The reactivity of retinol with peroxynitrite, which is a strong oxidant and has been reported to induce several biological damages, was investigated. 13-cis-14-nitroretinol (1), 13-trans-14-nitroretinol (2), 13-apo-β-carotenone (3), retinal (4), 11,14-epoxyretinol (5), and 11,15-epoxyretinol (6) were identified as reaction products of retinol with peroxynitrite. From these results, it was observed that retinol can undergo a nitration reaction with peroxynitrite. Furthermore, the formation mechanisms of 1, 2, and 3 from retinol with peroxynitrite are discussed.  相似文献

The in vitro reactivities of astaxanthin toward peroxynitrite were investigated and the reaction products after scavenging with peroxynitrite were analyzed in order to determine the complete mechanism of this reaction. A series of carotenoids, 13-apo-astaxanthinone (1), 12′-apo-15′-nitroastaxanthinal (2), 12′-apo-astaxanthinal (3), 10′-apo-astaxanthinal (4), 9-cis-14′-s-cis-15′-nitroastaxanthin (5), 14′-s-cis-15′-nitroastaxanthin (6), 13-cis-14′-s-cis-15′-nitroastaxanthin (7), 10′-s-cis-11′-cis-11′-nitroastaxanthin (8), 13,15,13′-tri-cis-15′-nitroastaxanthin (9), 9-cis-astaxanthin (10), and 13-cis-astaxanthin (11), were isolated from the reaction products of carotenoids with peroxynitrite. Our previous studies achieved for the first time the isolation of nitro derivatives from the reaction of astaxanthin with peroxynitrite. Here we identify the major remaining reaction products of this reaction and investigate the stabilities of the nitro astaxanthins.  相似文献

【背景】我国甘蔗生产中氮肥过量施用严重,导致生产成本居高不下,充分发挥甘蔗与内生固氮菌的联合固氮作用,减少氮肥施用量,对促进我国甘蔗产业可持续发展具有重要意义。【目的】筛选优势甘蔗内生固氮菌,对其基本特性、联合固氮效率及促生长功能进行评价。【方法】从甘蔗根系分离到一株内生固氮菌GXS16,利用乙炔还原法测定固氮酶活性,通过PCR扩增nifH基因确定菌株为固氮菌;通过形态观察、Biolog检测和16S rRNA基因序列分析等对菌株进行分类;通过接种盆栽甘蔗检测菌株的促生长作用,采用15N同位素稀释法检测菌株相对固氮效率。【结果】菌株GXS16固氮酶活性为2.42μmol-C2H4/(h·mL),根据菌株培养性状和菌体形态观察、Biolog检测、16S rRNA、nifH、acdS基因序列分析结果,菌株GXS16属于伯克氏菌属(Burkholderia);菌株GXS16还具有1-氨基环丙烷-1-羧酸脱氨酶(1-Aminocyclopropane-1-Carboxylate Deaminase,ACC)活性及合成生长素吲哚乙酸...  相似文献

Abstract

Aim: This study investigated the effects of polar Butia odorata fruit extract on metabolic, inflammatory, and oxidative stress parameters in rats submitted to a hyperlipidaemia condition induced by tyloxapol.

Methods: Animals were divided into 3 groups: saline, saline plus tyloxapol, and B. odorata extract plus tyloxapol. Animals were treated for 15?days with a saline solution or B. odorata fruit extract and after hyperlipidaemia was induced by tyloxapol.

Results: Treatment with B. odorata extract reduced serum triglyceride, total cholesterol, C-reactive protein, and adenosine deaminase and butyrylcholinesterase activities when compared to the tyloxapol group. HDL-cholesterol and paraoxonase 1 activity were higher in B. odorata extract treated animals when compared to tyloxapol-treated animals. No differences were observed in hepatic oxidative stress parameters. Phenolic compounds present in B. odorata fruit extract were identified and quantified by LC-MS/MS.

Conclusion: These findings indicated that phenolic rich B. odorata extract has hypolipidemic and anti-inflammatory effects in hyperlipidemic rats.  相似文献

Abstract

Liposomes loaded with the rhenium compound (bis-dimethylsulfoxido-cis-tetrachlorodi-μ-pivalatodirhenium(III) (cis–Re2((CH3)3CCOO)2Cl4?2DMSO, I) and cisplatin in the molar ratio of 4:1 as well as those loaded only with I were synthesized and characterized by scanning electron microscopy, transmission electron microscopy, dynamic light scattering and electronic absorption spectroscopy. The relative stability of liposomes loaded with I is reflected by a minimal change in the electronic absorption spectra over a period of 8 days whereas the stability of those loaded with both drugs is lower, which we ascribe to the formation of new Re-Pt species inside the liposomes. Furthermore, the investigations of the co-encapsulation effects on the anticancer activity of the Re-Pt system were undertaken. Importantly, the co-encapsulated liposomes exhibit synergistic or additive anticancer activities in vivo, e.g. introduction of these liposomes into tumor-bearing rats demonstrated their antianemic, nephro- and hepato-protecting effects. These liposomes, which are active in cancer treatment, protect the dirhenium compounds from hydrolysis and preserve the biological properties of the Re-Pt hybrid. This study reveals the importance of combined therapy in nanotechnology and medicine.  相似文献

trans -[PtCl4(NH3)(thiazole)] (1), trans-[PtCl4(cha)(NH3)] (2), cis-[PtCl4(cha)(NH3)] (3) (cha =cyclohexylamine), and cis-[PtCl4(NH3)2] (4) has been investigatedat 25 °C in a 1.0 M aqueous medium at pH 2.0–5.0 (1) and 4.5–6.8 (24) using stopped-flow spectrophotometry. The redox reactions follow the second-order rate law , where k is a pH-dependent rate constant and [GSH]tot the total concentration of glutathione. The reduction takes place via parallel reactions between the platinum(IV) complexes and the various protolytic species of glutathione. The pH dependence of the redox kinetics is ascribed to displacement of these protolytic equilibria. The thiolate species GS is the major reductant under the reaction conditions used. The second-order rate constants for reduction of compounds 14 by GS are (1.43±0.01)×107, (3.86±0.03)×106, (1.83±0.01)×106, and (1.18±0.01)×106 M−1 s−1, respectively. Rate constants for reduction of 1 by the protonated species GSH are more than five orders of magnitude smaller. The mechanism for the reductive elimination reactions of the Pt(IV) compounds is proposed to involve an attack by glutathione on one of the mutually trans coordinated chloride ligands, leading to two-electron transfer via a chloride-bridged activated complex. The kinetics results together with literature data indicate that platinum(IV) complexes with a trans Cl-Pt-Cl axis are reduced rapidly by glutathione as well as by ascorbate. In agreement with this observation, cytotoxicity profiles for such complexes are very similar to those for the corresponding platinum(II) product complexes. The rapid reduction within 1 s of the platinum(IV) compounds with a trans Cl-Pt-Cl axis to their platinum(II) analogs does not seem to support the strategy of using kinetic inertness as a parameter to increase anticancer activity, at least for this class of compounds. ABSTRACT

Organophosphorus compounds (OP) are mainly used in agriculture as pesticides. Unfortunately, each year many rural workers are intoxicated by these compounds and, many times, the diagnosis of the exact molecule causing the intoxication can be tardy, exposing the patients to a huge risk of death. One way of preventing this delay is the use of enzymatic biosensors like the enzyme Esterase 2 from Alicyclobacillus acidocaldarius (AaEST2), which is an efficient fluorescent biosensor for OP identification. However, although this enzyme has been well studied experimentally, the complete understanding of the energy transfer processes that occur between AaEST2 and OPs is still obscure, making it difficult the accurate identification of the OP. In order to better understand this process, we applied in this work molecular docking and molecular dynamics studies, together with the Förster fluorescence resonance energy transfer (FRET) theory, to achieve a better understanding of the fluorescence profiles that are described in the literature and correlate them to individual OPs. Our results suggest that the pesticides chlorpyrifos, diazinon, parathion and paraoxon are all capable of quenching the residue Trp85 from AaEST2, triggering fluorescence. This supports our hypothesis that AaEST2 can be used as a fluorescent biosensor for the detection of organophosphorus compounds.  相似文献

【目的】进一步了解兴义喀斯特洞穴可培养放线菌资源及产活性代谢产物的能力。【方法】选取多种分离培养基,利用稀释直接涂布平板法对贵州黔西南兴义市多个喀斯特洞穴的土壤和岩石进行可培养放线菌资源分离;利用三种发酵培养基对相关放线菌进行生物产物初筛。【结果】根据16S rRNA基因序列的比对分析,将分离得到的251株放线菌分别归类到44个属,其中链霉菌属(Streptomyces)占分离菌株的比例为24.30%,小单孢菌属(Micromonospora)占比11.95%,红球菌属(Rhodococcus)占比9.16%,微杆菌属(Microbacterium)占比7.17%,诺卡氏菌属(Nocardia)占比6.37%,该五类放线菌为该地区可培养放线菌的优势菌群。对70株细菌进行活性次级代谢产物筛选,其中35株放线菌对指示菌具有抑制活性,且主要类群为链霉菌属和小单孢菌属。【结论】贵州兴义喀斯特洞穴中存在丰富多样的放线菌类群,且蕴藏大量具有产生活性次级代谢产物能力的菌株,为医药产业提供潜力菌株资源,极具进一步发掘和研究的价值。  相似文献

Abstract

Metal-based ethanolamines, (L1)–(L4) coordinated with Co(II), Cu(II), Ni(II) and Zn(II) metals in 1:2 (metal:ligand) molar ratio to produce new compounds have been reported. These compounds were screened for their bactericidal/fungicidal activity against a number of bacterial (Escherichia coli, Shigella flexneri, Pseudomonas aeruginosa, Salmonella typhi, Staphylococcus aureus and Bacillus subtilis) and fungal strains (Trichophyton longifusus, Candida albicans, Aspergillus flavus, Microsporum canis, Fusarium solani and Candida glabrata) alongside against a shrimp species known as Artemia salina. The screening results indicated that metal complexes have significantly higher activity than uncomplexed ligands against one or more bacterial/fungal species due to chelation. The ligand (L4) displayed good bacterial and fungal activity as compared to other ligands. The antibacterial results revealed that the Zn(II) complex (16) of (L4) was found to be the most active complex and Co(II) complex (14) of the same ligand (L4), demonstrated the highest antifungal activity.  相似文献

In our searching program for novel sorbicillin related compounds, three novel compounds, spirosorbicillinols A (1), B (2), and C (3), were isolated from the fermentation broth of the USF-4860 strain isolated from a soil sample. The planar structures of compounds 13 were determined from spectroscopic evidence and degradation reaction, and that of 1 was the same as that of 2. The relative stereochemistries of compounds 13 were determined by 1H-1H coupling constants, the elucidation of HMBC and NOESY spectra in detail. 1 and 2 were stereoisomers at C8 position, each other. We propose that compounds 1 and 2 were formed by exo and endo intermolecular Diels-Alder reaction between sorbicillinol as a diene and scytolide (proposed precursor-1) as a dienophile, respectively. Similarly, we propose that compound 3 was formed by an endo intermolecular Diels-Alder reaction between sorbicillinol and proposed precursor-2.  相似文献

【目的】选择了几株隶属于新鞘氨醇杆菌属(Novosphingobium)的有和没有多环芳烃(PAHs)降解能力的细菌,在基因组水平比较分析它们的趋化通路,并探究其中一些菌株对芳香化合物和三羧酸(TCA)循环中间代谢产物的趋化性。【方法】通过基因组的比较分析,研究了几株新鞘氨醇杆菌的趋化蛋白组成和趋化基因分布,并采用滴定法和游动平板法检测了相关菌对芳香族化合物和TCA循环中间代谢产物的趋化性。【结果】N.pentaromativorans US6-1、N.pentaromativorans F2、Novosphingobium indicum K13、Novosphingobium stygium DSM 12445、Novosphingobium sp.C2AC等5株新鞘氨醇杆菌对芳香族化合物和TCA循环中间代谢物具有不同程度的趋化性;所选的7株已完成基因组测序的新鞘氨醇杆菌N.pentaromativorans F2、N.pentaromativorans US6-1、Novosphingobium sp.PP1Y、Novosphingobium sp.AP12、Novosphingobium sp.Rr 2-17,Novosphingobium sp.B-7和Novosphingobium sp.DSM 19370均含有趋化蛋白MCP、CheW、CheA、CheB、CheR和CheY,且亲缘关系最近的US6-1、F2和PP1Y 3株菌的che基因簇中的基因排列一致,为che W-Y-D-B-R-A-(X);新鞘氨醇杆菌的趋化系统属Fla类型。【结论】几株新鞘氨醇杆菌都具有较为完整的趋化通路,且对多种芳烃及其代谢物的趋化性各不相同,其中菌株US6-1趋化现象最明显。  相似文献

【目的】提出一种合成微生物组的理性构建策略,用于构建郫县豆瓣蚕豆醅初始发酵的微生物组合菌剂。【方法】采取自下而上的合成微生物组理性构建策略,以相对丰度、频率和特征向量中心度作为核心微生物属的选择指标,分析确定蚕豆醅发酵核心微生物;设计模拟原位体系的全合成培养基,并利用该培养基快速、稳定地检测核心微生物包括产香性能在内的发酵特征。基于核心微生物的产香互补性能进行双菌组合发酵实验,结合核心微生物之间的生长相互作用,设计三菌组合发酵菌剂并验证其发酵性能。【结果】本研究确定并分离了郫县豆瓣蚕豆醅发酵过程中的 9种核心微生物。检测核心微生物产生的挥发性风味化合物,发现酵母菌类、乳酸菌类和其他类微生物之间存在产香互补关系。然后,结合微生物间的生长抑制关系设计了由乳酸片球菌、肉葡萄球菌及异变假丝酵母组成的三菌组合菌剂。与企业原位发酵样品相比,三菌组合菌剂产生的挥发性化合物数量达到原位样品的63.1%,化合物种类结构较为相似。与原位样品相比,组合菌剂样品氨基酸态氮浓度提升了21.8%。【结论】本研究提出了一种自下而上的合成微生物组理性构建策略,基于此策略设计了郫县豆瓣蚕豆醅发酵组合菌剂。使用该组合菌剂作为起始发酵剂发酵的郫县豆瓣蚕豆醅具有良好的风味谱和优异的氨基酸态氮水平。本研究在合成微生物组构建与发酵食品工艺改造方面具有较大的科学与应用价值。  相似文献

Abstract

As a result of a literature survey and examination of specimens, we believe that the identification of three species of propaguliferous Pohlia in China, Pohlia flexuosa, Pohlia proligera, and Pohlia leucostoma needs to be clarified. Pohlia flexuosa has two differing types of propagule: one short and oblong and the other long and thread-like; both propagules have inconspicuous leaf primordia, and can occur on the same stem. Propagules of P. proligera are shortly clavate to long vermicular, with one to three unicellular or multicellular prong-like leaf primordia arising at the apex of the propagule, and the leaf primordia are approximately one-fourth of the total length of the propagules. Pohlia leucostoma has long filiform propagules, with two or three short peg-like leaf primordia about one-tenth of total propagule length.  相似文献

Abstract

In Euphorbiaceae tribe Hippomaneae is the pantropical genus Microstachys, comprised by 24 species, mostly from Brazil. Palynological studies with several representatives of the genus are scarce. The purpose of this study was to describe the pollen morphology of Microstachys and find possible differentiating characteristics between its species. Ten species of the genus were analysed, represented by 21 herbarium collections. Their pollen grains were acetolysed, measured and photographed under light microscopy (LM) and scanning electron microscopy (SEM). The analysed species possess pollen in monads, of small to medium size, isopolars, amb distincticly trilobed, oblate-spheroidal, prolate-spheroidal or subprolate, tricolporate with narrow and very long colpi, almost fused at the poles sometimes forming an invagination with tapering ends and margo of the colporus wide to very wide and psilate; endoapertures lalongate to very lalongate with tapering ends, presence of a narrow costae distinct in the endoaperture; exine microreticulate homobrochate, walls with simple columella; at the mesocolpus the sexine separates from the nexine forming cavea; and sexine always thicker than the nexine. Microstachys is a stenopalynous genus, despite its species varying in pollen size, shape and width of the cavea, margo of the colporus, and length of the endoapertures, demonstrating taxonomic value, and indicating a contribution towards future systematic analyses.  相似文献

A new species,Deprea nubicola, is described. Although the corolla is similar in some aspects to that ofD. bitteriana, D. nubicola is distinguished by its diminutive leaves, a low density of hairs in the corolla annulus, glabrous filaments, and a distinctive long-pedicellate, subulate-lobed fruiting calyx.Deprea nubicola shares withD. ecuatoriana a fruiting calyx that tightly invests the berry. The distribution ofD. nubicola is limited to the Sierra Nevada de Santa Marta of northern Colombia. A key to the species ofDeprea is provided.  相似文献

The activity on Aspergillus spp. growth and on ochratoxin A production of two novel chromene dimers (3) was evaluated. The results of the bioassays indicate that the chromene dimer 3a inhibited mycelia growth by approximately 50% (EC50) at 140.1 μmol L−1 for A. niger, 384.2 μmol L−1 for A. carbonarius, 69.1 μmol L−1 for A. alliaceus and 559.1 μmol L−1 for A. ochraceus. When applied at concentrations of 2 mmol L−1, 3a totally inhibited the growth of all Aspergillus spp. tested. Furthermore, ochratoxin A production by A. alliaceus was reduced by about 94% with a 200 μmol L−1 solution of this compound. A moderate inhibitory effect was observed for the analogous structure 3b on ochratoxin A production but not in mycelia growth. No inhibition was registered for compounds 2a and 2b, used as synthetic precursors of the dimeric species 3.  相似文献

【目的】探究土霉素残留对蔬菜自然发酵过程中微生物群落演替和代谢产物动力学的影响,为评估抗生素残留对蔬菜发酵的影响提供理论基础。【方法】超高效液相色谱-串联质谱法测定土霉素残留;高效液相色谱法测定有机酸、电子鼻和气相色谱-质谱联用测定挥发性成分和高通量技术测定微生物种类。【结果】蔬菜自然发酵过程中,土霉素残留从4.00 mg/L下降到2.53 mg/L;不含抗生素残留的蔬菜发酵含有同型和异型乳酸发酵,而土霉素残留的蔬菜发酵仅含有同型乳酸发酵;同时,其特征微生物由Lactobacillus pentosus和Lactobacillus plantarum转变为Lactobacillus paratarrginis、Lactobacillus buchneri和Lactobacillus kisonensis;土霉素残留明显影响了乳酸、柠檬酸、乙酸、香茅醇、3-辛醇、异硫氰酸烯丙酯、乙酸香叶酯、乙烯基硬脂醚和异硫氰酸苯乙酯等代谢产物的含量。【结论】土霉素残留影响了蔬菜乳酸发酵的类型、微生物群落的演替、有机酸和挥发性化合物的形成过程,因此应将抗生素残留纳入发酵蔬菜原料的质量控制指标。  相似文献

Background: There is a growing interest in understanding the gas exchange between the atmosphere and seasonally snow-covered regions, especially in light of projections that climate change will alter the timing and extent of seasonal snow cover. In snow-covered ecosystems, gas fluxes are due both to microbial activity in the snow-covered soils and to chemical and physical reactions with the various gases and/or dissolved constituents in the snowpack. Niwot Ridge, in the Colorado Rocky Mountains, has one of the most extensive sets of measurements of winter gas exchange globally.

Aims: Our goal was to examine the temporal patterns and environmental controls on Niwot Ridge of gas fluxes for gases with different sources and sinks.

Methods: Here, we review the concentrations and fluxes that have been measured for carbon dioxide, nitrous oxide, methane, nitrogen oxides, ozone, gaseous elemental mercury and volatile organic carbon compounds.

Results and Conclusions: We looked for similarities and differences among the gases, but in many cases, the origin, fate and controls of these fluxes still need to be determined. However, we believe that many of the biologically driven reactions are the result of exponential growth of a winter microbial community during the long period of stable environmental conditions under the seasonal snowpack.  相似文献
