Abstract

Seminal integument and germination of Cercis siliquastrum L. – A histological study of the seminal integument of Cercis siliquastrum L. has been carried out, in order to explain whether the obstacle to the germination, removed by a cold-treatment, was caused by a mechanical action of the integument itself. We have noted that both the cold-treated seeds which germinate and the non-treated control seeds which don't germinate show in the long run the same modifications. We have therefore come to the conclusion that the inability to germinate found in the non-treated seeds is not due to the mechanical resistance of the integument and we think it necessary to return to further researches, also at a biochemical level.  相似文献

Luzula spicata L. seeds are completely dormant at maturity. A germination inhibitor is present at the micropylar end. Normally, the only effective means of eliciting germination is a precise scarification of the micropylar end which inactivates the inhibitor. Exogenous application of gibberellic acid, kinetin, KNO3, and thiourea have no affect on the dormancy of unscarified seeds. Scarification of the hilar end of the seed does not elicit germination, but when gibberellic acid is applied to the hilar scarified seeds moderate germination results. Presumably, these seeds are dormant due to a deficit of endogenous gibberellin; a condition which can be overcome by the application of gibberellic acid to seeds scarified at a site in itself ineffective in producing germination. Apparently the gibberellic acid serves to initiate amylase activity in the endosperm, overcoming the inhibitor block. Luzula spicata seed dormancy is apparently unique in that a germination inhibitor is operative in conjunction with the commonly recognized gibberellin-amylase mechanism.  相似文献

Abstract

Consequences and significance of dormancy Prolongation in «Triticum durum» Desf. — Different environment conditions have effect on the dormancy duration: this has been ascertained also for durum wheat Cappelli. Starting from this acquisition, the author does the embryo exchange between seeds of Cappelli different for dormancy degree. Experiments were carried out with three seed lots of Betula nana collected in 1967 from different localities in Norway. Seeds were stratified for 0-20 days in dark at +2-+3 °C on filter papers moistened with distilled water, or treated with solution of GA3 for 24 h at room temperature, and then moved into special germination boxes that were placed in different temperature conditions. All the seed lots had conditional dormancy. Quantitatively, the dormancy was different in the different seed lots (pronenances), but there were no qualitative difference in the reaction to stratification gibberellic acid and to germination temperature. Differences between seed lots may have been due to different stage of seed development. The dormancy was deepest at low temperatures(12 and 15°C) decreasing gradually with increasing temperature (to 24 °C). The dormancy was effectively broken by a short stratification (from 5 to 15 days), and by treatment with gibberellic acid. The deeper the dormancy and the lower the germination temperature the longer the stratification that was needed for maximum germination. Similarly, the concentration of GA3 needed for maximum germination increased with decreasing temperature and with increasing dormancy.  相似文献

以紫斑牡丹种子为试验材料,考察不同浓度赤霉素(GA_3)处理下种子生根情况,同时对其种皮纹饰进行了扫描电镜(SEM)观察,并对胚乳结构进行了荧光倒置显微镜、透射电镜(TEM)观察,以揭示GA_3对紫斑牡丹种子下胚轴解除休眠过程中种子种皮、胚乳结构变化和生根的影响。结果表明:(1)GA_3处理能够促进紫斑牡丹种子生根,以300 mg/L GA_3处理的种子生根效果最好,与对照相比可提前14.66 d生根,生根率可达62.33%。(2)扫描电镜观察发现,紫斑牡丹种皮表面含有蜡质,种子萌发进程中蜡质明显不断增加,种皮表面网眼逐渐消失,种皮皱缩明显。(3)光学显微镜观察发现,GA_3处理的紫斑牡丹种子胚乳细胞中含有较多淀粉体和蛋白体,并随着种子萌发进程逐渐积累,可为种子萌发提供足够的养分。(4)透射电镜观察发现,GA_3处理的紫斑牡丹种子胚乳细胞内大分子营养物质不断降解,线粒体、内质网、高尔基体等细胞器相继出现,种子下胚轴休眠解除,细胞壁较薄后期消融,利于营养物质传送。研究认为,适宜浓度GA_3处理可有效加快紫斑牡丹种皮结构的改变和胚乳细胞内物质代谢进程,促进种子萌发,且300 mg/L GA_3处理紫斑牡丹种子24 h可以获得最好的生根效果。  相似文献

Abstract

Qualitative and quantitative analysis of free and bound amino acids and amides during dormancy and the most important phases of the first cell cycle was carried out in tubers of Helianthus tuberosus.

In the dormant tuber arginine was confirmed to be the most abundant amino acid. A high amount of asparagine was also present; on the contrary glutamine was found in very low concentrations. During the progression of dormancy, all the free amino acids and amides declined while aspartic and glutamic acid increased.

During the G1 phase of the first cell cycle induced by 2,4-D, all the free amino acids and amides decreased with the exception of glutamic acid.

At 18, 20, 24 h of activation with 2,4-D, corresponding to the S phase and the beginning of mitosis, bound amino acids were also determined. In these phases of the cell cycle they increased reaching a maximum at 20 h; on the other hand the free amino acid and amide content, especially aspartic acid, asparagine and arginine, decreased with the exception of glutamic acid, alanine and phenylalanine.  相似文献

Application of gibberellic acid (GA3) to dormant Avena fatua L. caryopses resulted in the termination of dormancy within 24 h as indicated by germination between 24 and 48 h. During the period of imbibition from 0 to 24 and 24 to 48 h changes occurred in protein and carbohydrate metabolism in GA-treated and untreated caryopses. Germination did not occur in untreated caryopses, therefore physiological changes in these caryopses were not associated with the termination of dormancy. GA-treatment increased the concentration of soluble and SDS-extractable protein in the endosperm tissue by 4 and 5%, respectively, over the 24 h untreated material; no changes were apparent when the protein profiles of GA-treated and untreated tissues were compared by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) 0, 24 and 48 h after imbibition. The concentration of hexose and sucrose in the GA-treated endosperm tissue increased 189 and 151 μmol, respectively, over the untreated material at 24 h. Gibberellic acid had no effect on starch metabolism in the endosperm tissue in the first 24 h, the period associated with the termination of dormancy. The concentration of hexose increased by 57 μmol and starch decreased by 80 μmol in the GA-treated embryo tissue within 24 h. Our results demonstrate that exogenously applied GA influences sucrose and hexose metabolism in the endosperm tissue. The specific effect of GA on starch and hexose metabolism in the dormant A. fatua caryopsis embryo tissue may be associated with the termination of dormancy.  相似文献

Abstract

New perspectives in the study of factors which control seed germination. — Seedlings of Triticum durum, cv. « Cappelli », coming from unirradiated embryos grafted on to irradiated endosperms (EM(u)/EN(i) of presoaked seeds (in distilled water for 24 h. at 20[ddot]C.) (treatment: X-rays, doses, 2,4,6,8,10 and 20 Kr), grow more than seedlings of control EM(u)/EN(u) (dose 0) (fig. 1). To have this reaction, it is necessary that the used seeds be after-ripe; at the various stages of seed ripening, be ginning from the milk stage, the phenomenon is not present.

On this basis, the author has thought that a natural inhibitor occurs in the after-ripe endosperm of « Cappelli », which is neutralized or destroyed by X-rays.

As an experimental demonstration, some trials have been made of growing wheat seedlings in Petri dish, on moistened (distilled water) filter paper, together with excised embryos or isolated endosperms (fig. 2): the after-ripe endosperm is able to depress the seedling growth (fig. 3). In the same experimental conditions, X-rays, dose 6 Kr, neutralize the inhibition effect given by the endosperm. (fig. 4).

A completely different situation occurs in wheat seed, during its ripening: endosperm is inactive, embryo produces inhibition effects on the seedling growth, which, also in this case, are reduced by X-rays.

These phenomena, put in relation with dormancy in Triticum durum, cv. « Cappelli », which is a relative dormancy, having its maximum at the milk stage, have led the author to the general conclusion that, during dormancy, a germination inhibitor occurs in the embryo of wheat seed; when dormancy is finished, the inhibitor appears in the endosperm, in a situation which becomes stable and definitive.  相似文献

Abstract

Light and growth-promoting compounds, such as gibberellic acids (GA3), are among the most important factors that can break physiological seed dormancy. Here, we investigate the effects of GA3 and light on germination of five species of Resedaceae that are known to have different levels of physiological dormancy. Seeds were incubated at 20/30?°C in both 12-hr photoperiod and complete darkness. To study the effect of growth hormone on germination, seeds were soaked for 24?h in different concentrations of GA3 before sowing. The annuals (Reseda aucheri and Oligomeris linifolia) and the perennial Ochradenus arabicus had deep physiological dormancy, and exogenous application of GA3 enhanced their germination in the light, but not in darkness; few or no seeds germinated in the dark in these species. Ochradenus aucheri and O. baccatus had intermediate and non-deep dormancy, respectively, and application of GA3 enhanced their germination in both light and darkness. Germination of the annual species was much slower than that of the shrubby perennials. Overall, these results indicate that conditions under which seed developed, matured and stored on maternal plants as well as incubation conditions should be taken into consideration when assessing germination behavior of the perennial species of Resedaceae.  相似文献

The interaction between hormones and salinity on seed germination of three halophytic taxa in the genus Suaeda: S. maritima (L.) Dum. var. flexilis Focke and var. macrocarpa Moq., and S. depressa (Pursh) Wats, was studied. Exogenous applications of kinetin and gibberellic acid (GA3) were applied in order to determine if either of these growth-promoting hormones would promote germination in the two dormant taxa, Suaeda depressa and S. maritima var. flexilis and to see if osmotically induced dormancy by NaCl could be alleviated. Our results indicate that gibberellic acid is capable of breaking dormancy in these species with dormant seeds, but kinetin proved to be ineffective. A seed dormancy that was induced by osmotic stress could also be alleviated by treatments with gibberellic acid. Endogenous concentrations of both cytokinins and gibberellins were measured in seeds exposed to osmotic stress (0.85 M NaCl), and we found a reduction in cytokinin activity in these three taxa. Gibberellin-like activity was reduced in S. depressa when seeds were soaked in 0.85 M NaCl.  相似文献

Summary Theparthenocarpic fruit (pat) allele causes a complex syndrome affecting different aspects of tomato reproductive development. This mutation affects stamen (reduced length and carpelloidy), ovule (arrested integument growth and unviability), and ovary (autonomous growth, i.e., parthenocarpy) development;pat mutant plants therefore have reduced male and female fertility. We studied the phenotypic expression patterns of thepat gene after treatments with gibberellic acid (GA3) and under different growth seasons (late spring and autumn) and genetic backgrounds (backcross [BC] population after interspecific cross). GA3 treatments were only effective in restoring carpelloid anthers to the wild-type phenotype. Compared to late spring, mutant plants grown in autumn had a lower frequency of carpelloid anthers and aberrant ovules and a higher seed set. Inflorescence position also affected thepat expression; upper inflorescences had low frequency of short anthers and aberrant ovules and an increased tendency to set seeds,pat expressivity was more variable in BC1 plants segregating after interspecific cross withLycopersicon pennellii than in the originalL. esculentum line. Abstract

Seed formation involves not only the embryo and endosperm development, but also the formation of a series of either ephemeral or non-ephemeral structures. In this article, we study several of those structures in Cytisus multiflorus and Cytisus striatus. The endosperm development is first nuclear and later cellular, except for the chalazal area, whose development is always nuclear. It generates, in the early developmental stages, a sac-like haustorium. As the seed develops, two structures seem to be closely related to nutrient mobilization to the embryo sac: on the one hand, a group of cells and a channel, located in the chalazal area and closely related between them and to the endosperm haustorium, which could be interpreted as a hypostase and on the other hand, an endothelium, derived from the inner integument, which later degenerates leaving no trace in the mature seed. All of these structures would be associated with the directionality of assimilates from ovule tissues to embryo sac. In mature seed and surrounding the embryo appears a unicellular layer of cells rich in proteins (aleurone layer), which is the origin of the outermost layer of the cellular endosperm. The seed coat is made up only of the outer integument.  相似文献

Summary In henbit (Lamium amplexicaule L.) seeds, micromolar levels of gibberellic acid (GA3) introduced at the onset of imbibition markedly stimulated germination. Sensitivity to GA3 stimulation is rapidly lost if the seeds are pretreated in darkness at 15° for 1 day or more on a water substrate. Loss in responsiveness to GA3 is hastened by pretreatment in far-red or by dark pretreatment at higher (25°) temperatures and is reduced by pretreatments at 5°. The relationship between barley grain dormancy and gibberellic acid (GA3) responsiveness of aleurone layers has been investigated. Barley (Hordeum distichum L. cvs Triumph and Kristina) grains were matured under defined conditions in a phytotron. Grains of Triumph plants grown under long-day/warm conditions had lower dormancy levels than grains of plants grown under short-day/cool conditions. Aleurone layers isolated from grains of long-day Triumph plants secreted more α-amylase and had a higher responsiveness to GA3 as measured by α-amylase secretion. Storage of the grains increased both the percentage of germination and the responsiveness of the aleurone to GA3. Use of different sterilization methods to break dormancy confirmed the correlation between germination percentage and aleurone layer GA3 responsiveness. The response of embryoless Triumph grains to GA3 was lower than that of the isolated aleurone layers, suggesting a role of the starchy endosperm in regulating the GA3 response of the aleurone layer. Grains of the cultivar Kristina harvested from short day- and long day-grown plants lacked dormancy, and their isolated aleurone layers had a similar responsiveness to GA3 as measured by α-amylase secretion. The data indicate that the physiological state of the aleurone layers contributes to the percentage germination of the grains.  相似文献

The physiological characteristics of seed dormancy in Nicotiana plumbaginifolia Viv. are described. The level of seed dormancy is defined by the delay in seed germination (i.e the time required prior to germination) under favourable environmental conditions. A wild-type line shows a clear primary dormancy, which is suppressed by afterripening, whereas an abscisic acid (ABA)-deficient mutant shows a non-dormant phenotype. We have investigated the role of ABA and gibberellic acid (GA3) in the control of dormancy maintenance or breakage during imbibition in suitable conditions. It was found that fluridone, a carotenoid biosynthesis inhibitor, is almost as efficient as GA3 in breaking dormancy. Dry dormant seeds contained more ABA than dry afterripened seeds and, during early imbibition, there was an accumulation of ABA in dormant seeds, but not in afterripened seeds. In addition, fluridone and exogenous GA3 inhibited the accumulation of ABA in imbibed dormant seeds. This reveals an important role for ABA synthesis in dormancy maintenance in imbibed seeds. Germination and post-germination events have been compared in seeds of Cercis siliquastrum whose dormancy was removed by fulfilling the natural chilling requirement or by exogenous GA3 application.Compared to the chilled ones, the GA3 treated seeds showed precocious embryo growth and an earlier reserve mobilization, which started before radicle emergence.Although the hormonal application was interrupted at seed germination, the plantlets of Cercis siliquastrum that originated from GA3-supplied seeds were taller than those from chilled ones. Moreover, they produced a greater number of leaves but a reduced root mass and had some difficulty in maintaining a good water balance.Thus, the treatment of Cercis siliquastrum seeds with exogenous GA3 broke dormancy and induced germination, but also caused long-lasting consequences on morphogenesis of the growing plantlet.  相似文献

Changes of soluble and ionically bound peroxidase and indoleacetic acid (IAA) oxidase activities were followed during peach seed development. Soluble peroxidase activity was located mainly in the embryo plus endosperm tissue, whereas wall ionically bound activities were found predominantly in the integument tissue. The different peroxidase isoenzymes present in the extracts were characterized by polyacrylamide gel electrophoresis and isoelectric focusing; the main soluble isoenzyme of embryo plus endosperm tissue was an anionic isoperoxidase of R F 0.07. Basic ionically bound isoenzymes were located only in the integument tissue, but two soluble anionic isoenzymes of R F 0.23 and 0.51 were also present in this tissue. In parallel, peroxidase protein content was estimated specifically using polyclonal antibodies. The kinetic data and the changes of seed IAA oxidase activity during fruit development suggested that basic peroxidase isoenzymes from ionically bound extracts of integument might be involved in IAA degradation.
