The present study investigated the effect of nitrogen source (NH4+; NO3) at different concentrations on the accumulation of biomass and secondary metabolites in adventitious root cultures of Hypericum perforatum L. Cultures were initiated in shake flasks by using half-strength Murashige and Skoog (MS) medium with B5 vitamins, 1.0 mg l−1 indole-3-butyric acid, 0.1 mg l−1 kinetin, 3% (w/v) sucrose, and different ratios of ammonium and nitrate (0:30, 5:25, 10:20, 15:15, 20:10, 25:5, and 30:0 mM, using NH4Cl and KNO3). The cultures were maintained in darkness. The medium supplemented with 5:25 (mM) NH4+/NO3 resulted in the optimum accumulation of biomass and total phenols and flavonoids. The antioxidant potential of a methanolic extract, measured as the 1, 1-diphenyl-2-picrylhydrazyl and 2, 2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activities, of H. perforatum adventitious roots showed that antioxidant activity was high from root extracts that were grown on higher concentrations of NO3 nitrogen (15, 20, and 25 mM). Further, assessment of hydrogen peroxide (H2O2) and malondialdehyde content of the root extracts revealed that cultures supplemented with higher levels of NO3 nitrogen (15–30 mM) were under oxidative stress, which boosted the levels of secondary metabolites in the adventitious roots. These results suggest that optimal adventitious root biomass could be achieved with the supplementation of cultures with 5:25 ratios of MS nitrogen sources.  相似文献

The present work deals with optimization of adventitious shoot culture of Bacopa monnieri for the production of biomass and bacoside A and has investigated the effects of macro elements (NH4NO3, KNO3, CaCl2, MgSO4 and KH2PO4) and nitrogen source [NH4 +/NO3 ] of Murashige and Skoog (Physiol Plant 15:473–497, 1962) medium (MS) on accumulation of biomass and bacoside A content. Optimum number of adventitious shoots (99.33 shoots explant−1), fresh weight (1.841 g) and dry weight (0.150 g) were obtained in the medium with 2.0× strength of NH4NO3. The highest production of bacoside A content was also recorded in the medium of 2.0× NH4NO3, which produced 17.935 mg g−1 DW. The number of adventitious shoot biomass and bacoside A content were optimum when the NO3 concentration was higher than that of NH4 +. Maximum number of shoots (70.00 shoots explant−1), biomass (fresh weight 1.137 g and dry weight 0.080 g) and also bacoside A content (27.106 mg g−1 DW) were obtained at NH4 +/NO3 ratio of 14.38/37.60 mM. Overall, MS medium supplemented with 2.0× NH4NO3 is recommended for most efficient bacoside A production.  相似文献

A mathematical computer-aided model CELLOP was constructed in which the desirability functions in a three-dimensional experimental design can be used to find the optimal growing conditions for plant cells. CELLOP is intended for the optimisation of 3 to 6 physical, chemical, or biological variables in the cultivation conditions of plant cell cultures. The model was used to optimise the culturing conditions (calcium, inorganic nitrogen, and sucrose concentrations) for coumarin-producing, spontaneously embryogenic cell lines of angelica Angelica archangelica L. subsp. archangelica and hogs fennel Peucedanum palustre (L.) Moench. For A. archangelica the overall optimum concentrations were 0.47 mM Ca2+, 5.06 mM NO3 , 0.40 mM NH4 +, and 96.25 mM sucrose. The dry mass was 24.7 % higher and the coumarin content 40.5 % higher than those achieved in the standard 75 % Gamborg B5 medium. For A. archangelica the highest embryogenic activity was reached in the media containing 1.25 mM Ca2+. In the case of P. palustre the overall optimum concentrations were 1.60 mM Ca2+, 2.84 mM NO3 , 0.23 mM NH4 +, and 85.10 mM sucrose. For P. palustre the dry mass production increased by 61.8 % and the coumarin content by 58.1 % compared to the values achieved in the Gamborg B5 medium. For P. palustre the highest embryogenic activity was reached in the presence of 50.0 mM NO3 and 4.01 mM NH4 +.  相似文献

为了提高西洋参不定根的诱导率和生长速度,该研究以西洋参鲜根为外植体,在基本培养基的基础上优化IBA、碳源、氮源和磷源等营养成分。结果表明:西洋参不定根诱导过程可以明显分为外植体脱分化(愈伤化)、再分化(根形成)和根伸长等三个阶段; MS基本培养基更有利于西洋参不定根的诱导,可能与MS培养基中矿质元素含量高有关;当培养基中IBA浓度达到2 mg·L-1时,外植体表面上不定根分布密度大,诱导率达到(96±3.5)%;培养基中添加蔗糖到30 g·L-1时,不定根的诱导效果最好,但继续提高浓度后不定根变短、直径变粗;培养基中NO3-∶NH4+和PO43-浓度分别为20∶10(总氮量30 mmol·L-1)和25.0mmol·L-1时,西洋参不定根诱导率达到最大。结果提示优化培养条件可以显著改善西洋参不定根的诱导和生长,为后续西洋参不定根规模化培养提供理论支持。  相似文献

Summary Foliar nutrition has been conceived as a possible means of overcoming the recalcitrance of Prosopis chilensis (Molina) Stuntz explants to standard in vitro culture. The foliar uptake of cations (K from 20 gl−1 KNO3 and Ca from 50 gl−1 CaCl2), anions (NO3 from 50 gl−1 KNO3 and PO4 from 50 gl−1 NaH2PO4), and glucose from a 100 mg l−1 solution studied. All of the nutrients examined were absorbed. The efficacy of foliar nutrition in prolonging the vigor of micropropagated P. chilensis shoot tips was compared with nutrients supplied as a liquid to the base of the stem (liquid) or as an agar-solidified medium (agar). A foliar-feeding apparatus was constructed that employed pressurization of the medium reservoir to drive the medium into the culture vessel with a passive return by a siphoning effect. The medium used was Murashige and Skoog with 30 gl−1 sucrose, 0.1 mgl−1 benzylaminopurine, and 1 mgl−1 indole-3-butyric acid. Over a 9-wk test period it was found that explants cultured by foliar nutrition performed significantly better than those grown on agar for shoot length, nodal production, and leaf retention; and better than liquid MS for node production. There was no significant difference among the three treatments in percentage survival, percentage rooting, or the mean number of roots.  相似文献

The effects of macronutrients (NO3 , NH4 + and PO4 3−) on cell growth and triterpenoids production inCentella asiatica cell suspension cultures were analyzed using the Box-Behnken response surface model experimental design. In screening and optimization experiments, PO4 3− as a single factor significantly influenced cell growth where increasing the phosphate level from 0.1 to 2.4 or 2.6 mM, elevated cell growth from 3.9 to 14–16 g/L. The optimum values predicted from the response surface model are 5.05 mM NH4 +, 15.0 mM NO3 and 2.6 mM PO4 3−, yielding 16.0 g/L cell dry weight with 99% fitness to the experimental data. While the NH4 +-NO3 interaction influenced cell growth positively in the optimization experiment, NH4 + and NO3 as single factors; and interactions of NO3 -PO4 3−, NH4 +-PO4 3− and NH4 +-NO3 were all negative in the screening experiment. Cell growth and the final pH level were positively affected by PO4 3−, but negatively affected by NH4 + and NH4 +-PO4 3− interactions. The different effects of factors and their interactions on cell growth and final pH are influenced by a broad or narrow range of macronutrient concentrations. The productions of triterpenoids however were lower than 4 mg/g cell dry weight.  相似文献

The effects of the concentration of PO4 3-, NO3 , Fe 2+, sucrose and inoculum size on the accumulation of blue pigment and growth of suspension cultures of Lavandula spica D.C. ( L. latifolia Vill.), were studied. The combination of 2.5 mM PO4 3-, 14.1 mM NO3 , 1 mM Fe 2+, 30 g l–1 sucrose and 10 g l–1fresh weight of inoculum, promoted a 7-fold enhancement on the productivity of blue pigment in comparison to the control medium. Among all the culture medium constituents tested, phosphate exerted the highest beneficial effect and Fe2+ showed to be essential for the accumulation of the pigment. High levels of sucrose (60 or 90 g l–1) did not stimulate the accumulation of blue pigment. In these conditions, cell growth and cell viability were drastically affected.  相似文献

Ruta graveolens in vitro cultures are a potential source of secondary metabolites (furanocoumarins) of significant medical interest. Experiments led in our laboratory showed that micropropagated shoots were richer in furanocoumarins than any other plant material. In order to optimize the molecule production by such cultivation systems, several factors related to the culture medium were studied. Effects of medium composition on biomass growth and furanocoumarin content were analysed and optimal conditions were determined for phosphate (300 mg l−1 of NaH2PO4), nitrate (2527 mg l−1 of KNO3), carbon source (10 g l−1 of sucrose) and phytohormones (2,4-dichlorophenoxyacetic acid (2,4-D) 50 μM and benzylaminopurine (BAP) 50 μM). Ruta shoot growth and furanocoumarin production were compared for optimized and standard culture conditions. Specific medium gave better results in terms of growth (tD equal to 6.9 days against 8.6 for standard conditions) but no significant differences appeared concerning metabolite concentrations. Protoplasts were isolated from embryogenic suspension cultures derived from avocado (Persea americana Mill.) zygotic embryos and nucellus in an enzyme digestion solution consisting of 1% cellulase Onozuka RS, 1% Macerase R10, 0.2% Pectolyase Y-23, 0.7 M mannitol. 24.5 mM CaCl2, 0.92 mM NaH2PO4 and 6.25 2-[N-morpholino]ethanesulfonic acid (1.5 ml) mixed with 0.7 M MS8P (2.5 ml). MS-8P medium consisted of Murashige and Skoog salts without NH4NO3, 1 mg l–1 thiamine HCl, 100 mg l–1 myo-inositol, 3.1 g l–1 glutamine and 8P organic addenda. Medium osmolarity was adjusted with 0.15 M sucrose and 0–0.55 M mannitol. Protoplast yields of 3.5×106 protoplasts g–1 were obtained. Growth and development of the protoplasts were significantly affected by osmolarity, nitrogen source, plating density and culture medium dilution. Under optimum conditions, proembryos developed directly from embryogenic protoplasts and subsequently into somatic embryos. Optimum conditions for somatic embryo development included the culture of protoplasts at a density of 0.8–1.6×105 ml–1 in 0.4 M MS8P for 2–3 weeks, followed by subculture in 0.15 M MS8P at a diluted density of 20–40× for 1 month in darkness to obtain somatic embryos. Mature somatic embryos were recovered on semisolid medium; however, a low frequency of plantlet recovery (≤1%) from protoplast-derived somatic embryos was observed. Humans have increased the availability of nutrients including nitrogen and phosphorus worldwide; therefore, understanding how microbes process nutrients is critical for environmental conservation. We examined nutrient limitation of biofilms colonizing inorganic (fritted glass) and organic (cellulose sponge) substrata in spring, summer, and autumn in three streams in Michigan, USA. Biofilms were enriched with nitrate (NO3 ), phosphate (PO4 3−), ammonium (NH4 +), NO3  + PO4 3−, NH4 + + PO4 3−, or none (control). We quantified biofilm structure and function as chlorophyll a (i.e., primary producer biomass) and community respiration on all substrata. In one stream, we characterized bacterial and fungal communities on cellulose in autumn using clone library sequencing and denaturing gradient gel electrophoresis to determine if community structure was linked to nutrient limitation status. Despite oligotrophic conditions, primary producer biomass was infrequently nutrient limited. In contrast, respiration on organic substrata was frequently limited by N + P combinations. We found no difference between biofilm response to NH4 + versus NO3 enrichment, although the response to both N-species was positively related to water column PO4 3− concentrations and temperature. Molecular analysis for fungal community composition suggested no relationship to nutrient limitation, but the dominant members of the bacterial community on cellulose were different on NO3 , PO43, and NO3  + PO4 3− treatments relative to control, NH4 +, and NH4 + + PO4 3− treatments, which matched patterns for biofilm respiration rates from each treatment. Our results show discrete patterns of nutrient limitation dependent upon substratum type and season, and imply changes in bacterial community structure and function may be linked following nutrient enrichment in streams.  相似文献

Withania somnifera is an important medicinal plant that contains withanolides and withaferins, both bioactive compounds. We have tested the effects of macroelements and nitrogen source in W. somnifera cell suspension cultures with the aim of optimizing the production of biomass and withanolide A. The effects of the macroelements NH4NO3, KNO3, CaCl2, MgSO4 and KH2PO4 at concentrations of 0.0, 0.5, 1.0, 1.5 and 2.0× strength and of the nitrogen source [NH4 +/NO3 (mM/mM) ratio of: 0.00/18.80, 7.19/18.80, 14.38/18.80, 21.57/18.80, 28.75/18.80, 14.38/0.00, 14.38/9.40, 14.38/18.80, 14.38/28.20, and 14.38/37.60 (mM)] in Murashige and Skoog medium were tested for biomass and withanolide A production. The highest accumulation of biomass [147.81 g l−1 fresh weight (FW) and 14.02 g l−1 (dry weight (DW)] was recorded in the medium containing a 0.5× concentration of NH4NO3, and the highest production of withanolide A content was recorded in the medium with 2.0× KNO3 (4.36 mg g−1 DW). The NH4 +/NO3 ratio also influenced cell growth and withanolide A production, with both parameters being larger when the NO3 concentration was higher than that of NH4 +. Maximum biomass growth (110.45 g l−1 FW and 9.29 g l−1 DW) was achieved at an NH4 +/NO3 ratio of 7.19/18.80, while withanolide A production was greatest (3.96 mg g−1 DW) when the NH4 +/NO3 ratio was 14.38/37.60 mM.  相似文献

Effects of phosphite (Phi) on phosphate (Pi) starvation responses were determined in Ulva lactuca L. by incubation in Pi‐limited (1 μM NaH2PO4) or Pi‐sufficient (100 μM NaH2PO4) seawater containing 0–3 mM Phi. Exposure to 1 μM NaH2PO4 decreased the growth rate and the content of free Pi and esterified‐P but increased the activities of extracellular alkaline phosphatase (EC 3.1.2.1) and intracellular acid phosphatase (ACP; EC 3.1.2.2); two ACP isozymes observed by activity staining on isoelectric focussing (IEF) gel were induced. The Km value of Pi uptake rate was decreased by incubation with 1 μM NaH2PO4 and the decrease in Km value was inhibited by 2 mM Phi, reflecting the operation of a high‐affinity Pi uptake system at low Pi concentrations. In the presence of Phi, the growth rate of Pi‐sufficient and Pi‐starved thalli decreased as Phi concentrations were increased from 0 to 2 mM. As Phi concentrations were increased from 0 to 2 mM, the free Pi contents in both Pi‐sufficient and Pi‐starved thalli decreased, but the esterified‐P contents in Pi‐starved thalli increased, whereas those in Pi‐sufficient thalli increased at 1 mM Phi and decreased at 2 mM Phi. Cell wall localized AP activity in both Pi‐sufficient and Pi‐starved thalli decreased as Phi concentrations were increased from 0 to 2 mM. Intracellular ACP activity in Pi‐starved thalli decreased as Phi concentrations were increased from 0 to 2 mM but was not affected in Pi‐sufficient thalli. The induction of ACP isozyme activity and high‐affinity Pi uptake system in Pi‐starved thalli was inhibited by Phi. The present investigation shows that Phi interrupts the sensing mechanisms of U. lactuca to Pi‐limiting conditions.  相似文献

Summary To improve proliferation of soybean cultures in liquid medium, the effects of sucrose; total inorganic nitrogen; content of No3 , NH4 +, Ca2+, PO4 3−, K+; NH4 +/NO3 ratio; and medium osmotic pressure were studied using cv. Jack. Sucrose concentration, osmotic pressure, total nitrogen content, and ammonium to nitrate ratio were found to be the major factors controlling proliferation of soybean embryogenic cultures. Growth decreased linearly as sucrose concentration increased from 29.7 mM to 175.3 mM. A sucrose concentration of 29.2 mM, a nitrogen content of 34.9 mM, at 1 to 4 ammonium to nitrate ratio were found to be optimal for the fastest proliferation of soybean embryogenic cultures. There was no significant effect on proliferation of cultures when concentrations of NH4 +, Ca2+, PO4 3−, and K+ were tested in the range of 3.50 to 10.50, 1.02 to 3.06, 0.68 to 2.04, and 22.30 to 36.70 mM, respectively. The relative proliferation of embryogenic cultures of four soybean genotypes was evaluated in Finer and Nagasawa medium and in the new medium formulation. Despite genotype-specific differences in growth, the genotypes tested showed a biomass increase in the new formulation equal to 278, 269, 170, and 251% for Chapman, F138, Jack, and Williams 82, respectively, relative to their growth on standard FN medium. Cotyledon explants of Panax ginseng produced somatic embryos directly on solid hormone-free MS medium containing 3% (w/v) sucrose while high concentration of NH4NO3 (60 mM) induced embryogenic callus. Ten subcultures of the embryogenic callus on hormone-free MS medium with 40 mM NH4NO3 gave hormone-independent proliferation of callus, which exhibited proliferation potential even on MS medium with a standard level of NH4NO3 (20 mM). Pulse treatment of callus with exogenous auxin or cytokinin (1.0 mg 1–1 2,4-D, 1.0 mg 1–1 kinetin) resulted in the loss of the hormone-independent characteristic and caused the callus to brown. For the suspension culture, embryogenic callus was transferred to MS liquid medium containing 3% (w/v) sucrose in an 500 ml Erlenmyer flask. Embryogenic cell clumps in full-strength MS liquid medium discharged toxic substances, resulting in strong suppression of cell growth. In 1/3-strength MS medium, exudation of toxic material did not occur. Embryogenic cell clumps were mass-grown on a large-scale in a bioreactor (20-1), showing a 7.1 increase of fresh weight in 1/3-strength MS medium with 3% (w/ v) sucrose after 5 weeks of culture. Total ginsenoside content of cultured embryogenic cell clumps was low and 6 times below naturally-cultivated ginseng roots.  相似文献

Oncidium cultivars gave different embryogenic responses of leaf explants when affected by auxins (2,4-D, IAA, IBA and NAA), cytokinins (2iP, BA, kinetin, TDZ and zeatin), sucrose, NaH2PO4, casein hydrolysate, peptone, and glutamine. The best embryogenic responses of cv. Sweet Sugar were at 20 g dm−3 sucrose, 85 mg dm−3 NaH2PO4 and 3 mg dm−3 kinetin, respectively. The development of somatic embryos on leaf explants of cv. Sweet Sugar was delayed for about 10 – 20 d in comparison with cv. Gower Ramsey. On growth regulator-free medium, about 40 % of leaf derived embryos of cv. Gower Ramsey were fused together in their basal parts and so called multiple-state embryos. However, under the same condition, the embryos of cv. Summary The purpose of this study was to investigate the phytotoxicity of nitrapyrin 2-chloro-6-(trichloromethyl)pyridine to sunflower (Helianthus annuus L.) under different N regimes and to see if N forms affect the phytotoxicity of nitrapyrin. Sunflower was grown in pot culture for 21 days and was fertilized with (NH4)2SO4, NH4NO3 and NaNO3 to provide 0, 100 and 200 ppm N and with nitrapyrin application of 0 and 20 ppm. All N-treated sunflower plants in all N regimes and regardless of titrapyrin treatment produced more root and shoot dry weights and contained a significantly higher N than untreated check. Nitrapyrin toxicity appeared as a curling of leaf margin and a tendril type of stem growth, the visible toxicity symptoms decreased in the order: (NH4)2SO4>NH4NO3>NaNO3. Furthermore nitrapyrin addition suppressed sunflower growth in each N regime, the suppressing effect being greater with (NH4)2SO4 and NH4NO3 than as with NaNO3. Although, shoot growth from plants receiving nitrapyrin was not significantly affected by any N regime, root growth of nitrapyrin-treated plants was somewhat restricted by NH4 +−N nutrition relative to NO3 −N nutrition.  相似文献

The question of how tropical trees cope with infertile soils has been challenging to address, in part, because fine root dynamics must be studied in situ. We used annual fertilization with nitrogen (N as urea, 12.5 g N m?2 year?1), phosphorus (P as superphosphate, 5 g P m?2 year?1) and potassium (K as KCl, 5 g K m?2 year?1) within 38 ha of old‐growth lowland tropical moist forest in Panama and examined fine root dynamics with minirhizotron images. We expected that added P, above all, would (i) decrease fine root biomass but, (ii) have no impact on fine root turnover. Soil in the study area was moderately acidic (pH = 5.28), had moderate concentrations of exchangeable base cations (13.4 cmol kg?1), low concentrations of Bray‐extractable phosphate (PO4 = 2.2 mg kg?1), and modest concentrations of KCl‐extractable nitrate (NO3 = 5.0 mg kg?1) and KCl‐extractable ammonium (NH4 = 15.5 mg kg?1). Added N increased concentrations of KCl‐extractable NO3 and acidified the soil by one pH unit. Added P increased concentrations of Bray‐extractable PO4 and P in the labile fraction. Concentrations of exchangeable K were elevated in K addition plots but reduced by N additions. Fine root dynamics responded to added K rather than added P. After 2 years, added K decreased fine root biomass from 330 to 275 g m?2. The turnover coefficient of fine roots <1 mm diameter ranged from 2.6 to 4.4 per year, and the largest values occurred in plots with added K. This study supported the view that biomass and dynamics of fine roots respond to soil nutrient availability in species‐rich, lowland tropical moist forest. However, K rather than P elicited root responses. Fine roots smaller than 1 mm have a short lifetime (<140 days), and control of fine root production by nutrient availability in tropical forests deserves more study.  相似文献

The effects of nitrogen form and phosphorus source on the growth, nutrient uptake and rhizosphere soil property of tea (Camellia sinensis L.) were investigated in a pot experiment. The experiment was performed with a compartmental cropping device, which enables the collection of rhizosphere soil at defined distances from the root of tea plant. Nitrogen was supplied as nitrate or ammonium in combination with soluble phosphorus as Ca(H2PO4)2 or insoluble P as rock phosphate. The leaf dry matter production of tea was significantly greater in the treatments with NH4 + than NO3 -, whereas dry matter production of root and stem was not significantly affected. Addition of phosphorus as either source did not influence the dry matter production. The concentrations of K in root, Mg and Ca in both the shoot and root supplied with NO3 - were significantly higher than in NH4 + and influence of P sources was minor. On the contrary, Al and Mn concentrations were significantly larger in NH4 --fed plants which could be attributed to remarkably increased availability of Al and Mn caused by acidification of the rhizosphere soil (the first 1-mm soil section from the root surface) with NH4–N nutrition. The concentration of N in shoot was also significantly higher in NH4- than in NO3-fed plants, indicating higher use efficiency of NH4–N. Whatever the phosphate source, rhizosphere pH declined in ammonium compared to in nitrate treatment. The pH decrease was much larger when no P or soluble P were applied and reached 0.85–1.30 units which extended to 3–5 mm away from the root surface. Exchangeable acidity, content of exchangeable Al and Mn were also considerably higher in the rhizosphere soils of NH4 + fed tea plants. Significant amounts of P dissolved from rock phosphate accumulated in rhizosphere of NH4 +, not NO3 -, suggesting that the dissolution of rock phosphate was induced by the proton excreted by tea root fed with ammonium. With soluble P addition, shoot and root P concentrations were greater in NH4 + than in NO3 - treatment and it appeared that this difference could not be sufficiently explained by the available P content in soil which was only slightly higher in NH4 + treatment. With rock phosphate addition, the shoot and root P concentrations were hardly affected by nitrogen form, although the available P content was much higher and accumulated in the rhizosphere soil supplied with ammonium. An efficient and reliable transformation system for a very important medicinal plant Papaver bracteatum was developed through optimization of several factors that affect the rate of effective A. rhizogenes-mediated transformation and growth rate of hairy root. Five bacterial strains, A4, ATCC15834, LBA9402, MSU440 and A13, and three explants types, hypocotyls, leaves and excised shoots were examined. The highest frequency of transformation was achieved using LBA9402 strain in the excised shoots. Several inoculation and co-cultivation media and different concentration of arginine were evaluated using LBA9402 strain and the excised shoots as explant. Interestingly, a drastic increase in the frequency of transformation (47.3 %) was observed when Murashige and Skoog medium containing 1 mM arginine and lacking NH4NO3 KH2PO4, KNO3 and CaCl2 was used. The effect of sucrose concentration and the ratio of NH4 +: NO3 ? on hairy root biomass was examined. Maximum biomass was obtained in 30 g/l sucrose and 20:10 mM ratio of NH4 + to NO3 ? on MS medium. Transgenic hairy root lines were confirmed by polymerase chain reaction (PCR) and Southern hybridization.  相似文献
