In a chemosystematic investigation of Wulfeniopsis amherstiana we have isolated four common iridoid glucosides as well as the esters 3″- and 4″-Cinnamoyl 6-O-rhamnopyranosylcatalpol. Furthermore the plant contained mannitol, arbutin, and four caffeoyl phenylethanoid glucosides, namely plantamajoside, aragoside and two new acetyl derivatives of the latter, named amherstianoside A and B. From Lagotis integrifolia we have obtained mannitol and three common iridoid glucosides together with the 8,9-unsaturated iridoids arborescosidic acid and anagalloside. The results show that the genus Wulfeniopsis is chemically different from Wulfenia and that it is closer related to Veronicastrum.  相似文献

Previous proposals for subdivision of the genus Cornus are reviewed. Twenty-seven authenticated Cornus species have been studied for their content of non-flavonoid glucosides. Two distinct groups of taxa can be distinguished on this basis: one, containing a hydroxycyclohexadienone glucoside, along with salidroside, its reduced counterpart, and the second, characterized by containing various iridoid glucosides. The two types of glucosides are apparently mutually exclusive.  相似文献

Countless reports describe the isolation and structural characterization of natural products, yet this information remains disconnected and underutilized. Using a cheminformatics approach, we leverage the reported observations of iridoid glucosides with the known phylogeny of a large iridoid producing plant family (Lamiaceae) to generate a set of biosynthetic pathways that best explain the extant iridoid chemical diversity. We developed a pathway reconstruction algorithm that connects iridoid reports via reactions and prunes this solution space by considering phylogenetic relationships between genera. We formulate a model that emulates the evolution of iridoid glucosides to create a synthetic data set, used to select the parameters that would best reconstruct the pathways, and apply them to the iridoid data set to generate pathway hypotheses. These computationally generated pathways were then used as the basis by which to select and screen biosynthetic enzyme candidates. Our model was successfully applied to discover a cytochrome P450 enzyme from Callicarpa americana that catalyzes the oxidation of bartsioside to aucubin, predicted by our model despite neither molecule having been observed in the genus. We also demonstrate aucubin synthase activity in orthologues of Vitex agnus-castus, and the outgroup Paulownia tomentosa, further strengthening the hypothesis, enabled by our model, that the reaction was present in the ancestral biosynthetic pathway. This is the first systematic hypothesis on the epi-iridoid glucosides biosynthesis in 25 years and sets the stage for streamlined work on the iridoid pathway. This work highlights how curation and computational analysis of widely available structural data can facilitate hypothesis-based gene discovery.  相似文献

From three of five investigated species of Griselinia a new iridoid glucoside, griselinoside, was isolated. It was found to be present also in foliage of Aralidium pinnatifidum and Toricellia angulata, accompanied in the former by aralidioside another novel iridoid glucoside. The structures and absolute configurations of the two iridoids were elucidated by NMR spectroscopy and chemical conversions. From G. littoralis and T. angulata the glucosides magnolioside and syringoside respectively were isolated. 13C NMR spectra are given for thirteen iridoid derivatives.  相似文献

Activity guided isolation of a MeOH extract of the aerial plant parts of Wulfenia carinthiaca Jacq . (Plantaginaceae), using a mushroom tyrosinase assay, resulted in the isolation of five phenylethanoid glucosides and four iridoid glycosides. Two of them, 2′‐O‐acetylisoplantamajoside and 2′,6″‐O‐diacetylisoplantamajoside, represent new natural products. Evaluation of the inhibitory activity of all isolated compounds revealed that the observed activity is not related to the isolated phenylethanoid glycosides but mainly due to the presence of the iridoid glycoside globularin (IC50 41.94 μm ; CI95% ± 16.61/11.89 μm ). Interestingly, structurally close related compounds (globularicisin, baldaccioside, and isoscrophularioside) showed no or only a weak tyrosinase inhibitory activity.  相似文献

Four iridoid glucosides, three of which are dimeric were isolated from the methanol extract of roots of Vietnamese Paederia scandens (Lour) Merrill together with the five known glucosides, paederoside, asperuloside, paederosidic acid, asperulosidic acid and geniposide. Seven sulfur-containing iridoid glucosides were also isolated. The structures of the iridoid glucosides were determined by a combination of high-resolution NMR, MS, IR and UV spectra, and chemical reaction such as acetylation.  相似文献

Twp new secoiridoid glucosides, abeliosides A and B, were isolated along with cantleyoside and sylvestroside II from Abelia grandiflora. On the basis of spectral and chemical evidence, these new glucosides were identified as esters of secologanic acid with an iridoid lactone which may arise from loganin. In addition to the known iridoid glucosides loganin, sweroside and cantleyoside, Dipsacus sylvestris has provided 4 novel bis-iridoid glucosides named sylvestrosides I–IV, composed of swerosidic acid, secologanic acid, loganin and loganin aglucone. Larvae and pupae of the alpine butterfly Euphydryas cynthia contain three iridoid glucosides: two of them, aucubin and catalpol, are very common in the Plantaginaceae, but the third, 6-O-glucopyranosylaucubin, has hitherto only been isolated from Odontites verna and Verbascum sinuatum (Scrophulariaceae). The amounts of the three iridoids found in the insects were 0.53, 0.31 and 1.48 % dry wt, respectively. Preliminary feeding experiments with an insectivorous bird indicated some unpalatability in the insect, which probably stems from the iridoid content.  相似文献

Besides the expected iridoid glucosides aucubin and catalpol as well as three known esters of the latter, Veronica beccabunga (brooklime) was shown to contain five carboxylated iridoid glucosides, namely gardoside, mussaenosidic acid, 8-epiloganic acid, arborescosidic acid and alpinoside. In addition to these compounds, the plant contained salidroside and a previously unknown caffeoyl phenylethanoid glycoside (CPG) which we have named chionoside J. The structure was elucidated mainly by 1D and 2D NMR spectroscopy to be 2″-(β-glucopyranosyl)-plantamajoside. The distribution of plantamajoside and its derivatives as well as that of carbocyclic iridoids with an 8,9-double bond is briefly discussed, and it is noted that such compounds are mainly confined to the tribe Veroniceae of the Plantaginaceae.  相似文献

Two iridoid glucosides namely; 6-acetylantirrinoside (1), 6'-O-p-hydroxybenzoylantirrinoside (2) were isolated from the aerial parts of Kickxia abhaica. Beside that, three known iridoid glucosides, antirrinoside (3), antirride (4) and mussaenosidic acid (5), one flavone glycoside (6) and a hexitol, d-mannitol (7) were isolated. The structures of the iridoid glucosides 1-2 were established by 1D and 2D NMR spectral data, including COSY, HMQC and HMBC experiments, as well as HRMS.  相似文献
