The susceptibility of 20 widely distributed mill and table olive varieties to Bactrocera oleae (Rossi) as affected by irrigation, and fruit diameter and oil content was evaluated in a 3‐year trial in Southern Spain. Bactrocera oleae was bivoltine life cycle in the experimental site, with significant differences among population size throughout the study. Even though the olive fruit fly damaged all varieties, significant differences in susceptibility were detected. Among the mill olive varieties “Nevadillo Blanco de Jaén” was the most susceptible, with average infestation levels ranging between 6.7% and 52.2% and between 10.3% and 69.2% under rainfed and irrigated conditions, respectively, and “Arbequina” was the least susceptible, with average infestation levels ranging between 0.6% and 12.7% and between 2.3% and 18.5% under rainfed and irrigated conditions, respectively. Among the table olive varieties, “Gordal Sevillana,” “Ascolana Tenera” and “Ocal” were the most susceptible (with average infestation levels reaching 39.7%, 36.5% and 33.3%, respectively), while “Callosina” was the least susceptible (with infestation levels of only 8.4%). Irrigation tended to promote both B. oleae infestation and its earlier occurrence compared to the rainfed condition. Even though the diameter and oil content were positively correlated with B. oleae fruit infestation (correlation coefficients ranged between 0.5 and 0.95), the present work reveals that other yet‐unknown factors may influence B. oleae oviposition preferences. The results of this study can be useful for breeding programmes to develop olive varieties resistant to B. oleae and provide key information for wide‐area olive fly pest management decisions.  相似文献

Enzymatically active cell wall isolaled from olive (Olea europaea) fruit was employed Hi investigate some hydrolytic enzymes bound to the cell wall and the changes in these during ripening. Seven glycosidases. β-glucosidase (EC 3.2.1.21) α-galactosidase (EC 3.2.1.22). β-galactosidase (EC 3.2.1.23). α-arabinosidase (EC 3.2.1.55), α-mannosidase (EC 3.2.1,24). β-xylosidase (EC 3.2.1.37) and β-N-acetylglucosamidase (EC 3.2.1.30). as well as Cx-cellulase (EC 3.2.1.4) and endo-polygalacturonase (EC 3.2.1.15). were identified in the cell wall preparation, at four stages of ripeness (mature green. changing colour, black and black-ripe). Activities of all these cell wall-associated enzymes fionicallv and covalently linked) were determined either by cell wall incubation with artificial substrate or after extraction from the cell wall with buffers of high salt concentration (Cx-cellulase). and were compared to those of forms solubilized from acetone powders with 500 nM citrate buffer (cytoplasmic and/or apoplastic plus ionically hound to cell wall) In general, the activities of low ionic strength buffer-soluble enzymes were found to be much higher than those of the bound enzymes. The bound enzymes are present in the fruit at the green colour stage, whereas the activities of the soluble enzymes only increased from the changing colour stage onwards. The tenacity of binding of enzymes to the wall was investigated by treating the walls with high salt and measuring residual activity. The nature of the ionic and covalent binding and the changes during ripening were also established for wall-hound glycosidase During ripening there was a marked change in the percentages of covalently- and tonically linked activities of β-glucosidase and β-galaclosidase: al the changing colour stages about 75–80% of the bound active in was present in high ionic strength buffer while al the black-ripe stage it was only 15–20. A possible role for these cell wall degradative enzymes in olive softening is discussed.  相似文献

Abstract

The response of mature olive (Olea europaea L.) trees to both regulated deficit irrigation and fertigation was studied for some consecutive years in sub-humid (Italy) and semi-arid conditions (Spain). All cultivars tested in these experiments were preferentially for table olive consumption (pickling and double-aptitude). Data from these long-term studies on field-grown, mature trees are summarized for assessing common effects of water and nutrient supply on plant behaviour and fruit yield. Seasonal yield variations were observed, and showed that the effect of irrigation treatments was somewhat more gradual in the “on” than in the “off” year. Differences amongst cultivars were always marked. Dry matter accumulation in pulp and pit was positively affected by progressively increasing the amount of water supplied to plants, regardless of years and cultivars. The ratio between pulp and pit fresh weight was negatively affected by irrigation, while fruit size was positively influenced by the gradual increase of irrigation water. Average nitrogen levels in leaves were generally above the deficiency threshold at both experimental plantations. Crop yield was not consistently increased by addition of soil nutrients. The effect of fertigation treatments was relatively uninfluential on oil accumulation in fruits. However, polyphenols in oil samples decreased progressively as the addition of nutrients increased. Critical aspects and strategic decisions in modern olive growing are the selection of cultivar, the supply of water, the option of fertilization, together with other orchard management practices. In this paper we outline various aspects of long-term studies in experimental olive plantations with the aim of highlighting inter-regulatory processes, involving plant growth and crop yield, which must be taken into account when planning future field research and modelling exercises.  相似文献

Pistachio fruit hull borer, Arimania komaroffi Ragonot (Lep.: Pyralidae), is one the most important pests of pistachio in Iran. The larvae spin web as well as bore into young fruits, and the infested fruits fall off the trees. The second-generation adult moths appear in August and September, and their offspring feed on the fruit hull. Results indicated the presence of α-amylase, α-glucosidase, β-glucosidase, α-galactosidase, β-galactosidase and some proteases in the digestive tract of the pest. Highest activities of α-amylase, α-glucosidase, β-glucosidase, α-galactosidase and β-galactosidase were at pH 10, 7, 7, 6 and pH 6, respectively. Highest activities of trypsin, chymotrypsin and elastase of larval midgut were at pH 11. Zymogram analysis of α-amylase, α-glucosidase, β-glucosidase, tryptic, chymotryptic and elastase using native-PAGE revealed 1, 1, 2, 3, 3 and 2 bands of activity respectively, in A. komaroffi. One band was disappeared in the presence of the inhibitor TLCK, but no further inhibition by the inhibitors TPCK was observed. The results can be of help for designing new strategies for controlling the pistachio fruit hull borer based on natural proteases and carbohydrase inhibitors.  相似文献

The susceptibility of olive cultivars to the olive fruit fly, Bactrocera oleae (Rossi) (Diptera: Tephritidae), has seldom been studied. This article examines factors associated with olive fruit fly infestation of 16 commonly planted Sicilian olive cultivars. Total infestation data were simultaneously correlated with categorical and quantitative factors using ordinal logistic regression. When all factors were included in the analysis, year, sampling date, cultivar, and fruit color were highly significant, but the quantitative factors fruit volume, fruit elongation, and fruit hardness were not. When the analysis was repeated excluding cultivar, all quantitative factors were significant, and elongation and volume were highly significant. Spherical, large, and hard fruit seemed to be preferred by B. oleae over fruit that are elongate, small, and soft. Therefore, fruit color, elongation, volume, and hardness provide useful information regarding the susceptibility of cultivars. In both organic and conventional olive cultivation, information about olive cultivar susceptibility to olive fruit fly will help orchard managers to produce quality oil and table olives while reducing treatments for olive fruit fly control.  相似文献

The photosynthetic contribution of a fruit to its carbon requirement throughout ontogeny and under different growing conditions was quantified in cucumber ( Cucumis sativus L. cv. Corona). In addition, the effects of shading on fruit dry matter accumulation and the diurnal course of the elongation rate were studied. Fruit darkening had no photomorphogenic effect on fruit growth, while the cumulative photosynthetic contribution of a fruit to its own carbon requirement ranged from 1 to 5%. During the day there was always a net CO2 efflux. The photosynthetic rate per fruit, calculated as the difference between rates of CO2 exchange in light and dark, increased during fruit ontogeny, while the photosynthetic rate per unit fruit surface area declined. The latter was not dependent on fruit size. The photosynthetic activity per unit surface area of fruits was estimated to be about 20–30% as efficient as that of leaves. The rate of calculated photosynthesis was reduced by 60–65% when the photosynthetically active radiation incident on the fruit decreased from 200 to 50 μmol m−2 s−1. Temperature (20–30°C) had no pronounced effect on the rate of calculated fruit photosynthesis when fruits of the same developmental stage (temperature sum) were compared. However, the relative photosynthetic contribution of a fruit to its carbon requirement increased when temperature decreased. Moreover, this contribution increased when irradiance increased or fruit growth was reduced by competing fruits. During fruit ontogeny the daily photosynthetic contribution was highest (up to 15%) in young and old fruits, with a small growth rate.  相似文献

Olive productivity should be improved through stimulating nutrition, particularly under poor fertility soils. Consequently, the objective of this study was to assess the efficacy of applying organic and bio-fertilizers on the physiological growth, yield and fruit quality of olive trees under newly reclaimed poor-fertility sandy soil in an arid environment. During a field experiment carried out at El-Qantara, North Sinai, Egypt over two consecutive seasons (2019–2020 and 2020–2021), olive Kalamata trees were evaluated under three organic fertilizer treatments alone or in combination with three bio-fertilizers treatments. Organic fertilizer was applied as goat manure (16.8 kg/tree/year), or olive pomace (8.5 kg/tree/year) in mid-December of each season vs. untreated trees. The bio-fertilizers were applied as N-fixing bacteria (150 g/tree) was inculated in early March of each season, or amino acid mixture (1.5%) was applied three times, at 70% of full bloom, 21 days after full bloom, and a month later in comparison to a non-fertilized trees (control). The cultivar used was Kalamata, a dual-purpose cultivar for oil and table olives whose value increases when processed as table olives. The results indicated that the goat manure followed by olive pomace significantly enhanced photosynthetic pigments (chlorophyll a, b, and carotenoids), leaf mineral contents (N, P, K, Ca, Mg and Fe), tree canopy volume, number of flowers per inflorescence, number of inflorescences per shoot, initial fruit set, fruit retention. For fruit quality, fruit length and width, fruit weight, and total fruit yield was increased compared to the non-fertilized control. Likewise, The bio-fertilizer N-fixing bacteria followed by the amino acid mixture significantly improved all of the aforementioned parameters. Accordingly, it is recommended, both environmentally and economically to utilize organic and bio-fertizers, particularly goat manure combined with N-fixing bacteria, in low-fertility soil to sustain olive production as well as reducing mineral fertilization.  相似文献

This study investigated the fungi diversity of fresh olive (Olea europaea L.) fruits, olive paste (crushed olives) and olive pomace (solid waste) and screened and quantified enzymatic activities with biotechnological applications. Fungi were randomly isolated from olive cultivars from Castilla La Mancha region (Spain). Identification included comparison of their polymerase chain reaction (PCR) amplicons of the ITS1-5.8S-ITS2 ribosomal DNA region, followed by nucleotide sequence analysis. Fourteen different species with DNA sequences of different similarities were identified, belonging to seven different genera (Aspergillus, Penicillium, Rhizomucor, Mucor, Rhizopus, Lichtheimia and Galactomyces). Aspergillus fumigatus, followed by Galactomyces geotrichum, Penicillium commune and Rhizomucor variabilis var. regularior were the most frequent species. Specific enzyme screening was assayed on agar plates, using cellobiose, carboxymethylcellulose (CMC), polygalacturonic acid and CaCl(2)/Tween 80 as substrates for β-glucosidase, carboxymethylcellulase (CMCase), polygalacturonase and lipase, respectively. Species exhibiting the best activities were: Aspergillus fumigatus (for β-glucosidase, CMCase and lipase); Rhizopus oryzae (for β-glucosidase and lipase); Rhizomucor variabilis (for β-glucosidase, CMCase and polygalacturonase); Mucor fragilis (β-glucosidase, CMCase and lipase); Galactomyces geotrichum (for β-glucosidase, polygalacturonase and lipase) and Penicillium commune and Penicillium crustosum (for lipase). The species that had shown the best enzymatic activities were grown on hemicellulose, cellulose and pectin and some activities were quantified (xylanase, cellulase, β-glucosidase and pectinase). An isolate of A. fumigatus and one of A. niger showed the best cellulase and xylanase activities, while no species presented good pectinase and β-glucosidase activities. The selected species with potential enzymatic activities could be used for future applications of industrial interest.  相似文献

Solar irradiance and air temperature are characterized by dramatic circadian fluctuations and are known to significantly modulate fruit composition. To date, it remains unclear whether the abrupt, yet predictive, diurnal changes in radiation and temperature prompt direct metabolic turn‐over in the fruit. We assessed the role of fruit insolation, air temperature, and source‐tissue CO2 assimilation in the diurnal compositional changes in ripening grape berries. This was performed by comparing the diurnal changes in metabolite profiles of berries positioned such that they experienced (a) contrasting diurnal solar irradiance patterns, and (b) similar irradiance but contrasting diurnal CO2 assimilation patterns of adjacent leaves. Grape carbon levels increased during the morning and decreased thereafter. Sucrose levels decreased throughout the day and were correlated with air temperature, but not with the diurnal pattern of leaf CO2 assimilation. Tight correlation between sucrose and glucose‐6‐phosphate indicated the involvement of photorespiration/glycolysis in sucrose depletion. Amino acids, polyamines, and phenylpropanoids fluctuated diurnally, and were highly responsive to the diurnal insolation pattern of the fruit. Our results fill the knowledge gap regarding the circadian pattern of source‐sink assimilate‐translocation in grapevine. In addition, they suggest that short‐term direct solar exposure of the fruit impacts both its diurnal and nocturnal metabolism.  相似文献

In the processing field, there is a saying that “seed drugs be stir-fried”. Bitter almond (BA) is a kind of seed Chinese medicine. BA need be used after being fried. To distinguish raw bitter almonds (RBA) from processed products and prove the rationality of “seed drugs be stir-fried”, we analyzed the RBA and five processed products (scalded bitter almonds, fried bitter almonds, honey fried bitter almonds, bran fried bitter almonds, bitter almonds cream) using RP-HPLC fingerprints and chemometric methods. The similarity between RBA and processed products was 0.733∼0.995. Hierarchically clustered heatmap was used to evaluate the changes in components. Principal component analysis (PCA) was used for classification, and all samples are distinguished according to RBA and five processing methods. Six chemical markers were obtained by partial least squares discriminant analysis (PLS-DA). The content and degradation rate of amygdalin and β-glucosidase activity were determined. Compared with RBA, the content and degradation rate of amygdalin, and β-glucosidase activity were increased in bitter almonds cream. The content and degradation rate were decreased, and β-glucosidase was inactivated in other processed products. The above results showed that stir-frying had the best effect. The results showed that processing can ensure the stability of RBA quality, and the saying “seed drugs be stir-fried” is reasonable.  相似文献

 田间自然条件下在葡萄园群体水平上多年的研究证明:叶幕PAR光能截留率与葡萄群体净光合速率、叶幕和单叶PAR光能截留率与果实干物质总量占地上部生物量干重的百分比、与果实总糖产量占果实本身干物质总量的百分比、以及与果皮色素产量占果实本身干物质总量的千分比之间呈现显著或极显著的线性正相关关系。说明利用叶幕结构变异调节叶幕PAR光能截留和分配,可以对光合同化物源库关系和果实中物质代谢方向进行有效的调控。PAR光能截留率较高和分配合理的叶幕,不但通过较高的群体光合速率为产量和品质形成提供了丰富的同化物“源”,而且通过调节器官间“库”关系使同化物以较高的比例流向果实,同时使果实中物质代谢过程有利于合成构成品质的要素。  相似文献

The developmental types of secretory cavities in Citrus remain controversial. The relationship between secretory cavity development and the accumulation of essential oil in fruits of Citrus species is also unknown. In order to develop better insights into these problems, histological, histochemical, and cytochemical methods were used to investigate secretory cavity development and the accumulation of essential oll at different developmental stages of fruits of Citrus medica L. var. sarcodactylis (Noot.) Swingle. The results indicate that the secretory cavity of the variety seemed to originate from an epidermal cell and a subepldermal cell. These two cells underwent successive divisions, resulting In the formation of two parts: (Ⅰ) a conical cap; and (Ⅱ) a globular gland. The formation of the lumen was schlzolysigenous. Regular changes in the size of vacuoles and the accumulation of essential oil were revealed during the process of secretory cavity development. In addition, when fruits were a light yellow or golden color, the structure of secretory cavities was well developed and the content of essential oil in a single fruit reached a maximum. It would be most appropriate to collect the fruit as a medicinal material at this time.  相似文献

An integrated process has been developed, consisting of the “adsorption–separation” of cellulase enzymes to selectively remove β-glucosidase, and multi-stage enzymatic hydrolysis of corncob residues from xylo-oligosaccharides manufacture with the β-glucosidase deficient cellulase, aiming to obtain a high yield of cello-oligosaccharides production. After the “adsorption–separation” process, 79.50% of the endo-glucanase was retained in substrate, whereas 90.67% of β-glucosidase was removed with the separated liquid fraction, utilizing the different adsorbability of these enzymes to the substrate. A three-stage enzymatic hydrolysis of corncob residues with the β-glucosidase deficient cellulase was proposed in which the first, the second and the third stage were conducted for 6, 6 h and 12 h, respectively. Analysis indicated that the removal of hydrolysis products (glucose and cello-oligosaccharides) at each stage improved cello-oligosaccharides productivity and enzymatic hydrolysis yield. The cello-oligosaccharides yield and enzymatic hydrolysis yield in three-stage enzymatic hydrolysis were significantly improved to 51.78% and 75.56%, respectively, which were 36.00% and 25.10% higher than single-stage hydrolysis with original cellulase enzymes.  相似文献

Olive fruit characteristics (weight, pulp/stone ratio, and oil and moisture content) and the iodine value (IV) of 31 new olive progenies (Olea europaea L.) were determined. To evaluate the effect of the genetic variability on these parameters, the new olive progenies, obtained through cross‐pollination between Tunisian and Mediterranean olive cultivars, were planted in a selected grove guaranteeing the homogeneity of the pedologic and climatic conditions. A strong genetic effect and significant differences between genotypes were obtained for the IV and the fruit characteristics evaluated. Discriminant analysis was used to classify the new progenies as distinct from each other, based on their IV, and their pulp and stone weight. An almost full discrimination of the olives from different genotypes was only achieved when the fruit characteristics (pulp and stone weight) and the IV data were analyzed together.  相似文献

The total proteins of four strawberry cultivars with differing resistance and/or susceptibility to grey mould were extracted by two different methods, phenol and sodium dodecyl sulphate (SDS). Compared to phenol extraction, SDS extraction produced higher protein yields but poor quality protein bands with a high degree of streaking indicating possible contamination. In contrast, the total proteins obtained by phenol extraction could be used in two-dimensional polyacrylamide gel electrophoresis (2-DE) because of their higher quality, and the protein spots were well separated for all tested genotypes. From all the cultivars studied in this research, we obtained 89 differential (unmatched) protein spots that may be related to fruit disease resistance or susceptibility. Using the SPSS 11.0 software, we obtained a cluster of the four strawberry cultivars. The cultivars “APF029-4” and “SJ8976-1” were in the same subcluster with the highest Pearson correlation coefficient (0.705). Afterwards, they were amalgamated into the second group with “Joliette”, a disease-resistant cultivar, which means that they might have a similar resistance to grey mould.  相似文献

The photosynthetic, optical, and morphological characteristics of a chlorophyll‐deficient (Chl‐deficient) “yellow” soybean mutant (MinnGold) were examined in comparison with 2 green varieties (MN0095 and Eiko). Despite the large difference in Chl content, similar leaf photosynthesis rates were maintained in the Chl‐deficient mutant by offsetting the reduced absorption of red photons by a small increase in photochemical efficiency and lower non‐photochemical quenching. When grown in the field, at full canopy cover, the mutants reflected a significantly larger proportion of incoming shortwave radiation, but the total canopy light absorption was only slightly reduced, most likely due to a deeper penetration of light into the canopy space. As a consequence, canopy‐scale gross primary production and ecosystem respiration were comparable between the Chl‐deficient mutant and the green variety. However, total biomass production was lower in the mutant, which indicates that processes other than steady state photosynthesis caused a reduction in biomass accumulation over time. Analysis of non‐photochemical quenching relaxation and gas exchange in Chl‐deficient and green leaves after transitions from high to low light conditions suggested that dynamic photosynthesis might be responsible for the reduced biomass production in the Chl‐deficient mutant under field conditions.  相似文献

为揭示灵武长枣(Ziziphus jujuba ‘Lingwu Changzao’)果实发育过程中阿拉伯半乳糖蛋白(AGPs)的分布规律,以膨大前期、快速膨大期、着色期和完熟期灵武长枣果实为试验材料,通过组织化学和免疫荧光定位的方法,研究了不同发育时期果实AGPs的分布特征。结果显示:βGlcY-AGPs形成的棕红色沉淀和MAC204抗体识别的抗原在各时期果实的外果皮及相邻的内部数层排列紧密的中果皮小细胞的细胞壁和细胞内部均有分布。中果皮大型卵圆形薄壁细胞的细胞壁和细胞内在膨大前期均有βGlcY-AGPs形成的棕红色沉淀和MAC204抗体识别的抗原分布,而在快速膨大期、着色期和完熟期均主要分布于薄壁细胞的细胞壁上,大部分细胞内部无分布;随着果实发育成熟,中果皮薄壁细胞间隙拉大排列更加松散,出现细胞破裂,βGlcY-AGPs形成的棕红色沉淀和MAC204抗体识别的抗原分布逐渐减少。各时期果实维管束中维管束鞘、木质部、韧皮部、形成层的所有细胞的细胞壁和细胞内部都分布有βGlcY-AGPs形成的棕红色沉淀和MAC204抗体识别的抗原,维管束数量和大小随果实发育及体积的进一步增大逐渐减少,βG...  相似文献

β-glucosidase (BG) was believed to take part in abscisic acid (ABA) synthesis via hydrolysis of ABA glucose ester to release active ABA during plant growth and development. However, there is no genetic evidence available to indicate the role of genes during fruit ripening. Here, the expression patterns of three genes (VvBG1, VvBG2, and VvBG3) encoding β-glucosidase were analyzed during grape fruit development, and it was found that β-glucosidase activity increased in grape fruit in response to various stresses. Furthermore, to verify the function of β-glucosidase during fruit ripening, heterogeneous expression of the VvBG1 gene in strawberry fruit was validated, and the results showed that the VvBG1 over-expression increased β-glucosidase and promoted the fruit ripening process in strawberry. In addition, we found that ABA contents increased in the VvBG1 over-expression of strawberry fruit, which induced fruit anthocyanin, soluble solid accumulation, and fruit softening. Moreover, genes related to coloring (CHS, CHI, F3H, and UFGT), softening (PG1, PL1, and EXP1), and aroma (SAAT, and QR) were up-regulated. This work will elucidate the specific roles of VvBGs in the synthesis of ABA and provide some new insights into the ABA-controlled grape ripening mechanism.  相似文献
