Riassunto Il mantenimento di collezioni di funghi e di lieviti, anche piccole, costituisce un problema di tempo e di validità, perchè si è costretti a passare mensilmente i ceppi e, tuttavia, è frequente l'inquinamento ed ancor più la pleomorfizzazione dei ceppi.Nel 1939Castellani propose un metodo di mantenimento basato sull'uso di fiale contenenti acqua distillata e chiuse alla fiamma.Con qualche variante la validità di tale metodo fu confermata daCastagnetta &Mungelluzzi nel 1961 e daBenedek nel 1963.Nella presente comunicazione gli AA. riferiscono sui risultati ottenuti con tale metodo in tre anni di studi, risultati che possono ritenersi eccellenti.Recentemente tale metodo è stato applicato al mantenimento di collezione di germi. Si è ottenuta la conservazione per sei e più mesi di ceppi di stafilococco e di altri germi, tra i qualiPseudomonas aeruginosa, Proteus e qualcheEscherichia.
Summary The preservation of collections of fungi and yeasts, even small ones, requires much care and much time, but there is always a vast number of strains which get lost by contamination and mainly due to the phenomenon of pleomorphism.In 1939Castellani proposed a method of preservation using vials containing distilled water, sealed over flame.With small variations this method was confirmed byCastagnetta &Mungelluzzi in 1961 and byBenedek in 1963.Our present communication refers to our experience with this method that we have being using for three years with excellent results.Recently this method was also successfully used for the collection of bacteria, to preserve staphylococci and other microorganisms, among thesePseudomonas aeruginosa, Proteus and sometimesEscherichia.

Abstract

About some « Carex » new for the Marche Region or for the Adriatic side of the Italian Peninsula. — We give information about the presence in the Marche region of six new or unpublished Carex in this district; they are: C. distachya Desf., C. Grioleti Roem., C. pallescens L., C. nitida Host, C. olbiensis Jord. and C. depauperata Good. Amongst them, we must consider C. Grioleti as new in the whole Adriatic basin, while C. nitida, C. olbiensis and C. depauperata result as new or unpublished in the whole Adriatic side of the Italian Peninsula, or in a good part of it. Information is given about the distribution and the ecology of every single species in this district; we linger especially over the C. Grioleti, for the particular importance and significance of this taxon.  相似文献

Abstract

HISTOGENESIS AND ANATOMICAL CHARACTERISTICS OF PORTIONS OF SOLANUM TUBEROSUM SPROUTS CULTIVATED IN VITRO. — Disks cut from sprouts of Solanum tuberosum L. have been cultured on White's basic or modified medium (White, 1943) with 20 mg/l KH22PO4 50 mg/l adenine, and 12 or 24 mg/l NAA.

Sections were prepared according to Morel's method (1948). Only on the disks cultured on White's modified medium with 12 mg/1/NAA growth glomerules and a normal meristematic layer were present, while there were no signs of cellular hypertrophy and organogenetic phenomena.

12 mg/l appears therefore to be the optimal dose of NAA for cultures of excised portions of potato sprouts. Adenine is however always necessary because it triplicates the growth activity of the tissues and because through its antagonism with regard to auxin it favours the genesis of the callus.  相似文献

Abstract

On the occurrence in Southern Italy of some benthic marine algae rare to the Mediterranean Sea. - The Authors report the occurrence along the coast of Southern Italy of the following species that resulted rare to the Mediterranean Sea: Antithamnion piliferum Cormaci et Furnari; Bonnemaisonia asparagoides (Woodward) C. Agardh (tetrasporophyte); Corynophlaea flaccida Kuetzing; Fosliella farinosa (Lamouroux) Howe: of this very common species, the occurrence of thalli with propagules is reported; Hypnea cervicornis J. Agardh; Jania adherens Lamouroux, new to Italy; Mesogloia lanosa P.L. et H.M. Crouan, new to Italy; Pneophyllum confervicolum (Kuetzing) Chamberlain f. minutulum (Foslie) Chamberlain, new to the Mediterranean Sea; Polystrata fosliei (Weber van Bosse) Denizot, new to Italy; Rhodymenia delicatula Dangeard; Spermothamnion johannis G. Abstract

A simple procedure for the biosynthetic preparation of carbon-C11 labelled D-glucose and D-fructose. — C 11-uniformely-labelled D-glucose and D-fructose have been prepared by exposing to C 11 O 2 leaves of Beta vulgaris var. saccharifera and Nicotiana tabacum. From 24 to 36 percent of the radioactivity supplied to the leaves was recovered in the two monosaccharides. The specific activity of D-glucose ranged from 8.7 to 18 microcuries per micromole while values from 8.3 to 20.8 were obtained in the case of D-fructose. The higher specific activities were always obtained when N. tabacum was employed. As compared to the radioactive monosaccharides further purified by two-dimensional paper chromatography, D-glucose and D-fructose prepared according to the proposed procedure showed a radiochemical purity of 98.7 and 99.8 percent, respectively.

After exposure to C 14 O 2 (final concentration 0.345 percent, by volume) for 24 hours at room temperature and 700 lux, the leaves were homogenized with quartz and extracted with water and 80 percent ethanol. The sugars were purified first by chromatography on a column of cocoanut charcoal, then by preparative paper chromatography. The time required for a complete experiment is of approximately 17 hours.

The procedure may be conveniently applied to preparations employing from a few microcuries up to 100 millicuries of BaC 14 O 3. No special equipment is required.  相似文献
