular Ca2+ concentration from 10−8 M to 10−3M also caused excess uptake of 45Ca2+ by cultured myocardial cells. B–16CW 1 cells, which did not show these morphological changes, did not take up excess 45Ca2+ on this treatment. Treatments, such as addition of verapamil or incubation at pH 6.3, which reduced the genesis of morphological changes, reduced the rate of 45Ca2+ uptake by myocardial cells. These facts show that the morphological changes of myocardial cells induced by increasing the extracellular Ca2+ concentration from low to normal are due to excess uptake of Ca2+ by the myocardial cells.
The morphological changes of cultured myocardial cells induced by increasing the extracellular Ca2+ concentration from low to normal were reversed on further incubation of the cells in medium with or without Ca2+.  相似文献

Abstract— The present study was undertaken to characterize the cholinergic system of primary cell cultures of mouse and rat CNS.
In confirmation of previous reports, primary cultures were found to contain choline acetyltransferase (ChAc). Furthermore they contain acetylcholine (ACh) as measured by two different bioassays. They also synthesize [3H]ACh from [3H]Choline offered to the cultures.
The formation of [3H]ACh is inhibited in the presence of hemicholinium-3 (10−6 m ) to 50% or ouabain (10−3 m ) to 20% of the values found in untreated cultures. Omission of Na + from the incubation solution also diminishes the [3H]ACh formation of the cells.
[3H]ACh is released upon depolarisation by K+ ions in a concentration dependent manner. The release can be prevented by lack of Ca2+ ions in the incubation solution.  相似文献

Even in the presence of glucose the growth of Marchantia polymorpha L. (cell line HYH-2F) requires light, and growth is more sensitive to 10−6 M 3-(3, 4-dichlorophenyl)-1, 1-dimethylurea than to 10−4 Antimycin A. The inability of the cells to grow in the dark is due to the low level of respiration. The respiration rate under light increased to four times the dark value. The values of the compensation ratio (the photosyntehtic rate/the respiration rate) for the oxygen exchange were below 1.0 daring the growth period, although oxygen evolution was found. At the early exponential phase, oxygen evolution was 0.373 μmol (mg cell dry weight)−1 h−1 [61.7 μmol (mg chlorophyll)−1 h−1]. M. polymorpha cells are unable to grow anaerobically in the light without a supply of carbon dioxide. When 1% carbon dioxide in nitrogen is supplied, photochemically produced oxygen and energy are sufficient for sustained growth although at significantly reduced yields in both cell dry weight and chlorophyll. Photosyntehtic CO2 assimilation rate was 0.13 μmol (mg cell dry weight)−1 h−1[11.3 μmol (mg chlorophyll)−1 h−1]. At least one-third of the carbon atoms in cellular constituents seem to be derived from atmospheric carbon dioxide, which indicates that M. polymorpha cells grow photomixotrophicaily.  相似文献

Iodination of cytotoxin P4, isolated from the venom of Naja nigricollis nigricollis, develops gradually and depends on the molar ratio between the free iodine and the cytotoxin reaching a maximum of two equivalents at a molar ratio of 250 or higher. The cytotoxic activity was also gradually decreased and was totally abolished when one equivalent of iodination was achieved. However, antigenic properties of the cytotoxin were preserved in the iodinated form. When the iodination of the cytotoxin was carried out with a carrier free radiolabeled iodide, the molar ratio was 0.05 resulting in labelling of only 2% of the cytotoxin molecules, which explains the cytotoxicity of the radiolabeled mixture.  相似文献

Abstract: Recessive mutations in the slyD (sensitivity to ly sis) gene were isolated by selecting for survival after induction of the cloned lysis gene E of bacteriophage φX174 [1]. The slyD mutation, transduced into the normal φX174 host, Escherichia coli C, confers an absolute block on the plaque-forming ability of the wild-type phage, indicating that slyD is required for E function. slyD encodes a protein with 196 residues. A segment corresponding to the first 142 residues of the predicted SlyD protein has significant similarity throughout its length to the FKBP family of peptidyl-prolyl cis-trans isomerases, or rotamases. The C-terminal 46 codons of slyD encode a remarkable histidine-rich peptide which is a metal-binding domain [2]. This sequence is dispensable for slyD function in E -mediated lysis. Although there is no obvious phenotype associated with the slyD genotype other than the resistance to E -mediated lysis, overexpression of slyD causes cells to filament and to increase significantly in diameter. Mutations in φX174 can restore the plaque-forming ability of the phage on a slyD host. These pos ( p lates on s lyD) mutants plate on E. coli C wild-type and slyD . Cytotoxin P4 was isolated from the venom of Naja nigricollis nigricollis in three steps and contained 55% of the crude cytotoxic activity. It had a molecular weight of 8 KD, was stable over a pH range of 1-11 and in boiling water for at least 15 min. It had no measurable enzymatic activities, but was destroyed by proteases. Concentrations of 0.8, 1, 1.2, 25. 20 and 45 ug/ml, were needed to destroy murine melanoma B16 and WEHI 3B leukemia, rat chondrosarcoma, mouse erythrocytes and spleen cells, and human erythrocytes, respectively, thereby showing preferential cytotoxicity to the examined tumor cells. It also prevented the development of the melanoma, leukemia and chondrosarcoma tumors in vivo when mixed with the cells prior to the injection into the animal.  相似文献

Abstract— Synthetic substance P initially increased cyclic AMP levels and subsequently induced neurite extension in cultured neuroblastoma N 18 cells. The magnitude of these effects depended on the concentration of fetal calf serum (FCS) in the culture medium, being more evident in the presence of a lower (0.1%) concentration of FCS.
In Eagle's medium containing 0.1% FCS, low concentrations of substance P (10−7-10−5 M) increased cyclic AMP levels and stimulated neurite extension.
In Eagle's medium containing 5%FCS, both substance P at concentrations of 10−5-10−3M and dibutyryl cyclic AMP at concentrations of 10−4-10−2M increased cyclic AMP levels and stimulated neurite extension. The activities of acetylcholinesterase, (Na++ K+)-, HCO3 and Mg2+ -stimulated-ATPase were also increased. Cell growth was inhibited.
Substance P at concentrations of 10-7-10−5M also stimulated the adenylate cyclase activity of a particulate fraction of N 18 in a concentration-dependent manner.  相似文献

Exoprotease activity of Leuconostoc oenos in stress conditions   总被引:1,自引:1,他引:0  
Exoprotease activity during 48 h of total energy and nutrient starvation was examined in Leuconostoc oenos X2L isolated from wine. Starved cells after 2 h of incubation at 30 °C in citrate buffer, 0.05 mmol 1−1 pH 5, showed greater extracellular proteolytic activity than at the onset of starvation. In the presence of 60 mg l−1 SO2 and 8% or 12% ethanol, the proteolytic activity was higher ; 10 mmol l−1 Ca2+ and Mg2+ produced an increase in protease activity during starvation. Glucose and 2-deoxyglucose (2-DOG) were found to repress synthesis by 80% and 100%, respectively. Cyclic adenosine 3'-5'-phosphate increased the exoprotease activity and reverted the repression by glucose and 2-DOG. De novo synthesis of proteins was required for the exoprotease activity by cells submitted to stress conditions. The absence of protease activity in the supernatant fluids from chloramphenicol-treated cells indicated that the activity is a result of deliberate release and not of passive cell lysis.  相似文献

The growth of Fucus vesiculosus L. germlings in chemically defined culture media containing a range of Cu concentrations (20–1000 nM) was monitored simultaneously with measurement of the Cu speciation in the media by competitive equilibrium-adsorptive cathodic stripping voltammetry. Fucus vesiculosus germlings were found to exude Cu-complexing ligands with conditional stability constants of the order of 1.6 × 1011. Ligand concentrations increased with increasing total dissolved Cu concentrations (CuT) until a concentration of 500–800 neq Cu·L−1 was reached. Concentrations of the ligand exceeded CuT in treatments containing 20 and 100 nM Cu, were similar to CuT in the 500-nM Cu treatment, but were less than CuT in the 1000-nM treatment. Therefore, [Cu2+] were calculated to be at concentrations of 10−11− 10−10 M in the 20- and 100-nM treatments, 10−9 M in the 500-nM treatment, and 10−7 M in the 1000-nM treatment. Growth rates were lowest at Cu2+ concentration > 10−9. These results are discussed within the context of the potential roles for exuded copper-complexing ligands.  相似文献

The effect of 253.7 nm ultraviolet radiation on elongation growth, medium acidification and changes in electric potential difference between vacuole and external medium in cells of maize ( Zea mays L.) coleoptile segments was investigated. It was found that irradiation with 390, 1170, 3900 and 5 850 J m−2 UV-C (ultraviolet radiation 253.7 nm) inhibited elongation growth, whereas at 195 J m−2 stimulation of growth was observed. The administration of IAA (10−5 M ) to the incubation medium of coleoptile segments partially abolished the inhibitory effect of UV-C. The pH of the incubation medium, measured simultaneously with growth, showed that the exposure of the segments to UV-C caused inhibition of H+-extrusion (or stimulation of H+ uptake). The presence of IAA (10−5 M ) in the incubation medium promoted (except after 5850 J m−2 irradiation) H+-extrusion to a level comparable with that produced by IAA in non-irradiated segments. In UV-C irradiated segments the potential difference underwent significant alterations. Irradiation of coleoptile segments with 390 J m−2 caused a transient depolarization, which was fully reversible within 30 min, while at higher doses depolarization was irreversible. The hyperpolarization of the membrane potential (MP) in cells of maize coleoptile induced by IAA was completely nullified by subsequent irradiation with UV-C. It is suggested that UV-C inhibited IAA-induced growth by a mechanism independent of cell wall acidification.  相似文献

Some classes of marine phytoplankton are believed to be more tolerant of high concentrations of trace metals than others, but the results of experimental tests of this hypothesis are ambiguous. Eleven species of phytoplankton representing five classes were grown in Aquil medium containing Cd concentrations between 10−8 and 10−5 M ([Cd2+]= 10−9.85 to 10−6.84 M), and growth rates and intracellular concentrations of Cd, C, N, and S were measured. The mean Cd2+ concentration (pCd50) that reduced the growth rate of each species to 50% of its maximum varied by 2.5 orders of magnitude, from 10−6.23 for Emiliania huxleyi to 10−8.79 for Synechococcus sp. Taxonomic trends in Cd resistance were not apparent in these data. Cadmium quotas (mol Cd·L−1 cell volume) were lowest in species of Bacillariophyceae (ANOVA, P < 0.001), suggesting that they might regulate Cd transport differently than other taxa. Cellular S:C molar ratios increased in four of seven phytoplankton grown at high pCd (7.37–6.84) compared to low Cd ion concentrations (no added Cd), a result of increases in S·L−1 cell volume. Nitrogen:carbon molar ratios were also higher in Cd-exposed phytoplankton, as changes in N and S were highly correlated ( r = 0.98, P < 0.0001). In two species that were examined, S:C ratios increased as a linear function of increasing Cd concentration. The results demonstrate large variability in Cd resistance among phytoplankton that is primarily a function of interspecific differences in Cd detoxification.  相似文献

Cells of the early chick blastoderms are either preparing for or undergoing regulated morphogenetic movements which culminate in the formation of a three-layered embryo. Information on the changes in the physical-chemical properties of cell surfaces may help in the understanding of this process. The binding of magnesium, manganese, strontium, barium and lanthanum to surfaces of early embryonic cells was estimated by the changes induced by these cations in the cells' electrophoretic mobilities (EPM). Cells show a positive EPM at concentrations of MgCl2 and MnCl2 at 3 × 10−2 M while SrCl2, and BaCl2 were not able to reverse the cells' charge at concentrations up to 6 × 10−2 M. CaCl3 reversed the cells' EPM at concentrations as low as 5 × 10−3 M.
Our results suggest that the surfaces of early embryonic cells have a high affinity for Mg and Mn. This is indicated by a reversal of polarity which cannot be detected in cells of differentiating or adult tissues at the cation concentrations used in these experiments.  相似文献

Abstract. Seedlings of Phaseolus vulgaris were exposed to solutions containing Cd2+ in the range 0 to 1 molm−3. Ethylene formation started following 3 h of exposure to 10−2, 10−1 and 1 mol m−3 Cd2+, peaked at 18 h and returned to a relatively low rate after 24 h. Cadmium-induced ethylene formation depended on the formation of 1-aminocyclopropane-1-carboxylic acid (ACC). Aminoethoxyvinylglycine (AVG, 0.1 mol m−3) inhibited ACC accumulation and ethylene production during exposure to 0.2 mol m−3 Cd2+.
Activity of soluble and ionically-bound peroxidase increased after 18 h of exposure to Cd2+ concentrations above 10−3 mol m−3 due to an increase in activity of cathodic isoperoxidases. Stimulation of soluble and ionically-bound peroxidase by 0.2 mol m−3 Cd2+ was reduced in the presence of 0.1 mol m−3 AVG.
Accumulation of soluble and insoluble ('ligninlike') phenolics was found in plants exposed to Cd2+ (10−2 mol m−3 or above) in the presence or absence of AVG. Deposition of insoluble (autofluorescing) material occurred in cell walls around vessels and was associated with reduced expansion and water content of leaves.  相似文献

Abstract: The effect of colchicine (0.5 mM) and of cytochalasin B (10−4 M) on the release of [35S]taurine from the isolated chick retina, upon stimulation by 68.5 mM-KCl, 10−5 M-veratridine and 10 mM-glutamate, was studied. Cytochalasin and colchicine effects on taurine release were compared with those on K+-stimulated release of [3H]dopamine and [3H]GABA. Colchicine caused a marked decrease of the [35S]taurine release evoked by the three stimulatory agents; it also decreased [3H]dopamine release without affecting that of [3H]GABA. Cytochalasin B significantly decreased the efflux of [35S]taurine stimulated by glutamate or veratridine without altering that evoked by 68.5 mM-KCl. Cytochalasin practically suppressed the [3H]dopamine-stimulated release and slightly decreased that of [3H]GABA. This drug produced a high increase in the spontaneous release of labeled GABA and taurine. These results suggest that the release of taurine and GABA from the chick retina probably occurs through different mechanisms. It is suggested that taurine release may be related to a process involving contractile proteins.  相似文献

Abstract— When butanol-water extracts of rat brain stem were incubated with [3H]5-HT, (5 × 10−7 m ), and the components resolved by chromatography on LH20 Sephadex, a peak representing approximately 70% of the eluted radioactivity was found in chloroform-methanol 4:1. The peak was not found in identically prepared extracts from rat diaphragm, neither was a similar peak found when brain extracts were incubated with [14C]ACh (10−6 m ), suggesting a degree of selectivity. Binding was not saturated at concentrations of 5 × 10−5 m -5-HT. The binding was highly sensitive to the presence of water, requiring about 15% (v/v) for optimum binding. The implications of these findings are discussed in terms of a possible '5-HT receptor'.  相似文献

The intracellular compartmentation of boron (B) in roots of sunflower plants precultured with 100 μ M B (high B) or 1 μ M B (low B) was studied using two independent approaches. In the first approach, short-term efflux studies using the stable isotopes 11B and 10B were carried out. In roots of high B plants, the calculated concentrations of B (nmol gFW −1) were 52.6 in the cell wall, 7.5 in the vacuole, 27.1 in the cytosol and 48.0 in the free space. In roots of low B plants, the concentrations of B (nmol gFW −1) were 43.4 in the cell wall, 2.8 in the vacuole, 17.9 in the cytosol and almost zero in the free space. Although the B supply differed by a factor 100, the B concentrations in the cytosol and the vacuole of low B plants were 66 and 37% of the respective concentrations in high B plants. This suggests an additional role for B in plant metabolism, besides its function in the cell wall. In the second approach, root B pools (cell sap and water-insoluble residue) were determined for comparison, and found to be in good agreement with the results from the efflux study.  相似文献

Abstract: The Ca2+/calmodulin-dependent phosphatase calcineurin may have physiological and pathological roles in neurons, but little is known about the roles of the enzyme in glial cells. We have previously reported that reperfusion of cultured astrocytes in Ca2+-containing medium after exposure to Ca2+-free medium caused Ca2+ influx followed by delayed cell death. In this study, we examined if calcineurin is involved in this Ca2+-mediated astrocytic injury. FK506, an inhibitor of calcineurin, protected cultured rat astrocytes against paradoxical Ca2+ challenge-induced injury in a dose-dependent manner (10−10–10−8 M ). Cyclosporin A at 1 µ M mimicked the effect of FK506. Rapamycin (1 µ M ) did not affect astrocyte injury, but it blocked the protective effect of FK506. Deltamethrin (20 n M ), another calcineurin inhibitor, had a similar protective effect, whereas okadaic acid did not. FK506 affected neither paradoxical Ca2+ challenge-induced increase in cytosolic Ca2+ level nor Na+-Ca2+ exchange activity in the cells, suggesting that the calcineurin is involved in processes downstream of increased cytosolic Ca2+ level. Immunochemical studies showed that both calcineurin A (probably the Aβ2 isoform) and B subunits were expressed in the cells. It is concluded that calcineurin is present in cultured astrocytes and it has a pathological role in the cells.  相似文献

Abstract: The adenylate cyclase activity of rat hippocampal plasma membranes can be stimulated by vaso-active intestinal polypeptide (VIP). Low concentrations (10−9 to 10−7M) of 5'-guanylyl-imido diphosphate (GppNHp) evoke a transient inhibition of the enzyme, which is followed by stimulation with increasing GppNHp concentrations (10−6 to 10−4M). Inclusion of ethyleneglycol - bis - (β - aminoethylether) - N,N' - tetraacetic acid (EGTA) during incubation abolishes the GppNHp inhibition while preserving GppNHp activation. The stimulation induced by GppNHp is amplified by VIP, but the inhibition is unaffected. Adenosine analogs and opiates are inhibitory ligands in the presence of GTP, and their effects can be reversed by the appropiate receptor antagonists, 3-isobutyl-1-methylxanthine and naloxone. Treatment of membranes with trypsin abolishes the GppNHp-induced inhibition without affecting the GppNHp stimulation. The inhibition induced by GppNHp is also abolished by EGTA treatment followed by washing, which coincides wtih a reduction in the adenosine- and opiate-mediated, GTP-dependent inhibition. The GppNHp inhibition can be restored in EGTA-treated but not in trypsin-treated membranes by addition of calcium-calmodulin but not by Ca2+ or Mg2+. Calcium-calmodulindepleted membranes lack calcium stimulation as well as GppNHp-induced inhibition, whereas untreated membranes and calcium-calmodulin-depleted membranes plus exogenous calcium-calmodulin showed calcium stimulation and GppNHp inhibition. These results suggest that calmodulin is involved in both Ca2+ stimulation and guanine nucleotide-mediated inhibition of rat hippocampal adenylate cyclase.  相似文献

Abstract: We investigated the interaction between Pb2+ and protein kinase C (PKC) in the Pb2+-induced release of norepinephrine (NE) from permeabilized adrenal chromaffin cells. Our analysis of endogenous PKC activity in permeabilized cells suggests that Pb2+ interacts with the adrenal enzyme at multiple sites. Pb2+ activates the enzyme through high-affinity ( K A(Pb) = 2.4 × 10−12 M ) interactions and inhibits the enzyme by competitive and noncompetitive interactions with nanomolar-( K i = 7.1 × 10−9 M ) and micromolar- ( K 'i = 2.8 × 10−7 M ) affinity sites, respectively. Activation of PKC by 12- O -tetradecanoylphorbol 13-acetate (TPA) in Ca2+-deficient, Pb2+-containing medium, enhances the Pb2+-induced NE release from permeabilized chromaffin cells by lowering the concentration of Pb2+ required for half-maximal activation of the secretory response from 7.5 × 10−10 to 5.7 × 10−11 M . The PKC inhibitors staurosporine and pseudosubstrate PKC (19–36) abolish the effect of TPA without affecting the Pb2+-induced secretion in the absence of TPA. These results indicate that (a) Pb2+ is a partial agonist of PKC, capable of both activating and inhibiting the enzyme and (b) synergistic activation of PKC by TPA and Pb2+ results in increased sensitivity of exocytosis to Pb2+ but is not obligatory for Pb2+-triggered secretion.  相似文献
