Summary Detached pistils of the clonal variety, Lilium longiflorum Aral No. 5, were submerged before pollination in 50°C water for 0, 1, 2, 3, 4, 5, 6 or 7 min and then immediately compatibly and incompatibly pollinated. Incompatibility, as indicated by pollen tube length after 48 h at 23.5°C, was eliminated by a 1–2 min submersion while compatibility was removed by a 4–5 min one. The window of incubation temperatures at which incompatible and compatible pollen tubes are clearly differentiated occurred between 15 and 30°C.  相似文献

Summary In order to identify the genotypic constitutions of incompatibility in the diploid species, Ipomoea leucantha Jacq. (K221), which is most closely related to the sweet potato, the progenies derived from the reciprocal crosses, backcrosses and testcrosses were analysed. All the plants examined were self-incompatible, and pollen germination was inhibited on the stigma after incompatible pollinations. No reciprocal differences were found in the incompatibility reactions. In the progenies three incompatibility groups were observed which showed the rather simple segregation ratios. The homozygous plants for incompatibility alleles were obtained in the progenies. The experimental results demonstrated a sporophytic type of incompatibility controlled by a single locus with multiple S-alleles exhibiting a dominance relationship in both the pollen and the stigma. The plants obtained in the progenies had the following genotypes: S 1 S 2, S 1 S 3, S 2 S 2, S 2 S 3 and S 3 S 3.  相似文献

Lycopersicon peruvianum (wild tomato) is a gametophytic self-incompatible (SI) species. One natural population has been shown to harbor a self-compatible (SC) allele. A stylar protein associated with the self-compatibility allele has been elucidated using SDS-PAGE. The temporal and spatial expression of this protein is presented and compared with protein expression of two SI alleles. Hybrids containing the SC and SI alleles were used in a backcrossing program to introgress the SC allele into SI backgrounds in six independent lines. Controlled pollinations and SDS-PAGE were used to identify and select classes of progeny. After four backcross generations (approximately 97% recovery of the SI backgrounds) the SC allele still confers self-fertility in lines that contain this allele, providing evidence that the mutation to SC occurred at the S-locus and that the associated protein is likely responsible.  相似文献

Summary The expressed activity in pollen and stigma was determined for both S alleles of sixteen S-alíele heterozygous genotypes and for one of the two S alleles of two additional heterozygotes. Activities were measured using pollen tube penetration and seed set data from reciprocal crosses between each S-allele heterozygote and its two corresponding S-allele homozygotes.In pollen the S-allele activities ranged from zero to 100% inhibition of pollen tube penetration and seed set, and in the stigma they ranged from 8 to 100% inhibition. Of the sixty-eight S-allele activities measured, thirty-three (48%) were 90 to 100% inhibition, nine (13%) were 80 to 89% inhibition and one to five were within each ten-unit range below 80% inhibition.In an S-allele heterozygote, each subset of two S alleles had an activity for each allele in both pollen and stigma which was highly repeatable among duplicate pollinations within and among successive years. Each subset of two S alleles had a specific S-allele interaction in the pollen, and the same or another specific interaction in the stigma. In pairings with six other S alleles, allele S 2 had four calculated levels of activity in pollen that ranged from 88 to 94%, and five levels in the stigmas between 15 and 94%. When paired in a heterozygote, alleles S 3 and S 5 had activities ranging between 42 and 59%, representing mutual weakening of S-allele activity. Also, heterozygote S 15 S 3 had pollen activities, respectively, of 25 and 6%, i.e. mutual weakening in the pollen.These results indicate that in heterozygous combination with a series of other S alleles, each S-allele may have activity in pollen and also in stigma that potentially is between zero and 100% inhibition. They further indicate that the defined sexual-organ X S-allele-interaction Types I, II, III and IV are extremes; all intermediate variations including complete weakening of both alleles are possible. Recessiveness is weakening of the activity of but one of the two S alleles. Summary Pollen tube growth was evaluated using an 18-step scale after both intra- and interspecific pollinations of genotypically widely differing diploid potato species and potato dihaploids expressing monofactorial gametophytic incompatibility. The results obtained account for a wide array of types of pollen tube growth resulting from crossing partners with distinct incompatibility behavior. Based on the assumption that inhibition of pollen tubes is the rule and solely prevented by the pollen itself, a model proposing one common cause underlying different mechanisms for both intraspecific selfincompatibility and interspecific incompatibility in diploids is put forward. Data supporting the model are presented from the experimental results of this study and from the literature. The strength of pollen-style interaction depends on particular S-alleles in combination with recognition and activity properties of the basic S-genotypes. The model is suitable to explain all former observations on incompatibility in diploids with a gametophytic system of incompatibility and, with modifications of the manner of phenotypic expression, also in plants with sporophytic incompatibility. The proposed scheme of pollen tube growth phenotypes permits prediction of pollen tube growth behavior in an intended cross combination. The model is based on both classical Mendelian genetics and recent molecular genetic insight.  相似文献

Summary Pontederia cordata L. (Pontederiaceae), a perennial diploid, possesses the rare genetic polymorphism tristyly. A controlled pollination programme was conducted over a three year period, under glasshouse conditions, on 36 clones of P. cordata var. cordata to examine the nature of the self-incompatibility system. The three major findings of the pollination study were: (1) the three floral morphs display different levels of self-incompatibility, (2) pollen from the two anther levels within a flower exhibits different compatibility behaviour in self-pollinations, (3) considerable individual genetic variation in the expression of self-incompatibility is evident among clones within floral morphs. Similar results were also obtained from a smaller study on 15 clones of P. cordata var. lancifolia conducted over a 6 month period. In common with other Pontederia species the mid-styled morph (M) of P. cordata produces large amounts of seed when self-pollinated with pollen from long-level anthers. A developmental model is proposed to explain the high level of self-compatibility of the M morph in Pontederia species. Self-pollination of segregating progenies from M and S morphs of known incompatibility status demonstrated that the expression of incompatibility is closely associated with style length. It is suggested that overall differences in incompatibility behaviour among the floral morphs may be due to the pleiotropic effects of major genes controlling sub-characters of the tristylous syndrome, rather than linked modifier genes. However, the variable expression of trimorphic incompatibility within floral morphs suggests that this variation may be polygenic in origin.  相似文献

Summary The events of the progamic phase of fertilization have been monitored by in vitro experiments in self compatible (SC), partial self-incompatible (PSI) and self incompatible (SI) lines. The duration of the progamic phase is about 30 h. Treatment with low concentrations of CO2 (3 to 5%) at high relative humidity (rH, 100%) had the following effects: pollen quality, which declines normally during flower ageing, was prematurely reduced; pollen adhesion and germination, both low in SI matings, were increased; the stigma callose response in SI matings was reduced to the low level of SC matings; and the number of pollen tubes in the style after SI matings significantly increased. CO2 concentrations of 4 to 6% applied for 8, 16 or 24 h at 100% rH proved to be the most effective treatment for blocking the SI response in cauliflower.  相似文献

Plants have evolved many systems to prevent inappropriate fertilization. Among them, incompatibility is a well-organized system in which pollen germination or pollen-tube growth is inhibited in pistils. Self-incompatibility (SI), rejecting self-pollen, promotes outbreeding in flowering plants. On the other hand, inter-species incompatibility, preventing gene flow among species to restrict outbreeding, usually occurs unilaterally, and is known as unilateral incompatibility (UI). In Brassicaceae, little is known about the molecular mechanism of UI, although S-locus genes involved in recognition of self-pollen have been characterized in the SI system. In the present study, we characterized novel UI observed between members of the same species, Brassica rapa; pollen of Turkish SI lines was specifically rejected by pistils of the Japanese commercial SI variety Osome. The incompatible phenotype of this intra-species UI closely resembled that of SI. Segregation analysis revealed that the pollen factor of this UI was not linked to the S-locus.The revised version was published online in December 2004 with corrections to figure 1.  相似文献

Summary By means of intra- and intermorph crosses it has been shown that the distyly in Anchusa officinalis L. is governed by a single diallelic locus with dominance. Presence of the dominant allele is reflected in the short-styled morph, while homozygous recessives become long-styled. Most of the short-styled plants are heterozygous, but homozygotes have been found among the raised progeny resulting from crossings. Diallel crosses have revealed that at least two incompatibility loci must exist. These and the morphological locus segregate independently. The type of incompatibility system is as yet unknown, though the great frequency of non-reciprocal incompatibility points to a sporophytic system.  相似文献

The mating systems of most ectomycorrhizal fungi have not been elucidated because of two reasons. One is the difficulty of obtaining homokaryotic isolates for mating tests caused by the low germination rate of basidiospores, and another is the difficulty of checking dikaryotization caused by the absence or inconsistent production of clamp connections on heterokaryotic mycelia under laboratory conditions. Basidiospore germination of a few ectomycorrhizal fungi has been induced by living roots of their host plants. Based on this information, we examined methods to obtain homokaryotic isolates of Rhizopogon rubescens using its host plant, Pinus thunbergii. The basidiospores of R. rubescens appeared to germinate well on an agar plate, on which axenic pine seedlings were grown in advance to induce germination, even when the seedlings were removed from the plate at the time of spore inoculation. To enhance the production rate of clamp connections on the heterokaryotic mycelia of R. rubescens, the culture medium composition was modified. The pH of the medium was critical for the production of clamp connections, and the optimal pH was higher for the production of clamp connections than for mycelial growth. These findings made it possible to conduct mating tests, and we found that the mating system of R. rubescens is bipolar with a multiallelic mating type factor.  相似文献

Summary Intraspecific and reciprocal interspecific crosses involving Zinnia angustifolia clones and Z. elegans lines showed that in both species, sporophytic self-incompatibility (SI) systems were present. Intensity of SI varied among clones and lines, and high self seed set was associated with a concomitant decrease in callose fluorescence in papillae and pollen tubes. Incomplete stigmatic inhibition of pollen germination and tube growth was observed in reciprocal interspecific crosses and associated with callose synthesis, suggesting S-gene activity. Seed set and progeny obtained following Z. angustifolia×Z. elegans matings was comparable to intraspecific compatible matings of Z. angustifolia although the rate of pollen tube growth through the style was slower. In Z. elegans × Z. angustifolia matings, additional prezygotic barriers were present and acted between pollen tube penetration of the stigma and syngamy. SI X SI interspecific incompatibility was essentially unilateral, with no embryos or progeny obtained when Z. elegans was the pistillate parent. 该研究以不结球白菜(Brassica campestris L. ssp. chinensis Makino)自交不亲和系品种‘矮脚黄’为实验材料,利用VIGS技术将BcMLPK基因构建到pTY载体,进行定量分析,研究其在自交不亲和中的功能。结果表明:(1) BcMLPK基因序列长为1 443 bp,编码480个氨基酸,将BcMLPK蛋白氨基酸序列与其他物种的MLPK氨基酸比较表明,BcMLPK基因与其他物种中该基因具有高度保守的结构域。(2) 成功构建pTY BcMLPK载体,采用基因枪法侵染‘矮脚黄’,其新生的真叶有明显的花叶症状,选取发病的叶片及花蕾进行实时定量qRT PCR,发现侵染的植株中BcMLPK基因表达量约为对照组的50%。(3) 花期对植株进行自交授粉,实验组有明显的结荚现象,而对照组则出现雌蕊未能明显伸长,显示受精异常的现象;统计结果表明实验组的自交不亲和指数和结实率均高于对照组。研究表明,不结球白菜MLPK基因的沉默能够抑制‘矮脚黄’自交不亲和相关基因BcMLPK的转录水平表达。  相似文献

A genomic clone of the S11 allele from the self-incompatibility locus (S locus) in Solanum chacoense Bitt. has been isolated by cross-hybridization to the S. chacoense S13 allele and sequenced. The sequence of the S11 allele contains all the features expected for S genes of the Solanaceae, and S11 expression, as assessed by northern blots and RNA-PCR, was similar to that of other S. chacoense S alleles. The S11 protein sequence shares 95% identity with the phenotypically distinct S13 protein of S. chacoense and is the gametophytic S allele with the highest similarity to an existing allele so far discovered. Only 10 amino acid changes differentiate the mature proteins from these two alleles, which sets a new lower limit to the number of changes that can produce an altered S allele specificity. The amino acid substitutions are not clustered, suggesting that an accumulation of random point mutations can generate S allele diversity. The S11 intron is unusual in that it could be translated in frame with the coding sequence, thus suggesting an additional mechanism for the generation of new S alleles.  相似文献

Inbreeding effects and incompatibility relationships were examined in strains of the egg parasitoid Trichogramma nr brassicae (Hymenoptera: Trichogrammatidae) from southeastern Australia. Crosses between strains provided weak evidence of incompatibility in a few cases. However sex ratio in crosses within strains tended to be more female-biased than in crosses between strains. Inbreeding was imposed for four generations (F>0.59) of sib mating. The fitness of inbred strains was compared to that of outbred strains generated by crossing the inbred strains. No effects of inbreeding were found for any of the four female traits examined (fecundity, body length, head width and hind tibia length), indicating that T. nr. brassicae is not subjected to inbreeding depression. Inbreeding effects were also not found for male mating success as expected for the haploid sex. There were differences among strains for all traits apart from fecundity, indicating heritable variation. Strain differences for fitness measures were uncorrelated with wasp size. The potential use of inbreeding in the quality control of Trichogramma for mass-release is discussed. Inbreeding may be a useful tool in minimising the effects of laboratory adaptation, thereby extending the useful life of a strain.  相似文献

沃尔巴克氏体(Wolbachia)作为节肢动物的胞内共生菌,可以引起宿主产生雌性化、孤雌生殖、杀雄和胞质不相容性(cytoplasmic incompatibility, CI) 4种生殖表型。其中CI是最常见的现象,表现为受感染的雄性昆虫与未感染或感染不兼容Wolbachia的雌性昆虫交配时引起胚胎死亡;而雌性感染同种Wolbachia时胚胎能够正常发育。CI是由被称为CI因子(cifA和cifB)的Wolbachia基因对调控的。其中,CifB作为毒剂在雄性中表达诱导产生CI,而CifA作为解毒剂在雌性中表达拯救CI。本文综述了CI因子结构、功能和作用机制的研究,以期为未来利用Wolbachia和CI进行蚊媒疾病和农业虫害的防控奠定基础。  相似文献

Drosophila melanogaster (Meigen) females from stocks collected at Melbourne (latitude 37°S) show partial incompatibility when mated with males from stocks collected at Townsville (latitude 19°S) on the east coast of Australia. The reciprocal cross is compatible. Eggs have reduced hatchability in the incompatible cross. The incompatibility is maternally inherited over three generations. Compatibility can be restored by culturing Townsville flies on medium with tetracycline for one generation and by using 2-week-old Townsville males.

Résumé Les souches de D. melanogaster récoltées à Melbourne (37°S) et Townsville (19°S) sur la côte Est de l'Australie montrent une incompatibilité partielle lorsque les femelles Melbourne sont accouplées aux mâles Townsville. Une telle incompatibilité n'est décelée, ni dans les croisements intrapopulations, ni dans le croisement réciproque. Le taux d'éclosion des oeufs est réduit d'environ 30% dans le croisement incompatible, mais la viabilité des larves n'est pas modifiée. Les éléments, mâle et femelle, de ce système d'incompatibilité sont hérités maternellement pendant 3 générations de croisements en retour. La compatibilité peut être intégralement rétablie en cultivant pendant une génération la souche Townsville avec un régime contenant de la tétracycline, et partiellement rétablie en utilisant des mâles âgés de 2 semaines.

该研究以杜鹃花属5亚属3组17亚组38种,计109个不育组合及91个可育组合的杂交结果为依据,对4个杂交不亲和指标与败育系数及败育频度参数进行了分析。结果表明:(1)杜鹃花属不同类群间杂交的不育组合比例约为54.5%,其不亲和与败育包括不能坐果(capsul aborted,Cab型)、坐果但不能形成种子(seed aborted,Sab型)和能形成种子但不能发芽(seed not germinated,Sng型) 3种情况,其中Cab与Sab类型均可能是前合子期不亲和与后合子期不亲和的复合表征,Sng型则可以肯定为后合子期种子发育阶段败育的情况,Cab∶Sab∶Sng=81∶13∶15;而可育组合中部分败育苗无疑属于"杂种不活hybrid inviability"的败育类型。(2)有关不育类型的分布与杜鹃花属植物亲本类群及其分类与亲缘关系具有明显关联,从同一亚组内、同一亚属内到不同亚属间杂交的不育类型的分布呈Sng型→Sab型→Cab型增加的趋势,杂交双亲分类上亲缘关系越密切Sng型的频度越高,反之关系越疏远Cab型的频率越高,亚属间远交往往止于Cab型。(3)"杂种不活"为败育苗的表现形式之一,从同一亚组、同一亚属到不同亚属间杂交,其败育等级频度分布通常也从无或轻度败育到严重败育方向发展,常绿杜鹃亚属内杂交的败育情况明显低于杜鹃亚属内杂交,这与后者多倍体亲本的介入相关,而上述2亚属间杂交的苗木败育则更加严重,但映山红(R. simsii)分别与百合花杜鹃(R. liliiflorum)和毛肋杜鹃(R. augustinii)的亚属间杂交未出现败育苗分布。  相似文献

The Western Australian annual of restricted distribution,Villarsia congestiflora, has distylous flowers and pollen-size heteromorphism. Results of a crossing program indicate that this species is strongly self-incompatible, that crosses among individuals of the same morph produce little or no seed, and that intermorph crosses produce copious seed. The species shares a number of morphological and ecological traits withV. capitata, another distylous, self-incompatible annual species of Western Australia with a greater area of distribution. Artificial hybrids between the two, however, showed reduced pollen stainability. Both species combine a number of specialized morphological features with a breeding system that is primitive for the familyMenyanthaceae.  相似文献
